This work focuses on the representatives of genus Echinops (Asteraceae, Cardueae) in the Aegean and Balkan regions, from the perspective of their genome evolution. Chromosome numbers were determined by orcein staining in 14 populations of nine taxa, and DNA contents were assessed by flow cytometry in 24 populations of nine taxa. A molecular phylogeny based on the internal transcribed spacer (ITS) and trnL-trnF and including first sequences for two taxa (Echinops sphaerocephalus subsp. taygeteus and E.?spinosissimus subsp. neumayeri) provided a framework for discussing genome changes. From a methodological point of view, similar C-DNA value estimates were obtained when measuring, for a same population, fresh leaves from adult plants collected in the field and from cultivated seedlings. Conversely, despite giving the appearance of being correct (e.g., low coefficient of variation), genome size assessed using silica gel-preserved material differs significantly from values obtained for the same populations with fresh material. Nevertheless, silica gel-preserved material may still provide rough estimates of genome size for, e.g., inferring ploidy level. Suitable—non-silica gel-based—DNA amounts assessed for 23 populations range from 2C?=?6.52?pg (E.?spinosissimus subsp. neumayeri) to 2C?=?9.37?pg (E.?bannaticus). Chromosome counts were established for the first time for Echinops graecus (2n?=?32), E.?sphaerocephalus subsp. albidus (2n?=?32), E.?sphaerocephalus subsp. taygeteus (2n?=?ca.?30), and E.?spinosissimus subsp. neumayeri (2n?=?28). Genome size and chromosome number are confirmed as crucial parameters for deciphering lineage diversification within the genus Echinops.  相似文献

Potential toxicity, costs, and drug‐resistant pathogens necessitate the development of new antileishmanial agents. Medicinal and aromatic plants constitute a major source of natural organic compounds. In this study, essential oils of Artemisia absinthium L. and Echinops kebericho Mesfin were investigated by GC and GC/MS analyses. Isolated oils were screened for antileishmanial activity against two Leishmania strains (L. aethiopica and L. donovani), and toxicity on the human monocytic leukemia (THP‐1) cell line and red blood cells in vitro. GC/MS Analysis revealed 65 compounds (93.74%) for Artemisia absinthium and 43 compounds (92.85%) for Echinops kebericho oil. The oils contained the oxygenated monoterpene camphor (27.40%) and the sesquiterpene lactone dehydrocostus lactone (41.83%) as major constituents, respectively. Both oils showed activity against promastigote (MIC 0.0097–0.1565 μl/ml) and axenic amastigote forms (EC50 0.24–42.00 nl/ml) of both leishmania species. Weak hemolytic effect was observed for both oils, showing a slightly decreased selectivity index (SI 0.8–19.2) against the THP‐1 cell line. Among the two oils tested, E. kebericho exerted strong antileishmanial activity that was even higher than that of amphotericin B with significant cytotoxicity. This study, therefore, demonstrated the potential use of both oils as source of novel agents for the treatment of leishmaniasis.  相似文献

The new species Echinops sahyadricus is discovered and described from northern Western Ghats, India. It is endemic to the Maharashtra state and probably restricted to few high mountain peaks in Western Maharashtra. The new species is distinguished from the related E. echinatus by adaxially glabrous to sparsely elgandular hairy leaf surfaces, larger non-cornigerous synflorescences up to 9 cm in diameter and numerous (19–24) glabrous phyllaries up to 27 mm long. An identification key to all Indian taxa of Echinops, illustration and colour plates are provided.  相似文献

A chemical analysis of essential oils from leaves of eleven Eucalyptus L’Herit taxa, grown in Viçosa, Brazil were carried out. The identification and quantification of essential oils constituents were carried out by GC‐FID and GC/MS. The leaves of E. camaldulensis and E. tereticornis presented the highest oil content (3.00% and 2.30% respectively). In total, 48 compounds were identified in the oils. Higher levels of 1,8‐cineole were found for oils produced by E. microcorys (66.2%), E. urophylla (65.4%) and E. camaldulensis (44.8%) and the hybrid E. urophylla × E. grandis (33.0%). The oil from E. saligna was composed mainly by α‐pinene (92.3%). High concentrations of α‐phellandrene were found in the oils produced by E. camaldulensis (22.9%) and E. robusta (36.6%). The oils from E. grandis and E. pilularis were rich in p‐cymene (59.5% and 46.0%, respectively). Samples with high levels of 1,8‐cineole were classified by principal component analysis (PCA) using the accumulated variance of the PC1 and PC2 into major groups. Other samples were grouped based on their content of p‐cymene; α‐phellandrene, α‐ and β‐eudesmol; α‐pinene. The PCA allowed the separation and classification of samples with the highest levels of different compounds, a procedure that can help in the decision of grouping oils from different sources for industrial use.  相似文献

The essential oils obtained by hydrodistillation of Geranium purpureum and G. phaeum were characterized by GC‐FID and GC/MS analyses (the former for the first time in general). In total, 154 constituents were identified, accounting for 89.0–95.8% of the detected GC peak areas. The investigated essential oils consisted mainly of fatty acids and fatty‐acid‐derived compounds (45.4–81.3%), with hexadecanoic acid and (E)‐phytol as the major components. The chemotaxonomic significance of the variations in the essential‐oil composition/production of the presently and previously investigated Geranium and highly related Erodium taxa from Serbia and Macedonia was assessed by multivariate statistical analyses. The main conclusions drawn from the high chemical similarity of the two genera, visible from the obtained dendrograms and biplots, confirm the close phylogenetic relationship between the investigated Geranium and Erodium taxa, i.e., that there is no great intergeneric oil‐composition variability. Changes in the composition and production of essential oils of the herein investigated taxa and 60 other randomly chosen species belonging to different plant genera were also statistically analyzed. The results put forward pro arguments for the oil‐yield–oil‐composition correlation hypothesis.  相似文献

Ripe cones of Juniperus communis L. (Cupressaceae) were collected from five wild populations in Kosovo, with the aim of investigating the chemical composition and natural variation of essential oils between and within wild populations. Ripe cones were collected, air dried, crushed, and the essential oils obtained by hydrodistillation. The essential‐oil constituents were identified by GC‐FID and GC/MS analyses. The yield of essential oil differed depending on the population origins and ranged from 0.4 to 3.8% (v/w, based on the dry weight). In total, 42 compounds were identified in the essential oils of all populations. The principal components of the cone‐essential oils were α‐pinene, followed by β‐myrcene, sabinene, and D ‐limonene. Taking into consideration the yield and chemical composition, the essential oil originating from various collection sites in Kosovo fulfilled the minimum requirements for J. communis essential oils of the European Pharmacopoeia. Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were used to determine the influence of the geographical variations on the essential‐oil composition. These statistical analyses suggested that the clustering of populations was not related to their geographic location, but rather appeared to be linked to local selective forces acting on the chemotype diversity.  相似文献

The chemical composition of the essential oils isolated from the aerial parts of Anthemis pignattiorum Guarino, Raimondo & Domina and A. ismelia Lojac . and the aerial parts and flowers of Anthemis cupaniana Tod . ex Nyman , three endemic Sicilian species belonging to the section Hiorthia, was determined by GC‐FID and GC/MS analyses. (Z)‐Muurola‐4(14),5‐diene (27.3%) was recognized as the main constituent of the A. pignattiorum essential oil, together with isospathulenol (10.6%), sabinene (7.7%), and artemisyl acetate (6.8%), while in the oil obtained from the aerial parts of A. ismelia, geranyl propionate (8.8%), bornyl acetate (7.9%), β‐thujone (7.8%), neryl propionate (6.5%), and τ‐muurolol (6.5%) prevailed. α‐Pinene was the main compound of both the aerial part and flower oils of A. cupaniana (18.4 and 13.2%, resp.). Also noteworthy are the considerable amounts of artemisyl acetate (12.7%) and β‐thujone (11.8%) found in the oil from the aerial parts and those of tricosane (9.8%) and sabinene (7.6%) evidenced in the flower oil. Furthermore, an update on the main compounds identified in the essential oils of all the Anthemis taxa studied so far was presented, and cluster analyses were carried out, to compare the essential oils of these taxa.  相似文献

In the present work, three varieties of Teucrium orientale, var. orientale, var. puberulens, and var. glabrescens, were collected and investigated for chemical composition of the oils. Subsequent gas chromatography (GC‐FID) and gas chromatography coupled to mass spectrometry (GC/MS) revealed high abundance of sesquiterpenes in the essential oils analyzed. All the oils contained β‐caryophyllene (22.6, 8.5, and 6.3%, resp.) and hexadecanoic acid (7.9, 12.8, and 13.1%). Germacrene D (24.6 and 33.4%) and bicyclogermacrene (6.7 and 8.5%) were found to be the main constituents of var. orientale and var. puberulens, respectively. The high percentages of β‐cubebene (26.9%), α‐cubebene (9.0%), and α‐copaene (7.2%) established the diversity of var. glabrescens. The qualitative difference between the essential oils allowed the differentiation between the varieties in agreement with the morphological observations described in Flora of Turkey for each variety studied. In addition, a cluster analysis of twelve Teucrium taxa based on the essential‐oil composition has been carried out. Hovewer, the analysis did not clearly reflect the infrageneric classification of the genus, it largely confirmed the relationships between the infraspecific taxa of Teucrium orientale and T. chamaedrys.  相似文献

The chemical composition of the essential oil isolated from the aerial parts of Pulicaria sicula (L.) Moris was characterized by GC‐FID and GC/MS analyses. The oil was particularly rich in oxygenated terpenoids. Among the oxygenated monoterpenes (content of 44.5%), the most abundant were borneol (23.7%), bornyl acetate (6.5%), and isothymol isobutyrate (6.2%). Caryophyllene oxide (10.2%), caryophylladienol I (4.3%), and caryophylla‐3,8(13)‐dien‐5β‐ol (4.4%) were identified as the main constituents among the oxygenated sesquiterpenes. Furthermore, a complete literature review on the composition of the essential oils of all the Pulicaria taxa studied so far was performed and a principal component analysis (PCA) was carried out.  相似文献

The chemical composition and antioxidant properties of the essential oil and EtOH extract of immortelle (Helichrysum italicum (Roth) G.Don subsp. italicum, Asteraceae) collected in Montenegro were evaluated. The essential oil was characterized by GC/MS analysis, and the content of total phenolics and flavonoids in the EtOH extract was determined using the Folin? Ciocalteu reagent. The free‐radical‐scavenging capacity (RSC) of both the essential oil and the EtOH extract was assessed with the 2,2‐diphenyl‐1‐pycrylhydrazyl (DPPH) method. Moreover, the inhibition of hydroxyl radical (.OH) generation by the EtOH extract of immortelle was evaluated for the first time here. Neryl acetate (28.2%) and γ‐curcumene (18.8%) were the main compounds in the essential oil, followed by neryl propionate (9.1%) and ar‐curcumene (8.3%). The chemical composition of the oils of the examined and additional 16 selected Helichrysum italicum taxa described in literature were compared using principal component (PCA) and cluster (CA) analyses. The results of the statistical analyses implied the occurrence of at least four different main and three subchemotypes of essential oils. Considering the antioxidant properties, the EtOH extract of immortelle exhibited similar potential as propyl gallate and quercetin, while the essential oil exhibited relatively weak DPPH.‐scavenging capacity.  相似文献

The chemical compositions of the essential oils obtained by hydrodistillation of the aerial parts of Croatian Eryngium alpinum L. and E. amethystinum L. were characterized by GC‐FID and GC/MS analyses. The main components identified were the sesquiterpene β‐caryophyllene (19.7%) in the oil of E. amethystinum and the oxygenated sesquiterpene caryophyllene oxide (21.6%) in the oil of E. alpinum. Overall, 32 and 35 constituents were detected in the essential oils of the aerial parts of E. alpinum and E. amethystinum, respectively, representing 92.4 and 93.1% of the total oil compositions. The essential oils of both Eryngium species were proved to reduce the number of lesions in the local host Chenopodium quinoa infected with cucumber mosaic virus and an associated satellite. This is the first investigation of antiphytoviral activity of essential oils of Eryngium species.  相似文献

The genus Lantana has many species complexes, and L. camara is one of the aggressive alien weedy species complexes; species delimitation in these complexes is a nightmare for taxonomists. We examined the diversity in the chemical composition of foliar essential oils among morphotypes of Lantana species complexes inhabiting the same ecological gradient, and its taxonomic and ecological significance. The yields of essential oils varied from 0.1 to 0.79% in foliar hydrodistillates of eleven morphotypes, and a total of 39 chemical constituents were detected by GC/MS. The quantitative and qualitative variability in the composition of essential oils among morphotypes was very high, and hence they represent chemotypes. The diversity observed in the composition of essential oils appears to be of genetic origin and thus of taxonomic value. The formation of distinct clusters and sub‐clusters at high distance cluster combine values also substantiates that the patterns of distribution of chemical constituents among morphotypes can be used in delimiting species and infraspecific taxa within the species complexes. The presence of β‐caryophyllene and other such compounds, which are known to prevent herbivory, in morphotypes of Lantana species complexes suggest that these compounds may provide selective advantage to Lantana over native species in the invasion of new and disturbed habitats.  相似文献

Here we report, for the first time, the results of detailed GC and GC/MS analyses of the essential oil of a rare taxon in Serbia, Hypericum elegans Stephan ex Willd . One hundred and sixty two constituents identified accounted for 98.6% of the oil. The major components of the oil were undecane (31.9%), α‐pinene (16.7%), nonane (6.1%), bicyclogermacrene (5.8%), 2‐methyloctane (3.7%), and germacrene D (3.6%). Non‐terpenoids as chemotaxonomic markers constituted the main fraction of H. elegans oil, whereby n‐alkanes were the most abundant contributors of this fraction. Based on these results and previously published ones, we performed an intrasectional multivariate statistical comparison of corresponding essential‐oil chemical compositions. Principal component analysis (PCA) and agglomerative hierarchical clustering (AHC) of the data on the volatile profiles of section Hypericum taxa revealed that H. elegans either represents an oil chemotype of its own (AHC) or could be considered related to H. perforatum (PCA).  相似文献

以新疆蓝刺头(Echinops ritro L.)全草为研究材料,通过硅胶柱色谱,Sephadex LH-20柱色谱,重结晶等技术对新疆蓝刺头化学成分进行分离纯化,通过理化性质分析及1H-NMR,13C-NMR等技术对化合物结构进行鉴定.结果表明,共分离出5个化合物,分别是三萜类化合物蒲公英甾醇乙酰酯(化合物1)、蒲公英甾醇(化合物2)、黄酮苷类化合物金丝桃苷(化合物3)、胡萝卜苷(化合物4)与β-豆甾醇葡萄糖苷(化合物5).其中化合物1,2,5首次从该种植物中分离,化合物3为首次从该属植物中分离.  相似文献

The composition of the essential oil isolated from leaves and flowers of Pulicaria incisa sub. candolleana E. Gamal ‐Eldin , growing in Egypt, was analysed by GC and GC‐MS. Forty‐nine and 68 compounds were identified from the oils of the leaves and flowers accounting for 86.69 and 84.29%, respectively of the total detected constituents. Both leaves and flowers oils were characterized by the high content of carvotanacetone with 66.01, 50.87 and chrysanthenone 13.26, 24.3%, respectively. The cytotoxic activity of both essential oils was evaluated against hepatocellular carcinoma cell line HEPG‐2, using MTT assay and vinblastine as a reference drug. Leaf oil showed higher activity with IC50 11.4 μg/ml compared with 37.4 μg/ml for flower oil. The antimicrobial activity of both oils was evaluated using agar well diffusion method towards two representatives for each of Gram positive and Gram negative bacteria as well as four representatives for fungi. The minimum inhibitory concentration of both essential oils against bacterial and fungal strains was obtained in the range of 0.49 – 15.63 μg/ml.  相似文献

The chemical compositions of the essential oils of seven natural populations of Seseli rigidum were analyzed. The essential‐oil yield ranged from 0.16 to 2.09%. Analysis of variance (ANOVA) revealed that there were no statistically significant differences in the mean essential‐oil yields between the populations, and no significant influence of the climate or soil type on the oil yield was observed. In all 67 analyzed samples, the polyacetylene falcarinol was the main compound, followed by octanal, methyl linoleate, α‐muurolene, 3‐butylphthalide, falcarinone, muurola‐4,10(14)‐dien‐1β‐ol, β‐sesquiphellandrene, salvial‐4(14)‐en‐1‐one, δ‐amorphene, spathulenol, and isospathulenol. The principal component analysis (PCA), the canonical discriminant analysis (CDA), and the cluster analysis (CA) revealed differentiation between the populations based on the climate. Three groups of populations were formed; the first group was composed of samples growing in regions with a humid climate, with oils having high falcarinol and low sesquiterpene contents, and the second and third groups comprised samples exposed to semi‐arid climate, with oils characterized by a lower falcarinol and higher α‐muurolene, δ‐amorphene, β‐sesquiphellandrene, and salvial‐4(14)‐en‐1‐one contents. The semi‐arid populations were divided into two groups, which were distinguished based on the oil contents of sesquiterpenes, falcarinone, and 3‐butylphthalide. On the other hand, no clear separation between populations based on the different soil types could be observed.  相似文献

Characterization by GC‐FID and GC/MS analyses of the Stachys officinalis (L.) Trevis . essential oil obtained by hydrodistillation of the aerial parts allowed the identification of 190 components that represented 97.9% of the total oil content. The main constituents identified were germacrene D (19.9%), β‐caryophyllene (14.1%), and α‐humulene (7.5%). Terpenoids were by far predominant (89.4%), with sesquiterpene hydrocarbons (69.1%) and oxygenated sesquiterpenes (14.8%) being the most abundant compounds detected in the oil. Based on the present and previously published results, multivariate statistical comparison of the chemical composition of the essential oils was performed within the species. Principal component analysis (PCA) and agglomerative hierarchical clustering (AHC) of the data on the volatile profiles of S. officinalis taxa revealed no pronounced differences among the samples originated from the Balkan Peninsula. Additionally, the oil was screened for in vitro antibacterial and antifungal activity using the broth microdilution assay. The oil's best antimicrobial activities were obtained against the mold Aspergillus niger (minimal inhibitory (MIC) and minimal fungicidal (MFC) concentrations of 2.5 and 5.0 mg/ml, resp.) and the yeast Candida albicans (MIC and MFC of 5.0 mg/ml).  相似文献

Seseli libanotis is an aromatic umbelliferous plant distributed sporadically on dry grassland edges in Europe and Western Asia. The essential oil composition in the different plant parts was studied from plants collected on nine sites in Austria and one site in Alto Adige, Italy. Monoterpenes such as α‐pinene, sabinene and β‐myrcene and the sesquiterpene germacrene D were present in all essential oils from the aerial parts. Inflorescences and fruits had the highest essential oil contents. These essential oils from four sites were rich in acorenone B, while from other three sites they had carotol as a major component. Osthole as furocoumarin occurred in some oils. The root essential oils were dominated by α‐pinene. The essential oil variability has been studied by principal component analysis (PCA) and discriminant analysis (DA) with plant parts or sampling site as a priori groups. PCA could well separate inflorescence and fruit essential oil samples from leaf and stem essential oil samples. DA differentiated well between the plant parts and most of the sampling sites.  相似文献

Analyses by GC and GC/MS of an essential‐oil sample obtained from dry fruits of Scandix balansae Reut. ex Boiss . allowed the identification of 81 components, comprising 91.4% of the total oil composition. Interestingly, the major identified volatile compounds were medium‐chain‐length n‐alkanes, i.e., tridecane (6.7%), pentadecane (13.4%), and heptadecane (19.3%), and a long‐chain homolog nonacosane (7.6%). A number of minor oil constituents, among them tetradecyl 3‐methylbutanoate, and octadecyl 2‐methylpropanoate, 3‐methylbutanoate, and pentanoate, turned out to have a restricted natural occurrence not only in umbellifers but also in the Plant Kingdom, whereas the last ester is a new natural compound in general. The identity of these rare plant constituents that present excellent chemotaxonomic marker candidates for Scandix species was unambiguously confirmed by co‐injection of the oil sample with appropriate standards, which were synthesized for this purpose and fully characterized (1H‐ and 13C‐NMR, IR, MS). To explore the possible applicability of the essential oils' compositional data in the taxonomy of Apiaceae, the herein studied and additional 58 oils obtained from Scandiceae taxa were compared using multivariate statistical analyses (MVA). MVA demonstrated that the evolution of the volatiles' metabolism of Scandiceae taxa was neither genera‐specific nor follows their morphological evolution.  相似文献
