The effects of growth-inhibiting substances from young cotton bolls on phosphorylation in both intact cotton plant and tissue slices of young cotton bolls were determined using 32P as a tracer. The results showed that the incorporation of 32P into the various organic phosphates including high-energy phosphate compounds, were decreased in abscissing bolls to about 1/2 as that of developing bolls. Infiltration of the growth-inhibiting substances into normal boll tissue caused a marked increase in respiration intensity, the synthesis of high-energy phosphate compouds was, however, decreased to 39% of that of the control tissue. When 10 ppm NAA was applied tvabscissing young cotton bolls, the incorporation of 32P into various fractions of organic phosphates increased, although never to the level of developing bolls. The above results suggested that uncoupling of respiration and oxidative phosphorylation might be one of the important effects of the growth-inhibiting substances in inducing the abscission of cotton bolls.  相似文献

The sediments of Nagara river and its bystreams were extracted with ether. The mutagenicities of the extracts were determined by the Ames test with Salmonella typhimurium TA100 with S9 mix. The extracts were fractionated on an activated silica-gel column with four kinds of organic solvent, namely iso-octane, iso-octane:benzene (1:1), benzene:ethyl acetate (1:1) and benzene:methanol (1:1). The highest mutagenicity was observed in the iso-octane-benzene fraction of Arata river by using TA100 with S9 mix. Chemical substances in this fraction were identified by GC-MS and GC, and several kinds of polycyclic aromatic hydrocarbons (PAH) were found. Several peaks were unidentified, but these PAH, especially benzo [b] fluoranthene and benzo [a] pyrene, may contribute substantially to the mutagenicity of this fraction.  相似文献

Dihydroxyacetone, glyceraldehyde, glyoxal, methyl glyoxal, and glyoxylic acid were found to show mutagenicity on Salmonella typhimurium TA 100. The mutagenicities of these substances were inhibited by the addition of S-9 or some free radical scavengers. The alkaline buffered solutions of these mutagenic substances were found to reduce Nitro Blue tetrazolium chloride. DNA was degraded by the addition of these mutagenic substances. It has also been confirmed that free radicals derived from autoxidation of these substances are responsible for their mutagenicity.  相似文献

The growth-inhibiting effect of inhibitor β, extracted from the potato variety Majestic, was compared with the effect of abscisic acid by means of the Avena coleoptile straight-growth test. Dose-response curves showed that total growth inhibition is attained by inhibitor β, but this is never caused by abscisic acid even in high unphysiological doses. By chromatographic methods inhibitor β can be separated into three active components. One of these is a phenolic substance with weak growth-inhibiting effect. The two others contribute equally to the inhibitory effect, and one of them is probably abscisic acid. Beside these inhibiting substances there are components without inhibitory effect, many of phenolic character, included in the inhibitor β complex.  相似文献

The combination of short-term bioassays and analytical chemical techniques has been successfully used in the identification of a variety of mutagenic compounds in complex mixtures. Much of the early work in the field of bioassay-directed fractionation resulted from the development of a short-term bacterial assay employing Salmonella typhimurium; this assay is commonly known as the Ames assay. Ideally, analytical methods for assessment of mutagenicity of any environmental matrix should exhibit characteristics including high capacity, good selectivity, good analytical resolution, non-destructiveness, and reproducibility. A variety of extraction solvents have been employed in investigations of mutagenicity of air particulate; sequential combination of dichloromethane followed by methanol is most popular. Soxhlet extraction has been the most common extraction method, followed by sonication. Attempts at initial fractionation using different extraction solvents have met with limited success and highlight the need for fractionation schemes applicable to moderately polar and polar mutagenic compounds. Fractionation methods reported in the literature are reviewed according to three general schemas: (i) acid/base/neutral partitioning followed by fractionation using open-column chromatography and/or HPLC; (ii) fractionation based on normal-phase (NP) HPLC using a cyanopropyl or chemically similar stationary phase; and (iii) fractionation by open-column chromatography followed by NP-HPLC. The HPLC methods may be preparative, semi-preparative, or analytical scale. Variations based on acid/base/neutral partitioning followed by a chromatographic separation have also been employed. Other lesser-used approaches involve fractionation based on ion-exchange and thin-layer chromatographies. Although some of the methodologies used in contemporary studies of mutagenicity of air particulate do not represent significant advances in technology over the past 30 years, their simplicity, low cost, effectiveness, and robustness combine to result in their continued application in modern laboratories.  相似文献

Fumes from phospholipids pyrolyzed at 500~700°C did not themselves show any mutagenicity on Salmonella strains, but when the pyrolyzates were treated with a sodium chloride precipitate, active carbon, or an anionic exchange resin, the filtrates were found to be mutagenic on Salmonella TA 100. Tests confirmed that the phospholipid pyrolyzates contained both mutagenic and inactivating substances of this mutagenicity. Low level mutagenicity was produced on Salmonella TA 98, but there was no such activity on the other strains. Preincubation of the pyrolyzates with S-9 mix had no activating effect on mutagenicity. The inactivating substances of the mutagenicity were isolated and identified as long chain fatty acids.  相似文献

To test the assumption that automobile exhausts contribute to soil mutagenicity, two soils with low levels of mutagenic activities were exposed to traffic exhausts at a heavily charged junction of German motorways (Autobahnen) for 3, 7, 10, 13, 17, 21, and 26 weeks. Indeed, in the presence of a metabolic activation system from rat liver (S9), an average increase of 8 and 9 (4 and 12) revertants per gram per week was found in Salmonella typhimurium TA 98 (TA 100). In the absence of S9, meaningful measurements were impossible on account of a concurrent dose dependent increase of toxicity. No correlation between the increase of mutagenicity and the contents of polycyclic aromatic hydrocarbons (PAH) could be detected. In another series, soils sampled at the roadside and at distances of 10 and 50m of five roads near Mainz expressed 10-20-fold higher mutagenicity (revertants per gram) under identical test conditions as compared with the average of agricultural soils. Toxic effects, however, again confounded the results and no correlation between the distance from roads and the levels of mutagenicity could be demonstrated. Subsequently, Soxhlet-extraction with the solvent sequence dichloromethane, acetone, and toluene/diethylketone was found to be an optimum procedure for soils at roadsides. The mass balance of solvent fractionation of such soils revealed that <2% each belonged to organic acids and bases, approximately 4% to fractions designed polar neutrals, approximately 8% to polar aromatics, approximately 7% to dichloromethane solubles, and approximately 79% to cylohexane solubles, among them approximately 63% acetone soluble compounds. The major part of mutagenicity (55-65%) was present in the fraction of polar aromatics, followed by polar neutrals and the acetone subfraction of cyclohexane solubles ( approximately 10% each) summarizing the results obtained with S. typhimurium TA 98, TA 98NR, YG 1021, YG 1024, TA 100, YG 1026, and YG 1029 with and without addition of S9. The modified tester strains, either deficient in nitroreductase (TA 98NR) or overproducing nitroreductase (YG 1021, 1026) or O-acetyl-transferase (YG 1024, 1026), indicated a major contribution of nitroarenes to soil mutagenicity. With respect to mutagenic PAH, high pressure liquid chromatography (HPLC) revealed that >90% of dibenz[a,h]anthracene (4.18mg/kg soil), benzo[a]pyrene (1.96mg), benzofluoranthenes (0.14mg), and benz[a]anthracene (0. 18mg) were present in the acetone subfraction of cyclohexane solubles. Concentrations and mutagenic activities, however, did not correlate. Additional preparative and analytical HPLC of the solvent fractions of polar neutrals and polar aromatics, resulted in the tentative identification of 2-nitrofluorene. Analysis of the vertical profile of soil revealed an increase of mutagenicity per gram from the surface to a maximum at 5-15cm depth and a subsequent decrease with very little activity remaining deeper than 35cm. In human lymphocyte cultures, the fraction of polar aromatics, 0.01-0. 3microg/ml, induced 11.27+/-4.76-20.70+/-6.19 sister-chromatid exchanges (SCE) per cell in the absence of S9 (solvent control: 10. 16+/-4.83 SCE per cell) and 12.77+/-6.53-17.87+/-4.93 SCE per cell in the presence of S9 (solvent control: 8.37+/-3.92 SCE per cell). However, no activities could be detected in the fractions of polar neutrals and non-polar neutrals. Again, negative results were obtained in the in vivo mouse bone marrow micronucleus assay at 2000mg/kg p.o. with all fractions.  相似文献

During phototropic curvature, indolyl-3-acetic acid (IAA) remains evenly distributed in the hypocotyl of sunflower (Helianthus annuus L.) and in the oat (Avena sativa L.) coleoptile. At the irradiated side, growth inhibiting substances accumulate. In sunflower, basipetal movement of a growth factor is not involved, since the top of the seedling can be covered or removed without affecting the photo-tropic response; this response, moreover, is independent of the rate of elongation growth. The chemical nature of the growth-inhibiting substances is only partly known. In the hypocotyl they occur in the neutral fraction: in sunflower cis-xanthoxin is one of them, in radish (Raphanus sativus L.) cis- and trans-raphanusanins, and possibly raphanusamide, are involved. The inhibitor(s) in the oat coleoptile are acidic. During curvature, their amount remains rather constant but the distribution changes with an accumulation at the irradiated side. It is concluded that phototropic curvature is brought about by an accumulation, at the irradiated side, of growth-inhibiting substances that unilaterally reduce cell elongation even though the IAA distribution is uniform.  相似文献

The Ames II bacterial mutagenicity assay is a new version of the standard Ames test for screening chemicals for genotoxic activity. However, the use of plastic micro-titer plates has drawbacks in the case of testing hydrophobic mutagens, since sorptive and other losses make it difficult to control and define the exposure concentrations, and they reduce availability for bacterial uptake or to the S9 enzymes. With passive dosing, a biocompatible polymer such as silicone is loaded with the test compound and acts as a partitioning source. It compensates for any losses and results in stable freely dissolved concentrations. Passive dosing using silicone O-rings was applied in the Ames II assay to measure PAH mutagenicity in strains TA98 and TAMix – a mixture of six different bacterial strains detecting six different base-pair substitutions – after metabolic activation by S9. Initially, 10 PAHs were tested with passive dosing from saturated O-rings, aiming at levels in the test medium close to aqueous solubility. Fluoranthene, pyrene and benzo(a)pyrene were mutagenic in both TA98 and TAMix, whereas benz(a)anthracene was mutagenic in TA98 only. The concentration-dependent mutagenic activity of benzo(a)pyrene was then compared for passive dosing and solvent spiking. With spiking, nominal concentrations greatly exceeded aqueous solubility before mutagenicity was observed, due to sorptive losses and limiting dissolution kinetics. In contrast, the passive dosing concentration-response curves were more reproducible, and shifted towards lower concentrations by several orders of magnitude. This study raises fundamental questions about how to introduce hydrophobic test substances in the Ames II assay with biotransformation, since the measured mutagenicity not only depends on the compound potency but also on its supply, sorption and consumption during the assay.  相似文献

49 substances permitted for use in food in the United States was tested for mutagenicity in the Ames Salmonella typhimurium assay and in Escherichia coli strain WP2. Four of these substances caused increases in revertant counts in S. typhimurium. Two of these four (papain and pepsin) were found to contain histidine, and therefore the results of the tests on these two substances could not be taken as demonstrating mutagenicity. The other two substances causing increases in revertant counts (hydrogen peroxide and potassium nitrite) were mutagenic. The results on one chemical, beta-carotene, were evaluated as inconclusive or questionable. The remaining 44 substances were nonmutagenic in the test systems used. It is concluded that, for those generally physiologically innocuous chemicals tested, there are very few 'false positives' in the bacterial test systems used.  相似文献

Summary Abscisic acid (ABA) moves basipetally and laterally in maize (Zea mays L.) root segments placed horizontally; its transport properties are thus similar to those of the growth-inhibiting substances produced by the root cap. 采用水提和酯提2种方式提取炭团菌液体培养菌液中高效抑制樟子松枯梢病菌活性物质。采用生长速率法测定提取物对病原菌菌丝生长的抑制率,采用悬滴法测定其对病原菌孢子萌发的抑制率。炭团菌液体培养菌液乙酸乙酯提取物稳定性试验选用紫外线、温度、pH、氧化剂与还原剂以及贮存时间5个因子。结果表明:炭团菌液体培养菌液的5种提取物对樟子松枯梢病菌均有一定的抑菌活性,乙酸乙酯提取物对病原菌菌丝生长的抑制率最高,达到73.20%;乙酸乙酯提取物、超声波提取物、正丁醇提取物对病原菌孢子萌发抑制率均超过90%。乙酸乙酯提取物对自然环境下的紫外线、温度具有很高的稳定性,并且有一定的抗氧化还原能力和良好的耐热性,长期贮存不影响其抑菌活性,具有很高的开发价值和很好的应用前景。  相似文献

通过鹅膏菌抑菌活性物质对杨树烂皮病病原菌金黄壳囊孢菌的菌丝生长及其孢子抑制效果的研究,分析鹅膏菌的抗真菌活性。将培养15 d的鹅膏菌液体发酵物分成培养液和菌丝体两部分,然后分别将培养液、菌丝体提取物和发酵液本身对金黄壳囊孢菌菌丝生长和孢子萌发抑制效果进行检测。结果表明:培养液对金黄壳囊孢菌菌丝生长的抑制率最高,为68.90%;三者对孢子萌发都有明显的抑制效果,即在处理后10 d孢子都没有萌发。菌丝体在超声波、加热和浸泡的条件下,利用乙醇,丙酮、乙酸乙酯和正丁醇进行提取,其提取物用于病原菌的抑制效果检测,结果表明:用乙醇和丙酮提取时,超声波、浸泡及加热处理,对金黄壳囊孢菌的抑制效果都与对照有显著差异(a=0.01),而正丁醇的提取物的抑制效果较差,与对照没有显著差异。通过不同提取物对金黄壳囊孢菌菌丝生长和孢子萌发的抑制效果进行分析,并对抑菌活性物质提取工艺进行筛选,得到抑菌活性物质提取的最佳方法为摇床培养,乙醇是最适提取溶剂,处理方式(超声波、浸泡或者加热)相互间差异不显著。  相似文献

A new method is described for the quantitative characterization of the genotoxic effect of chemicals. The method is based on the determination of the inactivation of bacteriophage T7 and on the application of a simple mathematical model valid for the processes during, or at least in the initial stage of the interaction of chemicals and phages. A value characteristic for the chemical is defined and it is determined from the inactivation kinetics. Typical inactivation kinetic curves and some problems of the application of the model as well as the mutagenicity index values determined for about 30 substances are presented. The substances examined have mutagenicity index values covering a range of six orders of magnitude. The obtained values are compared with the results of different mutagenicity/carcinogenicity tests and discussed on the basis of data in the literature. The presented method is proposed to be applied for quantitative mutagenicity screening of chemicals.  相似文献

Mutagenicity of organic extracts from Beijing Guanting Reservoir sediments was investigated with TA98 and TA100 of Salmonella typhimurium. TA98 and TA100 were employed to detect frameshift mutation and base-pair substitution mutation, respectively. For TA100, no positive result was found, while TA98 was more sensitive and pro-mutagenic frameshift mutagens were mainly detected in sediments. Sediments from northern and southern Guanting Reservoir were at potential mutagenic risk. No mutagenicity was found in the sediments from the entrance of the tributaries, but strong mutagenicity was observed in the sediments from the outlet of the reservoir. Chemical analysis was also performed, and poor correlation was found between mutagenicity and organochlorine pesticides (OCPs). However, significant positive correlation was found for polycyclic aromatic hydrocarbons (PAHs) (r = 0.603–0.946), which showed that PAHs were dominated for the tested mutagenicity in the sediments. Polychlorinated biphenyls (PCBs) might induce mutagenicity or promote the mutagenicity of other substances.  相似文献

Refined corn bran (RCB), a dietary fiber derived from the mechanical refining of corn hulls, effectively adsorbed various environmental mutagens. When RCB was added at a concentration of 10 mg/ml to an aqueous solution of dinitropyrene (DNP), 91.6% of the mutagenicity towards Salmonella tester strain TA98 disappeared. Under similar conditions decreases in mutagenicity of DNP using wheat bran and cellulose powder were 58.4% and 43.0%, respectively. The adsorption of DNP to the fibers appeared irreversible since little mutagenicity was recovered by washing the treated fibers with aqueous buffer solutions of various pHs. Even with an organic solvent (methanol: ammonium hydroxide 50:1), only 2/3 of the mutagenicity of DNP was recovered. RCB could similarly adsorb mutagenic heterocyclic amines such as IQ, Trp-P-1, Trp-P-2, Glu-P-1, and Glu-P-2.  相似文献

The possible mutagenicity of the water-soluble contents of cigarette smoke (WSCS) was evaluated by using the Vicia faba root tip micronucleus test. The results showed significant changes in micronucleus frequency which were caused by each different concentration of WSCS. This indicates that the Vicia faba root tip micronucleus test might be used as one kind of mutagenic detection method for cigarette smoke. A comparative evaluation on the mutagenicity of 10 brands of cigarettes was carried out. Results confirmed that various degrees of mutagenicity were found for all of the brand cigarettes, among them, Huaihai was the highest, while Camellia was the lowest. The micronucleus frequencies were reduced by adding tea polyphenol, nicotinamide adenine, vitamin C and sodium selenite to the WSCS. The results suggest that these added substances might reduce the genetic injury induced by cigarette smoke.  相似文献

Organic extracts of diesel-exhaust particles show direct mutagenic activity in the Salmonella typhimurium bacterial mutagenicity assay. Nitro-aromatic compounds are believed to be responsible for part of the mutagenicity. A previously unidentified polyfunctional nitro-aromatic compound, 2-nitro-9-fluorenone (2N-Fone) was isolated from diesel-exhaust particles using a two-step fractionation scheme consisting of Sephadex LH20 chromatography and silica-gel thin-layer chromatography. Positive identification was by gas chromatography/mass spectroscopy and coelution with an authentic standard. Direct and indirect mutagenicities of 2N-Fone in several bacterial strains were also determined. The results indicated that 2N-Fone produces 60-70 rev/nmole of direct mutagenic activity, and is about 1/5 to 1/10 as mutagenic as 1-nitropyrene.  相似文献
