The sesquiterpene lactone, 2-methyl-2-butenoic acid dodecahydro-4-(hydroxymethyl)-10a-methyl-8-methylene-3,7-dioxooxineno[5,6]cyclodeca[1,2-b]furan-9-yl ester [1aR*-[1aS*,4R*,5aS*,8aR*,9R*(E)]], argophyllone-B, was isolated from acetone extracts from the leaves of Helianthus argophyllus. Its structure has been determined by single crystal X-ray analysis. Complete 1H NMR and 13C NMR assignments have been made.  相似文献

The structure of a new ten-membered lactone, achaetolide, isolated from cultures of Achaetomium cristalliferum is deduced from its mass and NMR spectra and from the study ofsomederivatives. The 13C NMR spectra of achaetolide enriched with [1-13C], [2-13C] and [1, 2-13C] acetate established its formation from eight intact acetate units via a precursor octaketide chain.  相似文献

Oxopantoyl lactone reductase has been purified to homogeneity from a crude extract of spinach leaves (Spinacia oleracea L.) using affinity chromatography on Red-Agarose and several subsequent ion exchange steps. The enzyme is monomeric with a relative molecular mass between 33,000 to 36,000. Affinity-purified antibodies directed against the homogenous enzyme have been used to determine the amount of oxopantoyl lactone reductase in the crude leaf extract as well as the chloroplast stroma. The overall purification factor has been determined to be 22,000. The subcellular location of the enzyme is chloroplastic. The final specific activity (strictly NADPH-dependent) is 4.5 μmole . min?1 . mg?1. The enzyme is also able to reduce isatin, bornanedione and acenaphthenequinone. The enzyme activity is strongly and uncompetitively inhibited by 2-keto-4-hydroxybutyrolactone and substituted 4,5-dioxopyrrolidines. An oxopantoate reductase associated with acetohydroxy acid isomeroreductase could be detected in the plant extract. Using a specific inhibitor of this latter enzyme or oxopyrrolidines, complementation studies with branched chain amino-acids and pantothenate have shown that oxopantoyl lactone reductase is likely to be involved in pantothenate biosynthesis. Furthermore, pantoyl lactone, the putative product of the reaction, together with β-alanine and ATP, has been shown to be the substrate of pantothenate synthase using a novel assay for pantothenate.  相似文献

Nimbolide was biosynthesized from [2-14C, (4R)4-3H1]mevalonic acid lactone in the leaves of Azadirachta indica. The nimbolide had a 3H:14C ratio of 3:5 which gives support to the suggestion of the involvement of a triterpenoid intermediate with a double bond at the Δ8(9)-position in the biosynthesis of nimbolide.  相似文献

Acid hydrolysis of the saponin of the seeds of Zizyphus jujuba afforded ebelin lactone, which yielded the sapogenin, jujubogenin, on Smith-de Mayo degradation. The mechanism of conversion of jujubogenin into ebelin lactone has been elucidated. Hovenoside G of Hovenia dulcis and bacoside A of Bacopa monniera which produce ebelin lactone on acid hydrolysis have also been found to yield jujubogenin on Smith-de Mayo degradation.  相似文献

One sesquiterpene lactone – 9α-hydroxy-3-deoxyzaluzanin C, three benzopyrans: desmethoxyencecalin (6-acetyl-2,2-dimethylchromene), desacetylripariochromen B and 6-(1-hydroxyethyl)-2,2-dimethylchromene, one coumarin – scopoletin and two eugenol derivatives were isolated from the roots of Tolpis barbata (L.) Gaertn, hitherto unexamined species. In the extract from aerial parts of the plant, five known phenolic compounds, namely: esculin, esculetin, chlorogenic acid (5-CQA), luteolin 7-O-glucoside and 3,5-dicaffeoylquinic acid (3,5-DCQA) were identified as major constituents. Except for the two coumarins – scopoletin and esculetin, which were previously obtained from Tolpis webbii Sch.Bip. and T. proustii Pit., the isolated and identified compounds have not been previously reported as constituents of Tolpis spp. Though benzopyrans were found in numerous species of the Asteraceae, their occurrence in the tribe Cichorieae has not been demonstrated before.  相似文献

In addition to the four new sesquiterpene lactones previously identified, a new acylated γ-butyrolactone glucoside, taraxacoside, was isolated from the roots of Taraxacum officinale. Its structure was elucidated mainly by 1H and 13C NMR studies as β-O-[4-O-(p-hydroxyphenylacetyl]β-D-glucopyranosyl]-β-hydroxy-γ-butyrolactone. This seems to be the first instance of the detection of a monocyclic five-membered, saturated lactone O-glycoside. Additionally, p-hydroxyphenylacetic acid was identified for the first time as an acylating acid in a sugar ester.  相似文献

Crepis conyzaefolia (Gouan) Dalle Torre seed oil contains about 3% of (?)-(S,S)-12-hydroxy-13-octadec-cis-9-enolide (1), a lactone of (?)-threo-12,13-dihydroxyoleic acid. Entamoeba histolytica is the causative agent of human amoebiasis, which mainly affects developing countries. Although several drugs are effective against E. histolytica trophozoites, the control of amoebiasis requires the development of new and better alternative therapies. Medicinal plants have been the source of new molecules with remarkable antiprotozoal activity. Incomptine A isolated from Decachaeta incompta leaves, is a sesquiterpene lactone of the heliangolide type which has the major in vitro activity against E. histolytica trophozoites. However the molecular mechanisms involved in its antiprotozoal activity are still unknown. Using a proteomic approach based on two-dimensional gel electrophoresis and mass spectrometry (ESI-MS/MS) analysis, we evidenced that 21 E. histolytica proteins were differentially expressed in response to incomptine A treatment. Notably, three glycolytic enzymes, namely enolase, pyruvate:ferredoxin oxidoreductase and fructose-1,6-biphosphate aldolase, were down-regulated. Moreover, ultrastructural analysis of trophozoites through electronic microscopy showed an increased number of glycogen granules. Taken together, our data suggested that incomptine A could affect E. histolytica growth through alteration of its energy metabolism.  相似文献

Three new 2,4-linked sesquiterpene lactone dimers (SLDs), faberidilactones F-H (1–3), have been isolated from Carpesium faberi. Unlike 1 and 2, 3 was a modified Diels-Alder adduct, characteristic by a 1′-OH and a Δ5′(6′) double bond. Furthermore, the relative configuration of 1′-OH in 3 was assigned as β-configuration by comparison the experimental NOESY data with those of its two possible epimers. The exo/endo stereochemistry of 1–3 was determined by the spectrographic features of 2,4-linked SLDs that we discovered in our previous works. Compounds 1 and 2 showed potent cytotoxicity against human leukemia (CCRF-CEM) cells with IC50 value of 5.62 and 3.74 μM, respectively.  相似文献

A new steroidal lactone of the Withanolide A series has been isolated from the supercritical fluid extract of Eucalyptus globulus L. (bark) as a major component (I) along with a known structurally similar steroidal lactone as minor component (II). The structural identification of the new lactone was accomplished by different spectroscopic techniques viz. 1H and 13C NMR, etc. The relative stereochemistry was unequivocally determined from the X-ray crystallography.  相似文献

It has been shown that the phenotypic dissociation of Bacillus subtilis SK1 and Salmonella typhimurium TA100 is induced by hexylresorcinol, an exogenous non-species-specific autoregulator of pleiotropic action, which is genotoxic for both pro-and eukaryotes. Nongenotoxic homoserine lactone, a chemical analogue of cell-density-responsive species-specific regulators, does not induce bacterial dissociation. The phage resistance of the S-and R-type variants of S. typhimurium TA100 induced by hexylresorcinol has been found to be the same as that of the S-and R-type salmonella variants obtained by the routine subculturing method.  相似文献

Quorum sensing affects the regulation of more than 300 genes in Pseudomonas aeruginosa, influencing growth, biofilm formation, and the biosynthesis of several products. The quorum sensing regulation mechanisms are mostly described in a qualitative character. Particularly, in this study, the kinetics of N-butyryl-homoserine lactone (C4-HSL) and rhamnolipid formation in P. aeruginosa PAO1 were of interest. In this system, the expression of the rhamnolipid biosynthesis genes rhlAB is directly coupled to the C4-HSL concentration via the rhl system. Batch cultivations in a bioreactor with sunflower oil have been used for these investigations. 3-oxo-dodecanoyl-homoserine lactone (3o-C12-HSL) displayed a lipophilic character and accumulated in the hydrophobic phase. Degradation of C4-HSL has been found to occur in the aqueous supernatant of the culture by yet unknown extracellular mechanisms, and production was found to be proportional to biomass concentration rather than by autoinduction mechanisms. Rhamnolipid production rates, as determined experimentally, were shown to correlate linearly with the concentration of autoinducer C4-HSL. These findings were used to derive a simple model, wherein a putative, extracellular protein with C4-HSL degrading activity was assumed (putative C4-HSL acylase). The model is based on data for catalytic efficiency of HSL-acylases extracted from literature (k cat/K m), experimentally determined basal C4-HSL production rates (q C4?-?HSL basal), and two fitted parameters which describe the formation of the putative acylase and is therefore comparatively simple.  相似文献

Samples of Artemisia arbuscula ssp. arbuscula, A. tridentata ssp. tridentata, ssp. wyomingensis, ssp. vaseyana and ssp. vaseyana f. spiciformis were collected from various locations in Montana and analyzed by TLC for their sesquiterpene lactone content. Artemisia tridentata ssp. tridentata and ssp. wyomingensis are distinct morphologically and chemically, whereas ssp. vaseyana has three distinct chemical groups not yet separated morphologically. Artemisia arbuscula ssp. arbuscula and A. tridentata ssp. vaseyana f. spiciformis are easily separated by morphology but have identical TLC patterns. It has been further shown that the sesquiterpene lactones produced for a particular species or subspecies are the same regardless of the time of the year collected, although the quantity varies from winter to summer.  相似文献

Liquid whole egg or egg white, inoculated with Escherichia coli 1485, Salmonella senftenberg ATCC 8400, or Salmonella typhimurium 84-I, was treated with concentrations of β-propiolactone ranging from 0.05 to 0.3%. Egg white containing 1 × 103 to 1 × 106 cells of E. coli 1485 per ml was sterilized in 1 hr at 27 C by lactone concentrations of 0.2 and 0.3%. Egg white containing 1 × 105 cells of S. senftenberg ATCC 8400 per ml was sterilized in 12 hr at 10 C by 0.1% lactone and in 2 to 3 hr by 0.3% lactone at the same temperature.

Liquid whole egg inoculated with 1 × 105 cells of either species of Salmonella was sterilized in 4 to 5 hr at 10 C with 0.2% lactone or in 2 to 3 hr by 0.3% lactone at this temperature. A mild heat treatment of either 15 min at 37 C or 1 min at 55 C markedly shortened the exposure times required for sterilization by β-propiolactone at 10 C.

After disinfection was complete, the lactone-treated liquid whole egg was reinoculated with low cell numbers of either species of Salmonella to determine the presence of residual lactone or toxic products. Liquid whole egg treated with 0.2% lactone would support the growth of salmonellae after 13 to 14 hr at 10 C. A heat treatment of 45 min at 37 C or 10 min at 55 C immediately after addition of 0.2% lactone allowed growth of the salmonellae in the lactone-treated liquid whole egg. No evidence of residual toxicity from the lactone treatment was found.

The amount of lactone needed to prevent the outgrowth of low cell numbers of either strain of Salmonella in liquid whole egg was quantitated. Liquid whole egg containing 0.06 to 0.07% lactone would not support salmonellae growth from inocula of 1 to 10 cells per ml of egg. Lactone concentrations above 0.08% prevented outgrowth of salmonellae inocula of 10 to 200 cells per ml of liquid whole egg.
