The surface are and crystallinity was measured on a cellulose powder made from Cladophora sp. algae. The algae cellulose powder was found to have a very high surface area (63.4 m2/g, N2 gas adsorption) and build up of cellulose with a high crystallinity (approximately 100%, solid state NMR). The high surface area was confirmed by calculations from atomic force microscope imaging of microfibrils from Cladophora sp. algae.  相似文献

通过室内培养模拟了高密度刚毛藻聚积腐烂过程,研究了刚毛藻在不同腐烂时间段下的溶解性有机物(DOM)的释放和附着微生物的群落变化。结果表明:在40d的分解实验中,刚毛藻生物量(干重)减少,且表现为前期损失快,后期损失减缓的趋势。在实验结束时(40d),干物质残留率为43.15%,质量损失了56.85%。在刚毛藻分解过程中, DOM在7—10d内快速释放达到最大值,而后降低。DOM的组分也变得复杂,荧光峰从区域Ⅰ、Ⅱ和Ⅳ逐渐转移到区域Ⅲ和Ⅴ。大量的简单芳香蛋白,如酪氨酸类物质被微生物转化为各种代谢物,并产生了腐殖质类物质。刚毛藻附着微生物优势菌为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和厚壁菌门(Firmicutes),相对丰度分别为6.54%—71.62%、16.83%—55.50%和0.95%—20.91%。在腐烂过程的不同阶段中,微生物的组成差异显著,表现为前期主要是Proteobacteria占主导优势,实验后期是Bacteroidetes占主导优势,细菌群落的演替与DOM组成的变化相关。  相似文献

研究于2019年冬季对赤水河流域开展刚毛藻多样性调查,共设计采样点38个,覆盖赤水河上、中、下游。调查发现:21个采样位点分布有刚毛藻目藻类,其中19个位点有刚毛藻分布。基于核糖体小亚基(SSU rDNA)、核糖体大亚基(LSU rDNA)和内转录间隔区(ITS)对采集样品进行系统发育分析,结果显示:(1)赤水河流域刚毛藻多样性较高,且该流域上、中、下游均有刚毛藻分布;(2)目前淡水刚毛藻类群包含至少10个支系,赤水河流域采集到的刚毛藻覆盖其中6个支系(分别是clade 1、clade 2、clade 4、clade 7、clade 9和clade 10);(3)相比基于SSU+LSU双分子标记构建的系统进化树,基于SSU+ITS+LSU三分子标记构建的进化树各支系支持度更高,可以较有效地将淡水刚毛藻不同支系区分开来。研究较好地展示了冬季赤水河流域刚毛藻的广泛分布及其分子多样性,丰富了中国淡水丝状绿藻的分类研究,也为赤水河段的水生态环境保护提供了基础数据支持。  相似文献

采用人工配制污水进行静态模拟实验,研究了镉对被海藻酸钙凝胶包埋固定的小球藻去除磷能力的影响.结果表明:在各光照及pH条件下,镉对固定化小球藻吸收磷的效果随着实验时间的推移而变化,在实验的第一天时影响最大;总体而言,镉抑制了固定化小球藻的除磷能力,但在某些条件下镉反而提高了藻对磷的吸收;固定化处理减弱了镉对小球藻除磷能力的影响.具体的光照、pH值与镉的影响效果的关系尚待进一步探讨.  相似文献

A microplate technique was developed to determine the conditions under which pure cultures of algae removed heavy metals from aqueous solutions. Variables investigated included algal species and strain, culture age (11 and 44 days), metal (mercury, lead, cadmium, and zinc), pH, effects of different buffer solutions, and time of exposure. Plastic, U-bottomed microtiter plates were used in conjunction with heavy metal radionuclides to determine concentration factors for metal-alga combinations. The technique developed was rapid, statistically reliable, and economical of materials and cells. Results (expressed as concentration factors) were in reasonably good agreement with literature values. All species of algae studied removed mercury from solution. Green algae proved better at accumulating cadmium than did blue-green algae. No alga studied removed zinc, perhaps because cells were maintained in the dark during the labeling period. Chlamydomonas sp. proved superior in ability to remove lead from solution.  相似文献

对三种泉溪大型藻类(脆弱刚毛藻, 弧形串珠藻和普生轮藻)藻体上附生藻类的种类、分布及环境因子与寄主本身对其的影响进行了研究。2004 年7 月至2005 年4 月在山西省辛安泉泉域采集三种大型藻类植物(每个季节采集一次)。研究结果表明, 三种大型藻类藻体上的附生藻类种类数不同, 普生轮藻上的附生藻类有49 种, 脆弱刚毛藻上37 种, 弧形串珠藻上6 种, 其中硅藻门种类数最多。这说明寄主本身结构的复杂程度在一定意义上会影响附生藻类的种类数及多样性。就季节变化而言, 三种大型藻类藻体上的附生藻类也表现出了明显的季节性,大致趋势为: 春季>秋季>夏季>冬季, 同时, 灰关联分析结果也表明, 环境因子在一定程度上对附生藻类也有影响, 最关键的影响因子是流速、水深和电导。这说明影响大型藻类植物藻体上附生藻类的影响因素是多方面的, 不仅有寄主本身, 同时流速、水深和电导这三种环境因子也起到了很关键的作用。    相似文献

作者于2012-2014年先后在湖北省采集到4株刚毛藻目的丝状绿藻。经鉴定,其形态特征和莫拉瓦刚毛藻Cladophora moravica Gardavsk(1986)非常相似;但相较于刚毛藻属各个种的形态,4株藻体更接近黑孢藻科的分类特征。基于SSU和LSU rDNA序列构建的系统发育树显示,4株刚毛藻目的丝状绿藻均属于黑孢藻科拟湖球藻属(Aegagropilopsis)。据此,作者建议将该种名称作为一个新组合处理,即莫拉瓦拟湖球藻Aegagropilopsis moravica(Dvoˇrák)Zhao et Liu com.nov。拟湖球藻属仅含有3个种:莫拉瓦拟湖球藻、硬枝拟湖球藻和棒状拟湖球藻,其形态特征与湖球藻相近且易混淆,故采用核基因组序列分析方法区分二者是非常有效的。  相似文献

Field observations of changes in the populations of aquatic weeds and phytoplankton have confirmed that aquatic weeds have antagonistic activity toward phytoplankton. Nutritional studies in the laboratory indicate that cultures of the aquatic weeds, Myriophyllum sp., Ceratophyllum sp., and duckweed (Lemma minor L.); liquid cultures of barley (Hordeum vulgare L., Dickson variety); and cultures of the filamentous green algae, Cladophora sp. and Pithophora oedogonium (Mont.) Withrock, will remain relatively free of epiphytes or competing phytoplankton if the cultures are nitrogen-limited. Field observations of Cladophora sp. have confirmed that the growth of epiphytes on the Cladophora is related to conditions of surplus available nitrogen compounds. It is proposed that this antagonistic activity may be due to a “nitrogen sink” effect in which the aquatic weeds or filamentous green algae prevent the growth of contaminating algae by competition for the limited nitrogen compounds available. However, the presence of bacteria-sized organisms which have selective toxicity to certain algae indicates that perhaps multiple factors exist. Discussed are the ecological implications of associations of certain algae with bacteria that have selective toxicities for other species of algae under certain environmental conditions such as nitrogen-limited growth.  相似文献

A chemically resistant cuticle fraction was isolated from 5 phaeophycean, 1 rhodophycean, and 11 chlorophycean marine algae using acid treatment alone, or acid treatment followed by leaching in cupra-ammonium. In Cladophora rupestris and Chaetomorpha melagonium this fraction consists of several alternate microfibrillar and amorphous layers similar in appearance to those seen in innermost carbohydrate-rich regions and amount to about 1/10 or more of the cell wall thickness. In Porphyra umbilicalis and Padina vickersiae it is a single layer less than I μ thick, accounting for 1/50–1/100 of the cell wall in Porphyra, and 1/5–1/10 of the cell wall in Padina. The cuticle fractions of all 4 algae contain surprisingly large amounts of protein (about 70% in Cladophora and 80% in Porphyra). Similarities in the behavior of cuticles obtained from the other 12 species studied suggest that they may have a similar protein-rich composition.  相似文献

The microalga Scenedesmus incrassatulus was grown under continuous regime in the presence of chromium(VI), cadmium(II) and copper(II), as single metal species and as mixtures of two or three metals, in a laboratory scale system. We used an artificial wastewater with low free ion activities (as determined by MINEQL+) due to the presence of EDTA (a strong chelating agent) but with total concentrations not suitable for acceptable environments. Chromium(VI) and cadmium(II) had positive interaction that increased the removal percentages of both these metals; we could not, however, detect any interaction with copper(II). S. incrassatulus was able to remove all the tested metals to some extent (25-78%), but bivalent metals were not removed as efficiently as reported in batch cultures, probably due to the high pH values there recorded. Chromium(VI) was more efficiently removed in continuous cultures than in batch culture, because the uptake of chromate could be favored by actively growing algae.  相似文献

Macroalgae have received much attention for heavy metal removal in treatment of domestic wastewater. In this report, the uptake capacity of a common freshwater green alga, Cladophora fracta, for heavy metal ions (copper, zinc, cadmium, and mercury) was evaluated. The equilibrium adsorption capacities were 2.388?mg Cu2+, 1.623?mg Zn2+, 0.240?mg Cd2+, and 0.228?mg Hg2+ per gram of living algae at 18°C and pH?5.0. The removal efficiency for Cu2+, Zn2+, Cd2+, and Hg2+ were 99, 85, 97, and 98%, respectively. Greater removal efficiency was achieved when the concentrations of metal ions were at very low level. The results indicated that living algae are suitable for removal and recovery of heavy metal ions from aqueous solutions and can be a potential tool to treat industrial wastewater.  相似文献

1984~2000年间,对娘子关泉域的藻类植物进行了15次采集,共采得藻类植物标本200余号。经鉴定,有107种,隶属7门,32科,49属。其中,以绿藻门的种类最多,有13科,19属,45种。其次是裸藻门,有2科,7属,26种。排在最后的是轮藻门,有1科,1属,3种。群落主要包括8类,即刚毛藻-红毛菜群落(Cladophora Bangia community),刚毛藻群落(Cladophora community),拖拉藻-轮藻群落(Chara Thorea community),弯枝藻-拖拉藻群落(Composopogon-Thorea community),水绵-双星藻群落(Spirogyra Zygnema community),无隔藻群落(Vaucheria community),串珠藻群落(Batrachospermum community)和泡状饶氏藻群落(Jaoa community)。其中,有的群落类型已趋于消失或消失,许多种类已成为濒危物种,其原因主要是人为因素造成藻类植物生存环境的改变或破坏。本文也提出了相应的保护对策。  相似文献

Cadmium removal by living cells of the marine microalga Tetraselmis suecica was tested in cultures exposed to different cadmium concentrations (0.6, 3, 6, 15, 30 and 45 mg/l). The EC50 for growth was 7.9 mg Cd/l after six days of exposure. The cadmium removed was proportional to the concentration of this metal in the medium and it was dependent on the time of exposure; cultures with higher cadmium concentration removed a higher amount of this metal. In cultures exposed to 0.6 mg/l, T suecica cells removed 98.1% of added cadmium with 0.392 x 10(-6) microg Cd/cell, whereas in cultures with 45 mg/l only 7.7% was removed with 16.052 x 10(-6) microg Cd/cell. The highest amount of cadmium removed per liter of culture was observed in cultures exposed to 6 mg/l, with 3.577 mg/l of cadmium. After six days of incubation, the higher proportion of cadmium was bioaccumulated intracellularly in all cultures except in 45 mg/l cultures, the percentage of intracellular cadmium being always more than 50%. The highest percentage of bioadsorbed cadmium (60.1%) was found in cells of cultures with the highest cadmium concentration (45 mg/l). Furthermore, a relation between intracellular cadmium and the concentration of sulfhydryl groups was observed.  相似文献

A mathematical model of the dynamics of radiocaesium transport in the aquatic phase—algae system is suggested in this work. Allowance is made for algae growth and for both reversible and irreversible absorption of this radionuclide by the algae. The algae biomass is divided hypothetically into two compartments with different exchange kinetics. The parameters of the model are time dependent. The model is quantified using experimental data for the concentrations of 137Cs in Cladophora glomerata filamentous algae and in water, obtained in situ in the environment of a nuclear power station. The model fits the data reasonably well and can be applied, for example, in bioindication of radioactivity in aquatic recipients in the environment of nuclear power stations.  相似文献

Larvae of the microcaddisflies Hydroptila waubesiana Betten and Oxyethira pallida (Banks) were given monocultures of eight genera of algae to test their suitability as food sources, and to investigate the degree of trophic specialization exhibited by these larvae. Only those larvae given green algae (Chlorophyta) survived and emerged as adults. Oxyethira pallida consumed Zygnema and Spirogyra, and exhibited the shortest larval-pupal period on Spirogyra (P < 0.05). Similarly, H. waubesiana consumed Zygnema and Cladophora, but exhibited the shortest larval-pupal period on Cladophora (P < 0.001). Diatoms and Hydrodictyon allowed H. waubesiana to attain partial growth only, while O. pallida did not consume either alga. Oscillatoria and Anabaena were not consumed by either species. The larvae of the two species tested appear to obtain adequate nourishment from only a narrow spectrum of algae, and one alga allowed significantly faster growth compared to the others tested in each case. Usually marine algae are an excellent source of pigments for different commercial sectors. Freshwater macroalgae can be exploited as a good source of biologically active compounds provided an appropriate extraction method is developed. The efficiency of four methods, like microwave‐assisted (MAE), ultrasound‐assisted extraction (UAE), supercritical fluid extraction (SFE) with ethanol as a co‐solvent, as well as conventional Soxhlet extraction were studied in the same conditions (time, solvent and temperature) for the recovery of chlorophylls and carotenoids from three freshwater green algae species: Cladophora glomerata, Cladophora rivularis and Ulva flexuosa. UV‐Vis spectrophotometry was used to determine chlorophyll a, chlorophyll b and total carotenoid content in obtained extracts. The results of this study showed that the advantages of novel extraction techniques (MAE and UAE) include higher yield and, in consequence, lower costs compared to traditional solvent extraction techniques. These methods were much more efficient in freshwater green algae pigment recovery than the classic Soxhlet extraction as well as SFE.  相似文献

Screening of marine microalgae for bioremediation of cadmium-polluted seawater   总被引:11,自引:0,他引:11  
Twenty four strains out of 191 marine microalgal strains exhibited cadmium (Cd) resistance. They were tested for their Cd removal ability in growth media containing 50 μM Cd. Six strains out of 19 green algae and one out of five cyanobacteria removed more than 10% of total Cd from the medium. The marine green alga Chlorella sp. NKG16014 showed the highest removal of Cd 48.7% of total. Cd removal by NKG16014 was further quantitatively evaluated by measuring the amount of cell adsorption and intracellular accumulation. After 12 days incubation, 67% of the removed Cd was accumulated intracellularly and 25% of the Cd removed was adsorbed on the algal cell surface. The maximum Cd adsorption (qmax) was estimated to be 37.0 mg Cd (g dry cells)−1 using the Langmuir sorption model. The Cd removal by freeze-dried NKG16014 cells was also determined. Cd was more quickly adsorbed by dried cells than that by living cells, with a qmax of 91.0 mg Cd (g dry cells)−1.  相似文献

A fluorescent pseudomonad (strain CW-96-1) isolated from a deep-sea vent sample grew at 30 degrees C under aerobic conditions in an artificial seawater medium and tolerated cadmium concentrations up to 5 mM. After 140 h, strain CW-96-1 removed > 99% of the cadmium from solution. Energy dispersive microanalysis revealed that the cadmium was removed by precipitation on the cell wall; sulfide production was confirmed by growth on Kligler's agar. Based on 16S ribosomal DNA sequencing and fatty acid analysis, the microorganism is closely related to Pseudomonas aeruginosa.  相似文献

Cladophora glomerata, a macrophytic green alga, is commonly found in the Great Lakes, and significant accumulations occur along shorelines during the summer months. Recently, Cladophora has been shown to harbor high densities of the fecal indicator bacteria Escherichia coli and enterococci. Cladophora may also harbor human pathogens; however, until now, no studies to address this question have been performed. In the present study, we determined whether attached Cladophora, obtained from the Lake Michigan and Burns Ditch (Little Calumet River, Indiana) sides of a breakwater during the summers of 2004 and 2005, harbored the bacterial pathogens Shiga toxin-producing Escherichia coli (STEC), Salmonella, Shigella, and Campylobacter. The presence of potential pathogens and numbers of organisms were determined by using cultural methods and by using conventional PCR, most-probable-number PCR (MPN-PCR), and quantitative PCR (QPCR) performed with genus- and toxin-specific primers and probes. While Shigella and STEC were detected in 100% and 25%, respectively, of the algal samples obtained near Burns Ditch in 2004, the same pathogens were not detected in samples collected in 2005. MPN-PCR and QPCR allowed enumeration of Salmonella in 40 to 80% of the ditch- and lakeside samples, respectively, and the densities were up to 1.6 x 10(3) cells per g Cladophora. Similarly, these PCR methods allowed enumeration of up to 5.4 x 10(2) Campylobacter cells/g Cladophora in 60 to 100% of lake- and ditchside samples. The Campylobacter densities were significantly higher (P < 0.05) in the lakeside Cladophora samples than in the ditchside Cladophora samples. DNA fingerprint analyses indicated that genotypically identical Salmonella isolates were associated with geographically and temporally distinct Cladophora samples. However, Campylobacter isolates were genetically diverse. Since animal hosts are thought to be the primary habitat for Campylobacter and Salmonella species, our results suggest that Cladophora is a likely secondary habitat for pathogenic bacteria in Lake Michigan and that the association of these bacteria with Cladophora warrants additional studies to assess the potential health impact on beach users.  相似文献
