This study reports the first record of Coniochaeta on Laurocerasus officinalis Roem. from the Nitra district. This is the first record of Coniochaeta for Slovakia and also for Europe. The fungus Coniochaeta prunicola Damm & Crous (Coniochaetales, Sordariomycetes, Ascomycota) was isolated from damaged leaves and twigs of host trees. Morphological analyses demonstrate that Coniochaeta prunicola and Coniochaeta velutina are distinct species.  相似文献

Coniochaeta gigantospora collected from submerged wood in a freshwater habitat in France is described and illustrated as a new species in the family Coniochaetaceae (Coniochaetales, Ascomycota). This placement is based on morphology and phylogenetic analyses of partial nuclear ribosomal 28S large subunit and complete internal transcribed spacer DNA sequence data. Coniochaeta gigantospora is distinguished from other Coniochaeta species in possessing unusually large, ellipsoid, nearly equilateral, olivaceous to olivaceous-brown ascospores.  相似文献

The yeast-like fungus Coniochaeta pulveracea was studied with regard to its novel lignocellulolytic activities and the possible effect thereof on yeasts from the woody phylloplane. An enrichment procedure was used to isolate C. pulveracea from a decaying Acacia tree, and the identity of the isolate was confirmed using morphology, as well as molecular and phylogenetic techniques. This isolate, as well as strains representing C. pulveracea from different geographical regions, were compared with regard to optimum growth temperature and enzyme activity to representatives of closely related species. These include strains of Coniochaeta boothii, Coniochaeta rhopalochaeta, and Coniochaeta subcorticalis. Plate assays for cellulase and xylanase activity indicated that all representatives of the above-mentioned species were able to produce extracellular hydrolytic enzymes and were also able to degrade birchwood toothpicks during a 50-day incubation period at 30°C. To test the ability of these fungi and their enzymes to release simple sugars from complex cellulosic substrates, filtrates obtained from liquid cultures of Coniochaeta, cultivated on carboxymethyl cellulose (CMC) as sole carbon source, were analyzed using high-performance liquid chromatography analysis. Consequently, the presence of mono- and disaccharides such as glucose and cellobiose was confirmed in these culture filtrates. Two subsequent experiments were conducted to determine whether these simple sugars released from woody material by Coniochaeta may enhance growth of phylloplane yeasts. In the first experiment, representatives of Coniochaeta were co-cultured with selected yeasts suspended in agar plates containing birchwood toothpicks, followed by examination of plates for colony formation. Results indicated that Coniochaeta growth on the toothpicks enhanced growth of nearby yeast colonies in the agar plates. In the second experiment, representatives of selected yeasts and Coniochaeta species were co-cultured on CMC and xylan-containing plates where after yeast colony formation was recorded on the plates. Saccharomyces cerevisiae strains, engineered to utilize specific wood degradation products, i.e., cellobiose or xylose, as sole carbon source were used as positive controls. While it was found that cellobiose released from CMC was assimilated by the yeasts, no evidence could be obtained that xylose released from xylan was used as carbon source by the yeasts. These ambiguous results could be ascribed to secretion of nutritious metabolic end products, other than the products of fungal xylanases.  相似文献

Two new species ofConiochaeta, isolated from Japanese soils, are described and illustrated:C. cephalothecoides, which is characterized by dark brown ascomata clothed with short setae, cylindrical asci and ovoid to almond-shaped or pyriform ascospores with a longitudinal germ slit; andC. dumosa, which is characterized by dark brown ascomata clothed with short setae and dense hyphal hairs, cylindrical asci and ellipsoid-fusoid ascospores with a longitudinal germ slit. These species are distinguished from most species of the genus by the unique cephalothecoid peridium of their ascomata. During a study on the biodiversity of yeasts and yeast-like ascomycetes from wild plants in Iran, four strains of yeast-like filamentous fungi were isolated from a healthy plant of Euphorbia polycaulis in the Qom Province, Iran (IR. of). All four strains formed small hyaline one-celled conidia from integrated conidiogenous cells directly on hyphae and sometimes on discrete phialides, as well as by microcyclic conidiation. Two strains additionally produced conidia in conidiomata that open by rupture. The internal transcribed spacer (ITS) sequences suggested the placement of these strains in the genera Collophorina (Leotiomycetes) and Coniochaeta (Sordariomycetes), respectively. Blast search results on NCBI GenBank and phylogenetic analyses of ITS, the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and the translation elongation factor 1α (EF-1α) sequences, and the nuclear large subunit ribosomal gene (LSU), partial actin (ACT), and β-tubulin (TUB) sequences, respectively, revealed the isolates to belong to three new species, that are described here as Collophorina euphorbiae, Coniochaeta iranica, and C. euphorbiae. A new species ofMicrosphaeropsis (Sphaeropsidales, Coelomycetes),M. rugospora, is described and illustrated. This fungus is characterized by its rapid growth on common media such as oatmeal and potato-carrot agars; semi-immersed to immersed, nearly globose, papillate pycnidias; pale yellowish brown, translucent, membranaceous peridium; monophialidic, ampulliform conidiogenous cells; and one-celled, dark brown, globose conidia ornamented with distinct tubercles. One new species of the subgenus Eriocera Macquart, 1838, Hexatoma (Eriocera) cleopatroides Men, sp. n. (Southern China: Anhui) is described and illustrated. A key to all of 78 known species from China in the subgenus is provided, which was solely based on literatures. The new species is similar to Hexatoma (Eriocera) cleopatra Alexander, 1933, but distinguishes from the latter by the prescutum entirely black with two ill-defined gray stripes, by the legs with fore and middle femora brown in basal half, black in apical half, with hind femora brown in basal one-fourth, and by the wings with cells c and sc more yellowish brown than the ground color.  相似文献

Endolichenic fungi within 17 lichen species in the area near Ny-Ålesund (Svalbard, High Arctic) were studied by a culture-based method. The 247 fungal isolates were obtained from 2712 lichen thallus segments. The colonization rate of endolichenic fungi ranged from 1.6 to 26.5 %, respectively. These isolates were identified to 40 fungal taxa, including 35 Ascomycota (10 orders), 4 Basidiomycota (3 orders), and 1 unidentified fungus. Thelebolales was the most abundant order, while Sordariales were the most diverse order. The common fungal taxa shared by more than 3 lichen species were Thelebolus microsporus (93 isolates), Coniochaeta hoffmannii (7 isolates), Sarocladium kiliense (33 isolates), Coniochaeta sp. 1 (5 isolates), Coniochaeta sp. 4 (28 isolates), and Coniochaeta sp. 2 (5 isolates). Low Sorenson’s similarity coefficients were observed among different lichen species, indicating that host-related factor may shape the endolichenic fungal communities in this region. In addition, no endolichenic fungal taxa were previously found in the Antarctica and Austrian Alps, suggesting endolichenic fungal communities in this region might be also shaped by the Arctic climate. The results demonstrate the existence of specific cultured endolichenic fungal species, which may be suitable objects for further study of their possible functional roles in the lichen thalli.  相似文献

Aspergillus taichungensis isolated from a soil sample collected in Taiwan is described as a new species. The new species is characterized by its restricted growth on Czapek's and malt extract agars and its white to light yellow colonies, radiate conidial heads, smooth and often diminutive conidiophores, hemispherical to elongate vesicles with biseriate aspergilla (conidiogenous cells), globose, micro-verrucose conidia and dark brown sclerotia. The species somewhat resemblesA. versicolor, A. terreus andA. flavipes, but differs in cultural and morphological details, and is considered to represent an interface species in the subgenusNidulantes.  相似文献

During study of endophytic fungal diversity in tropical region, a new species of Arthrinium was isolated from healthy petiole of Jatropha podagrica. It is characterized by aseptate, dark brown, circular, lenticular conidia with hyaline margin, broad, hyaline conidiophores with numerous brown septa and ampulliform–lageniform conidiophore mother cells. The new species superficially resembles A. phaeospermum and A. saccharicola in having circular lenticular conidia, but differs in conidia and conidiophore size, and shape of conidiophore mother cells. Phylogenetic study using sequence analysis of ITS region (ITS1–5.8S–ITS2) separates the new species (A. jatrophae) from other related species of Arthrinium.  相似文献

Warming of Arctic and alpine regions has a substantial impact on high-altitude/-latitude ecosystems. Shifting biomes due to climate change may lead to adjustments in species distributions and potential extinctions. Therefore, detailed monitoring is requisite to assess biologically meaningful shifts in community composition and species distributions. Some Arctic-alpine lichens have been shown to be particularly sensitive to climatic shifts associated with global change. However, accurate identification of lichenized fungal species remains challenging and may limit the effective use of lichens in climate change research. Given the inherent difficulties in accurate identification of lichenized fungi and the potential value of efficient identifications for bio-monitoring research, we investigated the utility of DNA barcode identification of the 13 brown Parmeliae (Ascomycota) species occurring in Greenland. For these species, we assessed monophyly and genetic distances using the nuclear ribosomal internal transcribed spacer region (ITS), the standard DNA barcode for fungi. We also compared intraspecific distance values to a proposed intra-interspecific threshold value for Parmeliaceae to identify nominal taxa potentially masking previously unrecognized diversity. Our results indicated that the 13 brown Parmeliae species occurring in Greenland can be successfully discriminated using the ITS region. All phenotypically circumscribed species were recovered as well-supported, monophyletic clades. Furthermore, our data supported a barcode gap among congeners for all brown Parmeliae species investigated here. However, high intraspecific genetic distances suggest the potential for previously unrecognized species-lineages in at least five species: Melanelia agnata, M. hepatizon, Montanelia disjuncta, M. panniformis, and M. tominii. Our research facilitates effective, long-term bio-monitoring of climate change in Greenland using lichens by providing accurate molecular identification of brown Parmeliae specimens.  相似文献

Pseudoidium javanicum is proposed as a new species based on analyses of 28S, ITS and IGS rDNA sequences, and morphological data. This new species was found on Acalypha wilkesiana var. marginata, A. argentea, and A. cristata collected from Cibodas Botanical Garden, Bogor (West Java Province, Indonesia). Our analyses showed that all these specimens have identical rDNA sequences and similar morphological characteristics. They form a distinct clade separated from other species of Erysiphaceae. Pseudoidium javanicum differs from Erysiphe acalyphae by having shorter conidiophores and foot cells 1–3 times as long as the 0–2 following cells. The conidial size of Ps. javanicum is also smaller than that of E. jatrophae.  相似文献

Homatula laxiclathra, new species, is here described from the Wei-He of the Yellow River drainage in Shaanxi Province, northern China. It is similar to H. berezowskii, H. longidorsalis, and H. variegata in the shared possession of a shallower body with a uniform depth, a character distinguishing all of them from all other congeners, but differs from these three species in the width of vertical brown bars on the caudal peduncle. This new species, along with H. berezowskii, differs from H. longidorsalis, and H. variegata in head length, caudal-peduncle depth, length of the dorsal adipose creast of the caudal peduncle, body squamation, and intestinal coiling. Homatula laxiclathra and H. berezowskii are further distinct in the caudal-fin shape and interorbital width.  相似文献

Amyloporia pinea sp. nov. is described and illustrated on the basis of collections from southern China. Morphology and phylogenetic analysis of rDNA ITS sequences support this new species. Morphologically, it is characterized by resupinate, annual basidiocarps, cream to yellowish buff pore surface when fresh, which becomes yellowish brown to clay-buff upon drying, a dimitic hyphal system with clamped generative hyphae and inamyloid skeletal hyphae, fusoid cystidioles, and cylindrical basidiospores; moreover, it causes a brown rot. Molecular phylogeny inferred from ITS sequence data suggested a close relationship between A. pinea and Amyloporia crassa sensu lato. Antrodia subxantha has amyloid skeletal hyphae, and grouped within the Amyloporia clade, hence, it is transferred to Amyloporia, and a new combination Amyloporia subxantha is proposed.  相似文献
