本实验探讨赤楠果实红色素的理化性质及提取条件。结果表明,该色素属花色苷类;其最佳提取工艺是:1%HCl-乙醇(1ml HCl:99ml 95%乙醇)溶液作为提取剂,料液比1/10,温度35℃,提取时间3h,提取次数3次。  相似文献

轮叶蒲桃(Syzygium grijsii)系桃金娘科(Myrtaceae)蒲桃属(Syzygium)常绿灌木,其开发前景较好,但其叶绿体基因组特征及系统发育关系尚未有相关报道。为弥补轮叶蒲桃基因组学方面的空缺,该文对轮叶蒲桃的叶绿体基因组进行了系统的研究。运用Illumina高通量测序,并在GetOrganelle平台进行完整组装,同时利用组装好的数据分析轮叶蒲桃叶绿体基因组的结构特征和系统发育关系,其中包括轮叶蒲桃叶绿体基因组结构、功能及特征、密码子偏好性分析、叶绿体基因组的比较分析和系统发育的分析。结果表明:(1)轮叶蒲桃叶绿体基因组大小为158 591 bp,包含129个基因。其中,rRNA基因8个,tRNA基因37个,蛋白编码基因84个。分析检测到39个重复序列和84个SSR位点。(2)密码子偏好性分析发现轮叶蒲桃叶绿体基因组中末端存在对A/U的偏性,使用最多的是编码亮氨酸的密码子。(3)与近缘种比较,轮叶蒲桃的边界长度保守,边界处的基因种类与多个蒲桃属物种相似;轮叶蒲桃叶绿体基因组在LSC和SSC区变异度较大,有45处0.010i<0....  相似文献

福建省不同居群轮叶蒲桃叶形态与表皮特征比较   总被引:1,自引:0,他引:1  
通过对福建省16个不同居群轮叶蒲桃叶片形态特征、叶表皮特征的观察及叶片主要性状的测量,探讨不同居群轮叶蒲桃叶型、叶表皮特征多样性,为属下种间鉴定提供依据。对16个居群轮叶蒲桃叶型多样性进行观察,采用直尺测量叶片主要性状如叶长、叶宽等;采用光学显微镜和扫描电镜对叶表皮特征进行观察。结果表明,16个居群轮叶蒲桃叶片中,闽西居群叶形为阔椭圆形;闽东、闽北、闽南等各地居群叶形为长椭圆形或近带形。不同居群轮叶蒲桃叶片表皮毛、角质层纹饰、气孔器形态与类型等表现一致,但表皮细胞垂周壁式样差异明显,具波状、浅波状、平直、弓形等几种类型,且在上下表皮间存在式样一致或有多元化现象。由此说明,福建省不同居群轮叶蒲桃叶形与表皮细胞垂周壁式样均处于较强烈分化的状态,表皮细胞垂周壁式样不能作为属种间分类的依据。  相似文献

为生产各种食用品、化妆品、医药药品等色素新产品提供重要原料,研究榕树叶红色素的提取条件和稳定性。以水作提取剂,在室温下浸提24h,用紫外分析法研究色素的稳定性。色素在330am有最大吸收峰,该色素水溶性较好,pH值对色素的稳定性影响较大,但热稳定性良好。苯甲酸钠、柠檬酸、醋酸、蔗糖及葡萄糖等食品添加剂对色素无明显影响,淀粉对其影响较大,耐光性较差。氧化剂、还原剂中除抗坏血酸对色素的影响明显外,其他均无明显影响,金属离子对其影响较小。  相似文献

本文采用正交实验法对黑果枸杞色素的提取和精制工艺进行了研究。结果表明,黑果枸杞色素的最佳提取条件为:以pH 3.0的80%乙醇作浸提剂,提取温度50℃,提取时间3 h,固液配比1:40;用X-5大孔吸附树脂对色素进行精制,以树脂柱径高比1:15、流速3 mL/minp、H 3.0、色素液浓度1 g/L为最佳吸附条件,色素的吸附量可达0.03715 g/mL湿树脂体积;而以95%乙醇做洗脱液,在pH 2.0、流速5 mL/min、3倍于柱床体积的洗脱液条件下解吸附效果最佳,色素回收率达到97.78%;制取的色素产品外观呈紫红色,色价为36.7。  相似文献

采用正交试验对紫薇花红色素的最佳提取条件进行研究,并探讨该色素的稳定性。结果表明,乙醇浓度40%,料液比1:40(g/mL),提取时间90 min。该色素属水溶性色素,耐室内自然光,热稳定性好;在酸性条件下稳定;金属离子Na+、K+、Mg2+对紫薇花色素基本无影响,而Ca2+、Zn2+、Al3+、Cu2+、Pb2+、Fe2+离子对色素有明显影响。  相似文献

目的:研究五味子色素的提取工艺条件及色素稳定性。方法:在单因素实验基础上利用L9(3^4)正交试验确定最佳提取工艺;以吸光度值的变化为指标,研究色素稳定性。结果:最佳提取工艺为∶料液比1∶25,乙醇浓度45%,浸提温度70℃,浸提时间40min;五味子色素光稳定性较好;在酸性、弱碱性条件下稳定,当pH≥12时出现颜色变化;对糖、氯化钠有一定稳定性;抗坏血酸和Na2SO3对色素影响较小;苯甲酸钠和H2O2对色素影响显著;金属离子Ca2+、Mg2+、Na+对色素无明显影响,Fe2+、Cu2+、Al3+对色素影响显著。  相似文献

本试验研究了红穗醋栗色素的理化性质,结果表明:红穗醋栗色素热稳定性很好,而光照对其有一定的降解作用。pH值对该色素影响明显,宜在酸性食品中应用。几种金属离子Na~ 、Ca~(2 )、Al~(3 )、Zn~(2 )。Cu~(2 )对该色素的色泽没有影响;而Fe~(3 )、Sn~(2 )则有不良影响。食品中常含的几种添加物葡萄糖、蔗糖、淀粉和抗坏血酸对该色素无不良影响。红穗醋栗色素对Na_2SO_3的还原性耐性较强,对H_2O_2的耐氧化性很差。防腐剂苯甲酸钠对该色素也有一定的不良影响。  相似文献

研究了优美红游动菌(Rhodoplanes elegans)的生长以及类胡萝卜素的产生,通过比较4种广泛采用的细胞破碎方法,对优美红游动菌(Rhodoplanes elegans)类胡萝卜素的提取条件进行了研究,结果表明:类胡萝卜素的产生与菌种的生长曲线相符合,菌种活化后培养45h,类胡萝卜素的含量趋于稳定;取此时培养液,采用超声波法破碎细胞(破碎功率640W,时间10min),类胡萝卜素提取效果最好,初提液类胡萝卜素的提取率约为7.62mg·(g干菌体)-1;酸热法对色素的破坏严重。这为进一步开发利用天然色素资源提供了一定的理论依据。  相似文献

研究了光照、温度、pH值、氧化剂(H2O2)和还原剂(Na2SO3)对黑莓(Rubus spp.)果实色素水溶液稳定性的影响。结果表明,黑莓果实色素为水溶性花色苷类化合物,对光和热有较好的耐受性,对pH、H2O2和Na2SO3较敏感,且耐还原性略强于耐氧化性。在小于pH4.0、温度低于50℃及避光条件下黑莓果实色素的稳定性较好,可作为食品色素添加剂使用。  相似文献

The cDNA encoding a visual pigment of the locust Schistocerca gregaria has been inserted into the germline of the ninaE mutant of Drosophila melanogaster by P-element-mediated transformation. Functional expression has been documented by recording light-regulated electroretinograms in transgenic flies. The spectral properties of the expressed visual pigment were determined with detergent-solubilized material, prepared from the eyecups of the transgenic D. melanogaster. The recombinant locust pigment, as well as the genuine pigment of the fruitfly (Rh1) that served as a control for transformation/expression, showed photoreversibility between the pigment and metapigment forms. The absorptions of the difference spectra identify the locust visual pigment as a short wavelength-absorbing, blue-light-sensitive photoreceptor. The absorption maxima are similar to those recorded on living locust animals. These results show that, although locust visual pigments contain 11-cis retinal as chromophore, the expressed protein is able to adopt 3-hydroxyretinal that is provided by the transgenic fruitflies. The electrophysiological recordings reveal that the locust visual pigment is able to induce phototransduction in the fruitfly. The reported results have two important consequences: On the one hand, the binding site of the locust opsin is apparently able to interact with the 3-hydroxyretinal from Drosophila in a way that the biological signal generated by the photoisomerization of the chromophore can be used by the protein to adopt a physiologically active conformation. On the other hand, despite the relatively large phylogenetic distance between both insect species, the extent of conservation between the protein domains thought to be involved in G-protein activation is striking.  相似文献

An account is given of the development of green pigment in the early larval stages of the echiuran worm, Bonellia viridis . The green pigment first appears in scattered epidermal cells of the gastrula, before the development of the trochal rings. It is contained in spherical vacuoles (0.5-1.0 μm in diameter). As the gastrula develops into the trochophore there is an increase in total pigment content, as well as an increase in number of pigment cells and size of pigment vacuoles. The increase in pigment cell number in the trochophore results from the proliferation of the pigment cells of the gastrula. The pigment content increases rapidly before the trochophores hatch but shows little relative increase after hatching whilst the trochophores remain undifferentiated. We suggest that the pigment may have a protective role in the free-living trochophore for camouflage or chemical defence. In trochophores that undergo male differentiation the pigmentation is lost. Since the males do not lead a free-living existence but live as commensals inside the uterus of the female camouflage or chemical defence would be unnecessary. In trochophores undergoing female differentiation there is an increase in pigment content of the preoral lobe which develops into the proboscis. Following the enlargement of the coelom, clusters of coelomic cells accumulate green pigment whilst still in the coelom and then migrate to the integument of the body wall.  相似文献

Comparative electrophysiological and histological studies were made on the functional significance of the secondary iris pigment migration for the sensitivity of the eye in the noctuid moth Cerapteryx graminis. The pigment position at different adapting light intensities was studied as well as the influence of different positions on the sensitivity of the eye. Adapting light intensities above a certain value hold the pigment in light position. At a 3 log units lower intensity the pigment is brought into dark position and at light intensities between these limiting values the pigment attains intermediate positions. The results indicate that at light intensities between the limiting values the pigment shifts closely follow the changes in intensity of the environmental light. With the pigment in dark position the eye is about 1000 times more sensitive than when the pigment is in light position, there being a close relationship between the sensitivity of the eye and the position of the pigment at intermediate positions.  相似文献

The nature and stoichiometry of pigments in the Fenna-Matthews-Olson (FMO) photosynthetic antenna protein complex were determined by native electrospray mass spectrometry. The FMO antenna complex was the first chlorophyll-containing protein that was crystallized. Previous results indicate that the FMO protein forms a trimer with seven bacteriochlorophyll a in each monomer. This model has long been a working basis to understand the molecular mechanism of energy transfer through pigment/pigment and pigment/protein coupling. Recent results have suggested, however, that an eighth bacteriochlorophyll is present in some subunits. In this report, a direct mass spectrometry measurement of the molecular weight of the intact FMO protein complex clearly indicates the existence of an eighth pigment, which is assigned as a bacteriochlorophyll a by mass analysis of the complex and HPLC analysis of the pigment. The eighth pigment is found to be easily lost during purification, which results in its partial occupancy in the mass spectra of the intact complex prepared by different procedures. The results are consistent with the recent X-ray structural models. The existence of the eighth bacteriochlorophyll a in this model antenna protein gives new insights into the functional role of the FMO protein and motivates the need for new theoretical and spectroscopic assignments of spectral features of the FMO protein.  相似文献

【目的】分离鉴定雪白白僵菌红色素,分析红色素与环孢菌素A对底物缬氨酸的竞争和相互影响。【方法】对红色素进行分离纯化,利用UV、IR和ESI-MS对红色素进行初步鉴定。采用缬氨酸分批补料培养,通过控制溶氧水平,以及添加红色素,分析红色素与环孢菌素A合成之间的竞争关联及相互影响。【结果】经鉴定,雪白白僵菌红色素分子式为C15H10O5,推测为含有芳环结构的蒽醌类化合物。在补加缬氨酸和高DO条件下,环孢菌素的产量高于低DO水平,相反红色素在低DO条件下合成量大于高DO水平。在不补加缬氨酸条件下,实验结果与补加缬氨酸培养一致,但是红色素和环孢菌素A的产量都显著降低。进一步添加外源纯化的红色素时,随着添加量的增加出现了环孢菌素A合成先减弱后增加的变化。【结论】发现并证实了雪白白僵菌红色素与环孢菌素A合成都以缬氨酸为共同底物,但两者的途径又相互独立。  相似文献
