To conserve a threatened plant species (Penthorum chinense Pursh) in Japan, seed germination responses to pretreatment (imbibition and/or chilled), temperature and light, and seed dispersal by water were examined. The seeds collected from abandoned paddy fields in a warm temperate region, central Japan, germinated in light (14 h photoperiod; light 22°C, dark 21°C) after a moist-chilled treatment. After this pretreatment, the seeds germinated well at 10–25°C (optimum temperature 15°C), but did not germinate in darkness even at the optimum temperature. Most of the seeds floated on distilled water, but 20–60% of the seeds that were collected from several populations sank in distilled water, indicating dimorphism in seed dispersal by water. The floating and sunken seeds did not show significant differences in weight and germination rate within a population. The addition of a surface-active agent in distilled water submerged the seeds, indicating that the buoyancy of the seeds is attributable to an oil coating on the seed surface that enhances the interfacial tension on the seeds. Three times the number of seeds sank in river water collected from a rural area than in distilled water. A greater number of seeds also sank in water that had increasing concentrations of linear alkylbenzenesulfonate, which is a major component of synthetic detergents. This suggests that the water dispersal of this species is suppressed by surface-active agents, including detergents, in river water.  相似文献

Diplotaxis harra (Forssk.) Boiss, an annual herb in the family of Brassicaceae, is widely distributed in many sandy and gypseous areas in southern Tunisia. Laboratory experiments were carried out to assess the effects of temperature and salinity on seed germination and recovery responses after seed transfer to distilled water. The germination responses of the seeds in complete darkness were determined over a wide range of temperatures (5, 10, 15, 20, 25 and 30 °C) and salinities (0, 50, 100, 150 and 200 mM NaCl). Germination was inhibited by either an increase or decrease in temperature from the optimal temperature (15 °C). Highest germination percentages were obtained under non-saline conditions and an increase in NaCl concentrations progressively inhibited seed germination. Rate of germination decreased with an increase in salinity at all temperatures but comparatively higher rates were obtained at 15 °C. Salt stress decreased both the percentage and the rate of germination. An interaction between salinity and temperature yielded no germination at 200 mM NaCl. Seeds were transferred from salt solution to distilled water after 20 days, and those from low salinities recovered at all temperatures. At NaCl concentration of 200 mM, the recovery of germination was completely inhibited.  相似文献

The effects of three salinities (0, 100 and 500 mM NaCl) and four constant temperatures (10, 20, 30 and 35 °C) on seed germination of Halostachys caspica (M. B.) C. A. Mey., Kalidium foliatum (Pall.) Mop. and Halocnemum strobilaceum (Pall.) Bieb. were investigated. After seeds were treated with different concentrations of NaCl at constant temperatures of 10–35 °C for 16 days, ungerminated seeds were transferred to distilled water for 10 days to investigate the total germination; after this time, the ungerminated seeds from the 10 and 20 °C treatments were then moved to 35 °C for another 5 days to determine the final germination. The three plant species in the present experiment are salt-resistant euhalophytes growing in high saline soils in the Zhungur Basin in Xinjiang, a northwest province of China.Compared with germination under control conditions, germination percentages of all three species were not affected by 100 mM NaCl at 10–35 °C, while severely inhibited by 500 mM NaCl; germination percentages were very low at 10 °C up to 100 mM NaCl for all species; the optimum temperature for germination of H. caspica and K. foliatum was 20–30 °C, while 35 °C for H. strobilaceum, up to 100 mM NaCl; seeds did not suffer ion toxicity for all species, as evidenced by the high total germination after ungerminated seeds pretreated with 500 mM NaCl were transferred to distilled water at constant temperatures of 10–35 °C for 10 days, and the high final germination after the ungerminated seeds from the 10 and 20 °C treatments were subsequently moved to 35 °C for another 5 days; Halostachys caspica had greater sensitivity to increasing temperatures from 10 and 20 °C to 35 °C compared with the other two species.  相似文献

The effect of the application of gibberellins to Tagetes minuta L. achenes (seeds) was determined at both 25°C, the optimal germination temperature, and 35°C, at which temperature the achenes are thermoinhibited. Both GA3 and GA4+7 accelerated germination at 25°C. Seed germination at 25°C was inhibited by paclobutrazol, but on subsequent application of GA4+7 rapid germination was induced. Following application of GA3 or GA4+7 to thermoinhibited seeds, a significantly higher final germination percentage was observed than in the distilled water control. However, endogenous gibberellin levels in germinating and thermoinhibited seeds did not differ significantly.  相似文献

Salsola affinis is a dominant annual inhabiting saline deserts of Xinjiang, China. Experiments were conducted to determine the effects of temperature, winged perianths and NaCl on seed germination and on germination recovery from the effects of saline conditions after transfer to distilled water. Freshly harvested seeds could germinate equally well in light and darkness at 5–30 °C. Attached winged perianths significantly inhibited germination, removal enhanced germination. However, germination was not inhibited in the presence of detached winged perianths in any of the temperature treatments. We suggest that the winged perianth is a mechanical barrier for radicle emergence, not a barrier for water uptake; hence, it inhibited germination. Germination of seeds from which the perianth had been removed was not affected by NaCl at concentrations below 0.4 mol/l, but it was significantly decreased by NaCl at concentrations of 0.6–2.0 mol/l. No seeds germinated at 4.0 mol/l NaCl. Seeds incubated in NaCl at concentrations of 0.05–4.0 mol/l for 14 days recovered after being transferred to distilled water. However, germination was lower than that in the non-saline control, indicating that a portion of the NaCl-treated seeds may lose their ability to germinate.  相似文献

A study was made to investigate possible formation by the crustose coralline algaLithophyllum yessoenseof multiple allelopathic-related substances against the settlement and germination of spores of various seaweeds. Seven different solvents (n-hexane, diethyl ether, acetone, ethyl acetate, acetonitrile, methanol, distilled water) and seawater were used to obtain crude extracts and secretory exudates from the coralline alga. The extracts and the algal conditioned seawater were tested for inhibitory activity against the settlement and germination of spores from 17 species representing 15 genera. Spore settlement of 14 species was inhibited over 90% by one or more extracts of the six organic solvents and conditioned seawater. The germination of spores from 13 species was inhibited by one or more extracts of all seven solvents and conditioned seawater. The species where spore settlement was not significantly affected showed strong inhibition of germination, andvice versa.  相似文献

Germination of macroconidia and/or microconidia of 24 strains of Fusarium solani, F. chlamydosporum, F. culmorum, F. equiseti, F. verticillioides, F. sambucinum, F. oxysporum and F. proliferatum isolated from fluvial channels and sea beds of the south-eastern coast of Spain, and three control strains (F. oxysporum isolated from affected cultures) was studied in distilled water in response to a range of water potentials adjusted with NaCl. (0, −13.79, −41.79, −70.37, −99.56 and −144.54 bars). The viability (UFC/ml) of suspensions was also tested in three time periods (0, 24 and 48 h). Conidia always germinated in distilled water. The pattern of conidial germination observed of F. verticilloides, F. oxysporum, F. proliferatum, F. chlamydosporum and F. culmorum was similar. A great diminution of spore germination was found in −13.79 bars solutions. Spore germination percentage for F. solani isolates was maximal at 48 h and −13.79 bars with 21.33% spore germination, 16% higher than germination in distilled water. F. equiseti shows the maximum germination percentage in −144.54 bars solution in 24 h time with 12.36% germination. This results did not agree with those obtained in the viability test were maximum germination was found in distilled water. The viability analysis showed the great capacity of F. verticilloides strains to form viable colonies, even in such extreme conditions as −144.54 bars after 24 h F. proliferatum colony formation was prevented in the range of −70.37 bars. These results show the clear affectation of water potential to conidia germination of Fusaria. The ability of certain species of Fusarium to develop a saprophytic life in the salt water of the Mediterranean Sea could be certain. Successful germination, even under high salty media conditions, suggests that Fusarium spp. could have a competitive advantage over other soil fungi in crops irrigated with saline water. In the specific case of F. solani, water potential of −13.79 bars affected germination positively. It could indicate that F. solani has an special physiological mechanism of survival in low water potential environments.  相似文献

In vitro culture is an important aid for ex situ conservation of rare, endemic or threatened plants. In this work, we establish an efficient method for the seed germination, seedling development, and axillary shoot propagation of Centaurea zeybekii Wagenitz. The seeds, collected from a wild population, were surface sterilised and cultured on various in vitro germination media. The effects of photoperiod and temperature on seed germination were also investigated. Germinations were obtained after 6 weeks in culture and the radicle emergence was evaluated as a main indicator. A high frequency of germination was obtained on distilled water supplemented with vitamines and 1 mg/L GA3. Although the seed germination frequencies were not affected by photoperiod, the highest germination frequency was obtained at 24 ± 2°C. A high frequency of axillary shoot proliferation was produced on MS medium supplemented with 1 mg/L BA. Then, the axillary shoots were separated and transferred to MS medium with or without plant growth regulators for rooting. Rhizogenezis was promoted after 6 weeks only in MS and 1/2 MS media containing 0.5 mg/L IBA. The rooting process was very slow and the percentage of shoot rooting was also very low (15%). The present study not only enables reinforcement of wild plant populations using ex situ growth of individuals, but it also helps to large number of aseptic seedling to use it in clonaly micropropagation studies.  相似文献

Larval cuticle ofHelicoverpa (Heliothis)zea and yeast extract added to a minimal medium (MM) induced germination of conidia ofNomuraea rileyi whereas sterile distilled water or MM alone did not. Yeast extract increased mycelial yield, but when cuticle was added, mycelial yield significantly decreased. 为探求促进广金钱草萌发及生长的有效途径,该文使用不同浓度的费氏中华根瘤菌、放射型根瘤菌以及田菁茎瘤固氮根瘤菌菌液分别浸染广金钱草种子,以蒸馏水浸泡种子为空白处理,研究其对广金钱草种子萌发指标(发芽率、发芽势、发芽指数、活力指数)的影响,并在温室培养40、60、80 d后分别测定广金钱草幼苗的生长指标(叶片数、分枝数、株高)及部分生理指标(叶绿素含量、含氮量)的变化。结果表明:(1)在1×107 CFU·mL-1浓度条件下,三种菌液浸种均获得最大萌发指标,其中田菁茎瘤固氮根瘤菌实验组效果最显著,发芽率、发芽势、发芽指数和活力指数高于对照组16.00%、9.33%、9.51和41.34%。(2)除田菁茎瘤固氮根瘤菌实验组外,其余实验组的广金钱草幼苗叶片数、分枝数及株高均低于对照组。(3)放射型根瘤菌实验组和田菁茎瘤固氮根瘤菌实验组幼苗叶绿素含量相比对照组分别增加1.47%和7.47%,含氮量分别增加0.57%和5.17%。综上所述,三种根瘤菌菌液浸种均可在不同程度上提高广金钱草种子的发芽能力和植株生长期的叶绿素和含氮量,其中田菁茎瘤固氮根瘤菌具有最大正向影响,可有效提高种子萌发能力并促进植株生长。该研究可为不同地区广金钱草的种植和栽培提供技术指导,为开发田菁茎瘤固氮根瘤菌作为植物促生长菌剂提供理论支持。  相似文献

The effects of eight different sodium salts on the activation of spores of Bacillus stearothermophilus NGB101 at 30°C were examined. Sodium nitrite was a potent activator spores of NGB101. Complete activation of spore populations was obtained after 6 h or less at 30°C. Activation of spores of NGB101 in solutions of sodium nitrite, like activation in distilled water, was temperature dependent, with optimal activation at 30°C. While a potent activator of spores of NGB101 at 30°C, sodium nitrite was ineffective as an initiator of germination at 65°C. Activation of spores of NGB101 produced marked increases in colony forming spores compared with nonactivated populations. Spore populations activated in solutions of sodium nitrite gave higher plate counts compared with spores activated in distilled water.  相似文献

Summary The distribution of membrane-associated calcium was investigated in pollen grains and tubes of the underwater pollinated angiospermNajas marina L. using chlorotetracycline (CTC). Tubes grown in distilled water (pH 6) showed the highest fluorescence in a subapical region that tapered basally into a fluorescent strand centrally located in the tube and extending back towards the pollen grain. The apical cap had low fluorescence as did the cytoplasm surrounding the fluorescent strand, the tube base and the pollen grain. Tubes grown in different pond waters (pH 8) revealed no intracellular CTC fluorescence. Instead there was an external fluorescence forming a distinct layer around the whole tube, frequently enhanced in a subapical region to form an external collar.Modification of the patterns of fluorescence could be induced by manipulation pH of the growth media and content of specific ions. For example tubes grown in distilled water with 10–3 M Mg2+ salts showed a similar CTC fluorescence as those grown in pond water. In contrast, Ca2+ enrichment had no visible influence on the patterns of fluorescence. The pattern of fluorescence displayed by tubes grown in distilled water, could be reproduced in pond water if the pH was artificially reduced to pH 6.Ultrastructurally, there was no detectable difference in the markedly polar distribution of organelles between pollen tubes grown in the various growth media. The secretory vesicles found in the pollen grain prior to germination become distributed throughout the pollen tube but are least concentrated in regions that show highest internal CTC fluorescence. In arid zones, precipitation distribution is extremely uneven, with saline‐waterlogging and dry–moist cycles appearing frequently, which negatively impact on seed germination and seedling establishment. The responses of two halophytes, Suaeda physophora and Haloxylon ammodendron, and a xerophyte, Haloxylon persicum, to saline‐waterlogging and dry–moist cycles were studied. The results showed that aeration increased seed germination for all species when seeds were submerged in NaCl, especially for xerophyte. Compared with S. physophora and H. ammodendron, seed germination, recovery germination, and total germination of H. persicum were much lower when seeds were submerged in 700 mm NaCl, especially for the recovery germination and total germination of nongerminated seeds when the seeds were desiccated and then transferred to distilled water. However, when the seeds were submerged in 700 mm NaCl with aeration, the seed germination, recovery germination, and total germination of nongerminated seeds transferred to distilled water increased dramatically for H. persicum. No adverse effect of desiccation was found on those values of nongerminated seeds pretreated in NaCl with or without aeration for the two halophytes. In conclusion, seeds of the two halophytes were more tolerant to waterlogging and dry–moist cycles than seeds of the xerophyte during emergence under saline conditions; these traits may be important for halophytes to survive extreme saline environments during the seed germination stage.  相似文献

Treatment of Trollius ledebouri seeds with gibberellins A4+A7 promotes germination. The efficacy of the treatment is dependent upon the duration of imbibition in distilled water prior to GA4+7 application. Presoaking increases both the final percentage germination attained and also its rate of achievement. No presoaking effect is exhibited by seeds induced to germinate by testa removal in the absence of GA4+7. Active washing of Trollius seeds enhances the presoaking effect and the eluent from washed seeds is inhibitory to germination. The results support the hypothesis that the presoaking effect exhibited by Trollius is the result of the leaching of a germination inhibitor from the seeds which is antagonistic to GA4+7. Additionally, treatment of Trollius seeds with the gibberellin-biosynthesis inhibitor (2-chloroethyl)-trimethylammonium chloride (CCC) prior to testa removal retards germination. The inhibitory effect of CCC on germination is overcome by GA4+7. Although CCC inhibits embryo growth during the presoaking of intact seeds, it does not affect the increased sensitivity of presoaked seeds to GA4+7. Therefore, although endogenous gibberellins may be involved in the germination process, they do not contribute to the presoaking phenomenon. The expansion of isolated endosperm tissue is not affected by CCC. However, the chemical markedly inhibits endosperm expansion in intact seeds and implicates the embryo as both the site of production of the germination inhibitor and of gibberellin. We tested the effects of cold stratification, temperature, light and NaCl on seed germination and germination recovery and of NaCl on radicle growth and radicle elongation recovery of Kalidium caspicum, a small leafy succulent shrub dominant in saline deserts in northwest China. In all conditions of temperature and light/darkness, germination percentages and rates of cold-stratified seeds were significantly higher than those of nonstratified seeds. Germination of a high percentage of both nonstratified and stratified seeds was inhibited by 0.2 M NaCl, and 0.6 M NaCl completely inhibited germination. Nongerminated seeds germinated after they were transferred from NaCl solutions to distilled water. Radicle elongation significantly decreased with increase in salinity, and it was completely inhibited by ≥1.0 M NaCl; radicle elongation recovered in young seedlings pretreated by 10 days of incubation in ≤0.4 M NaCl. Results show that seed germination and early seedling growth of K. caspicum are salt tolerant, and these characteristics help explain why this species can survive and dominate salt habitats, such as those in the Junggar desert in Xinjiang, northwest China.  相似文献

Kochia scoparia is one of the most common annual halophytes foundin the Great Basin. Seeds were collected from a population growing in asalt playa at Faust, Utah and were germinated at 5 temperature regimes(12 h night/12 h day, 5–15 °C, 10–20 °C, 15–25 °C,20–30 °C and 25–35 °C) and 6 salinities (0, 200, 400,600, 800 and 1000 mM NaCl) to determine optimal conditions forgermination and recovery of germination from saline conditions after beingtransferred to distilled water. Maximum germination occurred in distilledwater, and an increase in NaCl concentration progressively inhibited seedgermination. Few seeds germinated at 1000 mM NaCl. A temperatureregime of 25 °C night and 35 °C day yielded maximumgermination. Cooler temperature 5–15 °C significantly inhibited seedgermination. Rate of germination decreased with increase in salinity.Germination rate was highest at 25–35 °C and lowest at5–15 °C. Seeds were transferred from salt solutions to distilled waterafter 20 days and those from high salinities recovered quickly at warmertemperature regimes. Final recovery germination percentages in high salttreatments were high, indicating that exposure to high concentration ofNaCl did not inhibit germination permanently.  相似文献

Biological soil crusts can affect seed germination and seedling establishment. We have investigated the effect of biological soil crusts on seed water status as a potential mechanism affecting seed germination. The seed water potential of two annual grasses, one exotic Bromus tectorum L. and another native Vulpia microstachys Nutt., were analyzed after placing the seeds on bare soil, on a crust that contains various lichens and mosses (mixed crust), or on a crust dominated by the crustose lichen Diploschistes muscorum (Scop.) R. Sant. (Diploschistes crust). Seed water potential and germination were similar on the bare soil and the mixed crust, except for the initial germination of V. microstachys, which was higher on the mixed crust than on the bare soil. For the two grasses studied, seed water potential was significantly higher on the bare soil and mixed crust than on the Diploschistes crust. These differences in water potential correlated with differences in germination, which was much lower on the lichen crust. Experiments were conducted under two watering regimens. Increasing the frequency of watering amplified the differences in seed water potential and germination between the Diploschistes crust and the other two surfaces. For a particular watering regimen, the bare soil, mixed crust, and Diploschistes crust received the same amount of water, but they reached significantly different water potentials. Throughout the experiments, the water potential of the soil and mixed crust remained above −0.6 MPa, while there was a marked decline in the water potential of the Diploschistes surface to about −4 MPa. To ascertain that water was the major factor limiting germination on the Diploschistes crust, we conducted germination tests in an environment with 100% relative humidity. Under these conditions, germination on the Diploschistes crust was similar to that on the bare soil. However, the seeds that germinated on the Diploschistes crust did not penetrate this surface and approximately 60% of their root tips became necrotic. Our results indicate that the presence of D. muscorum can inhibit seedling establishment by two mechanisms: a reduction in seed water absorption and an increase in root tip mortality.  相似文献

Summary The effect of culture filtrates of some common rhizosphere fungi on the seed germination ofDichanthium annulatum, Bothriochloa pertusa andSetaria glauca was studied. Surface sterilized seeds were soaked in the fungal culture filtrates for 24 hours and after several washings in distilled water transferred to moist filter paper in Petri dishes and the percentage germination recorded. No significant inhibition or stimulation of germination was noticed in any of the treatments. The only exception wasTrichoderma viride which inhibited the germination ofD. annulatum seeds to some extent.  相似文献

以不同渗透势的PEG-6000溶液模拟干旱胁迫条件, 研究云锦杜鹃种子的萌发、幼苗生长和累积吸水率对干旱胁迫的响应。结果显示:种子于第14 d开始萌发,萌发期为8 d,总萌发率为45.5%±1.7%。在渗透势为-0.3~-0.6 MPa的溶液中, 种子的累计吸水率在第5 d就超过对照(蒸馏水中), 种子的萌发和幼苗的生长也优于对照, 说明云锦杜鹃种子不适合过于湿润的土壤环境中萌发。但溶液的渗透势≥-0.9 MPa时, 种子的吸水减缓、萌发率下降, 同时幼苗的生长也慢, 而当渗透势≥-1.8 MPa时, 种子不能充分吸涨、萌发, 说明云锦杜鹃种子对干旱胁迫比较敏感。另外, 经PEG溶液浸种2天后移入蒸馏水中,萌发率均有一定程度的提高, 其中经-0.3~-1.5 MPa的PEG溶液浸种后, 萌发率显著高于对照。提示用一定浓度范围的PEG溶液浸种可使休眠种子活化, 提高种子繁育的效率。  相似文献

Biological soil crusts dominated by drought-tolerant mosses are commonly found through arid and semiarid steppe communities of the northern Great Basin of North America. We conducted growth chamber experiments to investigate the effects of these crusts on the germination of four grasses: Festuca idahoensis, Festuca ovina, Elymus wawawaiensis and Bromus tectorum. For each of these species, we recorded germination time courses on bare soil and two types of biological soil crusts; one composed predominantly of the tall moss Tortula ruralis and the other dominated by the short moss Bryum argenteum. On the short-moss crust, the final germination percentage was about half of that on bare soil. Also, the mean germination time was 4 days longer on short-mosses than on bare soil. In contrast to the short-moss crust, the tall-moss crust did not reduce the final germination percentage but increased the mean germination time. Similar results were observed in the four grasses studied. To investigate the mechanism by which moss crusts affected germination, we analyzed the water status of seeds on bare soil and moss crusts. Six days after seeding, the water content of seeds on bare soil was approximately twice that of seeds on tall- or short-moss crust. Analysis of the time course of changes in seed weight and water potential in Bromus tectorum revealed that overtime seeds on tall mosses reached higher water content than those on short mosses. The increase in the water content of seeds on tall mosses occurred as the seeds gradually fell through the moss canopy. Taken together, our results indicate that biological soil crusts with distinct structural characteristics can have different effects on seed germination. Furthermore, this study revealed that a biological soil crust dominated by short mosses had a negative effect on seed water status and significantly reduced seed germination.  相似文献
