We have previously shown that the coral cell signal, host release factor (HRF) from the scleractinian coral Plesiastrea versipora (Lamarck) stimulates the release of glycerol from its symbiotic dinoflagellate, Symbiodinium sp. Glycerol is a precursor for algal triacylglycerol (TG) and starch, and we have previously observed that HRF reduces the amount of newly synthesized TG in Symbiodinium sp. We have now examined the effect of P. versipora HRF on starch synthesis in isolated Symbiodinium. HRF had no effect on starch synthesis after 2 h photosynthesis (16.3+/-3.0 microg starch per 10(6) algae) compared with algae in seawater (13.9+/-1.2 microg starch per 10(6) algae). However, after 4 h incubation in HRF, there was a reduction (0-76%), in the amount of newly synthesized starch which was correlated with the amount of HRF (10-76 microg/ml). Reducing algal synthesis of both TG and starch in parallel with stimulating glycerol release may provide a mechanism to regulate the population density of intracellular symbiotic algae while still ensuring the transfer of photosynthetically fixed carbon to the animal host in the form of glycerol.  相似文献

Investigation of a Philippine specimen of the red alga Ceratodictyon spongiosum and its sponge symbiont Haliclona cymaeformis led to the isolation of p-sulfooxyphenylpyruvic acid, whose structure was elucidated using spectroscopic methods, with the Z-enol geometry determined through analysis of (3)J(C,H) coupling constants. The metabolite was tested for tyrosine kinase inhibition using a (3)H-thymidine incorporation assay, but was found inactive.  相似文献

Cnidarians - corals and their relatives - dominate the shallow, illuminated sea-floor in tropical nutrient-poor seas, mainly because their food sources are both heterotrophic, by predation, and autotrophic, from their intracellular symbiotic microalgae, the zooxanthellae. This trophic flexibility is rare in the animal kingdom, and the cellular usage and biological lifetime of the organic food molecules derived from both sources have never been investigated. Here we show that autotrophically-derived carbon is stored mostly in fatty tissue and is quickly consumed by the organism, while predation-derived carbon is incorporated throughout the animal’s body and remains there for a much longer time. We hypothesize that the simple photosynthate molecules are used primarily for immediate energetic processes like respiration, whereas the more complex heterotrophic organic molecules are utilized to build cellular structures such as proteins and membranes. Our study reveals, for the first time, the different fates and roles of autotrophy and heterothrophy in zooxanthellate coelenterates.  相似文献

From a strain of the fungus Emericella variecolor derived from the marine sponge Haliclona valliculata, two new natural products, evariquinone and isoemericellin, were isolated after HPLC-UV, -MS, and -NMR studies of the extract and their structures were elucidated by mass spectrometry and NMR experiments. Evariquinone showed antiproliferative activity towards KB and NCI-H460 cells at a concentration of 3.16 microg/ml. Furthermore, the fungus was found to produce the known metabolites stromemycin, shamixanthone, and 7-hydroxyemodin. Chemical degradation, NMR decoupling experiments, and spin-system simulation provided evidence for the double bonds in stromemycin to be all E-configured. ROESY experiments established the monosaccharide moiety to be glucose.  相似文献

Summary. Preparation of homogeneous endoderm cells and culture is a prerequisite to understanding the cellular and molecular mechanism of endosymbiosis in the cnidarian-dinoflagellate association. During the cell isolation from the stony coral Euphyllia glabrescens, various amounts of symbiotic endoderm cells were found to release their symbionts (Symbiodinium spp., or zooxanthellae in generic usage) into the culture. Due to the bulky occupation by zooxanthellae inside the endoderm cell, the symbiotic endoderm cells, or zooxanthellae in hospite, are difficult to be distinguished from released zooxanthellae by microscopic examination. We now report a method for this identification using a fluorescent analogue of sphingomyelin, N-[5-(5,7-dimethyl boron dipyrromethene difluoride)-1-pentanoyl]-D-erythro-sphingosylphosphorylcholine (C5-DMB-SM). Incubation of symbiotic endoderm cells with C5-DMB-SM–defatted bovine serum albumin (DF-BSA) complex results in bright fluorescent membrane staining. Nevertheless, the membrane staining of free-living or released zooxanthellae by this complex is significantly decreased or even diminished. This method has provided a fast and reliable assay to identify symbiotic endoderm cells and will greatly accelerate the progress of endosymbiosis research. A survey of 54 species of symbiotic cnidarians that included hydrozoan corals, anemones, gorgonians and scleractinian corals was conducted in the Mexican Caribbean for the presence of mycosporine-like amino acids (MAAs) in the host as well as the Symbiodinium fractions. The host fractions contained relatively simple MAA profiles, all harbouring between one and three MAAs, principally mycosporine-glycine followed by shinorine and porphyra-334 in smaller amounts. Symbiodinium populations were identified to sub-generic levels using PCR-DGGE analysis of the Internal Transcribed Spacer 2 (ITS2) region. Regardless of clade identity, all Symbiodinium extracts contained MAAs, in contrast to the pattern that has been found in cultures of Symbiodinium, where clade A symbionts produced MAAs whereas clade B, C, D, and E symbionts did not. Under natural conditions between one and four MAAs were identified in the symbiont fractions, mycosporine-glycine (λmax = 310 nm), shinorine (λmax = 334 nm), porphyra-334 (λmax = 334 nm) and palythine (λmax = 320 nm). One sample also contained mycosporine-2-glycine (λmax = 331 nm). These data suggest that Symbiodinium is restricted to producing five MAAs and there also appears to be a defined order of appearance of these MAAs: mycosporine-glycine followed by shinorine (in one case mycosporine-2-glycine), then porphyra-334 and palythine. Overall, mycosporine-glycine was found in highest concentrations in the host and symbiont extracts. This MAA, unlike many other MAAs, absorbs within the ultraviolet-B range (UVB, 280-320 nm) and is also known for moderate antioxidant properties thus potentially providing protection against the direct and indirect effects of UVR. No depth-dependent changes could be identified due to a high variability of MAA concentrations when all species were included in the analysis. The presence of at least one MAA in all symbiont and host fractions analyzed serves to highlight the importance of MAAs, and in particular the role of mycosporine-glycine, as photoprotectants in the coral reef environment.  相似文献

The quality of an insect as a host to an entomopathogenic nematode infective juvenile depends in part on whether or not the insect is already infected and on the stage of that infection. Previous research has shown that nematode response to hosts can change after infection and that, for uninfected hosts, CO(2) can be an important cue used by infective stage juveniles during attraction. We hypothesized that CO(2) production from an insect changes after it is infected, and that these changes could influence nematode infection decisions. Changes in CO(2) released by two insect species (Galleria mellonella and Tenebrio molitor) after infection by one of four nematode species (Steinernema carpocapsae, Steinernema feltiae, Steinernema glaseri, or Steinernema riobrave) were measured. Measurements were taken every 2h from time of initial exposure to nematodes up to 224 h after infection. Dead (freeze-killed) and live uninfected insects were used as controls. Infected G. mellonella showed two distinct peaks of CO(2) production: one between 20 and 30 h and the other between 70 and 115 h after exposure to the nematodes. Peaks were up to two times higher than levels produced by uninfected insects. Infected T. molitor showed only one peak between 25 and 50h. We found differences in peak height and timing among nematode and insect species combinations. The influence of these changes in CO(2) production on IJ attraction and infection behavior remains to be determined.  相似文献

This paper reported the isolation and the phylogenetic analysis of a free-living Gymnodinium-like dinoflagellates (G. sp. (15)) from the sea water collected at Jiaozhou bay, Qingdao, Shandong Province, P.R. China. Phylogenetic reconstruction analysis with Neighbor-Joining (NJ) method using sequences of variable regions (V1+V2+V3) of the small subunit (SSU) rDNA indicated that G. sp. (15) was a Symbiodinium sp., which was closely related to a symbiont of anemones, S. californium, and the free-living strain, Gymnodinium varians. These three strains formed a new clade (which had been designated as clade F) with 100% bootstrap support. Sequence comparison showed that sequences of the internal transcribed spacer (ITS) of these three strains were highly homologous, suggesting that they might belong to one species.  相似文献

The enzyme urease has been located in the cell wall of recently isolated phycobionts from Evernia prunastri and Xanthoria parietina lichens. Cytochemical detection is achieved by producing a black, electron-dense precipitate of cobalt sulfide proceeding from CO(2) evolved from urea in the presence of cobalt chloride. Cellular fractionation reveals that about 80% of total urease activity was associated to the cell wall on both phycobionts whereas only 20% was recovered as soluble protein.  相似文献

The cell line RIRI-PX1 was established from neonate larval tissues of Papilio xuthus by performing primary cultures in the modified Grace medium that was supplemented with 20% fetal bovine serum (FBS). The cell line primarily consisted of spindle-shaped and spherical cells which attached themselves to the flask. The population-doubling times (PDTs) at the 50th and 60th passage were 42.5 h and 42.1 h respectively. The average chromosome numbers of RIRI-PX1 cell line from passage 5 to passage 50 ranged from 103 to 199. It was confirmed that RIRI-PX1 cell line was derived from P. xuthus by comparing the mitochondrial cytochrome c oxidase subunit I gene (COI) of RIRI-PX1 cells and P. xuthus eggs. This cell line was susceptible to the Autographa californica nucleopolyhedrovirus (AcNPV) and produced high yield of polyhedral occlusion bodies (43.9 OBs/cell) after 10 days of infection by AcNPV. The virus titer of AcNPV infected RIRI-PX1 cells was 3.25 × 107 TCID50/ml. We concluded that the RIRI-PX1 cell line is established from the neonate larvae tissues successfully and the cells of the cell line are sensitive to AcNPV.  相似文献

A range of biologically active secondary metabolites with pharmacological application has been reported to occur in marine sponges. The present study was undertaken to provide a set of data on the safety of a hydro-alcoholic extract (ALE) and an aqueous fraction (AQE) from Aplysina fulva Pallas, 1766 (Aplysinidae, Verongida, Porifera). Salmonella typhimurium strains TA97, TA98, TA100 and TA102, Escherichia coli strains PQ65, OG40, OG100, PQ35 and PQ37 and Balb/c 3T3 mouse fibroblasts were used to detect induction of DNA lesions by ALE and AQE. Assays used for these analyses were a bacterial (reverse) mutation assay (Ames test), the SOS-chromotest and the comet assay. Both extracts presented identical infrared 2-oxazolidone spectra. ALE treatment induced a higher frequency of type-4 comets, indicative of increasing DNA migration, in the alkaline comet assay. ALE also induced a weak genotoxic effect, as expressed by the induction factor (IF) values in the test with E. coli strain PQ35 (IF = 1.5) and by cytotoxic effects in strains PQ35, PQ65 and PQ37. Positive SOS induction (IF = 1.7) was detected in strain PQ37 treated with diluted AQE. No genotoxic effects were observed in strains PQ35, PQ65, OG40 and OG 100 after treatment with AQE dilutions. Using the bacterial (reverse) mutation test and survival assays with or without S9 mix, after 60 min of pre-incubation, we observed for strain TA97 treated with ALE a weak mutagenic response (MI = 2.2), while cytotoxic effects were seen for strains TA98, TA100 and TA102. AQE did not show mutagenic activity in any of the strains tested, but a weak cytotoxic effect was noted in strain TA102. Our data suggest that both ALE and AQE from A. fulva induce DNA breaks leading to cytotoxicity and mutagenicity under the conditions used.  相似文献

A root nodule bacterium, Sinorhizobium meliloti CCNWSX0020, resistant to 1.4 mM Cu2+ was isolated from Medicago lupulina growing in mine tailings. In medium supplied with copper, this bacterium showed cell deformation and aggregation due to precipitation of copper on the cell surface. Genes similar to the copper-resistant genes, pcoR and pcoA from Escherichia coli, were amplified by PCR from a 1.4-Mb megaplasmid. Inoculation with S. meliloti CCNWSX0020 increased the biomass of M. lupulina grown in medium added 0 and 100 mg Cu2+ kg−1 by 45.8% and 78.2%, respectively, and increased the copper concentration inside the plant tissues grown in medium supplied with 100 μM Cu2+ by 39.3%, demonstrating that it is a prospective symbiotic system for bioremediation purposes.  相似文献

A mutant strain of Klebsiella pneumoniae, termed GEM167, was obtained by γ irradiation, in which glycerol metabolism was dramatically affected on exposure to γ rays. Levels of metabolites of the glycerol reductive pathway, 1,3-propanediol (1,3-PD) and 3-hydroxypropionic acid (3-HP), were decreased in the GEM167 strain compared to a control strain, whereas the levels of metabolites derived from the oxidative pathway, 2,3-butanediol (2,3-BD), ethanol, lactate, and succinate, were increased. Notably, ethanol production from glycerol was greatly enhanced upon fermentation by the mutant strain, to a maximum production level of 21.5 g/l, with a productivity of 0.93 g/l/h. Ethanol production level was further improved to 25.0 g/l upon overexpression of Zymomonas mobilispdc and adhII genes encoding pyruvate decarboxylase (Pdc) and aldehyde dehydrogenase (Adh), respectively in the mutant strain GEM167.  相似文献

Naringenin is a flavanone compound that alters critical cellular processes such as cell multiplication, glucose uptake, and mitochondrial activity. In this study, we used the social amoeba, Dictyostelium discoideum, as a model system for examining the cellular processes and signaling pathways affected by naringenin. We found that naringenin inhibited Dictyostelium cell division in a dose-dependent manner (IC(50) approximately 20 microM). Assays of Dictyostelium chemotaxis and multicellular development revealed that naringenin possesses a previously unrecognized ability to suppress amoeboid cell motility. We also found that naringenin, which is known to inhibit phosphatidylinositol 3-kinase activity, had no apparent effect on phosphatidylinositol 3,4,5-trisphosphate synthesis in live Dictyostelium cells; suggesting that this compound suppresses cell growth and migration via alternative signaling pathways. In another context, the discoveries described here highlight the value of using the Dictyostelium model system for identifying and characterizing the mechanisms by which naringenin, and related compounds, exert their effects on eukaryotic cells.  相似文献

In yeast the P-type Ca(2+)-ATPase of the Golgi apparatus, Pmr1p, is the most important player in calcium homeostasis. In Kluyveromyces lactis KlPMR1 inactivation leads to pleiotropic phenotypes, including reduced N-glycosylation and altered cell wall morphogenesis. To study the physiology of K. lactis when KlPMR1 was inactivated microarrays containing all Saccharomyces cerevisiae coding sequences were utilized. Alterations in O-glycosylation, consistent with the repression of KlPMT2, were found and a terminal N-acetylglucosamine in the O-glycans was identified. Klpmr1Delta cells showed increased expression of PIRs, proteins involved in cell wall maintenance, suggesting that responses to cell wall weakening take place in K. lactis. We found over-expression of KlPDA1 and KlACS2 genes involved in the Acetyl-CoA synthesis and down-regulation of KlIDP1, KlACO1, and KlSDH2 genes involved in respiratory metabolism. Increases in oxygen consumption and succinate dehydrogenase activity were also observed in mutant cells. The described approach highlighted the unexpected involvement of KlPMR1 in energy-yielding processes.  相似文献

We have previously described an attenuated line of Leishmania infantum (H-line), selected by culturing promastigotes in vitro in the presence of gentamicin. To elucidate the molecular basis for this attenuation, we undertook a comparative proteomic analysis using multiplex 2-dimensional (2D) difference gel electrophoresis. Eighteen proteins that showed significant and reproducible changes in expression were identified. Many of these were components of the thiol-redox control system in Leishmania and this observation, validated by Western blot, prompted us to investigate the sensitivity of the attenuated line to oxidative stress. The attenuated line was found to be significantly more susceptible to hydrogen peroxide, a change which may explain the loss of virulence. In a direct assay of trypanothione-dependent peroxidase activity, hydrogen peroxide metabolism in the H-line was significantly lower than in wild type. Furthermore, trypanothione reductase activity was significantly lower in the H-line, suggesting that gentamicin selection may result in pleiotropic affects on thiol metabolism in Leishmania. A putative RNA-binding protein was very strongly up-regulated in the attenuated line, suggesting a possible target for gentamicin in Leishmania.  相似文献

Cytochrome P450 monooxygenases (P450s) are commonly involved in biosynthesis of endogenous compounds and catabolism of xenobiotics, and their activities rely on a partner enzyme, cytochrome P450 reductase (CPR, E.C.1.6.2.4). Two CPR cDNAs, GhCPR1 and GhCPR2, were isolated from cotton (Gossypium hirsutum). They are 71% identical to each other at the amino acid sequence level and belong to the Class I and II of dicotyledonous CPRs, respectively. The recombinant enzymes reduced cytochrome c, ferricyanide and dichlorophenolindophenol (DCPIP) in an NADPH-dependent manner, and supported the activity of CYP73A25, a cinnamate 4-hydroxylase of cotton. Both GhCPR genes were widely expressed in cotton tissues, with a reduced expression level of GhCPR2 in the glandless cotton cultivar. Expression of GhCPR2, but not GhCPR1, was inducible by mechanical wounding and elicitation, indicating that the GhCPR2 is more related to defense reactions, including biosynthesis of secondary metabolites.  相似文献

The effects of cell size of epipelic diatoms and sediment Chl a content (as an index of algal biomass) on the ingestion rate of Hydrobia ulvae adults and juveniles were investigated in experimental microcosms. Results showed that both adults and juveniles ingested small and large diatoms without exhibiting cell size selection behaviour. The functional response of H. ulvae, juveniles (<4 mm) and adults (>4 mm), over a wide range of sediment Chl a content, was characterized by an increase of the ingestion rate according to a power law. Ingestion rate varied from 0.75 to 10 ng Chl a ind−1 h−1 for juveniles and from 1 to 52 ng Chl a ind−1 h−1 for adults, in the range ca. 10-100 μg Chl a (g dry weight sediment)−1. The ingestion rate was about three times higher for adults than for juveniles. Based on these experimental results, we further proposed a mechanistic approach, using an individual based-model, to identify simple feeding mechanisms that might be involved in H. ulvae functional response.  相似文献
