An inexpensive, easily constructed sampler for collecting interstitial water and sediment, seperately or combined is presented. The instrument has been developed for use in wetlands and shallow water as a tool for taxonomical and ecological investigations. Different to other samplers, our sampling depth is down to more than one meter at defined depths. Sampling showed highly reproducable sampling results for both, hydrophysical-chemical and biological analysis, which will be shown in the article.  相似文献

The airborne pollen concentration in the city of Badajoz was measured in two locations 2.9 km apart. The measurements were taken from January to June between 2009 and 2012 using Hirst-type volumetric aerobiological samplers. One sampler was placed at the Faculty of Science (FS) and the other at the Agricultural Engineering School (AES) of the University of Extremadura, Spain, on terraces located 16 and 6 m above ground, respectively. The two sets of hourly and daily pollen concentrations were compared regarding the meteorological parameters and the distribution of local pollen sources. A total of 46 pollen types were counted, with a mean total concentration of 43 pollen grains/m³ in the winter and 336 pollen grains/m³ in the spring. In the winter, pollen grains from (in decreasing order) Cupressaceae, FraxinusPhillyrea, Urticaceae spp., Alnus glutinosa and Urtica membranacea types represented 77 % of the total. In the spring, 89 % of the total was represented by pollen grains from (in decreasing order) Quercus, Poaceae, Olea, Pinaceae and Plantago. The FS/AES ratio was 0.57 in the winter and 1.31 in the spring. While a Wilcoxon test applied to the daily total concentration data showed statistically significant differences between the two sites, a correlation study based on the Spearman coefficient showed statistically significant correlations in both the winter and spring. The results were similar when comparing the separate pollen types, except for Urticaceae spp., which showed no statistically significant correlation. The meteorological data studied showed a statistically significant correlation with the daily concentrations. A comparison of the hourly concentration data showed no correspondence with the time of maximum concentration. The local distribution of pollen sources explained some of the differences found between the two sites. Overall, the results indicate that a single aerobiological sampler may be sufficiently representative to register the daily pollen grain data of an urban area of approximately 3 km or greater in diameter, although it underestimates the influence of heterogeneity in the distribution of the local flora.  相似文献

Results of measurements of the resuspended radioactive aerosols in the Chernobyl area are presented which were obtained soon after the Chernobyl reactor accident and in a European project in 1992–1993. The measurements were carried out with the intention of obtaining a data base for dose assessment of resuspended radioactive particles. Potential significant dose contributions may result from inhalation and secondary contamination due to resuspended radionuclides. In this first article of a series of three papers, the instrumentation and the measurement uncertainties are discussed. An effort was made to sample quantitatively giant aerosol particles (particles larger than 10 μm aerodynamic diameter) as well. The comparison of the samplers shows, in general, an agreement of concentration measurements of 137Cs and 7Be within a factor of two. One sampler was identified with larger discrepancies and needs additional investigation of its sampling characteristics; for another device, the recalibration of the analysing system is recommended. Ordinary integrating samplers have a loss of about 30% in 137Cs activity compared to an isokinetic sampler collecting giant particles as well. The mean ratio of 137Cs activity concentration between an instrument sampling only particles larger than 10 μm and an ordinary integrating sampler is 0.39 ± 0.15 during anthropogenic-enhanced resuspension. These findings demonstrate the significant contribution of giant particles to resuspended airborne radioactivity. The results of this study concerning integral measurements during wind-driven resuspension proved to be in good agreement with previously published data on resuspension. Previous field and laboratory studies investigating airborne Burkholderia pseudomallei have used a variety of different aerosol samplers to detect and quantify concentrations of the bacteria in aerosols. However, the performance of aerosol samplers can vary in their ability to preserve the viability of collected microorganisms, depending on the resistance of the organisms to impaction, desiccation, or other stresses associated with the sampling process. Consequently, sampler selection is critical to maximizing the probability of detecting viable microorganisms in collected air samples in field studies and for accurate determination of aerosol concentrations in laboratory studies. To inform such decisions, the present study assessed the performance of four laboratory aerosol samplers, specifically the all-glass impinger (AGI), gelatin filter, midget impinger, and Mercer cascade impactor, for collecting aerosols containing B. pseudomallei generated from suspensions in two types of culture media. The results suggest that the relative performance of the sampling devices is dependent on the suspension medium utilized for aerosolization. Performance across the four samplers was similar for aerosols generated from suspensions supplemented with 4% glycerol. However, for aerosols generated from suspensions without glycerol, use of the filter sampler or an impactor resulted in significantly lower estimates of the viable aerosol concentration than those obtained with either the AGI or midget impinger. These results demonstrate that sampler selection has the potential to affect estimation of doses in inhalational animal models of melioidosis, as well as the likelihood of detection of viable B. pseudomallei in the environment, and will be useful to inform design of future laboratory and field studies.  相似文献

A modified large-volume sampler was demonstrated to be an efficient device for the collection of mono-disperse aerosols of rhodamine B and poly-disperse aerosols of bacterial cells. Absolute efficiency for collection of rhodamine B varied from 100% with 5-μm particles to about 70% with 0.5-μm particles. The sampler concentrated the particles from 950 liters of air into a flow of between 1 and 2 ml of collecting fluid per min. Spores of Bacillus subtilis var. niger were collected at an efficiency of about 82% compared to the collection in the standard AGI-30 sampler. In the most desirable collecting fluids tested, aerosolized cells of Serratia marcescens, Escherichia coli, and Aerobacter aerogenes were collected at comparative efficiencies of approximately 90, 80, and 90%, respectively. The modified sampler has practical application in the study of aerosol transmission of respiratory pathogens.  相似文献

SUMMARY. 1. As three previous comparative studies of deep-water samplers for benthic macroinvertebrates in rivers highlighted the need for a quantitative sampler for use on stony substrata, a new air-lift sampler was developed. It can be operated from a small boat by two people, weighs 13.5–20.0 kg, depending on the length of riser used, and extends the maximum range of substrata that may be sampled quantitatively from 16–32 mm to 128—256 mm. The sampling area is isolated by forcing a collecting cylinder into the substratum, and rapid evacuation of the contents is assisted by a vibrator.
2. All the major specifications of the sampler were determined experimentally in a large tank using three sizes of substrata and plastic pellets to represent invertebrates. The sampler performed accurately to a depth of at least 8 cm on substrata ranging from gravel (2–4 mm) to large stones (32–36 mm long).
3. The performance of the sampler was compared with that of a Ponar grab and Pearson el at. air-lift sampler at two sites on a large river and also with a Naturalist's dredge and a diver-operated Hess-Waters sampler at one of the sites where there were large stones up to 280 mm long. In terms of both mean taxa per sample and mean numbers m−2, samples taken using the new air-lift sampler provided estimates comparable to or belter than those obtained with the other samplers.  相似文献

The importance of the transport of pollen by air movement into houses was evaluated using six to eight simultaneously collecting rotorod-type samplers, creating either a sampler line from outdoors to inside the room, or a sampler grid inside a room. The number of incoming pollen grains was highly dependent on the outdoor concentration. The highest concentrations inside (1–2 m distance) and outside (1 m) the room were 600 and 3,250 grains/m3, respectively, in the Betula pollen season and 1,980 and 5,080 grains/m3 in the Pinus season. The pollen concentration and the indoor/outdoor (I/O) ratio decreased as the distance from the ventilation opening increased. Inside the room at a distance of 1–2 m 28%, and at a distance of 3–5 m 12%, of the outside concentration was recorded. In the lower part of the opening the mean proportion was 63% and in the upper part of the opening it was 40%. Efficient ventilation with two open windows increased the I/O ratio and enabled the pollen to spread throughout the room. During the Pinus pollen season 3–35% of the outdoor concentration was simultaneously recorded at six locations inside the room with two open windows and only 0.1–3.6% with one open window. At the same point in the room the I/O ratio varied from <1 to 35%, depending on the sampling conditions. Only a minor effect on the I/O ratio was found between small and large ventilation windows and the door, although it was expected that more air and pollen grains would come indoors through a larger opening.  相似文献

Using Surber-type samplers and dip-net samplers, we assessed the efficiency of nets having pore sizes of 720 μm and 320 μm for determining standing crop and percentage composition of the stream fauna, and for collecting representative size-class specimens of Ephemeroptera and Plecoptera to be used in life-cycle studies. Except for one species, samples collected with either the 720-μm or 320-μm dip-net led to the same general inferences about the species' life cycle. Of fifty possible sample comparisons, there were twelve samples where the size-class frequencies of particular species collected in the 720-μm dip-net were significantly different from the size-class frequencies of the 320-μm dip-net; for five of these samples a deficit of large nymphs (> 5.0 mm) in the 320-μm net mainly contributed to the significant χ2 values. On one date, we used double-bag samplers with both th e 720-μm and 320-μm nets attached to either the Surber or dip-net sampler. Approximately 50% of the insects by numbers passed through the 720 fxm mesh ofeach sampler, but only 5% by volume-biomass. Shape of the insect as well as body length was important in assessing mesh-size efficiencies. The 720-μm mesh of the double-bag dip-net sampler retained most of the Nemoura dnctipes (having stout appendages) and Epeorus longimanus (flattened) nymphs 2.0 mm in body length and larger; whereas most Baetis (streamlined) nymphs smaller than 3.0 mm and all Paraleuctra (needle-like shape) nymphs passed through the 720-μm mesh.  相似文献

Two commercially available soil water samplers and a ceramic sampler constructed in our laboratories were evaluated for their ability to recover viruses from both tap water and secondary sewage effluent. The ceramic sampler consistently gave the best recoveries of viruses from water samples. SUMMARY. From 16 air-lift samplers described in the literature, three were selected for operation from a small boat: Mackey, Pearson et al. and Verollet & Tachet samplers. Random samples (number of sampling units n= 10) were taken in a large tank with a known number of dried peas, representing invertebrates, amongst stones of uniform size. Separate experiments were performed with three sizes of stones (modal sizes 2–4, 16–20 and 32–36 mm). Stratified-random samples (usually n= 10) were taken in rivers at lour sites with modal particle sizes of < 0.1 2–4. 32–128 and 64–128 mm. The samplers were compared with a Ponar grab in the tank experiments and the field, and with a Naturalist's dredge in the field. Rates of pumping at different air-flows were established for each sampler. Water-How increased with increasing air-How and increasing submergence of the riser-pipe. The Mackey sampler achieved the highest water-flow and lifted more substratum and larger stones (64–128 mm) than the other samplers. The Verollet & Tachet sampler had the lowest pumping rate and failed to lift stones > 8 mm long, whilst the Pearson et al. sampler, with an intermediate performance, lifted mud and stones < 32 mm long. In the tank experiments, the area from which the Mackey and Pearson et al. samplers collected peas varied with air-flow, operating time and type of substratum. The Maekey sampler over-sampled and the Pearson et al. sampler usually under-sampled their respective sampling areas, except the latter sampler at high air-flows gave approximately quantitative results comparable to those of the Ponar grab for peas on the surface of stones 16–20 mm. No sampler collected much at a depth of 3 cm on larger stones (32–36 mm), and the Verollet & Tachet sampler failed completely on all substrata. In field trials, the Mackey and Pearson et al. samplers provided good qualitative samples except on large stones (32–128mm). The Verollet & Tachet sampler lifted little material. The relative abundance of taxa often differed between samplers. Estimates of the number of invertebrates per m2 differed widely between samplers except for the Pearson et al. and Ponar grab which gave similar quantitative results at three sites. Values were very low for the Verollet & Tachet sampler and Naturalist's dredge and always very high for the Mackey sampler which often grossly over-sampled its sampling area. Therefore, these three samplers cannot be considered as quantitative. The relationship between the variances and the means of samples of peas in the tank and most invertebrates in the field followed a power law with values of the exponent b in the range 1.13–2.51. The present study completes the evaluation of 14 samplers used to sample benthic macro-invertebrates in deep rivers, and the more important conclusions from the comparative studies of seven grabs, four dredges and three air-lift samplers are summarized in a table.  相似文献

Root morphology of 5-year-old trees of Dacryodes edulis (G. Don) H. J. Lam of seed and two vegetative (cutting and marcot) origins were assessed in Cameroon. Roots of D. edulis trees of seed and vegetative origins were totally excavated and their root morphology described and quantified. Trees of seed origin were characterized by a tap root, reaching depths of about 1.2 m. Contrarily trees of cutting origin showed three strong vertical roots (d > 5 mm) with the longest reaching depths of 1.31 m. Trees of marcot origin were observed to have thick, relatively short prominent vertical roots, reaching depths of 1.15 m. At fixed lateral intra-row distances of 50 cm from tree bases, trees of seed and marcot origins recorded high root densities at soil depths of 0–20 cm; both differed significantly (p = 0.032) from those of trees of cutting origin. At soil depth of 60–100 cm, trees of cutting origin recorded the highest root density which however differ significantly (p = 0.016) from those of trees of both seed and marcot origins. D. edulis trees of seed origin had a tap root system, whereas trees of vegetative origin (cuttings and marcots) had developed prominent main adventitious roots growing vertically (sinker roots), into greater depths and a wider/deeper lateral root spread than seedlings. This could be a vital adaptation to the absence of a genetically determined tap roots in trees of vegetative origins so as not to compromise the acquisition of soil-based resources (water and dissolved ions) and anchorage.  相似文献

The diving ability of juvenile animals is constrained by their physiology, morphology and lack of experience, compared to adults. We studied the influences of age and mass on the diving behaviour of juvenile (2–3-year-old females, n = 12; 3–5-year-old males, n = 7) New Zealand (NZ) sea lions (Phocarctos hookeri) using time–depth recorders (TDRs) from 2008 to 2010 in the NZ subantarctic Auckland Islands. Diving ability (e.g. dive depth, duration and bottom time per dive) improved with age and mass. However, the percentage of each dive spent at the bottom, along with percentage time at sea spent diving, was comparable between younger and lighter juveniles and older and heavier juveniles. These suggest that younger and older juveniles expend similar foraging effort in terms of the amount of time spent underwater. Only, 5-year-old male juveniles dove to adult female depths and durations and had the highest foraging efficiency at depths >250 m. It appears that juvenile NZ sea lions attain adult female diving ability at around 5 years of age (at least in males), but prior to this, their performance is limited. Overall, the restricted diving capabilities of juvenile NZ sea lions may limit their available foraging habitat and ability to acquire food at deeper depths. The lower diving ability of juvenile NZ sea lions compared to adults, along with juvenile-specific constraints, should be taken into consideration for the effective management of this declining, nationally critical species.  相似文献

An investigation into the insular shelf and submerged banks surrounding Tutuila, American Samoa, was conducted using a towed camera system. Surveys confirmed the presence of zooxanthellate scleractinian coral communities at mesophotic depths (30–110 m). Quantification of video data, separated into 10-m-depth intervals, yielded a vertical, landward-to-seaward and horizontal distribution of benthic assemblages. Hard substrata composed a majority of bottom cover in shallow water, whereas unconsolidated sediments dominated the deep insular shelf and outer reef slopes. Scleractinian coral cover was highest atop mid-shelf patch reefs and on the submerged bank tops in depths of 30–50 m. Macroalgal cover was highest near shore and on reef slopes approaching the bank tops at 50–60 m. Percent cover of scleractinian coral colony morphology revealed a number of trends. Encrusting corals belonging to the genus Montipora were most abundant at shallow depths with cover gradually decreasing as depth increased. Massive corals, such as Porites spp., displayed a similar trend. Percent cover values of plate-like corals formed a normal distribution, with the highest cover observed in the 60–70 m depth range. Shallow plate-like corals belonged mostly to the genus Acropora and appeared to be significantly prevalent on the northeastern and eastern banks. Deeper plate-like corals on the reef slopes were dominated by Leptoseris, Pachyseris, or Montipora genera. Branching coral cover was high in the 80–110 m depth range. Columnar and free-living corals were also occasionally observed from 40–70 m.  相似文献

Asterias amurensis is a starfish native to the northern Pacific that was introduced into southern Australia in the 1980s. It is widely viewed as one of Australia’s most serious invasive marine pests, but there are few methods available to control new or established populations. The role of Coscinasterias muricata in controlling the distribution of Asterias in Port Phillip Bay, and its potential to eliminate new infestations of Asterias were investigated. Laboratory feeding trials, where alternative mussel prey were available, showed Coscinasterias consumed Asterias at the rate of ~45/year. Field surveys in Port Phillip Bay showed that most Coscinasterias occurred at depths shallower than 15 m, while most Asterias occurred at depths greater than 15 m, and the ratio of Asterias/Coscinasterias only exceeded 45 at depths greater than 15 m. Consequently, if laboratory and field feeding rates are similar, Coscinasterias would be expected to exert significant control over Asterias populations at depths shallower than 15 m, and augmenting Coscinasterias populations at sites of new Asterias infestations may help eliminate newly-established populations.  相似文献

The present study describes the airborne pollen grain concentrations at two different heights (1.5 m and 15 m, respectively). The survey was carried out in 1991 and 1992, using two Burkard spore-traps, both set up at the University of Córdoba, Faculty of Sciences. Generally, and for all herbaceous plants, pollen detection started and ended around the same date on both samplers. However, in the case ofOlea europaea, the pollen was detected in advance by the sampler located at 1.5 m compared with the one located at 15 m, probably due to the fact that olives growing close to the low sampler flower before the great olive plantations located some 60 km south of the city. No significant differences between the counts of both samplers have been observed, except in the case of Urticaceae, where the sampler situated on top of the building recorded higher pollen concentrations in both years. Similar annual peaks of Urticaceae are probably due to the buoyancy of their small, light grains and the explosive pollination mechanism which liberates pollen grains from the anthers of the Urticaceae family, includingUrtica andParietaria.  相似文献

The abundance of many reef fish species varies with depth, but the demographic processes influencing this pattern remain unclear. Furthermore, while the distribution of highly specialized reef fish often closely matches that of their habitat, it is unclear whether changes in distribution patterns over depth are the result of changes in habitat availability or independent depth-related changes in population parameters such as recruitment and mortality. Here, we show that depth-related patterns in the distribution of the coral-associated goby, Paragobiodon xanthosoma, are strongly related to changes in recruitment and performance (growth and survival). Depth-stratified surveys showed that while the coral host, Seriatopora hystrix, extended into deeper water (>20 m), habitat use by P. xanthosoma declined with depth and both adult and juvenile P. xanthosoma were absent below 20 m. Standardization of S. hystrix abundance at three depths (5, 15 and 30 m) demonstrated that recruitment of P. xanthosoma was not determined by the availability of its habitat. Reciprocal transplantation of P. xanthosoma to S. hystrix colonies among three depths (5, 15 and 30 m) then established that individual performance (survival and growth) was lowest in deeper water; mortality was three times higher and growth greatly reduced in individuals transplanted to 30 m. Individuals collected from 15 m also exhibited growth rates 50% lower than fish from shallow depths. These results indicate that the depth distribution of this species is limited not by the availability of its coral habitat, but by demographic costs associated with living in deeper water.  相似文献

A suction sampler designed to minimize sampling error, and quantitatively collect benthic macroinvertebrates from coarse substrata under various flow regimes is described. This sampler uses a 12 V bilge pump (capacity 4400 1 h–1) to suck animals into a collecting net from substrata disturbed by a single operator. Macroinvertebrates were collected more rapidly by this sampler than by a Surber sampler or a Boulton suction sampler. Sources of sampling error are discussed for all three samplers. The electric pump sampler described here is suitable for sampling habitats where depth and velocity vary spatially and with time.  相似文献

Coral reefs are subjected to unprecedented levels of disturbance with population growth and climate change combining to reduce standing coral cover and stocks of reef fishes. Most of the damage is concentrated in shallow waters (<30 m deep) where humans can comfortably operate and where physical disturbances are most disruptive to marine organisms. Yet coral reefs can extend to depths exceeding 100 m, potentially offering refuge from the threats facing shallower reefs. We deployed baited remote underwater stereo-video systems (stereo-BRUVs) at depths of 10–90 m around the southern Mariana Islands to investigate whether fish species targeted by fishing in the shallows may be accruing benefits from being at depth. We show that biomass, abundance and species richness of fishery-targeted species increased from shallow reef areas to a depth of 60 m, whereas at greater depths, a lack of live coral habitat corresponded to lower numbers of fish. The majority of targeted species were found to have distributions that ranged from shallow depths (10 m) to depths of at least 70 m, emphasising that habitat, not depth, is the limiting factor in their vertical distribution. While the gradient of abundance and biomass versus depth was steepest for predatory species, the first species usually targeted by fishing, we also found that fishery-targeted herbivores prevailed in similar biomass and species richness to 60 m. Compared to shallow marine protected areas, there was clearly greater biomass of fishery-targeted species accrued in mesophotic depths. Particularly some species typically harvested by depth-limited fishing methods (e.g., spearfishing), such as the endangered humphead wrasse Cheilinus undulatus, were found in greater abundance on deeper reefs. We conclude that mesophotic depths provide essential fish habitat and refuge for fishery-targeted species, representing crucial zones for fishery management and research into the resilience of disturbed coral reef ecosystems.  相似文献
