【目的】食线虫细菌对线虫的侵染机制报道甚少,限制了生防细菌的在农业实践中的广泛应用。本研究中,我们构建了一种对细菌杀线虫活性进行快速高效筛选的方法来快速寻找与线虫侵染功能相关的基因。【方法】将培养在固体平板上的野生型解淀粉芽孢杆菌FZB42及其突变株接种于5mL液体快杀培养基中,37℃培养24h后,在24孔板中分别加入200μL菌液和50-60条L4龄秀丽隐杆线虫(Caenorhabditis elegans),记录变化明显的时间点及线虫存活数,将相对于原始菌株杀线虫能力明显下降的突变株选出,并进行多次复筛。同时以传统的固体平板杀线虫法作为对照。【结果】用液体快杀法在21h时筛选出了与原始菌株相比杀线虫活性显著下降的2株突变株F1和F2,与传统的固体平板杀线虫法结果一致,且极大地缩短了筛选时间。【结论】该研究为下一步鉴定食线虫细菌中的毒力基因、进而阐明病原细菌侵染宿主的分子机制提供了良好的生物材料,并缩短了研究周期。  相似文献

【背景】相似穿孔线虫是一种重要的检疫性有害生物,其寄主范围广,危害具有毁灭性,已给我国农业生产造成潜在威胁。由于目前大多化学杀线剂毒性较高,筛选获得对相似穿孔线虫具有拮抗活性的微生物菌株对线虫防控具有重要意义。【方法】广泛从染病香蕉植株根际采集土壤样本,运用等比稀释涂布平板法分离菌株;采用摇瓶发酵法制备拮抗菌株上清液和发酵液。通过室内生物测定和盆栽试验,筛选出对相似穿孔线虫具有拮抗作用的细菌菌株,并通过16S rDNA的PCR扩增和序列分析,对拮抗菌株进行种类鉴定。【结果】获得5株对相似穿孔线虫具有较理想拮抗活性的细菌菌株。其中,菌株HD-86的作用效果最好,其发酵上清液处理24 h后的相似穿孔线虫死亡率达100%。盆栽试验表明,菌株HD-86发酵液处理42和70 d后对相似穿孔线虫的相对防效分别为77.34%和90.51%,均优于对照药剂阿维菌素。【结论与意义】菌株HD-86对相似穿孔线虫具有很强的拮抗作用,将其鉴定为洋葱伯克氏菌Burkholderia cepacia。本研究可为相似穿孔线虫生防制剂的研发和病害的防控提供依据。  相似文献

具杀线虫活性植物内生细菌的筛选和活性产物   总被引:1,自引:0,他引:1  
【目的】植物寄生线虫是危害植物的重要病原物,为了筛选到能在植物体内稳定定殖并且对植物寄生线虫具有较高杀线虫活性的植物内生细菌生防菌。【方法】以松材线虫为靶标,用直接触杀法进行筛选。对高活性菌株采用正交实验优化发酵条件,测定发酵液杀线虫活性的稳定性,并对菌株进行鉴定。【结果】从6种植物中分离筛选出13株对松材线虫具有较高杀线虫活性的植物内生细菌菌株,这些菌株的发酵上清液对松材线虫处理24h杀线虫率均达到了100%;其中BCM2、SZ5、CCM7和DP1这4个菌株的杀线虫活性较高,发酵上清液稀释3倍处理24h杀线虫率均达到95%以上,DP1和SZ5菌株达到了100%;并发现部分菌株发酵液能使线虫虫体发生渗漏或消解。发酵条件优化后能使发酵液杀线虫效果提高4倍。4株高活性菌株产生的杀线虫物质均对蛋白酶稳定、耐热不耐酸碱且长时间保藏活性不下降。经过鉴定DP1和CCM7是枯草芽孢杆菌(Bacillus subtilis),BCM2和SZ5是蜡样芽孢杆菌(Bacillus cereus)。【结论】经济作物体内存在一定数量的能产生杀线虫活性物质的内生细菌,其中一些细菌产生的杀线虫物质具有较强的稳定性。认为杀线虫活性的植物内生细菌具有很大的生防潜力。  相似文献

动物肠道细菌群落在联系宿主与生态系统功能中发挥着至关重要的作用。【目的】本研究旨在评估绿肥翻压和水稻生长不同时期对土壤细菌和线虫肠道细菌群落组成和结构的影响,并探究土壤细菌和线虫肠道细菌群落间的潜在关联关系。【方法】基于盆栽试验,结合16S rRNA基因高通量测序技术,分析黑麦草翻压和对照处理下水稻生长的前期(返青期)和后期(收获期)土壤细菌和线虫肠道细菌群落,结合网络分析研究土壤细菌网络互作对线虫肠道细菌群落的潜在影响。【结果】黑麦草翻压对土壤细菌和线虫肠道细菌群落组成和结构没有显著影响(P>0.05);水稻生长后期样品比前期样品具有更高的α多样性。基于随机森林机器学习法获得的土壤细菌和线虫肠道细菌生物标志物之间存在广泛的显著相关关系,为土壤细菌群落变化调控线虫肠道细菌群落组成提供了有力的证据。共现网络分析表明土壤细菌之间的正相互作用显著促进了土壤细菌和线虫肠道细菌之间的正相互作用(P<0.01),进而影响了线虫肠道细菌之间的网络互作。结构方程模型进一步表明土壤养分含量的降低主要通过降低土壤细菌之间正相互作用,从而间接影响线虫肠道细菌之间的互作。【结论】土壤细菌互作可能在...  相似文献

【目的】研究从棉田土壤中筛选得到的高产铁载体细菌产铁载体能力、分类地位和抑菌活性。【方法】通过改良蔗糖-天冬氨酸培养基选择性筛选产铁载体细菌,通过分光光度计法测定铁载体活性,通过混菌法测定产铁载体细菌上清液对棉花枯萎病致病菌尖孢镰刀菌(Fusarium oxysporum)的抑菌效果,采用形态学、生理生化鉴定及16S rDNA序列系统发育分析对高产铁载体菌株进行鉴定。【结果】从棉田土壤中筛选到162株产铁载体细菌,30株产铁载体能力较强的细菌中21株具有较高产铁载体能力,菌株SS05的铁载体活性单位达到98.3%;在低铁条件下,SS05上清液对F.oxysporum具有显著的抑制作用;SS05与莫哈韦芽孢杆菌(Bacillus mojavensis)最为接近。【结论】SS05是高产铁载体菌株,与莫哈韦芽孢杆菌(Bacillus mojavensis)最为接近,在低铁培养条件下其上清液对F.oxysporum具有显著的抑制作用。  相似文献

【目的】筛选亚低温条件下抗线虫和促进植物生长发育的菌株。【方法】采用线虫击倒率测定、室内耐低温测定、拮抗测试和盆栽生物测定相结合的方法进行功能菌株的筛选;采用表型特征、生理生化特征、16S r RNA基因序列测定相结合的多相分类技术对筛选的菌株进行鉴定。【结果】从根结线虫多发的设施黄瓜和番茄病土中分离出细菌和放线菌297株;经对根结线虫击倒率的初步筛选,得到校正击倒率大于70%的活性菌株9株;通过复筛获得1株在亚低温条件下同时具有防线虫、防土传病原菌病、促生特性的生防菌株S205;菌株S205被鉴定为抗生素链霉菌(Streptomyces antibioticus)。【结论】获得一株在亚低温条件下同时具有抗线虫活性和促进植物生长发育的生防菌株S205,该研究对解决亚低温条件下根结线虫的防治具有重要意义。  相似文献

【目的】比较7种植物源挥发物反式-β-法尼烯、橙花叔醇、6-甲基-5-庚烯-2-酮、顺-3-己烯乙酸酯、丁香酚、芳樟醇、顺式茉莉酮在玉米田中对天敌昆虫的诱集效果,为利用植物源挥发物对玉米田害虫进行生物防治提供参考。【方法】将7种植物源挥发物制作挥发物引诱球,利用水盆诱捕器、粘虫板和五点取样法调查百株虫量,比较7种植物源挥发物对捕食性和寄生性天敌昆虫的诱集效果。【结果】在7种植物源挥发物的筛选中,橙花叔醇对黄缘蜾蠃Anterhynchium flavomarginatum较其它6种化合物和对照的诱集效果有明显差异,顺式茉莉酮对黑带食蚜蝇Episyrphus balteatus较其它6种化合物和对照的诱集效果有明显差异,反式-β-法尼烯和顺式茉莉酮对龟纹瓢虫Propylaea japonica较其它5种化合物和对照的诱集效果有明显差异;在橙花叔醇和顺式茉莉酮的小区试验中,2种挥发物对龟纹瓢虫和异色瓢虫Harmonia axyridis的诱集效果明显优于对照,橙花叔醇对黑带食蚜蝇和虎斑食虫虻Astochia virgatipes的诱集效果明显优于对照。【结论】在7种植物源挥发物中,对玉米田天敌昆虫的诱集效果,橙花叔醇较好,顺式茉莉酮和反式-β-法尼烯次之。  相似文献

【背景】土壤中铁主要以难溶态铁氧化物的形式存在,有效性较低,产铁载体细菌对铁氧化物的活化是提高铁利用效率的有效途径。【目的】从林木土壤筛选产铁载体细菌,并观察菌株对难溶性铁氧化物的利用效应,可为土壤微生物资源开发及其在养分调控中的作用提供理论依据。【方法】通过CAS检测法从林木根系附近表层土壤中分离产铁载体细菌,借助生物培养实验分析温度和pH对微生物生长和铁载体产生的影响,通过振荡平衡实验,探究细菌产铁载体对铁氧化物的活化效应。【结果】通过CAS检测法从林木根系附近表层土壤中分离得到12株产铁载体细菌,16S rRNA基因扩增子测序初步鉴定结果显示筛选细菌均为假单胞菌属。选取铁载体产生能力和生长活性较高的两株细菌ARSB02和CNRSB01作为重点研究对象,结果显示,不同条件下CNRSB01的生物量和铁载体产量均高于菌株ARSB02,22 h时菌株ARSB02和CNRSB01的铁载体活性单位分别达到67.07%和84.60%。pH 5.0-8.0的范围内两株细菌可以保持较好的铁载体产生能力,菌株ARSB02和CNRSB01在pH7.0时铁载体产生能力最强,铁载体活性单位分别达到38.9...  相似文献

一株水稻纹枯菌拮抗细菌的分离与鉴定   总被引:1,自引:0,他引:1  
【目的】从土壤中分离并鉴定水稻纹枯菌拮抗细菌,测定其体外抑菌和温室防治效果。【方法】采用系列稀释法和平板对峙法筛选拮抗细菌,基于形态、生理特征及16S rDNA序列鉴定其分类地位,采用种子细菌化温室试验测定其防效。【结果】从蔬菜根际土壤中筛选出一株纹枯菌拮抗细菌,命名为kwkjT4。菌株具有明显的体外抑菌活性,对水稻纹枯病的温室防效与井冈霉素相当,初步鉴定为假紫色色杆菌(Chromobacterium pseudoviolaceum)。最适生长条件为pH 7.0,温度32°C,培养时间为36 h;抑菌活性物质产生的最适培养条件为pH 6.0,温度28°C,培养时间为48 h;表明两者并不一致。【结论】kwkjT4菌株在水稻纹枯病的生物防治中具有潜在的应用价值。这是C.pseudoviolaceum拮抗纹枯菌的首次报道。  相似文献

【目的】从土壤中分离、筛选和鉴定具有抑制病原真菌活性等生防效果的菌株,为进一步开发利用具有良好定殖能力的生防菌株奠定基础。【方法】利用对峙试验筛选拮抗菌并评价其拮抗性能,根据其形态特征、生理生化特性、16S r RNA基因序列测定及Gen Bank序列相似性分析进行分离菌株的分类鉴定,并通过福林酚法测定该菌株的蛋白酶活力。【结果】从山东泰安各种类型土壤中分离得到侧孢短芽孢杆菌(Brevibacillus laterosporus),保藏编号为AMCC100017。平板对峙试验结果表明该菌株对多种植物病原真菌均有较强的拮抗作用,尤其是对镰刀菌属致病菌拮抗效果明显。另外,本试验还初步验证该菌能产生较高活性的胞外蛋白酶。【结论】侧孢短芽孢杆菌AMCC 100017在作物真菌病害生物防治方面,有较好的开发和利用潜力,并可望应用于线虫生物防治。  相似文献

Competition is a major type of interaction between fungi and bacteria in soil and is also an important factor in suppression of plant diseases caused by soil-borne fungal pathogens. There is increasing attention for the possible role of volatiles in competitive interactions between bacteria and fungi. However, knowledge on the actual role of bacterial volatiles in interactions with fungi within soil microbial communities is lacking. Here, we examined colonization of sterile agricultural soils by fungi and bacteria from non-sterile soil inoculums during exposure to volatiles emitted by soil-derived bacterial communities. We found that colonization of soil by fungi was negatively affected by exposure to volatiles emitted by bacterial communities whereas that of bacteria was barely changed. Furthermore, there were strong effects of bacterial community volatiles on the assembly of fungal soil colonizers. Identification of volatile composition produced by bacterial communities revealed several compounds with known fungistatic activity. Our results are the first to reveal a collective volatile-mediated antagonism of soil bacteria against fungi. Given the better exploration abilities of filamentous fungi in unsaturated soils, this may be an important strategy for bacteria to defend occupied nutrient patches against invading fungi. Another implication of our research is that bacterial volatiles in soil atmospheres can have a major contribution to soil fungistasis.  相似文献

【目的】明确MeJA对茶树挥发物的诱导作用。【方法】采用顶空活体取样法对不同浓度MeJA处理后的茶苗挥发物进行抽提,并利用GC-MS对挥发物进行鉴定。【结果】不同剂量MeJA显著地影响茶树挥发物的种类组成和释放量,50μL MeJA处理可显著诱导茶树释放香叶烯、萜品油烯、罗勒烯等10种单萜类化合物,法呢烯、橙花叔醇和红没药烯等7种倍半萜类化合物,苯甲醇、苯乙腈和吲哚等5种氨基酸衍生物,以及3种未知化合物;而100μL MeJA处理仅能诱导茶树释放7种化合物。不同挥发物对MeJA处理的响应时间不同,但其释放量都具有昼高夜低的趋势。并且,释放量的大小明显受到光照强度的影响。【结论】外用MeJA喷雾处理可诱导茶树挥发物的产生和释放。  相似文献

Axenic, washed conidia of Fusarium solani f. sp. phaseoli, Aspergillus flavus, and Verticillium albo-atrum were placed on washed Difco purified agar discs along with an inorganic salt solution containing various levels of carbon and nitrogen substrates. These discs were exposed to volatiles from six soils (pH 5.1-8.6). Fusarium solani macroconidial germination was inhibited mostly by volatiles from soils of pH 5.1, 6.1, 7.0, and 7.5, but high levels of glucose and NH4Cl reversed this inhibition, raising germination to that of no-soil, no-carbon or nitrogen controls. Conidial germination of A. flavus was inhibited mainly by volatiles from high pH (7.0, 7.8, and 8.6) soils, and increased levels of glucose plus an amino acid mixture nullified this inhibition. Volatiles from soils of pH 5.1, 6.1, and 7.5 stimulated A. flavus conidial germination. Assays after the removal of CO2 from the air above soil of pH 5.1 demonstrated that volatiles inhibitory to A. flavus were produced by this soil. Assays indicated that a KOH-soluble compound was a fungistatic soil volatile to F. solani macroconidial germination. The nullification by carbon and nitrogen substrates of F. solani and A. flavus inhibition caused by soil volatiles parallels that for soil fungistasis. Conidial germination of V. albo-atrum was markedly stimulated by volatiles in all soils tested, and was not affected by removal of CO2. Inhibitory soil volatiles may increase the nutritional requirements for spore germination of certain fungi.  相似文献

Volatiles play major roles in mediating ecological interactions between soil (micro)organisms and plants. It is well‐established that microbial volatiles can increase root biomass and lateral root formation. To date, however, it is unknown whether microbial volatiles can affect directional root growth. Here, we present a novel method to study belowground volatile‐mediated interactions. As proof‐of‐concept, we designed a root Y‐tube olfactometer, and tested the effects of volatiles from four different soil‐borne fungi on directional growth of Brassica rapa roots in soil. Subsequently, we compared the fungal volatile organic compounds (VOCs) previously profiled with Gas Chromatography–Mass Spectrometry (GC–MS). Using our newly designed setup, we show that directional root growth in soil is differentially affected by fungal volatiles. Roots grew more frequently toward volatiles from the root pathogen Rhizoctonia solani, whereas volatiles from the other three saprophytic fungi did not impact directional root growth. GC–MS profiling showed that six VOCs were exclusively emitted by R. solani. These findings verify that this novel method is suitable to unravel the intriguing chemical cross‐talk between roots and soil‐borne fungi and its impact on root growth.  相似文献

【目的】为明确茉莉酸诱导的菜豆对西花蓟马Frankliniella occidentalis和南方小花蝽Orius similis的行为反应。【方法】采用四臂嗅觉仪测定了西花蓟马和南方小花蝽对不同浓度茉莉酸诱导菜豆后的行为反应,并用气相色谱-质谱联用仪测定了不同浓度茉莉酸处理后菜豆挥发物的成分。【结果】不同浓度茉莉酸处理的菜豆植株对西花蓟马和南方小花蝽分别有不同程度的驱避和吸引作用,以1 mmol/L的茉莉酸处理植株对西花蓟马的驱避作用最强,0.1 mmol/L的茉莉酸处理植株对南方小花蝽的吸引作用最强。不同处理菜豆的挥发物在含量和成分上存在显著差异,(Z)-3-己烯丙酸酯、2-异丙基-甲氧基毗嗦只有在茉莉酸处理植株中检测到。结合不同浓度茉莉酸处理植株对西花蓟马和南方小花蝽的行为反应及菜豆挥发物含量的变化趋势,推测(E)-2-己烯醛对西花蓟马有驱避作用,(E)-2-乙酸叶醇酯对南方小花蝽具有引诱作用。【结论】茉莉酸处理菜豆后,植物挥发物种类和含量发生了变化,在增强菜豆植株抗虫性的同时,还可增强捕食性天敌南方小花蝽的搜索和捕食能力。  相似文献

【背景】窖泥微生物的种类及其代谢产物类型是影响浓香型白酒发酵过程中丁酸和己酸等白酒中主要有机酸合成的影响因素之一。【目的】揭示浓香型白酒不同窖龄窖泥细菌群落结构,研究厌氧细菌产酸性能,阐明窖泥细菌与白酒中有机酸合成的相关性。【方法】通过Illumina HiSeq高通量测序,基于16S rRNA基因序列分析不同窖龄窖泥细菌的组成。分离获得厌氧细菌,通过比较菌株产丁酸和己酸能力来分析窖泥的微生物代谢特性。【结果】洋河酒窖泥细菌主要分布于梭菌纲(Clostridia)、拟杆菌纲(Bacteroidia)、互营养菌纲(Synergistia)和芽孢杆菌纲(Bacilli)。20年窖龄的窖泥中氢孢菌属(Hydrogenispora)和瘤胃梭菌属(Ruminiclostridium)丰度显著增加。窖泥细菌间相关性分析表明,瘤胃梭菌属(Ruminiclostridium)为窖泥中影响最大的核心微生物,很多微生物与梭菌属(Clostridium)菌株之间多为相互促进关系。通过传统可培养方法共分离得到梭菌目(Clostridiales)的20株厌氧菌。其中梭菌属(Clostridium)菌株产酸能力高...  相似文献

Endoparasitic and predatory nematophagous fungi are widely distributed throughout the maritime Antarctic, being recorded along the Antarctic Peninsula as far south as 68° S. Fungi were recorded from 71% of the sites examined with Cephalosporium balanoides and Dactylaria gracilis being the commonest recorded endoparasite and predator, respectively.Endoparasites with adhesive and nematode-attracting conidia were shown to be more abundant and to have a competitive advantage in the Antarctic ecosystem over those parasites requiring their conidia to be ingested before infection could occur. Predators able to form traps spontaneously on germination were shown to be far more abundant than those species with a more saprophytic mode of existence, with constricting rings being the most commonly isolated trapping mechanism. Species capturing nematodes by three-dimensional networks were restricted to bird-associated sites indicating that they are able to grow saprophytically in such organically enriched material.Nematophagous basidiomycetes and phycomycetes were absent except for a single Myzocytium sp. isolated from heated soil.  相似文献

In response to insect feeding on the leaves, cotton (Gossypium hirsutum L.) plants release elevated levels of volatiles, which can serve as a chemical signal that attracts natural enemies of the herbivore to the damaged plant. Pulse-labeling experiments with [13C]CO2 demonstrated that many of the volatiles released, including the acyclic terpenes (E,E)-[alpha]-farnesene, (E)-[beta]-farnesene, (E)-[beta]-ocimene, linalool, (E)-4,8-dimethyl-1,3,7-nonatriene, and (E/E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene, as well as the shikimate pathway product indole, are biosynthesized de novo following insect damage. However, other volatile constituents, including several cyclic terpenes, butyrates, and green leaf volatiles of the lipoxygenase pathway are released from storage or synthesized from stored intermediates. Analysis of volatiles from artificially damaged plants, with and without beet armyworm (Spodoptera exigua Hubner) oral secretions exogenously applied to the leaves, as well as volatiles from beet armyworm-damaged and -undamaged control plants, demonstrated that the application of caterpillar oral secretions increased both the production and release of several volatiles that are synthesized de novo in response to insect feeding. These results establish that the plant plays an active and dynamic role in mediating the interaction between herbivores and natural enemies of herbivores.  相似文献

Arthrobotrys conoides Drechsler,Arthrobotrys oligospora Fressenius andMonacrosporium rutgeriensis R. C. Cooke, Pramer belong to the peculiar group of predactious fungi which trap and kill nematodes. We have found that these cultures produce nematode-attracting and nematicidal substances the production of which is potentiated in the presence of nematodes. 植物释放的挥发性单萜在生态系统中起着重要的作用,这些单萜不仅能以空气为载体对其他植物显示直接的化感作用,而且也能以土壤为载体进行间接的化感作用。通过对三裂叶豚草挥发物对植物种子萌发和土壤微生物种群影响及其化学成分的鉴定研究,验证了以单萜类物质为主的三裂叶豚草挥发物可以经土壤载体对其他植物及土壤微生物显示化感效应。还对植物挥发物化学成分鉴定的GC (气相色谱) 和GC-MS (气相色谱-质谱联用) 方法存在的一些误区进行了澄清,这将有助于对生态系统中植物挥发物化感作用的研究和认识。  相似文献
