禽流感病毒(Avian influenza virus,AIV)不仅严重危害禽类,而且对人类生命健康造成严重威胁。鸽作为留鸟,具有作为AIV从野生鸟类传播至人类中间宿主的潜能。鸽对AIV的易感性以及在病毒传播中的作用却存在争议。通过分析AIV自然感染、人工感染鸽的流行病学以及实验研究数据,同时回顾了禽流感病毒感染鸽的机制,发现随着病毒的进化和时间的推移,鸽群AIV的感染率也在递增;尤其随着近年具有双受体结合特性的高致病性禽流感病毒(highly pathogenic avian influenza virus,HPAIV)clade2.3.4.4分支H5Nx毒株的出现,其感染鸽后排毒量上升以及鸽体间直接接触传播能力增强。为了有效防控AIV的跨种间传播,有必要加强对鸽感染AIV的流行病学监测和传播特性的研究,特别需要密切关注具双受体结合特性的H5Nx和H7N9 HPAIV对鸽易感性的发展趋势。  相似文献

H9N2亚型禽流感病毒(AIV)于1966年从北美的火鸡群中首次被分离以来。自20世纪90年代以来,H9N2病毒已经在世界多个国家和地区发展成地方流行并形成了稳定的种系,是当前禽流感流行的主要亚型之一,严重危害养禽业的发展。此外,H9N2亚型AIV也可以感染猪,甚至能够直接感染人,具有重要的公共卫生意义。H9N2亚型AIV还可以为H5N1、H7N9以及H10N8等能够直接感染人并致死的AIV提供部分甚至整套内部基因,潜在危害备受关注。对H9N2亚型AIV在我国的流行和进化特点进行了简要综述。  相似文献

建立一步法RT-PCR检测方法,对禽流感病毒(Avian influenza virus,AIV)的血凝素(Hemagglutinin,HA)分型进行了研究。参照AIV的HA基因序列设计1对引物,对H9和H5亚型AIV进行了扩增,产物大小分别为579bp和177bp。经测试,该引物不与新城疫病毒等鸡的其它传染性病原及鸡肌肉组织的核酸发生交叉反应。敏感性分析发现,从50pg的AIV总RNA中亦能扩增到目的条带。结果表明,此次利用1对引物建立的一步法RT-PCR方法简便适用,可以在一次反应中同时将H9和H5亚型AIV进行快速检测和分型。另外,两个亚型的扩增产物均包含了HA裂解位点在内的基因序列,可通过测序推导氨基酸顺序以预测H5或H9亚型禽流感病毒的潜在毒力。  相似文献

目的 检测和鉴定A型、H5亚型禽流感病毒 (AIV) ,研发一种高效实用的检测手段。方法 根据Ming ShiuhLee报道的文献设计、合成引物 ,采用反转录和PCR一步法对A型、H5亚型禽流感病毒cDNA进行扩增和电泳鉴定 ,组装成禽流感病毒RT PCR试剂盒 ,对H1~ 15亚型AIV参考株、38份AIV国内分离株进行检测试验。结果 建立了A型、H5亚型禽流感病毒RT PCR检测方法 ,并在此基础上组装试剂盒 ,用A型试剂盒检测时 ,全部AIV毒株均为阳性 ,能检测 1 10 2 4血凝单位禽流感病毒 ;用H5亚型试剂盒检测时 ,仅有H5亚型AIV参考株和 19株H5亚型AIV分离株呈阳性 ,其余H1~H4、H6~H15参考株和H7、H9分离株以及 1株H5分株均为阴性 ,能检测1 6 4血凝单位禽流感病毒。 2种试剂盒对实验感染鸡病料检出率均为 10 0 %。结论 研制的AIVA型、H5亚型RT PCR试剂盒具有特异性强、敏感性高、稳定性和重复性好的特点。  相似文献

L-Arabinose isomerase (AI), a key enzyme in the microbial pentose phosphate pathway, has been regarded as an important biological catalyst in rare sugar production. This enzyme could isomerize L-arabinose into L-ribulose, as well as D-galactose into D-tagatose. Both the two monosaccharides show excellent commercial values in food and pharmaceutical industries. With the identification of novel AI family members, some of them have exhibited remarkable potential in industrial applications. The biological production processes for D-tagatose and L-ribose (or L-ribulose) using AI have been developed and improved in recent years. Meanwhile, protein engineering techniques involving rational design has effectively enhanced the catalytic properties of various AIs. Moreover, the crystal structure of AI has been disclosed, which sheds light on the understanding of AI structure and catalytic mechanism at molecular levels. This article reports recent developments in (i) novel AI screening, (ii) AI-mediated rare sugar production processes, (iii) molecular modification of AI, and (iv) structural biology study of AI. Based on previous reports, an analysis of the future development has also been initiated.  相似文献

载脂蛋白AI(apolipoproteinAI,apoAI)是高密度脂蛋白HDL的主要组成成分 ,流行病学研究表明apoAI的含量决定血浆中HDL的高低 ,而HDL具有胆固醇逆向转运的功能 ,起到降低冠心病发生概率的作用。通过大肠杆菌表达系统来生产apoAI的蛋白原proapoAI,蛋白的表达形式为包涵体 ,通过疏水柱进行柱上复性。同时在proapoAI和apoAI之间设计一个酸水解位点 ,通过将蛋白原proapoAI进行酸水解来得到成熟蛋白apoAI。最终得到的成熟蛋白apoAI在结构分析和脂结合特性上都与天然的apoAI相似 ,从而为该蛋白的工业生产上提供了一个良好的基础。  相似文献

Conversion of AI to AII has been studied in the mesenteric circulation of the isolated perfused cat terminal ileum. Infusion of AI through the mesenteric circulation induced a significantly potentiated response when the venous return was superfused over the rat colon and the rabbit aortic strip. Addition of converting-enzyme inhibitor, SQ 20881 to the perfusion medium competitively prevented the potentiation of AI on the assay organs without altering its direct effects. The percent conversion of AI to AII was found to be 68 in the mesenteric circulation. In contrast, infusion of AII through the mesenteric circulation has lost about 40% of its biological activity as measured on the same assay organs. SQ 20881 abolished the inactivation of AII in the mesenteric circulation. It is concluded that the mesenteric circulation of the isolated perfused cat terminal ileum is one of the major conversion areas of AI to AII. SQ 20881 prevented the conversion of AI to AII as well as abolishing the inhibition of AII passing through the mesenteric circulation.  相似文献

The interactions of lysophosphatidylcholine and synthetic 1,2-dimyristoyl-sn-glycerophosphocholine (DMPC) liposomes with the isolated HDL-apolipoproteins, apo AI and apo AII, has been studied by microcalorimetry. Complex formation is a highly exothermal process characterized by a maximal enthalpy of about --200 kcal/mol of apoprotein when added to DMPC at 28 degrees C in 0.05 M sodium carbonate/bicarbonate buffer, pH 9.6. For the apo AI apoprotein, the binding consists of two processes, one endothermal occurring at low phospholipid/protein ratios and one exothermal predominant at higher phospholipid levels. The endothermal process has been attributed to a lipid-induced disaggregation of the apo AI while the exothermal process is similar to the binding of apo AII or apo HDL to phospholipids. The binding of a constant AI and apo AII, demonstrates the existence of a maximal association at a 1 : 1 molar ratio of the apolipoproteins. The sequential binding of DMPC to apo AI and apo AII suggests the existence of cooperativity between the two apoproteins in phospholipid binding as apo AII promotes the incorporation of apo AI into a protein-phospholipid complex.  相似文献

A human apolipoprotein AI (apo AI) minigene and two mutants were cloned into the vector pUHD10-1 for expression studies in COS cells under the control of the strong CMV (cytomegalovirus) enhancer and the own apo AI promoter. In the mutated apo AI minigene (mutant M1) the positions of the triplets of Gln(-2)-Gln-1 at the C-terminus of the prosequence were exchanged against Gln(-8)-Ala-7, the recognition site of the signal peptidase of the wild type human apo AI. The prosequence has been deleted in mutant M2 and the presequence linked directly to the N-terminus of the mature apo AI form. We report here on expression studies in COS cells, a cell line, which does not express apo AI. They were transfected by electroporation with pUHD10-1 constructs, which contain a) the wild type apo AI minigene and b) the two mutant apo AI minigenes with mutations described above. The following results were obtained: a) the wild type and mutant apo AI constructs were efficiently transcribed and translated in COS cells, b) the expression of the wild type preproapo AI minigene in COS cells led to the secretion of proapo AI (29 kDa), that of the mutant (M2) gene, devoid of the prosequence of mature apo AI (28.4 kDa), whereas the product of mutant gene M1 (31 kDa) with the recognition site of the signal peptides transposed to the C-terminus of the prosequence remained uncleaved within the COS cells.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Artificial intelligence (AI) has already been implemented widely in the medical field in the recent years. This paper first reviews the background of AI and radiotherapy. Then it explores the basic concepts of different AI algorithms and machine learning methods, such as neural networks, that are available to us today and how they are being implemented in radiotherapy and diagnostic processes, such as medical imaging, treatment planning, patient simulation, quality assurance and radiation dose delivery. It also explores the ongoing research on AI methods that are to be implemented in radiotherapy in the future. The review shows very promising progress and future for AI to be widely used in various areas of radiotherapy. However, basing on various concerns such as availability and security of using big data, and further work on polishing and testing AI algorithms, it is found that we may not ready to use AI primarily in radiotherapy at the moment.  相似文献

Cervical anatomy limits the use of transcervical intrauterine artificial insemination (TC AI) in sheep. We have developed an instrument to cope atraumatically with the cervix; although this instrument has not affected fertilization rate or pregnancy rate through Day 3, the effects on sperm transport and pregnancy after Day 3 are not known. The objective of the present study was to determine whether our TC AI instrument affected sperm transport, pregnancy rates, or lambing rate. In Experiment 1, ewes were assigned to two treatments: TC AI using the new TC AI instrument (n=10) or AI via laparotomy using a laparoscopic AI instrument (n=10). Twenty hours after artificial insemination, the uterine horns and oviducts were recovered and flushed to collect spermatozoa. Sperm transport did not differ (P>0.05) between the two treatments. In Experiment 2, ewes were assigned to three treatments: TC AI using the new TC AI instrument+sham intrauterine AI via laparotomy (n=29); sham TC AI+intrauterine AI via laparotomy using a laparoscopic AI instrument (n=29); and sham TC AI+intrauterine AI via laparotomy using the new TC AI instrument (n=30). On Day 14 after AI, uteri were collected and flushed to recover blastocysts. Transcervical deposition of semen reduced (P<0.05) Day 14 pregnancy rate (17.2% versus 61%), but intrauterine deposition of semen using the TC AI instrument via midventral laparotomy increased (P<0.05) Day 14 pregnancy rate (76.6% versus 44.8%). In Experiment 3, ewes were assigned to two treatments: sham cervical manipulation (n=40) or cervical manipulation to mimic TC AI (n=40). Immediately after treatment, each ewe was mated with a ram and watched until the ram mounted and ejaculated into the ewe. Treatment did not affect Day 30 or 50 pregnancy rate (67.5 and 66.2%, respectively), determined ultrasonically, or lambing rate (62.5%). The differences between Days 30 and 50 pregnancy rates and lambing rate were not significant. In Experiment 4, ewes were assigned to two treatments: TC AI (n=99) or laparoscopic AI (n=99). Transcervical AI reduced (P<0.01) Day 30 (TC AI versus laparoscopic AI; 5.0% versus 46.0%) and Day 50 pregnancy rates (4.0% versus 41.0%), determined ultrasonically, and lambing rate (4.0% versus 41.0%). Although the TC AI procedure significantly reduced pregnancy and lambing rates, large numbers of spermatozoa deposited at natural insemination seemed to compensate. Because our TC AI procedure has all but eliminated any visual evidence of trauma, and because the procedure does not seem to affect sperm transport or embryonal survival until Day 3, we speculate that cervical manipulation associated with TC AI may activate pathways that interrupt pregnancy between Days 3 and 14.  相似文献

The AI polypeptide chain from human high density serum lipoprotein has two accessible conformational states in aqueous solution. L-alpha-Palmitoyl lysophosphatidylcholine induces the transition between these two states at an equilibrium concentration of ligand of 2 X 10(-5)M, and the protein has a maximum binding capacity of 95 to 100 mol of lipid/mol of protein. The present study, together with previous investigations in this laboratory, suggests that the conformational state of AI in the presence of high levels of bound amphiphiles is similar to the in vivo state, and further, that this complex does not result from the insertion of AI into amphiphilic micelles. The mode of interaction of AI with amphiphilic ligands is shown to be significantly different from that of membrane proteins thus far investigated.  相似文献

The impact of artificial intelligence (AI) on the environment is the subject of discourse, with arguments for both positive and negative effects. There is a fine line between AI for good and AI for environmental degradation. Today, companies want to seize the benefits of AI, which distinctively involves reducing the company's carbon footprint. However, AI's carbon emissions differ as per the techniques involved in training it. As the saying goes, a coin always has two sides. Therefore, it cannot be denied that AI can be an effective tool for combating climate change, but its role in contributing to carbon emissions cannot be ignored. Multiple studies indicate that AI could be the game-changer in staving off anthropogenic climatic changes due to the deterioration of the environment and global warming. This double-edged relationship and interdependency of AI and carbon emissions are represented through a system of systems (SoS) approach. SoS states that a plan is created through multiple smaller systems, creating complexity in the design and vice versa. A complex system can be assumed as the world in general, where two individual independent systems AI and carbon emissions, when in interaction, create a complex complementary and contradictory relation, adding to the convolution of the system. This connection is demonstrated by conducting a network analysis and calculating the carbon emissions of six machine learning (ML) algorithms and deep learning (DL) models with different datasets but the same hyperparameters on a carbon emission calculator created through AI algorithms. The primary idea of this study is to encourage the AI society to create efficient AI models that may be used without compromising environmental issues. The focus should be on practicing sustainable AI, that is, sustainability from data collection to model deployment, throughout the lifecycle of AI.  相似文献

L-Arabinose isomerase (AI) catalyzes the isomerization of L-arabinose to L-ribulose. It can also convert d-galactose to d-tagatose at elevated temperatures in the presence of divalent metal ions. The araA genes, encoding AI, from the mesophilic bacterium Bacillus halodurans and the thermophilic Geobacillus stearothermophilus were cloned and overexpressed in Escherichia coli, and the recombinant enzymes were purified to homogeneity. The purified enzymes are homotetramers with a molecular mass of 232 kDa and close amino acid sequence identity (67%). However, they exhibit quite different temperature dependence and metal requirements. B. halodurans AI has maximal activity at 50 degrees C under the assay conditions used and is not dependent on divalent metal ions. Its apparent K(m) values are 36 mM for L-arabinose and 167 mM for d-galactose, and the catalytic efficiencies (k(cat)/K(m)) of the enzyme were 51.4 mM(-1)min(-1) (L-arabinose) and 0.4 mM(-1)min(-1) (d-galactose). Unlike B. halodurans AI, G. stearothermophilus AI has maximal activity at 65-70 degrees C, and is strongly activated by Mn(2+). It also has a much higher catalytic efficiency of 4.3 mM(-1)min(-1) for d-galactose and 32.5 mM(-1)min(-1)for L-arabinose, with apparent K(m) values of 117 and 63 mM, respectively. Irreversible thermal denaturation experiments using circular dichroism (CD) spectroscopy showed that the apparent melting temperature of B. halodurans AI (T(m)=65-67 degrees C) was unaffected by the presence of metal ions, whereas EDTA-treated G. stearothermophilus AI had a lower T(m) (72 degrees C) than the holoenzyme (78 degrees C). CD studies of both enzymes demonstrated that metal-mediated significant conformational changes were found in holo G. stearothermophilus AI, and there is an active tertiary structure for G. stearothermophilus AI at elevated temperatures for its catalytic activity. This is in marked contrast to the mesophilic B. halodurans AI where cofactor coordination is not necessary for proper protein folding. The metal dependence of G. stearothermophilus AI seems to be correlated with their catalytic and structural functions. We therefore propose that the metal ion requirement of the thermophilic G. stearothermophilus AI reflects the need to adopt the correct substrate-binding conformation and the structural stability at elevated temperatures.  相似文献
