This study investigated the variability of amino acid (AA) digestibility of rye grains in laying hens. Relationships between AA digestibility and physical properties (thousand seed weight, test weight, falling number, and extract viscoelasticity), chemical composition (proximate nutrients, non-starch polysaccharides, AA, minerals, and inositol phosphates), gross energy concentration, and in vitro solubility of nitrogen (N) of the grains were also examined. Twenty rye genotypes were grown under standardised agronomic and environmental conditions as part of a collaborative research project known as “GrainUp”. Each genotype was added to a basal diet at 500 g/kg at the expense of maize starch to produce 20 rye diets. The experimental design comprised four Latin Squares (6 × 6) distributed over two runs, resulting in 12 experimental periods. Caecectomised laying hens (LSL-Classic) were individually kept in metabolism cages. Excreta were collected quantitatively for 4 d, and AA digestibility of the rye genotypes was determined using a regression approach. The digestibility of AA was generally low but varied significantly among the 20 rye genotypes, especially for Lys (digestibility range 35–59%), Met (57–75%), Thr (34–54%), and Trp (36–71%). Nevertheless, physical and chemical characteristics as well as the in vitro solubility of N correlated in only a few cases with AA digestibility. Multiple linear regression was used to calculate equations to predict AA digestibility based on the analysed characteristics. However, their explanatory power, as judged by the adjusted R2, was not sufficiently precise for practical application (below 0.6 for most AA). In conclusion, the AA digestibility of rye grain is generally low and varies significantly between crop genotypes. Equations based on its physical and chemical characteristics are not sufficiently precise to be useful for feed formulation.  相似文献

Fat coating of soybean meal (SBM) can reduce its protein degradability in the rumen, but the encapsulation of SBM with palmitic (PA) and stearic acids (SA) has not yet been investigated, despite both fatty acids are common energy sources in dairy cow diets. This study aimed to evaluate the effects of applying a novel method, using either 400 or 500 g fat/kg (treatments FL40 and FL50, respectively), which was enriched in PA and SA at different ratios (100:0, 75:25, 50:50, 25:75 and 0:100), on physical and chemical characteristics, ruminal degradability, solubility and in vitro intestinal protein digestibility (IVIPD) of the obtained products. Encapsulation of SBM in fat resulted in greater mean particle size and lower bulk density and protein solubility than unprotected SBM (USBM). Treatment FL50 resulted in increased (p < 0.01) rumen-undegraded protein (RUP) compared to USBM. There were no differences in RUP of SBM when different PA: SA ratios were used. The mean RUP content of treatments FL40 and FL50 (306 and 349 g/kg, respectively) was greater compared to USBM (262 g/kg, p < 0.05), but lower than that for a standard heat-treated SBM (431 g/kg). Values of IVIPD did not differ among SBM, heat-treated SBM and FL40 and FL50 samples, all being greater than 97.8%. In conclusion, encapsulation of SBM with fats enriched in PA and SA proved to be effective in reducing protein solubility and increasing RUP without depressing protein digestibility in the intestine. For validation of the method, in vivo research to investigate the effects of these products on the production of dairy cows is warranted.  相似文献

This study investigated the effects of food quality on digestive enzyme activities, in vitro protein digestibility and histological traits of the midgut gland in juvenile crayfish Cherax quadricarinatus. Animals of a wide weight range were fed different diets: two commercial diets with high or low lipid content (high lipid and low lipid, respectively) and were compared with a reference diet (RF) previously formulated for this species. Proteinase, lipase and amylase activities were significantly influenced by diet and weight. Specific trypsin activity was significantly higher for crayfish fed with the HL diet. Trypsin activity depended on diet and weight. Protein digestibility showed that HL was the most digestible diet and RF the least. The weight of the animals did not affect protein digestibility. Structural disorganization, hypertrophy of B‐cells and presence of large vacuoles in R‐cells were mainly observed in juveniles fed with HL, indicative of malnutrition. Thus, our data suggest that the HL diet would not be the most appropriate for C. quadricarinatus, while RF diet would be more convenient for culture of this species.  相似文献

This study included 33 samples with main focus on unprotected or rumen-protected rapeseed and soybean feedstuffs, which were analysed using an enzymatic in vitro procedure (EIVP) in order to determine intestinal crude protein (CP) digestibility (IPD) of ruminally undegraded CP. The EIVP involved the sequential digestion of samples with a protease from Streptomyces griseus, pepsin-HCl and pancreatin. Briefly, the EIVP started with determination of true protein. Feeds were incubated for 18 h in a buffer solution at a constant ratio (41 U/g) of S. griseus protease activity to feed true protein. The dried residues were incubated in pepsin-HCl solution for 1 h, and residues from this step were incubated in pancreatin solution for 24 h. Results appeared to have lower IPD dimensions than literature data of previous studies. In addition, a negative correlation became apparent between acid detergent fibre and IPD, as well as a positive correlation between CP, true protein and IPD. The EIVP in its current, strictly standardised form can be applied to develop a database that can be used for protein evaluation systems for establishing tabular values of IPD. Nevertheless, future studies may be hindered since sufficient reference values, i.e. in vivo data, are completely missing.  相似文献

The effects of prebiotics on digestibility, short-chain fatty acid (SCFA) concentrations and bacterial populations in the faeces and immunity in dogs were evaluated by meta-analyses. Overall, data from 15 published studies containing 65 different treatment means of 418 observations from different breeds of dogs were included in the data set. Feeding of prebiotics to dogs did not affect the nutrient intake (P > 0.10), nor did prebiotics change (P > 0.10) the digestibility of dry matter (DM) and fat. However, crude protein (CP) digestibility tended to decrease quadratically (P = 0.06) with increasing dosages of prebiotics, although the degree of prediction was low (R(2) = 0.33). The concentration of total SCFA (P = 0.08; R(2) = 0.90) tended to increase linearly, whereas concentration of acetate (R(2) = 0.25), propionate (R(2) = 0.88) and butyrate (R(2) = 0.85) increased quadratically with increasing dosage of prebiotics in the faeces of dogs. The numbers of beneficial bifidobacteria (P < 0.01; R(2) = 0.62) increased quadratically, but lactobacilli (P < 0.01; R(2) = 0.66) increased linearly with increasing supplementation of prebiotics. The changes in healthy bacterial numbers were affected by the interaction of initial bacterial numbers and dose of prebiotics; bacterial numbers increased relatively more when initial bacterial numbers were low. Dietary composition did not influence the response of prebiotics on lactobacilli and bifidobacterial numbers in this study. The numbers of pathogenic Clostridium perfringens and Escherichia coli were not affected by prebiotics. Prebiotics did not affect the serum immunoglobulin (Ig) concentrations such as IgG, IgA and IgM in dogs. Although prebiotics may tend to have an adverse effect on CP digestibility, prebiotics at doses up to 1.40% food intake (DM basis) might increase the beneficial bacterial populations and SCFA concentrations in the faeces of dogs. Thus, the feeding of prebiotics has a great prospective to improve the intestinal health of dogs.  相似文献

The influence of physical treatment‐expansion and flaking‐on crude proteins degradability in the rumen was studied in maize, maize‐gluten feed, rape extracted meal and in the expanded one at 120°C and 150°C, rape cake, wheat and flaked wheat by in sacco method. The enzymatic digestibility of crude protein in the rumen undegraded residues of the above mentioned feeds was determined by an enzymatically in vitro method.

The treatment of feed decreased significantly the original solubility and theoretical degradability of crude proteins, and the amount of undegraded crude proteins was increased. Positive influence on the amount of enzymatically digested crude protein was determined in rape expanded at 120 °C and 150 °C (60, 61 and/or 68%). Flaking of wheat had a similar effect. Enzymatic digestibility at undegraded rests where increased by 8–10% after the heat treatment and it remained almost unchanged in expanded maize‐gluten feed.  相似文献

The objectives of this study were to determine the chemical composition, energy content and amino acid digestibility for corn gluten meals (CGM) and to develop prediction equations for estimating energy content and amino acid digestibility for growing pigs based on the chemical characteristics of these meals. The 15 CGM tested were obtained from seven Chinese companies. Experiment 1 was conducted to determine the digestible (DE) and metabolisable energy (ME) of the 15 CGM. The 18 growing barrows (38 ± 4 kg) were assigned to three 6 × 6 Latin square designs. The 15 CGM test diets were formulated to contain 19.20% CGM, which replaced 20% of the energy supplied by corn and crystalline amino acid in the basal diet. Experiment 2 was conducted to determine the apparent (AID) and standardised (SID) ileal digestibility of the crude protein (CP) and amino acids in the 15 CGM using chromic oxide as an inert marker. The 18 growing barrows (25 ± 2 kg) fitted with a simple T-cannula were assigned to three 6 × 6 Latin square designs. The 15 test diets contained 35% of one of the 15 CGM as the sole source of amino acids in the diet. The results showed a considerable variation in the chemical composition of CGM within and among plants. On dry matter basis, the DE and ME content of the CGM ranged from 18.8 to 21.0 MJ/kg and from 18.0 to 19.9 MJ/kg, respectively. There were no significant differences in the AID and SID for CP, arginine, lysine, glycine and proline among the 15 CGM, however, for all the other amino acids, significant differences were found for their AID and SID. With R 2 values exceeding 0.50, the DE of CGM can be predicted accurately from CP and fibre content and ME from starch and fibre content. Suitable prediction equations for SID of methionine were also developed.  相似文献

The effect of two feeding systems (indoors and at pasture) on intake and digestion of fresh grass was studied at two stages of regrowth (21 and 35 days of regrowth) in two parallel experiments. In Experiment 1, 10 adult Martinik rams weighing, on average, 50.5 (±0.9) kg, including four fitted with rumen cannula, were randomly allocated to two groups according to a 2 × 2 Latin Square design. These rams consumed a 21-day regrowth of Digitaria decumbens grass diet during two successive 28-day periods, indoors (five rams) or at pasture (five tethered rams). In Experiment 2, 10 other Martinik rams weighing, on average, 45.5 (±0.9) kg, including four fitted with rumen cannula, were randomly allocated to two groups according to a 2 × 2 Latin Square design. These rams consumed a 35-day regrowth of D. decumbens grass diet during two successive 28-day periods, either indoors (five rams) or at pasture (five tethered rams). For the indoors groups, in vivo organic matter digestibility (OMD) was measured by total collection of feces. In addition, OMD was estimated indoors and at pasture using the fecal CP (CPf) method (OMDCPf). Organic matter intake (OMI) was then estimated using OMDCPf and fecal organic matter output (OMICPf). Correlations of 0.49 and 0.77 were found between in vivo OMD and OMDCPf (P < 0.05) and between OMI and OMICPf (P < 0.001), respectively. OMDCPf was 1.8% (P < 0.05) and 2.7% (P < 0.01) lower indoors than at pasture at 21 and 35 days of regrowth, respectively, whereas OMICPf indoors was 1.1 and 1.16 times that registered at pasture at 21 and 35 days of regrowth, respectively. The higher OMDCPf at pasture was linked to the higher selective behavior of rams at pasture, whereas the differences in OMICPf between the two feeding systems were linked to differences in the total bulk density of the grass. These studies show that differences in OMDCPf and OMICPf exist between animals fed indoors and at pasture with the same forage and that these differences may vary according to the stage of regrowth of the grass offered.  相似文献

The aims of this study were to provide a controlled fermentation process of blue lupin seeds (Lupinus angustifolius, cv. Neptun), monitor the changes in seed composition and determine the influence of the fermentation on the apparent ileal digestibility (AID) of crude protein (CP) and amino acids in growing pigs, compared with raw lupin seeds. The fermentation with bacteria and yeast was conducted for 24 h at 25ºC under aerobic conditions. Seed fermentation increased the contents of CP, fibre, fat and ash and most of the analysed amino acids but reduced the levels of the nitrogen-free extractives. Furthermore, fermentation decreased the contents of raffinose family oligosaccharides and phytic acids but increased the alkaloid content. The AID was estimated on three barrows (mean initial body weight 25 kg), surgically fitted with a T-cannula in the distal ileum. The pigs received three diets, each for 6 d, within three experimental periods (3 × 3 Latin Square design). The diets contained soybean meal (Group SBM), raw lupin seeds (Group RL) or fermented lupin seeds (Group FL) as solely protein sources. Fermentation had a positive impact on the AID of CP and methionine, cysteine, isoleucine, leucine, phenylalanine and valine (p < 0.05). Feeding raw or fermented lupin seeds did not affect the microbial status of the ileal digesta. Moreover, ammonia content in the caecal digesta did not differ between Groups RL and FL, although it was significantly higher than in Group SBM. It can be concluded that the fermentation process modified the chemical composition of nutrients in seeds, which can influence the digestibility and utilisation of the fermentation product in animal diets compared to raw seeds.  相似文献

The in vitro digestibility of two-stage dried olive cake (TSDOC) and olive leaves (OL) unsupplemented or supplemented with increasing amounts of urea (U) or sunflower meal (SM) (0, 1.5, 2 and 2.5 g/100 g organic matter (OM) of the by-product) was determined. Chemical and amino acid composition, in vitro digestibility, in situ rumen degradability of crude protein and amino acids, and in situ–in vitro intestinal digestibility of SM CP and amino acids was determined. The in sacco rumen degradability and in vitro intestinal availability of CP and individual amino acids were also determined. Results obtained in Granadina goats and Segureña wethers were compared. SM provides arginine, glycine and aspartic and glutamic acids. The addition of increasing amounts of U or SM improved (P<0.001) the IVDMD and IVOMD of both TSDOC and OL. There was no effect (P>0.05) of the rumen inoculum origin on in vitro TSDOC digestibility. In contrast, values for OL were higher (P<0.001) for goats versus sheep. In sacco ruminal CP degradability of SM was relatively high, and similar in sheep and goats (ED=0.78 and 0.75 for sheep and goats). Individual amino acid ruminal degradability had different values, being lowest for methionine, leucine, proline, tyrosine and cysteine. Values obtained for individual amino acids differed from those of CP. Apparent intestinal digestibility of undegraded protein (AIDUP) of SM was high (0.86 and 0.98, respectively, for sheep and goats). The intestinally absorbable protein (IADP) was low (18.9 and 24.0 for sheep and goats, respectively). Results indicate that goats and sheep have the same capacity for TSDOC digestion, but goats showed a better capacity than sheep for OL utilisation. Although the amino acids supply to the intestine from SM is not important it could be a good supplement for low degradable protein feedstuffs such as TSDOC and OL.  相似文献

Summary Several different classes of chemical carcinogens induced the transformation of human fibroblasts grown in vitro. Characteristics of the events that occur from time of treatment through the expression of neoplastic transformation are presented. The S-phase appeared to be the portion of the cell cycle most vulnerable to insult. Staging of the cells by blocking them in G1 before releasing them to proceed through scheduled DNA synthesis (S) was required to induce reproducible transformation. Compounds such as insulin were added to the cells upon release from the block to sensitize the cells to the carcinogen that was added during S. Growth of the transformed cells as distinct from nontransformed cells was promoted by growth in medium supplemented with 8X nonessential amino acids. Carcinogen-treated cells in the early stage of transformation exhibited abnormal colony morphology and were able to grow at 41°C, in air atmosphere, and in medium supplemented with only 1% serum. In addition, the transformed cells were insensitive to KB cell lysate and exhibited density independent, as well as anchorage independent, growth (i.e., growth in 0.33% agar). Cells that grew in soft agar also produced undifferentiated mesenchymal tumors in preirradiated nude mice. Cell wall-bound hydroxycinnamic acids and the composition of lignin were studied in relation to the digestibility of a collection of 91 maize silages in wethers. Total lignin and guaiacyl content showed the highest correlation coefficients with digestibility. Using the above-mentioned chemical parameters, eight equations were also developed to predict digestibility. The prediction of organic matter digestibility produced a high adjusted R 2 value (0.487) using total lignin, guaiacyl, esterified ferulic acid and esterified p-coumaric acid content as predictors. The prediction of in vivo dry matter digestibility produced a higher adjusted R 2 value (0.516) using the same variables as predictors. Cell wall digestibility depends on a multiplicity of factors and it is not possible to attribute a causal effect on in vivo digestibility to any single factor. However, total lignin, guaiacyl and p-coumaric acid content emerge as good predictors of digestibility.  相似文献

Aims: To improve the digestibility of paddy straw to be used as animal feed by means of selective delignification using white rot fungi. Methods and Results: Solid state fermentation of paddy straw was carried out with some white rot fungi for 60 days. Different biochemical analyses, e.g. total organic matter (TOM) loss, hemicellulose loss, cellulose loss, lignin loss and in vitro digestibility, were carried out along with laccase, xylanase and carboxymethyl cellulase activity. The results were compared with that of a widely studied fungus Phanerochaete chrysosporium, which degraded 464 g kg?1 TOM and enhanced the in vitro digestibility from 185 to 254 g kg?1 after 60 days of incubation. Straw inoculated with Phlebia brevispora possessed maximum crude protein. Conclusions: All the tested white rot fungi efficiently degraded the lignin and enhanced the in vitro digestibility of paddy straw. Phlebia brevispora, Phlebia radiata and P. chrysosporium enhanced the in vitro digestibility almost to similar levels, while the loss in TOM was much lesser in P. brevispora and P. radiata when compared to P. chrysosporium. Significance and Impact of the Study: The study reflects the potential of P. brevispora and P. radiata as suitable choices for practical use in terms of availability of organic matter with higher protein value, selective ligninolysis and better digestibility.  相似文献

The effects of different drying treatments (oven- andfreeze-drying) on protein extractability of three subtropical brownseaweeds, Sargassum hemiphyllum, S. henslowianum and S. patens, as well as in vitro protein digestibility and amino acid profileof their protein concentrates (PCs) were investigated. When comparedwith freeze-drying, oven-drying not only improved significantly (p <0.05, two-way ANOVA, Tukey-HSD) the protein extractability of thethree Sargassum species but also the protein quality (in vitroprotein digestibility) of their PCs. Therefore, oven-drying was found to bemore suitable for protein extraction of brown seaweeds. The relationshipsbetween the protein extractability and total phenolic content in seaweedsand between in vitro protein digestibility and total phenolic contentin PCs were also discussed.  相似文献

It is well known that energy plays an important role in sow growth and development. Increasing the utilization of lipids will be beneficial to sows. Emulsifiers are substances which stabilize mixtures and prevent oil and water from separating, thereby enhancing the digestion of lipids. This study was conducted to evaluate the effect of dietary emulsifier (lysophospholipids (LPL)) supplementation in diets differing in fat contents on growth performance, nutrient digestibility and milk composition in lactating sows, as well as performance and fecal score in piglets. A total of 32 multiparous sows (Landrace×Yorkshire) were used in a 21-day experiment. On day 110 of gestation, sows were weighed and moved into the farrowing facility, randomly assigned in a 2×2 factorial arrangement according to their BW with two levels of LPL (0 and 30 mg/kg) and two levels of fat (4.75% and 2.38% fat; 13.66 and 13.24 MJ/kg). BW loss and backfat thickness loss were decreased (P<0.05) by LPL supplementation. Backfat thickness at weaning was higher (P<0.05) in sows fed LPL supplementation diets. The apparent total tract digestibility of dry matter, nitrogen, gross energy and crude fat in sows fed LPL diets was increased (P<0.05) compared with those fed non-LPL diets. Sows fed the high-fat diets had higher (P<0.05) milk fat on day 10 and milk lactose on day 20 than those fed the low-fat diets. Milk fat and lactose concentrations in LPL supplementation treatments was increased (P<0.05) compared with non-LPL treatments on day 10 and day 20, respectively. Positive interaction effects (P<0.05) between fat and LPL were observed for milk fat concentration on day 10. In conclusion, LPL addition decreased BW loss and backfat thickness loss, improved nutrient digestibility and milk fat as well as milk lactose concentrations. In addition, there was a complementary positive effect of dietary fat and LPL supplementation on milk fat concentration in lactating sows.  相似文献

The growth response of a double-mutant fatty acid auxotroph of yeast Saccharomyces cerevisiae to exogenous saturated fatty acids of a homologous series from 12:0 to 16:0, each supplied with oleate, linoleate, linolenate, or cis11- eicosenoate, cannot be explained in terms of the efficiency of incorporation of the fatty acids into phospholipids or alteration of membrane fluidity. There is, however, a negative correlation between growth and levels of 12:0 plus 13:0 in phospholipids, as well as a positive correlation between growth and levels of 14:0, 1 5:0, and 1 6:0. We, therefore, conclude that the predominant factor in these phospholipid fatty acyl chain modifications is maintenance of an optimal concentration of C14:0 through C16:0 in phospholipids of this organism.  相似文献

The effects of cadmium (10?6, 10?5, 10?4, and 10?3 M) on the growth and development of stem apical meristem and fatty acid (FA) composition of particular lipid fractions (neutral lipids, glyco-, and phospholipids) were investigated in the shoots of Karelian birch (Betula pendula var. carelica (Mercklin) Hämet-Ahti) produced in vitro. Low cadmium concentration (10?6 M) slightly stimulated shoot morphogenesis. However, at the concentration of 10?4 M, shoot growth and development was terminated, and the concentration of 10?3 M turned out to be critical (the meristem died within the first 5 days). The presence of Cd in the medium considerably affected FA composition in the shoots. The most pronounced changes occurred in the fraction of glycolipids, and the extent of adverse influence significantly depended on metal concentration in the medium. At the Cd concentration of 10?4 M, the content of di-and trienoic FAs decreased by 3–4 times, whereas the level of monoenoic FAs rose by 5 times. The revealed differences in the FA composition of lipids in birch were considered as an indicator of adverse effect of cadmium on the structure and functions of chloroplasts and therefore on photosynthesis.  相似文献

采用f/2培养基,在3000lx,5000lx,8000lx光照强度时对杆状裂丝藻(Stichococcus bacillaris,MACC(中国海洋大学微藻种质库)/C19)和3株小球藻(Chlorellasp.MACC/C95,MACC/C97和MACC/C102)等4株海洋绿藻的总脂含量及脂肪酸组成进行了研究。单因子方差分析结果表明,光照强度对4株海洋绿藻的相对生长率、EPA(二十碳五烯酸,20∶5(n-3))含量及PUFA(多不饱和脂肪酸)总量,C19、C97和C102的脂肪含量和SFA(饱和脂肪酸)总量,C19、C95和C102的MUFA(单不饱和脂肪酸)总量均有显著影响(P0.05)。C19和C95的总脂含量随光照强度的增加总体呈降低趋势,C97和C102的总脂含量则随光照强度的升高而明显升高。C19的EPA含量随光照强度的增加呈现低-高-低趋势,C95在中低光照强度下EPA含量较高,C97和C102的EPA含量则随光照强度的增加而明显降低,4株绿藻的AA(花生四烯酸,20∶4(n-6))含量和PUFA总量均随光照强度的增加而降低。多重比较结果表明:C19在高光照强度(8000lx)时总脂含量显著低于其它各组,低光照强度(3000lx)和中光照强度(5000lx)处理间没有显著性差异(12.9%—12.7%),C97和C102则分别在高光照强度时总脂含量显著高于其它各处理(分别为40.6%和33.3%)。C19的EPA含量在中光照强度水平显著高于其它处理(12.7%),C97和C102的EPA含量分别在低光照强度水平显著高于其它处理(分别为12.0%和10.5%),C95的EPA含量在高光照强度水平显著低于其它处理,低光照强度和中光照强度处理间没有显著性差异(14.0%—14.7%);C19和C95的PUFA总量均在高光照强度水平显著低于其它处理,在低光照强度和中光照强度处理间没有显著性差异(分别为19.1%—19.4%和21.3%—21.3%),C97和C102的PUFA总量均在低光照强度水平显著高于其它处理(分别为19.2%和18.9%)。4株绿藻的主要脂肪酸成份为14∶0、16∶0、16∶1(n-7)、16∶4(n-3)、18∶1(n-9)、18∶3(n-3)、20∶4(n-6)和EPA。  相似文献
