The ganglioside pattern of goldfish brain was investigated after adaptation (acclimatization, acclimation) to different temperatures. Adaptation at lower ambient temperature causes a higher proportion of polysialogangliosides to be formed in fish brain.  相似文献

The ability of ectothermic vertebrates to adapt to thermal fluctuations in their environment is mainly based upon adaptive changes within the CNS. These changes are thought to be correlated with functional, metabolism changes in the central nervous system, especially in neuronal membranes. Gangliosides, being highly enriched in synaptic membranes (1)show characteristic perculiarities concerning concentration and molecular composition with regard to their sialylation status (2,3,4). In order to get further information concerning the biological function of gangliosides with respect to thermal adaptation, it was of interest to investigate possible correlation between the nervous ganglioside metabolism of different brain regions after various temperature adaptations.  相似文献

Comparative studies on brain gangliosides of more than 60 vertebrate species show correlations between concentration and the level of evolutionary organization: poikilothermic lower vertebrates (fish, amphibs, reptiles) contain about 110 to 700 μg ganglioside bound NeuAc/g. fresh wt., homeothermic birds and mammals, on the other side, 500 to 1000 μg. The composition of brain gangliosides in poikilotherms is much more complex and variable (more multisialogangliosides) as compared with homeotherms (domination of less polar fractions). There are distinct correlations between brain ganglioside composition and state of thermal adaptation: Fishes being adapted to habitates with extreme temperatures (antarctic icefish — tropic fish) are characterized by quite opposite ganglioside patterns (domination of high versus less polar fractions). During seasonal acclimatization and experimental acclimation of fish to cold or during hibernation and early postnatal development of mammals poly-sialylations of brain gangliosides occur. With regard to this the individual brain structures react differently.

The results are taken for evidence that variations in the composion of synaptic membrane-bound gangliosides may induce long-term alterations in viscosity and permeability of the neuronal membrane by which the neuronal transmission might be kept on a constant level during the process of temperature adaptation.  相似文献

Bacterial artificial chromosome (BAC) libraries are important tools for genomic research. We have constructed seven genomic BAC libraries from three fish species, rainbow trout (Oncorhynchus mykiss), carp (Cyprinus carpio) and tilapia (Oreochromis niloticus). The two rainbow trout BAC libraries have average insert sizes of 58 and 110 kb. The average size of inserts in the carp BAC library is 160 kb. The average insert sizes of the four tilapia BAC libraries are 65, 105, 145 and 194 kb, respectively. These libraries represent good coverage of each genome (2-64 x coverage). The libraries can be screened by conventional colony hybridization and provide a starting point for the construction of high-density filtres or polymerase chain reaction (PCR) screening approaches. These BAC libraries will facilitate the positional cloning of quantitative trait loci (QTLs) for a variety of economically important traits in these species.  相似文献

The core temperature of the rainbow trout Oncorhynchus mykiss (3·5 kg) dropped to 1·0° C during the first 6 h of chilling at 0·5° C, remained stable until 24 h, and dropped significantly to 0·7° C after 39 h. Blood plasma osmolality increased and muscle moisture content decreased gradually with increasing chilling time. After 39 h of chilling, the rainbow trout experienced 40 mosmol l-1 higher blood plasma osmolality and 2·8% less muscle moisture content compared with initial values. In the Atlantic salmon Salmo salar (5·3 kg), core temperature dropped to 1·3° C and blood plasma osmolality increased significantly during the first 6 h of chilling at 0·5° C, but remained relatively stable throughout the rest of the experimental period. After 39 h of chilling, the salmon experienced 20 mosmol l-1 higher blood plasma osmolality and 0·5% less muscle moisture content compared with initial values. In rainbow trout muscle moisture content was inversely related to blood plasma osmolality indicating reduced seawater adaptation with increasing hours of chilling. No such relationship was observed in the Atlantic salmon. Hence, changes in plasma osmolality and muscle moisture in the Atlantic salmon do not indicate osmoregulatory failure since the new levels, once established, were maintained throughout the chilling time.  相似文献

The levels of glycogen and glucose, and the activities of several key enzymes of glycogenolysis, glycolysis and the pentose phosphate shunt were assessed in gills of rainbow trout of two sizes (97.03 ± 3.03 g and 165.69 ± 5.01 g) during gradual transfer to sea water (0, 9, 18 and 28 ppt). The results indicated changes, mainly size-independent, in carbohydrate metabolism of gills during gradual adaptation to sea water. Glucose availability in gills increased as a result of both higher glycogenolysis and greater supply of exogeneous glucose. This glucose was mainly used for increased glycolysis, providing the ATP needed to perform the hypo-osmoregulatory work occurring in gills during adaptation to sea water.  相似文献

To establish the time required for fish to acclimate to experimental conditions, comprising holding within a spatially restrictive environment, rainbow trout were transferred from rearing tanks (1500 1) to smaller volume holding tanks (50 1). The fish were maintained singly, in pairs, in groups of five fish, and in groups of 10 fish, and selected indices of physiological stress were monitored at intervals following the onset of confinement. In all cases transfer and confinement provoked a stress response in the fish, indicated by significantly elevated plasma cortisol levels, and significantly reduced numbers of circulating lymphocytes. Recovery of these parameters to levels similar to unstressed control fish, was most rapid among the fish confined singly, requiring 2 to 4 weeks, although weight gain, indicative of feeding, was apparent within 1 to 2 weeks. Acclimation of the fish confined as pairs was severely confounded by the development of hierarchical relationships between the fish. This resulted in some individuals displaying rapid acclimation and growth, in contrast to other fish within the groups which failed to acclimate, exhibited signs of chronic stress, and ultimately succumbed to bacterial infections. Similar, though less severe, evidence of social stress was observed among the fish confined as groups of five. A decline in the coefficient of variance for weight within the fish confined as groups of 10 suggested an absence of growth inhibition due to social interaction among these fish. The implications of these results for experimental design are discussed.  相似文献

This study presents data on myosin Va localization in the central nervous system of rainbow trout. We demonstrate, via immunoblots and immunocytochemistry, the expression of myosin Va in several neuronal populations of forebrain, midbrain, hindbrain and spinal cord. The neuronal populations that express myosin Va in trout constitute a very diverse group that do not seem to have many specific similarities such as neurotransmitters used, cellular size or length of their processes. The intensity of the immunoreactivity and the number of immunoreactive cells differ from region to region. Although there is a broad distribution of myosin Va, it is not present in all neuronal populations. This result is in agreement with a previous report, which indicated that myosin Va is approximately as abundant as conventional myosin II and kinesin, and it is broadly involved in neuronal motility events such as axoplasmatic transport. Furthermore, this distribution pattern is in accordance with what was shown in rats and mice; it indicates phylogenetic maintenance of the myosin Va main functions.  相似文献

We examined the developmental rate of hybrids between rainbow trout (Salmo gairdneri) and two subspecies of cutthroat trout: westslope cutthroat trout (Salmo clarki lewisi) and Yellowstone cutthroat trout (Salmo clarki bouvieri). These taxa show considerable genetic divergence at 42 structural loci encoding enzymes; the mean Nei's d between the rainbow trout and the two species of cutthroat trout is 0.22. We used four measures of developmental rate: time of hatching and yolk resorption, rate of increase in activity of four enzymes, and time of initial detection of seven isozyme loci. The two cutthroat trout subspecies reached hatching and yolk resorption earlier than rainbow trout. Cutthroat trout had higher relative enzyme activities than rainbow trout from deposition of eye pigment to hatching. There was no difference in the rate of increase in enzyme activity or time of initial expression of these loci between these species. Hybrids showed developmental rates intermediate or similar to that of the parental species using all measures. Our results indicate an absence of regulatory and developmental incompatibility between these taxa.This research was supported by NSF Grants ISP-8011449 and BSR-8300039. Heat shock (25° C) of 10° C-acclimated rainbow trout Oncorhynchus mykiss led to increases in heat shock protein 70 (hsp70) mRNA in blood, brain, heart, liver, red and white muscle, with levels in blood being amongst the highest. Hsp30 mRNA also increased with heat shock in all tissues with the exception of blood. When rainbow trout blood was heat shocked in vitro , both hsp70 and hsp30 mRNA increased significantly. In addition, these in vitro experiments demonstrated that blood from fish acclimated to 17° C water had a lower hsp70 mRNA heat shock induction temperature than did 5° C acclimated fish (20 v. 25° C). The hsp30 mRNA induction temperature (25° C), however, was unaffected by thermal acclimation. While increases in hsp70 mRNA levels in blood may serve as an early indicator of temperature stress in fish, tissue type, thermal history and the particular family of hsp must be considered when evaluating stress by these molecular means.  相似文献

A suite of 26 PCR‐based markers was developed that differentiates rainbow (Oncorhynchus mykiss) and coastal cutthroat trout (O. clarki clarki). The markers also differentiated rainbow from other cutthroat trout subspecies (O. clarki), and several of the markers differentiated between cutthroat trout subspecies. This system has numerous positive attributes, including: nonlethal sampling, high species‐specificity and products that are easily identified and scored using agarose gel electrophoresis. The methodology described for developing the markers can be applied to virtually any system in which numerous markers are desired for identifying or differentiating species or subspecies.  相似文献

To assess the influence of water temperature on silver uptake, rainbow trout Oncorhynchus mykiss ( c . 50 g; held at 13° C) were exposed to 0·1 μM AgNO3 in ion‐poor water for 1 week at 4 and 16° C without previous temperature acclimation. To assess the influence of temperature on elimination of previously accumulated Ag, rainbow trout were exposed to 0·1 μM AgNO3 in ion‐poor water for 1 week at 12° C, then were randomly divided amongst two Ag‐free water containers, differing only in temperature (3 and 16° C), for 2 months. In the uptake study greater accumulation of Ag was seen in the gills, plasma and especially the livers and bile of 'warm' rainbow trout (16° C) compared to 'cold' rainbow trout (4° C), which can be explained by the higher metabolic rates of the warmer fish. In the depuration study there was no net elimination of Ag from the livers and bile but there was biphasic elimination of Ag from the gills and plasma of 'warm' and 'cold' fish, but with few differences between them. This indicated that temperature‐dependent processes were less important in Ag elimination than in Ag uptake. Toxicokinetic modelling of Ag uptake by livers indicated four‐fold greater uptake of Ag by 'warm' rainbow trout compared to 'cold' rainbow trout (one compartment uptake model). Elimination of previously accumulated Ag from the plasma was best fitted by a two compartment rate‐constant based model, with approximately half the plasma Ag load eliminated within 24 h, followed by slower elimination of Ag over 2 months.  相似文献

We describe 12 diagnostic single nucleotide polymorphism (SNP) assays for use in species identification among rainbow and cutthroat trout: five of these loci have alleles unique to rainbow trout (Oncorhynchus mykiss), three unique to westslope cutthroat trout (O. clarkii lewisi) and four unique to Yellowstone cutthroat trout (O. clarkii bouvieri). These diagnostic assays were identified using a total of 489 individuals from 26 populations and five fish hatchery strains.  相似文献

Muscle cellularity at a developmental stage around the time of hatching was examined in rainbow trout which had been reared from the eyed stage at three different temperature regimes (5, 10 and 15° C) and different O2 tensions [70% of air saturation value (ASV) at 5° C, 100% of ASV at all temperatures, and 150% of ASV at 10 and 15° C]. It was found that, as has been shown for other species, there was a difference in muscle fibre numbers and fibre cross-sectional areas between some of the regimes. There was a decrease in fibre number at the intermediate and higher temperature, and a decrease in fibre size at the high temperature. The temperature effects observed were modified by the applied changes in O2 tension. An increased O2 tension at 10° C led to an increase in fibre size whereas a decrease in O2 tension at the low temperature resulted in a decrease in fibre number. The largest total white muscle cross-sectional area was achieved at 10° C under high O2 conditions. Temperature and O2 tension therefore had a clear effect on muscle cellularity and there was a significant interaction between the two parameters.  相似文献

In trout, fast growth stimulated white muscle fibre hypertrophy ( P 0·001) and hyperplasia ( P <0·01) in outer fibres but not in deep fibres. Glycogen was most prevalent in outer fibres ( P <0·01) and in brook trout ( P <0·01) that on average had three to four times larger fibres than rainbow trout.  相似文献

Glucokinase (GK) catalyses the first step of hepatic glycolysis to store the excess dietary glucose. After rainbow trout Oncorhynchus mykiss were fed a single meal with dietary glucose and fructose, there was induction of GK expression, although no specific effect of dietary fructose was observed. Summary The influence of trout serum on the attachment and spreading of isolated trout hepatocytes maintained in primary culture at different temperatures was evaluated. Hepatocytes were obtained from young rainbow trout (Salmo gairdneri) by collagenase dissociation and maintained in modified Leibowitz L15 medium at 10° or 27° C for 24 h in plastic dishes previously coated with type I bovine collagen. In the absence of serum, fewer than 10% of hepatocytes attached and none of them spread on the collagen substrate. Trout serum at concentrations as low as 1.25% in the medium resulted in a pronounced concentration-dependent increase in hepatocyte attachment, as determined by direct counts by phase contrast microscopy, or by percentage of lactate dehydrogenase activity attached to the dishes after washing away unattached cells. Attachment rates were greater at the lower temperature (10° C). Trout serum also substantially increased the proportion of attached hepatocytes that spread as monolayers on the collagen substrate, especially at 10° C. By comparison, fetal bovine serum had little influence on the attachment or spreading of trout hepatocytes. These studies demonstrate a simple inexpensive method for preparing attached monolayer trout hepatocyte cultures. This procedure may be useful in toxicologic or functional studies in which fish hepatocyte attachment is an operational requirement.  相似文献
