C-Glycosylation of 5,7-dihydroxy-3′,4′,5′-trimethoxyflavone was carried out with acetobromo-α-d-glucose, -α-d-galactose, -α-d-xylose, -β-l-arabinose and -αt-L-rhamnose. The respective 6-C-glycosides and 6, 8-di-C-glycosides (excepted for galactose) were isolated and permethylated. MS and TLC comparison confirmed the proposed structures of five natural tricetin-derived C-glycosides.  相似文献

Examination of a variety of arabinose containing C-glycosylflavones has shown that the sign and intensity of the CD band at 250–275 nm (charge-transfer band) reflect not only the point of attachment of the sugar to the flavone but also depend upon the absolute and anomeric configuration, ring-size and ring-conformation in addition to the preferred rotameric conformation of the sugar about the C-aryl, C-l″ bond. A change in stereochemistry of arabinose from the α to β anomer resulted in sign inversion of the 250–275 nm CD band for 6-C-l-arabinosylflavones. Furthermore, a 6-C-arabinosylflavone containing α-l-arabinose exhibited an oppositely signed charge-transfer CD band in comparison to one which contained α-d-arabinose. 6,8-Di-C-glycosylflavones containing arabinose and glucose exhibited CD bands resulting from contributions due to both sugars, if the arabinose was not present as the β-pyranose form (1C4, conformation).  相似文献

Permethylated 6-C-diglycosyl-8-C-glycosylflavones and 6-C-glycosyl-8-C-diglycosylflavones gave well defined EIMS including the molecular peak and a fragmentation pattern characteristic of the 6-C-glycosyl residue. X″′-O-glycosides (8-C-disaccharides) are thus easily distinguished from X″-O-glycosides (6-C-disaccharides) and, in the latter, the position of the O-glycosidic bond should be deduced from the MS, after acid hydrolysis. Three new C-glycosylflavones have been characterized in this way from Spergularia rubra and Stellaria holostea.  相似文献

The biosynthesis of the pyrrolidine ring of nicotine has been studied using short-term steady-state exposures of Nicotiana glutinosa seedlings to 14CO2. The pyrrolidine ring of the labeled nicotine has been degraded in a systematic manner to ascertain the radioactivity at each carbon, and a new method has been developed for obtaining C-2′ with complete radiochemical integrity. Some of the labeling patterns obtained were symmetrical while others were clearly unsymmetrical. The duality of the labeling patterns found in these 14CO2 biosyntheses, together with other data on pyrrolidine ring biosynthesis which are critically examined, is best rationalized by postulating two biosynthetic pathways for formation of the pyrrolidine ring, one involving a symmetrical precursor and the other an unsymmetrical one.  相似文献

Nine C-glycosyldeoxyanthocyanidins, 6-C-β-glucopyranosyl-7-O-methylapigeninidin, 6-C-β-glucopyranosyl-7-O-methylluteolinidin, 6-C-β-(2″-O-β-glucopyranosylglucopyranosyl)-7-O-methylapigeninidin, 6-C-β-(2″-O-β-glucopyranosylglucopyranosyl)-7,4′-di-O-methylapigeninidin, 8-C-β-glucopyranosylapigeninidin, 8-C-β-(2″-O-α-rhamnopyranosylglucopyranosyl)apigeninidin, 8-C-β-(2″-O-α-(4″′-O-acetylrhamnopyranosyl)glucopyranosyl)apigeninidin, 6,8-di-C-β-glucopyranosylapigeninidin (8), 6,8-di-C-β-glucopyranosyl-4′-O-methylluteolinidin (9), have been synthesized from their respective C-glycosylflavones (yields between 14% and 32%) by the Clemmensen reduction reaction using zinc-amalgam. The various precursors (C-glycosylflavones) of the C-glycosylanthocyanidins were isolated from either flowers of Iris sibirica L., leaves of Hawthorn ‘Crataegi Folium Cum Flore’, or lemons and oranges. This is the first time C-glycosylanthocyanidins have been synthesized. The structures of all flavonoids including the flavone rotamers were elucidated by 2D NMR techniques and high-resolution electrospray MS. The distribution of the various structural forms of 8 and 9 are different at pH 1.1, 4.5, and 7.0, however, the two pigments undergoes similar structural transformations at the various pH values. Pigments 8 and 9 with C-C linkages between the sugar moieties and the aglycone, were found to be far more stable towards acid hydrolysis than pelargonidin 3-O-glucoside, which has the typical anthocyanidin C-O linkage between the sugar and aglycone. This stability may extend the present use of anthocyanins as nutraceuticals, pharmaceuticals or colorants.  相似文献

Biogenetic pathways for leucodrin formation in Leucadendron spp. are discussed and were tested by feeding labelled d-galactose, d-glucose and l-ascorbic acid to cut stem ends of L. argenteum. It was shown that the two aldohexoses are better precursors and preferentially incorporated into that portion of leucodrin derivable from p-coumaric acid. When this route is suppressed by feeding the labelled sugars in the presence of a large amount of phenylalanine) label is predominantly incorporated in the carbohydrate part of leucodrin. The biogenesis of leucodrin therefore may proceed via coupling of l-galactono-γ-lactone and p-coumaric acid.  相似文献

The unfolding-refolding of proteins is a cooperative process and, as judged by equilibrium properties, occurs in one step involving the native,N, and the unfoldedU, conformational states. Kinetic studies have shown that the denatured protein exists as a mixture of slow-(U)Sand fast-(U)Frefolding forms produced by proline peptidecis-trans isomerization. Proline residues inU Fare in the same configuration as in the native protein while they are in non-native configuration inU S. For protein folding to occur quicklyU Smust be converted intoU F. The fact that the equilibrium and kinetic properties of are the same as those found for prolinecis-trans isomerization taken together with the absence of slow phase in the kinetics of refolding of a protein devoid of proline, support this view. However, the absence of a linear correlation between half-time of reactivation of denatured enzymes and their proline-contents, as well as the dissimilarities in the kinetic properties of in unfolding and refolding experiments are not consistent with the model. Conformational energy calculation and experimental results on refolding of proteins suggest that some proline residues are non-essential. They will not block protein folding even in wrong isomeric form. The native-like folded structure with incorrect proline isomers will serve as intermediate state(s) in which these prolines will more readily isomerize to the correct isomeric form. The picture becomes more complex when one considers the consequence ofcis-trans isomerism of non-proline residues on protein folding.  相似文献

Three 6,8-di-C-glucosylflavones: 6,8-di-C-β-d-glucopyranosylapigenin, luteolin and chrysoeriol were isolated from the whole plant of Spergularia rubra. Two new compounds, 7,2″-di-O-β-d-glucopyranosyl-6-C-α-l-arabinopyranosylapigenin and 6-C-α-l-arabinopyranosylapigenin (isomollupentin), were also characterized. Structural assignments were based on 1H NMR and MS spectra and on comparison with synthetic samples. MS fragmentation patterns of the new di-O-glucosyl compound PM derivative and of its acid hydrolysis product are given.  相似文献

Abstract

Hydrolysis studies of photo-switchable N-maleyl-amino acid-(4-phenylazophenyl) esters of Ala, Gly, Met, Phe, and Pro were performed using the proteases trypsin and chymotrypsin. It has been found out that the cis-isomers were hydrolyzed faster than the trans-isomers. In dependence of the amino acid in the P1 position the velocity graduation is Phe?>?Met?>?Ala?>?Gly?>?Pro for trypsin and Phe???Met?>?Ala?>?Gly?>?Pro for chymotrypsin for both isomers.  相似文献

An acidic compound isolated from seed of the legume Peltophorum africanum has been characterised on the basis of FAB-MS, EIMS, 13C and 1H NMR as trans-4-hydroxypipecolic acid-4-sulphate. This is the first naturally occurring sulphate ester of a non-protein amino acid to be described. The possible systematic significance of the distribution of the ester within Peltophorum and related genera is considered.  相似文献

After a 15 sec incubation in d-glucose-14C(U), 53–70% of the intracellular radioactivity in immature internodal tissue of sugarcane was in glucose-6-phosphate, and the remainder was in free glucose. Two unmetabolized glucose analogs, 2-deoxy-d-glucosce and 3-O-methyl-d-glucose, were transported at rates comparable to glucose but neither of these analogs was phosphorylated. Doubly-labeled d-glucose-1-14C-6-phosphate-32P was dephosphorylated prior to deposition in the inner space, and 14C was transported into this tissue twice as rapidly as 32P. It was also shown that 32P in exogenously supplied glucose-6-32P was not the source of phosphate for the intracellular synthesis of glucose-6-P. Galactose transport was similar to that of glucose in that the first major product recovered intracellularly was a phosphorylated sugar, i.e. 14C-galactose-1-P, when the tissue was incubated in d-galactose-14C(U). Although fructose, glucose, and galactose competed for transport into this tissue, free fructose and glucose predominated in the tissue extract after a 15-sce incubation in d-fructose-14C(U). This contrasted sharply, with the products of 14C-glucose transport which were comprised of phosphorylated sugars after 15 sec.  相似文献

A new xanthone and twelve known compounds were isolated from the aerial parts of Gentiana corymbifera. The new xanthone was shown to be 3-methylcorymbiferin (1,8-dihydroxy-3,4,5-trimethoxy-9H-xanthen-9-one) by spectral and chemical procedures. The chemotaxonomic implications are discussed.  相似文献

Six known tricin and apigenin di-C-glycosides, including 2″-O-ferulylisoschaftoside, have been identified in gametophytic material of Metzgeria conjugata. cis →trans isomerization. A study of the pH and ionic strength dependence of the isomerization reaction rate of the photochrome associated with metmyoglobin was perfomed. A comparative investigation of the reaction carried out in the presence of three proteins, metmyoglobin, apomyoglobin, and human albumin, indicates a specific influence of the heme pocket environment on the reaction. In order to clarify, in dipalmitoyl phosphatidylcholine multilayers, the effect of the 34°C thermal pretransition on the acyl chain intramolecular disordering process, Raman spectra of dipalmitoyl phosphatidylcholine gels at 20 and 34°C were compared in the 1000–1200 cm−1 skeletal C-C stretching region. In addition to an overall intensity decrease associated with a change in chain packing characteristics, the growth of intensity in the 1080–1090 and 1122 cm−1 regions in the (34-20°C) difference spectrum clearly indicates that the thermal pretransition is accompanied by an increase in the population of hydrocarbon chain gauche rotamers toward the center of the bilayer.  相似文献

河套蜜瓜果实发育过程中糖积累与蔗糖代谢相关酶的关系   总被引:3,自引:0,他引:3  
以河套蜜瓜为试材,采用外部形态观测与内部生理指标测定相结合的方法,对其果实发育过程中果实生长模式以及果实中蔗糖、果糖、葡萄糖和淀粉含量以及蔗糖代谢相关酶活性进行测定,以揭示河套蜜瓜果实生长发育过程中糖的代谢积累与相关酶的关系.结果显示:(1)河套蜜瓜果实生长速率呈单"S"曲线,果实发育早期以积累葡萄糖为主,进入成熟期后蔗糖积累量迅速增加,最终由蔗糖和己糖共同构成果实品质.(2)在河套蜜瓜果实成熟期前,蔗糖磷酸合成酶(SPS)活性维持较低水平,进入成熟期后,SPS活性迅速升高;蔗糖合成酶(SS)活性在成熟期前为分解活性大于合成活性,成熟期后表现为合成活性大于分解活性;在整个果实发育期,酸性转化酶(AI)活性较低,中性转化酶(NI)活性始终高于AI.(3)在果实整个发育期,蔗糖含量与蔗糖代谢酶的净活力呈极显著正相关,蔗糖代谢相关酶共同作用决定果实中蔗糖含量.研究表明,在河套蜜瓜果实发育前期,以蔗糖分解代谢为主,且蔗糖合成酶和中性转化酶是催化蔗糖分解的关键酶;果实成熟期间,蔗糖代谢转为合成方向为主,蔗糖合成酶和蔗糖磷酸合成酶在蔗糖积累中起主导作用.  相似文献
