Maximising seed longevity is crucial for genetic resource preservation and longevity of orthodox seeds is determined by environmental conditions (water content and temperature). The effect of water content (down to 0.01 g·H2O·g?1) on seed viability was studied at different temperatures for a 5‐year storage period in taxonomically related species. Seeds of seven Brassicaceae species (Brassica repanda, Eruca vesicaria, Malcolmia littorea, Moricandia arvensis, Rorippa nasturtium‐aquaticum, Sinapis alba, Sisymbrium runcinatum) were stored at 48 environments comprising a combination of eight water contents, from 0.21 to 0.01 g·H2O·g?1 DW and six temperatures (45, 35, 20, 5, ?25, ?170 °C). Survival curves were modelled and P50 calculated for those conditions where germination was reduced over the 5‐year assay period. Critical water content for storage of seeds of six species at 45 °C ranged from 0.02 to 0.03 g·H2O·g?1. The effect of extreme desiccation at 45 °C showed variability among species: three species showed damaging effects of drying below the critical water content, while for three species it was neither detrimental nor beneficial to seed longevity. Lipid content could be related to longevity, depending on the storage conditions. A variable seed longevity response to water content among taxonomically related species was found. The relative position of some of the species as long‐ or short‐lived at 45 °C varied depending on the humidity at which storage behaviour was evaluated. Therefore, predictions of survival under desiccated conditions based on results obtained at high humidity might be problematic for some species.  相似文献

Success of ex situ storage of germplasm for trade and conservation essentially depends upon the precision of the protocol employed for the assessment of germination potential. Active oxygen species and antioxidative enzymes during natural ageing (NA) and controlled deterioration (CD) was monitored during the loss of seed vigour and germination potential in neem seeds showing intermediate seed storage behaviour. Higher levels of SOD, CAT and APX were strongly and positively associated with germination and vigour. The loss of CAT and APX activity estimated quantitatively and number of isoenzymes were closely accompanied with the simultaneous increase in the amounts of H2O2 and OH-radical. The decline in germination and vigour was negatively related with the levels of H2O2 and OH-radical and enhanced electrolyte leakage. The amounts of OH-radical were positively correlated with the decline in DNA content and DNA damage. The levels of SOD isoenzymes initially increased as the germination index of seeds declined from 5250 to 762 and 882 under NA and CD conditions, respectively. Increasing activity of SOD in the ageing seeds were associated with the accumulation of H2O2. The role of antioxidative enzymes in maintaining signalling and damaging amounts of AOS as well as revelations of different pathways of ageing during NA and CD in the ageing neem seeds were discussed.  相似文献

Germination and vigour of accelerated aged (AA) and naturally stored onion seeds were examined. Accelerated ageing was conducted at 40 °C and 100 % relative humidity (RH). Non aged seeds were stored for 34 months at 3 or 15 °C and 40, 60 or 90 % RH. To restore seed viability, stored and aged seeds were matriconditioned with Micro-Cel E. A distinct loss of germination was observed after 5 days of accelerated ageing. Naturally stored seeds maintained high viability for 34 months, when stored at 3 °C and 40, 60 and 90 % RH or at 15 °C and 40 %. An increase of RH to 60 and 90 % at 15 °C caused loss of germination and vigour. Matriconditioning improved germination and increased endogenic ethylene release and in vivo ACC oxidase activity of both aged and stored seeds.  相似文献

Seeds with low vigour as a result of ageing showed a large improvementin seed quality following a 2 h soak in water, reflected inan increased rate of germination, higher emergence in soil,and the retention of high germination after 24 h rapid ageingin a controlled deterioration vigour test. Similar, but smallerimprovements were seen after a priming treatment using polyethyleneglycol. Seeds dried to their initial moisture content aftertreatment retained these beneficial effects. The treatmentshad little effect on high vigour seed. These findings suggestthat seed invigoration by hydration treatments results fromthe repair of previously sustained deterioration. The objectives of the present work were to investigate whether loss of sunflower (Helianthus annuus L.) seed viability was affected by the embryo moisture content (MC) during seed pretreatment at 35°C, and was related to changes in energy metabolism and in the antioxidant defence system. Non‐dormant seeds were equilibrated at MC of the embryonic axis ranging from 0.037 to 0.605 g H2O g?1 dry matter (DM) for 1 day at 15°C, and they were then placed at 35°C for various durations up to 14 days before the germination assays at 15°C. As expected, the higher the MC, the faster was seed deterioration. There existed a negative linear relationship between the time taken for germination to drop to 50% (P50) and the embryonic axis MC ranging from 0.108 and 0.438 g H2O g?1 DM. In dry seeds, adenosine triphosphate (ATP), adenosine diphosphate (ADP) and adenosine monophosphate represented 6.3, 14.8 and 70.9% of the adenylate pool, respectively, and the energy charge (EC) was very low (0.14). ATP and ADP levels and EC increased sharply during the first day of equilibrium of seeds at a MC above 0.158 g H2O g?1 DM. Subsequent controlled deterioration at 35°C resulted in a decrease in the adenylate pool, and consequently in ATP level. The higher the energy metabolism during ageing, the lower was seed viability. Loss of seed viability was associated with an accumulation of H2O2, and then of malondialdehyde (MDA) suggesting that lipid peroxidation was not the only cause of seed deterioration. When there was a sublinear relationship between H2O2 content in the embryonic axis and seed viability, MDA accumulation only occurred when 50% of the seed population died within 7 days, i.e. when MC was higher than 0.248 g H2O g?1 DM. Ageing was associated with a decrease in the activity of superoxide dismutase, catalase and glutathione reductase, the main enzymes involved in cell detoxification. The involvement of seed MC, as key factor of ageing is discussed with regards to energy metabolism and the regulation of active oxygen species accumulation.  相似文献

We evaluated the effects of treatments with thermotherapy (80°C for 24 h) and dipping in sodium hypochlorite (NaOCl; 0.8% for 10 min) on the germination percentage, germination rate, vigour index and ToMV inactivation of seeds from three pepino (Solarium muricatum) accessions (96–5, B–2 and OV–8) and two wild relatives (S. tabanoense EC–26 and S. caripense EC–40). Thermotherapy decreased the germination percentage (especially in the wild species), germination rate in wild species but not in cultivated pepino, and vigour index in all cases. Sodium hypochlorite increased germination, except for nonthermotreated seeds of OV–8, improved the germination rate in wild species and the vigour index in all cases except for non thermotreated OV–8. Seed coats were altered by both treatments, especially by NaOCl. Thermotherapy was totally effective in the inactivation of seed-borne ToMV, while NaOCl was not. Thermotherapy combined with NaOCl allows effective seed disinfection of S. muricatum seeds without negatively affecting seed germination. For the wild species, although thermotherapy reduces germination, the higher number of seeds per fruit in these species reduces the magnitude of the problem. Even when no disinfection is necessary, treatments with NaOCl are always advisable as they improve germination.  相似文献

The influence of seed priming and ageing treatments on viabilityand rate of germination of tomato (Lycopersicon esculentum Mill.)seeds was examined under both long-term and controlled-deteriorationstorage conditions. Seeds of a single lot of tomato were eitherprimed or aged to increase or decrease the rate of germination(Argerich and Bradford, 1989). They were then stored at 6% moisturecontent (dry weight basis) at either 4 ?C or 30 ?C for 1 year.Both viability and germination rate were unaffected by eitherstorage temperature in control seeds, or by 4 ?C storage inprimed or aged seeds. At 30 ?C, however, viability and germinationrate of primed and aged seeds was markedly reduced after 6 monthsof storage. The temperature dependence of the germination rateand the spread of germination times within the population wasalso adversely affected by high temperature storage, particularlyfor primed seeds. Under controlled deterioration conditions(13.5% moisture content and 50 ?C), the rate of loss of viabilitywas greater for primed seed than for control or aged seeds.The relationship between seed viability and the mean germinationrate, however, was not influenced by the seed treatments. Thesedata are analysed in relation to current models of seed deteriorationduring storage and seed repair during priming. The results indicatethat enhancement of seed germination rates by priming treatmentssimultaneously lowers the resistance of seeds to deterioration.Primed tomato seeds must, therefore, be considered to be vigorousseeds with a reduced storage life. The deterioration of seeds during prolonged storage results in a reduction of viability and germination rate. DNA damage is one of the major cellular defects associated with seed deterioration. It is provoked by the formation of reactive oxygen species (ROS) even in the quiescent state of the desiccated seed. In contrast to other stages of seed life, DNA repair during storage is hindered through the low seed water content; thereby DNA lesions can accumulate. To allow subsequent seedling development, DNA repair has thus to be initiated immediately upon imbibition. Poly(ADP‐ribose) polymerases (PARPs) are important components in the DNA damage response in humans. Arabidopsis thaliana contains three homologues to the human HsPARP1 protein. Of these three, only AtPARP3 was very highly expressed in seeds. Histochemical GUS staining of embryos and endosperm layers revealed strong promoter activity of AtPARP3 during all steps of germination. This coincided with high ROS activity and indicated a role of the nuclear‐localised AtPARP3 in DNA repair during germination. Accordingly, stored parp3‐1 mutant seeds lacking AtPARP3 expression displayed a delay in germination as compared to Col‐0 wild‐type seeds. A controlled deterioration test showed that the mutant seeds were hypersensitive to unfavourable storage conditions. The results demonstrate that AtPARP3 is an important component of seed storability and viability.  相似文献

The germination requirements and the basis of the optimal water content before and after cryopreservation were studied for ten endangered Brazilian species of Bromeliaceae. Constant and alternating temperature regimes were used to determine the best conditions for seed germination. The relationship between seed water content and relative humidity was evaluated using water sorption isotherms at 15 °C. Seeds were cryostored at four water contents (3, 5, 7 and 9%) and three storage periods (0, 180 and 365 days), and loss in viability and vigour were estimated. Fresh seeds of all species showed maximum germination in < 30 days at temperatures between 20 and 30 °C, indicating the absence of a physical/morphological dormancy. A sigmoidal relationship between seed water content and relative humidity was observed with no apparent differences in sorption characteristics among the species. The optimum water content for cryopreservation of most of these species was c. 7%. Ultra‐drying (3% seed water content) had a detrimental effect on seed viability and vigour. Our experiments suggested orthodox storage behaviour for all species of Bromeliaceae examined as they are able to survive desiccation and freezing. This study has shown the feasibility of ex situ conservation in seed cryobanks of endangered bromeliads from the Brazilian Atlantic Forest to support future reintroduction of these species in nature. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 176 , 567–576.  相似文献

The laboratory germination (criterion radicle emergence) of seven seed lots of winter wheat cv. Slejpner was similar. However, they differed in vigour as demonstrated by differences in germination after controlled deterioration carried out at a range of seed moisture contents, at two temperatures and for different times. A vigour assessment for each lot was quantified by calculating a value for the seed lot constant Ki, of the viability equation. Germination in lower water potentials reduced the uptake of water, radicle and coleoptile emergence and radicle and coleoptile extension. There was no difference in the water uptake of seed lots differing in vigour. However, seed lots of lower vigour showed less radicle emergence, less coleoptile emergence and shorter radicles than higher vigour seed lots in low water potentials. Similarly, controlled deterioration resulted in reduced radicle and coleoptile emergence and growth compared to unaged seed, and also to a greater sensitivity to low water potentials. Accelerated ageing is an accurate test indicator of seed vigor and storability that helps to understand the mechanisms of cellular and biochemical deterioration that occur during seed ageing. This study was carried out to elucidate the mechanisms of ageing in macaw palm embryos. Seeds were artificially aged during 4, 8 and 12 days at 45 °C and 100% relative humidity. After ageing, seeds were tested for viability (tetrazolium), electrical conductivity, lipid peroxidation (MDA) and hydrogen peroxide (H2O2) content. Part of the aged seeds was imbibed for 8 days and then determined the hydrogen peroxide content and the activity of antioxidant system enzymes (superoxide dismutase, catalase and glutathione reductase). Ageing reduced the embryo viability from 8 days of treatment and increased malondialdehyde content (MDA) and solute leakage. Hence, membrane permeability correlated with both loss of viability and lipid peroxidation. Imbibition after ageing significantly increased H2O2 content along with superoxide dismutase activity. Catalase activity was significantly higher than control in embryos aged from 8 days and imbibed, and glutathione reductase activity did not change. Our results suggest that macaw palm seed deterioration during accelerated ageing is closely related to lipid peroxidation, and that enzymatic antioxidant system is not completely efficient in reducing reactive oxygen species after imbibition, a critical phase to germination. Moreover, accelerated ageing test can be used as a reliable model to understand the mechanisms involved in palm seeds deterioration.  相似文献

In this paper, Glycine max L. seeds under accelerated ageing condition (40°C and 100% relative humidity) were used as experimental material to study the relationships between seed viability and cell death, production and scavenging of reactive oxygen species (ROS) during accelerated ageing. Water content of seeds gradually increased, while the final germination percentage, germination rate of seeds and fresh weight of seedlings produced decreased with increasing accelerated ageing time. The accelerated ageing time (T 50) when final seed germination decreased to 50% was about 10.5 days. During the period of accelerated ageing, the viability of root cells was lost gradually as manifested by the increase in staining with Evans blue. The respiration rate of seeds, ·O2 production rate, and H2O2 content of axes increased, peaked at the 10 days of accelerated ageing, and then decreased. Activities of superoxide dismutase, ascorbate peroxidase, catalase, and glutathione reductase of axes decreased; and malondialdehyde contents of axes markedly increased. A sceme to explain relationships between seed vigor, cell death, and production and scavenging of ROS during accelerated ageing was suggested. The allelopathic potential of two cultivars of sunflower (Helianthus annuus L.) Lech and Ogrodowy was studied. Leaf aqueous extracts of sunflower at 2.5, 5, and 10 % (m/v) concentrations were applied to determine their effect on mustard (Sinapis alba) seed germination and seedlings growth under laboratory conditions. Increasing concentration of aqueous extracts of sunflower inhibited seed germination, up to almost complete failure of germination, observed in the presence of 10 % extract from sunflower cv. Ogrodowy. The decrease in germinability was well correlated with increased membrane deterioration, assayed as electrical conductivity and enhanced lipid peroxidation, detected as increased malondialdehyde content.  相似文献

Seeds of onion (Allium cepa) cv. Ailsa Craig were exposed to various doses of gamma radiation (0, 10, 20, 40, 80 and 100 krad) and subjected to accelerated ageing (RH 100%, 42°C) for 12 h. Radiation doses significantly affected the percentage of normal seedlings, abnormality types (%) and seedling growth. Seed viability, conductivities of seed leachates, final germination (%), germination speed and time to reach 50% germination (T 50) were not affected by the seed irradiation. Accelerated ageing after irradiation had significant influence on seed viability, conductivities of seed leachates, final germination (%) and percentage of normal seedlings. Germination speed, T 50 and seedling growth were not affected by the accelerated ageing. However, all the parameters studied were significantly influenced by the interaction of radiation doses and accelerated ageing. Accelerated ageing generally enhanced the damaging effects of irradiation on seeds. Therefore, it was concluded that onion seeds should not be exposed to adverse environmental conditions after irradiation.  相似文献

The non-enzymatic modifications of proteins through Amadori and Maillard reactions play an important role in the loss of seed viability during storage. In the present study, the contribution of sugar hydrolysis and lipid peroxidation to Amadori and Maillard reactions, and to seed deterioration was investigated in mung-bean (Vigna radiata Wilczek). The contents of glucose and lipid peroxidation products in seed axes increased significantly during storage. The accumulation of Amadori products in seed axes was correlated to the lipid peroxidation, whereas the accumulation of Maillard products was closely correlated to sugar hydrolysis. The rate of accumulation of Maillard products was not well correlated to the content of Amadori products in both seed axes and protein/glucose model system, reflecting the complex nature of Amadori and Maillard reactions. The content of Amadori products in seed axes increased during the early stages of seed ageing, whereas the content of Maillard products increased steadily during the entire period of storage. The accumulation of Maillard products in seed axes was associated with the decline of seed vigour. These data suggest that, during seed ageing, sugar hydrolysis and lipid peroxidation are coupled with non-enzymatic protein modification through Amadori and Maillard reactions.  相似文献

  • Changes in seed lipid composition during ageing are associated with seed viability loss in many plant species. However, due to their small seed size, this has not been previously explored in orchids. We characterized and compared the seed viability and fatty acid profiles of five orchid species before and after ageing: one tropical epiphytic orchid from Indonesia (Dendrobium strebloceras), and four temperate species from New Zealand, D. cunninghamii (epiphytic), and Gastrodia cunninghamii, Pterostylis banksii and Thelymitra nervosa (terrestrial).
  • Seeds were aged under controlled laboratory conditions (3-month storage at 60% RH and 20 °C). Seed viability was tested before and after ageing using tetrazolium chloride staining. Fatty acid methyl esters from fresh and aged seeds were extracted through trans-esterification, and then analysed using gas chromatography–mass spectrometry.
  • All species had high initial viability (>80%) and experienced significant viability loss after ageing. The saturated, polyunsaturated, monounsaturated and total fatty acid content decreased with ageing in all species, but this reduction was only significant for D. strebloceras, D. cunninghamii and G. cunninghamii.
  • Our results suggest that fatty acid degradation is a typical response to ageing in orchids, albeit with species variation in magnitude, but the link between fatty acid degradation and viability was not elucidated. Pterostylis banksii exemplified this variation; it showed marked viability loss despite not having a significant reduction in its fatty acid content after ageing. More research is required to identify the effect of ageing on fatty acid composition in orchids, and its contribution to seed viability loss.

However, rRNA in immature seed is more vulnerableto damage by ageing than rRNA in mature seed; thus, amountsof rRNA per seed in aged seed are indicators of vigour and viability.On imbibition of aged or unaged seed, more rRNA fragmentationoccurs in immature seed than in mature seed, and the time atwhich the effects of synthesis outweigh the effects of degradationis earlier during germination in mature than in immature seed.
