Caffeoyl-catalpol, isoferuloyl-catalpol, protocatechuoyl-catalpol, benzoyl-catalpol, p-hydroxybenzoyl catalpol (catalposide), vanilloyl-catalpol and cinnamoyl-aucubin have been isolated from several Veronica species (Scrophulariaceae) in a more or less pure state. The first four compounds have never been recorded in plants before. A PC survey of forty-three species of the genus sensu lato has shown a general presence of aucubin and catalpol. They are accompanied in many of the species by a complex mixture of esters, especially esters of catalpol with aromatic acids. In only one species have aucubin esters been found. Loganin has been identified in five species, this being the first time that this compound has been found in the Scrophulariaceae. Loganin is accompanied by unidentified loganin esters in the same five species. From the systematic point of view the complex mixtures of esters of catalpol in Veronica seem to be noteworthy. Species of Globularia, Erinus, Scrophularia Verbascum, Wulfenia, Catalpa and Plantago investigated either totally lacked ester glucosides in leaves or contained mainly different types of esters. The possible systematic meaning of these results is briefly discussed.  相似文献

Plant secondary metabolites play an important role in constitutive and inducible direct defense of plants against their natural enemies. While induction of defense by aboveground pathogens and herbivores is well-studied, induction by belowground organisms is less explored. Here, we examine whether soil microorganisms and nematodes can induce changes in levels of the secondary metabolites aucubin and catalpol (iridoid glycosides, IG) in roots and root exudates of two full-sib families of Plantago lanceolata originating from lines selected for low and high constitutive levels of IG in leaves. Addition of soil microorganisms enhanced the shoot and root biomass, and the concentration of aucubin in roots of both Plantago lines without affecting IG levels in the rhizosphere. By contrast, nematode addition tended to reduce the root biomass and enhanced the stalk biomass, and increased the levels of aucubin and catalpol in root exudates of both Plantago lines, without affecting root IG concentrations. The Plantago lines did not differ in constitutive levels of aucubin and total IG in roots, while the concentration of catalpol was slightly higher in roots of plants originally selected for low constitutive levels of IG in leaves. Root exudates of “high IG line” plants contained significantly higher levels of aucubin, which might be explained by their higher root biomass. We conclude that soil microorganisms can induce an increase of aucubin concentrations in the roots, whereas nematodes (probably plant feeders) lead to an enhancement of aucubin and catalpol levels in root exudates of P. lanceolata. A potential involvement of secondary metabolites in belowground interactions between plants and soil organisms is discussed.  相似文献

Two species of Plantago, namely P. Alpina and P. altissima were investigated. From the former, nine iridoid glucosides and verbascoside were isolated. Together with the known iridoids gardoside, geniposidic acid, 8-epi-loganic acid, mussaenosidic acid, aucubin, monomelittoside and melittoside, two new glucosides were found: 10-O-acetylgeniposidic acid and alpinoside, another compound with a 10-O-acetyl group. From P. altissima verbascoside and isoverbascoside were isolated together with the known iridoids gardoside, 8-epi-loganic acid, catalpol, aucubin, and hookerioside as well as the new compound desacetylhookerioside.  相似文献

In a chemical investigation of the water soluble compounds in Paederota lutea eight known iridoids were isolated together with a new one with a 8,9-double bond, namely paederotoside (10-O-benzoyl-6'-O-alpha-arabino(1-->6)-beta-glucopyranosyl arborescosidic acid) and the 6-hydroxy-flavone glucoside 4'-O-methylscutellarein 7-O-beta-glucopyranoside. The known iridoid glucosides were 8-epiloganic acid, gardoside, aucubin, catalpol and the 6-O-esters of catalpol: veronicoside, catalposide, amphicoside and verproside. The compounds isolated show that Paederota has a glycoside content almost identical to that of a general Veronica species, and this is in good agreement with the results from recent investigations of the phylogeny of Veronica and its closest relatives, where Paederota is placed as a sister-group next to Veronica. In an analysis of the iridoid glucosides from some of these relatives, it is shown that Veronica, Paederota, Picrorhiza and Veronicastrum are all characterized by containing 6-O-esters of catalpol. Some less closely related taxa namely: Lagotis, Wulfenia, Plantago, Aragoa and Globularia instead contain 10-O-esters of catalpol or aucubin.  相似文献

In a continued chemosystematic investigation of the water-soluble compounds in Veronica sect. Hebe, we have investigated two more of the species formerly classified as Parahebe. Both species contained mannitol in considerable amounts and in addition some glucosides of iridoid acids. Veronica cheesemanii was characterised by aucubin and its esters: 2′-O-benzoylaucubin and an aucubin diester named cheesemanioside. The main iridoid compounds in Veronica hookeriana were catalpol and its ester verminoside, but this species also contained the sugar ester methyl 1-O-benzoyl-3-α-glucuronosylglycerol and a caffeoyl phenylethanoid glycoside (CPG) named parahebeoside, a 2′-O-β-xylopyranosyl derivative of the known plantamajoside. The results show that the studied species of the former genus Parahebe are very different with regard to their chemical content. This is in agreement with the DNA sequence data and implies the genus was polyphyletic as previously circumscribed.  相似文献

Summary We investigated the effects of genotype, habitat, and seasonal variation on production of the iridoid glycosides, aucubin and catalpol, in leaves of the common weed Plantago lanceolata. Two genotypes, one each from a lawn and an adjacent abandoned hayfield population, were clonally replicated in the greenhouse, and then planted back into the two habitats. One quarter of the plants from each treatment were harvested on each of four dates, at approximately two-week intervals. Over the course of the growing season, and in both habitats, we found a significant increase in the concentration of both aucubin and catalpol in P. lanceolata leaves. The genotypes differed in their response to environmental variation, both in time and between sites, as indicated by significant genotype x date and genotype x site interactions. Early in the season, habitat (lawn or field) had a greater effect on iridoid glycoside concentration than did plant genotype, but later in the season, plant genotype was more influential in determining the iridoid glycoside concentration. Thus, the relative palatability of Plantago genotypes to specialist and generalist herbivores may vary in time and space.  相似文献

Hairy root lines through the infection of Agrobacterium rhizogenes strain (A4) were established from shoot tips and leaves of Rehmannia glutinosa Libosch. Ten lines of hairy roots were selected on the basis of biomass increase in half-strength Gamborg medium (1/2 B5). Transgenic status of the roots was confirmed by polymerase chain reaction using rolB and rolC specific primers. Iridoid glycosides (catalposide, loganin, aucubin and catalpol) and phenylethanoid glycosides (verbascoside and isoverbascoside) identified using HPLC?CESI?CMS, and their contents were compared with untransformed root culture and roots of 1-year-old field-grown plants of R. glutinosa by RP-HPLC. The growth and production of secondary metabolites in ten hairy root lines varied considerably as to the media. Woody plant (WP) medium displayed higher growth in terms of fresh (FW) and dry weights (DW) compared to 1/2 B5 medium. High-yielding hairy root lines produced higher amounts of loganin, catalposide, verbascoside and isoverbascoside in comparison to the untransformed root culture and roots of 1-year-old field-grown plants. The highest amounts of catalposide and loganin in transformed roots were 4.45?mg?g?1 DW (RS-2 hairy root line) and 4.66?mg?g?1 DW (RS-1 hairy root line), respectively. Aucubin and catalpol were detected in some lines in trace amounts. The highest amounts of verbascoside (16.9?mg?g?1 DW) and isoverbascoside (3.46?mg?g?1 DW) were achieved in RS-2 root line. The contents of catalposide, verbascoside and isoverbascoside in high-producing lines were several times higher than in untransformed root culture and roots of R. glutinosa plants grown in soil. Loganin and aucubin could not be detected in roots of field-grown plants. However, the levels of catalpol were much lower in the in vitro roots.  相似文献

In a chemosystematic investigation of three Southern hemisphere species of Veronica, namely the Australian Veronica derwentiana Andrews and Veronica perfoliata R.Br. (formerly Derwentia species), and the New Zealand Veronica catarractae G. Forster (formerly a species of Parahebe), the water-soluble constituents were isolated and identified by spectroscopic methods. Apart from other iridoid glucosides common to the genus, three unusual substituted benzoyl esters of aucubin (derwentiosides A–C) were obtained from V. derwentiana and a chlorinated iridoid glycoside (catarractoside) from V. catarractae in addition to other iridoids common to the genus. The chemical profile of V. perfoliata is similar to that of Northern hemisphere species of Veronica because of the presence of characteristic 6-O-catalpol esters. The profile of V. derwentiana is unique, since 6-O-esters of aucubin rather than of catalpol dominate, however, the acyl groups are the same as those present in catalpol esters found in some other Veronica sections. V. catarractae also contains one of the catalpol esters characteristic of Veronica, but in addition three 6-O-rhamnopyranosyl substituted iridoid glycosides, one of which is 6-O-rhamnopyranosylcatalpol. Esters of the latter compound are previously only known from the more derived species in recent phylogenetic trees of sect. Hebe to which V. catarractae now also belongs, but as a more basal member.  相似文献

From Verbascum sinuatum, besides aucubin, harpagide, 6-O-β-d-xylopyranosylaucubin and sinuatol (6-O- α-l-rhamnopyranosylaucubin), a new iridoid glycoside, sinuatoside, has been isolated and its structure elucidated as 6-O-(3-O-β-d-xylopyranosyl)α-d-galactopyranosyl aucubin on the basis of spectral data and chemical modifications. A new highly oxygenated iridoid glucoside, urphoside B (1) was isolated from the Veronica pectinata var. glandulosa together with seven known iridoid glucosides, aucubin, catalpol, veronicoside, catalposide, verproside, amphicoside and 6-O-veratroyl catalpol. The planar as well as the stereo structures of the isolated compounds were determined by means of extensive 1D- and 2D-NMR spectroscopy and confirmed by HR-Mass.  相似文献

Diet of herbivorous insects can influence both the herbivores and their natural enemies. We examined the direct and indirect effects of diet on the interactions between the polyphagous herbivore Trichoplusia ni Hübner (Lepidoptera: Noctuidae) and its polyembryonic parasitoid Copidosoma floridanum Ashmead (Hymenoptera: Encyrtidae). To determine how host plant species and host plant iridoid glycoside content affect host caterpillars and their parasitoids, parasitized and unparasitized T. ni were given leaves of either Plantago lanceolata L., which contains the iridoid glycosides aucubin and catalpol, Plantago major L. (Plantaginaceae), which contains only aucubin, or Taraxacum officinale F.H. Wigg (Asteraceae), which contains neither. Survival of unparasitized T. ni was much lower when fed P. major compared with the other two host plants, whereas pupae were smallest when fed T. officinale and developed most slowly when fed P. lanceolata as larvae. Neither aucubin nor catalpol were detected in intact Plantago‐fed T. ni larvae or their hemolymph, and only trace amounts of aucubin were detected in frass, suggesting that these compounds are mostly metabolized in the midgut and are not encountered by the parasitoid. Copidosoma floridanum clutch size was almost doubled when reared from P. lanceolata‐fed T. ni compared with T. officinale‐fed larvae and tripled compared with P. major‐fed larvae, although the percent of parasitoids surviving to adulthood was uniformly high regardless of host diet. The observed variation in C. floridanum fitness among host diets is likely mediated by the effect of the diets on host quality, which in turn may be influenced more by other factors in the host plants than their iridoid glycoside profiles. Interactions between plant metabolites, generalist herbivores like T. ni, and their parasitoids may be predominantly indirect.  相似文献

The application of gas-liquid chromatography-mass spectrometric (g.l.c.-m.s.) analysis to a number of sialic acid-containing polysaccharides of meningococcal origin has been studied. Methylation of these polysaccharides by the Hakomori conditions resulted in both O- and N-methylation. Methanolysis of the methylated polysaccharides from serogroup C [(2→9)-linked], colominic acid [(2→8)-linked], and serogroups Y and W-135 [both (1→4)-linked], yielded the respective 4,7,8,4,7,9-, and 7,8,9-tri-O-methyl derivatives of methyl N-acetyl-N-methyl-β-D-neuraminate methyl glycoside. As model compounds, methyl N-acetyl-4,7,8,9-tetra-O-methyl-α-D-neuraminate methyl glycoside and its N-methyl derivative were also synthesized. All of the methylated derivatives could be identified on the basis of their typical fragmentation-patterns, indicating that this method is applicable to the determination of the position of linkages to sialic acid residues in biopolymers.  相似文献

In a continued chemosystematic investigation of the water-soluble compounds in Veronica sect. Hebe, four additional species were investigated. In comparison to other, Northern Hemisphere (NH) species of Veronica, those belonging to the New Zealand species in sect. Hebe are apparently more variable in chemical content. In addition to the compounds characteristic for NH Veronica, namely mannitol, aucubin, catalpol and 6-O-esters of catalpol as well as some caffeoyl phenylethanoid glucosides (CGPs), Veronica topiaria (syn. Hebe topiaria) also gave an unusual 6-O-ester of aucubin named topiarioside. The former Hebe species Veronica cupressoides and Veronica stenophylla each provided one of the two previously undescribed disaccharide esters named hebitol I and II, respectively, and the former plant also provided a CPG named cuproside, a 6-O-β-glucopyranosyl derivative of the known hebeoside. The last species, namely Veronica hulkeana (syn. Heliohebe hulkeana) only contained compounds common to other species of Veronica. The taxonomic results are discussed and it is concluded that carbohydrate esters are common in sect. Hebe. The data so far obtained indicate that the occurrences of esters of 6-O-rhamnopyranosylcatalpol are confined to the most derived species in the section.  相似文献

Larvae and pupae of the alpine butterfly Euphydryas cynthia contain three iridoid glucosides: two of them, aucubin and catalpol, are very common in the Plantaginaceae, but the third, 6-O-glucopyranosylaucubin, has hitherto only been isolated from Odontites verna and Verbascum sinuatum (Scrophulariaceae). The amounts of the three iridoids found in the insects were 0.53, 0.31 and 1.48 % dry wt, respectively. Preliminary feeding experiments with an insectivorous bird indicated some unpalatability in the insect, which probably stems from the iridoid content.  相似文献

Background and aims

Nitrate leaching from intensively cropped soils represents a huge environmental problem. In order to diversify the range of nitrogen management strategies, this investigation is focused on the effects of ribwort plantain, Plantago lanceolata L., and its allelochemicals on soil N mineralization.

Methods

High-performance liquid chromatography was used in this study for phytochemical analysis of the major allelochemicals aucubin, catalpol, and verbascoside. Soil incubation experiments demonstrated a significant suppression of soil N mineralization caused by the incorporation of the iridoid glycoside (IG) aucubin, leaf material of two varieties (P. lanceolata cv. Libor and cv. Arterner), and an aqueous extract of P. lanceolata leaves.

Results

Throughout the growing season, the two varieties conspicuously differed in aucubin and verbascoside contents as well as in leaf dry weight. In soil incubation experiments, incorporated leaf material of both varieties affected long-term low soil nitrate concentrations. Experimental aucubin application resulted in an inhibitory effect on soil N mineralization. This was not true for the IG catalpol. Furthermore, we observed a negative relationship between IG concentrations and inorganic soil nitrogen concentrations when the soil was incubated with aqueous P. lanceolata leaf extract of different concentrations.

Conclusion

This study enforced the hypothesis that allelochemicals of P. lanceolata have an active role in a suppression effect on soil N mineralization. Further research may be necessary to investigate the specific effects of P. lanceolata allelochemicals on the nitrogen cycle.  相似文献

A novel flavone glycoside has been obtained from the whole plant of Veronica filiformis and identified by means of 13C NMR spectroscopy as isoscutellarein 4′-methyl ether 7-O-β-(6?-O-acetyl-2″-O-allosylglucoside). The related isoscutellarein glycoside is also present. This is the first report of 2-allosylglucose as a disaccharide unit of flavonoids. 13`C NMR data on some A-ring trioxygenated flavonoids are also presented.  相似文献

The iridoid glucoside, ajugol, and the phenylethanoid glucoside, cornoside, have been isolated from species of Veronica (Plantaginaceae) for the first time. The presence of these compounds has been screened in 18 plant accessions belonging to 15 species of Veronica (Plantaginaceae), by isolation or NMR spectroscopy of crude extracts. In addition, the distribution of iridoids in the genus has been reviewed, using mainly the published data of isolated compounds. Using the recent expansion and reclassification of the genus based on DNA-sequence results as the model, we find that the genus is rather homogeneous with regard to the distribution of iridoid glucosides, aucubin and/or catalpol as well as 6-O-esters of catalpol being universally present in 10 of the 12 subgenera for which data exist. Only the two subgenera Pocilla and Chamaedrys deviate from this pattern. Pocilla is heterogeneous; in this subgenus, species in subsect. Agrestes contain the standard iridoid garniture, while species in subsect. Biloba do not contain the 6-O-esters of catalpol, but ajugol instead. Veronica intercedens (subsect. Subracemosae) differs from the remainder of the subgenus in only containing 5-hydroxylated iridoids (melittoside and globularifolin) and is so far the only species within the genus in which such compounds have been detected. These chemical differences are clearly reflected in the DNA-based phylogram of the subgenus. Subg. Chamaedrys appears homogeneous in lacking iridoids or only containing these in small amounts, but instead half of the investigated species contained the phenylethanoid glucoside cornoside. The distribution of this compound in angiosperms is reviewed; cornoside often substitutes iridoid glucosides in plants where these are expected to be present. The chemical results of Veronica fit in very well with the phylogenetic implications of the DNA-sequence results.  相似文献

1. To investigate the kinetics of ophidian l-amino acid oxidase, V and Km were determined for phenylalanines that were substituted in every ring position with groups of various size and reactivity, and for a few ring-substituted tryptophans and histidines. The venom of one representative from each of three major classes of poisonous snakes, Naja melanoleuca, Vipera russelli and Crotalus adamanteus, served as a source of the ophidian l-amino acid oxidase. Both crude and crystalline enzyme from the venom of C. adamanteus were tested. 2. The introduction of a benzene ring into glycine and alanine caused some increase of V and a very marked depression of Km. 3. With the exception of fluorine, residues in the ortho position of phenylalanine led to a decrease of V. The rates induced by various substitutions follow the pattern: metaparaortho. Within the halogen series, the effects become more pronounced with increasing atomic number. 4. Ring substitution in heterocyclic amino acids also affected the V values markedly. For methyl-substituted tryptophans the pattern was: 5-methyl ≥ 6-methyl ≥ 4-methyl. In a few instances ring substitution accounts for a considerable elevation of V, as shown for β-quinol-4-ylalanine and its 6-methoxy derivative. 5. The kinetic constants appear to be unaffected by relatively high concentrations of the corresponding d-amino acids. 6. A general principle that permits a uniform interpretation of a vast body of information is suggested. It is based on the assumption that most substrates form not only eutopic but also dystopic complexes with the enzyme. The latter, in contrast with the former, do not permit the formation of reaction products. K values for eutopic and dystopic complexes are computed. Similar concepts have been presented to elucidate the action of α-chymotrypsin (Hein & Niemann, 1962) and of monoamine oxidase.  相似文献

The peptide N-acetylmuramyl-l-alanyl-d-isoglutamine (MDP), which has adjuvant activities, and 17 of its derivatives and analogs were synthesized and assayed to elucidate the structure necessary for adjuvant activity in induction of experimental allergic encephalomyelitis (EAE) in guinea pigs. The results revealed the importance of the d configuration and the α-carboxamide group of the isoglutaminyl residue of MDP for adjuvant activity. Replacement of the l-alanyl residue of MDP by d-alanine, but not by l-serine or glycine, resulted in a marked decrease in the activity. The β-methyl glycoside of MDP was found to be more active than the α-methyl derivative. 6-O-Stearoyl-N-acetylmuramyl-l-alanyl-d-isoglutamme showed activity.  相似文献
