The photobiology of two reef corals and the distribution of associated symbiont types were investigated over a depth gradient of 0-60 m at Scott Reef, Western Australia. Pachyseris speciosa hosted mainly the same Symbiodinium C type similar to C3 irrespective of sampling depth. By contrast, Seriatopora hystrix hosted predominantly Symbiodinium type D1a or D1a-like at shallow depths while those in deeper water were dominated by a Symbiodinium C type closely related to C1. The photosynthesis/respiration (P/R) ratio increased consistently with depth at the two sampling times (November 2008 and April 2009) for P. speciosa and in November 2008 only for S. hystrix, suggesting a reduction in metabolic energy expended for every unit of energy obtained from photosynthesis. However, in April 2009, shallow colonies of S. hystrix exhibited decreased P/R ratios down to depths of approximately 23 m, below which the ratio increased towards the maximum depth sampled. This pattern was mirrored by changes in tissue biomass determined as total protein content. The depth of change in the direction of the P/R ratio correlated with a shift from Symbiodinium D to C-dominated colonies. We conclude that while photobiological flexibility is vital for persistence in contrasting light regimes, a shift in Symbiodinium type may also confer a functional advantage albeit at a metabolic cost with increased depth.  相似文献

We studied the effects of a hypocaloric diet (D, n = 24, age: 32.2 +/- 1.4 yr, body mass index: 34.7 +/- 0.5 kg/m2) and a hypocaloric diet associated with exercise training (D + T, n = 25, age: 32.3 +/- 1.3 yr, body mass index: 32.9 +/- 0.4 kg/m2) on muscle metaboreflex control, muscle sympathetic nerve activity (MSNA, microneurography), blood pressure, and forearm blood flow (plethysmography) levels during handgrip exercise at 10% and 30% of maximal voluntary contraction in normotensive obese women. An additional 10 women matched by age and body mass index were studied as a nonadherent group. D or D + T significantly decreased body mass index. D or D + T significantly decreased resting MSNA (bursts/100 heartbeats). The absolute levels of MSNA were significantly lower throughout 10% and 30% exercise after D or D + T, although no change was found in the magnitude of response of MSNA. D + T, but not D, significantly increased resting forearm vascular conductance. D + T significantly increased the magnitude of the response of forearm vascular conductance during 30% exercise. D or D + T significantly increased MSNA levels during posthandgrip circulatory arrest when muscle metaboreflex is isolated. In conclusion, weight loss improves muscle metaboreflex control in obese women. Weight loss reduces MSNA, which seems to be centrally mediated. Weight loss by D + T increases forearm vascular conductance at rest and during exercise in obese individuals.  相似文献

The percentages of dry weigth, ash, carbon and nitrogen in Chondrus crispus Stackh. and Furcellaria lumbricalis (Huds.) Lamour. were determined for plants distributed along a depth gradient. Analyses were made of samples collected from June throug August. Dry weights and ash were also determined for other major perennial species, but on fewer samples. In all instances the trend was an increase in ash as a percentage of the dry weight and a decrease in the percentage dry weight and organic matter, with depth. The percentage of carbon also decreased with depth, reflecting the changes in organic matter. However, the percentage of nitrogen increased with depth, thus the C/N ratio decreased with depth. These changes in gross chemical composition are assumed to be analogous to seasonal changes in composition at a given depth or level in the littoral zone.  相似文献

Diabetes, as a chronic metabolic disease, can impair the immune function of monocytes/macrophages (MMs). However, it is unclear how MM immune response to inflammation with the development of diabetes, and whether immune response around the inflammatory foci depends on the depth in tissue. Footpad provides a classical physiological site for monitoring cellular behavior during inflammation, but limited to the superficial dermis due to the strong scattering of footpad. Herein, we used confocal microscopy to monitor the motility of MMs in deeper tissue around inflammatory foci with the development of type 1 diabetic (T1D) mice through a switchable footpad skin optical clearing window. Delayed‐type hypersensitivity (DTH) model was elicited on the footpad of T1D. Results demonstrated that progressive T1D led to the gradually potentiated MM recruitment and increased expression of monocyte chemoattractant protein‐1 during DTH, but MM migration displacement, motion velocity and motility coefficient were significantly attenuated. Besides, MMs from the deeper dermis had a much larger migration displacement than those from superficial dermis at early stages of DTH but an opposite tendency at late stages for non‐T1D, while progressive T1D obscured this difference gradually. This study will be helpful for investigating the influences of progressive metabolic diseases on immune response. Whereas many cardiac symptoms of thyrotoxicosis resemble those of the hyperadrenergic state, circulating catecholamines are reduced or normal in this condition. To test the hypothesis that the thyrotoxic heart is hypersensitive to catechol-amines, we studied beta-adrenergic signaling in a transgenic (TG) mouse in which the human type 2 iodothyronine deiodinase (D2) gene is expressed in myocardium. Because D2 converts T4 to T3, the active form of thyroid hormone, the D2 TG mouse exhibits mild, chronic thyrotoxicosis that is limited to the myocardium. In the current study, we determined that cAMP accumulation in response to either norepinephrine or forskolin treatment was increased in isolated ventricular myocardiocytes and membrane-enriched fractions prepared from these D2 TG hearts as compared with wild type. This increase in adenylyl cyclase (AC) Vmax could not be explained by changes in AC isoform expression or changes in the long or short forms of stimulatory G-protein Gsalpha, which were approximately 10% decreased in D2 TG membranes. However, Western analysis and ADP-ribosylation studies suggest that the increase in AC Vmax is mediated by a decrease in the expression of inhibitory G proteins (Gialpha-3 and/or Goalpha). These data suggest that cardiac thyrotoxicosis leads to increased beta-adrenergic responsiveness of cardiomyocytes via alterations in the regulatory G-protein elements of the AC membrane complex.  相似文献

Genome size evolution is a dynamic process involving counterbalancing mechanisms whose actions vary across lineages and over time. Whereas the primary mechanism of expansion, transposable element (TE) amplification, has been widely documented, the evolutionary dynamics of genome contraction have been less thoroughly explored. To evaluate the relative impact and evolutionary stability of the mechanisms that affect genome size, we conducted a phylogenetic analysis of indel rates for 2 genomic regions in 4 Gossypium genomes: the 2 coresident genomes (A(T) and D(T)) of tetraploid cotton and its model diploid progenitors, Gossypium arboreum (A) and Gossypium raimondii (D). We determined the rates of sequence gain or loss along each branch, partitioned by mechanism, and how these changed during species divergence. In general, there has been a propensity toward growth of the diploid genomes and contraction in the polyploid. Most of the size difference between the diploid species occurred prior to polyploid divergence and was largely attributable to TE amplification in the A/A(T) genome. After separating from the true parents of the polyploid genomes, both diploid genomes experienced slower sequence gain than in the ancestor, due to fewer TE insertions in the A genome and a combination of increased deletions and decreased TE insertions in the D genome. Both genomes of the polyploid displayed increased rates of deletion and decreased rates of insertion, leading to a rate of near stasis in D(T) and overall contraction in A(T) resulting in polyploid genome contraction. As expected, TE insertions contributed significantly to the genome size differences; however, intrastrand homologous recombination, although rare, had the most significant impact on the rate of deletion. Small indel data for the diploids suggest the possibility of a bias as the smaller genomes add less or delete more sequence through small indels than do the larger genomes, whereas data for the polyploid suggest increased sequence turnover in general (both as small deletions and small insertions). Illegitimate recombination, although not demonstrated to be a dominant mechanism of genome size change, was biased in the polyploid toward deletions, which may provide a partial explanation of polyploid genomic downsizing.  相似文献

2DG causes cytotoxicity in cancer cells by disrupting thiol metabolism while Doxorubicin (DOX) induces cytotoxicity in tumor cells by generating reactive oxygen species (ROS). Here we examined the combined cytotoxic action of 2DG and DOX in rapidly dividing T47D breast cancer cells vs. slowly growing MCF-7 breast cancer cells. T47D cells exposed to the combination of 2DG/DOX significantly decreased cell survival compared to controls, while 2DG/DOX had no effect on MCF-7 cells. 2DG/DOX also disrupted the oxidant status of T47D treated cells, decreased intracellular total glutathione and increased glutathione disulfide (%GSSG) compared to MCF-7 cells. Lipid peroxidation increased in T47D cells treated with 2DG and/or DOX, but not in MCF-7 cells. T47D cells were significantly protected by NAC, indicating that the combined treatment exerts its action by increasing ROS production and disrupting antioxidant stores. When we inhibited glutathione synthesis with BSO, T47D cells became more sensitive to 2DG/DOX-induced cytotoxicity, but NAC significantly reversed this cytotoxic effect. Finally, 2DG/DOX, and BSO significantly increased the %GSSG in T47D cells, an effect which was also reversed by NAC. Our results suggest that exposure of rapidly dividing breast cancer cells to 2DG/DOX enhances cytotoxicity via oxidative stress and via disruptions to thiol metabolism.  相似文献

Health benefits of physical activity may depend on a concomitant weight loss. In a randomized, controlled trial, we compared the effects of endurance training with or without weight loss to the effect of weight loss induced by an energy‐reduced diet in 48 sedentary, moderately overweight men who completed a 12‐week intervention program of training (T), energy‐reduced diet (D), training and increased diet (T‐iD), or control (C). An energy deficit of 600 kcal/day was induced by endurance training or diet in T and D and a similar training regimen plus an increased dietary intake of 600 kcal/day defined the T‐iD group. Primary end point was insulin sensitivity as evaluated by HOMA‐IR (mainly reflecting hepatic insulin sensitivity) and hyperinsulinemic, isoglycemic clamps (primarily reflecting peripheral insulin sensitivity). Body mass decreased in T and D by 5.9 ± 0.7 and 5.3 ± 0.7 kg, respectively, whereas T‐iD and C remained weight stable. Total and abdominal fat mass were reduced in an additive manner in the T‐iD, D, and T groups by 1.9 ± 0.3/0.2 ± 0.1, 4.4 ± 0.7/0.5 ± 0.1, and 7.7 ± 0.8/0.9 ± 0.1 kg, respectively. HOMA‐IR was improved in T, D, and T‐iD, whereas insulin‐stimulated glucose clearance and suppression of plasma nonesterified fatty acids (NEFAs) were increased only in the two training groups. Thus, loss of fat mass (diet or training induced) improves hepatic insulin sensitivity, whereas peripheral insulin sensitivity in skeletal muscle and adipose tissue is increased by endurance training only. This demonstrates that endurance training per se increases various metabolic health parameters and that endurance training should preferably always be included in any intervention regimen for improving metabolic health in moderately overweight men.  相似文献

天山云杉森林土壤有机碳沿海拔的分布规律及其影响因素   总被引:4,自引:0,他引:4  
对森林土壤有机碳库含量的估测及其影响因素的研究一直是学术界关注的热点。在水热梯度上可能会存在森林土壤有机碳的分布规律,但多数在混交林内开展的工作因无法区分群落类型变化的影响而无法准确反映出森林土壤有机碳在水热梯度上的变化规律。天山云杉森林为纯林类型,在天山山脉巨大山体上呈带状分布(平均海拔下限1750 m至平均海拔上限2760m),存在水热梯度,能够排除混交林中群落类型变化对土壤有机碳的影响。因此,在天山云杉森林带按海拔梯度设置系列样地并采样,用重铬酸钾-氧化外加热法测定土壤有机碳含量并研究土壤有机碳密度沿海拔的分布规律,分析水热配比关系与植物群落(生物量)对该规律的影响。结果表明:①1 m深度的标准土壤剖面上,各海拔梯度的土壤有机碳密度随着剖面深度的增加呈减少的趋势;②各海拔梯度的有机碳主要集中在土壤表面0—40 cm范围内,所占的比例约占全剖面的60%—70%,具有明显的表聚现象;③在天山北坡中段云杉森林带的海拔下限到海拔上限标准剖面总土壤有机碳密度出现不显著的先下降后上升再下降的双峰变化,峰值出现在海拔1800—2000 m与海拔2400—2600 m,海拔2600—2800 m的有机碳密度最小;④云杉纯林在不同海拔的平均胸径呈先减少后增加再减少的趋势,与土壤总有机碳密度变化规律较吻合;⑤天山云杉森林土壤有机碳密度沿海拔的变化是水热梯度变化及受其影响的森林长势二者共同作用的结果。  相似文献

Background

Surface lightness perception is affected by scene interpretation. There is some experimental evidence that perceived lightness under bi-ocular viewing conditions is different from perceived lightness in actual scenes but there are also reports that viewing conditions have little or no effect on perceived color. We investigated how mixes of depth cues affect perception of lightness in three-dimensional rendered scenes containing strong gradients of illumination in depth.

Methodology/Principal Findings

Observers viewed a virtual room (4 m width×5 m height×17.5 m depth) with checkerboard walls and floor. In four conditions, the room was presented with or without binocular disparity (BD) depth cues and with or without motion parallax (MP) depth cues. In all conditions, observers were asked to adjust the luminance of a comparison surface to match the lightness of test surfaces placed at seven different depths (8.5–17.5 m) in the scene. We estimated lightness versus depth profiles in all four depth cue conditions. Even when observers had only pictorial depth cues (no MP, no BD), they partially but significantly discounted the illumination gradient in judging lightness. Adding either MP or BD led to significantly greater discounting and both cues together produced the greatest discounting. The effects of MP and BD were approximately additive. BD had greater influence at near distances than far.

Conclusions/Significance

These results suggest the surface lightness perception is modulated by three-dimensional perception/interpretation using pictorial, binocular-disparity, and motion-parallax cues additively. We propose a two-stage (2D and 3D) processing model for lightness perception.  相似文献

Remediation of polycyclic aromatic hydrocarbons (PAHs) contaminated alkaline saline soil with phreatophyte or "water loving plants" was investigated by spiking soil from the former lake Texcoco with 100 mg phenanthrene (Phen) kg(-1) soil, 120 mg anthracene (Ant)kg(-1) soil and 45 mg benzo(a)pyrene (BaP) kg(-1) soil and vegetating it with Athel tamarisk (Tamarix aphylla L Karst.). The growth of the Athel tamarisk was not affected by the PAHs. In soil cultivated with Athel tamarisk, the leaching of PAHs to the 32-34 cm layer decreased 2-fold compared to the uncultivated soil. The BaP concentration decreased to 39% of the initial concentration at a distance smaller than 3 cm from the roots and to 45% at a distance larger than 3cm, but 59% remained in unvegetated soil after 240 days. Dissipation of Ant and Phen decreased with depth, but not BaP. The biodegradation of PAHs was affected by their chemical properties and increased in the presence of T. aphylla, but decreased with depth.  相似文献

土壤碳氮沿海拔梯度变化及其耦合关系是山地生态系统碳氮循环研究的重要内容。为分析不同土层土壤有机碳,土壤全氮及有机碳活性组分在海拔梯度上的分布规律及相互之间的耦合关系,选取亚高山物种岷江冷杉(Abies faxoniana)原始林为研究对象,以卧龙邓生野牛沟岷江冷杉原始林2920—3700 m的样地调查数据为基础,分析不同土层土壤碳氮及活性组分沿海拔的变化规律,总结土壤有机碳稳定性沿海拔主要规律,从土壤有机碳活性组分和碳氮关系的角度揭示其对土壤有机碳沿海拔变化的影响。结果表明:1)腐殖质层土壤有机碳(SOC)随海拔升高逐渐增加,与温度显著负相关,轻组有机碳(LFOC)及颗粒态有机碳(POC)随海拔上升均表现先增加后降低的趋势,土壤全氮(TN)随海拔变化不显著,但林线处LOFC、POC和TN均显著增加;0—10 cm土壤有机碳及全氮则表现为双峰特征,峰值分别在3089 m和3260 m处,与年均温度无显著关系。2)LFOC及POC在腐殖质层和0—10 cm土层中所占比例较大,是表征土壤有机碳含量沿海拔变化规律的主要活性组分,腐殖质层LFOC/SOC和POC/SOC随海拔上升逐渐增高,0—10 cm层则逐渐降低,暗示腐殖质层有机碳稳定性沿海拔逐渐降低,0—10 cm有机碳稳定性逐渐升高。3)SOC与TN显著正相关,SOC是影响TN的主要因子,但腐殖质层TN与有机碳活性组分无显著相关关系。4)土壤C/N和微生物量C/N在3177 m大于25:1,是引起土壤有机碳含量显著降低的主要因素。  相似文献

At a given level of serum cholesterol, patients with T2D have an increased risk of developing atherosclerosis compared with nondiabetic subjects. We hypothesized that T2D patients have an increased interstitial fluid (IF)-to-serum gradient ratio for LDL, due to leakage over the vascular wall. Therefore, lipoprotein profiles in serum and IF from 35 T2D patients and 35 healthy controls were assayed using fast performance liquid chromatography. The IF-to-serum gradients for VLDL and LDL cholesterol, as well as for apoB, were clearly reduced in T2D patients compared with healthy controls. No such differences were observed for HDL cholesterol. Contrary to our hypothesis, the atherogenic VLDL and LDL particles were not increased in IF from diabetic patients. Instead, they were relatively sparser than in healthy controls. The most probable explanation to our unexpected finding is that these lipoproteins are more susceptible to retainment in the extravascular space of these patients, reflecting a more active uptake by, or adhesion to, tissue cells, including macrophages in the vascular wall. Further studies are warranted to further characterize the mechanisms underlying these observations, which may be highly relevant for the understanding of why the propensity to develop atherosclerosis is increased in T2D.  相似文献

Hepatic iodothyronine deiodinases (Ds) are involved in the conversion of thyroid hormones (THs) which interacts with growth hormone (GH) to regulate posthatch growth in the chicken. Previous studies suggest that leptin-like immunoreactive substance deposited in the egg may serve as a maternal signal to program posthatch growth. To test the hypothesis that maternal leptin may affect early posthatch growth through modifying hepatic activation of THs, we injected 5.0μg of recombinant murine leptin into the albumen of breeder eggs before incubation. Furthermore, chicken embryo hepatocytes (CEHs) were treated with leptin in vitro to reveal the direct effect of leptin on expression and activity of Ds. In ovo leptin administration markedly accelerated early posthatch growth, elevated serum levels of total and free triiodothyronine (tT3 and fT3), while that of total thyroxin (tT4) remained unchanged. Hepatic mRNA expression and activity of D1 which converts T4 to T3 or rT3 to T2, were significantly increased in leptin-treated chickens, while those of D3 which converts T3 to T2 or T4 to rT3, were significantly decreased. Moreover, hepatic expression of GHR and IGF-I mRNA was all up-regulated in leptin-treated chickens. Males demonstrated more pronounced responses. A direct effect of leptin on Ds was shown in CEHs cultured in vitro. Expression and activity of D1 were increased, whereas those of D3 were decreased, in leptin-treated cells. These data suggest that in ovo leptin administration improves early posthatch growth, in a gender-specific fashion, probably through improving hepatic activation of THs and up-regulating hepatic expression of GHR and IGF-I.  相似文献

Our aim was to study the potential mechanisms responsible for the improvement in glucose control in Type 2 diabetes (T2D) within days after Roux-en-Y gastric bypass (RYGB). Thirteen obese subjects with T2D and twelve matched subjects with normal glucose tolerance (NGT) were examined during a liquid meal before (Pre), 1 wk, 3 mo, and 1 yr after RYGB. Glucose, insulin, C-peptide, glucagon-like peptide-1 (GLP-1), glucose-dependent-insulinotropic polypeptide (GIP), and glucagon concentrations were measured. Insulin resistance (HOMA-IR), β-cell glucose sensitivity (β-GS), and disposition index (D(β-GS): β-GS × 1/HOMA-IR) were calculated. Within the first week after RYGB, fasting glucose [T2D Pre: 8.8 ± 2.3, 1 wk: 7.0 ± 1.2 (P < 0.001)], and insulin concentrations decreased significantly in both groups. At 129 min, glucose concentrations decreased in T2D [Pre: 11.4 ± 3, 1 wk: 8.2 ± 2 (P = 0.003)] but not in NGT. HOMA-IR decreased by 50% in both groups. β-GS increased in T2D [Pre: 1.03 ± 0.49, 1 wk: 1.70 ± 1.2, (P = 0.012)] but did not change in NGT. The increase in DI(β-GS) was 3-fold in T2D and 1.5-fold in NGT. After RYGB, glucagon secretion was increased in response to the meal. GIP secretion was unchanged, while GLP-1 secretion increased more than 10-fold in both groups. The changes induced by RYGB were sustained or further enhanced 3 mo and 1 yr after surgery. Improvement in glycemic control in T2D after RYGB occurs within days after surgery and is associated with increased insulin sensitivity and improved β-cell function, the latter of which may be explained by dramatic increases in GLP-1 secretion.  相似文献

In order to test the hypothesis that fish distribution is related to water depth, three exploited reservoirs were sampled at day and at night using a Simrad EK60 echosounder. Acoustic surveys were carried out between February and June 2008, which focused on the fish size, species density (fish per ha) and biomass (kg ha?1) along vertical and longitudinal gradients. It was evident in the surface layer (0–3 m) during daytime that the larger fish sizes occupy waters near the dam or the middle of Lakhmess and Sejnane reservoirs. In the upper layer at nighttime, a gradient of fish size is shown proportional to the depth at Lakhmess Reservoir and inversely proportional at the man‐made Sejnane Lake. At Lakhmess and Laabid reservoirs, fish density was significantly higher in deep water (3926 ± 1409 and 624 ± 258 fish per ha) rather than in the upper layer (988 ± 2 and 8 ± 2 fish per ha in daytime, respectively), while at Sejnane Reservoir the number of fish per area was similar along the vertical gradient. The biomass was significantly higher in waters deeper than 3 m at Sejnane Reservoir whereas at Lakhmess it was higher in the surface layer. Fish biomass increased from upstream (0.16 ± 0.05 kg ha?1) to middle (3.07 ± 2.96 kg ha?1) and downstream (3.82 ± 2.30 kg ha?1 at night) areas in the man‐made Laabid Lake while a similar longitudinal gradient occurred in the deepest Sejnane Reservoir. The vertical gradient in fish distribution is discussed. From the dam to the tributary of the entire water column, it was concluded that fish biomass distribution was governed by depth and was most abundant in areas with deep waters.  相似文献

Microbial mercury methylation and methylmercury decomposition were examined in Lake Clara, an oligotrophic northern Wisconsin seepage lake, using radioisotopic tracers. Methylation activity was near background in the water column, was greatest in the profundal surficial sediments, and decreased with depth in sediment cores. Active demethylation occurred in the water column but was variable. Demethylation was greatest in the surficial sediments and decreased slightly with sediment depth. The methylation/demethylation ratio (M/D) was >1 in the water column, exhibited a sharp peak in surface sediments, and decreased in deeper sediments. Methylation and demethylation activity varied in surface sediments collected along a lake transect. The M/D ratio in surface sediments ranged from 1.4 to 5.8. Methylation in attached microbial communities was near background, while demethylation was high. The M/D ratios in the attached communities were all <0.20. Methylation activity in surface sediments incubated at in situ temperature increased from spring to late summer and decreased in the fall. Demethylation increased from early to midsummer and then declined. The M/D ratio in surface sediments increased from mid- to late summer, and decreased in the fall. These results indicate that the greatest potential for methylation in Lake Clara occurs in the surficial sediments and that methylation in surficial sediments is greatest from mid-July through September. In addition, the net rate of methylmercury production may be significantly affected by demethylation.  相似文献

为探究植物对环境变化的适应策略,在安徽省金寨县天马国家自然保护区,以不同海拔高度(750、850、1000、1150 m)杉木(Cunninghamia lanceolata)成熟林为对象,采用土钻法获取土壤细根样品,分别测定了不同海拔不同土层(0—10 cm、10—20 cm、20—30 cm)土壤细根生物量、形态特征参数和碳氮含量。结果表明:(1)随海拔梯度增加,0—30 cm土层细根生物量、根长密度、比根长、表面积密度、体积密度均呈先减少后增加趋势,在海拔750 m生物量最大,其余指标在海拔1150 m最大;随土层深度增加,同一海拔细根生物量、根长密度、表面积密度、体积密度均呈减少趋势。(2)随海拔梯度增加,0—30 cm土层细根C和N含量呈先增加后减少趋势,C/N比呈先减少后增加再减少趋势;随土层深度增加,同一海拔细根C含量呈先减少后增加趋势,N含量呈降低趋势,C/N比呈上升趋势。(3)细根N含量与生物量、根长密度和体积密度显著正相关,C/N比与生物量、根长密度、表面积密度和体积密度极显著负相关。(4)土壤水分对细根生物量及其形态指标影响显著。  相似文献

Type 2 diabetes (T2D) is characterized by impaired insulin secretion, insulin insensitivity and decreased beta-cell mass. Multiple genes contribute to T2D. The chromosome 12q13.1 region is in linkage to T2D in different populations, including our Italian dataset. CHOP is a candidate gene for the linkage, as it is located in the chromosome 12q13.1 region, and may contribute to T2D by increasing beta-cell apoptosis susceptibility and by impairing insulin sensitivity. Our goal was to identify any potential CHOP gene variants contributing to T2D in our Italian early-onset T2D families, which show linkage to the CHOP region. We directly sequenced the CHOP gene in 28 Italian probands of the linked T2D families and in 115 control subjects. We performed genotype and haplotype association tests with T2D of the identified single nucleotide polymorphisms (SNPs). We performed model-free and parametric association haplotype tests with T2D. We identified three SNPs [5'UTR-c.279T > C, 5'UTR-c.120A > G and + nt30C > T (F10F)] in CHOP. These SNPs are in complete linkage disequilibrium. The genotype association test showed an association trend with T2D of TT (F10F) and AG (-c.120A > G). The haplotype association test provided significant results for the haplotypes T/C (frequency = 0.33) and C/T (frequency = 0.01) (at 5'UTR-c.279T > C and + nt30C > T, respectively) under non-parametric analysis (P-value = 0.0000), recessive model (P-value = 0.0000) and additive model (P-value = 0.0014). Our data show that CHOP described haplotypes T/C and C/T, as an additive and as a homozygous variant, contribute significantly to T2D in our Italian early-onset group. We conclude that the CHOP T/C and C/T haplotype contributes to our T2D linkage signal on chromosome 12q13.1.  相似文献
