To test thehypothesis that intracellular Ca2+activation of large-conductanceCa2+-activatedK+ (BK) channels involves thecytosolic form of phospholipase A2 (cPLA2), we first inhibited theexpression of cPLA2 by treating GH3 cells with antisenseoligonucleotides directed at the two possible translation start siteson cPLA2. Western blot analysis and a biochemical assay of cPLA2activity showed marked inhibition of the expression ofcPLA2 in antisense-treated cells.We then examined the effects of intracellularCa2+ concentration([Ca2+]i)on single BK channels from these cells. Open channel probability (Po) for thecells exposed to cPLA2 antisenseoligonucleotides in 0.1 µM intracellularCa2+ was significantly lower thanin untreated or sense oligonucleotide-treated cells, but the voltagesensitivity did not change (measured as the slope of thePo-voltagerelationship). In fact, a 1,000-fold increase in[Ca2+]ifrom 0.1 to 100 µM did not significantly increasePoin these cells, whereas BK channels from cells in the other treatmentgroups showed a normalPo-[Ca2+]iresponse. Finally, we examined the effect of exogenous arachidonic acidon thePoof BK channels from antisense-treated cells. Although arachidonic aciddid significantly increasePo,it did so without restoring the[Ca2+]isensitivity observed in untreated cells. We conclude that although [Ca2+]idoes impart some basal activity to BK channels inGH3 cells, the steepPo-[Ca2+]irelationship that is characteristic of these channels involves cPLA2.

In a variety of disorders, overaccumulation of lipid in nonadipose tissues, including the heart, skeletal muscle, kidney, and liver, is associated with deterioration of normal organ function, and is accompanied by excessive plasma and cellular levels of free fatty acids (FA). Increased concentrations of FA may lead to defects in mitochondrial function found in diverse diseases. One of the most important regulators of mitochondrial function is mitochondrial Ca2+ ([Ca2+]m), which fluctuates in coordination with intracellular Ca2+ ([Ca2+]i). Polyunsaturated FA (PUFA) have been shown to cause [Ca2+]i mobilization albeit by unknown mechanisms. We have found that PUFA but not monounsaturated or saturated FA cause [Ca2+]i mobilization in NT2 human teratocarcinoma cells. Unlike the [Ca2+]i response to the muscarinic G protein-coupled receptor agonist carbachol, PUFA-mediated [Ca2+]i mobilization in NT2 cells is independent of phospholipase C and inositol-1,4,5-trisphospate (IP3) receptor activation, as well as IP3-sensitive internal Ca2+ stores. Furthermore, PUFA-mediated [Ca2+]i mobilization is inhibited by the mitochondria uncoupler carboxyl cyanide m-chlorophenylhydrozone. Direct measurements of [Ca2+]m with X-rhod-1 and 45Ca2+ indicate that PUFA induce Ca2+ efflux from mitochondria. Further studies show that ruthenium red, an inhibitor of the mitochondrial Ca2+ uniporter, blocks PUFA-induced Ca2+ efflux from mitochondria, whereas inhibitors of the mitochondrial permeability transition pore cyclosporin A and bongkrekic acid have no effect. In cultured porcine aortic smooth muscle cells,sphingosylphosphorylcholine (SPC), ATP, or bradykinin (BK) induced arapid dose-dependent increase in the cytosolicCa2+ concentration([Ca2+]i)and also stimulated inositol 1,4,5-trisphosphate(IP3) generation. Pretreatmentof cells with pertussis toxin blocked the SPC-induced IP3 generation and[Ca2+]iincrease but had no effect on the action of ATP or BK. In addition, SPCstimulated the mitogen-activated protein kinase (MAPK) and increasedDNA synthesis, whereas neither ATP nor BK produced such effects. Boththe SPC-induced MAPK activation and DNA synthesis were pertussis toxinsensitive. SPC-induced MAPK activation was blocked by treatment ofcells with the phospholipase C inhibitor, U-73122, or the intracellularCa2+-ATPase inhibitor,thapsigargin, but not by removal of extracellular Ca2+. Lysophosphatidic acidinduced cellular responses similar to SPC in a pertussistoxin-sensitive manner in terms of[Ca2+]iincrease, IP3 generation, MAPKactivation, and DNA synthesis. Platelet-derived growth factor (PDGF)also induced a[Ca2+]iincrease, MAPK activation, and DNA synthesis in the same cells; however, the PDGF-induced MAPK activation was not sensitive to pertussis toxin and changes in[Ca2+]i.SPC-induced MAPK activation was inhibited by pretreatment of cells withstaurosporine, W-7, or calmidazolium. Our results suggest that, inporcine aortic smooth muscle cells, MAPK is not activated by theincrease in[Ca2+]iunless a pertussis toxin-sensitive G protein is simultaneously stimulated, indicating the role ofCa2+ in pertussis toxin-sensitiveG protein-mediated MAPK activation.

Calreticulin (CRT), a Ca2+-binding molecular chaperone in the endoplasmic reticulum, plays a vital role in cardiac physiology and pathology. Oxidative stress is a main cause of myocardiac apoptosis in the ischemic heart, but the function of CRT under oxidative stress is not fully understood. In the present study, the effect of overexpression of CRT on susceptibility to apoptosis under oxidative stress was examined using myocardiac H9c2 cells transfected with the CRT gene. Under oxidative stress due to H2O2, the CRT-overexpressing cells were highly susceptible to apoptosis compared with controls. In the overexpressing cells, the levels of cytoplasmic free Ca2+ ([Ca2+]i) were significantly increased by H2O2, whereas in controls, only a slight increase was observed. The H2O2-induced apoptosis was enhanced by the increase in [Ca2+]i caused by thapsigargin in control cells but was suppressed by BAPTA-AM, a cell-permeable Ca2+ chelator in the CRT-overexpressing cells, indicating the importance of the level of [Ca2+]i in the sensitivity to H2O2-induced apoptosis. Suppression of CRT by the introduction of the antisense cDNA of CRT enhanced cytoprotection against oxidative stress compared with controls. Furthermore, we found that the levels of activity of calpain and caspase-12 were elevated through the regulation of [Ca2+]i in the CRT-overexpressing cells treated with H2O2 compared with controls. Thus we conclude that the level of CRT regulates the sensitivity to apoptosis under oxidative stress due to H2O2 through a change in Ca2+ homeostasis and the regulation of the Ca2+-calpain-caspase-12 pathway in myocardiac cells. apoptosis; calcium; endoplasmic reticulum  相似文献

How the endoplasmic reticulum (ER) and mitochondria communicate with each other and how they regulate plasmalemmal Ca2+ entry were studied in cultured rat brown adipocytes. Cytoplasmic Ca2+ or Mg2+ and mitochondrial membrane potential were measured by fluorometry. The sustained component of rises in cytoplasmic Ca2+ concentration ([Ca2+]i) produced by thapsigargin was abolished by removing extracellular Ca2+, depressed by depleting extracellular Na+, and enhanced by raising extracellular pH. FCCP, dinitrophenol, and rotenone caused bi- or triphasic rises in [Ca2+]i, in which the first phase was accompanied by mitochondrial depolarization. The FCCP-induced first phase was partially inhibited by oligomycin but not by ruthenium red, cyclosporine A, U-73122, a Ca2+-free EGTA solution, and an Na+-free solution. The FCCP-induced second phase paralleling mitochondrial repolarization was partially blocked by removing extracellular Ca2+ and fully blocked by oligomycin but not by thapsigargin or an Na+-deficient solution, was accompanied by a rise in cytoplasmic Mg2+ concentration, and was summated with a high pH-induced rise in [Ca2+]i, whereas the extracellular Ca2+-independent component was blocked by U-73122 and cyclopiazonic acid. The FCCP-induced third phase was blocked by removing Ca2+ but not by thapsigargin, depressed by decreasing Na+, and enhanced by raising pH. Cyclopiazonic acid-evoked rises in [Ca2+]i in a Ca2+-free solution were depressed after FCCP actions. Thus mitochondrial uncoupling causes Ca2+ release, activating Ca2+ release from the ER and store-operated Ca2+ entry, and directly elicits a novel plasmalemmal Ca2+ entry, whereas Ca2+ release from the ER activates Ca2+ accumulation in, or release from, mitochondria, indicating bidirectional mitochondria-ER couplings in rat brown adipocytes. plasmalemmal calcium entry; calcium release; mitochondrial depolarization; FCCP  相似文献

In vascular endothelial cells, depletion of intracellularCa2+ stores elicited capacitativeCa2+ entry (CCE) that resulted inbiphasic changes of intracellular Ca2+ concentration([Ca2+]i)with a rapid initial peak of[Ca2+]ifollowed by a gradual decrease to a sustained plateau level. Weinvestigated the rates of Ca2+entry, removal, and sequestration during activation of CCE and theirrespective contributions to the biphasic changes of[Ca2+]i.Ca2+ buffering by mitochondria,removal byNa+/Ca2+exchange, and a fixed electrical driving force forCa2+ (voltage-clamp experiments)had little effect on the CCE signal. The rates of entry ofMn2+ andBa2+, used as unidirectionalsubstitutes for Ca2+ entry throughthe CCE pathway, were constant and did not follow the concomitantchanges of[Ca2+]i.Pharmacological inhibition of the plasma membraneCa2+ pump, however, abolished thesecondary decay phase of the CCE transient. The disparity between thebiphasic changes of[Ca2+]iand the constant rate of Ca2+entry during CCE was the result of a delayed,Ca2+-dependent activation of thepump. These results suggest an important modulatory role of the plasmamembrane Ca2+ pump in the netcellular gain of Ca2+ during CCE.

The role of theNa+/Ca2+exchanger in intracellular Ca2+regulation was investigated in freshly dissociated catfish retinalhorizontal cells (HC).Ca2+-permeable glutamate receptorsand L-type Ca2+ channels as wellas inositol 1,4,5-trisphosphate-sensitive and caffeine-sensitiveintracellular Ca2+ stores regulateintracellular Ca2+ in these cells.We used the Ca2+-sensitive dyefluo 3 to measure changes in intracellularCa2+ concentration([Ca2+]i)under conditions in whichNa+/Ca2+exchange was altered. In addition, the role of theNa+/Ca2+exchanger in the refilling of the caffeine-sensitiveCa2+ store followingcaffeine-stimulated Ca2+ releasewas assessed. Brief applications of caffeine (1-10 s) producedrapid and transient changes in[Ca2+]i.Repeated applications of caffeine produced smallerCa2+ transients until no furtherCa2+ was released. Store refillingoccurred within 1-2 min and required extracellularCa2+. Ouabain-induced increases inintracellular Na+ concentration([Na+]i)increased both basal free[Ca2+]iand caffeine-stimulated Ca2+release. Reduction of external Na+concentration([Na+]o)further and reversibly increased[Ca2+]iin ouabain-treated HC. This effect was not abolished by the Ca2+ channel blocker nifedipine,suggesting that increases in[Na+]ipromote net extracellular Ca2+influx through aNa+/Ca2+exchanger. Moreover, when[Na+]owas replaced by Li+, caffeine didnot stimulate release of Ca2+ fromthe caffeine-sensitive store afterCa2+ depletion. TheNa+/Ca2+exchanger inhibitor 2',4'-dimethylbenzamil significantlyreduced the caffeine-evoked Ca2+response 1 and 2 min after store depletion.

The effects of inhibitors of CaMKII on intracellular Ca2+ signaling were examined in single calf pulmonary artery endothelial (CPAE) cells using indo-1 microfluorometry to measure cytoplasmic Ca2+ concentration ([Ca2+]i). The three CaMKII inhibitors, KN-93, KN-62, and autocamtide-2-related inhibitory peptide (AIP), all reduced the plateau phase of the [Ca2+]i transient evoked by stimulation with extracellular ATP. Exposure to KN-93 or AIP alone in the presence of 2 mM extracellular Ca2+ resulted in a dose-dependent increase of [Ca2+]i consisting of a rapid and transient Ca2+ spike followed by a small sustained plateau phase of elevated [Ca2+]i. Exposure to KN-93 in the absence of extracellular Ca2+ caused a transient rise of [Ca2+]i, suggesting that exposure to CaMKII inhibitors directly triggered release of Ca2+ from intracellular endoplasmic reticulum (ER) Ca2+ stores. Repetitive stimulation with KN-93 and ATP, respectively, revealed that both components released Ca2+ largely from the same store. Pretreatment of CPAE cells with the membrane-permeable inositol 1,4,5-trisphosphate (IP3) receptor blocker 2-aminoethoxydiphenyl borate caused a significant inhibition of the KN-93-induced Ca2+ response, suggesting that exposure to KN-93 affects Ca2+ release from an IP3-sensitive store. Depletion of Ca2+ stores by exposure to ATP or to the ER Ca2+ pump inhibitor thapsigargin triggered robust capacitative Ca2+ entry (CCE) signals in CPAE cells that could be blocked effectively with KN-93. The data suggest that in CPAE cells, CaMKII modulates Ca2+ handling at different levels. The use of CaMKII inhibitors revealed that in CPAE cells, the most profound effects of CaMKII are inhibition of release of Ca2+ from intracellular stores and activation of CCE. Tunicamycin is anucleoside antibiotic that inhibits protein glycosylation andpalmitoylation. The therapeutic use of tunicamycin is limited inanimals because of its toxic effects, particularly in cerebralvasculature. Tunicamycin decreases palmitoylation of the endothelialisoform of nitric oxide synthase, stimulates nitric oxide synthesis,and increases the concentration of intracellular calcium([Ca2+]i)in bovine aortic endothelial cells (B. J. Buckley and A. R. Whorton.FASEB J. 11: A110, 1997). In the present study,we investigated the mechanism by which tunicamycin alters[Ca2+]iusing the Ca2+-sensitive dye fura2. We found that tunicamycin increased[Ca2+]iwithout increasing levels of inositol phosphates. When cells wereincubated in the absence of extracellularCa2+,[Ca2+]irapidly rose in response to tunicamycin, although a full response wasnot achieved. The pool of intracellularCa2+ mobilized by tunicamycinoverlapped with that mobilized by thapsigargin. Extracellular nickelblocked a full response to tunicamycin when cells were incubated in thepresence of extracellular Ca2+.The effects of tunicamycin on[Ca2+]iwere partially reversed by washing out the drug, and the remainder ofthe response was inhibited by removing extracellularCa2+. These results indicate thattunicamycin mobilizes Ca2+ fromintracellular stores in a manner independent of phospholipase Cactivation and increases the influx ofCa2+ across the plasma membrane.

This study examines theCa2+ influx-dependent regulationof the Ca2+-activatedK+ channel(KCa) in human submandibulargland (HSG) cells. Carbachol (CCh) induced sustained increases in theKCa current and cytosolic Ca2+ concentration([Ca2+]i),which were prevented by loading cells with1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA). Removal of extracellularCa2+ and addition ofLa3+ orGd3+, but notZn2+, inhibited the increases inKCa current and[Ca2+]i.Ca2+ influx during refill (i.e.,addition of Ca2+ to cells treatedwith CCh and then atropine inCa2+-free medium) failed to evokeincreases in the KCa current but achieved internal Ca2+ storerefill. When refill was prevented by thapsigargin,Ca2+ readdition induced rapidactivation of KCa. These dataprovide further evidence that intracellularCa2+ accumulation provides tightbuffering of[Ca2+]iat the site of Ca2+ influx (H. Mogami, K. Nakano, A. V. Tepikin, and O. H. Petersen. We suggestthat the Ca2+ influx-dependentregulation of the sustained KCacurrent in CCh-stimulated HSG cells is mediated by the uptake ofCa2+ into the internalCa2+ store and release via theinositol 1,4,5-trisphosphate-sensitive channel.

The presentstudy used real-time confocal microscopy to examine the effects of the2-adrenoceptor agonistsalbutamol on regulation of intracellularCa2+ concentration([Ca2+]i)in myotubes derived from neonatal mouse limb muscles.Immunocytochemical staining for ryanodine receptors and skeletal musclemyosin confirmed the presence of sarcomeres. The myotubes displayedboth spontaneous and ACh-induced rapid (<2-ms rise time)[Ca2+]itransients. The[Ca2+]itransients were frequency modulated by both low and high concentrations of salbutamol. Exposure to -bungarotoxin and tetrodotoxin inhibited ACh-induced[Ca2+]itransients and the response to low concentrations of salbutamol but notthe response to higher concentrations. Preexposure to caffeineinhibited the subsequent[Ca2+]iresponse to lower concentrations of salbutamol and significantly blunted the response to higher concentrations. Preexposure to salbutamol diminished the[Ca2+]iresponse to caffeine. Inhibition of dihydropyridine-sensitive Ca2+ channels with nifedipine orPN-200-110 did not prevent[Ca2+]ielevations induced by higher concentrations of salbutamol. The effectsof salbutamol were mimicked by the membrane-permeant analog dibutyryladenosine 3',5'-cyclic monophosphate. Thesedata indicate that salbutamol effects in skeletal muscle predominantly involve enhanced sarcoplasmic reticulumCa2+ release.  相似文献

Pancreatitis is an inflammatory disease of pancreatic acinar cells whereby intracellular calcium concentration ([Ca2+]i) signaling and enzyme secretion are impaired. Increased oxidative stress has been suggested to mediate the associated cell injury. The present study tested the effects of the oxidant, hydrogen peroxide, on [Ca2+]i signaling in rat pancreatic acinar cells by simultaneously imaging fura-2, to measure [Ca2+]i, and dichlorofluorescein, to measure oxidative stress. Millimolar concentrations of hydrogen peroxide increased cellular oxidative stress and irreversibly increased [Ca2+]i, which was sensitive to antioxidants and removal of external Ca2+, and ultimately led to cell lysis. Responses were also abolished by pretreatment with (sarco)endoplasmic reticulum Ca2+-ATPase inhibitors, unless cells were prestimulated with cholecystokinin to promote mitochondrial Ca2+ uptake. This suggests that hydrogen peroxide promotes Ca2+ release from the endoplasmic reticulum and the mitochondria and that it promotes Ca2+ influx. Lower concentrations of hydrogen peroxide (10–100 µM) increased [Ca2+]i and altered cholecystokinin-evoked [Ca2+]i oscillations with marked heterogeneity, the severity of which was directly related to oxidative stress, suggesting differences in cellular antioxidant capacity. These changes in [Ca2+]i also upregulated the activity of the plasma membrane Ca2+-ATPase in a Ca2+-dependent manner, whereas higher concentrations (0.1–1 mM) inactivated the plasma membrane Ca2+-ATPase. To clarify the contribution of intracellularCa2+ concentration([Ca2+]i)-dependent and -independentsignaling mechanisms in arteriolar smooth muscle (aSM) to modulation ofarteriolar myogenic tone by nitric oxide (NO), released in response toincreases in intraluminal flow from the endothelium, changes in aSM[Ca2+]i and diameter of isolated rat gracilismuscle arterioles (pretreated with indomethacin) were studied byfluorescent videomicroscopy. At an intraluminal pressure of 80 mmHg, [Ca2+]i significantly increased andmyogenic tone developed in response to elevations of extracellularCa2+ concentration. The Ca2+ channelinhibitor nimodipine substantially decreased[Ca2+]i and completely inhibited myogenictone. Dilations to intraluminal flow (that were inhibited byN-nitro-L-arginine methyl ester)or dilations to the NO donorS-nitroso-N-acetyl-DL-penicillamine (that were inhibited by the guanylate cyclase inhibitor1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one) were notaccompanied by substantial decreases in aSM[Ca2+]i. 8-Bromoguanosine cGMP and thecGMP-specific phosphodiesterase inhibitor zaprinast significantlydilated arterioles yet elicited only minimal decreases in[Ca2+]i. Thus flow-induced endothelialrelease of NO elicits relaxation of arteriolar smooth muscle by acGMP-dependent decrease of the Ca2+ sensitivity of thecontractile apparatus without substantial changes in thepressure-induced level of [Ca2+]i.

A sustained increase in the cytosolic Ca2+ concentration ([Ca2+]i) can cause cell death. In this study, we found that, in cultured porcine aortic smooth muscle cells, endoplasmic reticulum (ER) stress, triggered by depletion of Ca2+ stores by thapsigargin (TG), induced an increase in the [Ca2+]i and cell death. However, the TG-induced death was not related to the [Ca2+]i increase but was mediated by targeting of activated Bax to mitochondria and the opening of mitochondrial permeability transition pores (PTPs). Once the mitochondrial PTPs had opened, several events, including collapse of the mitochondrial membrane potential, cytochrome c release, and caspase-3 activation, occurred and the cells died. TG-induced cell death was completely inhibited by the pan-caspase inhibitor Z-VAD-fmk and was enhanced by the Ca2+ chelator 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA), suggesting the existence of a Ca2+-dependent anti-apoptotic mechanism. After TG treatment, Ca2+-sensitive mitogen-activated protein kinase (MAPK) activation was induced and acted as a downstream effector of phosphatidylinositol 3-kinase (PI 3-kinase). The protective effect of Z-VAD-fmk on TG-induced cell death was reversed by BAPTA, PD-098059 (an MAPK kinase inhibitor), or LY-294002 (a PI 3-kinase inhibitor). Taken together, our data indicate that ER stress simultaneously activate two pathways, the mitochondrial caspase-dependent death cascade and the Ca2+-dependent PI 3-kinase/MAPK anti-apoptotic machinery. Cytoplasmic Ca2+concentration ([Ca2+]i) variation is akey event in myoblast differentiation, but the mechanism by which itoccurs is still debated. Here we show that increases of extracellular Ca2+ concentration ([Ca2+]o)produced membrane hyperpolarization and a concentration-dependent increase of [Ca2+]i due to Ca2+influx across the plasma membrane. Responses were not related toinositol phosphate turnover and Ca2+-sensing receptor.[Ca2+]o-induced[Ca2+]i increase was inhibited byCa2+ channel inhibitors and appeared to be modulated byseveral kinase activities. [Ca2+]i increasewas potentiated by depletion of intracellular Ca2+ storesand depressed by inactivation of the Na+/Ca2+exchanger. The response to arginine vasopressin (AVP), which inducesinositol 1,4,5-trisphosphate-dependent[Ca2+]i increase in L6-C5 cells, was notmodified by high [Ca2+]o. On the contrary,AVP potentiated the [Ca2+]i increase in thepresence of elevated [Ca2+]o. Other clones ofthe L6 line as well as the rhabdomyosarcoma RD cell line and thesatellite cell-derived C2-C12 line expressed similar responses to high[Ca2+]o, and the amplitude of the responseswas correlated with the myogenic potential of the cells.

We examined the effects of dissolved nitric oxide (NO) gas oncytoplasmic calcium levels ([Ca2+]i) in C6glioma cells under anoxic conditions. The maximum elevation (27 ± 3 nM) of [Ca2+]i was reached at 10 µM NO. Asecond application of NO was ineffective if the first was >0.5 µM.The NO donor diethylamine/NO mimicked the effects of NO. Acute exposureof the cells to low calcium levels was without effect on the NO-evokedresponse. Thapsigargin (TG) increased [Ca2+]iand was less effective if cells were pretreated with NO. Hemoglobin inhibited the effects of NO at a molar ratio of 10:1. 8-Bromo-cGMP waswithout effect on the NO-evoked response. If cells were pretreated withTG or exposed chronically to nominal amounts of calcium, NO decreased[Ca2+]i. The results suggest that C6 gliomacells have two receptors for NO. One receptor (NOA)elevates [Ca2+]i and resides on theendoplasmic reticulum (ER). The other receptor (NOB)decreases [Ca2+]i and resides on theplasmalemma or the ER. The latter receptor dominates when the level ofcalcium within intracellular stores is diminished.

AlF4-is known to generate oscillations in intracellular Ca2+ concentration ([Ca2+]i) by activating G proteins in many cell types. However, in rat pancreatic acinar cells, AlF4--evoked [Ca2+]i oscillations were reported to be dependent on extracellular Ca2+, which contrasts with the [Ca2+]i oscillations induced by cholecystokinin (CCK). Therefore, we investigated the mechanisms by which AlF4- generates extracellular Ca2+-dependent [Ca2+]i oscillations in rat pancreatic acinar cells. AlF4--induced [Ca2+]i oscillations were stopped rapidly by the removal of extracellular Ca2+ and were abolished on the addition of 20 mM caffeine and 2 µM thapsigargin, indicating that Ca2+ influx plays a crucial role in maintenance of the oscillations and that an inositol 1,4,5-trisphosphate-sensitive Ca2+ store is also required. The amount of Ca2+ in the intracellular Ca2+ store was decreased as the AlF4--induced [Ca2+]i oscillations continued. Measurement of 45Ca2+ influx into isolated microsomes revealed that AlF4-directly inhibited sarco/endoplasmic reticulum Ca2+-ATPase (SERCA). The activity of plasma membrane Ca2+-ATPase during AlF4- stimulation was not significantly different from that during CCK stimulation. After partial inhibition of SERCA with 1 nM thapsigargin, 20 pM CCK-evoked [Ca2+]i oscillations were dependent on extracellular Ca2+. This study shows that AlF4- induces [Ca2+]i oscillations, probably by inositol 1,4,5-trisphosphate production via G protein activation but that these oscillations are strongly dependent on extracellular Ca2+ as a result of the partial inhibition of SERCA. cholecystokinin; plasma membrane adenosine 5'-triphosphatase; G proteins; caffeine  相似文献

We investigatedthe role of intracellular calcium concentration([Ca2+]i) in endothelin-1 (ET-1) production,the effects of potential vasospastic agents on[Ca2+]i, and the presence of L-typevoltage-dependent Ca2+ channels in cerebral microvascularendothelial cells. Primary cultures of endothelial cells isolated frompiglet cerebral microvessels were used. Confluent cells were exposed toeither the thromboxane receptor agonist U-46619 (1 µM),5-hydroxytryptamine (5-HT; 0.1 mM), or lysophosphatidic acid (LPA; 1 µM) alone or after pretreatment with the Ca2+-chelatingagent EDTA (100 mM), the L-type Ca2+ channel blockerverapamil (10 µM), or the antagonist of receptor-operated Ca2+ channel SKF-96365 HCl (10 µM) for 15 min. ET-1production increased from 1.2 (control) to 8.2 (U-46619), 4.9 (5-HT),or 3.9 (LPA) fmol/µg protein, respectively. Such elevated ET-1biosynthesis was attenuated by verapamil, EDTA, or SKF-96365 HCl. Toinvestigate the presence of L-type voltage-dependent Ca2+channels in endothelial cells, the [Ca2+]isignal was determined fluorometrically by using fura 2-AM. Superfusionof confluent endothelial cells with U-46619, 5-HT, or LPA significantlyincreased [Ca2+]i. Pretreatment ofendothelial cells with high K+ (60 mM) or nifedipine (4 µM) diminished increases in [Ca2+]i inducedby the vasoactive agents. These results indicate that 1)elevated [Ca2+]i signals are involved in ET-1biosynthesis induced by specific spasmogenic agents, 2) theincreases in [Ca2+]i induced by thevasoactive agents tested involve receptor as well as L-typevoltage-dependent Ca2+ channels, and 3) primarycultures of cerebral microvascular endothelial cells express L-typevoltage-dependent Ca2+ channels.

During hypoxia, the level of adenosine in the carotid bodies increases as a result of ATP catabolism and adenosine efflux via adenosine transporters. Using Ca2+ imaging, we found that adenosine, acting via A2A receptors, triggered a rise in cytoplasmic [Ca2+] ([Ca2+]i) in type I (glomus) cells of rat carotid bodies. The adenosine response could be mimicked by forskolin (but not its inactive analog), and could be abolished by the PKA inhibitor H89. Simultaneous measurements of membrane potential (perforated patch recording) and [Ca2+]i showed that the adenosine-mediated [Ca2+]i rise was accompanied by depolarization. Ni2+, a voltage-gated Ca2+ channel (VGCC) blocker, abolished the adenosine-mediated [Ca2+]i rise. Although adenosine was reported to inhibit a 4-aminopyridine (4-AP)-sensitive K+ current, 4-AP failed to trigger any [Ca2+]i rise, or to attenuate the adenosine response. In contrast, anandamide, an inhibitor of the TWIK-related acid-sensitive K+-1 (TASK-1) channels, triggered depolarization and [Ca2+]i rise. The adenosine response was attenuated by anandamide but not by tetraethylammonium. Our results suggest that adenosine, acting via the adenylate cyclase and PKA pathways, inhibits the TASK-1 K+ channels. This leads to depolarization and activation of Ca2+ entry via VGCC. This excitatory action of adenosine on type I cells may contribute to the chemosensitivity of the carotid body during hypoxia. Stimulation ofsingle Ehrlich ascites tumor cells with agonists (bradykinin, thrombin)and with arachidonic acid (AA) induces increases in the freeintracellular Ca2+ concentration([Ca2+]i)in the presence and absence of extracellularCa2+, measured using theCa2+-sensitive probe fura 2. Sequential stimulation with two agonists elicits sequential increasesin[Ca2+]i,unlike addition of the same agonist twice. Bradykinin and thrombin haveadditive effects on[Ca2+]iin Ca2+-free medium. Thephosphoinositidase C inhibitor U-73122 inhibits the agonist-inducedincreases in[Ca2+]i,whereas ryanodine has no effect. Pretreatment of cells in Ca2+-free medium with thapsigarginabolishes the bradykinin-induced increase in[Ca2+]ibut not the response to thrombin. The AA-induced response is notinhibited by U-73122 and cannot be mimicked by the inactive structuralanalog trifluoromethylarachidonyl ketone. Pretreatment of the cellswith 50 µM AA (but not with 10 µM AA) abolishes the agonist-inducedincrease in[Ca2+]i.Thus bradykinin, thrombin, and AA induce increases in[Ca2+]iin Ehrlich cells due to Ca2+ entryand release from intracellular stores. Thrombin causes release ofCa2+ from an intracellular storethat is insensitive to bradykinin and is not depleted by thapsigarginbut is depleted by AA.
