MicroRNAs(miRNAs)是一类大约22个核苷酸的非编码RNA.它能通过调控生长因子表达,引发肿瘤形成、细胞增殖和组织器官发育.本文通过构建miR-129-5p靶基因序列的双荧光素酶报告载体分析了miR-129-5p与靶基因之间的关系,应用脂质体转染技术和实时荧光定量技术观察了miR-129-5p在小鼠乳腺上皮细胞中的表达量及其变化,通过CASY细胞活力仪检测转染后的细胞增殖与活力变化,采用Western 印迹方法检Igf-1的变化.结果表明:miR-129-5p在小鼠乳腺青春期表达最高,成功构建了Igf-1基因 3′UTR荧光素酶报告载体, miR-129-5p抑制其荧光素酶活性(P <0.01),转染抑制子后miR-129-5p表达降低(P < 0.01),胰岛素样生长因子(Igf-1)表达增强(P <0.05),细胞增殖和活力增强(P <0.01),结果提示:miR-129-5p可能通过抑制靶基因蛋白Igf 1的表达,进而抑制小鼠乳腺上皮细胞增殖和活力.  相似文献

微RNA-375(microRNA-375,miR-375)是最早在胰腺组织中发现的微RNA,参与胰岛的形成和发展,具有调节胰岛素分泌的功能. 新近研究发现,miR-375在多种肿瘤组织(包括呼吸系统、消化系统、泌尿生殖系统、皮肤和妇科肿瘤组织)中和食管鳞状细胞癌、肝癌和胰腺癌患者外周血中明显表达异常. miR 375可通过调控多个靶基因(如:3-磷酸肌醇依赖性蛋白激酶1、14-3-3ζ、Janus激酶2、p53、丝裂原活化蛋白激酶、Wnt、血管内皮生长因子、胰岛素样生长因子 1受体、星形胶质细胞升高基因 1/异黏蛋白、地塞米松诱导的Ras相关蛋白1和特异蛋白1等相关基因),参与肿瘤的发生和发展过程. 提高细胞内miR 375的水平能够抑制肿瘤细胞(如:头颈鳞癌、胃癌、食管鳞癌、黑色素瘤和乳腺癌等肿瘤细胞)的增殖和迁移. 因此,具有抑癌活性的miR 375是一种有临床价值的新型肿瘤分子标志物和肿瘤靶向治疗的新靶点.  相似文献

先前研究表明,miR-186-5p在人类许多恶性肿瘤中扮演抑癌基因的作用,但其在肺腺癌上皮-间质转化(epithelial-mesenchymal transition,EMT)中的作用并不明确。本研究旨在证明,miR-186-5p可通过靶向调控PTTG1抑制肺腺癌细胞的上皮-间质转化。我们首先分析了miR-186-5p在人肺癌细胞中的表达。荧光定量PCR(QRT-PCR)结果显示,与人正常肺上皮细胞BEAS-2B相比,肺腺癌细胞SPC-A1、A549中的miR-186-5p表达量明显降低。为研究miR-186-5p在肺腺癌细胞中的功能,利用GV369-miR-186-5p表达载体,实现了在A549细胞中的过表达。基因转染结合Transwell侵袭结果显示,与对照质粒转染的A549细胞相比,过表达miR-186-5p的A549细胞的体外侵袭能力明显下降。Western印迹检测细胞中EMT相关标志物揭示,GV369-miR-186-5p转染的A549细胞中的上皮-钙黏着蛋白(E-cadherin)表达明显上调,而神经-钙黏着蛋白(N-cadherin)和波形蛋白(vimentin)表达明显下调。同时,GV369-miR-186-5p转染引起其靶基因--垂体肿瘤转化基因1(pituitary tumor-transforming gene 1,PTTG1)编码蛋白在A549细胞中明显降低。重要的是,过表达miR-186-5p与敲减PTTG1均可导致上皮-钙黏着蛋白表达上调,而神经-钙黏着蛋白和波形蛋白下调|而miR-186-5p和PTTG1表达载体共转染后,3种EMT相关标志物在A549的表达与对照细胞的表达无明显差异,提示过表达PTTG1可抵消miR-186-5p对EMT相关标志物表达的影响。综上所述,miR-186-5p可通过靶向调控PTTG1抑制EMT的发生,进而抑制肺腺癌细胞的侵袭转移。  相似文献

目的探讨LncRNA AC130710通过miR-129-5P/WNT4轴对子宫内膜癌细胞(HEC-1A细胞)增殖、凋亡及上皮间质转化(EMT)的影响及机制研究。 方法通过实时荧光定量PCR检测LncRNA AC130710、miR-129-5P和WNT4在子宫内膜癌细胞(HEC-1A细胞)和人子宫内膜上皮细胞(HEEC)中的表达。细胞分别转染(1)siRNA NC、AC130710 siRNA、WNT4 siRNA;(2)inhibitor NC、miR-129-5P inhibitor;(3)pcDNA-3.1 (+)+mimics NC、pcDNA-AC130710+mimics NC、pcDNA-3.1 (+)+miR-129-5P mimics、pcDNA-AC130710+miR-129-5P mimics。MTT实验检测LncRNA AC130710、miR-129-5P和WNT4的表达对HEC-1A细胞增殖能力的影响;Western blot检测LncRNA AC130710、miR-129-5P和WNT4的表达对HEC-1A细胞凋亡相关蛋白B淋巴细胞瘤-2基因相关蛋白X (Bax)、剪切的半胱氨酰天冬氨酸特异性蛋白酶-3 (cleaved caspase-3)、cleaved caspase-9和B淋巴细胞瘤-2基因(Bcl-2)表达的影响;Western blot检测LncRNA AC130710、miR-129-5P和WNT4的表达对HEC-1A细胞EMT的影响。miRanda和双荧光素酶报告基因实验分析LncRNA AC130710和miR-129-5P之间的关系,TargetScan数据库分析miR-129-5P与WNT4的相关性,双荧光素酶报告基因检测miR-129-5P与WNT4的相互作用;RT-qPCR法检测LncRNA AC130710通过miR-129-5P对WNT4表达的影响。两组间比较采用独立样本t检验,多组间比较采用单因素方差分析,两两比较采用LSD-t检验。 结果与HEEC细胞比较,HEC-1A细胞中AC130710表达水平(1.86±0.21比0.85±0.06)、WNT4表达水平(1.88±0.26比1.08±0.12)升高;HEC-1A细胞中miR-129-5P表达水平(0.89±0.16比1.76±0.08)降低。与转染siRNA NC比较,转染AC130710 siRNA细胞内Bax、cleaved caspase-3、cleaved caspase-9、E-cadherin蛋白相对表达水平[(1.37±0.14比0.84±0.21),(1.08±0.16比0.37±0.07),(1.26±0.24比0.39±0.06),(1.87±0.17比1.32±0.26)]上升,Bcl-2、N-cadherin、Snail和Vimentin蛋白相对表达水平[(0.38±0.08比1.18±0.14),(0.36±0.04比0.85±0.24),(0.35±0.09比1.12±0.18),(0.42±0.10比1.26±0.27)]下降;与转染inhibitor NC比较,转染miR-129-5P inhibitor细胞的Bcl-2、N-cadherin、Snail和Vimentin蛋白相对表达水平[(0.98±0.07比0.65±0.08),(1.39±0.15比0.68±0.09),(0.95±0.08比0.42±0.06),(1.16±0.16比0.54±0.02)]上升,Bax、cleaved caspase-3、cleaved caspase-9、E-cadherin蛋白相对表达水平[(0.27±0.09比0.85±0.13),(0.48±0.05比1.16±0.28),(0.52±0.14比1.19±0.15),(0.43±0.09比1.08±0.26)]下降;与转染siRNA NC比较,转染WNT4 siRNA细胞的Bcl-2、N-cadherin、Snail和Vimentin蛋白相对表达水平[(0.23±0.08比0.84±0.12),(0.28±0.09比1.14±0.17),(0.42±0.23比1.06±0.15),(0.35±0.08比1.13±0.08)]降低,Bax、cleaved caspase-3、cleaved caspase-9、E-cadherin蛋白相对表达水平[(0.96±0.12比0.42±0.08),(1.13±0.25比0.45±0.06),(1.54±0.23比0.72±0.12),(1.87±0.24比1.26±0.18)]上升。 结论LncRNA AC130710可通过miR-129-5P/WNT4轴调控子宫内膜癌HEC-1A细胞增殖、凋亡及EMT。  相似文献

摘要 目的:探讨长链非编码核糖核酸(LncRNA)KCNQ1OT1靶向调控miR-124-3p/高迁移率族蛋白B1(HMGB1)轴对高糖诱导肾小球系膜细胞(HMC)增殖、凋亡及纤维化的影响。方法:将人HMC分为对照组(NC组)、高糖组(30 mmol/L葡萄糖)、阴性序列(si-NC)组、KCNQ1OT1小干扰核糖核酸(RNA)(si-KCNQ1OT1)组、si-KCNQ1OT1+模拟对照序列(miR-NC)组、si-KCNQ1OT1+miR-124-3p抑制剂(miR-124-3p inhibitor)组,各组在转染后进行高糖处理。实时荧光定量聚合酶链式反应(RT-qPCR)检测LncRNA KCNQ1OT1信使核糖核酸(mRNA)、miR-124-3p mRNA、HMGB1 mRNA表达;四甲基偶氮唑盐比色法(MTT)检测细胞增殖活性;流式细胞术检测细胞凋亡率;蛋白印迹法(Western blot)检测HMGB1蛋白、增殖相关蛋白[细胞周期蛋白1(CyclinD1)]、细胞凋亡蛋白[半胱氨酸蛋白酶-3(caspase-3)、半胱氨酸蛋白酶蛋白9(caspase-9)]、细胞纤维化蛋白[纤维连接蛋白(FN)、细胞黏附分子-1(ICAM-1)]表达;双荧光素酶报告基因实验验证LncRNA KCNQ1OT1与miR-124-3p与HMGB1之间的靶向关系。结果:与NC组比较,高糖组KCNQ1OT1 mRNA、HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显上升,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显下降(P<0.05);与高糖组、si-NC组比较,si-KCNQ1OT1组KCNQ1OT1 mRNA、HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显下降,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显上升(P<0.05);与si-KCNQ1OT1组、si-KCNQ1OT1+miR-NC组比较,si-KCNQ1OT1+miR-124-3p inhibitor组HMGB1 mRNA及HMGB1蛋白、CyclinD1蛋白、FN蛋白、ICAM-1蛋白表达、HMC活性(24 h、48 h和72 h)明显上升,miR-124-3p mRNA、caspase-3蛋白及caspase-9蛋白表达、HMC凋亡率明显下降(P<0.05)。较miR-NC组与KCNQ1OT1-WT共转染组而言,miR-124-3p mimic组与KCNQ1OT1-WT共转染组细胞荧光素酶活性明显降低(P<0.05);较miR-NC组与HMGB1-WT共转染组而言,miR-124-3p mimic组与HMGB1-WT共转染组细胞荧光素酶活性明显降低(P<0.05)。结论:LncRNA KCNQ1OT1可以靶向下调miR-124-3p mRNA表达,上调HMGB1 mRNA及HMGB1蛋白表达,促进高糖诱导HMC增殖,抑制凋亡,促进细胞纤维化发展。  相似文献

B3型柯萨奇病毒(Coxsackie virus B3,CVB3)感染与1型糖尿病发病及胰岛β细胞损伤有关,但机制并不清楚。病毒性心肌炎的研究证实CVB3通过调控微小RNA(microRNAs,miRNAs)的表达引起心肌细胞损伤。本研究的目的是观察CVB3感染的胰岛β细胞中miRNAs表达的变化及生物学意义,进而初步探究CVB3感染引起胰岛β细胞损伤的分子机制。本研究培养了人胰岛β细胞株,感染CVB3后采用miRNAs芯片及荧光定量PCR检测miRNAs表达的变化;转染miR-阴性对照(NC)或miR-146a-5p后感染CVB3,检测细胞活力、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)及白介素-1β(IL-1β)的含量、IRAK-1及TRAF-6的表达水平;采用双荧光素酶报告基因实验验证miR-146a-5p靶向IRAK-1及TRAF-6。结果显示:CVB3组中miR-101a-3p、miR-140-5p、miR-146a-5p、miR-146b-5p、miR-340-5p的表达水平均低于对照组且miR-146a-5p表达降低最显著;miR-NC...  相似文献

摘要 目的:探究微小核糖核酸(miR)-152-3p调控果蝇Notch同源物1(Notch1)/Delta样配体4(DLL4)通路对家兔深II度烧伤创面血管生成的影响。方法:将50只新西兰家兔随机分为对照组、模型组、miR-152-3p拮抗剂(antagomir)组、miR-152-3p antagomir阴性对照+空载组、miR-152-3p antagomir+Notch1敲低组,每组10只,除对照组外其余各组家兔构建深II度烧伤模型,分组给药处理后,实时荧光定量聚合酶链式反应(qRT-PCR)检测各组家兔创面组织miR-152-3p与Notch1、DLL4 mRNA表达;检测各组家兔创面愈合率及微循环血流灌注值(MPD);免疫组织化学染色检测各组家兔创面微血管密度(MVD);酶联免疫吸附反应(ELISA)检测各组家兔血清血管内皮细胞生长因子(VEGF)及促血管生成素1(Ang1)水平;免疫印迹检测各组家兔创面组织VEGF、Ang1与Notch1/DLL4通路蛋白表达;双荧光素酶报告基因实验检测兔脐静脉内皮细胞中miR-152-3p对Notch1及DLL4的靶向调节。结果:与对照组相比,模型组家兔创面组织miR-152-3p与Notch1、DLL4 mRNA表达升高(P<0.05),创面MPD及MVD、血清VEGF及Ang1水平、创面组织VEGF与Ang1蛋白表达降低(P<0.05)。与模型组相比,miR-152-3p antagomir组家兔创面组织miR-152-3p mRNA表达降低(P<0.05),创面愈合率、创面MPD及MVD、血清VEGF及Ang1水平、创面组织Notch1、DLL4 mRNA及蛋白表达、创面组织VEGF与Ang1蛋白表达升高(P<0.05);miR-152-3p antagomir阴性对照+空载组家兔各指标无明显差异(P>0.05);与miR-152-3p antagomir组相比,miR-152-3p antagomir+Notch1敲低组家兔创面组织miR-152-3p mRNA表达无明显差异(P>0.05),创面愈合率、创面MPD及MVD、血清VEGF及Ang1水平、创面组织Notch1、DLL4 mRNA及蛋白表达、创面组织VEGF与Ang1蛋白表达降低(P<0.05)。miR-152-3p可靶向下调兔脐静脉内皮细胞中Notch1及DLL4的表达。结论:敲低miR-152-3p可通过上调Notch1/DLL4通路而增强家兔深II度烧伤创面血管生成,进而促进其创面愈合。  相似文献

摘要 目的:探讨微小核糖核酸(miRNA)-324-5p、miR-605-3p在脑胶质瘤组织的表达及与临床病理参数和预后的关系。方法:选取2018年1月~2019年12月徐州医科大学附属医院收治的90例脑胶质瘤患者。收集术中部分瘤组织和瘤旁组织,采用实时荧光定量聚合酶链式反应(qRT-PCR)检测miR-324-5p、miR-605-3p表达。根据脑胶质瘤组织中miR-324-5p、miR-605-3p表达的平均值分为高表达组和低表达组,采用Kaplan-Meier法分析不同miR-324-5p、miR-605-3p表达脑胶质瘤患者生存情况,采用多因素Cox回归分析脑胶质瘤患者预后的影响因素。结果:与瘤旁组织比较,脑胶质瘤组织中miR-324-5p、miR-605-3p表达降低(P<0.05)。不同分化程度、淋巴结转移、世界卫生组织(WHO)中枢神经系统肿瘤分类的脑胶质瘤患者miR-324-5p、miR-605-3p表达比较有差异(P<0.05)。90例脑胶质瘤患者3年总生存率为36.67%(33/90)。Kaplan-Meier生存曲线分析显示,miR-324-5p高表达组、miR-605-3p高表达组总生存率高于miR-324-5p低表达组、miR-605-3p低表达组(P<0.05)。多因素Cox回归分析显示,低分化、淋巴结转移和WHO中枢神经系统肿瘤分类Ⅲ~Ⅳ级为脑胶质瘤患者死亡的独立危险因素,miR-324-5p和miR-605-3p升高为独立保护因素(P<0.05)。结论:脑胶质瘤组织中miR-324-5p、miR-605-3p呈低表达,与分化程度、淋巴结转移、WHO中枢神经系统肿瘤分类有关,miR-324-5p、miR-605-3p低表达还可导致不良预后。  相似文献

目的: MiR-378a-5p是一种被认为在多种肿瘤发生过程中具有抑制肿瘤生长的微小RNA。然而miR-378a-5p在鼻咽癌中的 作用尚未见报道。因此,本文旨在通过临床样本的miRNA 表达谱分析以及细胞学实验从而揭示miR-378a-5p在鼻咽癌肿瘤发生过程中的作用。方法与结果:我们通过生物信息学的方法获取了鼻咽癌临床样本中miR-378a-5p的表达信息并通过与正常组织的 对比发现miR-378a-5p在鼻咽癌肿瘤组织中表达水平显著降低(P<0.01)。其次,我们发现高表达miR-378a-5p的鼻咽癌CNE-1 细 胞增殖速度显著较对照组降低(约40%~50%)。克隆形成实验证实了瞬时转染miR-378a-5p的鼻咽癌CNE-1 细胞的克隆形成数 量显著减弱。我们通过将稳定表达miR-378a-5p的CNE-1 细胞注射到裸鼠体内形成移植瘤并记录肿瘤生长曲线,结果显示 miR-378a-5p高表达组的裸鼠移植瘤体积明显较对照组小约50%,肿瘤重量显著降低(对照组0.33 g,处理组0.15 g)。结论:本研究通过对临床样本的分析以及在细胞和动物水平的实验验证揭示了miR-378a-5p具有抑制鼻咽癌肿瘤细胞增殖和肿瘤生长的作用。  相似文献

摘要 目的:研究血清外泌体长链非编码核糖核酸(lncRNA)前列腺癌基因表达标记1(PCGEM1)、微小核糖核酸(miR)-129-5p与非小细胞肺癌(NSCLC)患者临床病理特征及预后的关系。方法:选取2016年2月-2018年1月南京脑科医院收治的125例NSCLC患者作为NSCLC组,同期选取体检的70例健康人群作为健康组。采集两组静脉血,提取血清外泌体;采用实时定量聚合酶链式反应(qRT-PCR)检测血清外泌体lncRNA PCGEM1、miR-129-5p表达情况;采用Pearson相关性分析lncRNA PCGEM1与miR-129-5p的关系。并分析血清外泌体lncRNA PCGEM1、miR-129-5p与NSCLC患者临床病理特征的关系。对NSCLC患者行5年随访,绘制Kaplan-Meier曲线分析预后情况,多因素Cox比例风险回归模型分析预后不良危险因素,受试者工作特征(ROC)曲线分析lncRNA PCGEM1、miR-129-5p对NSCLC预后的预测价值。结果::NSCLC组lncRNA PCGEM1相对表达量高于健康组,miR-129-5p相对表达量低于健康组(P<0.05)。血清外泌体lncRNA PCGEM1相对表达量与miR-129-5p表达呈负相关(r= -0.420,P<0.05)。血清外泌体lncRNA PCGEM1、miR-129-5p表达与患者TNM分期、分化程度、淋巴结转移有关(P<0.05)。Kplan-Meier生存曲线显示,lncRNA PCGEM1低表达组5年生存率69.05%高于lncRNA PCGEM1高表达组35.53%,miR-129-5p高表达组5年生存率68.09%高于miR-129-5p低表达组33.80%。多因素Cox比例风险回归显示,TNM分期III期、有淋巴结转移、lncRNA PCGEM1高表达、miR-129-5p低表达为NSCLC患者预后不良的独立危险因素(P<0.05)。ROC曲线显示,lncRNA PCGEM1、miR-129-5p联合检测对NSCLC预后的预测曲线下面积(AUC)为0.865,预测价值高于两者单独预测。结论:NSCLC患者血清外泌体lncRNA PCGEM1表达上调、miR-129-5p表达下调,二者表达与NSCLC患者TNM分期、分化程度、淋巴结转移有关,且与患者预后密切相关,对NSCLC预后不良具有较好预测价值。  相似文献

Reduced expression of microRNA-129 (miR-129) has been reported in several types of tumor cell lines as well as in primary tumor tissues. However, little is known about how miR-129 affects cell proliferation in gastric cancer. Here, we show that all miR-129 family members, miR-129-1-3p, miR-129-2-3p, and miR-129-5p, are down-regulated in gastric cancer cell lines compared with normal gastric epithelial cells. Furthermore, using the real-time cell analyzer assay to observe the growth effects of miR-129 on gastric cancer cells, we found that all three mature products of miR-129 showed tumor suppressor activities. To elucidate the molecular mechanisms underlying down-regulation of miR-129 in gastric cancer, we analyzed the effects of miR-129 mimics on the cell cycle. We found that increased miR-129 levels in gastric cancer cells resulted in significant G0/G1 phase arrest. Interestingly, we showed that cyclin dependent kinase 6 (CDK6), a cell cycle-associated protein involved in G1-S transition, was a target of miR-129. We also found that expression of the sex determining region Y-box 4 (SOX4) was inversely associated with that of miR-129-2-3p and miR-129-5p but not of miR-129-1-3p. Together, our data indicate that all miR-129 family members, not only miR-129-5p, as previously thought, play an important role in regulating cell proliferation in gastric cancer.  相似文献

近年来,越来越多的证据表明,长非编码RNAs在肿瘤发生发展中发挥重要作用。位于12号染色体的长非编码RNA RP4-816N1.7(简称RP4)在乳腺癌细胞中的作用未见报道。我们通过实时荧光定量PCR证实,RP4在乳腺癌细胞中的表达量普遍低于其在正常乳腺上皮细胞MCF-10A中的表达量。RP4在MCF-7和MDA-MB-231中表达量分别比其在MCF-10A中的表达量下调21.57%和91.33%。过表达RP4可明显抑制乳腺癌细胞增殖。敲低RP4可显著增加乳腺癌细胞的增殖能力。生物信息学预测,RP4可能与miR-183-5p.1结合,且叉头蛋白O1(FOXO1)可能是miR-183-5p.1的潜在靶标。实时荧光定量PCR结果提示,RP4可下调miR-183-5p.1,而miR-183-5p.1也可下调RP4和FOXO1的表达。双荧光素酶报告基因结果证实,miR-183-5p.1可与RP4结合,下调其表达,也能与FOXO1 3′UTR结合,抑制其mRNA和蛋白质水平的表达量。最后,本文通过BrdU实验证实,RP4通过FOXO1抑制乳腺癌细胞的增殖。总之,RP4通过内源性结合miR-183-5p.1,上调FOXO1表达,进而抑制乳腺癌细胞增殖。  相似文献

miR-129-5p is implicated in many diseases, such as laryngeal cancer and breast cancer. In this study, we studied the mechanism underlying the role of BMP2 in intervertebral disc degeneration (IDD). We used a luciferase assay system to determine the relationship between BMP2 and miR-129-5 expression. In addition, Western blot and real-time PCR were used to confirm the regulatory relationship between miR-129-5p and its targets, while flow cytometry was used to evaluate the effect of miR-129-5p on the apoptosis of neural progenitor cells (NPCs). BMP2 was confirmed as a direct target of miR-129-5p. Furthermore, the expression of miR-129 was downregulated along with upregulated BMP2 expression in IDD patients. Meanwhile, BMP2 was validated as the target of miR-129-5p in cells transfected with miR-129-5p and BMP2 siRNA. Also, compared with NPCs transfected with blank/scramble controls or miR-129-5p inhibitors, the NPCs treated with miR-129-5p mimics or BMP2 siRNA exhibited evidently elevated viability and inhibited apoptosis. Infection by human papillomavirus (HPV) can cause cervical intraepithelial neoplasia (CIN) and cancer. Down-regulation of E6 and E7 expression may be responsible for the positive clinical outcomes observed with IFN treatment, but the molecular basis has not been well determined. As miRNAs play an important role in HPV induced cervical carcinogenesis, we hypothesize that IFN-β can regulate the expressions of specific miRNAs in cervical cancer cells, and that these miRNAs can mediate E6 and E7 expression, thus modulate their oncogenic potential. In this study, we found that miR-129-5p to be a candidate IFN-β inducible miRNA. MiR-129-5p levels gradually decrease with the development of cervical intraepithelial lesions. Manipulation of miR-129-5p expression in Hela cells modulates HPV-18 E6 and E7 viral gene expression. Exogenous miR-129-5p inhibits cell proliferation in Hela cells, promotes apoptosis and blocks cell cycle progression in Hela cells. SP1 is a direct target of miR-129-5p in Hela cells. This study is the first report of a cellular miRNA with anti-HPV activity and provides new insights into regulatory mechanisms between the HPV and the IFN system in host cells at the miRNA level.  相似文献

Cervical cancer is a critically malignant tumor with the second mortality of females worldwide. MicroRNAs (miRNAs) are short but regulatory non-coding RNAs playing a pivotal role in many biological processes including tumorigenesis. However, the exact role of miR-140-3p in cervical cancer remains to be elucidated. Here we identified that miR-140-3p was significantly reduced in cervical cancer tissues by comprehensive analysis of TCGA data, hinting that higher expression level of miR-140-3p predicted a good clinical prognosis. Quantitative real-time PCR (RT-qPCR) assay was performed to confirm the negative correlation between miR-140-3p expression level and human cervical cancer tissues as well as various cervical cancer cell lines. To clarify the certain role of miR-140-3p, forced expression by microRNA mimics was applied in Caski and C33A cells, showing that miR-140-3p overexpression significantly impeded the proliferation of cervical cancer cells by cell count kit (CCK-8) assay. Western blot analysis of cell cycle-related proteins Cyclin A, Cyclin B1 and Cyclin D1 have further confirmed the cell cycle arrest was induced by the ectopic expression of miR-140-3p. Annexin-V based FACS analysis also found the simultaneous appearance of early apoptotic cell population in miR-140-3p overexpression cells. The protein level of BCL-2 was attenuated in accompany with elevated Bax and Cleaved caspase-3 protein, indicating miR-140-3p overexpression induced early apoptosis. Mechanistically, we demonstrated that miR-140-3p could target the 3′UTR of RRM2 which has been proved to be highly involved in the onset of cancer. Furthermore, upregulation of miR-140-3p and RRM2 failed to inhibit the proliferation of human cervical cancer cells, revealing that RRM2 served as the target downstream gene of miR-140-3p abolishing its ability as a tumor suppressor. Overall, we figured out the new role of miR-140-3p in cervical cancer and concluded that miR-140-3p was a candidate of cancer control in preclinical.  相似文献

Lung cancer, predominantly non-small cell lung cancer (NSCLC), remains the leading cause of cancer-related deaths worldwide. Although epidermal growth factor receptor (EGFR) signaling is important and well studied with respect to NSCLC progression, little is known about how miRNAs mediate EGFR signaling to modulate tumorigenesis. To identify miRNAs that target EGFR, we performed a bioinformatics analysis and found that miR-542-5p down-regulates EGFR mRNA and protein expression in human lung cancer cells (H3255, A549, Hcc827). We observed increases in EGFR association with Ago2 in miR-542-5p-transfected cells. Interestingly, we observed an inverse correlation of miR-542-5p expression and EGFR protein levels in human lung cancer tissue samples, suggesting that miR-542-5p directly targets EGFR mRNA. Furthermore, we found that miR-542-5p inhibited the growth of human lung cancer cells. Our findings suggest that miR-542-5p may act as an important modulator of EGFR-mediated oncogenesis, with potential applications as a novel therapeutic target in lung cancer.  相似文献

Growing evidence suggested that the long noncoding RNAs (lncRNAs) regulate several pathophysiological processes in tumorigenesis and may be new biomarkers for tumor therapy. In this study, we studied the expression and role of lncRNA MT1JP in the development of bladder cancer. We demonstrated that the expression of MT1JP was downregulated in bladder tumor samples and cell lines. Ectopic expression of MT1JP suppressed cell proliferation, cycle, and invasion in bladder cancer. In addition, our result suggested that miR-214-3p overexpression decreased the luciferase activity of wild-type MT1JP but not mutated-type MT1JP and elevated expression of MT1JP decreased miR-214-3p expression in the bladder cancer cell. Furthermore, we indicated that the expression of miR-214-3p was upregulated in bladder tumor samples and cell lines. Ectopic expression of miR-214-3p promoted cell proliferation, cycle, and invasion in bladder cancer. MT1JP suppressed cell proliferation, cycle, and invasion via negative modulation of miR-214-3p in bladder cancer. These data suggested that lncRNA MT1JP acts a tumor suppressor gene in bladder cancer progression, considering MT1JP as a new therapeutic target in bladder cancer.  相似文献
