When Trichophyton mentagrophytes colonies were placed on a medium containing 150 micrograms/ml of Congo red, a dye which prevents chitin fibrillogenesis, their growth rate was reduced. The newly formed mycelium, examined under an ultraviolet microscope, consisted of thick, curled and branched hyphae endowed with swollen tips and subapical bulges. Short-time exposure revealed that the major sites of dye accumulation were the extension zones. Normally structured hyphae arose from aberrant mycelia when they were transferred onto a dye-free medium. The phenomena observed suggest that Congo red alters the wall properties of the extension zone, by inhibiting the gradual conversion of chitin chains, synthesized at the extreme tip, into microfibrils of increasing size and density.  相似文献

Meiotic prophase in Sordaria humana has been analyzed by three-dimensional reconstructions of 3 leptotene, 2 zygotene, 10 pachytene and 3 diplotene nuclei. Several notable features emerged. The lateral components of the synaptonemal complexes (SC) are hollow tubes which show dilations of variable sizes from late leptotene to early diplotene. These bulges occur before pairing. Their number decreases as soon as the SC are completely formed, but their mean size increases. Bulges can be present in all parts of the lateral components including telomeres and nucleolar organizer region, but their distribution along bivalents is not random. The remarkably uniform width of the SC central region, normally observed in other species is not observed in S. humana. Although as a general rule the bulges rarely affect the homologous components at identical sites, they often either fill or partially cover the central region. The recombination nodules are not clearly connected with the bulges. This work provides also additional insight into the development of both SC and the nodules. At late leptotene, the homologues are aligned before SC formation. One case of interlocking has been observed at early pachytene. Nodules are present from zygotene to diplotene. They are not evenly distributed along the bivalents during pachytene. The mean number of nodules, constant from late pachytene to diplotene, is equal to the mean number of chiasmata.  相似文献

近年来,尽管西瓜产业不断发展壮大,但轮作土壤种植西瓜易产生枯萎病害导致世界范围内的西瓜严重减产。通过对峙培养实验和抑菌实验探讨了重阳木内生真菌司氏角担子菌(Ceratobasidum stevensii)B6菌株对西瓜枯萎病病原菌尖孢镰刀菌西瓜专化型(Fusarium oxysporum f. sp.Niveum,FON)的拮抗作用,并初步分析了其作用机制。平板拮抗试验的结果表明,内生真菌B6生长过程中不是通过产生抑菌带来抑制FON菌丝,而是利用自身的生长优势将FON完全覆盖。显微观察B6与FON菌丝的接触部位,发现FON菌丝外侧附着B6顶端菌丝形成的胞样结构,表明FON菌丝生长仅受到B6菌丝的抑制。抑菌试验结果显示,B6产生的挥发性物质可以抑制FON的生长和产孢,并使其菌丝分枝明显减少;B6的发酵液对FON的生长和产孢没有抑制作用。因此,推测B6主要通过释放某些挥发性物质产生拮抗作用而抑制FON的生长。  相似文献

Zalokar , Marko . (Yale U., New Haven, Conn.) Enzyme activity and cell differentiation in Neurospora. Amer. Jour. Bot, 46(7): 555–559. Illus. 1959.—Morphological differences were observed in vegetative cells of Neurospora of different ages and in different parts of the mycelium. The surface layer of mycelium grown in standing cultures could be separated from the deep layer. The first contained most of the growing hyphae rich in protoplasm, while the second contained heavily vacuolated hyphae laden with fat droplets. Specific activities of several enzymes were studied in conidia, young hyphae, and the surface and deep layers of mature mycelium. Succinic dehydrogenase was low in conidia and about 10 times more active in mature mycelium. The surface layer had twice the activity of the deep layer. Aldolase increased about 3 times after the germination of conidia; it was slightly lower in the surface than in the deep layer of mycelium. Tryptophan synthetase exhibited only small differences between conidia and mycelium and was slightly lower in the surface than in the deep layer. β-galactosidase was formed in appreciable amounts only after prolonged growth and had a much higher specific activity in the deep layer. The results were discussed in connection with cell differentiation and aging.  相似文献

Hyphae of higher fungi are compartmentalized by septa. These septa contain a central pore that allows for inter‐compartmental and inter‐hyphal cytoplasmic streaming. The cytoplasm within the mycelium is therefore considered to be a continuous system. In this study, however, we demonstrate by laser dissection that 40% of the apical septa of exploring hyphae of Aspergillus oryzae are closed. Closure of septa correlated with the presence of a peroxisome‐derived organelle, known as Woronin body, near the septal pore. The location of Woronin bodies in the hyphae was dynamic and, as a result, plugging of the septal pore was reversible. Septal plugging was abolished in a ΔAohex1 strain that cannot form Woronin bodies. Notably, hyphal heterogeneity was also affected in the ΔAohex1 strain. Wild‐type strains of A. oryzae showed heterogeneous distribution of GFP between neighbouring hyphae at the outer part of the colony when the reporter was expressed from the promoter of the glucoamylase gene glaA or the α‐glucuronidase gene aguA. In contrast, GFP fluorescence showed a normal distribution in the case of the ΔAohex1 strain. Taken together, it is concluded that Woronin bodies maintain hyphal heterogeneity in a fungal mycelium by impeding cytoplasmic continuity.  相似文献

The growth pattern of Mucor globosus cultured on a medium with or without cyclic adenosine monophosphate (cAMP) was examined. Branching remarkably increased in a mycelium grown on an agar medium containing cAMP. In submerged culture containing cAMP, some sporangiospores grew spherically and formed yeast-like cells, and others showed hyphal growth. These hyphae showed septation and swelling and formed spore-like structures. When these hyphae were transferred to cAMP-free medium, a germ tube emerged from each compartment. These results show that cAMP has two different effects on the development of hyphae: one is the promotion of branching, and the other is the suppression of polarized growth.  相似文献

New fossil remains have been discovered from the well-known Lakhanda Microbiota (1015–1025 Ma, Uchur-Maya Region, Southeastern Siberia). The microfossils have characters observed in microscopic fungi, such as a reticulate mycelium, formed by anastomizing hyphae of non-cellular structure and of variable width. The fungal mycelium shows that hyphae grew in a certain direction, both forwards and backwards. As their length increased, the hyphae became curved and merged with neighboring hyphae to form enclosed spaces. The presence on hyphae of knoblike trapping structures and secretion of adhesive enzymes suggest a similarity with modern nematophagous fungi. Microorganisms associated with the adhesive hyphae consist of colonial green unicellular algae and thin bacterial sheaths, which may indicate mutually beneficial relationships between morphologically and biologically different partners. Microfossils of fungal origin are systematically described. They include Aimonema ramosa gen. et sp. nov. and the green unicellular alga Eoprotoderma neruenica gen. et sp. nov., forming a symbiotic association.  相似文献

Summary Conidia ofFusarium oxysporum f. sp.vasinfectum started to germinate on the roots of cotton (Gossypium barbadense L.) 6 h after inoculation and formed a compact mycelium covering the root surface. 18 h later, penetration hyphae branched off and infected the root. The number of penetration hyphae increased with the number of conidia used for inoculation. The optimal temperature for penetration was between 28 and 30 °C. The highest numbers of penetration hyphae were found in the meristematic zone, 40 percent less in the elongation and root hair zones, and none in the lateral root zone. The fine structure of the infection process was studied in protodermal cells of the meristematic zone and in rhizodermal cells of the elongation zone. The penetration hyphae were well preserved after freeze substitution and showed a Golgi equivalent consisting of three populations of smooth cisternae. Plant reactions were found already during fungal growth on the root surface. In the meristematic zone, a thickening of the plant cell wall due to an apposition of dark and lightly staining material below the hyphae occurred. This wall apposition increased in size around the hypha invading the plant cell and led to the formation of a prominent wall apposition with finger-like projections into the host cytoplasm. In the elongation zone, the deposits around the penetration hypha appeared less thick and the dark inclusions were less pronounced. High pressure freezing of infected cells revealed, thatF. oxysporum penetrates and grows within the host cells without inducing damages such as plasmolysis, cell degeneration or even host necrosis. Betaine-type lipids—diacylglyceryltrimethylhomoserines (DGTS)—were revealed in the mycelium of the basidial fungus Flammulina velutipes obtained by surface cultivation on agarized malt extract. DGTS accumulation was shown to occur at the late stages of culture development under deficiency of a complex of nutrients, including nitrogen, phosphorus, potassium, and trace elements. Induction of the synthesis of betaine lipids in F. velutipes occurred against the background of a decreased rate of growth of the vegetative mycelium, formation of monilioid hyphae, and inhibition of fructification. The relationship between DGTS formation and the environmental factors (temperature, illumination) was studied. It was established that the most active DGTS accumulation occurred at 15°C in the dark.  相似文献

 The mycelium of Suillus bovinus slowly absorbed [U-14C]glucose and other tracers from droplets placed on the cords, translocated them to the peripheral hyphae and exuded them into fluid drops on the hyphal tips. The exudate was characterized by 1H NMR spectroscopy and by sugar and amino acid analysis. The exuded compounds were mainly carbohydrates and peptides. Acetic acid and oxalic acid were also present in the exudate along with a number of unidentified compounds. Released ions (K, Na, Cl, P, Mg and Ca) were identified by X-ray microanalysis. The mycelium was shown to reabsorb up to 65% of the exuded 14C compounds in 2 days. Glucose, mannitol, glutamic acid (pH 3.2), and Rb+ (as well as other mineral ions) were all readily absorbed by the mycelium, while oxalic acid at pH 4.2 and glutamic acid at pH 6.5 were not. Exudation of fluid droplets on the surface of the hydrophobic mycorrhizal fungus S. bovinus may represent an ecophysiologically important function of the extramatrical hyphae, which provides an interface for interaction with the immediate hyphal environment and its other microorganisms where the peripheral hyphae exchange their photosynthetically derived products for nutrients to be used later by the pine host. We hypothesize that actively absorbed carbohydrates from the root are translocated to the peripheral hyphae along a concentration gradient of sugars and polyols by means of active translocation and diffusion in cell elements and by acropetal water transport in the cord vessels. The histopathological changes of the second instar nymph of the mealybug Phenacoccus fraxinus infected with Lecanicillium lecanii strain 3.4505 were investigated using light, scanning and transmission electron microscopy. The results demonstrated that L. lecanii 3.4505 could infect P. fraxinus in a short period. At 24 h post-inoculation, the conidia of L. lecanii 3.4505 adhered to the indented gloves or intersegmental folds of the insect body surface. Subsequently, the germinated conidia produced germ-tubes, appressoria and extended hyphae, which tightly adhered to the cuticle. Penetration of cuticle could be achieved either by peg form appressoria or directly by hyphae. Also, the conidia and hyphae could secrete massive mucilages causing visible damage to the host cuticle. After 48 h, the body wall, tissues and organs, including cuticle, trachea, fat body, muscle, Malpighian tubules and nerve ganglion, were destroyed by ramification of hyphae as a result of infection. The endoplasmic reticulum hypertrophied and formed obvious fingerprint agglomerates, and the mitochondria swelled and deformed in the haemocytes. Finally, the mycelium fully occupied the entire haemocoel. The entire bodies were wrapped in a white mycelium, with the mycelium extending radically outward.  相似文献

In the A-factor regulatory cascade leading to the onset of streptomycin biosynthesis and aerial mycelium formation in Streptomyces griseus, the A-factor receptor protein (ArpA) serves as a DNA-binding repressor and A-factor releases the repression by binding to ArpA and dissociating it from the DNA. Mutants defective in arpA therefore produce streptomycin and aerial hyphae in the absence of A-factor. A gene that inhibits streptomycin production and aerial hyphae formation in an arpA mutant was cloned on a high-copy-number plasmid and found to encode a eukaryotic-type adenylate cyclase (CyaA). Consistent with this, an exogenous supply of cAMP at high concentration almost abolished streptomycin production and aerial hyphae formation. On the other hand, cAMP at lower concentrations stimulated or accelerated these developmental processes. The effects of cAMP were detectable only in arpA mutants, and not in the wild-type strain; an exogenous supply of cAMP or cyaA disruption in the wild-type strain caused almost no effect on these phenotypes. Thus the effects of cAMP became apparent only in the arpA-defective background. cAMP at high concentrations inhibited stringent response factor ppGpp production, which is important for the onset of antibiotic biosynthesis. cAMP also influenced the timing of tyrosine phosphorylation of more than nine proteins. These findings show that a cAMP regulatory relay for physiological and morphological development functions in a concerted and interdependent way with other signal transduction pathways. When Trichophyton mentagrophytes thalli are placed on a medium containing 300 microgram/ml of coumarin, their growth rate is drastically reduced and the newly formed mycelium consists of curled and branched hyphae showing subapical bulges and swollen tips. Under the electron microscope, the most relevant abnormalities concern the cell walls which are often thickened by aberrantly shaped zones that are usually smeared unevenly over the surface of the "primary" wall instead of being deposited in discrete ridges. An ultracytochemical analysis suggests that the irregular thickenings are formed of chitin. Arguments suitable to explain the phenomena observed are discussed.  相似文献

The extramatrical mycelia of Suillus bovinus, Rhizopogon luteolus and R. vinicolor, all examples of hydrophobic (ho), mat-forming mycorrhizal fungi, were examined while associated with their hosts in the unsterilized rhizoscope, and efforts were made to produce and examine similar structures in vitro. Comparisons were made with four hydrophilic (hi) mycorrhizal fungi, Thelephora terrestris, Cenococcum geophilum, Laccaria laccata and Hebeloma crustuliniforme. The ho fungi formed linear structures (coarse, rhizomorph-like cords, with vessels in the center) and fans, both in the rhizoscope and in vitro. The same was seen in mycorrhizal mycelia in forest soils. These cords did not themselves give rise to the fans peripherally, and were not proper rhizomorphs, but were created continuously from single exploring air hyphae in the preexisting fan. Thus the ho exploring hyphae aggregated into strands, which grew in thickness only when no suitable, exploitable substrate was found. The assembly of hyphae creating ho cords was seen in the air as well as on inert hydrophilic (glass) or hydrophobic (plastic) surfaces, but never in water. It is hypothesized that the ho cell wall surface glues hyphae together while cords are formed. Water disturbed strands and mantles already formed. The ho exploring hyphae could also create ho mycelial patches (as in a mat) at the water-air interface of a number of substrates. The periphery of these patches seemed to be composed of shorter exploiting hyphae penetrating different water-soaked substrates. Exploring, aerial hyphal tips of the ho fungi were shown to excrete water droplets from openings in the ho cell wall surface, both in vitro and in the rhizoscope. In the rhizoscope, droplet excretion was apparently directly governed by photosynthesis in the shoot of the seedling. It is proposed that the drop exudation represents a kidney-like function of the extramatrical hyphae and a bridge to drier soil particles to initiate nutrient uptake by the hyphae. The ecological function of the different extramatrical structures of ho fungi are discussed. The ho cords or hyphae may translocate water only in the vessels or symplastically and not in the cell walls. The ho property may be essential among the S-selected (stress-tolerant) factors in these forest fungi. The transfer from water-repelling exploring structures into more hi exploiting structures in water contact with surrounding soil debris is, therefore, of great importance. The hi fungi did not form rhizomorph-like strands, in most cases, but an extending hyphal mycelium, representing foraging, exploring and exploiting structures at the same time. In the field, short strands may be found. On the hi fungi droplets were also produced but readily fused into a water sheath around the hypha. Trichoderma reesei was grown using purified cell walls ofNeurospora crassa as a primary source of carbon. The resulting culture medium contained an undefined mixture ofN. crassa cell-wall digesting enzymes. Protoplasts (cell lacking wall) were formed when youngN. crassa hyphae were treated withTrichoderma mixture. The vast majority of protoplasts resynthesized cell-wall material when washed free of cell-wall digesting enzyme; of these, about 40% regenerated a mycelium.  相似文献

The formation of microsclerotia of Pyrenochaeta lycopersici Schneider & Gerlach was observed in culture, in isolated tomato root segments and in roots of intact tomato plants. In culture microsclerotia formed in the aerial mycelium, and submerged in the agar; in tomato roots they formed in single cells of the outer cortex. The microsclerotium of P. lycopersici is structurally unspecialized and has cell walls of similar thickness throughout. Microsclerotia are thought to have powers of long survival in soil. In isolation the microsclerotia of P. lycopersici maintained viability over 2 years with consistently high germinability. Transverse hyphae were never seen in sections of microsclerotia, suggesting that germ-tubes arise from the outermost layers of cells.  相似文献

Six new types of developmental mutants were obtained from the bald variantbld-1 after treatment with mutagens (UV light, γ radiation, nitrous acid) and after natural selection Formation of the aerial mycelium was fully suppressed in thebld-2 type growing on media both with glucose and with starch. The other types were bald only on starch media, forming the aerial mycelium on media with glucose; typesspo-2, spo-3, spo-4 andspo-5 differed in size, shape and surface structure of spores, the typewhi formed asporogenous aerial hyphae.  相似文献

The mutant strainClaviceps purpurea 244 forming hyphae composed mainly from sclerotiumlike cells was found to sporulate both in liquid and solid media, particularly in the form of terminal chlamydospores (4.0 × 6.5 μm). Chlamydospores produced during submerged cultivation germinated, new chlamydospores being formed directly from germ tubes, or, occasionally, conidia (the so-called microcycle) or new vegetative mycelium were formed. The ultrastructures of the chlamydospores and vegetative cells was identical. The cytoplasm was filled with ribosomes and contained lipid inclusions and vacuoles with membrane invaginations. Strain 244 cultivated under submerged conditions produced 150 μg/ml clavins, with elymoclavin predominating (82 %). The parent strainClaviceps purpurea 129 only produced chlamydospores on the vegetative mycelium, whereas no microcycle was detected; under submerged conditions it produced mainly agroclavin (85 %) at a concentration of 4 mg/ml.  相似文献
