The aqueous, ethyl acetate, methanolic and Total Oligomer Flavonoids (TOF) enriched extracts, obtained from the aerial parts of Cyperus rotundus, were investigated for their contents in phenolic compounds. Antioxidative activity using the NBT/riboflavin assay system, antimicrobial activity against Gram positive and Gram negative bacterial reference strains as well as antigenotoxic activity tested with the SOS chromotest assay were also studied. Significant antibacterial activity against reference strains; Staphylococcus aureus, Enterococcus faecalis, Salmonella enteritidis and Salmonella typhimurium, was detected in the presence of ethyl acetate and TOF enriched extracts. In addition to their antimicrobial activity, the same extracts showed a significant ability to inhibit nitroblue tetrazolium reduction by the superoxide radical in a non enzymatic O2.− generating system, and were also able to reduce significantly the genotoxicity induced by nifuroxazide and Aflatoxin B1. The antioxidant, antimicrobial and antigenotoxic activities exhibited by C. rotundus depend on the chemical composition of the tested extracts.  相似文献

The essential oils (EOs) obtained from the leaves of Iryanthera polyneura Ducke trees was chemically Assessed and tested for the ability of inhibiting the growth of Candida albicans, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Streptococcus mutans and S. sanguinis. The oil was also tested against breast (MCF‐7) and prostate (PC‐3) cancer cell lines. Minimum bactericidal concentrations (MBCs) and 50 % inhibition concentrations (IC50) values were obtained. EOs were active against Gram‐positive bacteria. Spathulenol, α‐cadinol and τ‐muurolol were major components of EOs. The oils showed a higher cytotoxicity against PC‐3 than MCF‐7 cells, although the oils were active against both cell types. Oils obtained from leaves collected in the dry season were more active against E. faecalis, S. aureus and PC‐3, while the oils obtained from leaves collected in the rainy season were more active against S. mutans, S. sanguinis and MCF‐7. The antibacterial and cytotoxic activities of the essential oils from the leaves of I. polyneura are related to the seasonal climate variation and are influenced by compounds that are minor components of the oils.  相似文献

Infectious diseases caused by bacteria and fungi are the major cause of morbidity and mortality across the globe. Multi-drug resistance in these pathogens augments the complexity and severity of the diseases. Various studies have shown the role of biofilms in multi-drug resistance, where the pathogen resides inside a protective coat made of extracellular polymeric substances. Since biofilms directly influence the virulence and pathogenicity of a pathogen, it is optimal to employ a strategy that effectively inhibits the formation of biofilm. Pomegranate is a common food and is also used traditionally to treat various ailments. This study assessed the anti-biofilm activity of a methanolic extract of pomegranate against bacterial and fungal pathogens. Methanolic extract of pomegranate was shown to inhibit the formation of biofilms by Staphylococcus aureus, methicillin resistant S. aureus, Escherichia coli, and Candida albicans. Apart from inhibiting the formation of biofilm, pomegranate extract disrupted pre-formed biofilms and inhibited germ tube formation, a virulence trait, in C. albicans. Characterization of the methanolic extract of pomegranate revealed the presence of ellagic acid (2,3,7,8-tetrahydroxy-chromeno[5,4,3-cde]chromene-5,10-dione) as the major component. Ellagic acid is a bioactive tannin known for its antioxidant, anticancer, and anti-inflammatory properties. Further studies revealed the ability of ellagic acid to inhibit the growth of all species in suspension at higher concentrations (>75?μg?ml?1) and biofilm formation at lower concentrations (<40?μg?ml?1) which warrants further investigation of the potential of ellagic acid or peel powders of pomegranate for the treatment of human ailments.  相似文献

Acacia catechu, commonly known as catechu, cachou and black cutch is an important medicinal plant and an economically important forest tree. The methanolic extract of this plant was found to have antimicrobial activities against six species of pathogenic and non-pathogenic microorganisms: Bacillus subtilis, Staphylococcus aureus, Salmonella typhi, Escherichia coli, Pseudomonas aeruginosa and Candida albicans. The maximum zone of inhibition (20 mm) was found to be exhibited against S. aureus. For this organism the minimum bactericidal concentration (MBC) of the crude extract was 1,000 μg/ml. The extract was found to be equally effective against gram positive and gram negative bacteria. The antimicrobial activity of the extract was found to be decreased during purification. The chemical constituents of organic plant extracts were separated by thin layer chromatography (TLC) and the plant extracts were purified by column chromatography and were further identified by Gas chromatography–mass selection (GC–MS) analysis. The composition of A. catechu extract had shown major components of terpene i.e. camphor (76.40%) and phytol (27.56%) along with other terpenes in minor amounts which are related with their high antibacterial and antifungal properties.  相似文献

A library of 175 dipeptidomimetics and tripeptidomimetics containing an α‐amino boronic acid or boronate has been synthesized, and the activity toward Mycobacterium tuberculosis, Candida albicans, Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli and Pseudomonas aeruginosa has been screened. Although there is no clear structure–activity relationship, several compounds exhibit promising activity against different pathogens. 为研究23种中草药的80%乙醇提取物对4种临床常见致病菌的体外抗菌活性,该研究用琼脂扩散法测定抑菌圈直径,微量肉汤培养基倍比稀释法测定最低抑菌浓度(minimum inhibitory concentration,MIC)和最低杀菌浓度(minimum bactericidal/fungicidal concentration,MBC/MFC)。结果表明:滇龙胆草、金丝梅、溪黄草等16种提取物对金黄色葡萄球菌的MIC/MBC值在0.19~3.12 mg·mL-1之间,有很强的抑菌活性。头花蓼、淡竹叶、半枝莲等14种提取物对铜绿假单胞菌的MIC/MBC值在1.56~6.25 mg·mL-1之间,有较强的抑菌活性。除槐角外,其余提取物对大肠埃希菌的MIC/MBC值均在3.12~12.5 mg·mL-1之间,有较强的抑菌活性。黄藤、藿香提取物对白色念珠菌的MIC/MFC值在0.78~6.25 mg·mL-1之间,有较强的抑菌活性; 滇龙胆草、金丝梅、水杨梅、苦参、胡椒、赶黄草、荜菝、淡竹叶提取物对白色念珠菌的MIC/MFC值在6.25~12.5 mg·mL-1之间,也具有一定抑菌活性。因此,所选中草药的抑菌效果均较好,大部分均具有广谱抗菌活性。其中,藿香、黄藤的提取物对白色念珠菌抑菌活性较强,金丝梅、水杨梅、仙鹤草、苦参、赶黄草、溪黄草的提取物对金黄色葡萄球菌抑菌活性很强,这几种中草药可为进一步追踪其活性单体化合物和作用机制提供一定的参考。  相似文献

BackgroundTerfezia claveryi truffles are known for their nutritional value and have been considered among traditional treatments for ophthalmic infections and ailments.ObjectivesWe sought to investigate the in vitro antimicrobial efficacy of several T. claveryi extracts from Saudi Arabia. Certain pathogenic fungi and gram-negative and gram-positive bacteria were included.MethodsDry extracts were prepared using methanol, ethyl acetate, and distilled water, while the latter was used for preparing fresh extracts. The extracts were microbiologically evaluated through the disc-diffusion agar method; the zones of inhibition of microbial growth were measured post-incubation. The minimum bactericidal concentration (MBC) and minimum inhibitory concentration (MIC) were determined in Müller-Hinton Broth through the microdilution susceptibility method. anti-biofilm activity was assessed for potent extracts.ResultsDry extracts showed potent activity (>16-mm inhibition zones) against gram-positive (Bacillus subtilis IFO3007 and Staphylococcus aureus IFO3060) and gram-negative (Pseudomonas aeruginosa IFO3448 and Escherichia coli IFO3301) bacteria. The activity against fungi was moderate (12–16-mm inhibition zones) for both Aspergillus oryzae IFO4177 and Candida albicans IFO0583; there was no activity against Aspergillus niger IFO4414 growth. Methanolic extract had the lowest MIC and MBC, exhibiting remarkable activity against B. subtilis growth. Fresh extract showed moderate activity against bacterial growth and inactivity against fungal growth. Methanolic extract showed potent anti-biofilm activity (IC50, 2.0 ± 0.18 mg/mL) against S. aureus.ConclusionsT. claveryi extracts showed antibacterial effects potentially suitable for clinical application, which warrants further in-depth analysis of their individual isolated compounds.  相似文献

Kephir is a fermented carbonated refreshing milk, with a slightly acidic aromatic taste and creamy foam composition which contains lactobacilli, leuconostocci, acetic acid bacteria, lactostreptococci and yeasts. Recent studies have demonstrated its antibacterial, immunostimulating, antitumoral and cholesterol-lowering activities.

Purpose

The purpose of this study was to investigate the antimicrobial activity of kephir against Bacillus subtilis spp. spizizenii ATCC 6633, Staphylococcus aureus ATCC 6538, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 8739, Salmonella enteritidis ATCC 13076, Pseudomonas aeruginosa ATCC 9027 and Candida albicans ATCC 10231. The kephir fermented for 24 h and 48 h, as well and after 7 days preservation at 4–8 °C was tested by in vitro disk diffusion method. The intensity of the antimicrobial activity was interpreted by comparison with two antibiotics, i.e. ampicillin and neomycin.

Results

The antimicrobial activity of 24 h as well as 48 fermented kephir, fresh or after 7 days preservation at 4–8 °C was similar and observed against B. subtilis, S. aureus, E. coli, E. faecalis and S. enteritidis. For E. coli, E. faecalis and S. enteritidis the antimicrobial activity was superior to both tested antibiotics and for B. subtilis and S. aureus to one antibiotic. The tested products exhibited no activity against P. aeruginosa and C. albicans.

Conclusion

Kephir is exhibiting large spectrum and strong antibacterial activity probably due to the complex viable probiotic strains association producing antimicrobial substances.  相似文献

Antimicrobial peptides and their analogues have become substitutes for antibiotics in recent years. The antimicrobial peptide analogue SAMP-A4-C8 (n-octanoic-VRLLRRRI) with high antimicrobial activity was found in our lab. We speculate that it may kill pathogens by some lethal mechanism of action. In the present investigation, the microbicidal activities of SAMP-A4-C8 and its mechanism of action were investigated. The results demonstrated that SAMP-A4-C8 had lethal activities against Staphylococcus aureus and Candida albicans by cell disruption. Based on its microbicidal activities, we believe that it is worth further research for its potential as drug candidate. The results showed that SAMP-A4-C8, with low propensity to induce the resistance of S. aureus and C. albicans, could kill the persister cells of S. aureus and C. albicans, exhibited biofilm forming inhibition activity and preformed biofilm eradication ability against S. aureus and C. albicans, and displayed therapeutic potential on pneumonia in S. aureus-infected mice by reducing lung inflammation. The present study provided a promising drug candidate in the war against multidrug resistance.  相似文献

Aims: To investigate the potential activation of hydrogen peroxide by a novel catalyst, reducing the concentration of hydrogen peroxide required and the time taken for microbial inactivation. Methods and Results: The antimicrobial properties of an iron‐based novel heterogeneous polyacrylonitrile catalyst in combination with hydrogen peroxide were examined against Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus using a modified version of the European suspension test. Antimicrobial activity against Bacillus cereus and Bacillus subtilis endospores was also investigated. Bactericidal activity was significantly increased when the polyacrylonitrile catalyst was combined with hydrogen peroxide. 0·2, 0·5 and 1% w/v hydrogen peroxide resulted in average log reductions of 4·76, 5·59 and 5·37 for E. coli, Ps. aeruginosa and Staph. aureus, respectively, after 60 min exposure at room temperature. The catalyst also significantly increased the activity of hydrogen peroxide against B. subtilis and B. cereus endospores. Conclusions: These studies have demonstrated the potential biocidal use of the novel polyacrylonitrile catalyst when combined with hydrogen peroxide. Significance and Impact of the Study: This is the first publication to demonstrate the enhanced activity gained using the novel heterogeneous catalyst to potentiate the activity of hydrogen peroxide as a biocide.  相似文献

Mangroves are abundant in bioactive natural substances that fight off pathogenic diseases. Different parts of R. apiculata, an abundant mangrove found in Bhitarkanika National Park, India were extracted with methanol and a mixture of solvents methanol/ethanol/chloroform (60 : 20 : 20) to evaluate their antimicrobial properties. The combination solvent extract of bark had the highest zone of inhibition (ZOI) of 18.62 mm against Pseudomonas aeruginosa and a ZOI of 17.41 mm against Streptococcus mitis. Bark extracts had the highest DPPH (43 %) and FRAP (96 %) activities. The combination solvent bark extract of R. apiculata had the highest ZOI of 20.42 mm (lowest MIC of 2.12 μg/ml) against Candida albicans and ZOI of 15.33 mm (MIC of 3.02 μg/mL) against Penicillium chrysogenum. Combination bark extracts of R. apiculata contained flavanols than methanolic extracts. The crude extract of R. apiculata bark made with a mixture of solvents containing more active ingredients could be used in novel drug formulation.  相似文献

A peptide antibiotic, gramicidin A, was covalently bound to cystamine self-assembled monolayers on gold surfaces. Each step of the surface functionalization was characterized by polarization modulation infrared reflection absorption spectroscopy and X-ray photoelectron spectroscopy. The antimicrobial activity of the anchored gramicidin was tested against three Gram-positive bacteria (Listeria ivanovii, Enterococcus faecalis, and Staphylococcus aureus), the Gram-negative bacterium Escherichia coli and the yeast Candida albicans. The results revealed that the adsorbed gramicidin reduced, from 60% for E. coli to 90% for C. albicans, the number of culturable microorganisms attached to the surface. The activity was proven to be persistent overtime, up to 6 months after the first use. The bacteria attached to the functionalized surfaces were permeabilized as shown by confocal microscopy. Taken together, these results indicate a bacteriostatic mode of action of the immobilized peptide. Finally, using green fluorescent protein-expressing bacteria, it was shown that the development of a bacterial biofilm was delayed on peptide-grafted surfaces for at least 24 h.  相似文献

Three sesquiterpene lactones (centaurepensin=chlorohyssopifolin A, chlorojanerin and 13-acetyl solstitialin A) isolated from the aerial parts of Centaurea solstitialis L. ssp. solstitialis (Asteraceae) were investigated for antimicrobial and antiviral activities. For the antimicrobial activity assessment, both standard and isolated strains of Escherichia coli, Pseudomonas aeruginosa, Enterococcus faecalis, Staphylococcus aureus, Candida albicans and C. parapsilosis were employed by the microdilution method. Herpes simplex type-1, a DNA virus, and Parainfluenza, an RNA virus, were employed for the determination of the antiviral activity of these three sesquiterpene lactones using Vero cell lines. Ampicilline, ofloxocine, ketoconazole, fluconazole, acyclovir and oseltamivir were used as the reference drugs. 13-Acetyl solstitialin A displayed remarkable antibacterial activity against isolated strains of E. faecalis at 1 μg/ml concentration, which was close to the effective concentrations of ampicillin. The same compound also showed significant activity against the DNA virus, being as potent as the reference compound acyclovir at maximum and minimum concentrations of 16–<0.00006 μg/ml. This is the first report showing that 13-acetyl solstitialin A possesses significant antiviral activity.  相似文献

The present study reports for the first time, the in vivo wound healing potential of Punica granatum L. peels. A 5% (w/w) methanolic extract based-ointment was formulated and evaluated for its wound healing in guinea pigs. The ointment was applied in vivo on the paravertebral area of twelve excised wounded models once a day for 10 consecutive days. The ointment significantly enhanced the wound contraction and the period of epithelialization as assessed by the mechanical (contraction rate, tensile strength), the biochemical (increasing of collagen, DNA and proteins synthesis) and the histopathological characteristics. Such investigation was encouraged by the efficiency of the methanolic extract as antimicrobial and antioxidant. Indeed, the extract showed antioxidant activity as strong as natural and synthetic compounds (Trolox, BHA, Quercetin). Furthermore, the extract exhibited significant antibacterial and antifungal activity against almost all tested bacteria: Pseudomonas aeruginosa ATCC 9027, Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922, Klebsiella pneumoniae, Salmonella anatum, Salmonella typhimurium, Streptococcus pneumoniae, and fungi Candida albicans, Candida glabrata, Trichopyton rubrum and Aspergillus niger. The formulated ointment might well find use as skin repair agent without hazard to human health based on these results and on the fact that it has been well established that the extracts of pomegranate used in conditions similar to those applied by traditional medicine, showed no toxic effects.  相似文献

An attempt was made to control waterborne pathogens by using medicinal plant extracts. One hundred and twenty-six water samples from filtration plants, tube wells, and water supplies were collected and analyzed for total and faecal Coliform bacteria as well as for total viable count. Results showed that waterborne pathogens were numerous and significantly higher than the World Health Organization's recommended guidelines. The methanolic and aqueous extracts of different parts of Colebrookia oppositifolia (Labiateae) were examined for antibacterial activities in vitro by an agar diffusion method. Antibacterial activity of leaves, shoots, and roots of Colebrookia oppositifolia was assessed against Gram positive and Gram negative bacteria that were isolated and identified from water samples by the API 20E method. Extract of roots showed more antibacterial activities against Staph. aureus and B. cereus var. mycoides, Pseudomonas aeruginosa, Klebsiella pneumonia, and Shigella flexneri at 37°C, than extracts from leaves and shoots. The lowest MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) were observed in roots as compared to shoots and leaves. These results suggest that there is an urgent need for improvement in existing water quality treatment. Secondly, the fruit extract can be practical for protection and to avoid risk of contamination by waterborne pathogens and to promote indigenous solutions for disease-control and environmental management.  相似文献

Purpose of the present study was to evaluate antioxidant, antibacterial, antifungal, and antiviral activities of the petroleum ether, chloroform, ethyl acetate and methanol extracts as well as the alkaloid fraction of Lycopodium clavatum L. (LC) from Lycopodiaceae growing in Turkey. Antioxidant activity of the LC extracts was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging method at 0.2 mg/ml using microplate-reader assay. Antiviral assessment of LC extracts was evaluated towards the DNA virus Herpes simplex (HSV) and the RNA virus Parainfluenza (PI-3) using Madin-Darby Bovine Kidney (MDBK) and Vero cell lines. Antibacterial and antifungal activities of the extracts were tested against standard and isolated strains of the following bacteria; Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, Acinobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis as well as the fungi; Candida albicans and C. parapsilosis. All of the extracts possessed noteworthy activity against ATCC strain of S. aureus (4 μg/ml), while the LC extracts showed reasonable antifungal effect. On the other hand, we found that only the chloroform extract was active against HSV (16–8 μg/ml), while petroleum ether and alkaloid extracts inhibited potently PI-3 (16–4 μg/ml and 32–4 μg/ml, respectively). However, all of the extracts had insignificant antiradical effect on DPPH. In addition, we also analyzed the content of the alkaloid fraction of the plant by capillary gas chromatography-mass spectrometry (GC-MS) and identified lycopodine as the major alkaloid.  相似文献

Aspergillus fumigatus causes severe problems in poultry production systems. Seven South African tree species were selected from the database of the Phytomedicine Programme based on its antifungal activity against the fungus Cryptococcus neoformans. The acetone leaf extracts of the selected species had minimum inhibitory concentrations (MICs) of 0.16 mg/ml and lower in the preliminary screening. The antibacterial and antifungal activities of hexane, dichloromethane, acetone and methanol extracts of the leaves were determined using a two-fold serial microdilution method against a range of commonly encountered animal pathogenic fungi (A. fumigatus, Candida albicans, C. neoformans, Microsporum canis and Sporothrix schenckii) and four nosocomial bacteria (Staphylococcus aureus, Enterococcus faecalis, Escherichia coli and Pseudomonas aeruginosa). The plant species investigated were Combretum vendae (A.E. van Wyk) (Combretaceae), Commiphora harveyi (Engl.) Engl. (Burseraceae), Khaya anthotheca (Welm.) C.DC (Meliaceae), Kirkia wilmsii Engl. (Kirkiaceae), Loxostylis alata A. Spreng. ex Rchb. (Anacardiaceae), Ochna natalitia (Meisn.) Walp. (Ochnaceae) and Protorhus longifolia (Bernh.) Engl. (Anacardiaceae). All the extracts had activity against at least one of the test organisms over an incubation period of 24 or 48 h. The MIC values of the non-polar and intermediate polarity extracts of O. natalitia, K. anthotheca, C. vendae, C. harveyi, and P. longifolia had MICs as low as 0.08 mg/ml against at least one of the tested bacteria. Furthermore, the acetone extracts of L. alata, K. wilmsii, O. natalitia and C. vendae had antifungal activities with MIC values ranging from 0.04 to 0.08 mg/ml against at least one of the tested fungi. The average MIC values of the plant extracts against the different bacteria ranged from 0.17 to 2.11 mg/ml, while the range was 0.23–1.98 mg/ml for fungi. The Gram-positive bacteria (S. aureus and E. faecalis) were more susceptible to the plant extracts than the Gram-negative bacteria (E. coli and P. aeruginosa). E. faecalis was the most susceptible microbe and C. vendae extracts were the most active against nearly all the bacteria tested. The acetone extract of L. alata was the most active against fungal pathogens, with activity against at least 3 fungal organisms. L. alata was selected for further work to isolate compounds active against A. fumigatus and other fungal pathogens.  相似文献

Plant defensins are antimicrobial peptides that exhibit mainly antifungal activity against a broad range of plant fungal pathogens. However, their actions against Candida albicans have not been extensively studied. The mRNA for γ-thionin, a defensin from Capsicum chinense, has been expressed in bovine endothelial cells. The conditioned medium of these cells showed antifungal activity on germ tube formation (60–70% of inhibition) and on the viability of C. albicans (70–80% of inhibition). Additionally, C. albicans was not able to penetrate transfected cells. Conditioned medium from these cells also inhibited the viability (80%) of the human tumor cell line, HeLa.  相似文献

The discovery and optimisation of a new class of benzothiazole small molecules that inhibit bacterial DNA gyrase and topoisomerase IV are described. Antibacterial properties have been demonstrated by activity against DNA gyrase ATPase and potent activity against Staphylococcus aureus, Enterococcus faecalis, Streptococcus pyogenes and Haemophilus influenzae. Further refinements to the scaffold designed to enhance drug-likeness included analogues bearing an α-substituent to the carboxylic acid group, resulting in excellent solubility and favourable pharmacokinetic properties.  相似文献
