Estrogenic activities of ethanol extract and its active components from Psoralea corylifolia L. were studied using various in vitro assays. The main components from ethanol extract were analyzed to be bakuchiol, psoralen, isobavachalcone, isobavachromene, and bavachinin. In a fractionation procedure, hexane and chloroform fractions showed estrogenic activity in yeast transactivation assay and E-screen assay. In yeast transactivation assay, ethanol extract, hexane, and chloroform fractions showed significantly higher activities at a concentration of 1.0 ng/ml, and bakuchiol at the concentration of 10−6 M was showed the highest activity, especially, which was higher than genistein at the same concentration. In E-screen assay, cell proliferation of bakuchiol (10−6 M) showed similar estrogenic activity with genistein (10−6 M). In ER binding assay, bakuchiol displayed the strongest ER-binding affinity (IC50 for ERα = 1.01 × 10−6 M, IC50 for ERβ = 1.20 × 10−6 M) and bakuchiol showed five times higher affinity for ERα than for ERβ.  相似文献

Bakuchiol was an active antifungal compound isolated from Psoraleae Fructus by means of bioassay-guided fractionation in our previous study. The present work aimed to investigate the underlying mechanisms and the therapeutic effect of bakuchiol in Trichophyton mentagrophytes-induced tinea pedis. After exposure to bakuchiol at 0.25-fold, 0.5-fold and 1-fold of minimum inhibitory concentration (MIC) (3.91 μg/ml) for 24 h, the fungal conidia of T. mentagrophytes demonstrated a significant dose-dependent increase in membrane permeability. Moreover, bakuchiol at 1-fold MIC elicited a 187% elevation in reactive oxygen species (ROS) level in fungal cells after a 3-h incubation. However, bakuchiol did not induce DNA fragmentation. In a guinea pig model of tinea pedis, bakuchiol at 1%, 5% or 10% (w/w) concentration in aqueous cream could significantly reduce the fungal burden of infected feet (p < 0.01–0.05). In conclusion, this is the first report to demonstrate that bakuchiol is effective in relieving tinea pedis and in inhibiting the growth of the dermatophyte T. mentagrophytes by increasing fungal membrane permeability and ROS generation, but not via induction of DNA fragmentation.  相似文献

In vitro production of the meroterpene bakuchiol by Psoralea drupacea Bge (Fabaceae) has been studied using aseptically-grown plants, callus cultures of different origin, cell suspensions and transgenic hairy root cultures. The effect of phytohormones and methyl jasmonate on bakuchiol production was also investigated. Bakuchiol was not detected in cell suspensions or hairy root preparations of P. drupacea. In contrast, aerial parts of P. drupacea grown in vitro were found to accumulate up to 11% dry weight of bakuchiol and can therefore be regarded as a potentially useful source of this antimicrobial compound.  相似文献

Chiba seed (Psoralea corylifolia L.) oil, which contains bioactive components such as bakuchiol, psoralen, and isosporalen, is recognized for its high medicinal value and is widely used in Chinese and Indian Ayurvedic medicinal systems. Several earlier studies have proved the antibacterial and antioxidant properties of the key components in the seed oil. In view of the importance of the bioactive components in the global market, this study was carried out to separate the key components by supercritical carbon dioxide extraction and further enrichment by the molecular distillation technique. The effects of the process variables such as pressure (400∼2,000 μm), temperature (140∼200°C), and wiper speed (120∼200 rpm) were studied based on response surface methodology. The predicted model was maximized for the bakuchiol content in the distillate. And a predicted maximum of 72% bakuchiol content was confirmed by the experiment under the optimized process conditions.  相似文献

An EtOH extract of fruits of Piper longum was found to exhibit a potent inhibitory effect against α-melanocyte-stimulating hormone (α-MSH)-induced melanin production in B16 mouse melanoma cells. Bioassay-directed fractionation led to the isolation of prenylated phenolic compounds bakuchiol, bavachin, and isobavachalcone. These compounds and the crude extract of the fruits of P. longum may have suppressive effects against pigmentation by melanin in the skin.  相似文献

Significant incorporations of labelled leucine, valine and isovaleric acid into the meroterpene, bakuchiol (1) isolated from the medicinal plant, Psoralea corylifolia have been observed. Degradation experiments show that labels from these substrates find their way into both phenylpropane derived as well as terpenic part of 1 thereby indicating that none of the known pathways is operative in the case of 1. It is suggested that these substrates are metabolised to CO2 which is then incorporated into 1.  相似文献

Specific incorporation of l-[U-14C]phenylalanine, [U-14C]cinnamic acid and p[2-14C]coumaric acid into bakuchiol has been observed in Psoralea corylifolia. Our findings show that the aromatic moiety along with two carbon atoms of the side chain are biosynthetically derived via phenylpropane pathway and not by the alternate pathway proposed earlier.  相似文献

为研究大叶土蜜树(Bridelia retusa)茎的化学成分及其抗神经炎活性,采用色谱技术从大叶土蜜树茎部分95%乙醇提取物中分离得到11个化合物。通过核磁共振波谱、质谱以及与文献数据比较,化合物结构鉴定为没食子酸(1)、木栓酮(2)、阿魏酸二十七烷脂(3)、芥子醛(4)、丁香醛(5)、丁香脂素(6)、补骨脂素(7)、补骨脂酚(8)、二十五烷酸(9)、亚油酸(10)和1-Linoleoylglycerol(11)。其中化合物3~11为首次从土蜜树属中分离得到。对化合物1~11的抗神经炎活性进行评价,发现化合物4、5、10和11对LPS诱导BV-2细胞NO生成具有显著抑制作用,其IC50分别为12. 57、8. 41、5. 86、5. 86μM。  相似文献

Osteoporosis is a disease of bones that leads to an increased risk of fracture. Fructus of Psoralea corylifolia L. (scurfpea fruit) is commonly utilized for treating bone fractures and joint diseases for thousands of years in China. This study was aimed to screen active principles, which might have the potency to stimulate osteoblasts proliferation and differentiation from scurfpea fruit. A HPLC method was established to analyze the main components in scurfpea fruit. Totally 11 compounds have been identified by comparing their retention time with correspondent standard substances. The MTT and ALP methods were utilized for the assay of osteoblasts proliferation and differentiation activity. Icariin, a prenylated flavonoid glycoside was treated as the positive control. Bavachin and isobavachin significantly stimulated cell proliferation, while bakuchiol exhibited stronger effect to enhance osteoblasts differentiation. All these compounds were found with a characterized structure that in each of their molecule backbones, a prenylated side chain was attached. These results lead to a hypothesis that prenyl group might be crucial to exhibit the activity. The structure–effect relationship of these compounds with prenyl group in mouse primary calvarial osteoblasts needs to be explored in further research.  相似文献

鱼类的耐低温性状是一种重要的经济性状。为了初步分析大黄鱼的耐低温性状, 文章采用15对荧光微卫星标记, 以SSR-PCR方法对大黄鱼低温耐受组和正常对照组F1代共40个个体进行了耐低温性状遗传差异分析。结果显示, 标记LYC0002在两组样品中共扩增出5个等位基因(片段大小分别为112 bp、110 bp、108 bp、106 bp和104 bp), 其中LYC0002112 bp等位基因在低温耐受组的出现频率达60%, 而在正常对照组中的频率为零, 表明该等位基因对大黄鱼的温度敏感特性有较明显的偏好性, 可能与某种耐低温基因存在一定的连锁关系。此外, 对LYC0002106 bp、LYC0002108 bp、LYC0002110 bp 和 LYC0002112 bp 4个等位基因分别进行了回收、克隆及测序。序列比对结果显示, LYC0002112 bp等位基因含有10个(CA)重复单元, 而其他3个等位基因依次缺失1个(CA)重复单元, 说明LYC0002在本研究样本中的突变方式为微卫星逐步突变模型(Stepwise mutation model, SMM)。  相似文献

Somatic chromosomes of both sexes and chromosome behaviour during spermatogenesis were studied in the aphid Schoutedenia lutea (van der Goot). Four long but unequal chromosomes in females were interpreted as X chromosomes (X1X1X2X2) with one member of an autosome pair attached to one X1, and the other member to one X2, so that the four long chromosomes were actually X1+A, X1, X2+A, X2. Males (normally XO in aphids) received X chromosomes corresponding in relative length to the two longest (X1+A, X2+A) in females. During spermatogenesis parallel pairing occurred in prophase 1 and the X1 and X2 chromosomes became associated via their autosomal segments. In anaphase I, the autosomal segment became detached from one of the X chromosomes and entered the non-viable (non-X-bearing) spermatocyte, while the viable spermatocyte received both X1 and X2 (either one of which still carried an autosome) and the haploid set of free autosomes. The consequences for sex determination and zygote formation of this unusual system are discussed; a stable chromosomal constitution for the zygote can be achieved only at the expense of considerable gamete wastage.  相似文献

The reaction between 1,3,5-tris(imidazol-1-ylmethyl)-2,4,6-trimethylbenzene (titmb) and [Ag(NH3)2][Hsal] (H2sal=salicylic acid) produces novel complex [Ag2(titmb)2][Hsal]2 · 3H2O (1). There are two unique silver centers in complex 1 with one serving as a trigonal node and the other one as a linear node. Interestingly, there are two sorts of titmb ligands in complex 1, one acting as a three-connecting ligand and the other one as a two-connecting (bridging) ligand. The silver centers are linked by titmb ligand to form 2D puckered layers containing M5(titmb)5 macrocyclic motifs, which is large enough to allow the bridging titmb ligands from the neighboring layers to be completely enclosed in. Namely, two neighboring independent layers interpenetrated each other in a parallel fashion without the occurrence of catenation to give a 3D structure.  相似文献

The face-capped octahedral cluster cis-[Re63-Se)8(PEt3)4(MeCN)2](SbF6)2, site-differentiated with protecting PEt3 and substitutionally labile MeCN ligands, reacts with bidentate ligands (L) 4,4′-bipyridyl, (E)-1,2-bis(4-pyridyl)ethene, and 1,2-bis(4-pyridyl)ethane to give the first examples of cluster-based molecular squares of the general formula cyclo-[Re63-Se)8(PEt3)4L]4(SbF6)8. The complexes were characterized by microanalysis, 1H and 31P NMR, and time-of-flight electrospray ionization mass spectrometry. Cyclic voltammetry reveals in each case one quasi-reversible oxidation and, for two compounds, one reversible and one irreversible reduction process.  相似文献

Three new magnetic compounds were synthesized by using 2-(2′-pyridyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (NIT2Py) and tris(2-benzimidazolymethyl)amine (NTB) as ligands. The structures and magnetic properties of the complexes with formula [Ni(NIT2Py)(NTB)](ClO4)2(CH3OH) 1, [Mn(NIT2Py)(NTB)](ClO4)22 and {[Zn(NIT2Py)2(CH3OH)2](ClO4)2}{[Zn(NTB)(H2O)](ClO4)2} 3 were characterized. Compounds 1 and 2 both have [M(NIT2Py)(NTB)] structural units, where the metal ion is in an octahedral environment bound to one NIT2Py through one pyridyl nitrogen atom and one nitroxide oxygen atom. However, compound 3, the chelating zinc ion has two crystallographically independent molecules in the asymmetric unit: one is six coordinated octahedral structure [Zn(NIT2Py)2(CH3OH)2](ClO4)2, and the other one is five coordinated pyramidal structure [Zn(NTB)(H2O)](ClO4)2. The magnetic behaviors of these compounds indicate that both the nickel ion and the manganese ion are antiferromagnetically coupled with the NIT2Py ligand with a coupling constant of −19.44 and −0.37 cm−1, respectively, whereas two NIT2Py ligands in compound 3 are ferromagnetically coupled with a coupling constant of 19.1 cm−1.  相似文献

The solid state conformational analysis of [Tyr4] cyclolinopeptide A has been carried out by x-ray diffraction studies. The crystal structure of the monoclinic form, grown from a dioxane-water mixture [a = 9.849 (5) Å, b = 20.752 (4) Å, c = 16.728 (5) Å, β = 98.83 (3)°, space group P21, Z = 2], shows the presence of five intramolecular N-H? O?C hydrogen bonds, with formation of one C17 ring structure, one α-turn (C13), one inverse γ-turn (C7), and two β-turns (C10, one of type III and one of type 1). The Pro1-Pro2 peptide unit is cis (ω = 5°) all others are trans. The structure is almost superimposable with that of cyclolinopeptide A.  Optical absorption, EPR and electrospray ionization mass spectrometries were used to characterize a stoichiometric reaction between mercurials and the soluble ba 3–CuA protein from Thermus thermophilus. Either one Hg(II) or two RHg(II)ions react(s) to destroy the unique spectral properties of the CuA center. EPR spectra of the resulting product indicate that one Cu from the binuclear CuA center is released into the medium as a Type 2 Cu(II) while the other remains EPR silent. Mass spectra indicate that either one Hg(II) or two RHg(II) ions remain(s) bound to the protein along with one Cu, which is assumed to be a Cu(I) ion. The latter is slowly released from the protein under aerobic conditions as additional Type 2 Cu(II), and this process is catalyzed by fungal laccase, which serves as a strong one-electron oxidant. In F-actin, [A(SH)5]n, prepared from rabbit skeletal muscle, two thiol groups react with 2,4-dinitrophenyl-glutathionyl-disulfide, DNPSSG, to form [A(SH)3(SSG)2]n. One of the two thiol groups reacts fast, (20 min), while the reaction of the second is slow (200 min). The fast reacting group has been identified as cysteine-373.In the presence of approximately one equivalent of ATP, only one of the thiol groups is reactive. The reaction product is [A(SH)4SSG]n. In comparison, the shielding effect of ADP is about 2 to 3 times smaller than that of ATP, while AMP is ineffective.The mushroom toxin phalloidin, which binds to polymeric actin, exhibits a similar protective activity as ATP and shields one thiol group from reaction with DNPSSG.We conclude from these data that in F-actin a second low affinity binding site for adenosin-nucleotides exists, which can be monitored by the reactivity of one of the two reactive thiol groups.  相似文献
