Sequential reduction of chromate and nitrate, two competitive electron acceptors, has been demonstrated for strains of Pseudomonas genus for both planktonic cells and cells immobilised in agar layers on the surface of synthetic membrane. Denitrification occurs practically after chromate depletion. This order of reduction process is consistent with redox potentials of the respective reactions. In a membrane bioreactor, competitive inhibition results in nitrate transfer through the membrane without transformation. Thus the receiving phase is contaminated with nitrate. To address this problem, a membrane has been used for spatial sequencing of chromate and nitrate reduction. Bacterial cells were immobilised in two layers with each layer placed on opposing sides of the membrane. By this means, chromate reduction is localised into the layer contacting the feed phase while nitrate reduction occurs in the layer facing the receiving phase. As a result, only traces of the pollutants are detected in the receiving phase.  相似文献

Microbial reduction of toxic Cr6+ to the less toxic Cr3+ is potentially a useful bioremediation process. Among the matrices tested for whole cell immobilization of an efficient chromate-reducing Streptomyces griseus strain, PVA-alginate was the most effective and was used for reduction of Cr(VI) in a bioreactor. Cr6+ reduction efficiency decreased as Cr6+ was increased from 2 to 12 mg l−1 but increased with an increase in biomass concentration. However, increasing the flow rate from 2 to 8 ml h−1 did not significantly affect Cr6+ reduction. The reduction was faster in simulated effluent than in synthetic medium and complete removal of 8 mg Cr6+ l−1 from effluent and synthetic medium occurred in 2 and 12 h, respectively. Our results indicate that immobilized S. griseus cells could be applied for the large-scale bioremediation of chromate-containing effluents and wastewaters.  相似文献

In recent decades, the production of palatinose has aroused great interest since this structural isomer of sucrose has a promising potential. Using immobilized in a hollow-fibre membrane reactor Serratia plymuthica cells, a complete conversion of concentrated sucrose solutions into palatinose was achieved. Under typical process conditions, the specific productivity of the membrane reactor was 16.8 g m−2 h−1 (flow rate 1.3 cm3 min−1 and substrate concentration 40%) in continuous mode of action. The activity of the biocatalyst (productivity of the system) decreased slowly with the increase of operation time until the 15th day and remained almost constant to the end of the experiment. The loss of activity was 11% after 90 days of continuous operation. Conversion rate of over 90% was reached for 36–48 h for all concentrations (40–60%) of the substrate solution in cycle mode of action of the bioreactor. The best productivity (18.1 g palatinose m−2 biocatalytic membrane) for the set period was observed during the recirculation of a 60% sucrose solution. The culture displayed a very good stability under the conditions of critical osmotic stress during the experiments. A microbiological analysis of the end product showed that the produced palatinose syrup measures up to the standards for products of this kind and can be used as an additive to different food products and functional foods.  相似文献

Acid blue-15, a complex and resonance-stabilized triphenylmethane (TPM) textile dye, resistant to transformation, was decolorized/degraded in an up-flow immobilized cell bioreactor. A consortium comprised of isolates belonging to Bacillus sp., Alcaligenes sp. and Aeromonas sp. formed a multispecies biofilm on refractory brick pieces used as support material. The TPM dye was degraded to simple metabolic intermediates in the bioreactor with 94% decolorization at a flow rate of 4 ml h–1.  相似文献

Pseudomonas aeruginosa (P. aeruginosa) was immobilized with polyvinyl alcohol (PVA), sodium alginate and multiwalled carbon nanotubes (MCNTs). After immobilization, the beads were subjected to freeze-thawing to enhance mechanical strength. When exposed to 80 mg/L Cr(VI), the immobilized bacteria were able to reduce 50% of them in 84 h, however the free cells were deactivated at this concentration. The beads were used to reduce 50 mg/L Cr(VI) for nine times, with the reduction efficiency above 90% in the first five times and 65% in the end.  相似文献

The impact of removable and irremovable fouling on the retention of viral and bacterial indicators by the submerged microfiltration membrane in an MBR pilot plant was evaluated. Escherichia coli, sulphite-reducing Clostridium spores, somatic coliphages and F-specific RNA bacteriophages were used as indicators. The membrane demonstrated almost complete removal of E. coli and sulphite-reducing Clostridium spores. However, there was no correlation with membrane fouling. The phage removal varied in accordance with the irremovable fouling, rising from 2.6 to 5.6 log10 units as the irremovable fouling increased (measured by the change in the transmembrane pressure). In contrast, removable fouling did not have any effect on the retention of viruses by the membrane. These results indicate that irremovable membrane fouling may affect the removal efficiency of MBRs and, therefore, their capacity to ensure the required microbiological standards for the permeate achieved.  相似文献

Selenium at high levels may cause adverse health effects on human beings and endanger aquatic lives due to its toxicity. Se(VI) reduction in continuous-flow reactors packed with Shigella fergusonii strain TB42616 immobilized by Ca2+-alginate gel beads was investigated under various hydraulic retention times (HRT) and influent Se(VI) concentrations. Removal efficiency up to 98.8 % was achieved after 96 days operation under an HRT of 5 days and an influent Se(VI) concentration of 400 mg/L. The results showed that the overall selenium removal efficiency was affected by the HRT and the bed height of the reactor but not the influent Se(VI) concentration. The steady-state data were analyzed using a mathematical model and Monod-type kinetics. Biokinetic parameters of half-velocity constants and maximum specific reduction rates were optimized using steady-state data obtained under a range of HRTs (0.73–5.0 days) at a constant influent Se(VI) concentration of 50 mg/L. The model was validated using steady-state data obtained under influent Se(VI) concentrations ranging from 10 to 400 mg/L while maintaining the HRT at 5.0 days. The high correlation coefficients between model calculated Se(VI) and Se(IV) concentrations and the experimental data indicate that the model is robust to predict the performance of the continuous-flow bioreactor.  相似文献

The application of membrane bioreactor (MBR) technology was investigated with the aim of evaluating its potential for cytostatic drug and cytotoxicity bioremoval. The toxicity removal was assessed from biomarker test. CP removal of up to 80% was achieved under the operating conditions studied (HRT of 48 h and a SRT of 50 days). The increase of TMP was associated with an increase of supernatant toxicity as if fouling led to retention of the toxicity. Peaks of supernatant cytotoxicity were correlated with peaks in supernatant humic acid contents. It may suggest that molecules with a toxic effect may be adsorbed or entrapped in humic acids substances. Our study then points out that advances in wastewater treatment using an MBR can provide a suitable process for lowering CP concentrations before discharge into the aqueous environment. However, a tertiary treatment is necessary if complete elimination of toxicity is targeted.  相似文献

The performance of a submerged membrane bioreactor (SMBR) has been investigated for abattoir wastewater (AW) treatment. The chemical oxygen demand (COD) of permeate has not exceeded 25 mg L−1 providing an average COD removal of 98%. Microbiological analysis showed that the SMBR has allowed a complete removal of fecal coliforms, Listeria and Salmonella. A significant reduction in the excess biomass production was also observed. In fact, the yield of biomass production (Yobs) ranged between 0 and 0.106 g suspended solids/g COD removed.The study of the dynamic of bacterial communities using the single strand conformation polymorphism (SSCP) method showed a significant change in the population structure and revealed a correlation between the sludge production yield and the bacterial communities.  相似文献

l-Lactic acid was produced from raw cassava starch, by simultaneous enzyme production, starch saccharification and fermentation in a circulating loop bioreactor with Aspergillus awamori and Lactococcus lactis spp. lactis immobilized in loofa sponge. A. awamori was immobilized directly in cylindrical loofa sponge while the L. lactis was immobilized in a loofa sponge alginate gel cube. In the loofa sponge alginate gel cube, the sponge serves as skeletal support for the gel with the cells. The alginate gel formed a hard outer layer covering the soft porous gel inside. By controlling the rate and frequency of broth circulation between the riser and downcomer columns, the riser could be maintained under aerobic condition while the downcomer was under anaerobic condition. Repeated fed-batch l-lactic acid production was performed for more than 400 h and the average lactic acid yield and productivity from raw cassava starch were 0.76 g lactic acid g–1 starch and 1.6 g lactic acid l–1 h–1, respectively.  相似文献

An intermittently aerated membrane bioreactor (IAMBR) system has been developed to improve the efficiency of nutrient removal, and for the stable treatment of organic matter which is contained as suspended solid (SS) in the influent. The important operating factors of an intermittently aerated bioreactor (IABR) are sludge retention times (SRTs) and carbon/nitrogen (C/N) ratios. Because research on IAMBR is young, this paper explores the effect of SRTs and C/N ratios on these systems. For SRTs of 20, 25, 30, and 40 days, there was little difference in the removal of COD, T–N, and T–P. In comparing C/N ratios of 4.5, 7, and 10, the COD concentration in permeate with a C/N ratio of 10 was most stable, although the concentration of organic matter in the influent was high. The removal efficiencies of T–N and T–P in permeate with a C/N ratio of 10 were the highest at 92.9% and 88.9%, respectively. This implies that a C/N ratio above 10 should be maintained for a nutrient removal efficiency of approximately 90%.  相似文献

(S)-3-Chloro-1-phenylpropanol is an important chiral precursor for numerous antidepressants such as tomoxetine. A high enantiomeric excess (e.e.) of (S)-3-chloro-1-phenylpropanol can be achieved by asymmetric reduction of 3-chloropropiophenone using Saccharomyces cerevisiae CGMCC 2266 cells immobilized in calcium alginate. Thermal pretreatment of the immobilized cells at 50 °C for 30 min resulted in high enantioselectivity (99% e.e.) and good percent conversion (80%). The effects of various conditions on the reduction reaction were investigated. The optimal conditions were found to be as follows: sodium alginate concentration, 2%; bead diameter, 2 mm; temperature, 30 °C; re-culture time, 24 h; and batch addition of the substrate. After reusing these three times, the immobilized cells retained approximately 60% of their original catalytic activity with their enantioselectivity intact.  相似文献

In this study, the influence of various parameters such as determination methods and step height and length of incremental flux on critical flux values were investigated. Experiments were carried out on a pilot-scale membrane bioreactor (MBR) treated municipal wastewater. Three of the five critical flux determination methods, such as flux linearity, 90% permeability and flux cycling conducted in this study, indicated a decline in critical flux values as the step height of incremental flux increased. However, the hysteresis method and the two-third (2/3) flux limitation method showed an increase and independence of critical flux to the step height of incremental flux, respectively. On the other hand, the step length of incremental flux had no obvious effect on critical flux values evaluated by all critical flux determination methods. Like critical flux, sustainable flux has negative relationship with the increase of step height but no influence of step length was found in this study.  相似文献

Growing attention has been paid to the dissemination of antibiotic resistance genes (ARGs) in wastewater microbial communities. The application of membrane bioreactors (MBRs) in wastewater treatment is becoming increasingly widespread. We hypothesized that the transfer of ARGs among bacteria could occur in MBRs, which combine a high density of bacterial cells, biofilms, and antibiotic resistance bacteria or ARGs. In this study, the transfer discipline and dissemination of the RP4 plasmid in MBRs were investigated by the counting plate method, the MIDI microorganism identification system, and quantitative polymerase chain reaction (qPCR) techniques. The results showed that the average transfer frequency of the RP4 plasmid from the donor strain to cultivable bacteria in activated sludge was 2.76 × 10−5 per recipient, which was greater than the transfer frequency in wastewater and bacterial sludge reported previously. In addition, many bacterial species in the activated sludge had received RP4 by horizontal transfer, while the genera of Shewanella spp., Photobacterium spp., Pseudomonas spp., Proteus spp., and Vibrio spp. were more likely to acquire this plasmid. Interestingly, the abundance of the RP4 plasmid in total DNA remained at high levels and relatively stable at 104 copies/mg of biosolids, suggesting that ARGs were transferred from donor strains to activated sludge bacteria in our study. Thus, the presence of ARGs in sewage sludge poses a potential health threat.  相似文献

A pilot-scale membrane bioreactor was used to treat urban wastewater using pure oxygen instead of air as a source of aeration, to study its influence on bacterial diversity and levels of enzyme activities (acid and alkaline phosphatases, glucosidase, protease, and esterase) in the sludge. The experimental work was developed in two stages influenced by seasonal temperature. Operational parameters (temperature, pH, BOD5, COD, total and volatile suspended solids) were daily monitored, and enzyme activities measured twice a week. Redundancy analysis (RDA) was used to reveal relationships between the level of enzyme activities and the variation of operational parameters, demonstrating a significant effect of temperature and volatile suspended solids. Bacterial diversity was analyzed by temperature-gradient gel electrophoresis of PCR-amplified partial 16S rRNA genes. Significant differences in community structure were observed between both stages. Sequence analysis revealed that the prevalent Bacteria populations were evolutively close to Alphaproteobacteria (44%), Betaproteobacteria (25%) and Firmicutes (17%).  相似文献

This study investigated the contribution of microfiltration to phosphorus removal in the sequencing anoxic/anaerobic membrane bioreactor. The phosphorus content in activated sludge was fractionated by the Schmidt–Thannhauser–Schneider method. The size distribution of phosphorus in the influent was analyzed to estimate the portion of particulate phosphorus rejected physically by the 0.2 μm microfiltration. The result was that along with the high removal of phosphorus (83%) the phosphorus content of activated sludge was measured as 58.66 mgP/gVSS corresponding to 5.87% on dry weight basis. About 9% of total phosphorus was chemically precipitated phosphates while 56% was stored inside the microbial cell by activity of PAOs, and 35% was the sum of minor intracellular compositions and the particulate residuals, which could be rejected completely by the microfiltration. The biological activity is the dominant way of phosphorus removal in the process. However, the microfiltration also contributed significantly to phosphorus removal by retaining the particulate phosphorus inside the system.  相似文献

In this work the performance of a Membrane bioreactor (MBR) was assessed for the removal of 3-15 mg/l of copper, lead, nickel and zinc from wastewater. The average removal efficiencies accomplished by the MBR system were 80% for Cu(II), 98% for Pb(II), 50% for Ni(II) and 77% for Zn(II). The addition of 5 g/l vermiculite into the biological reactor enhanced metal removal to 88% for copper, 85% for zinc and 60% for nickel due to adsorption of metal ions on the mineral, while it reduced biomass inhibition and increased biomass growth. The metal ions remaining in soluble form penetrated into the permeate, while those attached to sludge flocs were effectively retained by the ultrafiltration membranes. The average heterotrophic biomass inhibition was 50%, while it reduced to 29% when lower metal concentrations were fed into the reactor in the presence of vermiculite. The respective autotrophic biomass inhibition was 70% and 36%. The presence of heavy metals and vermiculite in the mixed liquor adversely impacted on membrane fouling.  相似文献

Detoxification of Cr(VI) under alkaline pH requires attention due to the alkaline nature of many effluents. An alkaliphilic gram-positive Bacillus subtilis isolated from tannery effluent contaminated soil was found to grow and reduce Cr(VI) up to 100% at an alkaline pH 9. Decrease in pH to acidic range with growth of the bacterium signified the role played by metabolites (organic acids) in chromium resistance and reduction mechanism. The XPS and FT-IR spectra confirmed the reduction of Cr(VI) by bacteria into +3 oxidation state. Chromate reductase assay indicated that the reduction was mediated by constitutive membrane bound enzymes. The kinetics of Cr(VI) reduction activity derived using the monod equation proved (Ks = 0.00032) high affinity of the organism to the metal. This study thus helped to localize the reduction activity at subcellular level in a chromium resistant alkaliphilic Bacillus sp.  相似文献

The major operational problem associated with membrane bioreactors (MBR) is membrane fouling, for which extracellular polymeric substances (EPS) are primarily responsible. In this work both the soluble and bound EPS (i.e. SMP and EPS) produced in an MBR system operating under sludge retention times (SRT) of 10, 15, 20 and 33 days were fractionized by means of membranes having variable molecular weight cutoffs (300 kDa, 100 kDa, 10 kDa & 1 kDa). The results show that increasing the SRT leads to a reduction of SMP and EPS and that these reductions are more pronounced for the SRTs in the range 10–20 days. This reduction is more significant for carbohydrates than for proteins. The decrease of SMP and EPS with increasing SRT from 10 to 20 days led to a significant decrease of the level of fouling. The further increase of SRT to 33 days did not significantly impact on the level of fouling as the SMP and EPS concentrations did not change much.  相似文献
