Deficiency in n-3 fatty acids has been accomplished through the use of an artificial rearing method in which ICR mouse pups were hand fed a deficient diet starting from the 2nd day of life. There was a 51% loss of total brain DHA in mice with an n-3 fatty acid-deficient diet relative to those with a diet sufficient in n-3 fatty acids. n-3 fatty acid adequate and deficient mice did not differ in terms of locomotor activity in the open field test or in anxiety-related behavior in the elevated plus maze. The n-3 fatty acid-deficient mice demonstrated impaired learning in the reference-memory version of the Barnes circular maze as they spent more time and made more errors in search of an escape tunnel. No difference in performance between all dietary groups in the cued and working memory version of the Barnes maze was observed. This indicated that motivational, motor and sensory factors did not contribute to the reference memory impairment. 相似文献

Adult male Wistar rats were trained to find an escape box in the Barnes maze in order to characterize the extinction process of a learned spatial preference. To do so, once they had fully acquired the spatial task, they were repeatedly exposed to the maze without the escape box. Multiple behavioral measurements (grouped into motor skill and spatial preference indicators) were followed up throughout the complete training process. Animals gained efficiency in finding the escape box during acquisition, as indicated by the reduction in the time spent escaping from the maze, the number of errors, the length of the traveled path, and by the increase in exploration accuracy and execution speed. When their retention and preference were tested 24 h later, all the subjects retained their enhanced performance efficiency and accuracy and displayed a clear-cut preference for the escape hole and its adjacent holes. Almost all motor skill indicators followed an inverse, though not monotonic, pattern during the extinction training, returning to basal levels after three trials without escape box, displaying a transient relapse during the fifth extinction trial. Preference indicators also followed a reverse pattern; however, it took seven trials for them to return to basal levels, relapsing during the eighth extinction trial. The abbreviated Barnes maze acquisition, evaluation, and extinction procedures described herein are useful tools for evaluating the effects of behavioral and/or pharmacological treatment on different stages of spatial memory, and could also be used for studying the neurophysiological and neurobiological underpinnings of this kind of memory. 相似文献

The authors examined spatial working memory in the Morris water maze during the activity and rest periods of Wistar rats. Wheel-running activity was measured continuously as a marker of circadian phase. To minimize possible masking effects on performance, animals were placed in constant dim light the day before testing and tested in similar light conditions. Three experiments were run, each of them using animals varying in their previous experience in the water maze. Half of the animals of each experiment were tested 2 to 3 h after activity onset (active group), and the other half were tested 14 to 15 h after activity onset (inactive group). In the three experiments, a significant phase effect was observed in the animals' performance in the water maze; animals tested in the active phase showed steeper acquisition curves. These phase effects on performance are due to the animals' search pattern and not to a better acquisition and maintenance of spatial information; rats tested in the inactive phase found the platform faster on the first trial of the test, when the information on the location of the platform had not been presented to the animals. This effect vanished as the amount of training in the pool increased. Finally, swimming speed also showed a temporal effect, suggesting the existence of a phase effect for motivation to escape from the water; rats tested during their inactive phase tended to swim faster. All together, the data suggest a modulating effect of the biological clock on performance in the water maze, particularly when the animals are less experienced. 相似文献

The radial arm water maze (RAWM) contains six swim paths (arms) extending out of an open central area, with an escape platform located at the end of one arm (the goal arm). The goal arm location remains constant for a given mouse. On day 1, mice are trained for 15 trials (spaced over 3 h), with trials alternating between visible and hidden platform. On day 2, mice are trained for 15 trials with the hidden platform. Entry into an incorrect arm is scored as an error. The RAWM has the spatial complexity and performance measurement simplicity of the dry radial arm maze combined with the rapid learning and strong motivation observed in the Morris water maze without requiring foot shock or food deprivation as motivating factors. With two sessions each day, 16 mice can be tested over 2 days. 相似文献

Social isolation starting from the 21st day of birth affected neither a short-term nor a long-term memory in male rats at primary acquisition learning in an 8-arm radial maze. A number of the short-term and long-term memory errors were substantially decreased during primary learning but the difference between groups was not significant. Isolates were faster to start a search in an individual trial and took less time to finish offa trial. During the reversal learning, when baited and non-baited arms were reversed, the isolates outperformed of socially reared rats on working but not reference memory task. In overall they made twice less working memory errors than socially reared animals. During the reversal learning the isolates were also faster than non-isolates in initiation and completion of a trial. Maternal separation of rat's pups on the postnatal days 1-21 for 4 hr per day did not affect either working or reference memory on both primary and reversal learning. The data obtained are discussed on basis of influence of stress in early postnatal life on hypothalamo-pituitary axis and its effects on behavior of adult animals. 相似文献

Effects of estrogens on learning of rats with chronic brain cholinergic deficit in Morris water maze

A chronic deprivation of brain cholinergic functions in rats caused by intracerebroventricular injection of neurotoxin AF64A increases the escape latency in Morris water maze test as compared to control sham-operated animals. Measurements and analysis of rat movement tracks using an original computerized "Behavioral Vision" system revealed the ability of 17 beta-Estradiol and its synthetic isomer J-861 (both administered daily in per os dose 0.2 mg/kg during 7 days before and 10 days after a single intracerebroventricular injection of AF64A) to improve learning of the animals. Directivity of search trajectories was estimated by a novel index of track straightness. The introduction of an index of "passive swimming" made it possible to reveal episodes of immobility in water-maze behavior of AF64A-injected animals. Unlike J-861, 17 beta-Estradiol almost completely eliminated these episodes. The newly developed indices (especially straightness) seem to be very useful in differentiating learning ability of rats from a decrease in their mobility in the Morris water-maze test, in particular, in case of the estrogens under study. 相似文献

Equal numbers of male and female rats of the King-Holtzman hybrid breed, divided into two age groups (group A1: 21–30 days old and group A2: 90–100 days old), were exposed to two levels of noise: N0 = 30 db and N1 = 90 db, (ref. 0.002µbar), and three levels of direct electric field intensities: E0 = electric field strength of zero; E1 = 1,600 v/m; and E2 = 16,000 v/m. Time and error scores of 240 rats running a modified Lashley left-right maze with an escape-from-water motive served as criteria. A randomized complete blocks design with replications (2 × 3 × 2 × 2 × 10) was selected for treatment by analysis of variance. The results indicate that (a) adult rats pre-exposed to electric fields showed significantly lower error scores than young rats, regardless of sex or the noise condition; (b) after exposure to electric fields all rats swam faster than their controls; (c) after exposure to noise females showed significantly higher time scores than males under the same conditions, regardless of age; and (d) when subjected to the combined effect of electric fields and noise males showed significantly lower time scores than females under the same conditions, regardless of age.
Zusammenfassung Zwei Altersgruppen (21–30 und 90–100 Tage) männlicher and weiblicher Ratten des King-Holtzman-Stammes wurden 2 Lärmintensitäten (30 db und 90 db) und 3 elektrischen Feldstärken (0, 1.600 v/m und 16.000 v/m) ausgesetzt und die Lernfähigkeit beim Schwimmen im modifizierten Lashley linksrechts Maze geprüft. Das Kriterium waren die Zeit und die Anzahl Fehler bei dem Versuch aus dem Wasser zu entkommen. Auswertung der Messungen durch Varianzanalyse unt er Nachbildung kompletter Blockentwürfe. Die Ergebnisse zeigen: (a) Erwachsene Ratten, die vorher elektrischen feldern ausgesetzt wurden, wiesen signifikant weniger fehler auf als junge Ratten, unabhängig vom Geschlecht und Lärmbedingungen; (b) nach der Exponierung in elektrischen Feldern schwammen alle Ratten schneller als die unbehandelten Kontrollen; (c) nach Lärmeexponierung zeigten weibliche Ratten signifikant höhere Trefferzeiten als Männchen unter den gleichen Bedingungen ohne Beziehung zum Alter und (d) bei gleichzeitiger Anwendung von elektrischen Feldern und Lärm wiesen die Männchen niedrigere Trefferzeiten auf als die Weibchen. Alter spielte dabei keine Rolle.
Resume Des rats de la race King-Holzman, mâles et femelles, de deux groupes d'âge (21 à 30 et 90 à 100 jours) ont été exposes à deux intensités accoustiques (30 et 90 db) et à 3 champs électriques (0, 1.600 et 16. 000 v/m). On a alors examiné leur capacité d'apprendre à nager dans un labyrinthe droite-gauche de Lashley modifié. Les critères d'examen furent le temps et le nombre de fautes commises pour sortir de l'eau. Les observations furent ensuite traitées par l'analyse de variances en tenant compte de répétitions (2 × 3 × 2 × 2 × 10). Les résultats montrent: (a) Les rats adultes préalablement exposés à des champs électriques on fait moins fautes que de jeunes individus et cela de façon significative quels que soient le sexe ou les conditions de bruit; (b) après avoir été exposés à des champs électriques, tous les rats ont nagé plus vite que les bêtes de contrôle; (c) après exposition au bruit, les femelles ont présenté des temps de sortie significativement plus élevés que les mâles pris dans des conditions identiques et cela quel qu'en soit l'âge et (d) après exposition simultanée au bruit et aux champs électriques, les mâles se sont trouvés moins habiles que les femelles, sans incidence de l'âge.相似文献

Zusammenfassung Eine gleiche Anzahl m?nnlicher und weiblicher Ratten (n = 120) des King-Holtzman Stammes, unterteilt in 2 Altersgruppen (21–30 und 90–100 Tage) wurden zwei Ger?uschintensit?ten (30 db und 90 db) und 3 Konzentrationen negativer Luftionen I0: nicht messbar niedrig, I1:7×106, I2:7×107 Ionen/cm3 ausgesetzt. Die Wirkung wurde geprüft an der Schwimmzeit und der Anzahl Fehler der Tiere im Lasley-Links-Rechts-Labyrinth. Das Motiv zum Schwimmen war, aus dem Wasser zu entkommen. Die Auswertung durch Varianzanalyse erfolgte unter Nachbildung randomisierter kompletter Blockentwürfe. Die Ergebnisse zeigen, dass (a) die M?nnchen bei negativ ionisierter Luft signifikant weniger Fehler machten und (b) die Weibchen unter allen Bedingungen schneller schwammen als die M?nnchen, ohne dass das L?rmniveau oder die Luftionisation darauf einen Einfluss hatten.
Resume Un nombre égal de rats males et femelles (120 de chaque) de la race King-Holtzman futdivisé en deux groupes d'age (21 à 30 jours et 90 à 100 jours). Ils furent exposés à 2 intensités auditives (30 et 90 db) et à 3 concentrations d'ions négatifs: I0 non mesurables (faible), I1 : 7 × 106 et I2: 7 × 107 ions/cm3. Les effets d'un tel traitement furent examinés au moyen de deux critères: le temps pris pour sortir de l'eau et le nombre de fautes commises dans un labyrinthe droitegauche de Laslay. Pour l'analyse des variances, on a adopté une répartition due au hasard avec répétitions (2×3×2×2×10). On peut en conclure que: (a) les males font moins d'erreurs dans l'air ionisé négativement et cela de fa?on significative et (b) les femelles ont nagé plus vite que les males dans toutes les conditions de l'essai. Ni le bruit ni le degré d'ionisation ne semblent avoir d'influence sur leur comportement.

目的将奖励性操作式条件反射方法应用于学习记忆研究。方法首次采用奖励性操作式条件反射方法,设置单次操作训练、连续多次操作训练、信号辨识与消退四种检测模式,研究Wistar大鼠和SHR大鼠学习记忆能力。结果奖赏训练中,SHR组鼻触次数显著增多(vs.Wistar&Donepezil,P〈0.05);单次操作训练中,三组动物对操作反应的获得无显著差异;连续多次操作学习任务中,SHR组错误操作次数增多(vs.Wistar,P〈0.05),奖赏获得次数减少(vs.Donepezil,P〈0.001),反应准确率显著降低(vs.Wistar&Donepezil,P〈0.05);信号辨识任务中SHR组错误反应次数显著增多(vs.Wistar&Donepezil,P〈0.05),而反应准确率降低,组间差异有显著性(vs.Wistar&Donepezil,P〈0.05);消退实验中,三组动物差异无显著性。神经递质检测发现,SHR组大鼠谷氨酸[(1.0639±0.07086)mg/g]、乙酰胆碱[(2.7760±0.2609)μg/g]与五羟色胺[(1.2200±0.1137)μg/g]含量均显著低于Wistar组大鼠(P〈0.05),多奈哌齐组大鼠乙酰胆碱含量显著增加[(3.9344±0.2747)μg/g](vs.Wistar,P〈0.05;vs.SHR,P〈0.001)。结论奖励性操作式条件反射方法可作为一种正性增强条件反射检测新方法应用于大鼠学习记忆研究。 相似文献

Studies of spatial learning and memory of birds in radial maze are reviewed. The radial maze variants (standard, giant and open-field analog) are described; procedural problems of bird testing are discussed. Radial maze task performance of birds is compared with that of laboratory rats as a well-studied standard. Specific features of spatial learning in various taxonomic avian groups (pigeon, tits, corvids, chickens and others) are considered. The results of spatial memory studies in closely-related species with different ecology (food storing behavior, degree of caching specialization) and experiments with migrant birds and homing pigeons are discussed. 相似文献

The effect of sleep deprivation by 'carousel' method on spatial memory consolidation in a Morris water maze was studied in Wistar male rats after one-day learning (in accordance to a protocol by Frick et al., 2000). It was found that after fast 3-hr learning the memory trace retains during 24-hr. Twenty four hour sleep deprivation followed learning impaired consolidation of spatial memory. So the rat model of a one-day learning is suitable for the studying of neurophysiological mechanisms of sleep deprivation effects on spatial memory consolidation. 相似文献

Learning is likely to be costly and thus subject to trade-off with other components of life history. An obvious prediction, therefore, is that investment in learning, and thus learning performance, will vary with individual life history strategy and the reproductive value of the learning outcome. We tested this idea in the context of social dominance in male laboratory mice, using a simple radial maze paradigm to compare the ability of high- and low-ranking male mice to track changing food location. We tested animals in randomly selected pairs before and after establishing aggressive rank relationships to distinguish intrinsic differences in learning ability from those attributable to acquiring high or low rank. There was no difference in learning between later dominants and subordinates prior to establishing rank relationships. After pairing, however, dominants showed a significantly greater percentage of correct responses, with the difference being greatest earlier in a sequence of trials. The percentage of correct responses also increased with the amount of aggression initiated during pairing. The results thus appeared to reflect a state-dependent change in learning associated with the aggressive social relationships formed during pairing. 相似文献

The experiments were carried out on rats of Wistar (W) and Krushinsky-Molodkina (KM) lines, differing in the level of task performance in conditions of multiple choice of food sources in a 12ray radial symmetrical maze. It is shown that in the radial maze, the rats of both lines use a number of behavioural tactics. To determine the influence of changes of environmental spatial characteristics while preserving the main experimental strategy--never revisit the places of reinforcement,--on the peculiarities of tactics manifestations maze modifications were elaborated. Besides the environmental procedure was changed. Under all conditions the rats of W line solved the task. According to the situation, they either preserved former tactics or developed some new ones. The rats of KM line could not solve the task in changed conditions. In most sessions, appearance or absence of given tactics was not adequate to new conditions. The presence of the necessary tactics not always facilitated the task solving. Thus, for the realization of adequate reactions, the presence of necessary tactics is not sufficient: their "tuning" is needed depending on situation. The possibility is supposed of genetic determination of rats ability to some tactics. However, it is emphasized, that tactics are not strongly fixed, as their realization depends on environmental conditions. 相似文献

In the present work, the enzyme L-asparaginase was injected into mice by an intrathecal or by an intraperitoneal route and produced a significant impairment of memory. The effects of nucleotides on learning and memory were studied in normal and basal forebrain-lesioned rats using a Morris water maze test. Chronic oral administration of a nucleotide mixture (500 mg/kg), containing an equal weight of the disodium salts of adenosine 5'-monophosphate, guanosine 5'-monophosphate, inosine 5'-monophosphate, cytidine 5'-monophosphate, and uridine 5'-monophosphate facilitated learning acquisition in normal rats. In basal forebrain-lesioned rats, administration of the nucleotide mixture showed a tendency to improve learning acquisition and memory retrieval. In the biochemical studies, no significant changes were observed in brain choline and acetylcholine levels by treatment with the nucleotide mixture at the doses tested in both normal and basal forebrain-lesioned rats. The nucleotides did not affect the monoaminergic systems in normal rats, but did cause some changes in these systems in basal forebrain-lesioned rats. The present studies indicate that nucleotides ameliorate learning and memory processes in normal rats, but not in basal forebrain-lesioned rats, and they also modulate the activity of the central monoaminergic systems under certain conditions. 相似文献
