Effects of NaCl on growth in vitro and contents of sugars, free proline and proteins in the seedlings and leaf explants of Nicotiana tabacum cv. Virginia were investigated. The fresh and dry mass of the seedlings decreased under salinity. These growth parameters in leaf explants decreased at 50 mM NaCl and increased up to 150 mM NaCl and then decreased at higher level of salinity. Free proline content in both seedlings and leaf explants increased and polysaccharide content decreased continuously with increasing of NaCl concentration. Reducing sugars, oligosaccharides, soluble sugars and total sugars contents in both seedlings and leaf explants decreased up to 150 mM NaCl and then increased at higher concentrations of NaCl.  相似文献

Effects of NaCl (150 mM), proline (10 mM) and their combination on growth and contents of chlorophyll, proline and protein of bean (Phaseolus vulgaris cv. Kizilhaç) seedlings in vitro were investigated. NaCl decreased seedling growth. Proline added to control seedlings did not change seedling growth but decreased chlorophyll and increased protein contents. When proline added to NaCl-treated seedlings growth was increased in comparison with NaCl-treated only. Thus, proline alleviated salinity stress in bean seedlings.  相似文献

The effects of NaCl on growth, contents of proteins and proline, and activities of catalase, peroxidase and polyphenol oxidase were investigated in seedlings and calli of Trigonella foenum-graecum L. and T. aphanoneura Rech. f. Seeds and hypocotyl explants were cultured on Murashige and Skoog medium supplemented with 0, 50, 100, 150 and 200 mM NaCl. Seed germination and the fresh and dry mass of the seedlings decreased significantly under salinity. In both species significant increases in protein content of seedlings over that of control were observed at 150 and 200 mM NaCl. Protein content in calli decreased at 200 mM NaCl over that of control. Protein content was higher in seedlings than in calli at all NaCl concentrations. Conversely, proline content was lower in seedlings than in calli at all the tested NaCl concentrations. NaCl caused changes in the activities of peroxidase, catalase and polyphenol oxidase in seedlings and calli.  相似文献

Calluses initiated from leaves and seedlings of the mangrove,Bruguiera sexangula, were isolated from the original tissues and subcultured. Effects of NaCl on growth and ion content of each callus were measured. The growth rate of calluses derived from leaves (leaf callus) gradually decreased as the NaCl concentration in the medium increased, while that of calluses derived from seedlings (seedling callus) was highest in the medium containing 100 mM NaCl. Concentrations of Na and Cl in both calluses increased with increasing the NaCl concentration in the culture medium. The concentration of K of leaf calluses greatly decreased at 300 mM NaCl, while the K concentration of seedling calluses decreased only slightly and remained relatively high even in the presence of 300 mM NaCl. Transient treatment of leaf calluses with media containing high concentrations of NaCl frequently induced regeneration of adventitious tissues.  相似文献

Using sand culture, we examined the responses of 6-month-old jack pine (Pinus banksiana Lamb.) seedlings to boron and salinity (sodium chloride and sodium sulfate) treatments. During 4 weeks of treatments, 60 mM NaCl and 60 mM Na2SO4 significantly decreased survival, new shoot length, number of new roots, shoot to root dry weight ratio and transpiration rates. When applied in absence of the salts, B had little effect on the measured variables. However, when applied together with salts, B decreased seedling survival, increased needle injury and altered tissue elemental concentrations in jack pine seedlings. In 2 mM B treatment, B concentration was higher in the shoots than in the roots. However, when 2 mM B was present in NaCl and Na2SO4 treatments, shoot boron concentration declined and greater proportion of B accumulated in the roots. This shift corresponded to a decline in transpiration rates. In plants treated with NaCl, Na accumulated primarily in the shoots, while in Na2SO4-treated plants Na accumulated mostly in the roots. Based on the electrolyte leakage and needle necrosis data, Cl appears to be the major factor contributing to seedling injury and B aggravates the injurious effects of NaCl. We suggest that Cl may contribute to Na and B toxicity in jack pine by altering cell membrane permeability leading to increased Na concentration in the shoots.  相似文献

In the present study, the physiological responses of Nitraria tangutorum Bobr. seedlings to NaCl stress and the regulatory function of exogenous application of salicylic acid (SA) were investigated. NaCl in low concentration (100 mM) increased while in higher concentrations (200–400 mM) decreased the individual plant dry weights (wt) of seedlings. Decreased relative water content (RWC) and chlorophyll content were observed in the leaves of seedlings subjected to salinity stress (100–400 mM NaCl). Furthermore, NaCl stress significantly increased electrolyte leakage and malondialdehyde (MDA) content. The levels of osmotic adjustment solutes including proline, soluble sugars, and soluble protein were enhanced under NaCl treatments as compared to the control. In contrast, exogenous application of SA (0.5–1.5 mM) to the roots of seedlings showed notable amelioration effects on the inhibition of individual plant dry wt, RWC, and chlorophyll content. The increases in electrolyte leakage and MDA content in the leaves of NaCl-treated seedlings were markedly inhibited by SA application. The SA application further increased the contents of proline, soluble sugars, and soluble protein. The activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) were up-regulated by NaCl stress and the activities of SOD, POD, and CAT were further enhanced by SA treatments. Application of SA in low concentration (0.5 mM) enhanced while in higher concentrations (1.0 and 1.5 mM) inhibited APX activities in leaves of NaCl-treated seedlings. These results indicate that SA effectively alleviated the adverse effects of NaCl stress on N. tangutorum.  相似文献

In order to clarify the role of endogenous growth inhibitors A-2α and A-2β in a dwarf pea plant, red light (emission peak 657 nm) treated, 9-d-old seedlings of dwarf pea (Pisum sativum L. cv. Progress No. 9) were transferred to darkness, and the resulting changes in growth rate and concentrations of A-2α and A-2β were monitored. The growth rate of the epicotyls increased, and the concentration of the inhibitors in the epicotyls decreased, according to sigmoidal time courses. The relationship between the logarithms of the concentration of the inhibitors and the corresponding growth rate was linear. These results suggest that A-2α and A-2β, may play an important role in the growth recovery process of the dwarf pea cultivar after termination of red light irradiation. Two-month-old seedlings of Bruguiera parvifora were treated with varying levels of NaCl (100, 200 and 400 mM) under hydroponic culture. Total proteins were extracted from leaves of control and NaCl treated plants after 7, 14, 30 and 45 d of treatment and analysed by SDS-PAGE. As visualized from SDS-PAGE, the intensity of several protein bands of molecular weight 17, 23, 32, 33 and 34 kDa decreased as a result of NaCl treatment. The degree of decrease of these protein bands seemed to be roughly proportional to the external NaCl concentration. The most obvious change concerned a 23 kDa-polypeptide (SSP-23), which disappeared after 45 d treatment in 400 mM NaCl. Moreover, the SSP-23 protein, which disappeared in B. parviflora under salinity stress, reappeared when these salinized seedlings were desalinized. These observations suggest the possible involvement of these polypeptides for osmotic adjustment under salt stress. NaCl stress also caused an increase in the activity of both acid and alkaline protease. The increasing activity of proteases functions as a signal of salt stress in B. parviflora, which induces the reduction of protein level.  相似文献

Increasing NaCl concentration (0, 50, 100 and 150 mM) progressively decreased growth and seed yield of clusterbean (Cyamopsis tetragonoloba Taub.) which was associated with decreased concentrations of potassium and calcium and increased concentration of sodium in the shoots. Supplemental calcium (2.5 and 5.0 mM) significantly ameliorated the adverse effects of NaCl due to enhanced Ca and K uptake and reduced Na uptake. Calcium also alleviated the negative effects of NaCl on activities of nitrogen metabolism enzymes as well as on contents of soluble protein and free amino acids. The influence of NaCl salinity on growth, dry-matter production and leaf photosynthesis of seedlings of Eucalyptus camaldulensis Dehnh. and Dalbergia sissoo Roxb. was studied by imposing 4 levels (40, 80, 120 and 160 mM) of NaCl in pot culture. Salinity up to 160 mM did not affect plant survival, but did affect plant growth and dry-matter production depending upon the species and salt concentration. NaCl reduced leaf number and dry-weight of all the plant components, but increased stem dry-weight, especially in E. camaldulensis. Salinization also stimulated total dry-matter production at all the salinity levels in E. camaldulensis but only at 40 mM in D. sissoo. The two species varied in protein and chlorophyll concentration and in leaf photosynthetic rate. Protein and chlorophyll concentration of the plants fell at all the levels of NaCl, except at 40 mM, where stimulation in the photosynthetic carbon assimilation of the plants occurred. However, no distinct relationship between leaf photosynthetic rate and dry-matter production was found. The study indicated that low salt concentrations generally stimulated growth, biomass production and rate of photosynthesis in both the species, and E. camaldulensis appeared more NaCl salt-tolerant than D. sissoo.  相似文献

The distribution of the endogenous growth inhibitors, A-2 and A-2, was determined in light-grown dwarf pea seedlings (Pisum sativum L. cv. Progress No. 9). Red-light-induced changes in this distribution in dark-grown pea seedlings was monitored. The concentration of both inhibitors in light-grown pea seedlings was maximal in the terminal bud and decreased to the lower parts of the seedlings; their concentration in internodes was 7–10 times that in leaves of the same nodes. After the onset of continuous red-light-irradiation, the concentration of the inhibitors in dark-grown pea seedlings increased rapidly in the terminal bud, epicotyl hook and third internode, and the longitudinal gradients of the inhibitors showed a close correlation with the red-light-induced growth inhibition of the stem. The present results suggest that the endogenous growth inhibitors, A-2 and A-2 may be responsible for the red-light-induced growth inhibition of the dwarf pea seedlings.  相似文献

The percentage germination of chickpea seeds (Cicer arietinum L.cv. PBG-1) gradually decreased with increasing concentration of NaCl in the growth medium and was completely inhibited with 200 mM NaCl. In the presence of 75 mM NaCl, only 51% of the seeds germinated. Gibberellic acid (GA3) and kinetin at 6 µM concentration induced the maximum increase in % germination and seedling growth under salt stress. However, IAA further inhibited both the germination and growth of stressed seedlings. The reduction in amylase activity in cotyledons of stressed seedlings was partially reversed with GA3 and kinetin whereas IAA did not show any positive effect. GA3 was more effective than kinetin in enhancing the reduced germination and seedling growth of chickpea seeds along with amylase activity in cotyledons under NaCl induced saline conditions. The reduced uptake of radiolabelled 14C sucrose by cotyledons and its reduced distribution in the shoots and roots of stressed seedlings was increased with addition of GA3 in the medium. Cotyledonary amylase was separated into amylase 1 and amylase 2 by sephadex G 150 column chromatography. The reduced activities of both amylase 1 and amylase 2 in cotyledons under salt stress was returned to near normal levels with GA3 and there was also an increase in starch utilization, resulting in its lower concentration in cotyledons of GA3-supplemented stressed cotyledons.  相似文献

The present work describes the intrachloroplast localization and the changes that took place in the thylakoid and stroma-located superoxide dismutases (SOD, EC 1.15.1.1) and ascorbate peroxidases (APX, EC 1.11.1.11), in response to long-term NaCl stress in Pisum sativum L. cv. Puget plants. Native PAGE using high chloroplast protein concentrations pointed to the presence of the two main Fe-SODs, together with CuZn-SODs, both in thylakoids and in the stroma. Western blot and immunogold labelling using the antibodies against chloroplastic Fe-SOD from Nuphar luteum also confirmed the chloroplastic localization of a Fe-SOD. Thylakoidal Fe-SOD activity was induced by a NaCl concentration as low as 70 mM, while CuZn-SOD was induced at 90 mM, although in severe stress conditions (110 mM) both activities were similar to the levels at 90 mM NaCl. NaCl stress also induced stromatic Fe-SOD and CuZn-SOD activities, although these inductions only started at higher NaCl concentration (90 mM) and were significant at 110 mM NaCl. The increase in activity of both Fe-SODs was matched by an increase in Fe-SOD protein. Chloroplastic APX isoenzymes behaved differently in thylakoids and stroma in response to NaCl. A significant increase of stromal APX occurred at 70 mM, whereas the thylakoidal APX activity was significantly and progressively lost in response to NaCl stress (70-110 mM). A significant increase in the H2O2 content of chloroplasts during stress and a reduction in the ascorbate level at 90 mM NaCl also took place, although the oxidized ascorbate pool at the highest NaCl concentration did not show significant changes. These results suggest that the loss of thylakoidal APX may be an important factor in the increase in chloroplastic H2O2, which also results from the increased thylakoid and stroma-located Fe-SOD and CuZn-SOD activities. This H2O2 may be involved in the induction of stromal APX. The up-regulation of the above enzymes in the described stress conditions would contribute to the adaptation of cv. Puget plants to moderate NaCl stress.  相似文献

The distribution of xanthoxin (Xan), was determined in light-grown, 20-d-old pea (Pisum sativum L. cv. Progress No. 9) seedlings. The cis,trans-xanthoxin (c,t-Xan) and the trans,trans-xanthoxin (t,t-Xan) were more abundant in the young leaves and terminal bud; their concentrations in leaves were 2 - 3 times those in internodes of the same nodes. After the onset of red-light-irradiation, the concentration of both Xan isomers in 7-d-old dark-grown pea seedlings increased after a 12-h lag time. The increased level of Xan was greatest in the terminal bud and decreased to lower parts of the seedlings. The ratio of c,t-Xan to t,t-Xan concentration in the seedlings was about 2:3.  相似文献

The changes in plant growth, relative water content (RWC), stomatal conductance, lipid peroxidation and antioxidant system in relation to the tolerance to salt stress were investigated in salt-tolerant Plantago maritima and salt-sensitive Plantago media. The 60 days old P. maritima and P. media seedlings were subjected to 0, 100 and 200 mM NaCl for 7 days. Reduction in shoot length was higher in P. media than in P. maritima after exposure to 200 mM NaCl, but 100 mM NaCl treatment did not show any effect on shoot length of P. maritima. Shoot dry weight decreased in P. media and did not change in P. maritima. Two hundred millimolar NaCl treatment had no effect on leaf RWC in P. maritima, but it was reduced in P. media. Salt stress caused reduction in stomatal conductance being more pronounced in P. media than in P. maritima. Activities of superoxide dismutase (SOD; EC 1.15.1.1), catalase (CAT; EC 1.11.1.6), glutathione reductase (GR; EC 1.6.4.2) decreased in P. media with increasing salinity. Ascorbate peroxidase (APX; EC 1.11.1.11) activity in leaves of P. media was increased and showed no change under 100 and 200 mM NaCl, respectively. However, activities of CAT, APX and GR increased under 200 mM NaCl while their activities did not change under 100 mM NaCl in P. maritima. SOD activity in leaves of P. maritima increased with increasing salinity. Concomitant with this, four SOD activity bands were identified in leaves of P. maritima, two bands only were observed in P. media. Peroxidase (POX; EC 1.11.1.7) activity increased under both salt concentrations in P. maritima, but only under 200 mM NaCl in P. media. Confirming this, five POX activity bands were identified in leaves of P. maritima, but only two bands were determined in P. media. Malondialdehyde levels in the leaves increased under salt stress in P. media but showed no change and decreased in P. maritima at 100 and 200 mM NaCl, respectively. These results suggest that the salt-tolerant P. maritima showed a better protection mechanism against oxidative damage caused by salt stress by its higher induced activities of antioxidant enzymes than the salt-sensitive P. media.  相似文献

Na+ has been implicated as a requirement for the inhibition of adenylate cyclase by hormones and neurotransmitters. This study examines effects of salt concentration on neuroblastoma plasma membranes that occur in the absence of an inhibitory hormone. The adenylate cyclase response to stimulatory agonists (GTP plus PGE1 (3), PGI2 or PGE2) was influenced by NaCl. As the [NaCl] increased to 150 mM, an increase in maximal activity and a decrease in apparent affinity was observed. At concentrations above 150 mM, NaCl decreased prostaglandin affinity and progressively decreased maximal activation. The GTP requirement was not altered by 30 or 150 mM NaCl in the presence of PGE1 or PGI2. The rate of Gpp(NH)p stimulated activity increased as the [NaCl] was increased in the assay. This increased rate was conserved when membranes activated in the presence of Gpp(NH)p and NaCl were reassayed in the absence of guanine nucleotide or salt. The salt evoked rate increase was proportionally greater at submaximal MgCl2 concentrations. The concentration requirement for Mg2+ was reduced by salt for adenylate cyclase in the presence of GTP or Gpp(NH)p. However, the enzyme stimulated by hormone exhibited a Mg2+ requirement that was low in the absence of salt and could not be further reduced by increased [NaCl]. Alternative monovalent cations (150 mM Li+, K+, Cs+, but not choline or tetramethylammonium) and anions (SO4=) substituted for NaCl. The observed effects were reversible upon washing the membranes and neither ouabain nor tetrodotoxin altered the response. These effects may result from a conformational alteration of a protein particularly sensitive to neutral salts in the assay.  相似文献

In the present study we tried to evaluate the effect of salicylic acid (SA) in alleviating the negative effects of salinity stress. NaCl stress (50 and 100 mM) declines the shoot and root length and maximum decrease was observed at 100 mM concentration of NaCl. Similarly shoot dry weight decreased by 57.14% and root dry weight by 67.24% with 100 mM NaCl stress. The pigments and leaf relative water content (LRWC) were also observed to decline with increase in NaCl concentration. However, supplementation of SA to NaCl stressed seedlings showed enhanced length and dry weight of shoot and root. The pigment and LRWC also increased by the application of SA in the present study. NaCl stress also enhanced proline and glycine betaine (GB) by 3.01 and 2.04 folds, respectively; further enhancement was recorded by the application of SA. Hydrogen peroxide (H2O2) and malondialdehyde (MDA) content also showed rise in accumulation, however, seedlings treated with SA and NaCl (100 mM + SA) declines the H2O2 accumulation to 1.90 from 2.45 folds and MDA to 1.69 from 2.34 folds over the control. Antioxidants were observed to increase with NaCl concentration and further increase was recorded by the application of SA. Indoleacetic acid (IAA) and indole butyric acid (IBA) decreased by 36.60 and 44.16%, respectively, and ABA increased by 750% with 100 mM NaCl. Addition of SA to NaCl stressed seedlings enhanced the IAA and IBA and decreased the ABA concentration to appreciable level. NaCl is also responsible for the higher accumulation of Na+ and Na+/K+ ratio and decreased uptake of Ca2+ and K+. Supplementation of SA decreased the Na+ accumulation and enhanced the uptake of Ca2+ and K+ in NaCl stressed seedlings. In conclusion, SA supplementation mitigates the negative effects of NaCl toxicity in faba bean seedlings through the modulation of different osmoprotectants, antioxidants and nutrients uptake.  相似文献

DNA topoisomerase is present in nuclei purified from the rootmeristems of Pisum sativum seedlings. The DNA topoisomeraseis solubilized from nuclei by 500 mol m–3 NaCl and relaxessupercoiled pBR322 DNA forming a series of DNA topoisomers whichmigrate electrophoretically between the supercoiled and opencircular forms. The presence of ATP in the incubation mixtureincreases the number of DNA topoisomers migrating electrophoreticallyin the region with slightly greater mobility than the open circularform. The formation of topoisomers with different linking numbersmight be the result of the activation of a different DNA topoisomerasewhich has a peculiar relaxing activity or introduces supercoilsinto the open circular form of pBR322 DNA. A low unknottingactivity with knotted P4 DNA is also present in the same nuclearpreparation. The hypothesis is made that both DNA topoisomerase I and IImight be present contemporaneously in these nuclei. The DNArelaxing activity seems to be stable and is activated by KC1.Partial purification by ion-exchange chromatography is not sufficientto separate these two DNA topoisomerases. Effects of NaCl (0.1 – 0.2 M) alone or in combination with 1 mM arginine on growth and endogenous polyamine (PA) content have been observed in two cultivars of rice differing in NaCl stress tolerance. The germination, seedlings fresh mass and water content decreased with increase in salinity in both the cultivars. This inhibition was partially alleviated by application of arginine. Cv. CSR-27 exhibited relatively better germination than cv. Bas-370 at different salinities. Total PA content increased in both the cultivars under NaCl stress alone and in combination with arginine. Putrescine to spermidine and spermine ratio was higher in NaCl-treated seedlings being more in cv. Bas-370 as compared to cv. CSR-27 and the ratio reversed to almost control level when arginine was applied along with NaCl.  相似文献

The effect of NaCl on antioxidant enzyme activities in potato seedlings   总被引:7,自引:0,他引:7  
The effect of NaCl on the growth and activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) were investigated in the seedlings of four potato cultivars (Agria, Kennebec; relatively salt tolerant, Diamant and Ajax; relatively salt sensitive). The shoot fresh mass of Agria and Kennebec did not changed at 50 mM NaCl, whereas in Diamant and Ajax it decreased to 50 % of that in the controls. In Agria and Kennebec, SOD activity increased at 50 mM NaCl, but no significant changes observed in Diamant and Ajax. At higher NaCl concentration, SOD activity reduced in all cultivars. CAT and POD activities increased in all cultivars under salt stress. Unlike the other cultivars, in Ajax seedlings, APX activity increased in response to NaCl stress. We also observed new POD and SOD isoenzyme activities and changes in isoenzyme compositions under salt stress. These results suggest that salt-tolerant potato cultivars may have a better protection against reactive oxygen species (ROS) by increasing the activity of antioxidant enzymes (especially SOD) under salt stress.  相似文献
