通过高精度的双向电泳技术对家蚕中部丝腺组织的蛋白质进行分离,采用基质辅助激光解析电离飞行时间质谱(matrix-assistedlaserdesorption/ionizationtimeofflightmassspectrometry,MALDI-TOF-MS)对其中一些表达量较高的蛋白点进行鉴定,并利用GPMAW(GeneralProtein/MassAnalysisforWindows)软件结合家蚕基因组预测的蛋白质数据库构建本地的肽质量指纹图谱数据库,对所得到的肽质量指纹图谱进行分析。研究发现,经过双向凝胶电泳及其图象分析技术,硝酸银染色和考马斯亮蓝染色分别能分离出500个以上和100个以上的蛋白点。这些蛋白质点主要集中在分子量15~90kD区域,等电点pH3·5~7之间。MALDI-TOF-MS鉴定的25个考染蛋白点中有60%以上的PMF(PeptideMassFingerprint)的信号峰较强。在数据库检索过程中,利用家蚕肽质量指纹数据库所得检索结果与在Mascot的检索结果相比,前者不仅能够准确鉴定出一些已有研究报道的蛋白,从而验证检索方法的可行性,而且还能够对一些已经被家蚕基因组数据库所预测但未曾报道的新蛋白质进行鉴定,从而建立了一整套适合于家蚕蛋白质组研究的方法,并为其它绢丝昆虫蛋白质组研究提供了重要参考。  相似文献

天麻蛋白质的双向电泳和肽质量指纹谱分析与鉴定   总被引:7,自引:0,他引:7  
采用双向聚丙烯酰胺凝胶电泳和质谱技术对天麻染菌球茎皮层和不染菌的新生球茎皮层进行了比较蛋白质组分析与鉴定。双向电泳后在分子量 1 2~97kD、等电点 3~ 1 0范围内 ,每块胶分离到约 90 0个蛋白质点。对新生球茎中表达量明显增加的 5个蛋白质点用基质辅助激光解吸 电离飞行时间质谱 (MALDI TOFMS)进行肽质量指纹谱的分析 ,并通过检索不同的数据库进行蛋白质鉴定与功能预测 ,初步认为第 4号蛋白点是一个与转录有关的RNA结合蛋白。同时本文在天麻蛋白质组样品制备、数据库检索策略以及蛋白质鉴定成功率等方面进行了探讨。  相似文献

文章用蛋白质组学方法初步分析大鼠脑皮质蛋白质的表达。提取大鼠脑皮质蛋白质,双向凝胶电泳分离,考马斯亮蓝染色,胰蛋白酶胶内酶解,用基质辅助激光解吸/电离飞行时间质谱对酶解后的肽段进行分析,根据肽质量指纹图谱,检索专业数据库(Swissprot),对蛋白质进行鉴定。鉴定出84个蛋白,分别属于代谢酶、细胞骨架蛋白、热休克蛋白、抗氧化蛋白、信号传导蛋白、蛋白酶体相关蛋白、神经元特异蛋白及神经胶质蛋白等。文章结果丰富了大鼠脑皮质蛋白质组数据库,为在大鼠模型上研究神经疾病奠定了基础。  相似文献

采用双向聚丙烯酰胺凝胶电泳和肽质量指纹谱,对耐热性不同的月季品种热激处理后进行蛋白质组分析与鉴定。双向电泳后,差异蛋白质点主要集中在分子量10-30kDa、等电点5~6范围内,对耐热品种在高温下特异表达的蛋白质点中选取3个进行肽质量指纹谱的分析,并检索不同的数据库进行蛋白质鉴定与功能预测,初步认为这些蛋白质点分别是eIF-SA、LEA蛋白和Hsp17.5。同时结合已报道其他植物中这3种蛋白的功能,对月季耐高温生理机制进行初步探讨。  相似文献

目的:采用蛋白组学研究家蝇抗菌肽cecropin对人肝癌BEL-7402细胞蛋白的影响,从蛋白质水平探讨抗菌肽抗肿瘤机制.方法:在测定肿瘤细胞生长指数的基础上,通过双向凝胶电泳分离差异蛋白,银染后进行图像分析,对表达差异2倍以上蛋白质点进行胶内酶解和MALDI-TOF-MS检测,获得肽质量指纹谱,应用Mascot搜索引擎在NCBI nr数据库中进行检索鉴定.结果:家蝇抗菌肽cecropin能够抑制BEL-7402细胞的生长,与对照组比较,抗菌肽处理组双向凝胶电泳共答定出12个差异表达蛋白点(上调蛋白6个,下调蛋白6个),大部分与细胞凋亡、细胞代谢、基因转录以及细胞骨架等相关.结论:这为深入研究抗菌肽抗肿瘤机制打下了基础.  相似文献

对双向凝胶电泳后银染显色的蛋白质点经脱色与原位还原和烷基化处理后,用TPCK胰蛋白酶进行酶解,采用带有C18反相载体的ZipTip^TM吸头进行脱盐处理,再进行MALDI-TOF肽质谱指纹纹图分析,然后将肽质数据在EMBL数据库中进行搜寻从而对蛋白南点进行鉴定。结果表明用该实验程序可对银染的单一蛋白南点进行快速肽质谱指纹图ipTip^TM的应用可以明显增加质谱分析的信噪比,提高分析灵敏度。用以上方  相似文献

在布氏杆菌(Brucella)的感染免疫过程中,单核巨噬细胞的应答起着非常关键的作用,而毒力不同的布氏杆菌引起的宿主反应截然不同。用双向电泳技术对THP-1单核细胞受毒力不同的布氏杆菌株侵袭后的全细胞蛋白谱进行差异比较和分析,共发现了38个差异表达的蛋白质点。这些点经过胶内酶切后进行MALDI-TOF质谱鉴定,每个蛋白质点的肽质量指纹图谱都在人类的蛋白质组数据库中用Mascot进行检索后,发现这些差异表达的蛋白主要集中在结构蛋白,信号传导途径和物质代谢等领域,还有一些功能未知的蛋白。这一结果为研究布氏杆菌的感染与致病机制提供了方向,对深入探讨病原菌-宿主的相互作用模式具有参考价值。  相似文献

目的:用蛋白质组学方法初步分析大鼠海马蛋白质的表达。方法:提取大鼠海马蛋白质样品后,用双向凝胶电泳对其分离,经考马斯亮蓝染色后,产生大鼠海马蛋白质双向凝胶电泳图谱。从凝胶上切割分离的蛋白质,经胰蛋白酶胶内酶解,通过基质辅助激光解吸/电离飞行时间质谱(MALDI-TOF-MS)对酶解后的肽段进行分析。根据肽段质谱数据,经数据库(NCBI)检索,对蛋白质进行鉴定。结果:鉴定了37种具有明确功能的蛋白质,它们分别属于代谢酶、细胞骨架蛋白、热休克蛋白、抗氧化蛋白、信号传导蛋白、蛋白酶体相关蛋白、神经元特异蛋白及神经胶质蛋白。另外,鉴定了3种未知功能蛋白。结论:为建立大鼠海马蛋白质组数据库提供必要的资料,为在大鼠模型上研究神经疾病发病机理奠定基础。  相似文献

目的:通过研究帕金森病和正常外周血单个核细胞(PBMC)的蛋白质组差异,初步探讨外周免疫系统与帕金森病的病理联系.方法:用固相pH梯度双向凝胶电泳分离人帕金森病和正常单个核细胞总蛋白质,考马斯亮蓝染色,PDQuest 2-DE软件分析,对部分差异蛋白质点进行基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)测定其胶内酶解后的肽质指纹图谱,用Mascot查询系统查询SWISS-PROT数据库.结果:获得了分辨率和重复性均较好的双向电泳考染图谱,对其中的21个差异蛋白质点分别进行肽质指纹分析,经数据库查询,初步鉴定为一些与蛋白降解、抗氧化应激、信号转导、细胞骨架、细胞周期调控等有关的蛋白质.结论:建立了帕金森病PBMC的双向凝胶电泳图谱,提示帕金森病和正常的PBMC的蛋白质表达具有差异.  相似文献

目的:研究PTEN缺失细胞的蛋白表达规律。方法:用双向电泳技术比较Pten^+/+MEFs与Pten^-/-EFs细胞的蛋白表达差异;用基质辅助激光解吸/电离飞行时间质谱(MALDI-TOF-MS)对差异蛋白进行质谱分析;用肽质量指纹图谱检索数据库对差异蛋白进行鉴定;用Northern印迹和Western印迹验证蛋白的差异表达。结果:与Pten^+/+MEFs细胞相比。Pten^-/-MEFs细胞有明显的差异性蛋白表达谱,Pten^-/-MEFs细胞中表达下降的蛋白有铜锌超氧化物歧化酶、过氧化物氧化还原酶5和6等,表达增加的蛋白有低分子量蛋白酪氨酸磷酸酶和丝切蛋白(cofflin)1。结论:PTEN的缺失引起细胞内多种蛋白表达改变,这些表达改变的蛋白可能与PTEN缺失后细胞癌变相关。  相似文献

High-grade serous ovarian cancer (HGSOC) is the most lethal gynecologic malignancy in women. Its low survival rate is attributed to late detection, relapse, and drug resistance. The lack of effective second-line therapeutics remains a significant challenge. There is an opportunity to incorporate the use of histone deacetylase inhibitors (HDACi) into HGSOC treatment. However, the mechanism and efficacy of HDACi in the context of BRCA-1/2 mutation status is understudied. Therefore, we set out to elucidate how HDACi perturb the proteomic landscape within HGSOC cells. In this work, we used TMT labeling followed by data-dependent acquisition LC-MS/MS to quantitatively determine differences in the global proteomic landscape across HDACi-treated CAOV3, OVCAR3, and COV318 (BRCA-1/2 wildtype) HGSOC cells. We identified significant differences in the HDACi-induced perturbations of global protein regulation across CAOV3, OVCAR3, and COV318 cells. The HDACi Vorinostat and Romidepsin were identified as being the least and most effective in inhibiting HDAC activity across the three cell lines, respectively. Our results provide a justification for the further investigation of the functional mechanisms associated with the differential efficacy of FDA-approved HDACi within the context of HGSOC. This will enhance the efficacy of targeted HGSOC therapeutic treatment modalities that include HDACi.  相似文献

该研究利用4个由高到低不同海拔的同质园实验,以青藏高原高寒草地优势植物垂穗披碱草(Elymus nutans)、矮嵩草(Kobresia humilis)和珠芽蓼(Polygonum viviparum)为对象,分析了植物个体根、茎、叶生物量分配及根冠比的变化规律及影响因素。结果表明:(1)植物个体根、茎、叶质量比和根冠比具有显著的种间差异;与垂穗披碱草和珠芽蓼相比,矮嵩草具有显著较高的根质量比而叶、茎质量比较低,所以其根冠比较高。(2)在向低海拔移栽的过程中,珠芽蓼叶质量比保持不变,茎质量比显著降低而根质量比显著升高,根冠比表现出显著上升的趋势;垂穗披碱草则相反,即叶、茎质量比显著升高而根质量比显著降低,根冠比表现出显著下降的趋势;矮嵩草根、茎、叶质量比和根冠比则无显著变化。(3)随着海拔降低,年均气温明显升高而年均降雨量明显降低,且在植物个体种源地和土壤基质保持一致的条件下,向低海拔移栽过程中温度是导致珠芽蓼根、茎、叶生物量分配及根冠比变化的重要因素,而水分是垂穗披碱草根、茎、叶生物量分配及根冠比变化的重要驱动因素;矮嵩草根、茎、叶生物量分配及根冠比受其遗传因素影响较大。因此,在将来暖干化的背景下,青藏高原高寒草地植物生物量的分配将会发生改变,导致它们对资源(光照、水分和土壤养分)获取和利用的变化而改变它们的种间关系,从而影响群落的物种多样性与组成,最终可能导致生态系统功能的变化。  相似文献

Structural characterization of peptides in the range of 500–5000 Da, using fast atom bombardment (FAB) and Cs+ ion liquid secondary ion mass spectrometry (SIMS), is reviewed. These include syntheitc peptides Kemptamide (mol wt 1516); GIF-C15 (mol wt 1875), an isolated natural product as an acylated pentapeptide; and polypeptides generated from enzymatic digests of proteins. MS data is shown to reveal molecular weight and sequence information as well as determine disulfide bonds between cysteine residues and glycosylation sites in the case of a glycopeptide. The complementarity of MS technique to classical biochemical methods for peptide characterization is highlighted. The reader is briefly acquainted with two newer ionization techniques namely, electrospray ionization (ESI) and matrix-assisted laser desorption ionization (MALDI). Synthetic chemists and biochemists can refer to the in-depth review articles that are cited throughout this article.  相似文献

Alpha-glucan phosphorylase catalyzes the reversible cleavage of α-1-4-linked glucose polymers into α-D-glucose-1-phosphate. We report the recombinant production of an α-glucan/maltodextrin phosphorylase (PF1535) from a hyperthermophilic archaeon, Pyrococcus furiosus, and the first detailed biochemical characterization of this enzyme from any archaeal source using a mass-spectrometry-based assay. The apparent 98 kDa recombinant enzyme was active over a broad range of temperatures and pH, with optimal activity at 80 °C and pH 6.5–7. This archaeal protein retained its complete activity after 24 h at 80 °C in Tris-HCl buffer. Unlike other previously reported phosphorylases, the Ni-affinity column purified enzyme showed broad substrate specificity in both the synthesis and degradation of maltooligosaccharides. In the synthetic direction of the enzymatic reaction, the lowest oligosaccharide required for the chain elongation was maltose. In the degradative direction, the archaeal enzyme can produce glucose-1-phosphate from maltotriose or longer maltooligosaccharides including both glycogen and starch. The specific activity of the enzyme at 80 °C in the presence of 10 mM maltoheptaose and at 10 mg ml–1 glycogen concentration was 52 U mg–1 and 31 U mg–1, respectively. The apparent Michaelis constant and maximum velocity for inorganic phosphate were 31 ± 2 mM and 0.60 ± 0.02 mM min–1 µg–1, respectively. An initial velocity study of the enzymatic reaction indicated a sequential bi-bi catalytic mechanism. Unlike the more widely studied mammalian glycogen phosphorylase, the Pyrococcus enzyme is active in the absence of added AMP.  相似文献

Imaging mass spectrometry (IMS) is two-dimensional mass spectrometry to visualize the spatial distribution of biomolecules, which does not need either separation or purification of target molecules, and enables us to monitor not only the identification of unknown molecules but also the localization of numerous molecules simultaneously. Among the ionization techniques, matrix assisted laser desorption/ionization (MALDI) is one of the most generally used for IMS, which allows the analysis of numerous biomolecules ranging over wide molecular weights. Proper selection and preparation of matrix is essential for successful imaging using IMS. Tandem mass spectrometry, which is referred to MSn, enables the structural analysis of a molecule detected by the first step of IMS. Applications of IMS were initially developed for studying proteins or peptides. At present, however, targets of IMS research have expanded to the imaging of small endogenous metabolites such as lipids, exogenous drug pharmacokinetics, exploring new disease markers, and other new scientific fields. 油松是中国特有的树种,具有重要的造林和绿化价值。但是油松胚珠败育是油松种子生产和繁殖面临的一个严重的问题,因此研究油松胚珠的发育机理和败育原因具有重要的理论和实践意义。双向电泳和质谱技术结合研究基因表达的蛋白质组技术正越来越广泛地应用到植物研究中,我们将该技术应用到油松胚珠发育的研究中,比较了两种蛋白质提取的方法,发现蔗糖提取方法比传统的TCA方法更适合于油松蛋白质的制备。将电泳分离得到的蛋白点用肽质量指纹图谱的方法进行鉴定,得到了满意的结果,优化了蛋白质质谱鉴定的条件。  相似文献

Abstract

We aimed to find new physiological effects of the Japanese diet. First, to determine the key components in serum from mice fed the 1975 diet, serum from mice fed the 1960, 1975, 1990 or 2005 Japanese diet was analyzed using CE-TOFMS and LC-TOFMS. Based on these results, the key components were determined by principal component analysis. Among the identified compounds, GABA was included. Therefore, a stress reduction effect was inferred as a novel physiological effect of this diet. Next, we tested whether the 1975 diet had an actual stress reduction effect in mice. Mice were given the 1975 diet or a control diet for 4 weeks, after which they were divided into restraint stress and non-stress groups. Mice fed the 1975 diet had significantly decreased stress parameters compared with those fed the control diet. These results provide the first evidence that the 1975 Japanese diet has a stress reduction effect.  相似文献

Accurate age estimations are essential to archaeological and forensic analyses. However, reliability for adult skeletal age estimations is poor, especially for individuals over the age of 40 years. This is the first study to show that body size influences skeletal age estimation. The ??can et al., Lovejoy et al., Buckberry and Chamberlain, and Suchey‐Brooks age methods were tested on 764 adult skeletons from the Hamann‐Todd and William Bass Collections. Statures ranged from 1.30 to 1.93 m and body masses ranged from 24.0 to 99.8 kg. Transition analysis was used to evaluate the differences in the age estimations. For all four methods, the smallest individuals have the lowest ages at transition and the largest individuals have the highest ages at transition. Short and light individuals are consistently underaged, while tall and heavy individuals are consistently overaged. When femoral length and femoral head diameter are compared with the log‐age model, results show the same trend as the known stature and body mass measurements. The skeletal remains of underweight individuals have fewer age markers while those of obese individuals have increased surface degeneration and osteophytic lipping. Tissue type and mechanical loading have been shown to affect bone turnover rates, and may explain the differing patterns of skeletal aging. From an archaeological perspective, the underaging of light, short individuals suggests the need to revisit the current research consensus on the young mortality rates of past populations. From a forensic perspective, understanding the influence of body size will impact efforts to identify victims of mass disasters, genocides, and homicides.
