From a study of the effects of Ca ions on the nodulation of subterranean clover in flowing culture solutions it is concluded that root infection or nodule initiation has a higher Ca requirement than either nodule development or host plant growth in the presence of fixed nitrogen.  相似文献

We have developed a rapid and reproducible transformation system for subterranean clover (Trifolium subterraneum L.) using Agrobacterium tumefaciens-mediated gene delivery. Hypocotyl segments from seeds that had been allowed to imbibe were used as explants, and regeneration was achieved via organogenesis. Glucose and acetosyringone were required in the co-cultivation medium for efficient gene transfer. DNA constructs containing four genes encoding the enzymes phosphinothricin acetyl transferase, [beta]-glucuronidase (GUS), neomycin phosphotransferase, and an [alpha]-amylase inhibitor were used to transform subterranean clover. Transgenic shoots were selected on a medium containing 50 mg/L of phosphinothricin. Four commercial cultivars of subterranean clover (representing all three subspecies) have been successfully transformed. Southern analysis revealed the integration of T-DNA into the subterranean clover genome. The expression of the introduced genes has been confirmed by enzyme assays and northern blot analyses. Transformed plants grown in the glasshouse showed resistance to the herbicide Basta at applications equal to or higher than rates recommended for killing subterranean clover in field conditions. In plants grown from the selfed seeds of the primary transformants, the newly acquired gene encoding GUS segregated as a dominant Mendelian trait.  相似文献

Clones of two partially resistant and two susceptible white clover, Trifolium repens, genotypes were exposed to eggs of Heterodera trifolii and nematode development in stained roots measured at 2, 4, 7, 11, 18, 23, and 37 days after inoculation. The differences in development between nematode populations in resistant and susceptible genotypes showed that resistance operated after infection during feeding and development. At 7 days after inoculation, counts of second-stage juveniles did not differ between genotypes, whereas at 37 days more adults had developed in the susceptible than in the resistant genotypes. In a separate experiment, cysts hosted by susceptible genotypes were larger and contained more eggs than those on resistant genotypes so that the product of the values for cysts per plant and for eggs per cyst resulted in a more sensitive measure of resistance than from using cysts per plant alone.  相似文献

Lipid depletion of berseem seed treated with 100 mg/1 gibberellic acid or maleic hydrazide was studied during germination. During the first 3 days after germination there was little lipid consumption but thereafter the depletion of lipid was rapid. Gibberellic acid was found to enhance the lipid utilization of total lipids and triglyceride components during germination. On the other hand, maleic hydrazide reduced the utilization of triglycerides of the germinating seed. The rate of utilization was rapid for unsaturated fatty acids and slower for saturated ones in treated seeds as compared to untreated seeds.  相似文献

White clover (Trifolium repens L.) stolons become buried inthe field. It was observed that this also occurred in the greenhousewhere the accepted mechanisms of burial, treading by livestockand earthworm casting, did not occur. It was also observed thatthe crown of seedling T. repens plants become closely appressedto the soil. Experiments showed that, regardless of varietyof T. repens or depth of planting, all seedling hypocotyls firstlift the cotyledons clear of the soil, then ‘ contract’towards the soil until the cotyledons are in contact with orbelow the soil surface. Auxanometers were used to measure therate and extent of this contraction and were also attached tostolon nodes in experiments which showed that stolons move downwardsrelative to the soil surface and that the speed and extent ofthis duration varied with soil type. A further experiment showedthat only rooted nodes show this behaviour. The force exertedby the contraction of nodal roots was estimated experimentallyas 0.21 N g-1fresh root. A mechanism for the root contraction,based on examination of root anatomy of seedling tap-roots andnodal roots, is suggested. This study was conducted to identify lines of subterranean clover (Trifolium spp.) with resistance to Meloidogyne arenaria (Neal, 1989) Chitwood, 1949, race 1; M. incognita (Kofoid and White, 1919) Chitwood, 1949, race 3; and M. javanica (Treub, 1885) Chitwood, 1949. A collection of 134 subterranean clover lines was evaluated and all had intermediate to high susceptibility. Root galling was negatively correlated with both seed and dry matter yields. Soil fumigation significantly reduced the nematode population in the field. Results indicate there is limited genetic resistance to root-knot nematodes among subterranean clover lines. Alternative sources of variation for this trait should be investigated.  相似文献

Red clover (Trifolium pratense L.) is a forage legume and an allogamous diploid plant (2n = 14; 440 Mb). Here, we examine the 7 prometaphase chromosomes of red clover using fluorescence in situ hybridization (FISH) with ribosomal RNA sequences, pericentromeric and telomeric repeats, as well as bacterial artificial chromosome (BAC) clones. Position of hybridization signals and chromosome condensation patterns were quantified by the help of the chromosome image analysis system ver. 4.0 (CHIAS IV). Fourteen BAC clones belonging to linkage groups (LG) 1-7 hybridized to individual chromosomes 4, 2, 6, 5, 1, 7, and 3, respectively. Quantitative analysis of FISH mapping and chromosome analysis using CHIAS IV allowed us to construct a quantitative idiogram that constitutes the comprehensive chromosome map of red clover. Chromosomal positions of the 26S rDNA locus were detected at a heterozygous locus on chromosome 6 in the variety HR, and polymorphisms of rDNA loci were observed in other varieties, although chromosomal positions of some BAC clones did not vary among HR and other varieties. These results demonstrate chromosomal collinearity among allogamous red clover varieties. This integration of genetic linkage and quantitative chromosome maps should provide valuable insight into allogamous legume genetics.  相似文献

With the aim of establishing the basic knowledge and resourcesneeded for applied genetics, we investigated the genome structureof red clover Trifolium pratense L. by a combination of cytological,genomic and genetic approaches. The deduced genome size was440 Mb, as estimated by measuring the nuclear DNA content byflow cytometry. Seven chromosomes could be distinguished bymicroscopic observation of DAPI stained prometaphase chromosomesand fluorescence in situ hybridization using 28S and 5S rDNAprobes and bacterial artificial chromosome probes containingmicrosatellite markers with known positions on a genetic linkagemap. The average GC content of the genomes of chloroplast, mitochondrionand nucleus were shown to be 33.8, 42.9 and 34.2%, respectively,by the analysis of 1.4 Mb of random genomic sequences. A totalof 26 356 expressed sequence tags (ESTs) that were grouped into9339 non-redundant sequences were collected, and 78% of theESTs showed sequence similarity to registered genes, mainlyof Arabidopsis thaliana and rice. To facilitate basic and appliedgenetics in red clover, we generated a high-density geneticlinkage map with gene-associated microsatellite markers. A totalof 7159 primer pairs were designed to amplify simple sequencerepeats (SSRs) identified in four different types of libraries.Based on sequence similarity, 82% of the SSRs were likely tobe associated with genes. Polymorphism was examined using twoparent plants, HR and R130, and 10 F1 progeny by agarose gelelectrophoresis, followed by genotyping for the primer pairsshowing polymorphisms using 188 F1 plants from the mapping population.The selected 1305 microsatellite markers as well as the previouslydeveloped 167 restriction fragment length polymorphism markerswere subjected to linkage analysis. A total of 1434 loci detectedby 1399 markers were successfully mapped onto seven linkagegroups totaling 868.7 cM in length; 405 loci (28%) were bi-parental,611 (43%) were specific to HR and 418 (29%) were specific toR130. Each genetic linkage group was linked to a correspondingchromosome by FISH analysis using seven microsatellite markersspecific to each of the linkage groups as probes. Transferabilityof the developed microsatellite markers to other germplasmswas confirmed by testing 268 selected markers on 88 red clovergermplasms. Macrosynteny at the segmental level was observedbetween the genomes of red clover and two model legumes, Lotusjaponicus and Medicago truncatula, strongly suggesting thatthe genome information for the model legumes is transferableto red clover for genetic investigations and experimental breeding.  相似文献

In a growth chamber experiment, we determined net photosynthetic rate (PN) and leaf developmental characteristics of cultivars of a relatively small-, intermediate-, and a large-leaf genotype grown under irradiance of 450-500 µmol(photon) m-2 s-1 (HI), shade [140-160 µmol(photon) m-2 s-1] (LI), and after a shade-to-irradiation (LI »HI) transfer. Differences in physiological responses of the genotypes were more pronounced in HI and LI»HI plants than in LI plants. The small- and intermediate-leaf sizes had greater PN in the first measured leaf than the large-leaf type by 70 and 63 % in HI plants, and by 23 and 18 % in LI»HI plants, respectively. Similar relationships were observed in the next developed leaf. The LI plants did not differ significantly in PN. Greater PN in the small- and intermediate-leaf size genotypes were not associated with greater total dry matter of the plant. Under irradiation, the large-leaf genotype accumulated more total nonstructural saccharides (TNS) and starch than the small- or intermediate-leaf size plants. TNS and starch concentrations in LI plants were about one-half those of HI and LI»HI plants. These results should help to develop management practices that capitalize upon the competitive features of white clover in mixed-species swards.  相似文献

The response to different combinations of daylength (14, 15and 16 h) and day/night temperature (20/10°C, 30/15°C)of four white clover (Trifolium repens) varieties in terms offloral initiation and development was studied. More plants flowered under long days (16 h) at 30°C thanin any other combination. Varietal differences in response todaylength were large but temperature was the factor controllinginitiation. Daylength was more important in regulating peduncle length,floret and ovule numbers than was temperature. The amount ofnectar produced was lowest at the higher temperature althoughnectar concentration was unaffected by temperature. Nectar concentrationwas lowest in the shortest daylength (14 h). Summary Berseem (Trifolium alexandrinum L.) and oats (Avena satina L.) were grown in a greenhouse on a soil with a toxic level of available molybdenum. Significant increase in yield of berseem was observed on application of phosphorus and sulphur. The oat crop responded only to P application. The content of Mo in plants was enhanced by application of phosphorus. The application of sulphur depressed the Mo content of plants. The effect of P application was more conspicuous than the depressing effect of S application on Mo content of plants. The decrease in Mo content with S application was more pronounced in the oat crop. An application of 100 ppm S as gypsum caused 57.6 per cent decrease in the concentration of Mo which was below the threshhold toxic level. But in berseem, an application of 100 ppm S even could not lower the Mo concentration to a safer level. Hardseededness breakdown in the burr of subterranean clover(Trifolium subterraneum L.) was found to be non-random in occurrencewith respect to the relative position of seed in the burr. Thissuggests the presence of a predeterminative mechanism of theorder of hardseededness breakdown. While seed moisture content did not exhibit a pattern of variationconsistent with that for hardseededness breakdown, seed weightvariation appeared to conform closely with the pattern of breakdown. It is postulated that, either differences in seed size per sewithin the burr may determine the order of hardseededness breakdown,or that such differences may reflect a disproportionate supplyof translocate, and hence promotors or inhibitors, of hardseedednessbreakdown to the seeds of the burr. The implications of this finding to the assessment of genotypesfor differences in level of hardseededness level have been outlined.  相似文献

Changes caused by separate calcium and phosphorus deficienciesin the impedance locus and associated parameters of petioleswere investigated. In samples of mixed ages the deficiencieswhere characterized by a significant change in 17 out of the18 parameters studied. Almost invariably the change caused byphosphorus deficiency was significantly greater than that causedby calcium deficiency. In a more detailed study of phosphorus deficiency it was foundthat there is often a significant interaction between age andparameter change. From the phase angle data it is concludedthat the plasma membrane ages more rapidly in deficient thanin non-deficient plants; nevertheless the physico-chemical compositionof the plasma membrane in newly formed cells is the same indeficient and non-deficient petioles of similar ages. The extent to which phosphate can be absorbed directly fromthe outer medium by stolon internodes and contribute to thetotal accumulation of phosphate by intact plants of white clover(Trifolium repens L. cv. Blanca) was assessed in hydroponicexperiments in a controlled environment room. The uptake ofphosphate by intact roots or stolons was measured by sealinga segment (6-0 mm long) across a flow-cell in which 32P-labellednutrient solution was circulated for 24 h, the rest of the rootsystem receiving unlabelled nutrient solution. The rate of uptakeof phosphate (µmol g–1 d–1 dry wt. basis)by roots was more than 300 times that by intact stolons. Pretreatmentof stolons by gentle abrasion to remove cuticle, so as to simulatethe condition of stolons in the field, increased the uptakeof phosphate 7-fold compared with that of intact stolons. However,the potential of stolons to contribute to the P status of whitedover in the field was calculated to be small (5%). When an incision was made through the hypodermal layer of stolons,the rate of phosphate uptake greatly increased, attaining 71%of that by root segments. This increase, which was greater athigher phosphate concentrations, indicates that the suberi.zedhypodermis constitutes a major barrier to the influx of phosphatein the stolon. After withholding phosphate for different time intervals, thesubsequent rate of phosphate uptake by roots was increased 2-3-foldafter 2 d phosphate deprivation and 3-4-fold after 6 d or 13d phosphate deprivation. A higher proportion of absorbed phosphatewas transported to shoots in phosphate-deprived plants. After1 d of uptake following restoration of the phosphate supply,the concentrations of labelled phosphate in shoots were greaterthan in control plants, although the concentrations of labelin roots was less. However, the rate of uptake of phosphateby stolons, following deprivation, was not significantly increased.These results suggest that the mechanism regulating the enhancedrate of phosphate loading into the xylem, initiated by a periodof phosphate deprivation, is specific to roots and is not inducedin stolons. The results are discussed in relation to the growth and acquisitionof phosphate by white clover in the field. The pattern of breakdown of seed coat impermeability of seedwithin the burrs of two cultivars of subterranean clover (Trifoliumsubterraneum L.) was closely associated with relative differencesin flowering sequence within the inflorescence. The patternof seed weight variation showed very little relationship withthe pattern of breakdown, while that for seed moisture contentshowed no relationship. Within a burr it was almost invariably the seed which developedfrom the latest flower within the inflorescence which was thefirst to become permeable. An ordering of the timing of breakdownwas evident only within the burr; there was no evidence fora pattern of breakdown of impermeability between the differentburrs along the runner. The objective of this work was to construct a model of aerialdevelopment of clover that takes into account morphogeneticresponses to the light environment, and to use it to analyseand understand these processes in terms of signal perceptionand integration. The plant model was interfaced with a MonteCarlo model that determines photosynthetically active radiation(PAR) and red/far-red ratio (R/FR) throughout the canopy, takinginto account the absorption, reflection and transmission oflight by individual leaves. Light intensity and quality weresensed by the plant model at discrete time intervals and atdiscrete sites of perception: apices, emerging internodes andpetiole tips. This input regulated the final size of internodesand leaves, the vertical positioning of leaves, and the branchingdelay. The empirical relations (regression functions) quantifyingthis regulation were derived from data reported in the literatureand original measurements. Simulations produced realistic visualizationsand quantitative characterizations of the modelled plants fordifferent light treatments. These results were in general agreementwith observations of real plants growing under similar conditions,suggesting that the dependence of organ size and position onlight treatments can be regarded as an integration of the responsesof individual plant organs to their local light environment.The model is used to describe the regulation of branch appearanceand the impact of self-shading on plant morphogenesis as a functionof local light environment.
