The effects of temperature on the dark relaxation kinetics of nonradiative energy dissipation in photosystem II were compared in lettuce (Lactuca sativa L.) chloroplasts and leaves of Aegialitis annulata R. Br. After high levels of violaxanthin de-epoxidation in the light, Aegialitis leaves showed a marked delay in the dark relaxation of nonradiative dissipation, measured as non-photochemical quenching (NPQ) of photosystem II chlorophyll a fluorescence. Aegialitis leaves also maintained a moderately high adenylate energy charge at low temperatures during and after high-light exposure, presumably because of their limited carbon-fixation capacity. Similarly, dark-sustained NPQ could be induced in lettuce chloroplasts after de-epoxidizing violaxanthin and light-activating the ATP synthase. The duration and extent of dark-sustained NPQ were strongly enhanced by low temperatures in both chloroplasts and leaves. Further, the NPQ sustained at low temperatures was rapidly reversed upon warming. In lettuce chloroplasts, low temperatures sharply decreased the ATP-hydrolysis rate while increasing the duration and extent of the resultant trans-thylakoid proton gradient that elicits the NPQ. This was consistent with a higher degree of energy-coupling, presumably due to reduced proton diffusion through the thylakoid membrane at the lower temperatures. The chloroplast adenylate pool was in equilibrium with the adenylate kinase and therefore both ATP and ADP contributed to reverse coupling. The low-temperature-enhanced NPQ quenched the yields of the dark level (Fo) and the maximal (Fm) fluorescence proportionally in both chloroplasts and leaves. The extent of NPQ in the dark was inversely related to the efficiency of photosystem II, and very similar linear relationships were obtained over a wide temperature range in both chloroplasts and leaves. Likewise, the dark-sustained absorbance changes, caused by violaxanthin de-epoxidation (A508nm) and energy-dependent light scattering (A536nm) were strikingly similar in chloroplasts and leaves. Therefore, we conclude that the dark-sustained, low-temperature-stimulated NPQ in chloroplasts and leaves is apparently directly dependent on lumen acidification and chloroplastic ATP hydrolysis. In leaves, the ATP required for sustained NPQ is evidently provided by oxidative phosphorylation in the mitochondria. Summary Cultures ofStaurastrum gracile maintained in a liquid medium in a 168 hours light: dark regime were asynchronous. Alteration of the regime to 1410 or 186 failed to induce synchrony, as did variation of the light and dark temperatures. The cultures were synchronized by eliminating a light period to give a total dark period of 32 hours. One such cycle gave 54% synchrony, the cells entering division at the onset of the second light period after treatment. The omission of a second light period 72 hours after the first produced no increase in the level of synchrony. The level was improved by omitting a further light period 48 hours after the first to give a 321632 induction treatment which resulted in 64% synchrony. In no instance did the cells divide between the dark treatment and the synchronized wave of cytokinesis. This preliminary mechanistic model of normal swimming and phototactic behaviour in individual Daphnia was constructed using data and assumptions based on experiments and observations. Swimming under constant light intensity is characterized by short periods of upward movements alternating with equal periods of downward movements. Two oscillators are proposed that generate these phases in swimming. Unexpected shifts in depth, as observed in D. magna and D. longispina, are also present in the swimming of the computer daphnid and thus seem to be inherent to the underlying mechanism. As in real daphnids, during relative decreases in light intensity of low velocity, positive phototactic upward swimming is stepwise. With increasing velocity in the change in light, these steps disappear. When the model is triggered by a natural increase in light at dawn, a small downward movement results. Migration distance can be increased to commonly found depths of migrating Daphnia by the introduction of a fish exudate factor into the model, which enhances the phototactic response. Since attenuation of light in the water affects the phototactic swimming response, it also influences migration distance. The results of model calculations agree quite well with an empirical relationship between Secchi disc depth and amplitude of diel vertical migration in a number of lakes.  相似文献

Changes in carbon isotope composition(13C) and leaf morphology associated withvegetative phase change were monitored in Metrosiderosexcelsa Sol. ex Gaertn. (family Myrtaceae). Plants of threeontogenetic states were used: juvenile seedlings, micropropagated plants in arejuvenated state, and reproductively mature plants bearing leaves with adultcharacteristics. The effects of temperature regime (32/24 °C,24/16 °C, and 16/8 °C day/night) and plantarchitecture (branched and single-stemmed plants) were studied in two separateexperiments. Although both juvenile and rejuvenated plants exhibited juvenileleaf morphology at the start of the experiments, there was no differencebetweenleaf 13C in these plants and that in adultplantsat this time (mean ca. –27%). Vegetative phase change occurred injuvenileand rejuvenated plants grown at 24/16 °C, and there was acorresponding increase in leaf 13C (from ca.–27% to –23%) in these two groups of plants. Leaf13C in adult plants remained relatively constant(ca. –26%) at 24/16 °C. There was little change in leaf13C in all plant states maintained at 32/24°C or 16/8 °C, and vegetative phase change didnot occur in juvenile and rejuvenated plants grown under these two temperatureregimes. Rejuvenated plants grown in a greenhouse also exhibited a progressivedevelopment of adult leaf morphology, accompanied by an increase in leaf13C, an effect that was more pronounced insingle-stemmed (from –26.4% to ca. –24%) than in branched plants. Itis suggested that increasing 13C in juvenile andrejuvenated plants undergoing phase change is a result of reduced sink strengthin single-stemmed plants, and to a lesser extent within each branch of branchedplants, causing reduced stomatal conductance and photosynthesis.  相似文献

Gynophore elongation and pod formation were studied in peanut plants (Arachis hypogaea L.) under light and dark conditions in vivo. The gynophores elongated until pod formation was initiated. Pod (3–20 mm length) development could be totally controlled by alternating dark (switched on) and light (switched off) conditions, repeatedly. Gynophore elongation responded conversely to light/dark conditions, compared to pods. In this study we aimed to correlate the light/dark effects with endogenous growth substances. The levels of endogenous growth substances were determined in the different stags of pod development. Gynophores shortly after penetration into the soil, white gynophores, released twice the amount of ethylene as compared to the aerial green ones, or to gynophores bearing pods. Ethylene inhibitors had no effect on the percent of gynophores that developed pods, but affected pod size which were smaller compared to the control. A similar level of IAA was extracted from gynophore tips of green gynophores, white gynophores and pods. This paper presents evidence from an initial trial in favor of claims that the liquid seaweed extract of Ascophyllum nodosum, marketed in the UK as Maxicrop, can reduce numbers of the two-spotted red spider mite Tetranychus urticae on crop plants. Seven applications of Maxicrop Triple over 20 days significantly suppressed the population build-up of two-spotted red spider mites on treated strawberry plants (Fragaria sp. var. The paper presents a five-phase model to describe batch culture of Spirulina maxima under limitations of light and nutrients nitrogen andsulfur. The general equation for the exponential, linear, decelerating andstationary phases of the growth curve took into account that (i) the specificgrowth rate was proportional to the local light intensity in thephotobioreactor; (ii) light attenuation was due to cell's absorption andreflection and observed the Lambert-Beer's law with a total absorptioncoefficient (T) that was the sum of absorption coefficient(a) and reflection coefficient (r); (iii) thecomposition of the alga and its absorption a changed withtime and (iv) the specific growth rate was influenced by nutrients ofnitrogen and sulfur in the culture medium according to a Monod's law.Two successive equations describing the death phase were proposed basedon the hypothesis that the death rate accelerated with the %PSC until itreached its maximum. From that point the %PSC decreased and the deathrate reduced. Our model fitted very well the cultures grown in the photobioreactor developed in our laboratory.  相似文献

The membrane potential (pd) of duck weed (Lemna gibba G1) proved to be energy dependent. At high internal ATP levels of 74 to 105 nmol ATP g-1 FW, pd was between -175 and -265 mV. At low ATP levels of 23 to 46 nmol ATP g-1 FW, pd was low, about -90 to -120 mV at pH 5.7, but -180 mV at pH 8. Upon addition of glucose in the dark or by light energy the low pd recovered to the high values. The active component of the pd was depolarized by the addition of hexoses in the dark and in the light. Hexose-dependent depolarization of the pd (= pd) followed a saturation curve similar to active hexose influx kinetics. Depolarization of the pd recovered in the dark even in the presence of the hexoses and with a 10fold enhancement in the light. Depolarization and recovery could be repeated several times with the same cell. Glucose uptake caused a maximum depolarization of 133 mV, fructose uptake half that amount, sucrose had the same effect as glucose. During 3-O-methylglucose and 2-deoxyglucose uptake the depolarizing effect was only slightly lower. The pd remained unchanged in the presence of mannitol. The glucose dependent pd and especially the rate of pd recovery proved to be pH-dependent between pH 4 and pH 8. It was independent of the presence of 1 mM KCl. Su is a nuclear encoded, semi-dominant aurea mutation in Nicotiana tabacum L. The homozygous plants (Su/Su) are pale yellow and non-photosynthetic while the heterozygous (Su/+) are photosynthetically competent and have a yellow-green phenotype which is distinct from that of green wild-type plants (+/+). We have examined the RNA and protein levels for a number of nuclear and plastid encoded chloroplast proteins under high and low light plant growth conditions. Under high light conditions, the light-harvesting chlorophyll a/b binding proteins (LHCP) were undetectable in the homozygous Su/Su plants, and the large subunit (LSu) and the small subunit (SSu) of ribulose bisphosphate carboxylase (Rubisco) and cytochrome b559 were severely deficient. However, only the nuclear encoded cab and plastid encoded psbE mRNA (encoding LHCP and cytochrome b559 respectively) were reduced significantly. In heterozygous Su/+ plants, the level of LHCP was reduced to 25% of that in wild-type plants while cab and psbE mRNA, LSu, SSu and cytochrome b559 remained at normal levels, suggesting that LCHP is more immediately affected by the Su mutant gene product than the rest of the photosynthetic proteins and mRNA examined. Under low light conditions, the levels of cab and psbE mRNA, LSu, SSu and cytochrome b559 in homozygous Su/Su plants were equivalent to those in wild-type plants except LHCP which remained undetectable. Similarly, the LHCP level in low light grown Su/+ plants still remained at 25% of wild-type level. These results indicate that the decrease in LHCP is independent of light conditions and has not resulted from photooxidation, whereas the depletion of other proteins and mRNA examined under high light growth conditions is a consequence of photooxidative damage to Su/Su plastids. Furthermore, transgenic Su/Su and Su/+ plants with a cauliflower mosaic virus 35S (CaMV 35S)-cab construct constitutively maintained high levels of cab mRNA but displayed the same pattern of diminished LHCP accumulation as their non-transformed counterparts when grown under both high and low light conditions. These results indicate that the Su mutation primarily causes depletion of LHCP. The depletion of LHCP leads to photooxidative damage which results in decreased cab mRNA levels and other pleiotropic lesions in Su/Su plants.  相似文献

Anthocyanin formation in 36h dark grown Sinapis alba L. seedlings and inhibition of hypocotyl elongation in 36h and 54h dark grown and 54h and 7 day light grown seedlings in response to continuous red light could be substituted for by hourly 5 min light pulses where the total fluence over the irradiation period is the same. These pulses are partially (36h) or almost totally (54h and 7 day) reversible by subsequent far-red (RG 9) light pulses. In contrast to 654 nm light, hourly light pulses with 552 nm, 449 nm and 715 nm can at best only partially substitute for continuous irradiation. Phytochrome was studied spectrophotometrically in Avena sativa L. seedlings that had been grown for 6 d in continous white fluorescent light from lamps. Greening was prevented through the use of the herbicide San 9789. When placed in the light, phytochrome (Ptot) decreased with first order kinetics (1/2 2 h) but reached a stable low level (2.5% of the dark level) after 36 h. This concentration of phytochrome remained constant in the light and during the initial hours of a subsequent dark period, but increased significantly after a prolonged dark period. Evidence suggests that the constant pool of phytochrome in the light is achieved through an equilibrium between synthesis of the red absorbing (Pr) and destruction of the far-red absorbing form (Pfr) of phytochrome. Summary Synaptic ribbons (SR) in pinealocytes of adult (120–130 day-old) male Chinese hamsters (Cricetulus griseus) were classified into types 1, 2 and 3; these have a central dense structure showing rod-like, various and ringlike profiles, respectively. The central structure of the type-2 SR usually appeared as round, oval or comma-like bodies, and occasionally as plates showing various profiles or clubshaped bodies. The quantity of each type of SR, expressed as the SR index, was determined over a 24-h period under a light/dark regime (LD) 1212 or LD 1410. On comparing the results obtained from adults with previously published data from young (60–70-days-old) animals under LD 1212, it was found that, in both young and adult animals, the type-1 and type-3 SR indices exhibited different 24-h variations, whereas the type-2 SR index remained constant over a 24-h period. In addition, the indices of the type-2 SR, but not those of the other SR types, were found to be significantly larger in adult than in young animals. In adult animals, the effects of the photoperiod were different between the three types of SR. A nocturnal increase in the type-1 SR index was observed under both LD 1212 and LD 1410, its time course being different for each of these photoperiods. Under LD 1410, the type-2 SR index showed a significant 24-h rhythm with larger values during the dark period; this was not observed under LD 1212. The type-3 SR index was almost the same under LD 1212 and LD 1410. The results suggest that pinealocyte SR of the Chinese hamster may be composed of three types of SR, each with a different functional role.  相似文献

By analyzing the volatiles from Tetranychus urticae-infested kidney bean plants (Phaseolus vulgaris) at different times for two days, we found that they were mainly produced in the light. Tetranychus urticae showed a higher oviposition rate and spent more time feeding during the day (in the light) than at night (in the dark). Infested leaves placed in the light attracted the predatory mite Amblyseius womersleyi, whereas those that were placed in the dark for at least 2h in daytime did not. This indicates that presence or absence of light affects the production of herbivore-induced plant volatiles. Amblyseius womersleyi dispersed more frequently and consumed more T. urticae eggs during the day (in the light) than at night (in the dark), whereas their oviposition rate did not differ between day and night. Presence or absence of herbivore-induced plant volatiles in the surroundings did not affect dispersal, predation or oviposition rates of A. womersleyi. These results show that A. womersleyis behavior coincides with the production pattern of herbivore-induced plant volatiles.  相似文献

Induction by light of phenylalanine ammonia-lyase (PAL; EC 4.3.1.5) and of anthocyanin in cotyledons of the mustard (Sinapis alba L.) seedling is strongly affected by a light pretreatment which operates through phytochrome. If PAL or anthocyanin is induced by a light pulse, the effectiveness of phytochrome (Pfr) is strongly increased by a light pretreatment; however, if the increase of the PAL level or synthesis of anthocyanin is elicited by continuous far-red light (operating via phytochrome in the High Irradiance Response), effectiveness of light is strongly reduced by the same light pretreatment. This reduction of effectiveness is correlated with a decrease of total phytochrome (Ptot) caused by the light pretreatment. It is argued that the observations are compatible only with the open phytochrome-receptor model as suggested by Schäfer (J. Mathem. Biol. 2, 41–56, 1975). The peaks of the time courses of the PAL levels under continous far-red light are located at 48 h after sowing and do not depend on the original level of phytochrome. In light-dark-synchronized cultures of the unicellular green algaChlamydomonas reinhardtii, release of zoospores from the wall of the mother cell normally takes place during the second half of the dark period. The recently isolated mutant ls, however, needs light for the liberation of zoospores when grown photoautotrophically under a 12 h light-12 h dark regime. The light-induced release of zoospores was found to be prevented by addition of the photosystem-II inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Furthermore, light dependence of this process was shown to be abolished when the mutant ls was grown either photoautotrophically under a 14 h light-10 h dark regime or in the presence of acetate. Our findings indicate that the light-dependency of zoospore liberation observed in cultures of this particular mutant during photoautotrophic growth under a 12 h light-12 h dark regime might be attributed to an altered energy metabolism. The light-induced release of zoospores was found to be prevented by addition of cycloheximide or chloramphenicol, antibiotics which inhibit protein biosynthesis by cytoplasmic and organellar ribosomes, respectively. Actinomycin D, an inhibitor of RNA synthesis, however, did not affect the light-induced liberation of zoospores.Sporangia accumulate in stationary cultures of the mutant ls. Release of zoospores was observed when these sporangia were collected by centrifugation and incubated in the light after resuspension in fresh culture medium. Doubled haploid (DH) progeny from a cross between the scald susceptible barley (Hordeum vulgare L.) cultivar Ingrid and the resistant accession CI 11549 (Nigrinudum) was evaluated for resistance in the pathogen Rhynchosporium secalis (Oudem) J.J. Davis. Two linked and incompletely dominant loci confer resistance CI 11549 against isolate 4004. One is an allele at the complex Rrs1 locus on chromosome 3H close to the centromere; the other is located 22 cM distally on the long arm. The latter locus is designated Rrs4. In BC3-lines into Ingrid from CI 2222 (another Nigrinudum) resistance seems governed by one locus close to the telomeric region of chromosome 7H, probably allelic to Rrs2. In neither case did we find any trace of the recessive gene rh8 reported to be present in Nigrinudum. Various resistance donors of Ethiopian origin designated as Nigrinudum, Jet or Abyssinian were identical to a great extent with respect to markers, but differed in resistance to different isolates of scald or in barley yellow dwarf virus (BYDV) resistance. The implications for their use as differentials in scald tests and screening of germplasm collections are discussed.  相似文献

Exogenous gibberellin removes the genetical suppression of mesocotyl elongation in dark-grown seedlings of the rice cultivar Nihon Masari (japonica type). This gibberellin effect can be cancelled by light. All light effects can be accounted for by phytochrome. Dose-response and fluence-response studies show that phytochrome induces a reduction of the sensitivity to exogenous gibberellins. A cytological analysis of cell elongation and cortical microtubules led to a model where gibberellin and red light regulate mesocotyl elongation by controlling microtubule orientation in the epidermis of the mesocotyl. This causes corresponding changes of cellular extension growth, which can account for a large part of the observed growth responses. Comparative studies involving antimicrotubular drugs and gibberellin-synthesis inhibitors in the rice cultivar Kasarath (indica type) and a hybrid cultivar suggest that some of the differences between the cultivars are due to differences in gibberellin-sensitivity.  相似文献

Chlorella pyrenoidosa was grown in three continuous cultures each receiving a different light regime during the light period of a diurnal cycle. Hourly samples taken during the light period were subjected to medium frequency light/dark oscillations of equal duration, ranging from 3 to 240 seconds. The oxygen consumption and production of each sample were measured with an oxygen electrode in a small oxygen chamber. Although the light/dark cycles had little overall influence on photosynthetic activity, the microalgae appeared to adapt to the light regime to which they were subjected. Large differences were found between the maximum chlorophyll-specific production rates (P infmax supB ), the chlorophyll-specific production rates (PB) and the respiration rates between the cultures and treated subsamples. Respiration rates increased during the light period, whilst PB either increased, or had a mid light period minimum or maximum. The culture which received an hourly light oscillation during the light period had the highest P infmax supB and lowest respiration rates, and it is suggested that these algae react as in nature, whereas either a sinusoidal or a block light pattern is unnatural. The latter light regime is commonly used in laboratory studies.  相似文献
