The glutathione (GSH) status and heavy metal tolerance were investigated in four Paxillus involutus strains isolated from different heavy-metal-polluted and non-polluted regions of Europe. The heavy metal burden in the habitats did not affect significantly either the heavy metal (Cr2O72−, Cd2+, Hg2+, Pb2+, Zn2+, Cu2+) tolerance and accumulation or the GSH production of the strains tested. Exposures to heavy metals increased the intracellular GSH concentrations in 12 from 24 experimental arrangements (four strains exposed to six heavy metals) independently of the habitats of the strains. The importance of GSH in heavy metal tolerance (high MIC values, ability to accumulate heavy metals and to grow in the presence of heavy metals) was thus demonstrated in this ectomycorrhizal fungus.  相似文献

Industrial wastewaters contain various heavy metal components and therefore threaten aquatic bodies. Heavy metals can be adsorbed by living or non‐living biomass. Submerged aquatic plants can be used for the removal of heavy metals. This paper exhibits the comparison of the adsorption properties of two aquatic plants Myriophyllum spicatum and Ceratophyllum demersum for lead, zinc, and copper. The data obtained from batch studies conformed well to the Langmuir Model. Maximum adsorption capacities (qmax) were obtained for both plant species and each metal. The maximum adsorption capacities (qmax) achieved with M. spicatum were 10.37 mg/g for Cu2+, and 15.59 mg/g for Zn2+ as well as 46.49 mg/g for Pb2+ and with C. demersum they were 6.17 mg/g for Cu2+, 13.98 mg/g for Zn2+ and 44.8 mg/g for Pb2+. It was found that M. spicatum has a better adsorption capacity than C. demersum for each metal tested. Gibbs free energy and the specific surface area based on the qmax values were also determined for each metal.  相似文献

We examined the uptake of five heavy metals (Cu, Fe, Ni, Zn, and Mn) in Ceratophyllum demersum L. (hornwort) and Potamogeton alpinus Balb. (pondweed) from Iset’ river, Ural region, Russia. This study was conducted in a territory that is highly urbanized where the surface waters are contaminated by a wide spectrum of pollutants. The environmental situation in this territory drastically deteriorated due to anthropogenic activity. The water quality in most of the water bodies in the Ural region is rather poor. In a comparative study ofC. demersum and P. alpinus, differential accumulation pattern was noted for heavy metals (HMs). Higher amounts of HMs accumulated in C. demersum compared to P. alpinus. Also it was shown that in leaves of C. demersum there were high amount of total phosphorus, nitrogen, organics acids and ash; high activity of guaiacol peroxidase; high content of non-enzymatic antioxidants viz., flavonoids, ascorbate, glutathione and proline; high amount of thiols (soluble and membrane bound) compared to P. alpinus.  相似文献

Soil contaminated with heavy metals (e.g., Pb2+, Cu2+, and Zn2+) was treated with aminopoly-saccharide chitosan alone or a strain of the bacterium Bacillus subtilis to determine their effects on metal ion accumulations. Phosphatase and Chitosanase production were also assayed. The combined bacterial-chitosan treatment showed the greater accumulation of Zn2+ followed by Cu2+ as compared to single treatments, while Pb2+ did not show a marked accumulation by either single or combined treatments.  相似文献

In the early growth phase of Sesamum indicum cv. PB-1, the decrease in fresh and dry mass was higher with 1.0 mM Cd2+ than with the same level of Pb2+ and Cu2+. Recovery from the metal stress was considerable in the root fresh weight and almost completely in the root dry weight when 10.0 mM (1.9 EC), calcium chloride was supplied to the growing seedlings along with the metal salts in various combinations. Accumulation of Pb2+, Cd2+ and Cu2+ was differential to the metals and the plant parts when supplied without or with 10.0 mM calcium chloride. The order of endogenous metal accumulation was Cu2+Cd2+Pb2+ and roots accumulated more metal than the leaves in the absence, as well as in the presence, of calcium chloride. Calcium chloride could recover loss of in vivo NRA in roots caused by either of the metal combinations, whereas the salt could recover the loss in leaf NRA caused only by Pb2+Cd2+ (1.0 mM each). Response of root and leaf NRA was on the other hand, different when the enzyme was assayed directly using an in vitro assay method, and the salt accelerated the loss in enzyme activity drastically. The organic-N content of root and leaf was, however, increased significantly (p < 0.001) with calcium chloride alone and with the metals supplied in various combinations. Our data indicate that instead of a high endogenous accumulation of Cu2+, Cd2+ and Pb2+ in roots and leaves the metal toxicity is recovered to a great extent in the presence of 10.0 mM calcium chloride in the root environment regarding growth and nitrate reduction of the roots and leaves of young sesame seedlings.  相似文献

We selected higher aquatic plants (HAP) and microalgae possessing a high sorption capacity in respect to heavy metals to form a consortium designed to purify contaminated aquatic ecosystems. Accumulation of heavy metals Cd2+, Cu2+, Pb2+, and Zn2+ was investigated in plants Pistia stratiotes, Elodea canadensis, and Lemna minor and green microalgae Chlorella vulgaris ВВ-2, Ankistrodesmus sp. ВI-1, Chlamydomonas reinhardtii В-4, and Scеnеdеsmus quadricauda В-1. It was found that intense accumulation of metals occurs in cultures of HAP Pistia stratiotes and Elodea canadensis. These plants are macroconcentrators of zinc, lead, and copper and microconcentrators of cadmium. Out of the examined cultures of microalgae, effective bioaccumulators of heavy metals were C. vulgaris ВВ-2 and Ankistrodesmus sp. ВI-1. It was shown that heavy metals are selectively taken up from the medium in the series Zn2+ > Cu2+ > Cd2+ > Pb2+. In order to produce a consortium of higher aquatic plants and microalgae for purification of polluted aquatic ecosystems, we investigated interaction of HAP P. stratiotes and E. canadensis with microalgae C. vulgaris ВВ-2 and Ankistrodesmus sp. ВI-1 in the course of their cocultivation. Neutral relations were detected between the cells of microalgae C. vulgaris ВВ-2 and Ankistrodesmus sp. ВI-1 and HAP E. canadensis. At the same time, the cells of Ankistrodesmus sp. ВI-1 and HAP P. stratiotes formed a symbiosis. Microscopic examination showed numerous points where the cells of microalgae Ankistrodesmus sp. ВI-1 were attached to the roots of P. stratiotes plants. We tested an opportunity to employ the association between P. stratiotes and Ankistrodesmus sp. ВI-1 for purification of simulated wastewater polluted with heavy metal ions. This consortium proved to be capable of eliminating contaminants from the sewage, reducing their level in the sewage to standard values, and active accumulation of heavy metal ions.  相似文献

The aim of this study was to characterize the impact of heavy metals on phenotypical markers of Pseudomonas aeruginosa. Twenty-two isolates of P. aeruginosa, either clinical (20) or secondary treated wasterwater (2), were used to inoculate micro-ecosystems of sterile distilled water or secondary waste effluent in the presence of subminimal inhibitory concentrations of a variety of heavy metals commonly encountered in the aquatic naturally habitat (Ca2+, Co2+, Cr3+, Cu2+, Hg2+, Ni2+, Zn2+). Micro-ecosystems were exposed to visible light at laboratory temperature and individual strains were reisolated after a 1-, 3-, or 6-month period. The re-isolates (129) were characterized using hierarchical classification analysis in order to define affinities among variants of P. aeruginosa. Subsequently, discriminant analysis was used to detect eventual relationships among the different phenotypical markers studied. Results of the hierarchical classification, based on qualitative or quantitative approaches, showed clearly that incubation of P. aeruginosa in the presence of heavy metals altered the studied phenotypical markers, namely serotype, phage type, MIC of metals, and pyocin type. Discriminant analysis showed that the studied phenotypical markers could be classified into four clusters: C1 (L1 and L2 phage types, Hg tolerance and/or resistance, S2 serotype), C2 (P2 pyocin type, Cd tolerance and/or resistance, S1 serotype), C3 (Co and Cr tolerance and/or resistance) and C4 (P1 pyocin type, Ni, Zn, and Cu tolerance and/or resistance).  相似文献

Biosorption of Heavy Metals by Marine Algae   总被引:7,自引:0,他引:7  
The ability of four different algae (three brown and one red) that have not been previously studied to adsorb Cr3+, Co2+, Ni2+, Cu2+, and Cd2+ ions was investigated. The metal uptake was dependent on the type of biosorbent, with different accumulation affinities towards the tested elements. The HCl-treated biomass decreased the metal biosorptive capacity particularly in the case of Cr3 adsorption with Laurencia obtusa. The extent of uptake of the different metals with the tested algae was assessed under different conditions such as pH, time of algal residence in solution with the metal, and concentration of algal biomass. The rate of uptake of the different metals was very fast in the first 2 h; thereafter the increase in metal uptake was insignificant. The amount of the metal uptake (5–15 mg range) increased steeply by increasing the weight of the biomass. An exception was L. obtusa, where a parallel increase of the uptake of different metals was observed on increasing the algal mass from 5 to 50 mg. We studied the influence of a synthetic cytokinin-like growth regulator (Cytodef) and heavy metal ions—Pb2+, Sr2+, Zn2+, and Ni2+—on generation of superoxide anion (O2⊙−), lipid peroxidation, and carotenoids content in leaves of 7-day-old cucumber plants (Cucumis sativus L., cv. Izyashchnyi). In some instances Cytodef reduced the toxicity of heavy metals: it mitigated the negative effect of metals on oxidative processes and elevated the concentration of antioxidants (carotenoids).  相似文献

The heavy metal resistant ciliate, Stylonychia mytilus, isolated from industrial wastewater has been shown to be potential bioremediator of contaminated wastewater. The ciliate showed tolerance against Zn2+ (30 μg/mL), Hg2+ (16 μg/mL) and Ni2+ (16 μg/mL). The metal ions slowed down the growth of the ciliate as compared with the culture grown without metal stress. The reduction in cell population was 46% for Cd2+, 38% for Hg2+, 23% for Zn2+, 39% for Cu2+ and 51% for Ni2+ after 8 days of metal stress. S. mytilus reduced 91% of Cd2+, 90% of Hg2+ and 98% of Zn2+ from the medium after 96 h of incubation in a culture medium containing 10 μg/mL of the respective metal ions. Besides this, the ciliate could also remove 88% of Cu2+ and 73% Ni2+ from the medium containing 5 μg/mL of each metal after 96 h. The ability of Stylonychia to take up variety of heavy metals from the medium could be exploited for metal detoxification and environmental clean-up operations.  相似文献

Yeast cells are capable of accumulation of various heavy metals, preferentially accumulating those of potential toxicity and also those of value. They retain their ability to accumulate heavy metals under a wide range of ambient conditions. In the present study it was shown that yeast cells in suspension accumulate heavy metal cations such as Cu2+, Co2+. The level of copper accumulation was dependent on the ambient metal concentration and was markedly inhibited by extremes of ambient pH. Temperature (5–40°C) and the presence of the alkali metal sodium had much smaller effects on the level of copper accumulation. This suggests that in waste-waters of pH 5.0–9.0, yeast biomass could provide an effective bioaccumlator for removal and/or recovery of the metal. During bioaccumulation and subsequent processes it is necessary to retain the biomass. It was shown in the present study that this could be achieved by cell immobilization. Immobilization allowed for complete removal of Cu2+, Co2+, and Cd2+ from synthetic metal solutions. The immobilized material could be freed of metals by use of the chelating agent ethylenediamine tetraacetic acid (EDTA) and recycled for further bioaccumulation events with little loss of accumulation capacity.Correspondence to: J. The influence of arsenic (As) on the growth and the antioxidant system of Lemna gibba L. exposed to five concentrations of As (0.0, 0.25, 0.5, 1.0, and 1.5 mg L?1) was studied. Although As exposure reduced relative growth rate, L. gibba continued to grow even after the high accumulation of this element after five days of exposure. The concentration of the superoxide anion was unaffected by As, whereas an increase of hydrogen peroxide concentrations was observed with the increasing of As concentration. Increasing concentrations of As also increased the enzyme activity of superoxide dismutase, peroxidase, and glutathione reductase and the total and nonprotein thiols, up to 0.5 mg L?1, whereas the anthocyanin content increased constantly with As concentration. Catalase and ascorbate peroxidase activities as well as the content of chloroplastic pigments were reduced in plants exposed to all As concentrations. These results support a major role of anthocyanins, nonprotein thiols, and antioxidant scavenging enzymes in L. gibba tolerance to toxic As concentrations.  相似文献

Trace metals always act as cofactors or coenzymes in many cellular processes. Deficiency or excess of some metals will affect the fermentation of lignocellulosic hydrolysate. In order to make sure the deficient or excessive states of metals in culture medium, metal contents analysis was conducted in Pichia stipitis ATCC 58784 cells, synthetic medium, and diluted acid hydrolysate of rice straw. The results showed that Cu, Ni, and Co were deficient, and Al was a little excessive. So the influences of Cu2+, Al3+, Ni2+, and Co2+ additions on the growth and ethanol production of ATCC 58784 were further researched. Low concentration additions of Cu2+ and Al3+ (<0.24 mM and <0.23 mM, respectively) improved biomass growth of ATCC 58784 by 34 and 13%, respectively; however, higher concentrations decreased biomass growth. On the other hand, addition of Cu2+ (0.39 mM) did not affect volumetric ethanol production significantly (P = 0.05) and addition of Al3+ (0.38 mM) showed no influence on volumetric ethanol production (P = 0.68). Addition of 0.074 mM Co2+ inhibited biomass growth of ATCC 58784 by 13% and volumetric ethanol production by 10%. The biomass growth and volumetric ethanol production of ATCC 58784 was arrested by the addition of 0.33 mM of Ni2+ by 53 and 65%, respectively.  相似文献

Summary The bioaccumulation of metals (Cu2+, Cr6+, Cd2+, Ni2+ and Zn2+) from three electroplating effluents by viable Saccharomyces cerevisiae, and the effect of glucose treatment on accumulation was determined. Pretreatment of the yeast cells with glucose increased the amount of metal removed, whilst direct addition of glucose to the yeast-effluent solution had no effect on the amount of metal accumulated.  相似文献

In the present study, the effect of copper (Cu2+) and lead (Pb2+) ions on the growth and lipid composition of various parts of the fern, Matteuccia sthruthiopteris, was examined. Plants were incubated in the presence or absence of 1, 10, 100 μM of Cu(NO3)2 or Pb(NO3)2. Cu2+ and Pb2+ ions at concentrations of 1 and 10 μM caused an increased growth of the roots and leaves. A higher concentration of Pb2+ did not show any effect on growth, whereas that of Cu2+ slowed down the growth of the whole plants. The roots accumulated more than 700 μg of Cu2+ and 400 μg of Pb2+ per 1 g dry weight when the plants were incubated with the higher concentrations of metals, whereas in the leaves the concentration of Cu2+ was much lower and did not exceed 12 μg/g dry weight. No accumulation of Pb2+ ions by leaves was detected. The lipid composition of photosynthetic leave tissues was shown to be affected by the presence of metal ions in the root medium at either concentration studied. Various changes in lipid classes were noted as responsive reactions of M. sthruthiopteris to the heavy metal ions in nutrient medium. Cu2+ ions decreased the content of total lipids, total phospholipids, and individual phosphatidylcholines and phosphatidylethanolamines, whereas Pb2+ ions caused a decrease in the content of total lipids and glycolipids. Changes in the lipid composition were more pronounced in the mature leaves than in the scrolls of the studied fern.  相似文献

Acinetobacter johnsonii has potential use in the remediation of heavy metal-contaminated wastewaters. For metal accumulation, cells must remain intact and metabolically active. The effect of possible accumulation targets, Cd2+, UO2 2+, Cu2+, and Ni2+ on cytoplasmic membrane integrity and polarity was investigated by flow cytometry using a mixture of two fluorescent dyes, propidium iodide and bis-oxonol. The former binds to DNA but cannot cross an intact cytoplasmic membrane, whilst the latter is anionic, lipophilic and stains depolarized cytoplasmic membranes. All four metals permeabilized the cytoplasmic membrane of some cells during the period of exposure. The effects of the metals differed in that Cu2+ and Cd2+ also generated an intermediate population of cells, having intact but depolarized cytoplasmic membranes. Hydrogenase-constitutive (Hupc) mutants of Bradyrhizobium japonicum were previously shown to accumulate more nickel than the wild-type strain. In a 2 h period Hupc strains JH101 and JH103 also accumulated 2- to 3-fold more Mg2+, Zn2+ and Cu2+, and about 4-fold more Co2+ and Mn2+ than the wild-type strain JH. Init uptake rates (first 10 min) by the Hupc strains were also greater for all the metals. The mutation in the Hupc strains affecting a trans-acting regulator of the hup structural genes appears to have also amplified a metal uptake/accumulation process common to many divalent metal ions. From efflux experiments (suspension of cells in metal-free medium after metal accumulation) to determine the degree of dissociation of each metal with the cells it was concluded that Zn2+, like Ni2+, was rapidly and tightly cell-associated. In contrast, about 50% of the accumulated Cu2+ and about 30% of the Mn2+ was effluxed within 2 h by both the Hupc and wild-type strains. Cobalt was more tightly cell-associated than Mn2+ or Cu2+, as the strains effluxed about 26% of the previously accumulated metal in 2 h. Even after accounting for effluxed metal, the Hupc strains retained more of each metal than the wild-type. The increased metal accumulation by Hupc strains could not be accounted for solely at the level of transport, as known metabolic inhibitors (carbonyl cyanide m-chlorophenylhydrazone and nigericin) of nickel transport partially inhibited (1 h) accumulation of only some (magnesium, zinc and copper) of the other metals. Hydrogenase-derepressed wild-type cells exhibited slightly higher (22–27% more) 2 h accumulation capacity for some of the metals (nickel, zinc and copper) than did non-derepressed cells, but not to the 2- to 4-fold greater level observed for Hupc strains compared with the wild-type. The Hupc strains JH101 and JH103 do not synthesize more capsular/cell wall carbohydrate than the wild-type strain.  相似文献

One key step of the bioremediation processes designed to clean up heavy metal contaminated environments is growing resistant cells that accumulate the heavy metals to ensure better removal through a combination of biosorption and continuous metabolic uptake after physical adsorption. Saccharomyces cerevisiae cells can easily act as cation biosorbents, but isolation of mutants that are both hyperaccumulating and tolerant to heavy metals proved extremely difficult. Instead, mutants that are hypersensitive to heavy metals due to increased and continuous uptake from the environment were considered, aiming to use such mutants to reduce the heavy metal content of contaminated waters. In this study, the heavy metal hypersensitive yeast strain pmr1∆ was investigated for the ability to remove Mn2+, Cu2+, Co2+, or Cd2+ from synthetic effluents. Due to increased metal accumulation, the mutant strain was more efficient than the wild-type in removing Mn2+, Cu2+, or Co2+ from synthetic effluents containing 1–2 mM cations, with a selectivity $ {\text{Mn}}^{{{\text{2}} + }} > {\text{Co}}^{{{\text{2}} + }} ~ > {\text{Cu}}^{{{\text{2}} + }} $ {\text{Mn}}^{{{\text{2}} + }} > {\text{Co}}^{{{\text{2}} + }} ~ > {\text{Cu}}^{{{\text{2}} + }} and also in removing Mn2+ and Cd2+ from synthetic effluents containing 20–50 μM cations, with a selectivity Mn2+ > Cd2+.  相似文献

Qualitative and quantitative composition of lipids was investigated in fresh-water vascular plant Hydrilla verticillata (L. fil.) Royle in the course of the accumulation and elimination of heavy metals (HM). The plants were incubated in 100μM solutions of metal nitrates for 10 days. The accumulation of Cu2+, Zn2+, and Pb2+ and their elimination from the plants depended on the duration of exposure and chemical nature of the metal. Accumulation of lead and copper salts was the greatest on the 3rd day, and zinc, on the 10th day. It was associated with changes in the composition of total lipids, polar lipids, and fatty acid (FA). Copper ions suppressed lipid metabolism stronger than other metals. Zn2+ and Pb2+ induced the accumulation of biomass and elevated the content of some phospholipids and glycolipids. The detected changes (decrease or increase) were observed both during the incubation with HM and within an afterstress period when the plants recovered in the medium free of metals. Judging by their effect on the content of lipids and FA, HM form a series: Cu2+ > Zn2+ > Pb2+. The responses of plant lipid metabolism to the metals of various chemical nature are discussed.  相似文献
