钙调素对花粉萌发和花粉管生长的效应 总被引:1,自引:0,他引:1
牛脑和玉米胚CaM能显著促进花粉萌发和花粉管生长(图1),而CaM抑制剂TFP、CPZ及另外两个专一性更强的抑制剂Compound48/80和W7均严重抑制甚至阻止花粉的萌发(图2,3)。用对CaM亲和性较低的W7同系物W5,在与W7同样浓度下,对花粉萌发和花粉管生长无明显影响。此外,W7对花粉萌发和花粉管生长的抑制效应可被外源CaM所消除(图4)。在花粉萌发过程中,其内源CaM含量显著上升,在花粉萌发率接近最大值时,花粉CaM含量达最高水平(图5)。上述结果表明CaM对花粉萌发和花粉管生长的调控起重要作用。 相似文献

We examined exocytosis during oscillatory growth in lily (Lilium formosanum and Lilium longiflorum) and tobacco (Nicotiana tabacum) pollen tubes using three markers: (1) changes in cell wall thickness by Nomarski differential interference contrast (DIC), (2) changes in apical cell wall fluorescence in cells stained with propidium iodide (PI), and (3) changes in apical wall fluorescence in cells expressing tobacco pectin methyl esterase fused to green fluorescent protein (PME-GFP). Using PI fluorescence, we quantified oscillatory changes in the amount of wall material from both lily and tobacco pollen tubes. Measurement of wall thickness by DIC was only possible with lily due to limitations of microscope resolution. PME-GFP, a direct marker for exocytosis, only provides information in tobacco because its expression in lily causes growth inhibition and cell death. We show that exocytosis in pollen tubes oscillates and leads the increase in growth rate; the mean phase difference between exocytosis and growth is –98° ± 3° in lily and –124° ± 4° in tobacco. Statistical analyses reveal that the anticipatory increase in wall material predicts, to a high degree, the rate and extent of the subsequent growth surge. Exocytosis emerges as a prime candidate for the initiation and regulation of oscillatory pollen tube growth. 相似文献

The effect of phospholipase C (PLC) signaling pathway on lily (Lilium davidii Duch.) pollen tube elongation was examined by means of microinjection. Pollen tube elongation was inhibited by microinjecting antibodies against animal PLCβ1-3 or inositol-1,4,5-triphosphate receptor (IP3R2, 3), but was not affected by antibodies against animal PLCβ4 or IP3R1. Pollen tube elongation was also stimulated significantly by microinjecting IP3. The results suggest that PLC-IP3 signaling pathway might present in pollen system and be involved in pollen tube growth. 相似文献

采用花粉离体萌发法研究不同培养基组分和培养条件对桃花粉萌发和花粉管生长的影响,同时对不同贮藏温度下的桃花粉寿命进行研究.结果表明:固体培养基与液体培养基对桃的花粉萌发率和花粉管长度影响差异不显著;10%蔗糖是大多数桃品种花粉的最适萌发条件;硼能提高桃花粉的萌发率,但对花粉管的生长没有促进作用;桃花粉在20℃~25℃的培养温度下萌发率最高,花粉管最长;桃花粉萌发率和花粉管长度在培养前3 h内上升最快,3~5 h上升趋势减弱,5 h后基本停止;随着贮藏温度的升高和贮藏时间的延长,花粉生活力呈降低的趋势. 相似文献

以黄连木花粉为试材,采用离体培养法研究了培养基组分和植物生长调节物质对黄连木花粉萌发和花粉管生长的影响.结果表明:花粉萌发和花粉管生长的适宜蔗糖浓度为15%,适宜培养温度为25℃;该培养条件下,花粉萌发率和花粉管长度分别达最大值63.3%和412.1 μm.硼酸、赤霉素(GA3)和吲哚乙酸(IAA)在一定浓度范围内,可以促进黄连木花粉萌发和花粉管生长,浓度过高时起抑制作用;最适宜黄连木花粉萌发和花粉管生长的硼酸浓度、赤霉素(GA3)和吲哚乙酸(IAA)浓度分别为100、50和15 mg/L. 相似文献

研究了果梅(Prunus mumeSieb.1et Zucc.)花粉在不同培养基组分、花粉不同培养密度和不同温度及培养时间的离体萌发和花粉管生长特性.结果表明:细叶青花粉萌发及花粉管生长最适宜的液体培养基为30mmol/L MES(pH 6.5)缓冲液中含20%蔗糖,0.01%硼酸,20%PEG-4000,0.03?(NO3)2?4H2O,0.02%Mg-SO4?7H2O;萌发率达45.03%,花粉管长度达597.2μm.适宜于果梅花粉萌发和花粉管生长的花粉粒密度为20~80粒/μL.培养温度过高或过低都不利于果梅花粉的萌发和生长,25℃时花粉萌发和花粉管生长最好.细叶青"、月世界"、莺宿"3个品种的平均萌发率为48.6%,平均花粉管长度为762.3μm.果梅花粉在不同培养温度下,萌发及生长不同,在25℃条件下花粉管生长速度最快,集中在0~12 h内,3个品种花粉管平均生长速度为58.5μm/h. 相似文献

Pine (Pinus roxburghii) pollen grown in suspension cultureswas used to study the effects of growth regulators and lightconditions on germination and pollen tube growth. Indol-3-ylacetic acid, gibberellic acid, ethylene, abscisic acid and cyclicAMP (cAMP) at low concentrations (110 mg 11) promotedgermination and tube growth. Addition of 1 and 10 mg 11cAMP to any of the growth regulators had a promotory effect.Pollen tube growth decreased in white light as compared to thedark, and was increased in red light. Fine structural events in the mature stylar transmitting tissue ofsweet pepper (Capsicum frutescens L. var. grossum Bally) have been investigated byelectron microscope with regard to pollen tribe grwoth. The transmitting tissue consists of parenchymatous cells with large intercellular spaces filled with an electron densesubstance. The pollen tubes grow through the intercellular spaces in the intercellular substance. Cells of this tissue are rich in organelles, especially the rough and tubular ER, and numerous lomasomes near the plasma membrane. It is demonstrated that they function as secretory cells. On the other hand, the fact that the transmitting tissue contains many large amyloplastids with several starch grains in the cytoplasm and numerous globular protein bodies in vacuoles, indicates that the transmitting tissue mayhave some nutritive value for the growth of pollen tubes. The results obtained from this observation are in agreement with those of mostspecies reported by other authors and support the conclusion that the transmitting tissue is not a collenchyma tissue and the nature of intercellular substances is essentially same as that of the middle lamella. 相似文献

以烟草为材料,通过半体内实验,就花柱和花粉胞外钙调素对花粉萌发和花粉管伸长的影响进行了观察。发现用EGTA及钙调素抗血清处理柱头或花粉均可抑制花粉在柱头上的萌发;向花柱引导组织中显微注射纯化钙调素可促进花粉管束伸长,而注射钙调素抗血清可抑制花粉管束伸长;同时证实玉米花柱和花粉细胞壁中均存在钙调素及钙调素结合蛋白,而且花粉和花柱细胞壁中钙调素结合蛋白的种类有差异。结果表明存在于花粉和花柱细胞外的钙调素对花粉萌发和花粉管伸长均有促进作用。 相似文献

为明确扇脉杓兰花粉形态结构及雄性生殖特性,利用扫描电镜、透射电镜和荧光显微镜对花粉形态和超微结构及花粉管生长过程进行观察。结果表明:(1)扇脉杓兰单粒花粉长球形,表面光滑无特征纹饰,有少量胶黏物质,一些表面有2个或以上的深凹陷,凹陷内有球形突起的内容物。(2)花粉壁分为由棒状的基柱小单元组成的外壁和纤维素果胶组成的内壁,有覆盖层;生殖细胞近圆形,细胞核大而致密;营养细胞多弧形,核质分散;花粉粒细胞质含有大量的线粒体、质体和小泡等细胞器,淀粉、蛋白质和多糖含量丰富。(3)花粉管萌发后沿子房壁方向伸长,授粉20d花粉管伸长生长并不明显,授粉30d伸长的花粉管中出现大量胼胝质塞,并且伸长方向转为胚珠中间,花粉管逐渐接近胚珠,在授粉后50d基本完成受精作用。研究认为,扇脉杓兰花粉发育正常,不阻碍有性生殖过程。 相似文献

Proper vesicle tethering and membrane fusion at the cell plate are essential for cytokinesis. Both the vesicle tethering complex exocyst and membrane fusion regulator KEULE were shown to function in cell plate formation, but the exact mechanisms still remain to be explored. In this study, using yeast two-hybrid (Y-2-H) assay, we found that SEC6 interacted with KEULE, and that a small portion of C-terminal region of KEULE was required for the interaction. The direct SEC6-KEULE interaction was supported by further studies using in vitro pull-down assay, immunoprecipitation, and in vivo bimolecular florescence complementation (BIFC) microscopy, sec6 mutants were male gametophytic lethal as reported; however, pollen-rescued sec6 mutants (PRsec6) displayed cytokinesis defects in the embryonic cells and later in the leaf pavement cells and the guard cells. SEC6 and KEULE proteins were co-localized to the cell plate during cytokine- sis in transgenic Arabidopsis. Furthermore, only SEC6 but not other exocyst subunits located in the cell plate interacted with KEULE in vitro. These results demonstrated that, like KEULE, SEC6 plays a physiological role in cytokinesis, and the SEC6-KEULE interaction may serve as a novel molecular linkage between arriving vesicles and membrane fusion machin- ery or directly regulate membrane fusion during cell plate formation in plants. 相似文献
