Summary We report on a thrombocytopenic female belonging to a pedigree with the Wiskott-Aldrich syndrome (WAS). Restriction fragment length polymorphism (RFLP) analysis with probe M27, closely linked to the WAS gene, demonstrated that she is a carrier of WAS. Both small-sized and normal-sized platelets were present, suggesting that, unlike the vast majority of WAS carriers, she does not manifest nonrandom X-chromosome inactivation in the thrombopoietic cell lineage. Study of X-chromosome inactivation by means of RFLP and methylation analysis demonstrated that the pattern of X-chromosome inactivation was nonrandom in T lymphocytes, but random in granulocytes. While this is the first complete report on the occurrence of thrombocytopenia in a carrier female of WAS as the result of atypical lyonization, it also suggests that expression of the WAS gene occurs at (or extends up to) a later stage than the multipotent stem cell along the hematopoietic differentiation pathway.  相似文献

Water chemistry of the Chascomús Pond (Province of Buenos Aires, Argentina) was studied during a year characterized by low precipitation. Supersaturation of calcium carbonate up to a calcite saturation index of about 20 in spring and summer was observed, together with a diminution of the concentration of dissolved organic matter, in particular humic substances (HS). These results suggested an adsorption between organic matter and calcium carbonate which should influence the growth of calcium carbonate crystals. This overview attempts to cover, from a personal viewpoint, the development of the heparin field during the last four decades. In particular, it emphasizes the metamorphosis of heparan sulfate (HS), from a disturbing contaminant in heparin production to the present-day key player in cell and developmental biology. Our understanding of the structural properties of the polysaccharides has been greatly promoted by studies of their biosynthesis. We now have a fairly detailed view of the various enzymatic reactions, that convert the initial [4GlcA1-4GlcNAc1-]n polymer into sulfated products with highly variable proportions of GlcA/IdoA and of N-acetyl, N-sulfate and O-sulfate substituents. It is also recognized that the variously substituted domains of the polysaccharide serve to interact, in more or less specific fashion, with a multitude of proteins, and that these interactions are essential to the biological functions of the proteins. Molecular genetics has unravelled the gene structures for almost all of the enzymes required to synthesize a heparin or HS chain, and has shown that several of these proteins exhibit genetic polymorphism. While differences in substrate specificity between enzyme isoforms may help to explain the structural variability of, in particular, HS chains, we still only partly understand the key features of heparin/HS biosynthesis and its regulation.  相似文献

This paper reports the influence of sulfate-reducing bacterial activity on acidogenic digestion of waste activated sludge (WAS). A series of experiments was conducted by feeding WAS to a 10-l, completely mixed, mesophilic reactor maintained at pH 5.0 under sulfidogenic and non-sulfidogenic conditions. Analyses of volatile fatty acids indicated that the productions of acetic and propionic acids were slightly increased by sulfidogenic activity at 218 mg/l level of sulfate when COD sulfate ratio was 55:1. Higher amounts of methanol, ethanol and hydrogen were observed in the non-sulfidogenic condition, but ammonia was lower than in the moderate sulfidogenic runs. At 0.63 kg VS/m3 d loading rate the hydrolysis was above 90% in both moderate sulfidogenic and non- sulfidogenic runs. The results of these experiments showed the possible influence of moderate sulfidogenesis in the protein degradation of WAS anaerobic digestion. However, the acid-phase digestion was adversely affected by increasing the sulfate concentration to 400 mg/l.  相似文献

We investigated the in vitro influence of humic substances (HS) extracted from de-inking paper sludge compost on the inhibition of Pythium ultimum by two compost bacteria, Rhizobium radiobacter (Agrobacterium radiobacter) and Pseudomonas aeruginosa. When low concentrations (5 or 50 mg l(-1)) of HS were added to the culture medium, fungal inhibition by R. radiobacter significantly increased (P<0.01) by 2-3%. In contrast, these low levels of HS had no effect on P. ultimum inhibition by P. aeruginosa. The Fe, chelated by HS, was in part responsible for the decrease of P. ultimum inhibition by the bacteria when increasing amounts of HS were added in the culture medium. The addition of 500 mg l(-1) of humic acids isolated from de-inking paper sludge compost or from fossil origin completely eliminated the inhibition of P. ultimum by R. radiobacter. This Fe effect also stimulated growth of R. radiobacter and reduced its siderophore production in a minimal medium supplemented with HS as sole source of Fe. The results showed that HS influence microbial antagonism when added to a culture medium. However, this effect varies with different factors such as the type of bacteria, concentration of HS, molecular weight and Fe content.  相似文献

An immunohistochemical study was undertaken, in an attempt to identify the acidic glycoprotein(s) present in colloid and in parenchymal cells in human fetal pituitary gland. As the colloid has been proposed to represent disintegrating cells, a series of antibodies against plasma glycoproteins and plasma proteins was applied; their presence intracellularly would generally be an indicator of plasma membrane leakage in dying parenchymal cells. In tissue sections from 9- to 20-week-old fetuses, the colloid showed prominent staining with an antibody to human fetuin/2 HS glycoprotein. Anti-2-HS glycoprotein labelled parenchymal cells in pars anterior and intermedia. Apart from a minor immunoreactivity for 1 glycoprotein, no other plasma glycoprotein was seen in colloid or parenchymal cells. An antibody against bovine fetuin showed staining of the colloid and of some parenchymal cells in pars distalis and intermedia; the plasma and stroma of the pituitary gland were unstained. In contrast, the anti-human plasma protein antibodies all stained the stroma. The presence of 2 HS glycoprotein in parenchymal cells and absence of other plasma glycoproteins imply integrity of the parenchymal cell plasma membrane. Thus, 2 HS glycoprotein is either synthesized locally or taken up specifically in the parenchymal cells, which are proposed to participate in the formation of colloid. It is suggested that 2 HS glycoprotein is part of a homeostatic system, which controls remodelling and physiological cell death during development.  相似文献

Summary Fetuin is a plasma protein present in high concentrations during fetal development in animals of the order Artiodactyla. Its role is not known. The human homologue of fetuin — 2HS glycoprotein — has been shown to be a negative acute phase protein in adult plasma. In the present study, the concentration of fetuin was measured in the serum of healthy cattle (Bovis bovis) and in animals with various injuries and inflammatory disorders. The levels were decreased by 30% in pregnancy but increased up to 10-fold in some trauma cases. A significant negative correlation between the concentrations of fetuin and albumin has also been found. Serine/threonine (O)-linked oligosaccharide on cell-surface CD43 has been reported to be abnormal in haemopoietic lineages of patients with the X-linked immunodeficiency, Wiskott-Aldrich syndrome (WAS). This defect largely appears to be the result of abnormal regulation of UDP-GlcNAc:Gal1-3GalNAc-R1-6-N-acetylglucosaminyltransferase (also known as core 2 GlcNAc-T), an enzyme in the Golgi apparatus that is subject to regulation during haemopoietic differentiation. To determine whether core 2 GlcNAc-T activity provides a reliable marker for WAS, we studied 12 unrelated WAS patients with respect to their expression of this enzyme activity. Compared with healthy subjects, the WAS patients showed levels of core 2 activity that were, on average, 2.5- and 3.9-fold higher in fresh lymphocytes and platelets respectively. These data suggest that altered core 2 GlcNAc-T activity is consistently found in lymphocytes and platelets of WAS patients and as such may provide a diagnostic marker for the disease. In view of the relatively limited amounts of blood sample generally available from infants and young children, we have also tested a more sensitive coupled assay that permits assessment of core 2 GlcNAc-T activity in very small samples of cells and which would therefore render this assay of wide clinical applicability.  相似文献

Differential emergence and diversity of bacterial communities from activated sludge in response to varied cultural conditions using 2,4-dichlorophenoxyacetic acid (2,4-D) were investigated by coupling molecular analyses based on 16S rDNA with functional genes. We employed three different cultural conditions: (1) a culture sequentially fed a high concentration (300 mg/L) of 2,4-D (HS); (2) a culture continuously fed a low concentration (10 mg/L) of 2,4-D (LC); and (3) a serial batch culture in which 1% (v/v) of culture was transferred to a fresh medium containing a high concentration (300 mg/L) of 2,4-D (HB). The HS and LC bioreactors were operated for 3 months and HB was repeatedly transferred for 1 month. The 2,4-D was stably degraded under all the cultural conditions tested. PCR amplification and cloning-based analysis of functional genes using community DNAs from the cultures revealed five different oxygenase genes that may be involved in the initial step of 2,4-D degradation. All five gene-types were present in HS, while one of the five genes, type V (tftA) was not detected in LC. Quantitative PCR analysis showed that in HS, Ralstonia eutropha JMP 134 type-tfdA4 (type I) was the most abundant in copy number (2.0 ± 0.1 × 107 copies/g DNA) followed by RASC type-tfdA (type II) (1.8 ± 1.0 × 106 copies/g DNA), putative cadA-like gene (type IV) (2.6 ± 0.8 × 105 copies/g DNA), cadA gene (type III) (1.3 ± 1.0 × 104 copies/g DNA), and tftA gene (type V) (3.5 ± 1.1 × 103 copies/g DNA). Similar results were obtained in LC. In contrast, HB contained only type I and type III genes, and the type I gene was five orders of magnitude greater in copy number than the type III gene. Denaturing gel gradient electrophoresis (DGGE) analysis of PCR, amplified 16S rDNA fragments of bacterial communities in the three different cultures showed low similarity coefficient values (0.35) when compared to the original activated sludge, suggesting that 2,4-D amendment caused a drastic change in the bacterial community. Particularly, HB showed only six bands (16–18 bands in the other cultures) and very low similarity coefficient values when compared to the other communities (0.10 to HS, 0.17 to LC, and 0.0 to original sludge). These results indicated that serial batch culturing (HB) resulted in a phylogenetically limited number of 2,4-D degrading bacteria carrying limited catabolic genes whereas more diverse 2,4-D degraders and catabolic genes were present in HS and LC. Therefore, the approach used for monitoring should be taken into account when one evaluates the population dynamics of contaminant-degrading bacteria at bioremediation sites.  相似文献

This work focuses on fermentation of pre-treated waste activated sludge (WAS) to generate volatile fatty acids (VFAs). Pre-treatment by high-pressure thermal hydrolysis (HPTH) was shown to aid WAS fermentation. Compared to fermentation of raw WAS, pre-treatment enabled a 2-5x increase in VFA yield (gVFA(COD)gTCOD(-1)) and 4-6x increase in VFA production rate (gVFA(COD) L(-1) d(-1)). Three sludges, pre-treated in full-scale HPTH plants, were fermented. One was from a plant processing a mix of primary sludge and WAS and the other two from plants processing solely WAS. The HPTH plants solubilised suspended matter, evidenced by a 20-30% decrease in suspended solids and an increase of soluble COD : total COD from 0.04 to 0.4. Fermentation of the three sludges yielded similar VFA concentrations (15-20gVFA(COD) L(-1)). The yields were largely independent of retention time (1 d-6 d) and temperature (42°C, 55°C). Also, the product spectrum depended mostly on the composition of the sludge rather than on operating conditions.  相似文献

Summary Eleven families segregating for the X-linked recessive immune deficiency disorder, Wiskott-Aldrich syndrome (WAS), were studied by linkage analysis with an alpha satellite DNA probe, pBamX-7, which detects polymorphism at the X chromosome centromere, locus DXZ1, as well as three other polymorphic markers defining loci on the proximal short arm of the X chromosome. Linkage has been established between WAS and DXZ1 ( ()=7.08 at =0.03) and WAS and the TIMP gene locus ( ()=5.09 at =0.0). We have also confirmed close linkage between DXZ1 and two marker loci, DXS14 and DXS7, previously shown to be linked to the WAS locus. The probe pBamX-7 detected allelic variation in all females tested, reflecting the high frequency of polymorphism at the centromere. One WAS carrier revealed a recombination between WAS and both marker loci DXZ1 and DXS14, indicating that WAS does not map between these loci. In conjunction with previous data from genetic mapping studies of WAS, these results confirm the pericentromerix Xp localization of WAS and demonstrate the usefulness of alpha satelite DNA probes as tools for genetic prediction in WAS as well as other pericentric X-linked diseases.  相似文献

Objective

To study the relationship between brain-gut peptides, gastrointestinal hormones and altered motility in a rat model of repetitive water avoidance stress (WAS), which mimics the irritable bowel syndrome (IBS).

Methods

Male Wistar rats were submitted daily to 1-h of water avoidance stress (WAS) or sham WAS (SWAS) for 10 consecutive days. Plasma hormones were determined using Enzyme Immunoassay Kits. Proximal colonic smooth muscle (PCSM) contractions were studied in an organ bath system. PCSM cells were isolated by enzymatic digestion and IKv and IBKca were recorded by the patch-clamp technique.

Results

The number of fecal pellets during 1 h of acute restraint stress and the plasma hormones levels of substance P (SP), thyrotropin-releasing hormone (TRH), motilin (MTL), and cholecystokinin (CCK) in WAS rats were significantly increased compared with SWAS rats, whereas vasoactive intestinal peptide (VIP), calcitonin gene-related peptide (CGRP) and corticotropin releasing hormone (CRH) in WAS rats were not significantly changed and peptide YY (PYY) in WAS rats was significantly decreased. Likewise, the amplitudes of spontaneous contractions of PCSM in WAS rats were significantly increased comparing with SWAS rats. The plasma of WAS rats (100 µl) decreased the amplitude of spontaneous contractions of controls. The IKv and IBKCa of PCSMs were significantly decreased in WAS rats compared with SWAS rats and the plasma of WAS rats (100 µl) increased the amplitude of IKv and IBKCa in normal rats.

Conclusion

These results suggest that WAS leads to changes of plasma hormones levels and to disordered myogenic colonic motility in the short term, but that the colon rapidly establishes a new equilibrium to maintain the normal baseline functioning.  相似文献

Summary As a potential measure of active biomass in an automated and continuous system, enzyme activities of activated sludge were sought that would allow a rapid and accurate determination. Using p-nitrophenylphosphate, bis-p-nitrophenylphosphate, p-nitrophenyl-- and -D-gluco- or galactopyranosides, and the p-nitroanilides of L-alanine, L-leucine, L-lysine, L-glutamic acid and L-phenylalanine, we could establish that the corresponding phosphatase, cyclic phosphodiesterase, glycosidase and aminopeptidase activities can be conveniently tested with diluted activated sludge within 10–20 min incubation at 30°C. The reactions were linear with time and concentration of activated sludge. The specific activities were in the range from 1 to 50 nmol substrate cleaved/min/mg protein at 30°C. They were diminished by starvation or poisoning with zinc powder, ZnCl2, HgCl2 or KCN. The enzymes were over 95% sedimented together with the flocs. Comparable activities were found in sludges from three independent sewage works in the Munich area. At the same time, dehydrogenase activities and ATP contents were investigated under several conditions.  相似文献

Several polymorphic mutations are located on the spectrin -chain; among these the variant termed IIa is characterized by an acid shift in the isoelectric point of the tryptic digest peptides 46 kDa and 35 kDa. In this variant a single amino acid substitution (alanine to aspartic acid) occured at position 972 of the spectrin -chain due to a point mutation (GCT to GAT) in the DNA. This variant, which seemed very rare in normal people, could be related to the recessive form of hereditary spherocytosis (HS) and could be absent in the dominant form of the disease. We have studied the IIa variant by denaturing electrophoresis of the spectrin tryptic digest peptides from 179 subjects: 46 controls, 78 patients with dominant (d) or non dominant (nd) HS and 55 relatives of the patients. The confirmation of the results was obtained at the DNA level in 41 subjects. The frequency of the chromosome bearing the IIa mutation was 7.6% in controls and higher (about 12–14%) in members of families with dHS as well ndHS. However, the family trees clearly showed that the mutation and the HS disease gene(s) were located on different chromosomes and inherited independently from each other. Furthermore, our study allows the conclusion that in most (if not all) cases of dHS, the IIa the variant is not the cause, is not a marker, and does not influence the phenotypic expression of the disease.  相似文献

Summary The ability of methanogenic bacteria to adapt to high concentrations of aluminium was investigated using a 9 Upflow Anaerobic Sludge Blanket (UASB) reactor fed semi-continuously with a synthetic waste containing glucose as the organic carbon source. It was found that an influent concentration of 2500 mg/ Al3+ could be tolerated by acclimatized methanogenic bacteria. However, if the concentration in the synthetic waste was further increased, inhibition of the methanogens resulted, with a corresponding decrease in digester performance. The aluminium precipitated as Al(OH)3 and accumulated in the sludge bed of the digester.  相似文献

Biomass behaviour and COD removal in a benchscale activated sludge reactor have been studied alternating anaerobic and aerobic conditions. Particular attention has been paid to the influence of the ratio of the initial substrate concentration (S 0) to the initial biomass concentration (X 0) on the reactor performance. Tests at very low ratios (S 0/X 0<2) demonstrate the existence of a threshold below which the reactor performance is seriously affected (S 0/X 0=0.5). Under conditions of total suppression of cell duplication, substrate maintenance requirements have also been calculated for the microbial consortium present in the activated sludges. The potential of 3,3,4,5-tetrachlorosalicylanilide (TCS) addition to an activated sludge continuous process to reduce excess sludge production by disrupting coupling between anabolism and catabolism was investigated. TCS was chosen as a metabolic uncoupler for continuous test in a lab-scale completely mixed activated sludge process. TCS reduced sludge yield by approximately 30% at a dosage of 40 mg/day. Substrate removal capability was not adversely affected by the presence of TCS, but effluent nitrogen concentration increased during the 60-day continuous operation. Sludge settleability of treated and control samples was qualitatively comparable and not significantly different. Microbial activities in terms of specific oxygen uptake rate were also enhanced, and the microbial population was altered. The results suggest that TCS is an effective chemical uncoupler that reduces sludge yield; process performance was not significantly affected by introduction of the uncoupler.  相似文献

High fermenter (volumetric) ethanol productivities (80 g/lh–1) were attained in a simple single-stage continuous-stirred-tank-reactor (CSTR) employing a flocculent mutant of Zymomonas mobilis with a feed containing 100g/l glucose.
