The expression of some insect P450 genes can be induced by both exogenous and endogenous compounds and there is evidence to suggest that multiple constitutively overexpressed P450 genes are co-responsible for the development of resistance to permethrin in resistant mosquitoes. This study characterized the permethrin induction profiles of P450 genes known to be constitutively overexpressed in resistant mosquitoes, Culex quinquefasciatus. The gene expression in 7 of the 19 P450 genes CYP325K3v1, CYP4D42v2, CYP9J45, (CYP) CPIJ000926, CYP325G4, CYP4C38, CYP4H40 in the HAmCqG8 strain, increased more than 2-fold after exposure to permethrin at an LC50 concentration (10 ppm) compared to their acetone treated counterpart; no significant differences in the expression of these P450 genes in susceptible S-Lab mosquitoes were observed after permethrin treatment. Eleven of the fourteen P450 genes overexpressed in the MAmCqG6 strain, CYP9M10, CYP6Z12, CYP9J33, CYP9J43, CYP9J34, CYP306A1, CYP6Z15, CYP9J45, CYPPAL1, CYP4C52v1, CYP9J39, were also induced more than doubled after exposure to an LC50 (0.7 ppm) dose of permethrin. No significant induction in P450 gene expression was observed in the susceptible S-Lab mosquitoes after permethrin treatment except for CYP6Z15 and CYP9J39, suggesting that permethrin induction of these two P450 genes are common to both susceptible and resistant mosquitoes while the induction of the others are specific to insecticide resistant mosquitoes. These results demonstrate that multiple P450 genes are co-up-regulated in insecticide resistant mosquitoes through both constitutive overexpression and induction mechanisms, providing additional support for their involvement in the detoxification of insecticides and the development of insecticide resistance.  相似文献

1. The acute toxicity of endosulfan was determined for the freshwater rotifer Brachionus calyciflorus. 2. The mean 24 hr LC50 value for endosulfan was 5.15 ppm with a coefficient of variation of 14.7%. 3. Rotifers were exposed at two sublethal concentrations (1.5-2.0 ppm) of endosulfan for bioaccumulation experiments, for an exposure time of 24, 48, 72 and 96 hr. The rotifers were fed with Nannochloris oculata (5 x 10(5) cell/ml). 4. The highest accumulation of endosulfan was found 24 hr after the start of the exposure to 1.5 ppm of the toxicant. A steady-state concentration in rotifer was reached between 24-48 hr, followed by a gradual decrease until 96 hr.  相似文献

利用闪烁管药膜法测定了2009年山东省德州、滨州、梁山、曲阜和聊城5个棉花产区绿盲蝽Lygus lucorum Meyer-Dür对硫丹、马拉硫磷、毒死蜱、灭多威、丁硫克百威、吡虫啉、联苯菊酯和氟虫腈8种杀虫剂的敏感性,筛选出适合各地区的高效防治药剂。结果表明:5个地区绿盲蝽种群对马拉硫磷、毒死蜱、丁硫克百威、联苯菊酯和吡虫啉处于敏感性阶段。不同种群之间对灭多威、硫丹和氟虫腈的敏感性差异较大,其中聊城种群为最敏感种群,滨州种群对灭多威、 硫丹和氟虫腈的LC50值分别为聊城种群的5.12,6.04和39.80倍;曲阜种群对灭多威、硫丹和氟虫腈的LC50值分别为聊城种群的22.12,5.48和22.80倍。两种群对此3种药剂的敏感性下降,而其余种群仍处于较敏感阶段。8种药剂对绿盲蝽成虫的毒力按大小依次排序为:氟虫腈>灭多威、联苯菊酯、硫丹>马拉硫磷、毒死蜱>丁硫克百威>吡虫啉。2009年7-10月间德州夏津绿盲蝽种群对8种杀虫剂的敏感性变化极微。  相似文献

The performance of an endosulfan tolerant strain of Trichogramma chilonis Ishii was evaluated against Helicoverpa armigera (Hübner) eggs on cotton plants in a net house. Under pesticide pressure, the tolerant strain could provide significantly higher parasitism in comparison to the susceptible strain. The parasitoid adult emergence was also significantly higher in the treatment with the tolerant strain in comparison to the susceptible strain, when parasitoids were released within 3 days of endosulfan spraying. Release of the tolerant strain of T. chilonis in conjunction with endosulfan spray application resulted in significantly higher pest mortality in comparison to the endosulfan treatment or release of T. chilonis alone. It could be inferred from the results that T. chilonis should be released only 3 days after endosulfan spray application as during the first 3 days after the spraying, endosulfan provided the required protection and adversely affected the performance of the parasitoid.  相似文献

A resistance monitoring program conducted for the polyphagous whitefly, Bemisia tabaci (Gennadius), in Imperial Valley, CA, USA generated a large set of LC50s for adults collected from broccoli, cantaloupe and cotton crops over a four-year period. A vial bioassay and, subsequently, a yellow-sticky card bioassay produced similar temporal profiles of relative susceptibilities to the pyrethroid insecticide bifenthrin. Both bioassays revealed that whiteflies collected from broccoli were significantly less susceptible to bifenthrin compared to the other two crops. A similar finding was observed for endosulfan and the mixture of bifenthrin+endosulfan in the yellow-sticky card bioassay. The possibility that seasonal differences contributed to the observed differences in susceptibility provided the impetus to conduct a reciprocal transfer experiment using broccoli (or kale) and cantaloupe grown simultaneously in the field and greenhouse. Whitefly adults collected from an organic farm over three consecutive weeks had significantly higher LC50s on kale than those collected the same day on cantaloupe. After culturing in the greenhouse on broccoli or cantaloupe and testing again, LC50s remained significantly higher on broccoli after one week and again at the F1 generation. In contrast, whiteflies originating on kale in the field and transferred to cantaloupes in the greenhouse had significantly reduced LC50s at the F1 generation. When tested against the bifenthrin+endosulfan mixture, significantly higher LC50s were generated for whiteflies reared on broccoli in the greenhouse at one week and the F1 compared to the field source from cantaloupes. The consistently higher LC50s for whiteflies on broccoli and other Brassica spp. crops, compared to cantaloupes or cotton, point to statistically significant host-plant influences that are expressed in both field-collected and greenhouse-reared populations of whiteflies.  相似文献

Selected commercial and technical grade pesticides were tested against the egg, preparasite and adult stages of Agamermis unka , a nematode parasite of the brown planthopper, Nilaparvata lugens . The commercial insecticide, diazinon (LC = 0.37 ppm), was most toxic to the 50 preparasites, followed by phenthoate (LC = 0.43 ppm), BPMC (LC = 0.44 ppm), IBP 50 50 (LC = 0.46 ppm), cartap hydrochloride (LC = 0.82 ppm) and buprofezin + isoprocarb 50 50 (LC = 1.11 ppm). The least toxic commercial pesticide tested was the fungicide, pencycuron 50 (LC = 2.19 ppm). Out of 12 technical grade insecticides tested, phenthoate, monocrotophos, 50 diazinon and carbofuran (LC = 0.37-0.46 ppm) were highly toxic to the preparasites, followed by 50 buprofezin, BPMC and fenitrothion (LC = 0.74-0.86 ppm). Fenthion, etofenprox, chlorpyrifos, 50 imidacloprid and MIPC (LC = 1.11-2.19 ppm) were the technical grade insecticides least toxic 50 to the preparasites. Most preparasites survived for up to 24 h at the low insecticide concentrations (0.63 and 0.31 ppm). Preparasites that were exposed to BPMC for 24 h at concentrations as high as 5.0 ppm and survived the treatments infected brown planthopper nymphs. Four selected insecticides-chlorpyrifos, BPMC, imidacloprid and carbofuran-had significant adverse effects on A. unka egg hatching. Eggs that were in the insecticide solution for 168 h fared poorly with imidacloprid having the best survival ( > 2% of the eggs hatching at 0.04 ppm). No eggs hatched from the other insecticide treatments. Three selected insecticides, BPMC, imidacloprid and chlorpyrifos, tested against adult A. unka showed that most adults survived the exposure to the insecticides between 0.31 and 2.5 ppm. At 5.0 ppm of BPMC or chlorpyrifos none of the adults survived, whereas with imidacloprid 70% of the adults survived. Egg deposition by the surviving adults was greatly reduced in those treated with the insecticides compared with those in the controls. Imidacloprid had some negative impact on the preparasites' ability to infect BPH nymphs, but it had the least detrimental effect of the insecticides tested on preparasite survival and on the eggs and adults of A. unka .  相似文献

Abstract  Insecticide resistance monitoring using a Potter precision spray tower with discriminating concentration and log dose probability techniques underpins the Australian insecticide management strategy for Frankliniella occidentalis Pergande. Abamectin, acephate, chlorpyrifos, dichlorvos, dimethoate, endosulfan, fipronil, malathion, methamidophos methidathion, methiocarb, methomyl, pyrazophos and spinosad are recommended for use against F. occidentalis but abamectin, methiocarb and pyrazophos are the only chemicals where insecticide resistance has not been detected. Although not registered, chlorfenapyr was effective against F. occidentalis and should be pursued for that purpose. In contrast, chlorpyrifos, dichlorvos and malathion resistance were detected at low to moderate levels throughout the study period putting their sustainable use for F. occidentalis control in doubt . Although it appears that acephate, dimethoate, endosulfan, fipronil, methamidophos, methidathion and spinosad remain effective, some populations contained a small percentage of thrips that survived exposure to a concentration that killed 100% of the susceptible strain. Subsequent laboratory selection of one such population separately with fipronil and spinosad caused an increase in resistance to these insecticides. These products must now be considered at risk. This is the first report of fipronil or spinosad resistance in populations of F. occidentalis.  相似文献

Individual and combined toxicity of three pesticides, endosulfan, phosphamidon, and aldicarb was evaluated in P. conchonius. The 48 hr LC50 was 21.36 and 446.5 ppm respectively for endosulfan and phosphamidon. When tested jointly, 48 hr LC50 for different ratios of these pesticides were 0.332 (IE:3P), 0.224 (IE:1P), and 0.178 ppm (3E:1P). The cotoxicity coefficients for these combinations were 1793, 3986, and 10009, respectively. An equitoxic mixture of endosulfan, phosphamidon, and aldicarb yielded a 48 hr LC50 of 130.5 ppm. An enhanced toxic impact is indicated when the pesticides are present together rather than as individual compounds.  相似文献

Abstract  The sensitivity of three reference strains of the western flower thrips Frankliniella occidentalis from Australia, Israel and Turkey was evaluated and their response did not differ significantly and so they are considered typical of susceptible F. occidentalis . Field strains of F. occidentalis from Antalya, Turkey were tested for resistance against five classes of insecticide during the years 2001–2003. Difference in the field strains response to abamectin, endosulfan, malathion and methomyl was low (<3.0-fold) and in many cases not significant. However, all strains were likely resistant to cypermethrin (1.6- to 12.2-fold at LC50 (lethal concentration)). The established agricultural practices and ecosystem patterns in the Antalya province are assumed to contribute to delay in insecticide resistance in F. occidentalis .  相似文献

The toxicity of a commercial formulation of the insecticide parathion‐methyl to the N2‐fixing filamentous cyanobacterium (blue‐green alga) Cylindrospermum, sp. was studied. A concentration of parathion‐methyl of 0.5 ppm caused growth increase in liquid growth media. The minimum inhibitory concentration of parathion‐methyl for both types (N2, fixing and nitrate supplemented) of liquid and solid media was 1.0 ppm. LC50 values were: 4.4 ppm (liquid, N2, fixing), 5.5 ppm (liquid, nitrate supplemented), 3.3 ppm (agar, N2‐fixing) and 4.0 ppm (agar, nitrate supplemented). LC100 values for N2‐fixing liquid and both types of agar media were 10.0 ppm, while for the liquid nitrate supplemented medium the LC100 was 12.0 ppm. Both akinete (spore) formation and germination were inhibited below the highest permissive concentration of 8.0 ppm, with the insecticide incorporated in the agar media. In soil, the LC50 and LC100 values for parathion‐methyl were 13.6 and 30 ppm, respectively. Both the dehydrogenase activity of heterocysts (monitored by 2,3,5‐triphenyl tetrazolium chloride reduction) and the nitrogen concentration of cultures (estimated by the micro‐Kjeldahl method) were affected by the insecticide, but the latter (N2‐fixation) was more sensitive. The Kruskal‐Wallis H test on the numbers of vegetative cells in the filaments revealed that the insecticide significantly affected the division of vegetative cells. The cyanobacterium could detoxify the growth medium containing high levels (30 and 40 ppm) of the insecticide in short‐term exposures at the expense of cell viability.  相似文献

Baseline toxicity levels to a novel semicarbazone insecticide, metaflumizone were established for 25 field populations of Colorado potato beetle, Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae),from North America. Excluding the susceptible laboratory strain, 50% lethal concentrations of metaflumizone ranged from 0.57 to 1.31 ppm, while response slopes ranged from 1.92 to 4.24 (average = 2.93), and were unrelated to the 50% lethal concentration (r = 0.06; P = 0.76). Beetle populations with known resistance to the neonicotinoid imidacloprid also exhibited the highest LC50 levels to metaflumizone suggesting at least the possibility of cross-resistance. Additional experiments using a potato leaf-dip bioassay as well as field efficacy evaluations confirmed the high level of toxicity of metaflumizone to L. decemlineata and demonstrated a potential benefit of tank mixing a low rate of the pyrethroid esfenvalerate with metaflumizone at one-tenth the recommended field rate. These research findings confirm that metaflumizone is highly active against L. decemlineata larvae and adults and could provide an effective alternative insecticide for potato pest management.  相似文献

Oscillatoria anguistissima could tolerate 50 ppm ZnSO4.7H2O, and a zinc-tolerant strain with maximum tolerance concentration (MTC) of 100 ppm ZnSO4.7H2O was obtained by stepwise transfer to higher concentrations. The adaptation was irreversible even after three generations in metal-free medium. In the presence of metal, the tolerant strain grew with a shorter lag period of 4 days as against 6 days in the case of the wild strain. The tolerant strain had higher MTC than that of the wild strain for other metals also, viz., Ni2+, Co2+, Cu2+ and Cd2+. The zinc resistance in the tolerant strain was a result of reduced uptake, since around 42% of the total metal was present on the surface as against only 30% in the wild strain. The calcium-stimulated uptake, as observed in the wild strain, was absent in the tolerant strain. Ultrastructural comparisons revealed no structural change in the tolerant strain on exposure to zinc, whereas in the wild strain a thick extracellular matrix was observed. Toxicological and neurophysiological studies were performed to characterize the resistance mechanism in a cyclodiene-resistant strain of Drosophila melanogaster (Maryland strain). Dieldrin had an LC50 of 0.058 ppm against the larvae of susceptible D. melanogaster (Oregon-R wild type) when formulated in the rearing media. The LC50 of the resistant Maryland strain was 10.8 ppm, giving a resistance ratio (LC50-Maryland/LC50-susceptible) of 186-fold. Suction electrode recordings were made from peripheral nerves of the larval central nervous system to test whether reduced nerve sensitivity played any role in the observed resistance. In susceptible preparations (n = 5), inhibition of nerve firing by 1 mM gamma-aminobutyric acid (GABA) was effectively antagonized within 3-10 min by 10 microM dieldrin. In contrast, 30 min incubations with 10 microM dieldrin had no effect on preparations from cyclodiene-resistant individuals (n = 5). Similarly, 10 microM picrotoxinin blocked GABA-dependent inhibition in susceptible nerve preparations (n = 3). In recordings from resistant insects (n = 4), picrotoxinin displayed either weak antagonism of GABA or hyperexcitation indistinguishable from susceptible preparations. These results demonstrate that cyclodiene resistance in the Maryland strain of D. melanogaster 1) is expressed in immature stages, 2) is present at the level of the nerve, and 3) extends to picrotoxinin, albeit at a reduced level compared with dieldrin. The possible role of an altered GABA receptor in this resistance is discussed.  相似文献

In a first series of experiments, the biological response of a continuous cell line of the beet armyworm, Spodoptera exigua, was tested with different groups of insecticides with different modes of actions: acetylcholinesterase inhibitors, acetycholine receptor agonists, inhibitors and uncouplers of oxidative phosphorylation, site I electron transport inhibitors, gamma-aminobutyric acid receptor inhibitors, chitin synthesis inhibitors, and juvenile hormone analogues. From the concentration response curves, 50% inhibition concentration (IC(50)) values were calculated. The most active compound in vitro was pyridaben with an IC(50) value of 0.0083 ppm. In a second series of experiments, the toxicity of these insecticide groups was determined on third-instar larvae of S. exigua, and lethal concentration with 50% kill (LC(50)) values were used in the evaluation of their in vivo biological activity. Toxicity bioassays showed that lufenuron was the most toxic (LC(50) = 0.098 ppm). To explain the discrepancies in biological responses in vitro with insect cells compared with in vivo conditions with whole third-instar larvae, the significance of different detoxifying enzyme systems was tested. P(450) monooxygenases, esterases, and glutathione S-transferases were measured in third-instar larvae and cells of S. exigua. Data are discussed in terms of the usefulness of insect cell cultures as tools in the screening for novel insecticide actions.  相似文献

Abstract Resistance to diflubenzuron in the Australian sheep blowfly, Lucilia cuprina , has rendered this insecticide incapable of preventing flystrike in sheep from a few districts in eastern Australia. Wool producers affected by this situation must find suitable alternatives to protect their flocks. Results of laboratory bioassays against one population demonstrated that, despite extremely high diflubenzuron resistance (Resistance Factor >791), it had only very low (2x) tolerance of cyromazine and dicyclanil. It is unlikely that this level of tolerance would have any practical impact on field control with either insecticide. Consequently, wool producers in districts where diflubenzuron-resistant flies are common can rotate insecticide treatment to either of these compounds to prevent flystrike in their flocks. However, unlike the highly diflubenzuron-resistant field strain, a laboratory strain selected for resistance to diflubenzuron (Resistance Factor = 617) was 10 times more resistant to dicyclanil than a susceptible strain but, like the field strain, was only two times more tolerant of cyromazine. Conversely, a field-derived strain selected in the laboratory for cyromazine resistance was 20 times more resistant to dicyclanil and 362 times more resistant to diflubenzuron than the reference susceptible strain.  相似文献

During 1995-1998, we tested 134 geographically discrete populations of Colorado potato beetle, Leptinotarsa decemlineata (Say), from the United States, Canada, Germany, France, and Poland for susceptibility to imidacloprid. Neonates were assayed on potato-based agar diet incorporated with imidacloprid and exposed on filter paper to esfenvalerate, azinphosmethyl, and carbofuran to characterize cross-resistance. In all 4 yr, Long Island populations were the most tolerant to imidacloprid, with LC50s ranging up to 29 times higher than the most susceptible populations. Responses to imidacloprid did not change significantly on farms where populations were assayed over time, except for those from Long Island, which doubled in overall tolerance to imidacloprid since 1995. Much of this tolerance was already present before imidacloprid was used on Long Island. Correlative analysis of the populations tested over the 4 yr indicated positive cross-resistance patterns with esfenvalerate and azinphosmethyl. This response was probably caused by preexisting metabolic and excretion mechanisms selected by previous exposure. There was no significant pattern of cross-resistance with carbofuran or bensultap. Regression slopes were also significantly negatively correlated with LC50 values for imidacloprid, indicating higher heterogeneity, which could lead in further resistance development. We discuss the relative sensitivity of diet-incorporated assays with neonates compared with other bioassay studies. Based on a pooled group of susceptible populations tested in 1995, a baseline LC50 of 0.39 ppm and a discriminating concentration of 8 ppm were suggested to detect early stages of resistance in "suspect" populations. We also suggest application strategies for imidacloprid that reduce selection pressure.  相似文献

Fourteen strains of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) had mortalities ranging from 5 to 100% when exposed to diatomaceous earth at 600 ppm for seven days. The most tolerant strain had a lethal dose for 50% of the population (LD50) of 413 ppm and the most susceptible strain had a LD50 of 238 ppm. Adults of the tolerant strain were lighter (2.0 mg) than the susceptible strain (2.6 mg). Tolerant adults lost water at lower rate (6 g h–1 than susceptible adults (12 g h–1), when held in wheat treated with 600 ppm diatomaceous earth for 24 h, than held at 5% r.h. with no food. Tolerant adults that were not exposed to diatomaceous earth lost water at a lower rate (3 g h–1) than susceptible adults (5 g h–1). Both strains, exposed and not exposed to diatomaceous earth died when their water content was between 33 and 37% of their total weight. Insects taken directly from the cultures had 52% (tolerant) and 53% (susceptible) of their total weight as water. Tolerant adults moved slower through grain and across filter paper than susceptible adults. Tolerant adults avoided wheat treated with diatomaceous earth at concentrations as low as 75 ppm, whereas the adults from the susceptible strain did not avoid diatomaceous earth, even at 600 ppm. The consequences of a strain tolerant to diatomaceous earth is discussed with respect to the use of diatomaceous earth to control stored-product insect infestations.  相似文献

Homalodisca coagulata Say, adults from three locations in California were subjected to insecticide bioassays to establish baseline toxicity. Initially, two bioassay techniques, petri dish and leaf dip, were compared to determine the most useful method to establish baseline susceptibility data under laboratory and greenhouse conditions. Comparative dose-response data were determined by both techniques to endosulfan, dimethoate, cyfluthrin, and acetamiprid. Toxic values were similar to some insecticides with both techniques but not for all insecticides, revealing susceptibility differences among the three populations of H. coagulata. In subsequent tests, the petri dish technique was selected to establish baseline susceptibility data to various contact insecticides. A systemic uptake bioassay was adapted to estimate dose-mortality responses to a systemic insecticide, imidacloprid. A 2-yr comparison of toxicological responses showed all three populations of H. coagulata to be highly susceptible to 10 insecticides, including chlorpyrifos, dimethoate, endosulfan, bifenthrin, cyfluthrin, esfenvalerate, fenpropathrin, acetamiprid, imidacloprid, and thiamethoxam. In general, two pyrethroids, bifenthrin and esfenvalerate, were the most toxic compounds, followed by two neonicotinoids, acetamiprid and imidacloprid. The LC50 values for all insecticides tested were lower than concentrations used as recommended field rates. Baseline data varied for the three geographically distinct H. coagulata populations with the petri dish technique. Adult H. coagulata collected from San Bernardino County were significantly more susceptible to select pyrethroids compared with adults from Riverside or Kern counties. Adults from San Bernardino County also were more sensitive to two neonicotinoids, acetamiprid and imidacloprid. The highest LC50 values were to endosulfan, which nonetheless proved highly toxic to H. coagulata from all three regions. In the majority of the tests, mortality increased over time resulting in increased susceptibility at 48 h compared with 24 h. These results indicate a wide selection of highly effective insecticides that could aid in managing H. coagulata populations in California.  相似文献

Endosulfan, a chlorinated hydrocarbon insecticide of cyclodiene subgroup acts as a contact poison in a wide variety of organisms. In the present study, the effect of endosulfan on the growth, alpha amylase activity and plasmid amplification was investigated in Bacillus subtilis system. The bacteria were grown in medium, incubated with different concentrations (32, 48, 64 and 80 microg/mL) of endosulfan. The bacterial growth was gradually seen after 1st day at up to 48 microg/L endosulfan. The 48 microg/L endosulfan inhibited approximately 50% of the bacterial growth. No growth was observed at and after 64 microg/L endosulfan, for all days (1-5). Also, no alpha amylase activity was found in the supernatant of the culture medium containing 64 and 80 microg/L endosulfan, whereas slight activity was observed with 32 and 48 microg/L endosulfan concentration. The amount of plasmid increased up to 50% in the presence of 32 microg/L endosulfan. Endosulfan had no effect on the alpha amylase activity in vitro.  相似文献

将室内选育成功的抗敌百虫淡色库蚊Culex pipiens pallens Coq.品系(RD)分为二个分系,一个不再用敌百虫处理,称之为RD衰退品系(RD139-x),34代后对敌百虫的敏感度增加了10倍,对溴氰菊酯的敏感度无显著变化.另一分系改用溴氰菊酯选育,命名为Rde品系,53代后对溴氰菊酯抗性达200倍左右,对敌百虫敏感度上升约10倍,对DDT的交互抗性高达118倍,对马拉硫磷、杀螟硫磷的敏感度与敏感品系(SEN,上海昆虫所保存)比较也有上升,呈负交互抗性现象.用高剂量溴氰菊酯处理幼虫、也证明Rde在20分钟内麻痹率比敏感品系低,可见抗性机制主要是抗击倒因子(Kdr).但增效醚(Pb)对溴氰菊酯明显增效,可见mfo酶也起重要作用,推测抗性为多因子遗传.  相似文献
