Healing of skin wounds in the African catfish Clarias gariepinus   总被引:1,自引:0,他引:1  
The African catfish Clarias gariepinus was used as a model for wound healing and tissue regeneration in a scale-less fish. A temporal framework of histological and cell proliferation markers was established after wound induction in the dorsolateral cranial region, by removing the epidermal and dermal layers, including stratum adiposum (SA). Wound closure and epidermis formation was initiated within 3 h post-procedure (hpp) with migration and concomitant proliferation of epidermal cells from the wound borders. The wound was covered by this primary epidermal front 12 hpp and fusion of the opposing epidermal fronts occurred within 24 hpp. Attachment of the newly formed epidermal layer to the underlying dermis was observed 48 hpp concomitant with a second wave of cell proliferation at the wound edge. Normal epidermal thickness within the wound was achieved 72 hpp. Formation of a basement membrane occurred by 120 hpp with concomitant emergence of the SA from the wound borders. Wound healing in C. gariepinus skin involved closure of the wound and re-epithelization through cell migration with a single wave of early cell proliferation not documented in other species. Furthermore, covering of the wound by epithelium as well as the reappearance of the basement membrane and SA occurred sooner than in other fish species.  相似文献

African catfish Clarias gariepinus hatched with morphologically immature features; however, sensory organs developed rapidly with fish growth. Although the eyes of newly hatched larvae were immature without pigment, in 2 day‐old larvae, the retina of the eyes had already developed except for the rod cells. No free neuromasts were observed in newly hatched larvae. In 1 day‐old larvae, however, free neuromasts were observed on the head and trunk. Free neuromasts increased with larval growth. Newly hatched larvae had simple round‐shaped otic vesicles; however, all sensory epithelia of the inner ear were observed until the larvae were 3 days old. Two day‐old larvae swam horizontally, had sharp teeth, commenced ingesting rotifers and also artificial feed (small‐size pellets) under both light and dark conditions; by then the larvae already had many taste buds. Three day‐old larvae showed negative phototaxis and cannibalism by eating their conspecifics. Most of the free neuromasts observed in this study had the peculiar feature of many microvilli around the sensory cells on the apical surface. Detected free neuromasts as ordinary type lateral‐line organs were not observed in previous reports in teleosts. In 10 day‐old larvae, there were two lines of free neuromasts on the flank and lower edge of the trunk; presumptive canal neuromasts were oval shaped and had begun to sink under the skin. The direction of maximum sensitivity of the neuromasts was parallel with the longitudinal axis of their elliptical apical surface.  相似文献

Tetracycline was used as a chemical tag in a mark‐recapture study to examine the pattern of increment formation in the otoliths of Tropheus moorii , a rock‐dwelling cichlid from Lake Tanganyika. A total of 256 fish were captured by divers and injected with tetracycline. Of these, nine were recaptured after either 1 or 2 years at liberty and eight retained tags within their otoliths. Comparison of the number of growth increments formed after the tag and the time at liberty demonstrated that increments were deposited on an annual basis in the otoliths of this species. Furthermore, there was a strong relationship between otolith mass and age suggesting that otoliths grew at a predictable rate throughout the life of the fish. Validation of an annual pattern of increment deposition allowed age and growth information to be derived from otoliths. This showed that T. moorii grew rapidly to attain adult size by 3 years of age. Males grew faster than females and also attained a larger size than females (8·74 v . 7·91 cm L S respectively). The longevity of some of these small freshwater fish was surprising; the oldest individual had an age of 10 years, while the average age of adults was 4 years.  相似文献

The African sharptooth catfish Clarias gariepinus lives in freshwater, is an obligatory air breather, and can survive on land during drought. The objective of this study was to elucidate how C. gariepinus defends against ammonia toxicity when exposed to terrestrial conditions. During 4 d of aerial exposure, there was no accumulation of urea in its tissues, and the rate of urea excretion remained low. Thus, exposure to terrestrial conditions for 4 d did not induce ureogenesis or ureotely in C. gariepinus. Volatilization of NH(3) was not involved in excreting ammonia during aerial exposure. In addition, there were no changes in levels of alanine in the muscle, liver, and plasma of C. gariepinus; nor were there any changes in the glutamine levels in these tissues. However, there were extraordinarily high levels of ammonia in the muscle (14 micromol g(-1)), liver (18 micromol g(-1)), and brain (11 micromol g(-1)) of fish exposed to terrestrial conditions for 4 d. This is the first report on a fish adopting high tolerance of ammonia in cells and tissues as the single major strategy to defend against ammonia toxicity during aerial exposure. At present, it is uncertain how C. gariepinus tolerates such high levels of ammonia, especially in its brain, but it can be concluded that, contrary to previous reports on two air-breathing catfishes (Clarias batrachus and Heteropneustes fossilis) from India, C. gariepinus does not detoxify ammonia to urea or free amino acids on land.  相似文献

African sharptooth catfish Clarias gariepinus were housed under continuous dim light (1 lx) or 12L:12D (350–0 lx) cycles. The number of skin lesions, as indicator of aggressive acts, and plasma cortisol levels, as indicator of stress‐axis activity, were measured at baseline as well as following a stressor (given in the light or dark phase). Results showed that (1) baseline plasma cortisol levels were not different between photoperiods, (2) the number of baseline skin lesions was highest for C. gariepinus housed under continuous dim light, (3) stressor‐induced peak levels of plasma cortisol were highest in the light phase and (4) the number of skin lesions following a stressor was highest in the dark phase. The higher number of stressor‐related skin lesions in the dark (active) phase suggests increased stressor‐induced aggression while in the active phase. In addition, the data suggest that housing under continuous dim light does not result in higher stress‐axis activity, as measured by baseline levels of cortisol, but does result in more stressor‐induced aggression, as measured by the higher number of skin lesions. The latter may be related to the fact that the continuous dim light photoperiod has twice the number of dark‐phase (active) hours in which stressor‐induced aggression is stronger compared to the 12L:12D photoperiod, which has a light phase in which stressor‐induced aggression is lower.  相似文献

Some aspects of the physiology and biochemistry of the African sharptooth catfish, Clarias gariepinus (Burchell, 1822), were studied along with an array of physicochemical characteristics of the water in Lake Maryût, Egypt. Data were compared to those of a reference fish hatchery. At least 11 of the conventional water pollutants (Cd, Cu, Fe, Pb, Mn, Hg, Ni, Zn, turbidity, chemical oxygen demand and ammonia nitrogen) were elevated in the most polluted main basin of the lake. In turn, serologic analysis of the indigenous catfish C. gariepinus pointed at functional impairment of the liver, heart and kidney as reflected by the elevated activities of alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase, creatine kinase, cholinesterase, glucose and creatinine. Reduced nucleic acid measurements [the ratio RNA/DNA and the relative RNA content (r)] indicated diminished protein synthesis and impaired growth in polluted fish. In some instances, glucose and nucleic acid measures were elevated in favour of fish from moderately polluted basins of the lake rather than in reference fish. Fish seemed to profit from the typical geographical habitat in the lake, regardless of the virtual contamination there, versus reference fish that seemed to suffer congestion stress and food competition in the confined and overcrowded commercial pond.  相似文献

Artificially fertilized eggs and yolk-sac larvae of a freshwater tropical/subtropical fish Clarias gariepinus receiving no external food were incubated at 22, 25 and 28° C until full yolk resorption. Developmental time, size and matter composition (CHNS-O Analyzer and ashing) were assessed at egg fertilization, hatching and yolk resorption; respiration was measured every 4–5 h. The course of acceleration of C. gariepinus embryonic developmental rate with temperature ( Q10 dev) was compared over the temperature range to those of Cyprinus carpio and Oncorhynchus mykiss ; they differed greatly, but were similar when compared on the basis of effective temperatures specific to each fish. Specific growth rates for energy (88, 150 and 183% per day at 22,25 and 28° C, respectively) as well as the conversion efficiencies of egg energy (64, 71 and 68%, respectively) and protein (71, 78 and 76%, respectively) in C. gariepinus larval tissues were higher than those known for the endogenous feeding period of coldwater and temperate fish species. In C. gariepinus at the end of yolk resorption, the carbon percentage and caloric values of dry weight, size (in terms of dry matter, minerals, protein and energy per larva) and transformation efficiencies were lowest at 22° C, highest at 25° C and had slightly decreased at 28° C. A tentative mechanism which leads to the positive or negative response of body size to temperature over the viable temperature range is defined.  相似文献

The otoliths of juveniles and adults of European anchovy Engraulis encrasicolus held in aquaria were marked by immersion in oxytetracycline hydrochloride (OTC) at concentrations between 350 and 410 mg l−1 for 12 h. Counts of microincrements between fluorescent bands validated the daily otolith increment formation. The otolith increments were easily readable at ×400 with average increment widths of c . 1·1 µm. Validation was successfully demonstrated in juveniles and adults maintained for short periods in the aquaria in the summer. For European anchovy captured as juvenile and reared to adults, however, increment formation appeared less than daily. The daily periodicity of the otoliths in juvenile European anchovy implies that counting of microincrements can be used to study their birth dates. The application of this technique to adults, however, may lead to the underestimation of actual age and further research needs to be done to clarify the reasons for the apparent loss of the daily rhythm over long periods.  相似文献

The sharptooth catfish, Clarias gariepinus (Burchell, 1822), is a widespread African freshwater fish species which occurs naturally from the Nile to the Orange River. Through translocation aquaculturists, farmers, anglers and engineers have helped to extend its range into many South African rivers where it did not occur naturally and where it is now an alien invasive species. Being a very hardy fish species it will have a major impact on a range of aquatic, amphibian and avian biota, impacts that should not be under-estimated and which urgently require study. Ironically, the illegal introduction of catfish into some dams has impacted on sport fisheries for another alien: bass. There is also a serious potential impact from alien catfish on freshwater eels. Anglers and farmers need to be educated about the adverse consequences of introducing catfish and a serious look needs to be taken at interbasin transfers of biota by engineers.  相似文献

The aim of this study was to evaluate the use of dried marine seaweed, Gracilaria arcuata for the first time as dietary ingredient in partial substitution of fishmeal on the growth performance, feed utilization and body composition of African catfish, Clarias gariepinus. Four experimental diets were formulated: D1 as a control group; D2; D3 and D4 which included 10%, 20% and 30% G. arcuata meal respectively. One hundred and eighty African catfish weighing 9.62 ± 0.42 g, (mean ± SE) was divided into four groups corresponding to the different feeding regimes. The final body weight of the fishes showed significant differences (P < 0.05) between the control (D1); D2 and other treated groups D3 and D4, with weights of 66.98, 59.60, 47.34 and 30.73 g recorded for D1, D2, D3 and D4, respectively. Significant differences (P < 0.05) were also evident in weight gain, specific growth rate, and feed utilization between treatment and control groups. However, no significant differences (P > 0.05) were observed between the control group and fishes fed D2 for all previous parameters. Protein productive value, protein efficiency ratio, daily dry feed intake and total feed intake were also significantly lower in fish fed with a diet containing G. arcuata than in the control group and D2 which contains 10% of G. arcuata. Overall, the results of the experiment revealed that African catfish fed a diet with G. arcuata included in 20% and 30% levels showed poorer growth and feed utilization than the control group and D2. However, the study recommended that C. gariepinus can accept this ingredient up to 10% in their diets. More defined experiments therefore seem to be necessary in order to determine the maximum level of this marine seaweed in diets with amino acid supplementation for African catfish.  相似文献

Transgenerational mass marking of viviparous fish larvae in vivo was validated by intra‐muscular injection of elemental strontium chloride (SrCl2) in gestating females and detection of the Sr in the otoliths of developing larvae. All otoliths of brown rockfish Sebastes auriculatus larvae produced from SrCl2‐injected females showed enriched Sr:Ca ratios near the otolith edges, and the signatures did not appear to be affected by the anterior, centre and posterior positions of larvae within the ovary. Results from the present study indicate that transgenerational marking is a highly reliable technique for marking large numbers of extremely small viviparous fish larvae.  相似文献

Summary The olfactory tract of the African catfish, Clarias gariepinus, consists of two tracts, the medial and lateral olfactory tract. Ovulated female catfish are attracted by male steroidal pheromones. Attraction tests with catfish in which the medial and lateral olfactory tract have been selectively lesioned show that the effects of these pheromones are mediated by the medial olfactory tract. The central connections of the medial and lateral olfactory tract have been studied by retro- and anterograde transport techniques using horseradish peroxidase as a tracer. Upon entering the forebrain, the medial olfactory tract innervates the posterior pars ventralis and pars supracommissuralis of the area ventralis telencephali and the nucleus preopticus periventricularis, the nucleus preopticus and the nucleus recessus posterioris. Application of horseradish peroxidase to the olfactory epithelium shows that part of the innervation of the area ventralis telencephali and the nucleus preopticus periventricularis can be attributed to the nervus terminalis, which appears to be embedded in the medial olfactory tract. The lateral olfactory tract sends projections to the same brain areas but also innervates the nucleus habenularis and a large terminal field in the area dorsalis telencephali pars lateralis ventralis. Furthermore, the medial olfactory tract carries numerous axons from groups of perikarya localized in the area dorsalis telencephali. Contralateral connections have been observed in the olfactory bulb, telencephalon, diencephalon and mesencephalon. It is suggested that processes of the medial olfactory tract innervating the preoptic region may influence the gonadotropin-releasing hormone system and in doing so may lead to behavioral and physiological changes related to spawning.  相似文献

This study investigated the long-term effects of total and partial replacement of dietary fish meal (FM) by a mixture of agricultural products on sperm quality of African catfish Clarias gariepinus. Four isonitrogenous and isoenergetic diets were formulated containing graded levels of either 50% FM and maize meal (diet 1); 25% FM mixed with crude sunflower oil cake (SFOC) and bean meal (BM) (diet 2); 12.5% FM mixed with sunflower oil cake, BM and ground nut oil cake (GOC) (diet 3) and 0% FM mixed with de-hulled sunflower oil cake (SFOCD), BM and ground nut oil cake (diet 4). Gonadosomatic index (GSI), sperm quality, plasma sex steroids (11-keto testosterone [11-KT]; testosterone [T]; estradiol-17beta [E2]) were evaluated on 10 to 24 fish fed on each diet. Sperm quality was assessed using computer-assisted sperm analysis (CASA). Total replacement of fish meal by plant products markedly increased sperm volume, spermatocrit, spermatozoa integrity, and sperm motility. Fish fed diet 3 (12.5% fish meal) provided intermediate results on sperm quality whereas the lowest values were obtained in fish fed diets 1 and 2. In fish fed 0% fish meal (diet 4), androgen levels were higher and estrogen levels were lower than in fish fed fish meal diets. Based on dietary lipid and fatty acid analyses, these results suggest a positive impact of short chain n-6 fatty acids on androgen synthesis and sperm quality. In conclusion, a combination of ground nut oil cake, bean meal and sunflower oil cake (preferably when the sunflower is dehulled) in African catfish diet improves the sperm quality.  相似文献

African catfish, Clarias gariepinus (B.), is one of the promising freshwater fish species in African aquaculture but the expansion of its farming needs more production of its larvae. The use of live food organisms at first feeding for larvae is still obligatory. That increases the cost of larvae production. Hence, the incorporating of exogenous enzymes especially protease in artificial microdiets may provide affordable alternatives for enhancing the larvae performance. The present study was carried out to evaluate the growth and survival of larvae or fingerlings of African catfish fed artificial diets incorporated with different protease levels. Four artificial diets were formulated and enriched with protease enzyme at levels of 0.0, 750, 1,000, and 1,250 unit/kg diet; after that diets were made into crumbles (100–200 µm diameter). After absorption of the yolk sac, diets were offered to fish larvae (3.6 ± 0.2 mg) in triplicates as a starter feed up to apparent satiation every two hours for 30 days. In another treatment, fish larvae were fed on newly hatched Artemia nauplii (2,500 Artemia/L) as a starter food. In another experiment, African catfish fingerlings (10.1 ± 1.6 g) were fed on the same diets up to satiation twice a day for 2 months. It was noticed that the dietary protease improved larval growth and survival but not as Artemia nauplii did where fish larvae fed on Artemia nauplii showed highest growth and survival followed by those fed a diet enriched with 1,250 unit/kg diet of protease. The mortality of larvae fed protease‐enriched diets as well as the control diet was occurred mostly at the first week reaching its maximum at the third week. The poor growth was observed with fish larvae fed the control diet. Meanwhile, catfish fingerlings fed protease‐enriched diets showed higher growth over those fed the control diet. The larvae survival (11.0%–41.7%) was enhanced by increasing protease levels and it was lower than that of fingerlings (95.6%–100.0%). Furthermore, protein retention and digestibility were significantly improved with protease supplementation over the control diet especially at a level of 1,000 unit/kg diet. As compared with the previous studies, live food should be used in larvae rearing for the first week after that a starter diet enriched with protease at levels of 1,250 unit/kg diet should be used. In case of fish fingerlings, the dry diets should be enriched with 1,100 unit/kg diet to improve diet digestibility and subsequently enhance their growth.  相似文献

Wild adult specimens of the Peruvian anchovy Engraulis ringens were captured and reared to validate the daily periodicity of otolith microincrement formation. The postcapture stress generated spontaneous spawning, making it possible to conduct a rearing trial on larvae first in an artificial nutrient‐enriched system (ANES) for 52 days followed by an artificial feeding regime in a culture tank until day 115 post‐hatch. Microincrements of the sagittal otoliths of sacrificed juveniles [mean ± s.d. total length (LT) = 5·13 ± 0·37 cm, range 5–6 cm; c.v. = 7·5%] showed very distinct light and dark zones. The slope of the relationship between the total number of increments after the hatch check and days elapsed after hatching was not significantly different from 1. The transfer from ANES to the artificial feeding regime induced a mark in the sagittal otoliths. The number of microincrements after this induced mark coincided with the number of days elapsed after the transfer date. In parallel experiments, adult E. ringens (mean ± s.d. LT = 14·92 ± 0·55 cm, range 13–16 cm) were exposed to one of two fluorescent marking immersion treatments with either alizarin red S (ARS; 25 mg l?1 per 6 h) or oxytetracycline hydrochloride (OTC; 200 mg l?1 per 10 h). The microincrements between fluorescent bands were distinct, ranging from 0·89 to 2·75 µm (mean ± s.d. =1·43 ± 0·28 µm; c.v. = 32%) and from 0·71 to 2·89 µm (1·53 ± 0·27 µm; c.v. = 35%) for ARS and OTC, respectively. The relationship between the number of microincrements between marks and the number of elapsed days for ARS and OCT treatments indicated that there was a significant correspondence between the number of increases observed and the number of days. Hence, daily microincrements of otoliths of E. ringens are likely to be formed in juveniles and adults under natural conditions.  相似文献

Determining the genetic characteristics of natural fish stocks is useful for conservation and aquaculture programs. For African catfish, Clarias gariepinus, genetic characterization could help identify populations suitable as brood stock for culture, and those in need of conservation. This study determined the genetic diversity, population structure, and demographic history of C. gariepinus from Lakes Victoria (LV), Kenyatta (LKE), Kamnarok (LKA), and Rivers Nyando (NR), Tana (TR) and Sosiani (SR) in Kenya. Using 128 DNA sequences of D-loop control region, 34 haplotypes were recovered, of which 79.4% were singletons. Only 7 haplotypes were shared between sites, implying little gene flow between sites. Number of haplotypes was highest in LKE and NR populations and lowest in SR. Haplotype diversity was highest in LV, and lowest in SR, while, nucleotide diversity was highest in LKA and lowest in LV. Phylogenetic analyses revealed five clusters: Lakes Victoria, Kamnarok and Kenyatta, and Rivers Tana and Nyando, from both maximum likelihood tree and minimum spanning network. This, together with significant F ST values among the sites imply population differentiation. Mismatch distributions were multi-modal in LKA, LKE, NR and TR, signifying demographic equilibria. Neutrality tests Tajima`s D values for the sampled populations were negative and significantly different, suggesting stable populations. These results show the existence of genetically distinct populations of C. gariepinus that require spatially explicit management actions such as reducing fishing pressure, pollution, minimizing habitat destruction and fragmentation for sustainable utilisation of stocks.  相似文献

Nine groups each of four fish were injected with a single intramuscular dose of the following preparations: Physiological saline (0.9% NaCl) as a control group, 0.5 ml kg−1 Ovaprim, 20 and 40 μg kg−1 BW of GnRHa, 8 and 16 mL kg−1 pimozide tablets and the following combination of GnRHa with pimozide (GP): 20 μg + 4 mg, 30 μg + 8 mg and 40 μg + 16 mg kg−1 BW. The primary oocyte diameter (POD) before hormone administration ranged from 943.3 to 1071.0 μm. The latency periods (LP) were in the range of 9.0 to 12.0 h after injection. The highest ovulation ratio (OR) was observed in groups Ovaprim, GP(30 + 8) and GP(40 + 16). Other treatments were effective for ovulation, the ovulation ratio in Groups G(40) and GP(20 + 4) were significantly higher than G(20) treatment. The ovulation index (OI) was in the range 62 to 77% and showed significant differences among groups. There was no significant difference in fertilization ratio (FR) among Ovaprim, GP(30 + 8) and GP(40 + 16) groups, while there were significant difference between the previous group and G(20) and G(40) groups. Control, P8, P16 showed negative results in all the parameters LP, OED, OR, OI and FR. Levels of sex steroids were analyzed on 6 and 12 h after initiation of treatments. A significant increase in plasma E2 with GP(30 + 8) injection was observed 6 and 12 h after injection, while there were no significant increase between all the other groups 6 h after injection. Treatments with GP(20 + 4) resulted in a significant increase in plasma T concentration in females compared with control after 6 h. In contrast, plasma T and E2 concentrations were lower during the combined GP(20 + 4), GP(30 + 8) and GP(40 + 16) after 12 h than after 16 h of injection. The combined treatments (GnRHa + PIM) are better compared with Ovaprim which gave the same results, they have some advantages, such as reliable response and low cost. Ovaprim is more than 3 to 5-fold of the cost of (GnRH + PIM). Therefore, this method could be useful tool for commercial catfish breeders to ensure spawning success.  相似文献

A study was undertaken to examine the effect of different amounts of dietary lysine (13 and 21 g kg?1 diet), lipid (80 and 160 g kg?1 diet) and L ‐carnitine (0.2 and 1.0 g kg?1 diet) on growth performance, proximate composition and amino acid metabolism of the African catfish (Clarias gariepinus). Juvenile African catfish (23 ± 1.5 g/fish) were stocked into 70‐L aquaria (16 aquaria, 28 fish/aquarium) connected to a recirculation system during a maximum period of 74 days. All groups were fed at a level of 24 g kg?0.8 day?1 in an experiment run at pair feeding. Animals receiving 1.0 g carnitine accumulated up to six times more carnitine in their tissues than animals receiving 0.2 g (P < 0.05). Acyl‐carnitine and free L ‐carnitine levels increased in the whole body and in tissues. Dietary L ‐carnitine supplements increased protein‐to‐fat ratios in the body, but did not affect growth rate. Protein‐to‐fat ratios were only affected when the biosynthesis capacity of L ‐carnitine was restricted due to low lysine levels and when there was a shortage of dietary fat. When lysine was offered at 21 g kg?1 feed, dietary L ‐carnitine supplements did not affect the amino acid concentrations of body tissues. Dietary L ‐carnitine supplements raised the concentration of glutamic acid > aspartic acid > glycine > alanine > arginine > serine > threonine in skeletal muscle tissue (P < 0.05). Total amino acid concentration in muscle and liver tissues (dry‐matter basis) increased from 506 to 564 and from 138 to 166 mg g?1, respectively, when diets were offered with high L ‐carnitine, low lysine and low fat levels. These data suggest that dietary L ‐carnitine supplementation may increase fatty acid oxidation and possibly decrease amino acid combustion for energy.  相似文献
