The pre-steady states of Pseudomonas species lipase inhibitions by p-nitrophenyl-N-substituted carbamates (1–6) are composed of two steps: (1) formation of the non-covalent enzyme–inhibitor complex (E:I) from the inhibitor and the enzyme and (2) formation of the tetrahedral enzyme–inhibitor adduct (E–I) from the E:I complex. From a stopped-flow apparatus, the dissociation constant for the E:I complex, KS, and the rate constant for formation of the tetrahedral E–I adduct from the E:I complex, k2 are obtained from the non-linear least-squares of curve fittings of first-order rate constant (kobs) versus inhibition concentration ([I]) plot against kobs=k2+k2[I]/(KS+[I]). Values of pKS, and log k2 are linearly correlated with the σ* values with the ρ* values of −2.0 and 0.36, respectively. Therefore, the E:I complexes are more positive charges than the inhibitors due to the ρ* value of −2.0. The tetrahedral E–I adducts on the other hand are more negative charges than the E:I complexes due to the ρ* value of 0.36. Formation of the E:I complex from the inhibitor and the enzyme are further divided into two steps: (1) the pre-equilibrium protonation of the inhibitor and (2) formation of the E:I complex from the protonated inhibitor and the enzyme.  相似文献

Cadmium-induced initial changes in the production of reactive oxygen species (ROS) and antioxidant mechanism were investigated in soybean (Glycine max L. cv. Don Mario 4800 RR) leaves. Whole plants (WP) and plants without roots (PWR) were exposed to 0.0, 10.0 and 40.0 μM Cd for 0, 4, 6 and 24 h. Compared to PWR, a higher level of endogenous Cd in WP was associated with a lower oxidative stress measured in terms of lipid peroxidation. Furthermore, O2 •− content decreased in the leaves of Cd-treated WP, whereas it increased in those of Cd-treated PWR. Although O2 •− accumulation in PWR was associated with a decrease in superoxide dismutase (SOD) activity, O2 •− diminution in WP leaves was not related to any increase in SOD activity. H2O2 content increased in the leaves of both Cd-treated WP and PWR, and it was concomitant with a corresponding decline in catalase (CAT) and ascorbate peroxidase (APX) activities. When diphenyl iodonium (DPI), an inhibitor of NADPH oxidase, was added, H2O2 content remained unchanged in Cd-treated WP, suggesting that NADPH oxidase does not participate in the early hours of Cd toxicity. Taken together, our results showed that early ROS evolution and oxidative damage were different in WP and PWR. This suggests that the response in soybean leaves during the early hours of Cd toxicity is probably modulated by the root.  相似文献

The symbiotic association of endophyte fungus, Neotyphodium lolii, and ryegrass improves the ryegrass resistance to drought. This is shown by a 30 % increase in the number of suckers in infected plants (E+), compared to plants lacking endophyte (E−), and by a higher water potential in the E+ than E− plants. The E+ plants have higher stomatal conductance (g s), transpiration rate, net photosynthetic rate (P N), and photorespiratory electron transport rate than the E− plants. The maximal photochemical efficiency (Fv/Fm) and the actual photochemical efficiency (ΦPS2) are not affected by the endophyte fungus. The increase in P N of the E+ plants subjected to water stress was independent from internal CO2 concentration. An increased P N was observed in E+ plants also in optimal water supply. Hence the drought resistance of E+ plants results in increased g s, P N, and photorespiratory electron transport rate. Studies on the chemical and biological properties of annual pack ice at a coastal station in Terra Nova Bay (74°41.72′S, 164°11.63′E) were carried out during austral spring at 3-day intervals from 5 November to 1 December 1997. Temporal changes of nutrient concentrations, algal biomasses, taxonomic composition, photosynthetic pigment spectra and P–E relationships were studied. Quantity, composition and degradation rates of organic matter in the intact sea ice were also investigated. In addition, microcosm experiments were carried out to evaluate photosynthetic and photo-acclimation processes of the sympagic flora in relation to different light regimes. High concentrations of ammonia were measured in four ice-cores (weighted mean values of the cores ranged from 4.3 ± 1.9 μM to 7.2 ± 3.4 μM), whereas nitrate and phosphate displayed high concentrations (up to 35.9 μM and 7.6 μM, respectively) only in the bottom layer (135–145 cm depth). Particulate carbohydrate and protein concentrations in the intact sea ice ranged from 0.5 to 2.3 mg l−1 and 0.2 to 2.0 mg l−1, respectively, displaying a notable accumulation of organic matter in the bottom colored layer, where bacterial enzymatic activities also reached the highest values. Aminopeptidase activity was extremely high (up to 19.7 μM l−1 h−1 ± 0.05 in the bottom layer), suggesting a rapid turnover rate of nitrogen–enriched organic compounds (e.g. proteins). By contrast, bacterial secondary production was low, suggesting that only a very small fraction of mobilized organic matter was converted into bacterial biomass (<0.01‰). The sympagic autotrophic biomass (in terms of chlorophaeopigments) of the bottom layer was high, increasing during the sampling period from 680 to 2480 μg l−1. Analyses of pigments performed by HPLC, as well as microscope observations, indicated that diatoms dominated bottom communities. The most important species were Amphiprora sp. and Nitschia cfr. stellata. Bottom sympagic communities showed an average P B max of 0.12 mgC mg Chl−1 and low photoadaptation index (E k=18 μE m−2 s−1, E m=65 μE m−2 s−1). Results of the microcosm experiment also indicated that communities were photo-oxidized when irradiance exceeded 100 μE m−2 s−1. This result suggests that micro- autotrophs inhabiting sea ice might have a minor role in the pelagic algal blooms. Accepted: 4 August 1999  相似文献

Soluble Organic Nitrogen Pools in Forest soils of Subtropical Australia   总被引:15,自引:0,他引:15  
Soil soluble organic N (SON) plays an important role in N biogeochemical cycling. In this study, 22 surface forest soils (0–10 cm) were collected from southeast Queensland, Australia, to investigate the size of SON pools extracted by water and salt solutions. Approximately 5–45 mg SON kg−1, 2–42 mg SON kg−1 and 1–24 SON mg kg−1 were extracted by 2 M KCl, 0.5 M K2SO4 and water, on average, corresponding to about 21.1, 13.5 and 7.0 kg SON ha−1 at the 0–10 cm forest soils, respectively. These SON pools, on average, accounted for 39% (KCl extracts), 42% (K2SO4 extracts) and 43% (water extracts) of total soluble N (TSN), and 2.3% (KCl extracts), 1.3% (K2SO4 extracts) and 0.7% (water extracts) of soil total N, respectively. Large variation in SON pools observed across the sites in the present study may be attributed to a combination of factors including soil types, tree species, management practices and environmental conditions. Significant relationships were observed among the SON pools extracted by water, KCl and K2SO4 and microbial biomass N (MBN). In general, KCl and K2SO4 extracted more SON than water from the forest soils, while KCl extracted more SON than K2SO4. The SON and soluble organic C (SOC) in KCl, K2SO4 and water extracts were all positively related to soil organic C, total N and clay contents. This indicates that clay and soil organic matter play a key role in the retention of SON in soil.  相似文献

This study investigated the effects on running economy (RE) of ingesting either no fluid or an electrolyte solution with or without 6% carbohydrate (counterbalanced design) during 60-min running bouts at 80% maximal oxygen consumption (O2max). Tests were undertaken in either a thermoneutral (22–23°C; 56–62% relative humidity, RH) or a hot and humid natural environment (Singapore: 25–35°C; 66–77% RH). The subjects were 15 young adult male Singaporeans [O2max = 55.5 (4.4 SD) ml kg−1 min−1]. The RE was measured at 3 m s−1 [65 (6)% O2max] before (RE1) and after each prolonged run (RE2). Fluids were administered every 2 min, at an individual rate determined from prior tests, to maintain body mass (group mean = 17.4 ml min−1). The O2 during RE2 was higher (P < 0.05) than that during the RE1 test for all treatments, with no differences between treatments (ANOVA). The mean increase in O2 from RE1 to RE2 ranged from 3.4 to 4.7 ml kg−1 min−1 across treatments. In conclusion, the deterioration in RE at 3 m s−1 (65% O2max) after 60 min of running at 80% O2max appears to occur independently of whether fluid is ingested and regardless of whether the fluid contains carbohydrates or electrolytes, in both a thermoneutral and in a hot, humid environment. Photosynthetic parameters of phytoplankton and sea ice algae from landfast sea ice of the Chukchi Sea off Point Barrow, Alaska, were assessed in spring 2005 and winter through spring 2006 using Pulse Amplitude Modulated (PAM) fluorometry including estimates of maximum quantum efficiency (F v/F m), maximum relative electron transport rate (rETRmax), photosynthetic efficiency (α), and the photoadaptive index (E k). The use of centrifuged brine samples allowed to document vertical gradients in ice algal acclimation with 5 cm vertical resolution for the first time. Bottom ice algae (0–5 cm from ice–water interface) expressed low F v/F m (0.331–0.426) and low α (0.098–0.130 (μmol photons m−2s−1)−1) in December. F v/F m and α increased in March and May (0.468–0.588 and 0.141–0.438 (μmol photons m−2s−1)−1, respectively) indicating increased photosynthetic activity. In addition, increases in rETRmax (3.3–16.4 a.u.) and E k (20–88 μmol photons m−2 s−1) from December to May illustrates a higher potential for primary productivity as communities become better acclimated to under-ice light conditions. In conclusion, photosynthetic performance by ice algae (as assessed by PAM fluorometry) was tightly linked to sea ice salinity, temperature, and inorganic nutrient concentrations (mainly nitrogen).  相似文献

The potential of the dried yeast, wild-type Schizosaccharomyces pombe, to remove Ni(II) ion was investigated in batch mode under varying experimental conditions including pH, temperature, initial metal ion concentration and biosorbent dose. Optimum pH for biosorption was determined as 5.0. The highest equilibrium uptake of Ni(II) on S. pombe, q e, was obtained at 25 °C as 33.8 mg g−1. It decreased with increasing temperature within a range of 25–50 °C denoting an exothermic behaviour. Increasing initial Ni(II) concentration up to 400 mg L−1 also elevated equilibrium uptake. No more adsorption took place beyond 400 mg L−1. Equilibrium data fitted better to Langmuir model rather than Freundlich model. Sips, Redlich–Peterson, and Kahn isotherm equations modelled the investigated system with a performance not better than Langmuir. Kinetic model evaluations showed that Ni(II) biosorption process followed the pseudo-second order rate model while rate constants decreased with increasing temperature. Gibbs free energy changes (ΔG°) of the system at 25, 30, 35 and 50 °C were found as −1.47E + 4, −1.49E + 4, −1.51E + 4, and −1.58E + 4 J mol−1, respectively. Enthalpy change (ΔH°) was determined as −2.57E + 3 J mol−1 which also supports the observed exothermic behaviour of the biosorption process. Entropy change (ΔS°) had a positive value (40.75 J mol−1 K−1) indicating an increase in randomness during biosorption process. Consequently, S. pombe was found to be a potential low-cost agent for Ni(II) in slightly acidic aqueous medium. In parallel, it has been assumed to act as a separating agent for Ni(II) recovery from its aqueous solution.  相似文献

The effects of cadmium (Cd) on germination, and antioxidative enzyme activity (AEA) involving superoxide dismutase, catalase, peroxidase, and ascorbate peroxidase, and on amounts of malondialdehyde and proline present within Achnatherum inebrians, were determined for specimens infected (E+) vs. non-infected (E−) by Neotyphodium gansuense, and cultivated in the presence of various concentrations of CdCl2 (0, 50, 100, 200 and 300 μmol/l). Under high Cd concentrations (100, 200 and 300 μM), E+ (vs. E−) specimens exhibited a higher germination rate and index, and higher values for shoot length, root length and dry biomass, but there was no significant difference (P > 0.05) under low Cd concentrations (0 and 50 μM). AEA and the proline content increased, but malondialdehyde content declined in the E+ (vs. E−) specimens under high Cd concentrations (100, 200 and 300 μM). There was no significant difference (P > 0.05) under low Cd concentrations (0 and 50 μM). Endophyte infection was concluded to be of benefit to the germination and anti-oxidative mechanisms within A. inebrians under plant exposures to high CdCl2 concentrations.  相似文献

To determine if heat exposure alters the hormonal responses to moderate, repeated exercise, 11 healthy male subjects [age = 27.1 (3.0) years; maximal oxygen consumption, O2max = 47.6 (6.2) ml · kg · min−1; mean (SD)] were assigned to four different experimental conditions according to a randomized-block design. While in a thermoneutral (23°C) or heated (40°C, 30% relative humidity) climatic chamber, subjects performed either cycle ergometer exercise (two 30-min bouts at ≈50% O2max, separated by a 45-min recovery interval, CEx and HEx conditions), or remained seated for 3 h (CS and HS conditions). Blood samples were analyzed for various exercise stress hormones [epinephrine (E), norepinephrine (NE), dopamine, cortisol and human growth hormone (hGH)]. Passive heating did not alter the concentrations of any of these hormones significantly. During both environmental conditions, exercise induced significant (P < 0.001) elevations in plasma E, NE and hGH levels. At 23°C during bout 1: E = 393 (199) pmol · l−1 (CEx) vs 174 (85) pmol · l−1 (CS), NE = 4593 (2640) pmol · l−1 (CEx) vs 1548 (505) pmol · l−1 (CS), and hGH = 274 (340) pmol · l−1 (CEx)vs 64 (112) pmol · l−1 (CS). At 40°C, bout 1: E = 596 (346) pmol · l−1 (HEx) vs 323 (181) pmol · l−1 (HS), NE = 7789 (5129) pmol · l−1 (HEx) vs 1527 (605) pmol · l−1 (HS), and hGH = 453 (494) pmol · l−1 (HEx) vs 172 (355) pmol · l−1 (HS). However, concentrations of plasma cortisol were increased only in response to exercise in the heat [HEx = 364 (168) nmol · l−1 vs HS = 295 (114) nmol · l−1). Compared to exercise at room temperature, plasma levels of E, NE and cortisol were all higher during exercise in the heat (P < 0.001 in all cases). The repetition of exercise did not significantly alter the pattern of change in cortisol or hGH levels in either environmental condition. However, repetition of exercise in the heat increased circulatory and psychological stress, with significantly (P < 0.001) higher plasma concentrations of E and NE. We studied the functional response of the freshwater unionid bivalve Anodonta anatina, feeding on five phytoplankton strains differing in food quality: the small green alga Scenedesmus obliquus, a toxic and a non-toxic strain of the filamentous cyanobacterium Planktothrix agardhii and a toxic and a non-toxic strain of the coccoid cyanobacterium Microcystis aeruginosa. On S. obliquus, A. anatina had a type II functional response with a maximum mass-specific ingestion rate (IRmax) of 5.24 mg C g DW−1 h−1 and a maximum mass-specific clearance rate (CRmax) of 492 (±38) ml g DW−1 h−1, the highest values for all the phytoplankton strains that were investigated. On toxic and non-toxic P. agardhii filaments, A. anatina also had a type II functional response, but IRmax and CRmax were considerably lower (IRmax 1.90 and 1.56 mg C g DW−1 h−1; CRmax 387 (±97) and 429 (±71) ml g DW−1 h−1, respectively) than on S. obliquus. Toxicity of P. agardhii had no effect on the filtration rate of the mussels. On the non-toxic M. aeruginosa (small coccoid cells), we also observed a type II functional response, although a type I functional response fitted almost as good to these data. For the colonial and toxic M. aeruginosa, a type I functional response fitted best to the data: IR increased linearly with food concentration and CR remained constant. CRmax and IRmax values for the (colonial) toxic M. aeruginosa (383 (±40) ml g DW−1 h−1; 3.7 mg C g DW−1 h−1) demonstrated that A. anatina filtered and ingested this cyanobacterium as good as the other cyanobacterial strains. However, on the non-toxic M. aeruginosa we observed the lowest CRmax of all phytoplankters (246 (±23) ml g DW−1 h−1, whereas IRmax was similar to that on toxic M. aeruginosa. The high maximum ingestion rates on S. obliquus and M. aeruginosa indicate a short handling time of these phytoplankton species. The high clearance rates on S. obliquus, toxic M. aeruginosa and P. agardhii reflect a high effort of the mussels to filter these particles out of the water column at low concentrations. The low clearance rates on non-toxic M. aeruginosa may be explained by the small size and coccoid form of this cyanobacterium, which may have impaired A. anatina to efficiently capture the cells. Although A. anatina had relatively high maximum clearance rates on non-toxic and toxic P. agardhii, this cyanobacterium does not seem to be a good food source, because of the observed high rates of pseudofaeces production and hence low ingestion rates.  相似文献

The goal of our study was to investigate the impact of fungal endophytes in tall fescue (Festuca arundinacea) on rhizodeposition and in turn, the soil microbial community. Sand-based, aseptic microlysimeter units were constructed for the collection of rhizodeposit solutions for chemical analyses from the roots of endophyte-free (E−) and endophyte-infected (E+) tall fescue plants. E+ plants were infected with Neotyphodium coenophialum, the most common endophyte found in tall fescue. Rhizodeposit solutions collected over nine weeks from E+ grass contained more organic carbon and carbohydrates than E−. These solutions were allowed to percolate through columns of plant-free soils to assess the response of the soil microbial communities. Soils to which solutions from E+ grass were applied had significantly higher respiration rates than those receiving solutions from E− grass, suggesting that microbial activity was stimulated by changes in the rhizodeposits. Culture-based assays of the soil microbial community (plate counts and community-level physiological profiling) suggest that the basic structure of the microbial community was not affected by application of rhizodeposit solutions from E+ plants as compared to E−. Our results indicate that the presence of a fungal endophyte may enhance rhizodeposition by tall fescue and could consequently influence microbial mineralization processes in the soil. In grasslands where nutrients may be limiting, hosting a fungal endophyte has the potential to enhance plant nutrient supply indirectly via a stimulatory effect on the soil microbial biomass. Leaf carbon gain simulation was performed forQuercus serrata seedlings with previously reported 6 day photosynthetic photon flux density (PPFD) histograms from 20 understorey microsites of a pine forest (Washitani & Tang 1991). This simulation was performed with or without an assumption of the acclimatization of photosynthetic capacity (Pmax) to microsite light availability. A constant ratio of respiration rate to Pmax, within, the range of 0.07–0.1, was assumed as a constraint. In relatively well illuminated microsites with a diffuse site factor above 0.1, predicted optimal Pmax was about 5 μmol m−2 s−1, with the predicted mean daily net carbon gain being about 50 mmol m−2 day−1. Each of the predicted optimal Pmax and the simulated mean daily net carbon gains with a constant Pmax (5 μmol m−2 s−1) or the predicted optimal Pmax was linearly related to the microsite light availability index, diffuse site factor. Simulated net carbon gain was negative at diffuse site factors below 0.04, if the constant of Pmax was assumed. The predicted linear relationship between net carbon gain and diffuse site factor could provide an ecophysiological basis for the observed linear dependency of the relative growth rate of biomass ofQ. serrata seedlings on the microsite diffuse site factor (Washitani & Tang 1991).  相似文献

Sequence alignment has been an invaluable tool for finding homologous sequences. The significance of the homology found is often quantified statistically by p-values. Theory for computing p-values exists for gapless alignments [Karlin, S., Altschul, S.F., 1990. Methods for assessing the statistical significance of molecular sequence features by using general scoring schemes. Proc. Natl. Acad. Sci. USA 87, 2264–2268; Karlin, S., Dembo A., 1992. Limit distributions of maximal segmental score among Markov-dependent partial sums. Adv. Appl. Probab. 24, 13–140], but a full generalization to alignments with gaps is not yet complete. We present a unified statistical analysis of two common sequence comparison algorithms: maximum-score (Smith-Waterman) alignments and their generalized probabilistic counterparts, including maximum-likelihood alignments and hidden Markov models. The most important statistical characteristic of these algorithms is the distribution function of the maximum score S max, resp. the maximum free energy F max, for mutually uncorrelated random sequences. This distribution is known empirically to be of the Gumbel form with an exponential tail P(S max > x) ∼ exp(−λx) for maximum-score alignment and P(F max > x) ∼ exp(−λx) for some classes of probabilistic alignment. We derive an exact expression for λ for particular probabilistic alignments. This result is then used to obtain accurate λ values for generic probabilistic and maximum-score alignments. Although the result demonstrated uses a simple match-mismatch scoring system, it is expected to be a good starting point for more general scoring functions.  相似文献

Chlorophyll (Chl) content, dry mass, relative water content (RWC), leaf mass per area (LMA), proline (Pro) content, malondialdehyde (MDA) content, superoxide dismutase (SOD) and peroxidase (POD) activity, P N-PAR response curves and gas exchange were studied to determine the effects of water stress on photosynthetic activity, dry mass partitioning and metabolic changes in four provenances of neem (Azadirachta indica A. Juss). The results indicated that provenance differences existed in the adaptation response to water stress that included changes to growth strategies coupled with ecophysiological and metabolic adjustments. As water stress increased, stomatal conductance (g s), net photosynthetic rate (P N), transpiration rate (E), and leaf RWC decreased while LMA increased in all provenances. Dry mass was reduced in droughted plants and the percentage increased in dry mass allocated to roots, and enzyme activities of SOD and POD were highest in neem originating from Kalyani (KA) provenance and lowest in neem originating from New Dehli (ND) provenance. In contrast, water stress increased MDA content least in KA and most in ND. Furthermore, neem originating from ND also had the greatest decrease in Chl a/b ratio while the ratio was least affected in neem originating from KA. These findings suggest neem originating from KA may have more drought resistance than neem originating from ND. The data from P N-PAR response curves are less clear. While these curves showed that drought stress increased compensation irradiance (I c) and dark respiration (R D) and decreased saturation irradiance (I s) and maximum net photosynthetic rate (P max), the extent of decline in P max was provenance dependent. P max under non-waterlimiting conditions was higher in neem originating from Jodhpur (MA) (about 14 μmol m−2 s−1) than in the other three provenances (all about 10 μmol m−2 s−1), but mild water stress had minimal effect on P max of these three provenances whereas P max of MA provenance declined to 10 μmol m−2 s−1, i.e. a similar value. However, under severe water stress P max of MA and KA provenances had declined to 40% of non-stressed values (about 6 and 4 μmol m−2 s−1, respectively) whereas the decline in P max of neem originating from Kulapachta (KU) and ND provenances was about 50% of nonstressed values (about 5 μmol m−2 s−1). These data suggest the P N responses of KU and ND provenances are most tolerant, and KA and MA least tolerant to increasing water stress, but also suggest MA provenance could be the most desired under both non-water-limiting and water-limiting conditions due to highest P max in all conditions.  相似文献

While global climate change in polar regions is expected to cause significant warming, the annual cycle of light and dark will remain unchanged. Cultures of three species of Antarctic sea ice diatoms, Fragilariopsis cylindrus (Grunow) Krieger, Thalassiosira antarctica Comber and Entomoneis kjellmanii (P.T. Cleve) Poulin and Cardinal, were incubated in the dark and exposed to differing temperatures. Maximum dark survival times varied between 30 and 60 days. Photosynthetic parameters, photosynthetic efficiency (α), maximum quantum yield (Fv/Fm), maximum relative electron transport rate (rETRmax) and non-photochemical quenching (NPQ), showed that dark exposure had a significant impact on photoacclimation. In contrast, elevated temperatures had a relatively minor impact on photosynthetic functioning during the dark exposure period but had a considerable impact on dark survival with minimal dark survival times reduced to only 7 days when exposed to 10°C. Recovery of maximum quantum yield of fluorescence (Fv/Fm) was not significantly impacted by temperature, species or dark exposure length. Recovery rates of Fv/Fm ranged from −5.06E−7 ± 2.71E−7 s−1 to 1.36E−5 ± 1.53E−5 s−1 for monthly experiments and from −9.63E−7 ± 7.71E−7 s−1 to 2.65E−5 ± 2.97E−5 s−1 for weekly experiments. NPQ recovery was greater and more consistent than Fv/Fm recovery, ranging between 5.74E−7 ± 8.11E−7 s−1 to 7.50E−3 ± 7.1E−4 s−1. The concentration of chl-a and monosaccharides remained relatively constant in both experiments. These results suggest that there will probably be little effect on Antarctic microalgae with increasing water temperatures during the Antarctic winter.  相似文献

Annual gross primary productivity in mesotrophic Shahidullah Hall pond (Dhaka, Bangladesh) was 1383.35 g C m−2 y−1 (arithmetic mean). Daily primary productivity (between 1.6 and 6.8 g C m−2 d−1 was correlated with chlorophylla, day length and dissolved silica. Chlorophylla related significantly withk, incident light, SRP, alkalinity and conductivity. A negative correlation existed between biomass and rainfall. Productivity, biomass, conductivity, alkalinity, and SRP increased after mid-winter.k, I k andZ eu varied according seasonally.P max related directly with temperature. The marine algal species in the Baltic Sea are few due to the low sea water salinity. One of the few species that can be found is Fucus vesiculosus. Even this species is affected by the low salinity and becomes smaller in size in the Baltic. In present work the photosynthesis of F. vesiculosus in the northern Baltic (Bothnian Sea) was compared to the photosynthesis of F. vesiculosus in the Atlantic. Oxygen evolution was measured before and after exposure to 2.3 W of UV-B (280–320 nm) radiation for 5 h, as well as after 48 h recovery in low light. The plants were kept in their own sea water salinity as well as in a changed salinity, this to examine possible correlations between salinity and photosynthesis. The results show a significant higher initial maximal photosynthesis (P max) for Atlantic plants (10.3 nmol O2 g−1 FW s−1) compared to Baltic plants (4.0 nmol O2 g−1 FW s−1). The Baltic plants were found more sensitive to UV-B with a 40–50% decrease of P max as well as a lower degree of recovery (60–70% compared to 75–95% for the Atlantic plants). The higher salinity (35 psu) had a positive effect on the Baltic F. vesiculosus with increased P max as well as increased tolerance to UV-B. The lower salinity (5 psu) had a negative effect on the Atlantic plants with a decreased P max as well as a lower tolerance to UV-B. Pigment content was found higher in Atlantic F. vesiculosus. The pigment content decreased then the Atlantic plants were transferred to 5 psu. The concentration of Chl a as well as the total content of violaxanthin, diadinoxanthin and zeaxanthin in Baltic plants increased when transferred to 35 psu. The Atlantic F. vesiculosus can not survive the low salinity in the northern Baltic (died within 8 weeks). It is likely that a long time acclimation or adaptation to low salinity has taken place for F. vesiculosus in northern Baltic. If this is an ecotypic or genotypic development it is too early to say.  相似文献

Benthic algal communities can play an important role in matter and energy flux of shallow lakes. Their contribution to total primary production of lakes has been largely unexplored. The aim of this study was to estimate the primary production of the epipsammic algal communities at different water depths in Lake Balaton (Hungary) with photosynthetic measurements performed in laboratory. The photosynthesis of the benthic algae of different origin was studied at nine different irradiance levels, in three replicates. The maximum photosynthetic rate (P max) was always higher in samples from the shallow parts than those from the deeper regions of the lake. Along the west–east longitudinal axis of the lake P max decreased in the southern part and increased in the middle of the lake as a consequence of differences in the chlorophyll-a concentrations. Knowing P max, I k, global radiation and extinction coefficient, the primary production (mg C m−2 day−1) of the epipsammic algal community was calculated at different water depths. In the shallow regions at 0.5 and 1 m water depth 75–95% and 60–85% of the production was attributable to the epipsammon. The percentage contribution of epipsammon was at 2 m water depth 20–65%. In the deeper pelagic region (>3 m) more than 85% of the primary production originated from the phytoplankton.  相似文献

The kinetic parameters Km and Vmax for urea uptake by Melosira italica were determined at 160 μeinsteins m−2 s−1 and in the dark. The transport systems showed an affinity for the substrate and a storing capacity in the dark (Km = 65.07 μM; Vmax = 2.18 nmoles 105 cells −1 h−1) greater than under 160 μE m−2 s −1 (Km = 111.2 μM; Vmax = 1.11 nmoles 105 cells−1 h−1). Similarly, a reduction in consumption rate of urea under increasing photon flux densities was observed. The use of an inhibitor (potassium cyanide) indicated that the uptake process requires metabolic energy. That urea transport is more important in darkness, may constitute a survival strategy in which this compound is utilized by cells mainly during heterotrophic growth.  相似文献
