We have previously shown that a range of nicotinamide containing ‘biomimetic coenzymes’ function as active analogues of NAD+ in the oxidation of alcohols by horse liver alcohol dehydrogenase (HLADH), despite their apparently astonishing lack of structural similarity to the natural coenzyme. The simplest structure as yet shown to exhibit activity is the biomimetic coenzyme CL4. To investigate the effect of the structure of this truncated artificial coenzyme on its activity, a range of close structural analogues of CL4 were designed, synthesized and characterized. The electrochemical reduction potentials of the analogues were strongly influenced by the nature of the groups attached to the pyridine ring. All of the analogues could be chemically reduced using sodium borohydride, to give compounds with altered UV‐visible absorption and fluorescence properties. An HPLC‐based assay suggested that two of the new analogues were coenzymically active in the oxidation of butan‐1‐ol by HLADH, with one displaying a significantly higher activity than CL4. The results demonstrate which features of the structures of the coenzymes lead to desirable electrochemical and spectroscopic properties, but suggest that the structural requirements for a functional coenzyme are quite stringent. 21,31-O-(Propane-1,2-diyl)cyclomaltoheptaose has been prepared from 2-O-allylcyclomaltoheptaose by mercuration in trifluoroacetic acid, followed by reduction with sodium borohydride. 2-O-(2,3-Epoxypropyl)cyclomaltoheptaose, prepared from 2-O-allylcyclomaltoheptaose by oxidation with dimethyldioxirane, was converted into 21,31-O-(3-hydroxypropane-1,2-diyl)cyclomaltoheptaose by treatment with trifluoroacetic acid. Both derivatives containing fused 1,4-dioxane rings are mixtures of stereoisomers, in which the methyl and hydroxymethyl group, respectively, that is linked to this ring, occupies an axial or an equatorial position.  相似文献

Pig heart succinate-coenzyme A transferase (succinyl-coenzyme A: 3-oxoacid coenzyme A transferase; E. C. 2.8.3.5.), a dimeric enzyme purified by affinity chromatography on Procion Blue MX-2G Sepharose, reacts with acetoacetyl-coenzyme A to form a covalent enzyme-coenzyme A thiolester intermediate in which the active site glutamate (E344) of both subunits each forms thiolester links with coenzyme A. Reaction of this dimeric enzyme-coenzyme A species with sodium borohydride leads to inactivation of the enzyme and reduction of the thiolester on both subunits to the corresponding enzyme alcohol, as judged by electrospray mass spectrometry. Reaction of the dimeric enzyme-coenzyme A intermediate with either succinate or acetoacetate, however, results in only one-half of the coenzyme A being transferred to the acceptor carboxylate to form either succinyl-coenzyme A or acetoacetyl-coenzyme A. Reaction of this latter enzyme species with borohydride caused no loss of enzyme activity despite the reduction of the remaining half of the enzyme-coenzyme A thiolester to the enzyme alcohol. That this catalytic asymmetry existed between subunits within the same enzyme dimer was demonstrated by showing that the enzyme species, created by successive reaction with acetoacetyl-coenzyme A and succinate, bound to Blue MX-2G Sepharose through the remaining available active site and could be eluted as a single chromatographic species by succinyl-coenzyme A. It is concluded that while both of the subunits of the succinate-coenzyme A transferase dimer are able to form enzyme-coenzyme A thiolester intermediates, only one subunit is competent to transfer the coenzyme A moiety to a carboxylic acid acceptor to form the new acyl-coenzyme A product. The possible structural basis for this catalytic asymmetry and its mechanistic implications are discussed.  相似文献

The metallochromic indicator Eriochrome Blue SE was used to measure light-induced internal movement of Mg2+ in intact chloroplasts. By dual-wavelength spectroscopy (measuring wavelength 554 nm, reference 592 nm) a light-induced, dark-reversible absorbance increase of Eriochrome Blue in samples of isolated intact chloroplasts was observed. The light/dark difference spectrum of Eriochrome Blue between 550 and 590 nm (reference wavelength 562 nm) indicated that this absorbance increase was caused by an increased concentration of free Mg2+ in a neutral or slightly alkaline chloroplast compartment.

The signal was seen only with intact, but not with broken, envelope-free chloroplasts, which had lost most of their divalent cations. This is interpreted to show that the indicator responds to an increase of Mg2+ concentration in the chloroplast stroma, which represents an efflux of Mg2+ from the intra-thylakoid space caused by light-dependent proton pumping.

As calculated from corrected values of the absorbance increase of Eriochrome Blue, the light-induced internal release of Mg2+ was close to 100 nequiv per mg chlorophyll at pH 7.6 and 250 nequiv at pH 7.1. This corresponds to a light-dependent increase in the concentration of free Mg2+ in the stroma of about 2 and 5 mM, respectively.  相似文献

Cuaq+ forms stable complexes with carbon monoxide in aqueous solutions. Furthermore it reacts very fast with aliphatic radicals. The reaction of Cu(CO)maq+ with methyl radicals, CH3 was studied using the pulse-radiolysis technique. The results point out that methyl radicals react with Cu(CO)aq+ to form an unstable intermediate with a CuII-C σ bond identified as (CO)CuII-CH3+, k = (1.1±0.2) × 109 M−1 s−1. This intermediate has a strong LMCT charge transfer band (λmax = 385 nm, max = 2500 M−1 cm−1) which is similar to the absorption bands of other transient complexes with CuII-alkyl σ bonds. The coordinated carbon monoxide in (CO)CuII-CH3+ inserts into the copper—carbon bond (or rather the coordinated methyl migrates to the coordinated carbon monoxide ligand) at a rate of (3.0±0.8) × 102 s−1 to form the copperacetyl complex (CO)mCuII-C(CH3)=O+max = 480 nm, max = 2100 M−1 cm−1). The rate of formation of (CO)CuII-CH3+ and of the insertion reaction are pH independent. The complex (CO)mCuII-C(CH3)=O+ is also unstable and decomposes heterolytically to yield acetaldehyde and Cuaq2+ as the final stable products. This reaction is slightly pH dependent. The same reactivity pattern has been observed for the Cu(COnaq+ complexes (n = 2 or 3). The results clearly point out that CO remains coordinated to transient complexes of the type CuII-alkyl.  相似文献

Homogeneous class II alcohol dehydrogenase (pi-ADH) has been isolated from human liver homogenates by chromatography on DE-52 cellulose, 4-[3-[N-(6-amino-caproyl)amino]propyl]pyrazole-Sepharose, SP-Sephadex C-50, and agarose-hexane-AMP, yielding an enzyme that has a significantly higher specific activity and is markedly more stable than that isolated by an earlier procedure. pi-ADH is composed of two identical 40 000-dalton subunits, contains 4 mol of zinc/dimer, and is readily inhibited by metal-chelating agents. The purified enzyme binds two molecules of coenzyme per dimer, exhibits an absorption maximum at 280 nm, epsilon 280 = 57 000, and exhibits an isoelectric point of 8.6. The class II isozyme catalyzes the oxidation of a variety of alcohols with Km values ranging from 7 microM to 560 mM and with kcat values from 32 min-1 to 600 min-1 and demonstrates a preference for hydrophobic substrates. The kcat/Km ratio for ethanol oxidation exhibits a pH maximum at 10.4.  相似文献

The behaviour of Pichia stipitis, Pachysolen tannophilus, Candida shehatae and Candida parapsilosis was investigated to select the most suitable yeast to convert xylose either to ethanol or to xylitol, with little or no formation of by-products. The aeration rate was used as a variable parameter. P. stipitis and C. parapsilosis were the most effective producers or ethanol and xylitol, respectively, both reaching productivities at very low levels of oxygenation. With P. stipitis, better ethanol productivity was attained under microaerobic conditions (KLa = 4·8 h−1) while with C. parapsilosis high yields and rates of xylitol production were detected at KLa values of about 16·3 h−1. P. tannophilus and C. shehatae showed lower performances under all conditions used while changes in oxygenation modified the ratio of ethanol to xylitol produced by these yeasts, suggesting that they are more dependent on the oxygen power input than P. stipitis and C. parapsilosis. The influence of oxygen transfer rates on ethanol and xylitol formation with the best producers is discussed.  相似文献

Abstract A cytochrome aa3 terminal oxidase was isolated from protoplast membrane vesicles of Micrococcus luteus grown under aerobic conditions. The purified complex showed similarities to cytochrome c oxidase (EC 1.9.3.1) of the electron transport chain of mitochondria and many prokaryotes. The enzyme was solubilized by subsequent treatment with the detergents CHAPS and n-dodecyl-β-d-maltoside and purified by ion-exchange chromatography using poly-L-lysine agarose and TMAE-fractogel-650 (S) columns, followed by hydroxyapatite chromatography. The purified complex is composed of two major subunits with apparent molecular masses of 54 and 32 kDa. After purification the isolated enzyme contains 12.1 nmol of heme A (mg protein)−1 and exhibits absorption maxima at 424 nm and 598 nm in the oxidized state and at 442 nm and 599 nm in the reduced state. The CO-difference spectrum shows peaks at 428 and 590 nm which is indicative of heme a 3, furthermore oxygen consumption was found to be sensitive to cyanide.  相似文献

A new functional macrocyclic ligand, 2,4-dinitrophenylcyclen (= 1-(2,4-dinitrophenyl)-1,4,7,10-tetraazacyclododecane), has been synthesized and isolated as its trihydrochloric acid salt (L·3HCl). The protonation constants (log Kn) for three secondary nitrogens of L were determined by potentiometric pH titration to be 10.10, 7.33 and <2 with I = 0.10 (NaNO3) at 25°C. The 2,4-dinitrophenylaniline chromophore was proven to be a good reporter signaling proton- and metal-binding events in the macrocyclic cavity. The UV absorption band (λmax 370 nm, 8200) of the 2,4-dinitrophenylaniline moiety at pH ≥ 9 becomes quenched as pH is lowered (to pH 3.1, where the major species is L·2H+), due to the strong protonation effect extended to the aniline moiety within the macrocyclic cavity. This is in sharp contrast to the pH-independent UV absorption (λmax 390 nm, 14 000) of a reference compound, N,N-diethyl-2,4-dinitroaniline. The UV absorption band of L is shifted to lower wavelengths with Zn2+max 320 nm), Cd2+max 316 nm) and Pb2+max 317 nm), while it almost disappears with Cu2+ and Ni2+. The 1:1 Zn2+ and Cu2+ complexes with L were isolated and characterized. The Zn2+ complex recognizes 1-methylthymine anion (MT) in aqueous solution at physiological pH to yield a stable ternary complex ZnL-MT. The X-ray crystal structure of ZnL-MT showed that Zn2+ is four-coordinate with three secondary nitrogens of L and the deprotonated imide anion that is cofacial to the 2,4-dinitrophenyl ring.  相似文献

An all-solid-state pellet electrode for ethanol determination has been developed and compared with the membrane-layered sensor. It is found that the new pellet electrodes have reliable responses (current vs. concentration) to ethanol from 0.1 to 10 m with response times of about 2 min. The current response decreases about 10% h−1 during continuous operation, compared with a drop of 50% h−1 with normal membrane electrodes. Fresh electrodes can be stored in a freezer for 2 weeks without apparent activity loss. The measurement procedure is convenient and it is probable that reproducible, disposable electrodes can be made at low cost. This format is general and can easily be extended to many other systems with β-NAD+/NADH as coenzyme.  相似文献

Low concentrations of metals of the first row transition series, Zn2+, Co2+, Mn2+, Ni2+, Cu2+ and Fe2+, and to a lesser extent the group IIa ions, particularly Mg2+, influenced the interaction of firefly luciferase [Photinus luciferin:oxygen 4-oxidoreductase (decarboxylating, ATP-hydrolysing), EC 1.13.12.7] with a number of triazine dyes. For example Cu2+ promoted the binding of luciferase to Cibacron Brilliant Blue (BR-II) and Cibacron Blue F3GA a dichloro and monochloro triazine dye, respectively. On the other hand Zn2+ prevented dye inactivation and even enhanced the enzyme activity. Specificity was observed in the interference of different metals interacting with different dye-protein. 彩色豆马勃能与松树、桉树形成外生菌根,研究其次生代谢产物对植物的影响具有重要意义。用水、乙醇和丙酮抽提彩色豆马勃的子实体,发现这些抽提物对稗草和水稻幼苗生长有极显着的抑制作用。丙酮抽提物对狼尾草和油菜幼苗生长有抑制作用。子实体的丙酮抽提物用硅胶柱色谱分离得到2个纯化合物,可鉴定为豆马勃内酯(Pisolactone)和麦角甾醇。该2个化合物在400μg·ml-1浓度下均显着抑制稗草幼苗根生长。豆马勃内酯在100μg·ml-1浓度下仍然极显着抑制稗草幼苗根生长.  相似文献

The free radical scavenging properties and possible antioxidant activity of folic acid are reported. Pulse radiolysis technique is employed to study the one-electron oxidation of folic acid in homogeneous aqueous solution. The radicals used for this study are CCl3O2, N3, SO4, Br2, √OH, and O•−. All these radicals react with folic acid under ambient condition at an almost diffusion-controlled rate producing two types of transients. The first transient absorption maximum is around 430 nm, which decays, and a simultaneous growth at around 390 nm is observed. Considering the chemical structure of folic acid, the absorption maximum at 430 nm has been assigned to a phenoxyl radical. The latter one is proposed to be a delocalized molecular radical. A permanent product has been observed in the oxidation of folic acid with CCl3O2 and N3 radicals, with a broad absorption band around 370–400 nm. The bimolecular rate constants for all the radical-induced oxidation reactions of folic acid have been measured. Folic acid is seen to scavenge these radicals very efficiently. In the reaction of thiyl radicals with folic acid, it has been observed that folic acid can not only scavenge thiyl radicals but can also repair these thiols at physiological pH. While carrying out the lipid peroxidation study, in spite of the fact that folic acid is considerably soluble in water, we observed a significant inhibition property in microsomal lipid peroxidation. A suitable mechanism for oxidation of folic acid and repair of thiyl radicals by folic acid has been proposed.  相似文献

The copper(II) complex of 4-methyloxazolidine- 4′-carboxylic acid and the nickel(II) complex of 3N,7N-(1,3,5,7-tetraazabicyclo [3.3.1]nonyl)diacetic acid were prepared and then treated with sodium borohydride to form the sodium salts of the respective acids. The saturated heterocyclic rings of the acids are retained in the reactions.The methyl esters of the acids were subsequently prepared under anhydrous conditions and characterized by gas chromatography/mass spectrometry (GC/MS).  相似文献

A modification of the resorcinol method of Kulka1 for the determination of ketoses is described. Though being a stopped enzyme test, it is much more sensitive than the carbazol method and by applying microtiter plates and measuring with an ELISA reader, a large number of tests can be performed within a short time, thereby facilitating initial velocity studies.

The test is linear up to a concentration of 2.5 m -xylulose even in the presence of 10 m -xylose and 2 m -fructose in the presence of 10 m -glucose. The sensitivity is 25 μ for xylulose and 38 μ for fructose. The test method is insensitive to perturbations of substances frequently used in isolation procedures such as ammonium sulfate, Triton X-100, PEG 6000, sodium dodecyl sulfate, and ethanol in moderate concentrations.  相似文献

The time course of aerobic photobleaching of various chlorophyll-protein complexes in vivo at high light intensities was studied with isolated Aspidistra elatior chloroplasts.

1. 1. Ca680 bleaching starts with the onset of irradiation and, initially, proceeds linearly with time. Washing the chloroplasts causes a nearly constant increase of the bleaching rate throughout the experiment.

2. 2. Ca670 does not appreciably, if at all, bleach initially; subsequently, bleaching proceeds linearly with time and at a slightly higher rate than that for Ca680. Washing makes Ca670 bleach concomitantly with the onset of illumination, and at a nearly constant rate.

3. 3. Bleaching at 665 nm is likely to start only after a relatively long period of illumination. Washing shows no effects during this period. Once bleaching has started, washing causes its rate to increase.

4. 4. No indication of the occurrence of “short-wave” chlorophyll a forms other than Ca670 and Ca665 was obtained.

5. 5. Cb bleaching starts concomitantly with illumination at a low rate. The rate increases more or less exponentially with time. Washing enhances bleaching in two steps.

6. 6. The importance of the results is discussed.

A procedure has been developed for purification of the cytochrome oxidase from Pseudomonas aeruginosa (EC 1.9.3.2) using DEAE- and CM-cellulose chromatography, gel filtration and crystallization. The final preparation was found to be homogeneous according to ultracentrifugal and disc electrophoretic criteria. The crystalline preparation also exhibited nitrite reductase activity. The spectrum of the enzyme characterizes it as cytochrome cd. At 280 nm E1 %1 cm was 18.5 after dry weight analysis.

The molecular weight of the cytochrome oxidase was calculated to be 119000 based on a sedimentation coefficient s° 20,w = 7.36 S, diffusion coefficient D 20,w = 5.36×10−7 cm2×s−1 and partial specific volume of 0.72 ml/g. The iron content of the enzyme (0.166 %) indicates that this entity contains four iron atoms per molecule. Succinylation of the enzyme produced two probably identical subunits containing both hemes c and d, having a sedimentation coefficient s° 20,w = 4.30 S and an approximate molecular weight of 65000. In dodecylsulphate-acrylamide gel electrophoresis the cytochrome oxidase also dissociates into two subunits with molecular weight of 63000.  相似文献

The mechanism of the charge separation and stabilization of separated charges was studied using the femtosecond absorption spectroscopy. It was found that nuclear wavepacket motions on potential energy surface of the excited state of the primary electron donor P* leads to a coherent formation of the charge separated states P+BA, P+HA and P+HB (where BA, HB and HA are the primary and secondary electron acceptors, respectively) in native, pheophytin-modified and mutant reaction centers (RCs) of Rhodobacter sphaeroides R-26 and in Chloroflexus aurantiacus RCs. The processes were studied by measurements of coherent oscillations in kinetics at 890 and 935 nm (the stimulated emission bands of P*), at 800 nm (the absorption band of BA) and at 1020 nm (the absorption band of BA) as well as at 760 nm (the absorption band of HA) and at 750 nm (the absorption band of HB). It was found that wavepacket motion on the 130–150 cm−1 potential surface of P* is accompanied by approaches to the intercrossing region between P* and P+BA surfaces at 120 and 380 fs delays emitting light at 935 nm (P*) and absorbing light at 1020 nm (P+BA). In the presence of Tyr M210 (Rb. sphaeroides) or M195 (C. aurantiacus) the stabilization of P+BA is observed within a few picosseconds in contrast to YM210W. At even earlier delay (40 fs) the emission at 895 nm and bleaching at 748 nm are observed in C. aurantiacus RCs showing the wavepacket approach to the intercrossing between the P* and P+HB surfaces at that time. The 32 cm−1 rotation mode of HOH was found to modulate the electron transfer rate probably due to including of this molecule in polar chain connecting PB and BA and participating in the charge separation. The mechanism of the charge separation and stabilization of separated charges is discussed in terms of the role of nuclear motions, of polar groups connecting P and acceptors and of proton of OH group of TyrM210.  相似文献

An ion-pair liquid high-performance chromatography method with solid-phase extraction for measuring urinary concentrations of sodium cromoglycate following inhalation has been developed and validated. Sodium cromoglycate was extracted from urine on a 100-mg phenyl cartridge (Isolute, Jones Chromatography) and then quantified on a 25-cm C8 Spherisorb 5 μm stationary phase with a mobile phase of methanol-0.045 M phosphate buffer-0.05 M dodecyl triethyl ammonium phosphate (550:447.6:2.4, v/v) pH 2.3, at 0.85 ml min−1 using nedocromil sodium as an internal standard and UV detection at 238 nm. The inter- and intra-day reproducibilities were 8.33 and 13.63%, respectively, at 0.25 mg l−1. The limit of determination for sodium cromoglycate was 0.25 mg l−1 (with a signal-to-noise ration of greater than 10:1). Following oral and inhaled administration of 20 mg of sodium cromoglycate to eight healthy volunteers, the mean and S.D. of sodium cromoglycate excreted in the urine at 0.5, 1 and 24 h post-dose were 0.02, 0.05 and 0.33%, and 0.16, 0.30 and 1.55% of the dose, respectively. The urinary recovery of sodium cromoglycate at 0.5 and 1 h following inhalation can therefore be used to compare the amount of drug reaching the respiratory tract using different sodium cromoglycate inhaled products or inhalation methods.  相似文献
