Using electrophoretic analysis of 11 enzyme systems, we studied the genetic structure and differentiation of eight natural populations of silver fir Abies alba Mill. in the Ukrainian Carpathians. Of 24 isozyme loci identified, 66.8% proved to be polymorphic. The mean numbers of alleles and genotypes per locus in the populations were respectively 3.1 and 4.5. Each A. alba tree was on average heterozygous at 15.9% of genes. In six populations, the genotypic distribution for all of the loci examined corresponded to Hardy-Weinberg proportions. The populations studied had low levels of subdivision (F ST = 0.018; G ST = 0.019 ) and differentiation. Allozyme spectra of peroxidase, esterase, superoxid dismutase, tyrosinase, alcohol dehydrogenase, lactate dehydrogenase, and acid phosphatase were examined in populations of sexual (Taraxacum serotinum and Pilosella echioides) and apomictic (T. officinale and P. officinarum) plant species. The heterozygosity in these populations (0.455–0.620) proved to be considerably higher than the average level characteristic of plant populations (0.058–0.185). The populations examined did not differ in the mean phenotype number , i.e., they exhibited the same diversity (3.188–3.380). The proportion of rare phenotypes h also did not differ between the sexual and apomictic species of the same genus, whereas this parameter in the Pilosella populations (0.150–0.174) was significantly higher than in the Taraxacum ones (0.093–0.114). The populations were characterized by numerous isozyme spectra (more than 11 per populations) and displayed multiple allelism (the mean allele frequency was 3.63–4.38 per locus). They exhibited a high percentage of rare (occurring at a frequency lower than 5%) spectra (35–80%). The karyological data on populations of Siberian fir (Abies sibirica Ledeb.) growing in lowland swamp and dry valleys are given. The diploid set of both populations included 24 chromosomes (2n = 24). Polykaryogram analysis revealed seven pairs of metacentric and five pairs of submetacentric chromosomes. The revealed differences between the populations included the absolute length of chromosomes, number of nucleolar organizer regions, and number of nucleoli. Changed chromosome numbers (mixoploidy and aneuploidy) as well as chromosomal aberrations were recorded. For the first time, mitosis was studied in this species and anaphase/telophase aberrations were revealed. A comparative molecular genetic study of cultured Saccharomyces strains isolated from the surface of berries and various fermentation processes showed that bakers yeast and black-currant isolates contain not only Saccharomyces cerevisiae but also S. cerevisiae × S. bayanus var. uvarum hybrids. The molecular karyotyping of bakers, brewers, and wine yeasts showed their polyploidy. The restriction enzyme analysis of noncoding rDNA regions (5.8S-ITS and IGS2) makes it possible to differentiate species of the genus Saccharomyces and to identify interspecies hybrids. The role of glutathione and other antioxidant systems in the response of Escherichia coli to acetamidophenol (paracetamol), rifampicin, and chloramphenicol was studied. The exposure of aerobically growing E. coli cells to acetamidophenol diminished the intracellular level of glutathione by 40% and the reduced-to-oxidized glutathione ratio in the cells by 50%, while it enhanced the expression of the antioxidant genes soxS and sodA by 2.7 and 1.8 times, respectively. Glutathione-deficient cells were more susceptible to acetamidophenol than were normal cells. All this suggests that acetamidophenol induces a mild oxidative stress in E. coli cells. The fatty acid (FA) composition of lipids of two geographic populations of larch (Larix gmelinii (Rupr.) Rupr.) was determined. One of these populations was grown on soil with a naturally high content of fluorides. This population exceeded the control one in the content of unsaturated fatty acids (FAs) in the lipid fraction of its calli. These FAs were characterized by the presence of 5-unsaturated FAs with irregular positions of double bonds, such as taxoleic, pinolenic, and sciadonic acids. The lysin content in maize endosperm protein is considered to be one of the most important traits for determining the nutritional quality of food and feed. Improving the protein quality of the maize kernel depends principally on finding a mutant with a higher lysine content. Two high-lysine mutant lines with opaque endosperm, QCL3024 and QCL3021, were isolated from a self-cross population derived from Robertsons Mutator stocks. The gene controlling this mutation is temporarily termed opaque-16 (o16). In order to illuminate the genetic locus and effect of the o16 gene, two F2:3 populations, one developed from a cross between QCL3024 and QCL3010 (a wild type line) and another from a cross between Qi205 (opaque-2 line) and QCL3021, were created, and F3 seeds from the F2 plants in the two populations were evaluated for lysine content. The distributions of lysine content and tests for their normality indicate that the lysine content in the two populations is regulated by the major gene of o16 and genes of o2 and o16, respectively. Based on two data sets of the linkage maps of the F2 plant marker genotypes and the lysine content of F3 seeds originating from the two F2:3 populations, the o16 gene was located within 5 cM, at either 3 or 2.2 cM from umc1141 in the interval between umc1121 and umc1141 on the long arm of chromosome 8, depending on the recombination rate in the two populations as determined by composite interval mapping. According to the data of the F2:3 population constructed from the o2 and o16 lines, the double recessive mutant effect was analyzed. The average lysine content of the F3 o2o2o16o16 families identified by the umc1066 and umc1141 markers was approximately 30% higher than that of the F3 o2o2 and o16o16 families, respectively. The lysine content of seven F3 families among nine F3 double recessive mutant families showed different increments, with an average increase of some 6% compared with that of the maternal o2 line. The potential application of the o16 mutant for maize high-lysine breeding may be to combine it with the o2 mutant bearing modifier genes, thus obtaining a mutant with much higher lysine content. For the purpose of pyramiding the o16 with o2 genes, the availability of closely linked markers of the o16 and o2 loci will facilitate marker-assisted selection and greatly reduce breeding time and effort.  相似文献

Heterozygous tandem duplications that appear in Escherichia coli conjugation matings segregate different types of haploid and diploid recombinants formed by unequal crossing over between sister chromosomes. As shown previously, the frequency of segregants in the extended duplication D104 (150 kb or more than 3 min of the genetic map) heterozygous for E. coli deo-operon genes (deoA deoB::Tn5/deoC deoD) is not decreased in strains with defective RecBCD and RecF recombination pathways. Analysis of a shorter duplication of this type (46 kb) showed that the frequency of segregants in the strain recBC sbcBC recF was similar to that in a strain with undamaged system of recombination. Megacodon stylophorus (Clarke) Smith is a perennial alpine herb endemic to the species-rich eastern Himalayan region. Its populations are locally scattered as isolated patches throughout this region. Genetic variation within and among six populations of this species was assessed using ISSR fingerprinting with 13 primers. High levels of genetic diversity exist within species (P = 69.83%, HT = 0.1949 and Hsp = 0.3047), while the within-population diversity is low (P = 11.21%, HE = 0.0532 and Hpop = 0.0792). Extraordinarily high levels of genetic differentiation were detected among populations based on various statistics, including Neis genetic diversity analysis (72.7%), Shannons diversity index (74.01%) and AMOVA (80.70%). That is, populations shared low levels of genetic identity (I = 0.8203 ± 0.0430). This genetic structure was probably due to severe genetic drift of the small-sized patchy populations resulting from postglacial habitat fragmentations. The observed genetic structure of the populations implies that as many populations as possible should be considered for any in situ and ex situ conservation practice on this species.  相似文献

We report the first systematic survey for the presence of Wolbachia endosymbionts in aphids and whiteflies, particularly different populations and biotypes of Bemisia tabaci. Additional agriculturally important species included were predator species, leafhoppers, and lepidopterans. We used a polymerase chain reaction (PCR)-based detection assay with ribosomal 16S rDNA and Wolbachia cell surface protein (wsp) gene primers. Wolbachia were detected in a number of whitefly populations and species, whitefly predators, and one leafhopper species; however, none of the aphid species tested were found infected. Single, double, and triple infections were detected in some of the B. tabaci populations. PCR and phylogenetic analysis of wsp gene sequences indicated that all Wolbachia strains found belong to group B. Topologies of the optimal tree derived by maximum likelihood (ML) and a ML tree in which Wolbachia sequences from B. tabaci are constrained to be monophyletic are significantly different. Our results indicate that there have been at least four independent Wolbachia infection events in B. tabaci. The importance of the presence of Wolbachia infections in B. tabaci is discussed. Galeuchet D. J. and Holderegger R. 2005. Conservation and re-introduction of Dwarf Bulrush (Typha minima) – vegetation surveys, monitoring and genetic analysis of origin. Bot. Helv. 115: 15–32.Typha minima was formerly widespread along fast flowing alpine rivers but is now red-listed as critically endangered. To assess its conservation perspectives, we surveyed the few remaining natural populations along the alpine part of the River Rhine from 1997 to 2002 and determined their genetic diversity using isozyme electrophoresis. Six of the populations became extinct or extremely small, probably due to shading by taller plants and trampling, while six other populations remained stable or increased, partly due to habitat restoration measures. The largest populations, with areas of more than 10000 m2, are found in secondary habitats which are regularly disturbed due to water regime management. Of the 19 investigated isozyme loci, only six were polymorphic. Allelic diversity (1.4–1.8) was low in all populations, and the number of multilocus genotpyes (1–18) was low for 11 of 13 investigated populations. Genetic diversity was also studied for ex-situ cultivations of T. minima in Swiss botanical gardens and reintroduced stands.These artificial populations (each with 1–3 multilocus genotypes) were genetically similar to natural populations (average genetic distance 0.094). For two ex-situ cultivations with unknown origin, the likely origin could genetically be defined. Hybridisation between two ex-situ cultivations of different origin (i.e. a potential risk of genetic introgression) was detected in one botanical garden. It is concluded that the long-term conservation of T. minima requires both the restoration of regularly disturbed, sparsely vegetated river margins and the re-introduction of plants from ex-situ cultivations with appropriate origin.Manuskript angenommen am 2. Summary Genetic parameters of 11 weedy rye populations located in California's northern mountain area and the adjoining Oregon border were compared with those of the putative parents, wild species Secale montamim and cultivated rye S. cereale. All weedy populations exhibited high levels of genetic variation as determined by isozyme analysis. On average, 44% of the isozyme loci were polymorphic, total genetic diversity was 0.30; and number of alleles per locus was 1.65. High genetic identities, averaging 0.994 ± 0.005 between populations, indicated that little genetic differentiation has occurred among these weedy populations since the initial colonization. Lack of population differentiation could be attributed to a wind-pollinated, self-incompatible breeding system resulting in extensive gene flow among weedy populations, and between weedy populations and local cultivars of rye. Multilocus outcrossing rates of weedy populations ranged from 0.86 to 0.97. The estimated levels of gene flow using the private-alleles method were high among weedy populations, and between cv Merced and weedy populations, with estimated Nm values of 14.50 and 8.21, respectively. The colonizing success of weedy rye is discussed and a strategy for its conservation recommended.  相似文献

Experiments were performed to optimize the macronutrients concentrations for in vitro rooting of Ceratonia siliqua micropropagated shoots. Several dilutions of Murashige and Skoog (MS) medium were tested: full-strength MS, half-strength MS ( MS), and MS + full N. The frequency of in vitro rooting was enhanced when the MS was used (50 % rooted shoots). Mature leaves from 20 – 30 year-old carob trees and from 2 year-old micropropagated plants were collected and the concentrations of macronutrients (N, P, K, Ca, Mg) assessed. Based on the mineral composition of the leaves a new medium was formulated and compared with the previous ones showing an increment of the rooting frequency to 80 %. Moreover, shoots rooted in the new medium did not show symptoms of apical necrosis that occurred in the other tested media.  相似文献

Background

The genetics of osteogenesis imperfecta (OI) have not been studied in a Vietnamese population before. We performed mutational analysis of the COL1A1 and COL1A2 genes in 91 unrelated OI patients of Vietnamese origin. We then systematically characterized the mutation profiles of these two genes which are most commonly related to OI.

Methods

Genomic DNA was extracted from EDTA-preserved blood according to standard high-salt extraction methods. Sequence analysis and pathogenic variant identification was performed with Mutation Surveyor DNA variant analysis software. Prediction of the pathogenicity of mutations was conducted using Alamut Visual software. The presence of variants was checked against Dalgleish’s osteogenesis imperfecta mutation database.

Results

The sample consisted of 91 unrelated osteogenesis imperfecta patients. We identified 54 patients with COL1A1/2 pathogenic variants; 33 with COL1A1 and 21 with COL1A2. Two patients had multiple pathogenic variants. Seventeen novel COL1A1 and 10 novel COL1A2 variants were identified. The majority of identified COL1A1/2 pathogenic variants occurred in a glycine substitution (36/56, 64.3 %), usually serine (23/36, 63.9 %). We found two pathogenic variants of the COL1A1 gene c.2461G?>?A (p.Gly821Ser) in four unrelated patients and one, c.2005G?>?A (p.Ala669Thr), in two unrelated patients.

Conclusion

Our data showed a lower number of collagen OI pathogenic variants in Vietnamese patients compared to reported rates for Asian populations. The OI mutational profile of the Vietnamese population is unique and related to the presence of a high number of recessive mutations in non-collagenous OI genes. Further analysis of OI patients negative for collagen mutations, is required.

Little is known about the occurrence and linkage between secreted insecticidal virulence factors in natural populations of Bacillus thuringiensis (Bt). We carried out a survey of 392 Bt strains isolated from various samples originating from 31 countries. The toxicity profile of the culture supernatants of these strains was determined individually against Anthonomus grandis (Coleoptera) and Spodoptera littoralis (Lepidoptera). We analyzed -exotoxin I production and searched for the genes encoding Vip1–2, Vip3, and Cry1I toxins in 125 of these strains. Our results showed that these insecticidal toxins were widespread in Bt but that their distribution was nonrandom, with significant linkage observed between vip3 and cry1I and between vip1–2 and -exotoxin I. Strains producing significant amounts of -exotoxin I were more frequently isolated from invertebrate samples than from dust, water, soil, or plant samples.  相似文献

The Atlas Rat cDNA Expression Array (BD Biosciences, United States) has been used to analyze changes in the expression of 588 genes in rat brain cells in response to a single administration of Ladasten, a 2-aminoadamantane derivative that has psychostimulating and anxiolytic effects. The analysis of hybridization on macroarrays, confirmed by the results of real-time quantitative RT-PCR, has demonstrated that Ladasten alters the expression of 12 genes in the rat brain. The GAT3 and CARBH genes are presumed to be pharmacologically important targets of Ladasten. The changes in their activity explain the mechanisms of the anxiolytic and mood-stabilizing effects of the drug. Ladasten has been shown to induce the genes whose products are involved in various signal pathways (APC, Rb, PKCIP, and PMCA), as well as the genes of cytoskeletal proteins (Tub1 and actin), synaptic proteins (SynIA&IB and PLP), and enzymes (Gapdh and NSE). Overall, 30 strains of hyperthermophilic archaea, representing seven species of the genera Thermococcus, Desulfurococcus, Thermoproteus, and Acidilobus, were tested for the presence of thermostable DNA polymerases. Thermostabilities of the polymerases varied distinctly among the strains within one species. Polymerases of five strains retained 60–100% activity upon incubation of the preparations at 95°C for 120 min. A new DNA polymerase was isolated from the strain Thermococcus litoralis Sh1AM, possessing the enzyme with the most promising properties, and characterized. Molecular weight of the enzyme is 90–100% kDa. The purified DNA polymerase preserved 50% of the initial activity upon incubation at 95°C for 120 min. The polymerase isolated displayed an associated 3–5 exonuclease activity. The error rate when extending a DNA strand was at least twofold lower compared with Taq polymerase. Two polymorphic sites were found in the human c-fms gene: one (G A) was in position 34047 in the last intron, and the other (dinucleotide TC CA) was in positions 34 293 and 34 294 in the 3"-untranslated gene region, 34 bp downstream of the translation stop codon. The polymorphic dinucleotide appeared to be immediately upstream of an octamer showing 100% homology to cis element –CAAACTTC–, which is responsible for controlled instability of mRNAs of several genes. Based on these data, functional significance was assumed for this polymorphism of the c-fms gene. Allele frequencies were established for several populations. The mutant allele of the polymorphism located in the intron were detected only in one family of ethnic Germans from the Altaiskii krai. Polymorphism of the second site, which is in the 3"-untranslated region of the c-fms gene, was observed in all Caucasoid and Mongoloid populations examined. Frequency of the rare allele varied from 19.7–25% in Arctic Mongoloids to 31–42.6% in Central Asian Mongoloids and was similar in two Caucasoid populations (22.6% in ethnic Russians and 26.5% in ethnic Germans). The wide distribution of the mutant allele in human populations of the two races was considered indicative of an adaptive role of the polymorphism in providing a certain level of the gene product, a receptor, in certain cell processes.  相似文献

Mutants capable of a high frequency of cytoduction (Hfc+) were obtained in a haploid strain of Saccharomyces cerevisiae, suggesting impaired cytogamy. Nine of the 68 Hfc+ mutants showed the antisuppressor effect with respect to mutations of the SUP35 and SUP45 genes, which code for translation termination factors, or to the [PSI +] factor, which is the prion form of Sup35. Cosegregation of the characters higher frequency of cytoduction and antisuppression was demonstrated for three Hfc+ mutants. One (HFC12-2) of the Hfc+ mutations exerted a dominant antisuppressor effect with respect to [PSI +] and had no effect on [PSI +] maintenance. Enzyme preparations were isolated from the culture liquid of five mutant strains of the cellulase producer Penicillium verruculosum. The hydrolytic activities of these preparations against unbleached eucalypt cellulose was compared to that of commercial preparations of Trichoderma reesei (T. longibrachiatum). In the majority of cases, P. verruculosum enzymes provided higher yields of reducing sugars (RSs) and glucose.
