The phylogenetics of Sternbergia (Amaryllidaceae) were studied using DNA sequences of the plastid ndhF and matK genes and nuclear internal transcribed spacer (ITS) ribosomal region for 38, 37 and 32 ingroup and outgroup accessions, respectively. All members of Sternbergia were represented by at least one accession, except S. minoica and S. schubertii, with additional taxa from Narcissus and Pancratium serving as principal outgroups. Sternbergia was resolved and supported as sister to Narcissus and composed of two primary subclades: S. colchiciflora sister to S. vernalis, S. candida and S. clusiana, with this clade in turn sister to S. lutea and its allies in both Bayesian and bootstrap analyses. A clear relationship between the two vernal flowering members of the genus was recovered, supporting the hypothesis of a single origin of vernal flowering in Sternbergia. However, in the S. lutea complex, the DNA markers examined did not offer sufficient resolving power to separate taxa, providing some support for the idea that S. sicula and S. greuteriana are conspecific with S. lutea. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 166 , 149–162.  相似文献

Abstract

A study of Sternbergia lutea Ker-Gawl lipidsoluble pigments during some phases of its biological cycle. — In the present paper we have examined the pigments of Sternbergia lutea and their variations during some particular phases of its biological cycle in order to obtain useful information on their probable function in the plant. Therefore, we have analyzed the pigments of the leaves, floral scape, tepals and anthers. The results obtained show that Sternbergia lutea is characterized by the same pigment composition as that of the other higher plants. In addition to this, three further carotenoids have been observed (mainly in the tepals and anthers). Chromatographic and spectrophotometric properties of these new pigments indicate their relationship with carotene isomers that have already been examined. During the phases of vegetative cycle of Sternbergia lutea chlorophylls and carotenoids increase concomitantly with the growth of the plant. Carotenes reach a significant maximum value only in anthers. In the course of the necrosis (that obviously occurs in different stages for the various organs) is observed a higher decrease of chlorophylls (in the green tissues) and of the carotenes (in the anthers) while lutein and violaxanthin increase greatly (in the tepals).  相似文献

In continuation of our research program on Mediterranean dietary plants, a bioassay‐guided fractionation of extracts from several accessions of Capparis sicula subsp. sicula and Capparis orientalis aerial parts was carried out. Antilipidemic activity of samples was assayed using inhibition of pancreatic lipase. To study the metabolic variability in Capparis species, HPTLC analyses were performed in order to characterize the species through the detection, isolation, and quantitative evaluation of rutin taken as significant chemical marker. The best activity was exerted by C. orientalis accession no. C10 and C. sicula subsp. sicula accession no. C6. The bioactivity evaluation of specific chemical markers, rutin and glucocapparin, led to the identification of a potent antilipidemic compound rutin. The HPTLC analysis showed large variation among the different analyzed samples with respect to rutin concentration. The chemical investigation showed a different composition between the species and between the collection zones. The variations showed by the studied accessions of caper could be attributed to exogenous factors. Capparis species contained predominantly quercetin rutinoside (rutin), accompanied by other constituents such as the glucosinolate glucocapparin. These rutin‐rich extracts exhibited pronounced dose‐dependent enzyme inhibitory activities toward pancreatic lipase.  相似文献

Three species of boid snakes are recognized in Madagascar, namely the genus Sanzinia (one species and two subspecies) and the genus Acrantophis (two species). In the present study, we studied the patterns of genetic variation of these species across Madagascar using a fragment of the mitochondrial 16S rRNA gene in 77 specimens. To support the phylogenetic relationships of the lineages identified, three further gene fragments (cytochrome b, 12S rRNA and c‐mos) were analyzed in a reduced but representative set of samples. The results obtained corroborate that the genus Sanzinia includes two highly divergent mitochondrial lineages that evolved independently from each other on the east versus the west side of Madagascar. Each of these lineages presents a further subdivision that separates northern from southern groups. The nuclear marker showed no variation among the Malagasy boas, indicating either very low substitution rates in this gene or relatively recent speciation events coupled with high mitochondrial substitution rates. Because the broad geographic sampling detected no admixture among haplotypic lineages within Sanzinia, it is hypothesized that these may represent distinct species. Ex situ cultivation in botanic gardens could be one possibility to preserve plant species diversity and genetic variation. However, old ex situ populations are often sparsely documented. We were able to retrieve three different ex situ populations and their source in situ populations of the endangered plant species Silene otites after 20–36 years of isolation. Furthermore, three additional wild populations were included in the analysis. Population genetic diversity and differentiation were analysed using AFLP markers. Genetic variation in the ex situ populations was lower than the variation found in the in situ populations. Strong differentiation (FST = 0.21–0.36) between corresponding in situ and ex situ populations was observed. Bayesian clustering approach also showed a distinct genetic separation between in situ and ex situ populations. The high genetic differentiation and loss of genetic diversity during spatial and temporal isolation in the ex situ populations can be attributable to small population sizes and unconscious selection during cultivation. Therefore, adequate sampling prior to ex situ cultivation and large effective population sizes are important to preserve genetic diversity. Near‐natural cultivation allowing for generation overlap and interspecific competition without artificial selection is recommended as being best for the maintenance of the genetic constitution. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, ??, ??–??.  相似文献

The perennial herbs Silene vulgaris and S. uniflora are closely related, partially sympatric and interfertile, yet morphologically distinct. We used nuclear (allozyme) and plastid (polymerase chain reaction–restriction fragment length polymorphism) DNA markers to investigate whether these species have a shared history of postglacial colonization and hybridization in Europe, as inferred from large‐scale patterns of geographic variation. The majority of plastid haplotypes and allozyme alleles were widespread and patchily distributed within both species and there was no geographic structure in the distributions of shared allozymes or haplotypes. The mosaic variation is consistent with a scenario in which repeated episodes of interspecific hybridization pre‐dated the largely allopatric range expansion of the two species during the postglacial period. Our overall results are not consistent with a scenario of extensive hybridization and introgression during the postglacial range expansion of the species or within their current areas of sympatry, but we found some evidence for local, postglacial evolution and hybridization in the Baltic region. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 166 , 127–148.  相似文献

The taxonomy and nomenclature of the genus Huperzia Bernh. in the Azores and Madeira have been reviewed. Plants collected in the Azores and Madeira were characterized morphologically. The independence between two endemic species common to Madeira and the Azores Islands –Huperzia suberecta (Lowe) Tardieu and Huperzia dentata (Herter) Holub – is clearly shown. A clear‐cut morphological separation between these taxa and Huperzia selago (L.) DNA restriction endonuclease fragment analysis was used to obtain new information on the genomic organization of ribosomal DNA (rDNA) of Brassica and allied genera. The total genomic DNA of 95 accessions of 52 species representing 16 genera was restricted with six enzymes, and the restriction fragments were probed with three ribosomal clones (pTA71, Ver 18‐6, and Ver 6‐5). Eleven repeat unit length classes were recognized. The repeat unit size classes of 8.9 kb and 9.5 kb were observed most commonly, being represented in 17 and 14 species, respectively. The restriction enzyme SacI produced three to six (generally three) bands with detectable hybridization to the probe pTA71. This probe–enzyme combination indicated a remarkable uniformity amongst Brassica and allied genera in the coding region of repeat units. By contrast, an extensive size variation in the restriction fragments could be localized in the intergenic spacer (IGS) region. Eleven IGS‐containing length variants were detected. Complex hybridization patterns, resulting from extensive repeat unit heterogeneity and taxon‐specific methylation of one or more cleavage sites, were obtained with the EcoRI + pTA71 combination. The relative homologies between the coding regions were evident from the presence of 1.5 kb in all the taxa, and 0.4‐, 1.3‐, and 1.7‐kb fragments in 33, 27, and 24 species, respectively. The SacI + pTA71 and EcoRI + pTA71 combinations were generally able to distinguish taxa both within and between genera. Three restriction endonuclease digests probed with three ribosomal clones yielded essentially identical fragmentation patterns across all the accessions within the cultivated species Brassica campestris, B. oleracea, and B. juncea. In B. napus, three and seven accessions exhibited restriction profiles similar to one and both diploid progenitor species, respectively. A new species of Gesneriaceae, Paraboea trisepala W.H.Chen & Y.M.Shui, from a karst cave in Guangxi, China is described and illustrated. The new species differs from other species of Paraboea by its three‐lobed calyx. Heterostyly is a genetic polymorphism in which plant populations possess two (distyly) or three (tristyly) morphs with flowers differing reciprocally in stigma and anther height. Sex organ deployment has been described as being highly variable among and within species of several distylous taxa belonging to different taxonomic groups. However, the number of studies considering within‐species disparities is still limited. For a better overview of the existing amount of variation that can occur within and between heterostylous species, we sampled 46 populations of six Melochia spp., a style‐polymorphic genus in Cuba. We characterized the floral morphology in all populations and described a set of ancillary characters per species. All of these Melochia spp. are distylous, except for the monomorphic M. nodiflora. The S‐morph produces fewer, larger pollen than L‐morphs, and has verrucose ornamentation. The L‐morph produces reticulate pollen and has larger stigmatic papillae than the S‐morph. The monomorphic M. nodiflora shows ancillary characteristics that are similar to the L‐morph individuals in the related species. As expected, there are differences in ancillary characters among species and also dissimilarities in reciprocity among and within species of Melochia. 1. Spatial variation of methane (CH4) efflux from the littoral zone of a meso‐eutrophic boreal lake was studied with a closed‐chamber technique for three summer days in 22 vegetation stands, consisting of three emergent and three floating‐leaved species. 2. Between‐species differences in CH4 emission were significant. The highest emissions were measured from the emergent Phragmites australis stands (0.5–1.7 mmol m?2 h?1), followed by Schoenoplectus lacustris > Equisetum fluviatile > Nuphar lutea > Sparganium gramineum > Potamogeton natans. Within‐species differences between stands were not significant. 3. In P. australis stands, the stand‐specific mean CH4 emission was significantly correlated with solar radiation, probably indicating the role of effective pressurised ventilation on CH4 fluxes. The proportion of net primary production emitted as CH4 was significantly higher in P. australis stands (7.4%) than in stands of S. lacustris and E. fluviatile (both 0.5%). 4. In N. lutea stands, CH4 efflux was negatively correlated with the mean fetch and positively with the percentage cover of leaves on the water surface. There were no differences in CH4 efflux between intact N. lutea leaves and those grazed by coleopteran Galerucella nymphaeae. In S. graminaeum and P. natans stands, CH4 effluxes were not related to any of the measured environmental variables. 5. For all vegetation stands, the biomass above water level explained about 60% of the observed spatial variation in CH4 emission, indicating the important role of plants as gas conduits and producers of substrates for methanogens in the anoxic sediment.  相似文献

The taxonomy of the endemic arboreal snail Satsuma albida species complex from Taiwan was unclear due to the animals' highly similar morphology, and their nocturnal and strict arboreal behaviour, leading to difficulties in collecting living specimens. This article is the first comprehensive comparative study on the systematics and taxonomy of this species complex using external morphology, anatomy of the reproductive system and molecular phylogeny. Consequently, two subspecies of S. albida are raised to species status, namely S. insignis and S. mollicula. Fourteen new species are also described. Fourteen of the 17 species showed polymorphism in banding pattern amongst populations and other species retained the whitish unity as seen in S. albida. Distributions of almost all taxa are geographically limited, with the exception of S. polymorpha sp. nov . The phylogeny of these species was reconstructed using 20 morphological characters and molecular data from the partial sequences of mtDNA CO1 and 16S rRNA genes, and the complete ITS2 sequence. Superficial similarities among unrelated species are often a result of convergent evolution and can cause considerable taxonomic confusion. A case in point is Satyrium eurycalcaratum , described here as a new species, which has been confused with several other Satyrium spp. with similar long‐spurred, white flowers. A phylogenetic analysis, based on molecular data, indicated that S. eurycalcaratum is not closely related to any of the species with which it has been previously confused. A comparative analysis of morphological characters in the seven South African Satyrium spp. with long‐spurred, white flowers showed that each of these, including S. eurycalcaratum , is characterized by a unique combination of traits. Despite the similarity in pollination syndrome characters, such as spur length and flower colour, variation in rostellum structure was particularly pronounced and four distinctive forms were present. There was no phylogenetic signal in patterns of interspecific rostellum variation, as some closely related species had different rostella, whereas some distantly related species shared similar rostellum structures. Extreme variation in early life‐history strategies is considered a moderately good predictor of genetic subdivision and hence dispersal for a range of marine species. In reality, however, a good deal of population differentiation must reflect historical effects, more subtle variation in life histories, and, particularly, the interaction of larvae with oceanographic processes. Using a combination of allozyme and microsatellite markers, we show that the large‐scale genetic structure of populations of three species (direct and planktonically developing cushion stars and a planktonic developing sea anemone that is also asexually viviparous) varies consistently, in line with the predicted capacity for dispersal within three geographic regions. We detected high levels of genetic subdivision for the direct developing cushion star (FST = 0.6), low levels for the planktonically developing cushion star (FST = 0.009), and intermediate levels for the sexual/asexual sea anmone (FST = 0.19). These patterns are exhibited despite the highly variable patterns of current movement and the presence of biogeographic barriers. Our results suggest that, although there is large scale genetic differentiation for two species, patterns of population connectivity are remarkably consistent within major regions and do not reflect variation in major oceanographic processes or genetic discontinuity coincident with biogeographic boundaries. © 2008 The Linnean Society of London, Biological Journal of the Linnean Society, 2008, 95 , 106–116.  相似文献

The present study aimed to infer evolutionary scenarios for Vipera latastei and Vipera monticola in the Iberian Peninsula and the Maghreb through the identification of spatial patterns in morphological character variation and biogeographic patterns in morphological variability distribution. Ten morphological traits from 630 vipers were analysed with geostatistic and ecological niche modelling in a geographical information system. Interpolation by Kriging was used to generate surfaces of morphological variation, which were combined with spatial principal components analysis (SPCA). Putative morphological differentiated groups generated by SPCA maps were tested with discriminant function analysis (DFA). Maximum entropy modelling and nine environmental variables were used to identify factors limiting the distribution of groups and areas for their potential occurrence. Groups supported by DFA were: Western Iberia, Eastern Iberia, Rif plus Middle Atlas, Algeria, and High Atlas. Their distribution is influenced by common environmental factors such as precipitation. Areas of probable sympatry between Iberian groups matched the morphological clines observed by geostatistics tools. Geographic variation patterns in V. latastei‐monticola are probably due to vicariant separation of Iberian and African populations during the opening of the Strait of Gibraltar, and population refugia during the Quaternary glaciations with secondary contact. The taxonomic status of northern Morocco and Algerian groups should be further investigated.
