The properties of ideal HABb are defined on the basis of the results of box titrations of a random set of HBAg-positive sera with diluted human HBAb in microimmunodiffuse reaction. Ideal HBAb makes it possible to determine relative strength of antibody under test. In contact experiments with different experimental conditions, electrochemical Ag+ solutions exhibited better antimicrobial effectiveness against bacteria, a yeast species, and a mold than did analogous silver solutions from inorganic salts. We discovered that Yeast Extract Mannitol (YEM) medium possessed immense potential to generate silver nanoparticles from AgNO3 upon autoclaving, which was evident from (i) alteration in color of the medium; (ii) peak at ∼410 nm in UV-Vis spectrum due to surface plasmon resonance specific to silver nanoparticles; and (iii) TEM investigations. TEM coupled with EDX confirmed that distinct nanoparticles were composed of silver. Yeast extract and mannitol were key components of YEM medium responsible for the formation of nanoparticles. PXRD analysis indicated crystalline geometry and Ag/Ag2O phases in nanoparticles generated with YEM medium, yeast extract and mannitol. Our investigations also revealed that both mannitol and yeast extract possessed potential to convert ∼80% of silver ions in 0.5 mM AgNO3 to nanoparticles, on autoclaving for 30 min at 121°C under a pressure of 1.06 kg/cm2. Addition of filter sterilized AgNO3 under ambient conditions to pre-autoclaved YEM medium and yeast extract brought about color change due to the formation of silver nanoparticles, but required prolonged duration. In general, even after 72 h intensity of color was significantly less than that recorded following autoclaving. Silver nanoparticles formed at room temperature were more heterogeneous compared to that obtained upon autoclaving. In summary, our findings demonstrated that (i) YEM medium and its constituents promote synthesis of silver nanoparticles; and (ii) autoclaving enhances rapid synthesis of silver nanoparticles by YEM medium, yeast extract and mannitol.  相似文献

We report equilibrium dialysis and electric dichroism studies of the two strong complexes (I and II) of silver ion with DNA. Cooperative conversion of DNA to the stronger type I complex results in a 9% length decrease, and a structure in which intercalated ethidium is perpendicular to the helix axis. Upon addition of more Ag+ to form the type II complex, the DNA length reverts to its original value and bound ethidium once again becomes tilted from the plane perpendicular to the helix axis. In both type I and type II Ag (I) - DNA complexes, ethidium binding is mildly cooperative. We interpret the results in terms of a sequence of silver-induced cooperative switches of DNA from its B-form structure with propeller twisted base pairs to a structure with flat base pairs in the type I complex, and back again to propellered base pairs in the type II complex.  相似文献

Abstract Radioisotope equilibration techniques have been used to determine the intracellular concentration of K+, Na+ and Cl?, together with the unidirectional ion fluxes across the plasmalemma of Porphyra purpurea. Influx and efflux of 42K+, 24Na+ and 36C1? are biphasic, the rapid, initial uptake and loss of tracer from individual thalli being attributable to desorption from extracellular regions. Cellular fluxes are slower and monophasic, cells discriminating in favour of K+ and Cl? and against Na+. A comparison between the equilibrium potential of individual ion species and the measured membrane potential demonstrates that there is an active component of K+ and Cl? influx and Na+ efflux. ‘Active’ uptake and ‘passive’ loss of K+ and Cl? are reduced when plants are kept in darkness, suggesting that a fraction of the transport of K+ and Cl? may be due to ‘exchange diffusion’ (K+/K+ and Cl?/Cl?antiport).  相似文献

The cell of Caloglossa leprieurli is a polycaryon. The sexual thalli are usually dioecious. Its life cycle involves the alternation of three generations. In the reproductive season, the appearance of tetrasporophyte and female and male gametophyte shows evident difference. The tetrasporophyte is big and flat. The branches are sparse. Many small red spots, i.e. tetrasporogial groups, can be seen with the naked eyes in the upper part of the branch. The terminal part of the female gametophyte is a little twisted with thick branches. The cystocarps are spherical and most of them are located in ventral side of the upper branch points. The male gametophyte is smaller with less branches. Its terminal part is a little twisted with pale colour. The pit connections of the vegetative cells of every thallus are all well developed. The carpogonial branch consists of four cells. Generally, it is formed by the division of the pericentral cell. After fertilization of the carpogonium, an auxiliary cell is formed by the division of supporting cell. Usually five to seven, even more than ten young procarps can be formed at the apical part of the reproductive branch. The procarps often occur on consecutive segments of pericentral ceils. But among most of them only a carpogonium is fertilized and developes into cystocarp. When a carpogonium is fertilized, the other one on this branch usually cease further development. The spermatangia are formed on the lateral wing cells of both sides of the upper part midrib of a reproductive branch and distributed on both dorsal and ventral surfaces. A vegetative cell is divided into three cells in parallel to the surface of the thallus. The cortical cells under the two outer surfaces forms three to four permatangial mother cells. Each spermatangial mother cell divides into two to four spermatangia. After the sperm dispersal of the mature spermatangia, the spermatangial mother cells are still retained on both sides of the middle discoid cells.  相似文献

Silver nanoparticles (Ag NPs) are increasingly used in many products and are expected to end up in the aquatic environment. Mussels have been proposed as marine model species to evaluate NP toxicity in vitro. The objective of this work was to assess the mechanisms of toxicity of Ag NPs on mussel hemocytes and gill cells, in comparison to ionic and bulk Ag. Firstly, cytotoxicity of commercial and maltose stabilized Ag NPs was screened in parallel with the ionic and bulk forms at a wide range of concentrations in isolated mussel cells using cell viability assays. Toxicity of maltose alone was also tested. LC50 values were calculated and the most toxic Ag NPs tested were selected for a second step where sublethal concentrations of each Ag form were tested using a wide array of mechanistic tests in both cell types. Maltose-stabilized Ag NPs showed size-dependent cytotoxicity, smaller (20 nm) NPs being more toxic than larger (40 and 100 nm) NPs. Maltose alone provoked minor effects on cell viability. Ionic Ag was the most cytotoxic Ag form tested whereas bulk Ag showed similar cytotoxicity to the commercial Ag NPs. Main mechanisms of action of Ag NPs involved oxidative stress and genotoxicity in the two cell types, activation of lysosomal AcP activity, disruption of actin cytoskeleton and stimulation of phagocytosis in hemocytes and increase of MXR transport activity and inhibition of Na-K-ATPase in gill cells. Similar effects were observed after exposure to ionic and bulk Ag in the two cell types, although generally effects were more marked for the ionic form. In conclusion, results suggest that most observed responses were due at least in part to dissolved Ag.  相似文献

A fucoidan consisting of L-fucose, sulfate and acetate in a molar proportion of 1:1.23:0.36 was isolated from the Pacific brown seaweed Fucus evanescens. The structures of its desulfated and de-O-acetylated derivatives were investigated by 1D and 2D (1)H and (13)C NMR spectroscopy, and the data obtained were confirmed by methylation analysis of the native and desulfated polysaccharides. The fucoidan was shown to contain a linear backbone of alternating 3- and 4-linked alpha-L-fucopyranose 2-sulfate residues: -->3)-alpha-L-Fucp(2SO(3)(-))-(1-->4)-alpha-L-Fucp(2SO(3)(-))-(1-->. Additional sulfate occupies position 4 in a part of 3-linked fucose residues, whereas a part of the remaining hydroxyl groups is randomly acetylated.  相似文献
