Ascending and descending projections to the inferior colliculus in the rat   总被引:1,自引:0,他引:1  
The ascending and descending projections to the central nucleus of the inferior colliculus (IC) were studied with the aid of retrograde transport of horseradish peroxidase (HRP). HRP-labelled cells were found in contralateral cochlear nuclei, where the majority of different cell types was stained. Few labelled cells were observed in the ipsilateral cochlear nuclei. HRP-positive neurones were found in all nuclei of the superior olivary complex on the ipsilateral side with the exception of the medial nucleus of the trapezoid body, which was never labelled either ipsilaterally or contralaterally. The largest concentration of HRP-labelled cells was usually observed in the ipsilateral superior olivary nucleus. Smaller numbers of labelled cells were present in contralateral nuclei of the superior olivary complex. Massive projections to the inferior colliculus were found from the contralateral and ipsilateral dorsal nucleus of the lateral lemniscus and ipsilateral ventral nucleus of the lateral lemniscus. Many neurones of the central and external nuclei of the contralateral inferior colliculus were labelled with HRP. Topographic organisation of the pathways ascending to the colliculus was expressed in the cochlear nuclei, lateral superior olivary nucleus and in the dorsal nucleus of the lateral lemniscus. HRP--positive cells were found in layer V of the ipsilateral auditory cortex, however, the evidence for topographic organisation was lacking.  相似文献

A cytophotometric study of sections stained with gallocyanin and chrome alum showed that monaural stimulation for 2 h and binaural stimulation for 1.5 h with rhythmic noise signals led to a marked increase in the cytoplasmic RNA content per cell in the principal and large multipolar neurons of the dorsal and ventral parts of the ventrolateral region of the central nucleus of the inferior colliculus. The increase in cytoplasmic RNA content in the principal cells of the ipsiand contralateral parts of this nucleus relative to the stimulated ear in the case of monaural stimulation and the increase in RNA content in response to binaural stimulation suggests a uniform distribution of bilaterally converging connections from the lower nuclei of the auditory system on the principal cells. The increase in cytoplasmic RNA in the large multipolar cells of the contralateral central nucleus in response to monaural stimulation is evidence of the predominantly contralateral projection to these cells. The results are evidence of convergence of binaural influences on the principal and large multipolar cells of the central nucleus of the inferior colliculus.I. P. The EEG of 10 right-handed subjects preceding saccades with mean values of latent periods were selected and averaged. Two standard paradigms of presentation of visual stimuli (central fixation stimulus-peripheral target succession): with a 200-ms inerstimulus interval (GAP) and successive single step (SS). During the period of central fixation, two kinds of positive potentials were observed: fast potentials of "inermediate" positivity (IP) developing 600-400 ms prior to saccade onset and fast potentials of "leading" positivity (LP), which immediately preceded the offset of the central fixation stimulus. Peak latency of the LP potentials was 300 ms prior to saccade onset in the SS paradigm and 400 ms in the GAP paradigm. These potentials were predominantly recorded in the frontal and frontosagittal cortical areas. Decrease in the latency by 30-50 ms in the GAP paradigm was associated with more pronounced positive potentials during the fixation period and absence of the initiation potential P-1' (or decrease in its amplitude). The obtained evidence suggest that the fast positive presaccadic potentials are of a complex nature related to attention, anticipation, motor preparation, decision making, saccadic initiation, and backward afferentation.  相似文献

Previous studies have demonstrated that despite its blindness, the subterranean blind mole rat (Spalax ehrenbergi) possesses a noticeable lateral geniculate nucleus and a typical cyto-architectural occipital cortex that are reciprocally connected. These two areas, as revealed by the metabolic tracer 2-deoxyglucose, are activated by auditory stimuli. Using single unit recordings, we show that about 57% of 325 cells located within the occipital cortex of anesthetized mole rats responded to at least one of the following auditory stimuli — white noise, pure tones, clicks, and amplitude modulated tones — with the latter two being the most effective. About 85% of cells driven by either contralateral or ipsilateral stimulation also responded to binaural stimulation; about 13% responded only to binaural stimulation; and 2% were driven exclusively by contralateral stimulation. Comparing responsiveness and response strength to these three modes of stimulation revealed a contralateral predominance. Mean latency (±SD) of ipsilateral and contralateral responses were 48.5±32.6 ms and 33.5±9.4 ms, respectively. The dorsomedial motor nuclei were demonstrated by the cobalt-labeling technique applied to the so-called somatic motor cranial nerves. The motoneurons constituting these nuclei are oval-shaped and smaller than the motoneurons in the ventrolateral motor nuclei. They give rise to ventral and dorsal dendrite groups which have extensive arborization areas. A dorsolateral cell group in the rostral three quarters of the oculomotorius nucleus innervates ipsilateral eye muscles (m.obl.inf., m.rect.inf., m.rect.med.) and a ventromedial cell group innervates the contralateral m. rectus superior. Ipsilateral axons originate from ventral dendrites, contralateral axons emerge from the medial aspect of cell bodies, or from dorsal dendrites, and form a "knee" as they turn around the nucleus on their way to join the ipsilateral axons. A few labeled small cells found dorsal and lateral to the main nucleus in the central gray matter are regarded as representing the nucleus of Edinger-Westphal. The trochlearis nucleus is continuous with the ventromedial cell group of the oculomotorius nucleus. The axons originate in dorsal dendrites, run dorsally along the border of the gray matter and pierce the velum medullare on the contralateral side. A compact dendritic bundle of oculomotorius neurons traverse the nucleus, and side branches appear to be in close apposition to the trochlearis neurons. A dorsomedial and a ventrolateral cell group becomes labeled via the abducens nerve. The former supplies the m. rectus lateralis, while the latter corresponds to the accessorius abducens nucleus which innervates the mm. rectractores. Neurons in this latter nucleus are large and multipolar, resembling the neurons in the ventrolateral motor nuclei. Their axons originate from dorsal dendrites and form a "knee" around the dorsomedial aspect of the abducens nucleus. Cobalt applied to the hypoglossus nerve reaches a dorsomedial cell group (the nucleus proper), spinal motoneurons and sympathetic preganglionic neurons. Of the dorsomedial motor cells, the hypoglossus neurons are the largest, and a branch of their ventral dendrites terminates on the contralateral side. Some functional and developmental biological aspects of the morphological findings, such as the crossing axons and the peculiar morphology of the accessory abducens nucleus, are discussed.  相似文献

A new technique for studying the physiology and pharmacology of locust central neurones is described. Somata isolated from neurones in the meso and metathoracic ganglia of third instar locusts (Schistocerca gregaria) were maintained for up to 4 weeks in co-culture (monolayer) with embryonic locust neurones. Most of the cultured cells became multipolar but a few were monopolar like their in vivo counterparts. They had diameters of 40-80 microns and "clean" (glial free) surface membranes. Cells 6-14 days in vitro were depolarized by acetylcholine and usually hyperpolarized by gamma-aminobutyrate, taurine and glycine. L-Glutamate and L-aspartate were inactive but further pharmacological studies are required to confirm this. Cultured larval neurones should provide excellent opportunities to study the molecular basis for drug-receptor interactions and voltage-sensitive membrane channels using the patch clamp technique.  相似文献

Metastable epialleles (MEs) are mammalian genomic loci where epigenetic patterning occurs before gastrulation in a stochastic fashion leading to systematic interindividual variation within one species. Importantly, periconceptual nutritional influences may modulate the establishment of epigenetic changes, such as DNA methylation at MEs. Based on these characteristics, we exploited Infinium HumanMethylation450 BeadChip kits in a 2-tissue parallel screen on peripheral blood leukocyte and colonic mucosal DNA from 10 children without identifiable large intestinal disease. This approach led to the delineation of 1776 CpG sites meeting our criteria for MEs, which associated with 1013 genes. The list of ME candidates exhibited overlaps with recently identified human genes (including CYP2E1 and MGMT, where methylation has been associated with Parkinson disease and glioblastoma, respectively) in which perinatal DNA methylation levels where linked to maternal periconceptual nutrition. One hundred 18 (11.6%) of the ME candidates overlapped with genes where DNA methylation correlated (r > 0.871; p < 0.055) with expression in the colon mucosa of 5 independent control children. Genes involved in homophilic cell adhesion (including cadherin-associated genes) and developmental processes were significantly overrepresented in association with MEs. Additional filtering of gene expression-correlated MEs defined 35 genes, associated with 2 or more CpG sites within a 10 kb genomic region, fulfilling the ME criteria. DNA methylation changes at a number of these genes have been linked to various forms of human disease, including cancers, such as asthma and acute myeloid leukemia (ALOX12), gastric cancer (EBF3), breast cancer (NAV1), colon cancer and acute lymphoid leukemia (KCNK15), Wilms tumor (protocadherin gene cluster; PCDHAs) and colorectal cancer (TCERG1L), suggesting a potential etiologic role for MEs in tumorigenesis and underscoring the possible developmental origins of these malignancies. The presented compendium of ME candidates may accelerate our understanding of the epigenetic origins of common human disorders.  相似文献

Summary Movements of the femoro-tibial joint of a locust hind leg are monitored by three classes of proprioceptors; a chordotonal organ (Usherwood et al. 1968), multipolar joint receptors (Coillot and Boistel 1968) and a strand receptor innervated by a single afferent with a central cell body (Bräunig 1985). All three classes are excited by imposed or voluntary extension of the tibia. The strand receptor (fe-tiSR) spikes tonically and at a frequency dependent upon the position of the joint whilst the multipolar joint receptors give overlapping information but for a more restricted range. The afferent from the strand receptor makes an excitatory connection with a spiking local interneurone in the midline group of the metathoracic ganglion. The central latency and consistency with which the EPSP follows each sensory spike suggests that the connection is direct. This interneurone also receives convergent inputs from neurones in the chordotonal organ, but not from multipolar joint receptors. Neither the strand receptor nor the multipolar joint receptors apparently synapse upon leg motor neurones that we have tested, in contrast to receptors in the chordotonal organ.  相似文献

The effects of ketamine on the activities of the mylohyoid nerve (a branch of the Vth nerve) and of both phrenic nerves were investigated in rabbits anaesthetized with halothane, paralyzed and artificially ventilated. Intravenous administration of ketamine elicited a marked prolongation of the phrenic inspiratory discharge (without significantly affecting its amplitude) and a depression of the mylohyoid expiratory activity. An elimination of the volume-related input from the lungs ("no-inflation manoeuvre") or deflation elicited under these conditions typical apneustic pattern of breathing. The response to tracheal occlusion at peak-inspiration was "classical". We conclude that ketamine inhibits the Vth nerve motor nucleus which is not only an important component of the central inspiratory-inhibitory neurones but also a "relay station" between the vagal and the central inspiratory "off-switch" mechanisms.  相似文献

By means of intracellular injection of the dye Lucifer yellow processes were revealed of the snail command neurones of escape behaviour LPa3 and RPa3 in pedal nerves ipsilateral to these neurones. A process of the neurone LPa3 was also found in the contralateral Nervus cutaneus pedalis secundus. Blockade of central chemical synapses by cadmium chloride did not lead to disappearance of motor reactions evoked by electrical stimulation of LPa3 and RPa3 neurones both on ipsi- and contralateral sides of the animal foot; this allowed to suggest a presence in the contralateral muscular pedal nerves of these neurones processes unrevealed by the used dyeing method.  相似文献

Summary Substance-P immunoreactivity has been located in semithin sections of mouse hypothalami and pituitaries and compared with the distribution of other hypothalamic peptides. In the mouse, nerve fibres and terminals reacting with antibodies against substance P (SP) were detected both in the external zone of the median eminence (ME) and in the neural lobe of the pituitary. Immunoreactive SP (ISP) axons of the ME did not react with antibodies against other peptides, i.e. arginine-vasopressin (AVP), oxytocin (OT), somatostatin and enkephalin, and were also negative with an antibody to serotonin. In the neural lobe, SP immunostaining occurred in AVP but not in OT axons. In the hypothalamus, ISP axons were widespread but conspicuously lacking in areas containing AVP neurones, i. e. in the suprachiasmatic nucleus and the clusters of AVP cells in the SO and PV nuclei. In contrast, multiple ISP endings were seen in contact with OT neurones. Immunoreactive cell bodies, only detected after colchicine treatment, belonged to two distinct classes of neurones: 1) single AVP neurones of the SO and PV nuclei; 2) specific (staining only for SP) neurones, scattered or grouped in different areas of the hypothalamus, not showing relationship with any circumscribed nucleus. These results reinforce the opinion that SP can be released as a neurohormone into the vascular portal system and can directly affect the pars distalis. The presence of immunoreactive SP in the neural lobe, which has not been reported in species other than the mouse, may have a different physiological significance.  相似文献

We examined factors that affect spatial receptive fields of single units in the central nucleus of the inferior colliculus of Eptesicus fuscus. Pure tones, frequency- or amplitude-modulated sounds, or noise bursts were presented in the free-field, and responses were recorded extracellularly. For 58 neurons that were tested over a 30 dB range of sound levels, 7 (12%) exhibited a change of less than 10° in the center point and medial border of their receptive field. For 28 neurons that were tested with more than one stimulus type, 5 (18%) exhibited a change of less than 10° in the center point and medial border of their receptive field.The azimuthal response ranges of 19 neurons were measured in the presence of a continuous broadband noise presented from a second loudspeaker set at different fixed azimuthal positions. For 3 neurons driven by a contralateral stimulus only, the effect of the noise was simple masking. For 11 neurons driven by sound at either side, 8 were unaffected by the noise and 1 showed a simple masking effect. Summary The central projections of ocellar interneurones in two species of trichopterous insects Agrypnia varia F. and Limnephilus flavicornis F. were analysed by use of cobalt iontophoresis. The interneurones were classified into three groups: large-, medium- and small-caliber neurones based on the diameters of the axons. Seven large-diameter neurones project from each lateral ocellus into the central nervous system. Of these, four neurones terminate in the posterior slope (three ipsilateral and one contralateral). Three neurones possess branches in the contralateral posterior slope and proceed down the cervical connective into the thoracic ganglia. Medium-sized neurones connect the neuropiles of the three ocelli to each other. Small-diameter neurones contact the contralateral lobula and medulla of the optic lobes and connect the three ocellar neuropiles. Large-diameter neurones of the median ocellus were found to terminate bilaterally or ipsilaterally in the posterior slope. In the posterior slope four different subregions can be recognised: (1) the dorso-lateral, (2) the ventro-lateral, (3) the lateral, into which large-diameter interneurones of the lateral ocelli send branches, and (4) the medial, innervated by interneurones of the median ocellus. Objective

Interaural level difference (ILD) is the difference in sound pressure level (SPL) between the two ears and is one of the key physical cues used by the auditory system in sound localization. Our current understanding of ILD encoding has come primarily from invasive studies of individual structures, which have implicated subcortical structures such as the cochlear nucleus (CN), superior olivary complex (SOC), lateral lemniscus (LL), and inferior colliculus (IC). Noninvasive brain imaging enables studying ILD processing in multiple structures simultaneously.

Methods

In this study, blood oxygenation level-dependent (BOLD) functional magnetic resonance imaging (fMRI) is used for the first time to measure changes in the hemodynamic responses in the adult Sprague-Dawley rat subcortex during binaural stimulation with different ILDs.

Results and Significance

Consistent responses are observed in the CN, SOC, LL, and IC in both hemispheres. Voxel-by-voxel analysis of the change of the response amplitude with ILD indicates statistically significant ILD dependence in dorsal LL, IC, and a region containing parts of the SOC and LL. For all three regions, the larger amplitude response is located in the hemisphere contralateral from the higher SPL stimulus. These findings are supported by region of interest analysis. fMRI shows that ILD dependence occurs in both hemispheres and multiple subcortical levels of the auditory system. This study is the first step towards future studies examining subcortical binaural processing and sound localization in animal models of hearing.  相似文献

The supra-oesophageal ganglion (brain) of Schistocerca gregaria Forskál has been studied at the light-microscopical level using Wigglesworth's osmic acid-ethyl gallate method. Particular attention was paid to the midbrain. A structural ground-plan is described which incorporates the ocellar nerve roots, the antenno-glomerular bundles, the corpora cardiaca nerves I roots, the antennal lobes, the corpora pedunculata and the central complex. The "undifferentiated" midbrain is described in terms of the distribution of the main features of 15 unique pairs of large neurones all of which have their cell bodies in the brain and which project, either ipsilaterally or contralaterally, to the circumoesophageal connectives. In addition a single unique neurone with bilateral distribution restricted to the brain is described. All these neurones provide a conspicuous and constant framework for further investigation. A ground-plan based on the distribution of 64 individual neurones (central complex system I) is presented for the central complex. This system indicates that the central complex is fundamentally organized on the basis of 16 repetitive groups of neurones.  相似文献

Experiments were performed in 13 chloralose-anaesthetized, curarized cat preparations (monitoring of rectal temperature, heart rate, expired pCO2 and EEG), in order to ascertain whether, and to what extent, the reactivity to ipsilateral skin shocks of the neurones of the anterior ectosylvian and anterior suprasylvian gyri (AEG and ASG, respectively) is dependent on the callosal output of the somatosensory areas of the contralateral hemisphere. Indeed, we knew from previous experiments that a high proportion of AEG and ASG neurones having bilateral peripheral receptive fields (PRFs) can be excited by direct stimulation of the contralateral homonymous areas, and that the callosal fibres originating in the latter carry somesthetic impulses related to contralateral PRFs. A preliminary analysis was carried out on the amplitude and latency relationships between the evoked potentials (EP) recorded simultaneously from the two hemispheres and from the corpus callosum (CC) following stimulation of the forepaw of one side. The results obtained showed good correlations between the onset and development of the EPs picked up from the hemisphere ipsilateral to the stimulated skin, on the one hand, and onset and development of the EPs recorded from the contralateral hemisphere and the corpus callosum, on the other. At a further stage of the experiments, the EPs elicited upon ipsilateral and contralateral skin shocks in the AEG-ASG area have been recorded and averaged before, during and after the reversible inactivation of callosal somesthetic transmission. This was achieved by applying polarizing currents (0.2-1 mA) to the rostral portion of the CC, adequacy and reversibility of this method having been tested by observing, respectively, suppression and prompt restoration of transcallosal EPs and of the asynaptic spiking produced by cortical cells when antidromically invaded from contralateral homotopic cortex. It was seen that during CC blockade the EPs elicited in the AEG-ASG areas did not show any change either in amplitude or time-course if brought about by contralateral peripheral stimulation, whereas those evoked by ipsilateral skin shocks exhibited significant reduction, which was related to the strength of CC polarization and to the reduction of transcallosal EPs. In control experiments similar effects were observed after ablation of somatosensory areas of the hemisphere which send off somesthetic callosal impulses, whereas strychninization of these areas caused effects opposite in sign, i.e., enhancement of the ipsilateral but not of the contralateral EPs in the areas of the untreated hemisphere. By testing the effects of CC polarization on single AEG-ASG neurones, it was observed that the responses of units linked only with contralateral PRFs (Group I; 7 units tested) were unaffected by callosal polarization. The discharges of neurones provided with wide and bilateral PRFs (Group II; 27 units tested) were not affected if elicited by contralateral PRF shocks but were deeply impaired (in 11 neurones out the 27) when provoked by ipsilateral PRF stimulation. The effect consisted chiefly of the disappearance of the first high peak of the PSTHs, and when recording intracellularly graded events, it was mirrored by a large decrease of the postsynaptic excitatory potentials elicited in Group II neurones by ipsilateral PRF shocks. A late scattered histographic component was identified in the PSTHs of such cells, which did not appear to be significantly altered during CC blockade. These effects were observed on the ipsilateral responses of 11 out of the 27 Group II neurones so tested whereas the ipsilateral PSTHs of the remaining 16 Group II neurones either did not undergo significant changes during the callosal blockade or escaped evaluation because of high spontaneous shifts of neural responsiveness. The results are discussed mainly with a view to the possible functional role of the specific somesthetic callosal fibres in defining ipsilateral reactivity for the wide-field cells of the AEG-ASG area.  相似文献

The present study was focused on the morphology of the diencephalic nuclei (likely involved in reproductive functions) as well as on the distribution of GnRH (gonadotropin-releasing hormone) in the rhinencephalon, telencephalon and the diencephalon of the brain of bluefin tuna (Thunnus thynnus) by means of immunohistochemistry. Bluefin tuna has an encephalization quotient (QE) similar to that of other large pelagic fish. Its brain exhibits well-developed optic tecta and corpus cerebelli. The diencephalic neuron cell bodies involved in reproductive functions are grouped in two main nuclei: the nucleus preopticus-periventricularis and the nucleus lateralis tuberis. The nucleus preopticus-periventricularis consists of the nucleus periventricularis and the nucleus preopticus consisting of a few sparse multipolar neurons in the rostral part and numerous cells closely packed and arranged in several layers in its aboral part. The nucleus lateralis tuberis is located in the ventral-lateral area of the diencephalon and is made up of a number of large multipolar neurones. Four different polyclonal primary antibodies against salmon (s)GnRH, chicken (c)GnRH-II (cGnRH-II 675, cGnRH-II 6) and sea bream (sb)GnRH were employed in the immunohistochemical experiments. No immunoreactive structures were found with anti sbGnRH serum. sGnRH and cGnRH-II antisera revealed immunoreactivity in the perikarya of the olfactory bulbs, preopticus-periventricular nucleus, oculomotor nucleus and midbrain tegmentum. The nucleus lateralis tuberis showed immunostaining only with anti-sGnRH serum. Nerve fibres immunoreactive to cGnRH and sGnRH sera were found in the olfactory bulbs, olfactory nerve and neurohypophysis. The significance of the distribution of the GnRH-immunoreactive neuronal structures is discussed.  相似文献

OBJECTIVE: To examine the validity of the presence of mucin, utilizing a semiquantitative, statistical approach, as a criterion for distinguishing low to intermediate grade mucoepidermoid carcinoma (ME) from Warthin's tumor (WT). STUDY DESIGN: We selected histologically established fine needle aspiration (FNA) cases of ME (26) and WT (30) from a six-year period (January 1995-January 2001). Mucicarmine staining was performed on the FNA preparations. The amount of mucin detected was rated on a scale of 1-3, as follows: 1 = no mucin present, 2 = mucin present in the background and < 5% of tumor cells, and 3 = mucin present in the background and > or = 5% tumor cells. The scores for the 30 WTs and 26 MEs were tabulated and analyzed for statistical significance utilizing the Student t test. RESULTS: The mean mucin score for the 26 MEs was 1.81, SD .63. The mean mucin score for the 30 WTs was 1.52, SD .74. There was no statistically significant difference (P = .122) in the amount of mucin present between MEs and WTs in the 56 aspirate specimens examined. CONCLUSION: Based on the results of this study, the presence of extracellular or intracellular mucin in FNA specimens of salivary gland tumors may not be a reliable criterion for distinguishing WT from ME.  相似文献
