Preparation of a New Chiral Stationary Phase Based on Macrocyclic Amide Chiral Selector for the Liquid Chromatographic Chiral Separations 下载免费PDF全文
A new chiral stationary phase (CSP) based on macrocyclic amide receptor was prepared starting from (1R,2R)‐1,2‐diphenylethylenediamine. The new CSP was successfully applied to the resolution of various N‐(substituted benzoyl)‐α‐amino amides with reasonably good separation factors and resolutions (α = 1.75 ~ 2.97 and RS = 2.89 ~ 6.82 for 16 analytes). The new CSP was also applied to the resolution of 3‐substituted 1,4‐benzodiazepin‐2‐ones and some diuretic chiral drugs including bendroflumethiazide and methylchlothiazide and metolazone. Acetylated and/or 3,5‐dimethylphenylcarbamated riboflavins were prepared and the resulting riboflavin derivatives as well as natural riboflavin were regioselectively immobilized on silica gel through chemical bonding at the 5’‐O‐ or 3‐N‐position of the riboflavin to develop novel chiral stationary phases (CSPs) for enantioseparation by high‐performance liquid chromatography (HPLC). The chiral recognition abilities of the obtained CSPs were significantly dependent on the structures of the riboflavin derivatives, the position of the chemical bonding on the silica gel, and the structures of the racemic compounds. The CSPs bonded at the 5’‐O‐position on the silica gel tended to well separate helicene derivatives, while the CSPs bonded at the 3‐N‐position composed of acetylated and 3,5‐dimethylphenylcarbamated riboflavins showed a better resolving ability toward helicene derivatives and bulky aromatic racemic alcohols, respectively, and some of them were completely separated into the enantiomers. The observed difference in the chiral recognition abilities of these riboflavin‐based CSPs is discussed based on the difference in their structures, including the substituents of riboflavin and the positions immobilized on the silica gel. 在模拟动物体生理条件下,用荧光猝灭、荧光偏振和紫外-可见吸收光谱法研究了槐定碱与牛血清白蛋白(BSA)结合作用。荧光猝灭数据显示,槐定碱与BSA发生反应生成了新的复合物,属于静态荧光猝灭。求出了不同温度(19、25、31、37℃)下槐定碱与BSA作用的结合常数分别为1.219×106,1.164×106,1.110×106和1.057×106L/mol,由van’tHoff方程式计算槐定碱与BSA反应的热力学参数:焓变ΔH和熵变ΔS值分别为-5.97kJ/mol和96.11J/(mol.K),表明槐定碱与BSA间的作用力以静电引力为主。以华法林和布洛芬(分别为siteI和siteII探针)为标记药物研究槐定碱在BSA上的结合位点,结果表明,槐定碱结合在BSA疏水空腔的siteI位点。 相似文献

Chiral Ligand‐Exchange Resolution of Underivatized Amino Acids on a Dynamically Modified Stationary Phase for RP‐HPTLC 下载免费PDF全文
The synthesis of Spi(τ‐dec), derived from the selective alkylation of L‐spinacine (4,5,6,7‐tetrahydro‐1H‐imidazo[4,5‐c]pyridine‐6‐carboxylic acid) at the τ‐nitrogen of its heteroaromatic ring, with a linear hydrocarbon chain of 10 carbon atoms, is described here for the first time. Spi(τ‐dec) was successfully employed in the past to prepare home‐made chiral columns for chiral ligand‐exchange high‐performance liquid chromatography. In the present article a new method is described, using Spi(τ‐dec) as a chiral selector in high‐performance thin‐layer chromatography (HPTLC): commercial hydrophobic plates were first coated with Spi(τ‐dec) and then treated with copper sulfate. The performance of this new chiral stationary phase was tested against racemic mixtures of aromatic amino acids, after appropriate optimization of both the conditions of preparation of the plates and the mobile phase composition. The enantioselectivity values obtained for the studied compounds were higher than those reported in the literature for similar systems. Synthesis and Enantioseparation Ability of Xylan Bisphenylcarbamate Derivatives as Chiral Stationary Phases in HPLC 下载免费PDF全文
Ten novel xylan bisphenylcarbamate derivatives bearing meta‐ and para‐substituents on their phenyl groups were synthesized and their chiral recognition abilities were evaluated as the chiral stationary phases (CSPs) for high‐performance liquid chromatography (HPLC) after coating them on macroporous silica. The chiral recognition abilities of these CSPs depended on the nature, position, and number of the substituents on the phenyl moieties. The introduction of an electron‐donating group was more attractive than an electron‐withdrawing group to improve the chiral recognition ability of the xylan phenylcarbamate derivatives. Among the CSPs discussed in this study, xylan bis(3,5‐dimethylphenylcarbamate)‐based CSP seems to possess the highest resolving power for many racemates, and the meta‐substituted CSPs showed relatively better chiral recognition than the para‐substituted ones. For some racemates, the xylan bis(3,5‐dimethylphenylcarbamate) derivative exhibited higher enantioselectivity than the CSP based on cellulose tris(3,5‐dimethylphenylcarbamate). Stereoselective high‐performance liquid chromatographic separations of eight sterically constrained cyclic β‐amino acid enantiomer pairs were carried out using the newly developed Cinchona alkaloid‐based zwitterionic chiral stationary phases Chiralpak ZWIX(+) and ZWIX(?). The effects of the mobile phase composition, the nature and concentrations of the acid and base additives, the counterions and temperature on the separations were investigated. The changes in standard enthalpy, Δ(ΔH°), entropy, Δ(ΔS°), and free energy, Δ(ΔG°), were calculated from the linear van't Hoff plots derived from the ln α vs. 1/T curves in the studied temperature range (10–50°C). The values of the thermodynamic parameters depended on the nature of the selectors and the structures of the analytes. Unusual temperature behavior was observed on the ZWIX(?) column: decreased retention times were accompanied by increased separation factors with increasing temperature. On the ZWIX(+) column only enthalpically, whereas on the ZWIX(?) column both enthalpically and entropically driven separations were observed. This paper describes a single-step method for the biomimetic synthesis of stably suspended magnetite nanoparticles in poly(vinyl alcohol) termed ferrofluids. The challenge is to synthesize water based stable magnetic colloids with a control over the particle size and morphology for biomedical applications. The polymer possibly plays a dual role of a surfactant and a functionalizing agent. Transmission electron microscopy, infrared spectroscopy and vibrating sample magnetometry were used to investigate the properties of the synthesized ferrofluids. It has a strong affinity towards the tryptophan residues in bovine serum albumin protein as determined from the fluorescence emission studies. For in vivo applications this could indirectly mean a resistance to immune response and thus ensure long-term circulation. The ability of the synthesized ferrofluid to bind a che-motherapeutic drug ceftriaxone and its ionic release was observed. The polymer hydroxyl group allows drug-binding and the magnetic property allows targeting to specific sites. Magnetic hybrid fluids with combined advantages of magnetism and polymer open up new perspectives for applications. 相似文献

Liquid Chromatographic Resolution of Mexiletine and Its Analogs on Crown Ether‐Based Chiral Stationary Phases 下载免费PDF全文
Mexiletine, an effective class IB antiarrhythmic agent, and its analogs were resolved on three different crown ether‐based chiral stationary phases (CSPs), one (CSP 1 ) of which is based on (+)‐(18‐crown‐6)‐2,3,11,12‐tetracarboxylic acid and the other two (CSP 2 and CSP 3 ) are based on (3,3’‐diphenyl‐1,1’‐binaphthyl)‐20‐crown‐6. Mexiletine was resolved with a resolution (RS) of greater than 1.00 on CSP 1 and CSP 3 containing residual silanol group‐protecting n‐octyl groups on the silica surface, but with a resolution (RS) of less than 1.00 on CSP 2 . The chromatographic behaviors for the resolution of mexiletine analogs containing a substituted phenyl group at the chiral center on the three CSPs were quite dependent on the phenoxy group of analytes. Namely, mexiletine analogs containing 2,6‐dimethylphenoxy, 3,4‐dimethylphenoxy, 3‐methylphenoxy, 4‐methylphenoxy, and a simple phenoxy group were resolved very well on the three CSPs even though the chiral recognition efficiencies vary with the CSPs. However, mexiletine analogs containing 2‐methylphenoxy group were not resolved at all or only slightly resolved. Among the three CSPs, CSP 3 was found to show the highest chiral recognition efficiencies for the resolution of mexiletine and its analogs, especially in terms of resolution (RS). Valacyclovir, a potential prodrug for the treatment of patients with herpes simplex and herpes zoster, and its analogs were resolved on two chiral stationary phases (CSPs) based on (3,3’‐diphenyl‐1,1’‐binaphthyl)‐20‐crown‐6 covalently bonded to silica gel. In order to find out an appropriate mobile phase condition, various mobile phases consisting of various organic modifiers in water containing various acidic modifiers were applied to the resolution of valacyclovir and its analogs. When 30% acetonitrile in water containing any of 0.05 M, 0.10 M, or 0.15 M perchloric acid was used as a mobile phase, valacyclovir and its analogs were resolved quite well on the two CSPs with the separation factors (α) in the range of 2.49 ~ 6.35 and resolutions (RS) in the range of 2.95 ~ 12.21. Between the two CSPs, the CSP containing residual silanol protecting n‐octyl groups on the silica surface was found to be better than the CSP containing residual silanol groups. The enantiomeric separation ability of the newly prepared chiral stationary phases containing acridino‐18‐crown‐6 ether selectors was studied by high‐performance liquid chromatography (HPLC). Four regioselective-carbamoylated cellulose derivatives having two different substituents at 2-, 3-, and 6-position were prepared and evaluated as chiral stationary phases (CSPs) for high-performance liquid chromatography. Investigations showed that the nature and arrangement of the substituents significantly influenced the chiral recognition abilities of the heterosubstituted cellulose derivatives and each derivative exhibited characteristic enantioseparation. Some racemates were better resolved on these derivatives than the corresponding homogeneously substituted cellulose derivatives including a commercial CSP, Chiralcel OD. Racemic compounds shown in this study were most effectively discriminated on cellulose 2,3-(3-chloro-4-methylphenylcarbamate)-6-(3,5-dimethylphenylcarbamate) and 2,3-(3,5-dimethylphenylcarbamate)-6-(3-chloro-4-methylphenylcarbamate). 相似文献

The enantiomeric separation of eight pesticides including bitertanol ( 1 ), diclobutrazol ( 2 ), fenbuconazole ( 3 ), triticonazole ( 4 ), imazalil ( 5 ), triapenthenol ( 6 ), ancymidol ( 7 ), and carfentrazone‐ethyl ( 8 ) was achieved, using normal‐phase high‐performance liquid chromatography on two cellulosed‐based chiral columns. The effects of isopropanol composition from 2% to 30% in the mobile phase and column temperature from 5 to 40 °C were investigated. Satisfactory resolutions were obtained for bitertanol ( 1 ), triticonazole ( 4 ), imazalil ( 5 ) with the (+)‐enantiomer eluted first and fenbuconazole ( 3 ) with the (—)‐enantiomer eluted first on Lux Cellulose‐2 and Lux Cellulose‐3. (+)‐Enantiomers of diclobutrazol ( 2 ) and triapenthenol ( 6 ) were first eluted on Lux Cellulose‐2. (—)‐Carfentrazone‐ethyl ( 8 ) were eluted first on Lux Cellulose‐2 and Lux Cellulose‐3 with incomplete separation. Reversed elution orders were obtained for ancymidol (7). (+)‐Ancymidol was first eluted on Lux Cellulose‐2 while on Lux Cellulose‐3 (—)‐ancymidol was first eluted. The results of the elution order at different column temperatures suggested that column temperature did not affect the optical signals of the enantiomers. The enantiomeric separation of 21 ruthenium (II) polypyridyl complexes was achieved with a novel class of cyclofructan‐based chiral stationary phases (CSPs) in the polar organic mode. Aromatic derivatives on the chiral selectors proved to be essential for enantioselectivity. The R‐napthylethyl carbamate functionalized cyclofructan 6 (LARIHC CF6‐RN) column proved to be the most effective overall, while the dimethylphenyl carbamate cyclofructan 7 (LARIHC CF7‐DMP) showed complementary selectivity. A combination of acid and base additives was necessary for optimal separations. The retention factor vs. acetonitrile/methanol ratio plot showed a U‐shaped retention curve, indicating that different interactions take place at different polar organic solvent compositions. The separation results indicated that π–π interactions, steric effects, and hydrogen bonding contribute to the enantiomeric separation of ruthenium (II) polypyridyl complexes with cyclofructan chiral stationary phases in the polar organic mode. 紫杉醇(Paclitaxel,商品名Taxol)是一种在红豆杉科(Taxaceae L.)红豆杉属(Taxus L.)生长缓慢的常绿乔木中分离提取的天然化合物。卡铂和紫杉醇均是目前临床上使用率很高的抗肿瘤药物,并在临床上经常配伍使用治疗不同的癌症。本研究以叶酸偶联的牛血清白蛋白作为药物载体,采用去溶剂技术制备了叶酸靶向卡铂—紫杉醇的白蛋白纳米粒,并研究了靶向制剂体外释放性质。研究结果表明:卡铂—紫杉醇白蛋白纳米粒平均粒径为199.4 nm,Zeta电位为-30.90 mV。卡铂包封率为91.4%;紫杉醇包封率为56.1%,药物总载药量为21%。其冻干粉复溶12 h后各项数据未发生较大变化,说明其具有良好的稳定性。体外释放结果表明叶酸—卡铂—紫杉醇白蛋白纳米粒与卡铂和紫杉醇原粉比较具有明显的缓释效果,体外释药时间可达120 h。 相似文献

合成了壳寡糖和稀土离子La3+的配合物,利用红外光谱、紫外光谱和差热-热重手段对其结构和性质进行了表征。采用抑菌圈法考察了壳寡糖、壳寡糖-La对革兰氏阳性菌金黄色葡萄球菌和革兰氏阴性菌大肠杆菌的抑菌活性。此外,紫外光谱、荧光光谱和循环伏安曲线法研究壳寡糖-La与牛血清白蛋白(BSA)相互作用。结果表明壳寡糖、壳寡糖-La对两种细菌均具有较强的抑菌活性,且壳寡糖-La的抑菌活性强于壳寡糖;壳寡糖-La使BSA的内源荧光猝灭,猝灭机制为静态猝灭,并计算了室温下壳寡糖-La与BSA的结合常数和结合位点数分别为6.35×104L/mol和1.29。 相似文献

利用光谱探针技术研究在酸性溶液中考马斯亮蓝G-250(Coomassie brilliant blue G-250,CBBG)与牛血清白蛋白(bovine serum albumin,BSA)相互作用机理,考察了不同实验条件对CBBG-BSA复合物吸收光谱的影响。实验结果表明:CBBG与BSA相互作用产生光谱蓝移主要是由CBBG与BSA间的疏水相互作用引起,而静电作用则是形成CBBG-BSA蓝色复合物的必要条件。同时,CBBG聚集体的聚集程度是影响CBBG-BSA蓝色复合物形成的重要因素。 相似文献

The chromatographic chiral resolution of 4-aryl-1,4-dihydropyridines ( 1–32 ), 4-aryl-2-thioxo-1,2,3,4-tetrahydropyrimidines ( 33–38 ), and 4-aryl-2-oxo-1,2,3,4-tetrahydropyrimidines ( 39–41 ) was studied on a tetraproline-immobilized chiral column synthesized in our lab. This tetraproline chiral stationary phase can resolve most of these compounds. The 4-aryl-2-thioxo-1,2,3,4-tetrahydropyrimidines ( 33–38 ) and 4-aryl-2-oxo-1,2,3,4-tetrahydropyrimidines ( 39–41 ) were more efficiently resolved than the racemic 4-aryl-1,4-dihydropyridines on the tetraproline chiralstationary phase. Analytes with 5,5-dimethyl groups ( 39–41 ) were less efficiently resolved than analytes without 5,5-dimethyl substituents ( 1–16 ). The 4-aryl-2-oxo-1,2,3,4-tetrahydropyrimidines ( 39–41 ) without a sulfur atom were much more efficiently resolved than 4-aryl-2-thioxo-1,2,3,4-tetrahydropyrimidines ( 33–38 ). No obvious electronic effects on the resolution of any of these analytes ( 1–41 ) were observed on the tetraproline chiral stationary phase.
