Rapid drying to baling moisture reduces weather-related risks when harvesting perennial grasses to be used as biomass feedstocks. Drying switchgrass can be challenging due to large, dense swaths and unfavorable autumn drying conditions, so new methods to improve drying are needed. Two techniques to enhance switchgrass drying rate were investigated, which are intensive conditioning and wide-swath drying. Intensive conditioning involved rolls that crushed the stem along its length accompanied by differential roll speed to disrupt the waxy epidermis of the stem. Wide-swath drying involved a post-conditioning tedding operation that distributed the crop across most of the cut width. A mower-tractor-intensive conditioner-tedder combination was configured so that intensive conditioning, wide-swath drying, or both could be achieved in a single pass. Although not consistent across all trials, intensive conditioning and wide-swath drying were about equally effective at improving switchgrass drying rate compared to conventional conditioning alone. However, the combination of intensive conditioning followed by wide-swath drying consistently resulted in the most rapid drying rate. In eight separate trials, this combination produced switchgrass moisture contents well below 20% (w.b.) on the second or third day after cutting. An empirical model of switchgrass drying rate was developed, which can be used to predict switchgrass drying time based on weather conditions, conditioning level, and swath density.  相似文献

Diet specialization in the trophically polymorphic cichlid fish Herichthys minckleyi was examined using gut contents. Individual H. minckleyi were categorized as having molariform, papilliform or undetermined pharyngeal jaws. The presence of enlarged flattened pharyngeal jaw teeth was used to categorize H. minckleyi as molariform, and the possession of only small pencil‐like pharyngeal teeth was used to classify fish as papilliform. Undetermined individuals (<50 mm standard length, L S) were not assigned to one of the two larger morphotypes. Arthropods were found to be generally rare in H. minckleyi gut contents, but when present, they were most frequently recovered from undetermined individuals. The percentage of plant material consumed by undetermined H. minckleyi was not as great as papilliforms ingested on average, and snail crushing by undetermined H. minckleyi was not evident. A significantly greater mean percentage of plant detritus was recovered from papilliforms compared to molariforms. Snails were crushed by molariforms more frequently than by papilliforms. When only molariforms and papilliforms that had crushed snails were compared, a greater number of snails were crushed by molariforms. No relationship was found between molariform L S and the number of snails crushed, but greater molariform tooth number, adjusted for L S, was indicative of recent snail crushing. The maintenance of H. minckleyi pharyngeal jaw variation could be promoted by intraspecific diet differentiation.  相似文献

Tissues of the hollow organs of the gastrointestinal tract are crushed in order to create an anastomosis. The processes taking place in the tissues have been inadequately studied. A model of the tissue crushing process is suggested and the results of theory and experiments are compared. Dependence of the rejection time of the crushing influence on the crushing force was demonstrated experimentally. The relationship obtained can be explained on the grounds that migration of cell destruction products from the zone of crushing to the zones of healing stimulates adequate cellular immunologic processes. The views developed in this paper may prove useful for the elaboration of new surgical techniques.  相似文献

Summary This paper considers prey size selection by four molluscivorous cichlids feeding on the intermediate host snail of Schistosoma parasites, Biomphalaria glabrata. Haplochromis ishmaeli obtains its prey by crushing the snails between the pharyngeal jaws, whereas H. xenognathus, H. sauvagei and Macropleurodus bicolor apply both pharyngeal crushing and oral shelling. The fishes crushed significantly more snails with the highest reward in biomass per second of crushing. Oral shelling occurred far less often than pharyngeal crushing. Encounter rates with prey showed significant variations between different size classes of prey. The fish have no overall knowledge of snail availability in a tank. The probability that a snail will be eaten at encounter, calculated from the number encountered and the number eaten, reflects the prey size preference of the fish. Those snails with the highest biomass/crushing-time ratio had the highest probability of being crushed; observed and predicted prey size preferences corresponded well. Although for oral shelling the potential reward in biomass per second is of the same magnitude as for crushing, the probability of successful shelling is very low. Apparently the fish prefer prey with lowest risks.  相似文献

Soil aggregates strongly influence C dynamics by affecting microbial activity. Our study tested the effect of soil crushing on C mineralization by laboratory incubation experiments of soil samples from a tropical deciduous forest ecosystem in Western Mexico. Soil samples were taken in January (dry season) and in October (rainy season). For each sampling date, the incubation experiment had a two factorial design with litter and macroaggregates (>250 μm) crushing as the main factors, both with two levels (with and without). At both sampling dates, the soil samples with intact macroaggregates had significantly higher C mineralization than the soil samples in which macroaggregates were crushed. The pH of leached solution was higher in the crushed soil samples than in uncrushed soil samples. The reduction of C mineralization caused by the disruption of soil aggregates is explained by the disturbance of environmental conditions within macroaggregates. The effect of macroaggregates crushing also reduced the differences of C mineralization between both seasons. We concluded that macroaggregates promote microbial activity by reducing the impact of variations in soil chemical and physical environmental conditions.  相似文献

The purpose of this research was to evaluate the influence of dry granulation parameters on granule and tablet properties of spray-dried extract (SDE) fromMaytenus ilicifolia, which is widely used in Brazil in the treatment of gastric disorders. The compressional behavior of the SDE and granules of the SDE was characterized by Heckel plots. The tablet properties of powders, granules, and formulations containing a high extract dose were compared. The SDE was blended with 2% magnesium stearate and 1% colloidal silicon dioxide and compacted to produce granules after slugging or roll compaction. The influences of the granulation process and the roll compaction force on the technological properties of the granules were studied. The flowability and density of spray-dried particles were improved after granulation. Tablets produced by direct compression of granules showed lower crushing strength than the ones obtained from nongranulated material. The compressional analysis by Heckel plots revealed that the SDE undergoes plastic deformation with a very low tendency to rearrangement at an early stage of compression. On the other hand, the granules showed an intensive rearrangement as a consequence of fragmentation and rebounding. However, when the compaction pressure was increased, the granules showed plastic deformation. The mean yield pressure values showed that both granulation techniques and the roll compaction force were able to reduce the material's ability to undergo plastic deformation. Finally, the tablet containing a high dose of granules showed a close dependence between crushing strength and the densification degree of the granules (ie, roll compaction force). Abortion was induced in 91 cows and heifers by three manual methods applied perrectum. The three methods were: (1) rupture of the amnion and crushing of the fetus in cows pregnant 35 to 66 days, (2) decapitation of the fetus in cows 66 to 120 days into gestation, and (3) rupture of the amnion without crushing the fetus in cows pregnant less than 70 days. All the cows aborted between 10 and 54 days after treatment except three out of the 29 in the group in which the amnion was ruptured but the fetus not crushed. The mean interval (21.5 days) from treatment to abortion was shorter (P ~-.07) when fetal decapitation was done between 70 and 120 days of gestation compared to the interval (26.5 days) when the amnion was ruptured and the fetus crushed between 35 and 66 days of gestation. The average interval from treatment to estrus with amnion rupture was 38 days; with fetal decapitation it was 32 days (P>.10). After abortion, estrus occurred in an average of 15 days in the amnion rupture group and in 12 days in the fetal decapitation group (P>.10).  相似文献

AIM: To study the influence of different methods of grape treatment in wineries on the diversity of the yeast species in spontaneous fermentations. METHODS AND RESULTS: Grapes were crushed and pressed in three different ways followed by spontaneous fermentation. The same grape material picked and crushed aseptically directly in the vineyard served as control. Yeasts isolated at different stages of the fermentation were characterized by 5.8S-ITS-RFLP. Yeasts of the Saccharomyces sensu stricto complex were additionally analysed by microsatellite polymerase chain reaction fingerprinting. The diversity of yeast species isolated from winery fermentations was much greater than from the vineyard fermentation in respect to yeasts of the genus Saccharomyces as well as non-Saccharomyces. CONCLUSIONS: Oenonogical methods alter significantly the yeast diversity in spontaneous fermentations of grape juice. SIGNIFICANCE AND IMPACT OF THE STUDY: Managing spontaneous fermentations successfully depends not only on choosing the suitable grapes but also on the crushing and pressing techniques leading to different yeast populations.  相似文献

Experiments with five samples of corn straw were carried out on a one-dimensional bench combustion test rig. The bed temperature distribution and the mass loss of fuel and gas components such as O2, CO, CO2 and NO were measured in the bed. The combustion of corn straw occurred in two stages, ignition front propagation and char oxidation. The average burning rate increased with an increase in the primary air flow until a critical point was reached, beyond which a further increase in the primary air flow resulted in a decreased burning rate. The mean concentration of NO reached a minimum value and then increased with increased primary air flow. The time taken for the drying front to reach the bottom of the bed was 800 s, 700 s, and 500 s; the temperatures in the high bed temperature zones were 900-935 degrees C, 800-850 degrees C and 700-743 degrees C; and the maximum concentrations of NO were 725 ppmv, 1287 ppmv, and 2730 ppmv, for whole corn stalks, hollow corn stalks and flaked corn stalks, respectively. The maximum concentrations of CO and NO were quite different between samples. There was only one peak in the distribution of NO concentration for sample B, but there were two peaks for whole corn stalks and sample A.  相似文献

The proximo-distal intra-axonal transport of acetylcholine (ACh) and cholinergic enzymes (choline acetyltransferase, CAT, and ACh-esterase, AChE) in rat regenerating sciatic nerve was studied by accumulation technique. Four types of axonal trauma were performed: freezing with solid CO2, crushing, ligating the nerve with remaining tight silk ligature, and cutting the nerve. Normal and sham-operated rats were used as controls. One to twenty-nine days later, the nerves were crushed about 15 mm proximal to the trauma. The nerve segment proximal to this crush was dissected out 12 hr later and assayed for ACh-content and enzyme activities. The increase in this segment 12 hr after crushing was taken as an indication of proximo-distal transport in the regenerating nerves. ACh transport did not seem to vary during regeneration as compared to controls. In contrast, the transport of both CAT and AChE was initially markedly depressed. Towards the end of the observation period (29 days), a recovery of CAT-transport occurred in all groups. Recovery of AChE-transport was marked in the freeze and crush groups. In the cut group no recovery was seen and in the ligated group only a small recovery occurred. Thus, in the nerves where regeneration was facilitated by the presence of intact connective tissue sheaths (freezing and crushing) recovery of transport occurred earlier than in cut or ligated nerves.  相似文献

The rising demand for bioethanol, the most common alternative to petroleum-derived fuel used worldwide, has encouraged a feedstock shift to non-food crops to reduce the competition for resources between food and energy production. Sweet sorghum has become one of the most promising non-food energy crops because of its high output and strong adaptive ability. However, the means by which sweet sorghum stalks can be cost-effectively utilized for ethanol fermentation in large-scale industrial production and commercialization remains unclear. In this study, we identified a novel Saccharomyces cerevisiae strain, TSH1, from the soil in which sweet sorghum stalks were stored. This strain exhibited excellent ethanol fermentative capacity and ability to withstand stressful solid-state fermentation conditions. Furthermore, we gradually scaled up from a 500-mL flask to a 127-m3 rotary-drum fermenter and eventually constructed a 550-m3 rotary-drum fermentation system to establish an efficient industrial fermentation platform based on TSH1. The batch fermentations were completed in less than 20 hours, with up to 96 tons of crushed sweet sorghum stalks in the 550-m3 fermenter reaching 88% of relative theoretical ethanol yield (RTEY). These results collectively demonstrate that ethanol solid-state fermentation technology can be a highly efficient and low-cost solution for utilizing sweet sorghum, providing a feasible and economical means of developing non-food bioethanol.  相似文献

Lignocellulosic wastes, including corn stalks and wheat straw, were pretreated and hydrolyzed with combined supercritical and subcritical hydrothermal technology. Soluble sugars were collected by pre-washing the crushed materials before hydrolysis. The effects of solid–liquid ratio, temperature, and reaction time on oligosaccharide production were investigated and the optimum supercritical conditions were found to be 20 mg/2.5 ml water, 384 °C, 17 s for corn stalks and 20 mg/2.5 ml water, 384 °C, 19 s for wheat straw. Subsequent subcritical processing of the hydrolyzate (with or without the water extract) from supercritical treatment was guided by a previous analysis of cellulose hydrolysis kinetics. The highest yield of fermentable hexoses from corn stalks (27.4% of raw material) was obtained at 280 °C, 27 s, and from wheat straw (6.7% of raw material) at 280 °C, 54 s. This study provides novel key parameters for fermentable hexose production from lignocellulosic feedstocks using combined supercritical and subcritical hydrothermal treatment.  相似文献

Inducible defences are widely used for studying phenotypic plasticity, yet frequently we know little about the cues that induce these defences. For aquatic prey, defences are induced by chemical cues from predators (kairomones) and injured prey (alarm cues). Rarely has anyone determined the separate and combined effects of these cues, particularly across phylogenetically diverse prey types. We examined how tadpoles (Hyla versicolor) altered their defences when 10 different prey were either crushed by hand or consumed by predators. Across all prey types, crushing induced only a subset of the defences induced by consumption. Consuming vs. crushing produced additive responses for behaviour but synergistic responses for morphology and growth. Moreover, we discovered the first extensive evidence that prey responses to different alarm cues depends on prey phylogeny. These results suggest that the amount of information available to the prey affects both the quantitative and qualitative nature of the defended phenotype.  相似文献

Background

Sweet sorghum is regarded as a very promising energy crop for ethanol production because it not only supplies grain and sugar, but also offers lignocellulosic resource. Cost-competitive ethanol production requires bioconversion of all carbohydrates in stalks including of both sucrose and lignocellulose hydrolyzed into fermentable sugars. However, it is still a main challenge to reduce ethanol production cost and improve feasibility of industrial application. An integration of the different operations within the whole process is a potential solution.

Results

An integrated process combined advanced solid-state fermentation technology (ASSF) and alkaline pretreatment was presented in this work. Soluble sugars in sweet sorghum stalks were firstly converted into ethanol by ASSF using crushed stalks directly. Then, the operation combining ethanol distillation and alkaline pretreatment was performed in one distillation-reactor simultaneously. The corresponding investigation indicated that the addition of alkali did not affect the ethanol recovery. The effect of three alkalis, NaOH, KOH and Ca(OH)2 on pretreatment were investigated. The results indicated the delignification of lignocellulose by NaOH and KOH was more significant than that by Ca(OH)2, and the highest removal of xylan was caused by NaOH. Moreover, an optimized alkali loading of 10% (w/w DM) NaOH was determined. Under this favorable pretreatment condition, enzymatic hydrolysis of sweet sorghum bagasse following pretreatment was investigated. 92.0% of glucan and 53.3% of xylan conversion were obtained at enzyme loading of 10 FPU/g glucan. The fermentation of hydrolyzed slurry was performed using an engineered stain, Zymomonas mobilis TSH-01. A mass balance of the overall process was calculated, and 91.9 kg was achieved from one tonne of fresh sweet sorghum stalk.

Conclusions

A low energy-consumption integrated technology for ethanol production from sweet sorghum stalks was presented in this work. Energy consumption for raw materials preparation and pretreatment were reduced or avoided in our process. Based on this technology, the recalcitrance of lignocellulose was destructed via a cost-efficient process and all sugars in sweet sorghum stalks lignocellulose were hydrolysed into fermentable sugars. Bioconversion of fermentable sugars released from sweet sorghum bagasse into different products except ethanol, such as butanol, biogas, and chemicals was feasible to operate under low energy-consumption conditions.

The monoclinic crystalline form of aldolase from rabbit skeletal muscle grown at 29 degrees C is catalytically active in the direction of aldol cleavage. Activity was assayed for in a crystallization buffer containing 45% saturated ammonium sulfate using chemically unmodified single crystals cut to precise dimensions. Diffusion effects on velocities from assays employing aldolase crystals do not appear to be limiting when cut single crystals are crushed. Assays of crushed crystals are linear with respect to both time and enzyme concentration. Kinetic constants are reported for both substrates fructose 1-phosphate and fructose 1,6-phosphate. Maximal velocities and binding constants determined differ by no more than a factor of 2 between the crystalline and the soluble state of the enzyme. Analysis of the kinetic constants for fructose 1-phosphate as substrate shows that binding of substrate does not change in going to the crystalline state. Release of product is reduced roughly 2-fold in the crystalline state. A similar conclusion can be reached in the case of fructose 1,6-phosphate as substrate provided the "on" steps of substrate and product are only diffusion limited but independent of the physical state of the enzyme. It is not possible to distinguish between a more sluggish conformational change during catalysis or simply tighter product binding in the crystalline state as compared to the soluble enzyme state.  相似文献

Morphological mutants of Caulobacter crescentus were isolated by selecting for cells that did not possess normal, buoyancy-conferring stalks. The most prevalent phenotype enriched by the selection was structurally deficient stalks (designated "Cds"), observable as crumpled or flattened stalks. The second phenotype ("Abs") was observed as spontaneous shedding of normal-appearing stalks. The goal of our study was to compare the skeletal muscle regeneration induced by two types of injury: either crushing, that causes muscle degeneration as a result of mechanical devastation of myofibers, or the injection of a cardiotoxin that is a myotoxic agent causing myolysis of myofibers leading to muscle degeneration. Regenerating muscles were analyzed at selected intervals, until the 14th day following the injury. We analyzed their weight and morphology. We also studied the expression of different myosin heavy chain isoforms as a molecular marker of the regeneration progress. Histological analysis revealed that inflammatory response and myotube formation in crushed muscles was delayed compared to cardiotoxin-injected ones. Moreover, the expression of myosin heavy chain isoforms was observed earlier in cardiotoxin-injured versus crushed muscles. We conclude that the dynamics of skeletal muscle regeneration depends on the method of injury.  相似文献

A substantial loss of embryos occurs between Days 30 and 40 of pregnancy in the pig under crowded intrauterine conditions, but it is not clear whether this loss affects the growth of adjacent conceptuses. Birth intervals are known to increase with decreasing litter size, but the factors responsible are unknown. Two possibilities are that increased birth weight associated with reduced litter size and the empty uterine space and resulting constricted uterine regions that occur in pigs with small litters may impair piglet delivery. To address these, pregnant gilts were laparotomized on Day 35 of pregnancy and one or two fetuses were manually crushed through the uterine wall on the ovarian or cervical end of each uterine horn to create an empty uterine space behind or in front of the litter of piglets, respectively, in relation to the route of delivery from the uterus. A subset of gilts was slaughtered at 105 days of gestation to confirm that the empty uterine spaces were successfully created and to determine their effects on placental and fetal weights of adjacent conceptuses. At slaughter, the lengths of all externally visible empty constricted regions of the uterus were measured. The uterine horns were opened and the lengths of each placenta were measured from the umbilicus toward the ovary and toward the cervix to assess whether placentas developed symmetrically, and then each fetus and placenta was weighed. Fetal crushing successfully created constricted empty uterine regions on the ovarian and cervical ends of the uterine horns. Ovarian-side placental lengths were greater than cervical-side for conceptuses adjacent to fetuses crushed on the ovarian end of the horn. Cervical-side placental lengths were greater than ovarian-side for conceptuses adjacent to fetuses crushed on the cervical end. Both placental and fetal weights were greater (10% and 6%, respectively, P<0.05) for conceptuses adjacent to crushed fetuses compared to nonadjacent conceptuses. Remaining gilts were farrowed to determine the effect of litter size, average birth weights, and treatment on birth intervals of piglets, which were monitored using 24-h video surveillance. The negative association between number of piglets born alive and average birth interval was confirmed and was not explained by litter size-induced reduction in litter average birth weights. Birth intervals and stillbirth rate did not differ between cervically- and ovarian-treated gilts. These results indicate that conceptus loss on Day 35 of gestation can benefit the growth of adjacent placentas and fetuses, but the benefit is small. Increased average birth weight and the presence of empty uterine space that occurs when litter size is reduced does not fully explain the effect of litter size on birth intervals.  相似文献

This is a study of the acid-fast staining characteristics of pine pollen and an investigation of factors causing loss of acid-fastness after pine pollen has been in contact with tisues or mucous membranes. Intact loblolly pine pollen was readily stained with cold carbol-fuchsin, and retained its acid-fastness after decolorization with 3% HC1 in 95% ethyl alcohol for 2 min, followed by methylene blue counterstain. Pine pollen resembles spermatozoa in ease of staining and resistance to decolorization. Acid-fastness was destroyed by crushing or by germination of the pollen grain, and by contact for several hours with serum or saline solutions, but was unchanged by exposure to 0.1% solution of streptomycin in water. Nonviable pine pollen did not lose acid-fastness after suspension for several days in serum or water. When counterstain was omitted, crushed or germinated pollen appeared red to pink after staining with carbol-fuchsin and decolorization with acid alcohol, thus indicating that lipids of an acid-fast nature were still present. The large size and biologic properties of pine pollen provide a unique means of studying the chemical and physical aspects of the acid-fast phenomenon.  相似文献
