The structure of the glandular tissue located beneath the eye sulcus in Andrena variabilis Smith was studied by means of light and electron microscopy. The glandular tissue was found to be constituted of single-layered, high cylindrical epithelial cells. Electron microscopy revealed strong foldings of the cell membrane on the basal pole of the gland cells. Next to the basal lamina, nerve fibres were found with great frequency. The central, perinuclear zone of the gland cells was rich in cell organelles. The lamellae of the rough-surfaced endoplasmic reticulum (rer) were mostly observed near the nuclear membrane. The vesicles and tubules of the smooth-surface endoplasmic reticulum (ser) were found to be congregated in contiguous compact regions. A well-developed and complicated canalicular system was enclosed between the rer and ser compartments, presumably serving as a passage for the secretory products. Membrane-bound electron-dense granules (secretory granules) were also a characteristic finding in the ser. In a small proportion of the gland cells, strong vacuolization indicated the degradation processes. From the light and electron microscopic characteristics of the gland cells, the existence of pheromone-producing secretory activity of exocrine type in the cephalic gland is presumed.  相似文献

The parotid gland of Dasyuroides byrnei was examined by light microscopy, and transmission and scanning electron microscopy. The acini were composed predominantly of seromucous cells with a few mucous cells. The seromucous cells were light or dark cells containing acidophilic spherical granules of moderate to high electron density and had well-developed cytoplasmic organelles-ordinary mitochondria and large mitochondria with tubular cristae, RER with vesicular or tubular elements, and Golgi apparatus with lamellae, vesicles and vacuoles. The mucous cells had basophilic amorphous granules of low electron density, like those of ordinary mucous cells. The intercalated ducts were composed of simple cuboidal light cells having a few electron-dense granules. The striated ducts consisted of tall columnar light cells containing numerous vesicles and mitochondria with tubular cristae, the same as found in acinar seromucous cells.  相似文献

The mandibular glands of 6 male and 6 female volcano rabbits were examined by means of light and transmission electron microscopy. The acinar cells of the glands were seromucous in nature, and contained faintly basophilic granules. The cells were classified into the light cells containing granules of low or moderate densities and the clear cells having polygonal granules of low density. The preacinar cells were occasionally observed at the site between acinus and intercalated duct. These cells had many weakly basophilic granules which contained fine granular materials of moderate density. The intercalated ducts were composed of light cells containing cored granules. The striated duct cells consisted of light cells and dark cells. Both of them contained a few vacuoles and vesicles, but no secretory granules. No sex-and age-related differences were observed in the mandibular gland of the volcano rabbit. The mandibular gland of the volcano rabbit was similar to the rabbit mandibular gland rather than the pika mandibular gland morphologically.  相似文献

The axon terminals of neurosecretory cells, containing elementary granules of neurosecretory materials, have been described on the basis of light and electron microscopy. No allochthonous neurosecretory elements were found in the sinus gland. The discharge of the granules from some terminals of the sinus gland occur with the changed salinity of the environment. There is a great difference in the structure of the sinus gland when salinity is changed. The structure of the sinus gland and its modification under experimental conditions indicate a possibility of neurohormonal regulation of the hydromineral balance.  相似文献

The parotid and mandibular glands of the cotton rat were examined by light and transmission electron microscopy. Parotid gland: Acinar cells were serous in nature, and contained electron-dense granules. Intercalated duct cells contained electron-dense granules. Striated duct cells had small granules of moderate and high electron densities. Mandibular gland: Acinar cells were seromucous in nature, and contained granules of low and moderate electron densities. Intercalated duct cells contained granules of moderate and high electron densities. Striated ducts were comprised of two portions - a secretory portion and a striated portion without granules. The secretory portion had many electron-dense granules. A sexual dimorphism was obserbed in these granules, which were smaller and fewer in females than in males.  相似文献

The innervation of the salivary gland of the cockroach Nauphoeta cinerea (Olivier) has been investigated with the use of light and scanning electron microscopy. Light microscopy of methylene blue stained glands reveals the presence of a dual innervation arising from the ventral nerve cord and the stomodeal nervous system; the principal innervation is that from the ventral nerve cord which passes to the gland via the reservoir ducts. Branches of these nerves form a plexus on the acinar surface, the axons of which exhibit swelling at irregular intervals. The presence of this surface plexus and the axonal swellings was confirmed by scanning electron microscopy both in normal glands and in those in which the basal lamina had been removed by means of an HCl-collagenase digestion method. No acinar plexus was seen to be formed by branches of the stomatogastric nerve that were associated with the gland. However, other branches of this nerve were clearly connected with a complex network of multipolar neurones on the surfaces of the anterior regions of both salivary reservoirs.  相似文献

Acervuli and fragments of pineal gland obtained from 33 subjects of both sexes and age ranging from 1 to 87 years, (30 autopsy and 3 biopsy specimens) were analyzed by light microscopy, transmission and scanning electron microscopy, X-ray diffraction and X-ray energy dispersive microanalysis. It was found that primary mineralization occurs in an organic matrix formed by pinealocytes and that hydroxyapatite also takes place in mineral deposition. From our analysis, the formation of acervuli appears to be age and sex independent and can be possibly related to the secretory activity of the gland.  相似文献

A method is presented for the preparation and quantitative transplantation of enzymatically monodispersed rat mammary gland cells. When injected into the interscapular white fat pads of hormonally primed recipient rats, such cells give rise to organized mammary structures. The early stages in formation and differentiation of these mammary gland structures were studied with light microscopy, autoradiography and electron microscopy. Under appropriate hormonal stimulation, hollow ‘alveolar spheres’ of secretory mammary epithelium develop, ducts outgrow from them, and with time an arborized glandular structure which displays the cell types and morphology of a normal mammary gland is formed. The applications of this technique to quantitative study of mammary cell survival, differentiation and neoplasia are discussed.  相似文献

Fragments of snake (Bothrops jararaca) venom gland were analysed by light and transmission electron microscopy in order to characterize the changes in collagen fibres types I and III in the intertubular gland septa during the secretory cycle. The snakes were sacrificed at 45 days (unmilked group), 6 h, 4 and 8 days after manual extraction of the venom. The fragments were fixed, processed according to standard histologic technique for embedding in paraffin, and stained with haematoxylin-eosin and Gomori's trichrome and submitted to Gomori's silver impregnation technique and picrosirius-polarization method. For transmission electron microscopy the fragments were fixed and processed for embedding in Spurr's medium. At the 45th day (the gland at rest), when the secretory activity was at a minimum, the septa were narrow and filled with densely packed collagen fibrils. At 6 h, the septa were enlarged and exhibited wide spaces filled with finely granular Alcian Blue-positive material. Until the 8th day, the septa were narrower and the histologic aspect resembled that of the gland at rest. The results demonstrated structural modifications in the glandular septa according to the different periods of the secretory cycle. These modifications can be associated with the transformation in the secretory epithelium during the venom synthesis cycle.  相似文献

Summary In the secretory epithelium of the chicken mandibular gland, glycoconjugates have been studied by means of histochemical methods of light and electron microscopy. In light microscopy, a series of histochemical procedures have been employed which included lectin—peroxidase—diaminobenzidine methods and a digestion technique with neuraminidase or-amylase. In electron microscopy, a battery of methods were used that corresponded to those employed in light microscopy. In the secretory cells of the chicken mandibular gland, vicinal diol- and sulphate-containing glycoconjugates with sialic acid,-d-mannose,-d-glucose and-d-galactose residues were visualized and the possible histophysiological significances of such glycoconjugates were discussed with special reference to the functions of the salivary gland.  相似文献

Early membrane injury in lethally irradiated salivary gland cells   总被引:3,自引:0,他引:3  
The early manifestations of radiation injury in salivary glands were investigated in the rat. The animals received a single X-ray dose in the range of 200-2000 rad to their neck area. Glandular changes during the first 24 hours were studied by light and electron microscopy and by measuring serum amylase activity. The amount of cell necrosis was quantitated and expressed as necrosis index (NI), Parotid NI and serum amylase activity 24 hours following irradiation were directly proportional to the X-ray dose. The submandibular gland cells were radioresistant and so were the mucous cells of the sublingual gland. The major increase in parotid acinar cell necrosis occurred between 12 and 24 hours after irradiation. However, more than 100 per cent increase in serum amylase level was detected prior to the onset of any significant cell necrosis. As early as two hours following irradiation signs of cell membrane injury were demonstrable in the parotid by electron microscopy and consisted of intracellular oedema, sequestered degenerative cell membranes, and an accumulation of intramitochondrial particles. None of these changes was detectable in the submandibular gland. The implication of membrane injury in the lethal effects of radiation on parotid cells is discussed.  相似文献

We used light and transmission electron microscopy to examine the morphology of the accessory glands of immature and mature adult males of Apis mellifera L. We also made an electrophoretic analysis of the protein content of the mature gland. The glands of the immature male actively secrete a mucous substance that can be seen in the lumen of the gland of the mature male. This secretion stains with mercury bromophenol blue and with periodic acid-Schiff reaction, which stain glyconjugates. The protein content was higher in the lumen secretion than in the gland wall extracts. The electrophoresis patterns of the wall extracts were different from those of the secretion found in the gland lumen.  相似文献

The mandibular gland of the Djungarian hamster was examined by light microscopy, and transmission and scanning electron microscopies. Its acinar cells reacted with periodic acid-Schiff (PAS) and were weakly stained with alcian blue (AB). There were intercellular canaliculi between the acinar cells. These cells therefore appeared to be seromucous. The acinar epithelium was composed of light cells containing various spherical secretory granules. The granular cells of the mandibular gland possessed many acidophilic granules exhibiting a positive reaction to PAS stain. They were frequently observed at the junction of the acini and intercalated ducts in all mandibular glands examined. All of these cells were light and contained secretory granules of varying size and density. The intercalated ducts consisted exclusively of light cells possessing a few round granules of high density in the apical region. The striated ducts were comprised of two portions--a secretory portion and a typical striated portion without secretory granules. The secretory portion consisted of light, dark and specifically light epithelial cells containing acidophilic granules, which exhibited a strongly positive PAS reaction. The epithelium of typically striated portions was composed of light and dark cells containing fine vacuoles in the apical region. The mandibular gland of the Djungarian hamster revealed no histological differences between sexes.  相似文献

The mechanism of silk formation inApis mellifera salivary glands, during the 5th instar, was studied. Larval salivary glands were dissected and prepared for light and polarized light microscopy, as well as for scanning and transmission electron microscopy. The results showed that silk formation starts at the middle of the 5th instar and finishes at the end of the same instar. This process begins in the distal secretory portion of the gland, going towards the proximal secretory portion; and from the periphery to the center of the gland lumen. The silk proteins are released from the secretory cells as a homogeneous substance that polymerizes in the lumen to form compact birefringent tactoids. Secondly, the water absorption from the lumen secretion, carried out by secretory and duct cells, promotes aggregation of the tactoids that form a spiral-shape filament with a zigzag pattern. This pattern is also the results of the silk compression in the gland lumen and represents a high concentration of macromolecularly well-oriented silk proteins.  相似文献

The microanatomy, fine structure and cytochemistry of scolex glands of Vitta riparia, Angularella beema and Trichocephaloidis megalocephala are examined by light and electron microscopy. Cytochemical tests include PAS, mercury bromphenol blue and performic acid-alcian blue for light microscopy and periodic acid-thiocarbohydrazide-OsO4 for electron microscopy. The scolex glands are organized in two spatially isolated syncytia, one located in the rostellum, the other in the rostellar sac. The cytoplasm of the gland syncytia is characterized by a high affinity for haematoxylin. Cytochemical tests indicate the presence of moderate amounts of glycoprotein in the glands. A specialized type of secretion—dark oval or ovoid bodies with diameter 0.3–0.5 μm are observed by EM in both the rostellum glands and the rostellar-sac glands of V. riparia and T. megalocephala , and in the rostellar-sac gland of A. beema . The rostellum gland of A. beema produces larger dark oval bodies measuring up to 0.9 μm. Lipid droplets are also observed in the glandular cytoplasm. Some tegumental cytons containing discoid bodies are found between the glandular perikarya. The structure and function of the captaculum in Graptacme calamuswere studied using light microscopy, videomicrography, and transmission electron microscopy. Graptacme calamusutilizes a longitudinal ciliary band to transport small food particles along the outstretched filament. The head of the captaculum, which bears the alveolar indentation, contains a large ganglion, at least three types of gland cells, circular and longitudinal muscles, and connective tissue. It is likely that gland 1 secretes mucus which is used in the transport of food particles and that gland 2 secretes material which promotes adhesion of the alveolus to the substratum. The function of gland 3 is unknown. Critical observation of the captaculum surface revealed that the cilia of the alveolar region have truncated tips unlike those found on other regions of the captaculum. These alveolar cilia probably aid in the distribution of adhesive substance and in adhesion.  相似文献

The microinjection of polycationic but not anionic molecules causes swelling of the rough endoplasmic reticulum (RER) in salivary gland cells of a fly larva. Ca-EGTA buffers, lanthanum chloride, lysozyme, bovine serum albumin, cationic and anionic ferritin were microinjected into salivary gland cells and their effects observed by light and electron microscopy. Immediately after the microinjection of polycationic molecules, the cytoplasm changed from transparent to opaque as the RER became swollen. Binding of polycationic molecules to the RER may cause the membrane to become permeable to some solute and swell due to osmotic forces.  相似文献

Orbital gland structure of the Atlantic bottlenose dolphin, Tursiops truncatus, was examined at the macroscopic, light microscopic, and electron microscopic levels. The gland completely encircles the ocular globe in a belt-like fashion near the conjunctival fornix but is considerably more developed medially. Duct openings are scattered throughout the fornix and over the surface of the palpebral conjunctiva. Microscopically, the gland has a tubuloalveolar arrangement; alveolar cells contain numerous secretory vesicles which can be interpreted as two structural types by light and electron microscopy. Histochemical staining demonstrates that both types contain glycosaminoglycans. Lipid analysis of the glandular secretion (dolphin tears) shows them to be non-oily and to contain only negligible amounts of cholesterol, triglycerides, phospholipids, and free fatty acids. The secretion is clear, slippery, and viscoelastic and well-adapted to protecting the eye and to reducing frictional forces between the eye surface and surrounding seawater.  相似文献
