We isolated 400 aerobic heterotrophic bacteria from the sediment of unpolluted and polluted sites in a fast-flowing south Wales river. Isolates were subjected to taxonomic tests and screened for the presence of plasmid DNA by alkaline lysis and agarose gel techniques. There were no significant differences between sites in either the total percentage of isolates containing plasmids (unpolluted site, 9.4%; polluted site, 15%) or in the percentage of non-Pseudomonas-like isolates containing plasmids (unpolluted site, 15%; polluted site, 10%). There were significantly more Pseudomonas-like isolates with plasmids at the polluted site than at the unpolluted site (unpolluted site, 7%; polluted site, 18%). This presumably reflected a response of the nutritionally versatile Pseudomonas-like isolates to conditions at that site. The majority (86%) of the plasmids detected had molecular masses between 35 and 312 megadaltons. These plasmids were large enough to carry genes for conjugal transfer, suggesting the possibility of such transfer in this environment.  相似文献

The large-scale effects of habitat alteration produced by oil-industry related pollution on the habitat use of four species of freshwater turtles (Pelusios castaneus, Pelusios niger, Pelomedusa subrufa, Trionyx triunguis) were studied in the River Niger Delta, southern Nigeria (West Africa) between 1996 and 2004. The numbers of turtle specimens observed during our study declined drastically in polluted sites, despite a nearly identical field effort. The number of specimens of all turtle species declined considerably at all habitat types, but complete disappearance in polluted areas was found only with regard to one habitat type for Trionyx triunguis and two habitat types for Pelomedusa subrufa. The mean values of species dominance and diversity indexes were not statistically significant between pristine and altered areas. Based on the interspecific similarity in proportional frequencies of turtle specimens found in each habitat type, a multivariate set of analyses (UPGMA) showed that the turtles were arranged in three ‘ecological’ clusters: a group formed by Pelomedusa subrufa at both polluted and unpolluted areas and Trionyx triunguis at polluted areas; (ii) a group formed by Pelusios castaneus in polluted areas and Pelusios niger in polluted areas; (iii) a group formed by Pelusios castaneus in unpolluted areas and Pelusios niger in unpolluted areas; however, this latter cluster was not very close, as the linkage distance was close to 80% of Euclidean distance. Habitat use similarity among turtles in both polluted and unpolluted study areas was evaluated by the use of two types of overlap formulas (Pianka and Czechanowski) and the use of Monte Carlo randomisations in order to control for the eventual role of chance in the actual data matrix. These data indicated that, for a pair of species (Pelusios niger vs. Pelusios castaneus), there was a statistically significant increase in the similarity of habitat use in the polluted areas vs. the unpolluted areas, and that this pattern was not dependent on the chance. Considering that these two species are ecologically and morphologically similar, we conclude that the most likely consequence at the community level is an increase in the intensity of interspecific competition for space between Pelusios niger and Pelusios castaneus in the polluted areas. Although the direction of the intensification of this competition process could not be easily predicted, it is likely that the species which is least adapted to life in main rivers and creeks may be disadvantaged over the other competitor. The general implications for habitat preservation are also discussed.  相似文献

The prevalence of allergic disease has increased world wide during the last decades. Pollen allergy is the most typical form of allergic disease. The increase in its frequency during recent years is the most evident. Environmental factors play an important role in the problem of pollen allergy in large cities. The aim of this research is determination of allergenicity of Canna pollen in polluted and non-polluted conditions, detection of their allergenic proteins and also elucidation of some microscopic effects of air pollutants on pollen structure and proteins. Mature and immature pollen grains of Canna indica were collected from polluted and non-polluted areas. Pollen grains were studied by scanning electron microscopy. Mice were sensitized by injection of pollen extract and an adjuvant for five times. Allergy potency of different pollen extracts were compared by means of skin test, blood eosinophills number and IgE levels in sensitized and treated animals. Pollen proteins were studied by SDS-PGE and allergenic proteins were detected by immunoblotting techniques. Scanning electron microscope study of the pollen grains showed that in polluted areas, air born particles accumulated on the surface of pollen and changed both pollen's shape and pollen's tectum. Also many vesicles were released out of polluted pollen and the pollen material agglomerated on the surface of pollen. SDS-PAGE showed that different proteins exist in mature and immature pollen. In pollen collected from polluted area, some of protein bands between 22 and 45 kDa were disappeared . Also in all polluted pollen grains, protein content of pollen decreased in response to air pollution causing the release of pollen proteins. According to our experiments and regarding induction of allergic symptoms, the polluted pollen is more effective than non-polluted one, and mature pollen has more allergy potency than immature one.  相似文献

The aim of this study was to define a model for the coupling between extracellular enzyme activity and substrate uptake by bacterial populations in natural waters. The balance between uptake of leucine and extracellular hydrolytic production of leucine from a peptide model substrate was investigated in a combined fluorescence-radiotracer experiment with [H]leucine as a marker for the leucine pool and l-leucine-4 methyl-7-coumarinylamide (Leu-MCA) as a marker for the pool of dissolved peptide substrates. Results show that at low concentrations of the model substrate the input and uptake processes of leucine are nearly balanced, whereas at high concentrations of the model substrate much more leucine is liberated than taken up. In addition, samples from one polluted and one less polluted station in the Kiel Fjord were investigated for their extracellular enzymatic and uptake properties in an annual cycle. It was found that turnover rates of leucine (T(r), percent per hour) and hydrolysis rates of Leu-MCA (H(r), percent per hour), as well as the quotient T(r)/H(r), reflect the impact of environmental conditions on decomposition processes at both sampling sites. The quotient T(r)/H(r) is interpreted as an indirect measurement of the pool size ratio (polymers/monomers), which may serve as an index of hydrolysis-uptake coupling in bacterial utilization of dissolved protein. Calculated on an annual average basis, turnover rates are ca. nine times higher than hydrolysis rates at the polluted station and ca. five times higher at the less polluted station. From the described model, this would mean that the relative fraction of polymers within the total dissolved organic carbon pool (with regard to the substrate combination dissolved protein-leucine) is about twice that at the polluted than at the less polluted station.  相似文献

The yellow carotenoid-based plumage coloration of great tit Parus major nestlings is found to be paler in polluted and urban environments. Because carotenoid pigmentation is often considered to be a condition dependent trait in birds we wanted to find out whether food-limitation and poor nestling condition could explain the pale plumage colour in a polluted area. P. major nestlings were supplemented with variable diets along a well known heavy metal pollution gradient around a copper smelter: two food treatments with carotenoids, one food treatment with little carotenoid and one unsupplemented control. Our field experiment showed that nestlings in the polluted area grew better with carotenoid rich diets, while such effect was not found in the unpolluted area. Nestlings showed higher plasma carotenoid (lutein) levels and higher plumage carotenoid chroma values in the unpolluted area than in the polluted area. However, plasma lutein levels or plumage colour were not associated with heavy metal levels in nestling faeces (a proxy for dietary exposure). Our results provide only weak evidence for carotenoid-based colouration to be condition-dependent in great tit nestlings as we found a positive relationship between body mass and carotenoid chroma in the non-supplemented control group only. The positive relationship between body mass and plumage colour intensity is more likely to be produced by the fact that good availability of caterpillars, an important food source for P. major, also means a good availability of carotenoids to nestlings. Our results suggest that main reason for pale nestling plumage in the polluted area is lower quality invertebrate food, and not nutrition-related oxidative stress.  相似文献

Chemical forms of zinc in light chestnut soil polluted by zinc, copper, cadmium, and lead, introduced separately or jointly, were studied. A change in the initial relations between the element forms was found for all types of pollution. An increase in pollution of any type led to the accumulation of the most mobile forms of zinc in the soil. It was revealed that ion exchange is of the greatest significance in the fixation of zinc in polluted soil.  相似文献

Environmental pollutants cause a potential hazard for survival in free-living animal populations. We modelled local survival (including emigration) by using individual mark-recapture histories of males and females in a population of a small insectivorous passerine bird, the pied flycatcher (Ficedula hypoleuca) living around a point source of heavy metals (copper smelter). Local survival of F. hypoleuca females did not differ between polluted and unpolluted environments. Males, however, showed a one-third higher local-survival probability in the polluted area. Low fledgling production was generally associated with decreased local survival, but males in the polluted area showed relatively high local survival, irrespective of their fledgling number. A possible explanation of higher local survival of males in the polluted area could be a pollution-induced change in hormone (e.g. corticosterone or testosterone) levels of males. It could make them to invest more on their own survival or affect the hormonal control of breeding dispersal. The local survival of males decreased in the polluted area over the study period along with the simultaneous decrease in heavy metal emissions. This temporal trend is in agreement with the stress hormone hypothesis.  相似文献

We compared the general activity and heavy metal resistance of non-specific esterases in two populations of the leaf beetle Chrysomela lapponica from habitats severely contaminated by heavy metals (mostly Ni and Cu) and two populations from unpolluted habitats. Concentrations of Ni and Cu in adult beetles from the most polluted site were 7.7 and 3.6 times higher that in beetles from unpolluted habitats. Larval esterases showed higher activity and lower susceptibility to heavy metals than esterases of adults. Larval esterase activity did not differ between populations from polluted and unpolluted sites, but adult beetles from polluted localities had lower esterase activity than beetles from unpolluted habitats. Both Cu and Ni sulfates in millimolar concentrations in vitro suppressed esterase activity of larvae from unpolluted habitats, but caused no negative effect on esterases of larvae from polluted sites. Similarly, inhibition of adult esterase activity by Ni was stronger in beetles from unpolluted localities than in beetles from polluted localities. This indicates that resistance of non-specific esterases to heavy metals is higher in leaf beetle populations from contaminated environment.  相似文献

Organic concentrates were recovered using XAD-2/8 resin adsorption from the leachates of municipal solid waste landfills and their mutagenic activities were tested for 8 months using the Ames Salmonella/microsome assay. Highly polluted leachates (COD and BOD > or = 40 mg/l) generally had equal or higher mutagenic activities than lightly polluted leachates (COD and BOD < 40 mg/l). But there was no clear difference in mutagenicity per amount of concentrate between the two leachates. These results suggest that the mutagenic activity of landfill leachate is decided to some degree by the organic concentration in the leachate. The mutagenic activities detected even in lightly polluted leachates were not so low as those of various kind of surface waters ever reported. It is suggested that it is important to investigate the mutagenic activity of the leachate for evaluation of the impact of landfill leachate on the environment.  相似文献

Allozyme variation was determined in two land snail species (Cepaea nemoralis and Succinea putris) from four localities in northern Belgium. In each locality we selected a polluted and a nearby, less-polluted, reference plot. We examined whether (i) genetic variability differed between the polluted and reference plots, (ii) populations from polluted plots experienced recent bottlenecks, and (iii) certain allele or genotype frequencies were associated with the pollution. Our results suggest that (i) about 13% of the genetic differentiation in C. nemoralis and 5% in S. putris was due to differences among polluted and reference plots, (ii) polluted and reference plots had comparable levels of genetic variation, but in C. nemoralis observed heterozygosities were higher in polluted plots, (iii) most plots showed significant evidence for recent bottlenecks, irrespective of the degree of pollution, so that bottlenecks seem poor indicators of pollution-induced stress in land snails, and (iv) mutagenic or pollution-induced modifications did not seem to account for new allozyme variants in polluted sites. The observed patterns of genetic variation may be explained by the action of genetic drift, pollution-mediated selection, restricted gene flow, or a combination of these processes.  相似文献

Fish from polluted waters are subject to increased prevalence of disease. Because they respond to bacterial pathogens by producing serum antibodies, it was possible to construct a seasonal serological record in three fish species from clean and polluted waters of the New York Bight. Antibody levels were determined by testing sera for agglutinating activity against 36 strains of bacteria. Evaluation of 5,100 antibody titrations showed the following. During warm months, summer flounder (Paralichthys dentatus) from the polluted area had significantly higher antibody levels and antibody to a greater diversity of bacteria than fish from the unpolluted area. Weakfish (Cynoscion regalis) from the same polluted area shared with summer flounder raised titers to many bacteria. The greatest proportion of raised titers was against Vibrio species, although prominent titers were also seen against Aeromonas salmonicida and Haemophilus piscium, bacteria usually associated with diseases in freshwater but not marine fish. Differences between polluted and clean waters were not as evident in winter flounder (Pseudopleuronectes americanus) during cold months. This could be due, in part, to reduced antibody production at colder temperatures. The data illustrate the usefulness of the serum antibody record in identifying environmental exposure to bacteria in marine fish and indicate that the polluted New York Bight apex has increased levels and diversity of bacteria during warm months.  相似文献

Analysis of chromosomal aberrations in bone marrow cells and determination of the level of fluctuating asymmetry (FA) for eight bilateral cranial traits were used to estimate effects of disturbed environment on natural populations of small mammals. During the spring and fall of 1994-2000 (except in 1999) black-striped mice (Apodemus agrarius) were collected from a polluted industrial area (Pancevo) and from an unpolluted reference site (Cer) in Serbia and Montenegro. Mice from the polluted area had significantly higher mean number of aberrant cells per individual than those in the reference site (p<0.01). Both sexes had the greater number of lesions in the polluted site than in the reference one. Also, there were no significant differences between sexes, neither within the same site/season nor between sites/seasons. Chi-square analysis of the difference between lesions per individual and aberrant cells per individual among sites showed that no site deviated from the expected difference (p=1.000). Developmental stability, assessed as FA, was reduced in the polluted area for two traits (width of lower jaw and length of the diastema lower jaw). Juvenile animals from the polluted area had significantly higher levels of FA compared to those from the reference site for three traits (length of the foramina incisiva, distance between incisor and the third upper molar, and length of the upper molars row). Juvenile animals from the polluted area had significantly higher levels of FA for three traits (length of the foramina incisiva, length of the lower molars row and width of lower jaw) compared to adult ones. In the polluted area seasonal differences were detected for two traits (length of the foramina incisiva and length of the upper molars row). Finally, Apodemus agrarius may be an important species for environmental quality evaluation studies based on an assay using a combination of end-points.  相似文献

Arabidopsis halleri, a close wild relative of A. thaliana, is a clonal, insect-pollinated herb tolerant to heavy metals (Zn, Pd, Cd) and a hyperaccumulator of Zn and Cd. It is of particular interest in the study of evolutionary processes and phytoremediation. However, little is known about its population gene flow patterns and the structure of its genetic diversity. We used five microsatellite loci to investigate the genetic structure at a fine spatial scale (10 cm to 500 m) in a metallicolous population of A. halleri. We also studied the contributions made by clonal propagation and sexual reproduction (seed and pollen dispersal) to the genetic patterns. Clonal diversity was high (D(G) > 0.9). Clonal spread occurs only at short distances (< 1 m). Both clonal spread and limited dispersal, associated with sexual reproduction, contribute to the significant spatial genetic structure revealed by spatial autocorrelation analysis. The shape of the autocorrelogram suggests that seed dispersal is restricted and pollen flow extensive, which may be related to intense activity by insect pollinators. Clonal spread was more extensive in the lowly polluted zone than in the highly polluted zone. This cannot be interpreted as a strategy for promoting the propagation of adapted genotypes under the harshest ecological constraints (highest heavy metal concentrations). The higher fine-scale spatial genetic structure found in the lowly polluted zone can be ascribed to plant densities that were lower than in the highly polluted zone. No evidence of genetic divergence due to spatial heavy metal heterogeneity was found between lowly and highly polluted zones.  相似文献

Soil bacterial biomass, phospholipid fatty acid pattern, pH tolerance, and growth rate were studied in a forest area in Finland that is polluted with alkaline dust from an iron and steel works. The pollution raised the pH of the humus layer from 4.1 to 6.6. Total bacterial numbers and the total amounts of bacterial phospholipid fatty acids in the humus layer did not differ between the unpolluted control sites and the polluted ones. The number of CFU increased by a factor of 6.4 in the polluted sites compared with the controls, while the bacterial growth rate, measured by the thymidine incorporation technique, increased about 1.8-fold in the polluted sites. A shift in the pattern of phospholipid fatty acids indicated a shift in the bacterial species composition. The largest proportional increase was found for the fatty acid 10Me18:0, which indicated an increase in the number of actinomycetes in the polluted sites. The levels of the fatty acids i14:0, 16:1ω5, cy17:0, 18:1ω7, and 19:1 also increased in the polluted sites while those of fatty acids 15:0, i15:0, 10Me16:0, 16:1ω7t, 18:1ω9, and cy19:0 decreased compared with the unpolluted sites. An altered pH tolerance of the bacterial assemblage was detected either as a decrease in acid-tolerant CFU in the polluted sites or as altered bacterial growth rates at different pHs. The latter was estimated by measuring the thymidine incorporation rate of bacteria extracted from soil by homogenization-centrifugation at different pHs.  相似文献

The rate of development and degree of survival of Asellus aquaticus eggs outside the marsupium of ovigerous females are affected by water quality and temperature. Eggs were maintained in polluted river water and relatively clean canal water. Developmental rates increase with increased temperature, but survival decreases. Eggs from polluted site ovigerous females survive better in clean water than in polluted water. Eggs from clean site ovigerous females maintained in polluted water have significantly lower survival rates than eggs from the polluted site at all temperatures tested. The developmental rate of clean site eggs is increased significantly in polluted water at 10–25 °C, possibly as a response to the stress imposed upon them.It is suggested that the method outlined might form the basis of a useful bioassay technique for measuring water quality.  相似文献

The hyporheic zone and its interactions with coarse surface sediments is increasingly reported by aquatic ecologists because the water exchanges between surface and subsurface are important factors for the understanding of the ecosystem functioning. However, the hyproheic oligochaete assemblages have received less attention than other assemblages such as crustaceans. In addition, studies investigating the incidence of pollution in watercourses have mostly focused on the benthic zone and have neglected the hyporheic zone. Some examples are given from an unpolluted glacial river (Roseg), polluted plains rivers (Moselle, Rhône) and a protected wetland in an urbanized environment. The hyporheic zone kept the memory of past and present incidences of pollution, in particular when downwellings of polluted surface waters to the hyporheic zone predominated. The Active hydrologic Exchange Describers between surface and subsurface (AED oligochaete species) were the same in the glacial river Roseg, the rivers Rhône and Moselle and the urbanized wetland. The predominance of pollution-tolerant species like Limnodrilus hoffmeisteri was observed in polluted groundwater as well as in polluted surface coarse sediments. Moreover, the urbanized wetland exhibited a high species richness, suggesting that the hyporheic zone is a reservoir of species. The oligochaete communities enable biologists to simultaneously assess the pollution incidence, the permeability of coarse habitats, the water exchanges between surface and subsurface, and give an approximate measure of the metabolic activities in the sediments. Consequently, the simultaneous study of surface and hyporheic oligochaete assemblages is of great interest when considering the ecological functioning of watercourses and the incidence of pollution inputs.  相似文献
