The application of an ultrafiltration procedure for fractionation of molecular weight of dissolved organic matter (DOM) extracellularly released by phytoplankton is described. Seven ultrafiltration membranes Diaflo (Amicon Corp. USA), with range 500--300,000 molecular weight (MW), were used for separation of different molecular weight compounds released by phytoplankton in Rhode River estuary of Chesapeake Bay, and their composition was determined. Percentage of extracellular release of DOM by phytoplankton varied from 3.92--68.07% of total carbon fixed in photosynthesis. The composition of algal extracellular products varied with different phytoplankton populations. However, two fractions of molecular weight compounds dominated in the composition of DOM released, i.e. a low molecular weight fraction of less than 1,000 MW and the fraction between 10,000--30,000 MW. The ultrafiltration procedure is effective for studying the composition of DOM released by phytoplankton in natural waters. 相似文献

土壤中溶解性有机质(DOM)是生物活性和物理化学反应活性都很活跃的有机组分,主要通过疏水吸附、分配、氢键、电荷转移、共价键、范德华力等多种作用与有机污染物结合,提高溶液中有机污染物的溶解度,改变土壤中有机污染物的吸附-解吸、迁移-转化等环境行为.DOM对有机污染物的吸附-解吸、迁移-转化过程的影响有双重性:一方面,DOM与有机污染物在土壤表面的共吸附可增加土壤对有机污染物的吸附容量,促进有机污染物在土壤中的吸持;另一方面,DOM对有机污染物的增溶作用,有利于土壤中有机污染物的解吸,提高移动性.作为光敏剂,DOM能提高土壤中有机物的光解反应速率.在一定条件下,DOM也可影响土壤中有机污染物的水解过程.DOM对土壤中有机污染物环境行为的影响与DOM和有机污染物的性质及其相互作用的介质条件密切相关. 相似文献

Liquid culture assays revealed a previously unreported capacity for Mycobacterium bovis BCG, M. gordonae, and M. marinum to oxidize CO and for M. smegmatis to consume molecular hydrogen. M. bovis BCG, M. gordonae, M. smegmatis, and M. tuberculosis H37Ra oxidized CO at environmentally relevant concentrations (<50 ppm); H2 oxidation by M. gordonae and M. smegmatis also occurred at environmentally relevant concentrations (<10 ppm). CO was not consumed by M. avium or M. microti, although the latter appeared to possess CO dehydrogenase (CODH) genes based on PCR results with primers designed for the CODH large subunit, coxL. M. smegmatis and M. gordonae oxidized CO under suboxic (10 and 1% atmospheric oxygen) and anoxic conditions in the presence of nitrate; no oxidation occurred under anoxic conditions without nitrate. Similar results were obtained for H2 oxidation by M. smegmatis. Phylogenetic analyses of coxL PCR products indicated that mycobacterial sequences form a subclade distinct from that of other bacterial coxL, with limited differentiation among fast- and slow-growing strains. 相似文献

Scavenging of dissolved organic matter (DOM) by particulate metal oxides like Fe(OH)3(s) is one of three processes that can influence the concentration and composition of DOM in aquatic systems. The other two possible processes include photodegradation and biodegradation. In combination, these processes alter the concentration and composition of DOM systematically with increasing time, measured as hydrologic residence time (HRT). The objective of this research was to determine the change in Fe(OH)3(s)-scavengable dissolved organic carbon (DOC) with increasing HRT (0–80 yr). In addition, DOC from allochthonous and autochthonous sources were included in this study. The susceptibility of DOC from surface waters to scavenging by Fe(OH)3(s) was found to decrease as a function of HRT, from approximately 90% to 79%. The lowest HRT system was operationally considered equivalent to allochthonous DOC, while autochthonous DOC was scavenged similarly to DOC from the 80 yr HRT system. These results indicate that scavenging of bulk DOC may be limited by metal loading in aquatic systems, and that the bulk of Fe(OH)3(s)-reactive DOC is from allochthonous sources. In addition, all surface waters treated with Fe(OH)3(s) contained approximately 1 mg l–1 of DOC that was resistant to scavenging (SD = 0.50, n = 5), which suggests that a refractory fraction of DOC persists in surface waters. 相似文献

The hyporheic zone is a region underneath streambeds that integrates surface and groundwater. Although its location is central to biogeochemical linkages between the riparian zone, dissolved nutrients, and benthic biota, the seasonal quality and likely sources of dissolved organic matter (DOM) in the hyporheic zone are not well understood. To investigate DOM characteristics in the hyporheic zone, water from the surface and subsurface (at depths 20, 60, and 100 cm below the streambed) was sampled every 4 weeks from 2007 to 2008 in a third-order stream in southern Ontario. Using UV spectroscopy, measures of spectral slopes, aromaticity, and A 254/A 365 ratios (molecular weight) were obtained. Temporal changes in these measures were consistent with watershed processes such as shedding of leaf litter in the fall, and photochemical and biofilm influence in the spring and summer. The fluorescence index (a measure of relative DOM source) suggested that at the surface and in the downwelling zone, DOM microbial sources increased with depth in the sediment, regardless of the season. Excitation–emission matrices (EEMs) showed seasonally distinct, protein-like DOM components of bacterial origin that were stronger in the fall. Leachates from specific allochthonous DOM sources—leaf litter from Betula papyrifera (white birch) and Thuja occidentalis (white cedar)—and an autochthonous source, biofilm, were isolated and incubated with unfiltered surface water. EEMs from these leachates indicated that these sources could indeed help explain observed patterns of DOM in surface and subsurface waters. These results suggest that although DOM sources were relatively constant, biogeochemical processing within the hyporheic zone resulted a DOM pool that was temporally dynamic and altered the nature of organic matter transported downstream into lakes and rivers. 相似文献

Pseudomonas acidovorans and Pseudomonas sp. strain ANL but not Salmonella typhimurium grew in an inorganic salts solution. The growth of P. acidovorans in this solution was not enhanced by the addition of 2.0 micrograms of phenol per liter, but the phenol was mineralized. Mineralization of 2.0 micrograms of phenol per liter by P. acidovorans was delayed 16 h by 70 micrograms of acetate per liter, and the delay was lengthened by increasing acetate concentrations, whereas phenol and acetate were utilized simultaneously at concentrations of 2.0 and 13 micrograms/liter, respectively. Growth of Pseudomonas sp. in the inorganic salts solution was not affected by the addition of 3.0 micrograms each of glucose and aniline per liter, nor was mineralization of the two compounds detected during the initial period of growth. However, mineralization of both substrates by this organism occurred simultaneously during the latter phases of growth and after growth had ended at the expense of the uncharacterized dissolved organic compounds in the salts solution. In contrast, when Pseudomonas sp. was grown in the salts solution supplemented with 300 micrograms each of glucose and aniline, the sugar was mineralized first, and aniline was mineralized only after much of the glucose carbon was converted to CO2. S. typhimurium failed to multiply in the salts solution with 1.0 micrograms of glucose per liter. It grew slightly but mineralized little of the sugar at 5.0 micrograms/liter, but its population density rose at 10 micrograms of glucose per liter or higher. Peatlands are important contributors of dissolved organic matter (DOM) to downstream aquatic systems. We investigated the effects of storm events on dissolved organic carbon (DOC) concentrations and DOM quality in a stream draining a Welsh peatland catchment. Intensive stream samples were collected and analysed for pH, DOC, dissolved organic nitrogen (DON), absorbance and fluorescence. Soil water samples and samples of sphagnum pore water were also collected, and a simple end-member mixing model was applied to account for changes occurring during the events. Fluorescence data were interpreted using parallel factor analysis (PARAFAC). DOC concentrations increased and pH decreased during the storm events. The soil water data and the mixing model indicated that this was due to a change of flow paths and draining of the DOC-rich acrotelm. Absorbance indices and the DOC/DON ratio suggested that the DOM released during events was less degraded. There was a striking, inversely related diurnal pattern in absorbance and fluorescence after the discharge peak. The diurnal pattern and a lack of fit with the mixing model suggested that fluorescing DOM was mainly produced in-stream. Fluorescence has been found to peak in the morning and decline during day-time due to photo-bleaching. We hypothesise that the input of additional DOM during events causes a change in the diurnal pattern, giving a peak at mid-day, when the processing of the additional DOM is highest. 相似文献

Sponges possess exceptionally diverse associated microbial communities and play a major role in (re)cycling of dissolved organic matter (DOM) in marine ecosystems. Linking sponge-associated community structure with DOM utilization is essential to understand host–microbe interactions in the uptake, processing, and exchange of resources. We coupled, for the first time, DNA-stable isotope probing (DNA-SIP) with 16S rRNA amplicon sequencing in a sponge holobiont to identify which symbiotic bacterial taxa are metabolically active in DOM uptake. Parallel incubation experiments with the sponge Plakortis angulospiculatus were amended with equimolar quantities of unlabelled (12C) and labelled (13C) DOM. Seven bacterial amplicon sequence variants (ASVs), belonging to the phyla PAUC34f, Proteobacteria, Poribacteria, Nitrospirae, and Chloroflexi, were identified as the first active consumers of DOM. Our results support the predictions that PAUC34f, Poribacteria, and Chloroflexi are capable of organic matter degradation through heterotrophic carbon metabolism, while Nitrospirae may have a potential mixotrophic metabolism. We present a new analytical application of DNA-SIP to detect substrate incorporation into a marine holobiont with a complex associated bacterial community and provide new experimental evidence that links the identity of diverse sponge-associated bacteria to the consumption of DOM. 相似文献

研究了来源于稻草腐熟物的外源水溶性有机质(DOM)和土壤本身固有的内源DOM对有机碳含量不同的3种土壤吸附菲的影响.结果表明,不同处理土壤对菲的吸附曲线均为线性,其吸附系数(Kd)与土壤有机碳含量(foc)正相关.去除内源DOM后,黄棕壤、红粘田和黑土吸附菲的Kd值增加了7.08%~21.4%,增加量(ΔKd)和增加幅度与foc正相关,表明土壤中存在的内源DOM抑制土壤对菲的吸附.而外源DOM对土壤吸附菲的影响与其浓度密切相关.在供试浓度范围(0~106 mg DOC·L-1)内,红粘田吸附菲的Kd值随加入外源DOM浓度的提高先增大后减小.外源DOM浓度为28 mg DOC·L-1时,红粘田吸附菲的Kd值增加了19.5%;而当外源DOM浓度≥52 mg DOC·L-1时,则明显抑制菲的吸附.内源和外源DOM对土壤吸附菲的影响,主要与DOM和菲在溶液中的结合作用、在土壤中的累积吸附效应等有关. 相似文献

为了了解滨海湿地土壤中溶解性有机质的结构特征及来源,2014年1月在胶州湾采集光滩、碱蓬、芦苇和大米草湿地的土壤样品,测定土壤溶解性有机质(DOM)含量,利用三维荧光技术进行光谱分析.结果表明: 胶州湾4种湿地类型土壤溶解性有机碳(DOC)含量表现为大米草湿地>光滩>碱蓬湿地>芦苇湿地,垂直剖面上随土层深度的增加DOC含量均呈减少的趋势.经光谱分析,胶州湾湿地土壤DOM的三维荧光光谱(3DEEMs)中出现了B、T、A、D和C等5种荧光峰,分别代表类酪氨酸、类色氨酸、类富里酸、类可溶性微生物副产物和类腐殖酸5种组分.利用荧光区域积分(FRI)法对5种组分进行定量分析,类色氨酸、类富里酸和类酪氨酸在DOM各组分含量中居前3位,类可溶性微生物副产物和类腐殖酸的含量次之,二者含量差异不显著.DOM的5种组分相互之间均呈显著正相关,与DOC含量呈显著正相关,与总磷、有效磷、总氮有不同程度的相关性.胶州湾4种湿地类型土壤DOM主要由生物相互作用内源产生,且腐殖化程度较低. 相似文献

Dissolved organic matter (DOM) is an important vehicle for the movement of nutrients from terrestrial to aquatic systems. To investigate how the source and composition of aquatic DOM change in both space and time, we used chemical, spectroscopic, and isotopic analyses to characterize DOM in a headwater catchment in the Colorado Front Range. Streamwater samples for DOM analyses were collected from 2 sites, a lightly vegetated alpine site and a forested, subalpine site, in the North Boulder Creek catchment during the snowmelt runoff season (May–September). Concentrations of dissolved organic carbon (DOC) peaked on the ascending limb of the snowmelt hydrograph at both the alpine (2.6 mg C l−1) and the subalpine sites (7.0 mg C l−1) and decreased sharply on the descending limb of the hydrograph. Fractionation of DOM into operationally defined humic and non-humic components showed that the fulvic acid content of DOC decreased through the season at both sites and that spectroscopic (fluorescence and ultraviolet) properties of the humic DOM fraction shifted in a manner consistent with an increase in the proportion of humic DOM derived from instream sources as compared to terrestrial catchment sources. Humic and non-humic fractions of DOM isolated near peak flow in June and during low flows in September showed a seasonal enrichment in 15N and 13C as well as a seasonal decrease in the ratio of aromatic to aliphatic carbon, both of which were correlated with a decrease in the C:N ratio of the DOM fractions. These results suggest that seasonal shifts in the isotopic and chemical characteristics of DOM are a result of changes in catchment sources of DOM. In particular, it appears that DOM production in alpine lakes is an important contributor to the streamwater DOM load during late season low flows, especially in the alpine reach of the catchment. Our results further suggest that stable isotopes of C and N are useful tools, particularly when combined with ancillary data such as elemental analyses and catchment discharge, for evaluating sources and transformations of DOM at the catchment scale. 相似文献

米槠次生林转换成米槠人工幼林和米槠人工促进天然更新幼林(以下简称"人促幼林")后,以这三种森林类型为研究对象,连续监测每次降雨后地表径流量及径流水中可溶性有机碳(DOC)的含量及通量,比较不同森林类型观测结果的差异,并分析降雨对实验结果的影响。结果表明:米槠人工幼林单次产流量是米槠次生林的1.5—19.0倍,观测期间总径流量为5.9倍;米槠人促幼林单次径流量和总径流量均与米槠次生林无显著差异(P0.05)。观测期间米槠次生林、人工幼林、人促幼林径流水DOC浓度值范围为5.9—18.4 mg/L,4.3—13.5 mg/L和3.2—9.9 mg/L,米槠次生林径流水浓度均值(12.6 mg/L)分别是米槠人促幼林(7.6 mg/L)和米槠人工幼林(5.3 mg/L)的1.6和2.4倍。回归分析表明,径流水中DOC浓度与降雨前土壤含水率呈显著相关;降雨前土壤含水率20.8%是一个临界值,含水率低于20.8%时,径流水DOC浓度与降雨前含水率呈显著正相关(P0.05);高于20.8%时,径流水DOC浓度与降雨前土壤含水率呈显著负相关(P0.05)。米槠人工幼林地表径流DOC输出通量是米槠次生林的0.7—5.4倍,观测期间总输出通量为2.1倍;米槠人促林DOC单次通量和观测期间总通量均与米槠次生林差异不显著(P0.05)。三种森林类型DOC输出通量均与降雨量呈显著相关(P0.05)。可见,米槠次生林转变成米槠人工幼林后DOC输出浓度降低,但径流量显著增加,导致DOC输出通量增加;而转变成米槠人促幼林后DOC输出浓度也降低,但径流量并未增加,因而并未增加DOC输出通量。 相似文献

Several algal species responsible for harmful algal blooms (HABs), such as Alexandrium fundyense, are mixotrophic under certain environmental conditions. The ability to switch between photosynthetic and heterotrophic modes of growth may play a role in the development of HABs in coastal regions. We examined the influence of humic dissolved organic matter (HDOM) derived from terrestrial (plant/soil) and microbial sources on the growth of A. fundyense. We found that a terrestrially derived HDOM, Suwannee River humic acid (SRHA), did enhance A. fundyense growth; however, a microbially derived HDOM, Pony Lake fulvic acid (PLFA) did not enhance growth. A. fundyense grows in association with bacteria in culture and we observed that bacterial cell densities were much lower in A. fundyense cultures than in bacteria‐only cultures, consistent with bacterial grazing by A. fundyense in culture. In bacteria‐only cultures with added algal exudates (EX), the addition of PLFA and SRHA resulted in a slight increase in bacterial cell density compared to cultures without HDOM added. Changes over time in the chemical quality of the HDOM in the A. fundyense cultures reflected contributions of microbially derived material with similar characteristics as the PLFA. Overall, these results suggest that the chemical differences between SRHA and PLFA are responsible for the greater effect of SRHA on A. fundyense growth, and that the differential effect is not a result of an effect on the growth of associated bacteria. 相似文献
