Are metals dietary carcinogens?   总被引:13,自引:0,他引:13  
Humans have been in contact with metals almost since the beginning of our existence. In fact, one cannot even think on human evolution without considering the great role played by metals in mankind's development. Metals are common moieties of molecules involved in a wide variety of biological processes, and hence are found in virtually all living organisms. Some metals are essential for human nutrition; others are found as contaminants in foodstuffs. One feature of the normal human diet which is frequently found is the simultaneous presence of both essential and toxic metals. Other factors important in the risk-evaluation analysis of metals are their pharmacokinetics, interactions among them and with other major components of the diet, and, especially, the great differences in the dietary habits of different populations and in the regional distribution of metals. In attempting to understand the role which dietary metals could play in human carcinogenesis, we found that the many factors involved and the lack of specific information made it difficult to reach firm conclusions on the hazards of dietary metals. We hope that this paper will raise the interest of genetic toxicologists in the subject and will consequently facilitate a risk analysis of the carcinogenic potential of dietary metals.  相似文献

Long-term exposure to extremely-low-frequency electromagnetic fields (ELF EMFs) greater than 0.4 microT has been linked, by epidemiological studies, to a small elevated risk of childhood leukaemia. Laboratory-based experiments have been claimed to show that ELF EMFs induce a variety of biological responses, although these claims are controversial. Recent experiments by Ivancsits et al. [Mutat. Res. 519 (2002) 1; Int. Arch. Occup. Environ. Health 76 (2003) 431; Mech. Age. Dev. 124 (2003) 847; H.W. Rüdiger, S. Ivancsits, E. Diem, O. Jahn, Genotoxic effects of ELF-EMF on human cells in vitro, Bioelectromagnetics Society 25th Annual Meeting, Maui, USA, 2003] suggest that ELF EMFs are genotoxic, on the basis of observations that intermittent exposures induce single-strand breaks (SSB) and double-strand DNA breaks (DSB) in the DNA of cultured human fibroblasts. A fundamental tenet of modern genetics is that mutations occur spontaneously, without regard to their effects on the organism's fitness. However, several authors have recently published evidence that suggests that the bacterium E. coli can respond to novel environmental conditions by inducing or directing specifically those mutations that enhance fitness in such environments. From January 1977 to June 1990, 1,670 patients with a liver mass or masses underwent transabdominal fine needle aspiration biopsy of the liver. Of those cases, 99 were diagnosed cytologically as "hepatocellular carcinoma" and 9 as "consistent with liver cell adenoma." Among the 99 patients with hepatocellular carcinoma, 3 were users of oral contraceptives. The nine patients with liver cell adenoma were all users of oral contraceptives. Of them, two developed foci or areas of liver cell dysplasia within the adenomas. The mean periods of oral contraceptive use among these three groups of patients were different. It appeared that patients started to develop liver cell adenoma after five years of oral contraceptive use (mean, 6.3 years). Foci or areas of liver cell dysplasia began to arise within liver cell adenomas after 8 years of oral contraceptive use (mean, 9 years), and the patients started to develop hepatocellular carcinoma after 10 years of contraceptive use (mean, 11 years). The cytologic features of liver cell dysplasia strikingly mimicked those of hepatocellular carcinoma. From this study, the foci or areas of liver cell dysplasia appear to be the missing link responsible for the transformation of liver cell adenoma to carcinoma. It is believed that liver cell adenomas are not premalignant and may undergo reversible change after withdrawal of causative agents, whereas foci or areas of liver cell dysplasia within the adenomas are irreversible, premalignant changes and may transform into hepatocellular carcinoma.  相似文献

A wide variety of oxidative DNA lesions are commonly present in untreated human and animal DNA. One of these lesions, 8-hydroxydeoxyguanosine, has been shown to lead to base mispairing (mutation) on DNA replication. Other lesions remain to be investigated in this respect. Oxidative DNA lesions on cell replication may, in appropriate circumstances, lead to proto-oncogene activation. Oxidative DNA damage, on fixation, may also lead to cytotoxicity followed by regenerative proliferation. The probable or possible importance of oxidative DNA damage is reviewed for various classes of carcinogens and natural processes, including metal ions, high-energy radiation, miscellaneous chemicals, tumor-promoting agents, polyhydroxyphenols/quinones, lipid metabolism, peroxisome proliferators and thyroid function. It is concluded that although the evidence needs considerable strengthening in many of these examples, the available information indicates the potential importance of oxidative DNA damage in the induction of tumors by these agents. It is also possible that non-cancerous degenerative diseases associated with aging are the result of the accumulation of lesions resulting from unrepaired oxidative DNA damage.  相似文献

Pediatric liver disease (PLD) is a major cause of severe morbidity and prolonged hospitalizations in children. Stratifying patients in terms of prognosis remains challenging. The limited knowledge about molecular mechanisms causing and accompanying PLD remains the main obstacle in a search for reliable prognostic biomarkers. A systematic search of MEDLINE via PubMed and Embase via OVID was conducted on studies published between August 2007 and August 2017. Molecular markers with a prognostic potential in terms of survival, need for liver transplantation or disease progression/regression were selected. In general, identified studies were single center smaller case-control studies or case series with a low level of evidence and a high risk of bias. Only 23 studies comprising 898 patients could be included, mostly focusing on biliary atresia, non-alcoholic fatty liver disease, viral hepatitis, and LT; and markers related to morphogenesis and fibrosis. Furthermore, molecular markers in metabolic pathways and inflammation shown to be relevant, however requiring further validation. Hence, further biological and clinical studies are needed to gain greater molecular insight into PLD.  相似文献

Statistically rigorous methods for summarizing and reporting trends in the intactness of biodiversity are a key element of effective biodiversity monitoring programs. There are four major approaches for translating complex monitoring data into easily communicated summary statistics: (1) traditional diversity indices such as species richness and Simpson's diversity, (2) species intactness indices based on occurrence, (3) species intactness indices based on abundance, and (4) multivariate community indices. We use simulated data to evaluate the effectiveness of 13 indices from these four categories based on statistical robustness, sensitivity to errors and noise in the data, ecological relevance, and ease of communication. We show that indices that calculate species intactness using equations like Buckland's arithmetic mean index are the most effective for use in large-scale biodiversity intactness monitoring programs. Traditional diversity indices are unsuitable for monitoring of biodiversity intactness, and multivariate indices can be highly sensitive to errors and noise in the data. Finally, we provide guidelines for the application of these indices in biodiversity intactness monitoring.  相似文献

Evidence is increasing suggesting that adding progestins to estrogen replacement therapy may be more harmful then beneficial, however it is debatable whether all progestins act equally on breast epithelial cells.

Experimental data with the comparison of various progestins in the same in vitro model present a rather high evidence that there may be differences between the various progestins regarding breast cancer risk. Especially of concern may be to differentiate between primary and secondary risk i.e. between benign and malignant breast epithelial cells.

The epidemiological studies and especially the Women's Health Initiative (WHI) trial, so far the only prospective placebo-controlled interventional study, demonstrate an increased risk under combined estrogen/progestin therapy, but they have the limitations that they up to now cannot discriminate between the various progestins mostly due to too small or not comparable patient numbers in the subgroups with the various progestins. However, there is evidence that the natural progesterone, possibly also the transdermal usage of synthetic progestins, may avoid an increased risk, but this must be proven in further clinical trials.  相似文献

Aims Coastal areas, and in particular coastal dunes, are ecosystems strongly affected by the invasion of alien plants. However, few attempts have ever been made to quantify alien species incidence in different communities along the coastal zonation. This work aims to analyze the distribution of alien plants along the coastal zonation of sandy shores on the Tyrrhenian coast, addressing specifically differences among plant communities in abundance of alien plants.Methods The study was performed on recent dunes (Holocene) of the central western coast of Italy. We selected dune landscapes where invasion processes were particularly evident. Vegetation plots were randomly sampled and through cluster analysis, we identified six plant communities corresponding to the typical zonation described for the Tyrrhenian sandy coast of Central Italy. We evaluated and compared frequency and abundance of invasion in these different communities. Further, we investigated how propagule pressure (measured using as proxy human structures) contributed to the observed invasion patterns.Important findings We found a relatively low total number of aliens but also a differential distribution pattern and strong abundance of some of the aliens in specific sectors of the vegetation zonation. The perennial community of transition dunes appears most affected by invasion processes, related almost exclusively to the frequent and widespread Carpobrotus aff. acinaciformis. This alien species reaches high cover values, apparently lowering cover of native species of transition dune plant communities. Higher levels of invasion in the transition dune can be partially explained because of greater propagule pressure in this section of the dune profile. Our findings thus have important conservation and management implications since transition dune communities with Crucianella maritima are rare and protected (sensu Habitat 92/43/EEC Directive) along the entire Italian coast.  相似文献

Summary The unprecedented rate of depletion of the stratospheric ozone layer will likely lead to appreciable increases in the amount of ultraviolet-B radiation (UV-B, 280–320 nm) reaching the earth's surface. In plants, photosynthetic reactions and nucleic acids in the mesophyll of leaves are deleteriously affected by UV-B. We used a fiber-optic microprobe to make direct measurements of the amount of UV-B reaching these potential targets in the mesophyll of intact foliage. A comparison of foliage from a diverse group of Rocky Mountain plants enabled us to assess whether the foliage of some plant life forms appeared more effective at screening UV-B radiation. The leaf epidermis of herbaceous dicots was particularly ineffective at attenuating UV-B; epidermal transmittance ranged from 18–41% and UV-B reached 40–145 m into the mesophyll or photosynthetic tissue. In contrast to herbaceous dicots, the epidermis of 1-year old conifer needles attenuated essentially all incident UV-B and virtually none of this radiation reached the mesophyll. Although the epidermal layer was appreciably thinner in older needles (7 y) at high elevations (Krumholtz), essentially all incident UV-B was attenuated by the epidermis in these needles. The same epidermal screening effectiveness was observed after removal of epicuticular waxes with chloroform. Leaves of woody dicots and grasses appeared intermediate between herbaceous dicots and conifers in their UV-B screening abilities with 3–12% of the incident UV-B reaching the mesophyll. These large differences in UV-B screening effectiveness suggest that certain plant life forms may be more predisposed than others to meet the challenge of higher UV-B levels resulting from stratospheric ozone depletion.  相似文献

A careful survey of the dinosaur footprints and trackways (identified as Eubrontes, Anchisauripus and Grallator) at the Mt. Tom site north of Holyoke, Massachusetts, and plots of their orientations reveal that an improbable percentage (70%) of the tracks are oriented in a nearparallel course. All but one of these coincident ancient traverses were probably made by the same kind of Triassic dinosaur (the footprints of which are referred to as Eubrontes). Comparable trackway orientation patterns have been reported by Albritton (1942) near Comanche, Texas for an Early Cretaceous dinosaur (iguanodobntid?) and Bird (1941, 1944) cited paralled sauporod trackways of Early Cretaceous age near the Paluxy River in Texas. Probability dictates that these sub-parallel traverses were not independent events and the presence of other deviating trackways at all three sites indicates that the trackmakers probably were not confined in their passage by physical barriers. Furthermore, the coincidental occurrence of such natural barriers at all of the sites mentioned here seems highly improbable. the combinedevidence of the Massachusetts site and the two Texas localities, together with the apparently widespread occurrence of dinosaur trackway lineation, strongly indicates gregarious habits for several different kinds of dinosaurs.  相似文献

We used a combination of field experiments and stable isotopes to examine mummichog growth and movement within a New England estuary. We documented physical and biological patterns within the estuary by caging individually-marked fish in enclosures at four locations along a coastal river and measuring environmental parameters (e.g., salinity, tidal inundation) and fish characteristics (e.g., gut-contents, growth, and stable isotope values) at each location. The upstream location was fresh (1 ppt) at low tide, and the downstream location was saline at high tide (32 ppt). The upstream and downstream locations had more tidal inundation than the intermediate location. Fish gut contents were dominated by terrestrial insects at the upstream location, by algae and detritus at the intermediate locations, and by aquatic insects at the downstream location. Fish grew fastest at the upstream location and slowest at the downstream location. Stable isotope values (δ13C and δ15N) of fish held in cages were significantly different at upstream, intermediate, and downstream locations. We transferred fish from one location to another in order to document how stable isotope values change when fish switch diets by moving within this estuary. Because differences in rates at which different tissue types approach the isotopic value of new diet sources can be used as a way to estimate the time since diet shift, we used the δ13C and δ15N values of liver and muscle as indicators of short term previous diet (liver) and longer term previous diet (muscle). We collected wild (uncaged) mummichogs from each location, and we compared their liver and muscle isotope values to values of fish that were transferred among locations. When fish were transferred from one location to another, their stable isotope values were intermediate between expected values at the previous and current locations. The liver approached stable isotope values representative of current location faster than muscle. Wild fish showed greater variation in stable isotope values than fish held in cages. Wild fish from the upstream location showed patterns in liver and muscle stable isotope values that were consistent with patterns in fish that were transferred from the downstream location to the upstream location (∼ 10 km away). These patterns in stable isotope values could have multiple causes including intra-season movement between downstream and upstream locations.  相似文献

A 5′-nucleotidase (5′-ribonucleotide phosphohydrolase, EC 3.1.3.5) was highly purified from rat liver. The preparation appeared homogeneous on the criteria of disc-gel electrophoresis.A pH optimum at about 6.5 was observed for all substrates tested. The activity of this enzyme was absolutely dependent on the presence of various bivalent metal salts. The highest V value was attained with MgCl2 and the concentration at half-enzyme saturation was lowest with MnCl2. The enzyme had markedly higher affinities for IMP, dIMP, GMP and dGMP than the other 5′-mononucleotides, although V values for all the substrates tested were in the same order of magnitude.The activity of this enzyme was stimulated by various alkali metal salts, some carboxylic acids and adenine nucleotides. When AMP was used as substrate, the substrate-velocity plot was sigmoidal and NaCl, Tris-maleate and ATP stimulated the enzyme by decreasing the sigmoidicity of the plot. When IMP was used as substrate, the substrate-velocity plot was hyperbolic and these three activators stimulated the enzyme by increasing the V and decreasing the Km value.Some of these results provided consistent evidence for the identity of this enzyme and the cytosol 5′-nucleotidase, the presence of which had been reported in crude preparations from rat liver.  相似文献

Summary Prealbumin plasma level is considered a good index of liver function in liver cirrhosis. However, plasma protein levels depend not only on liver function, but also on amino acid supply which is consequent to nutritional status.In 12 cirrhotics we measured prealbumin plasma levels and the lower limb venous-artero difference of amino acid plasma levels in blood samples taken from femoral vein and femoral artery in post-absorptive conditions considered as a direct index of protein release from peripheral tissues and an indirect index of protein nutritional status.In arterial and in venous plasma amino acid sum was 1.86±0.40 (mean + sd) and 2.00 ± 0.04 mMol/l respectively.Prealbumin plasma levels were found directly correlated with the venousartero difference of amino acid plasma levels (r = 0.57p < 0.05) and of glutamate + glutamine levels (r = 0.73p < 0.007).In conclusions, these data suggest that prealbumin plasma levels are linked to amino acid supply from peripheral tissues in cirrhotics.  相似文献

Many animals, plants and protists contain non-parasitic microorganisms and these endosymbioses are widely assumed to be mutualistic. Most of the microorganisms possess metabolic capabilities, such as the ability to fix nitrogen, photosynthesize or degrade cellulose, that their partners utilize. However, as discussed in this article, there is scant evidence that the microorganisms benefit from such associations, and it is unclear how the benefit or harm incurred by microorganisms that require the association can be demonstrated.  相似文献
