The occurrence pattern of the marsh fritillary was studied within a patch network on the Baltic island Öland, Sweden. Presence/absence was established for potentially suitable habitat patches (n = 158) on calcareous moist grassland and analyzed in a multiple logistic regression model where patch area, patch isolation and nine habitat quality variables were included as explanatory variables. Larval food plant density was positively, and patch isolation negatively, correlated to the presence of Euphydryas aurinia. Area did not contribute to the explanation of the occurrence pattern. Significant interactions between larval food plant density times patch isolation, and larval food plant density times vegetation height, show that with low food plant density the butterfly primarily occurs in patches with a vegetation height of 4–10 cm, within a distance of 250 m from nearest occupied patch. In patches with a high food plant density the butterfly occurs in patches where the vegetation height is higher, 4–16 cm, and the distance to nearest occupied patch can be longer, up to 1.4 km. This study supports earlier findings in other regions, suggesting that a network of adjacent patches with a high food plant density and a vegetation height within the preferred threshold, despite their size, is an apparent conservation goal.  相似文献

In this study, silk fibroin nanofibrous scaffolds were developed to investigate the attachment and proliferation of primary human meniscal cells. Silk fibroin (SF)–polyvinyl alcohol (PVA) blended electrospun nanofibrous scaffolds with different blend ratios (2:1, 3:1, and 4:1) were prepared. Morphology of the scaffolds was characterized using atomic force microscopy (AFM). The hybrid nanofibrous mats were crosslinked using 25 % (v/v) glutaraldehyde vapor. In degradation study, the crosslinked nanofiber showed slow degradation of 20 % on weight after 35 days of incubation in simulated body fluid (SBF). The scaffolds were characterized with suitable techniques for its functional groups, porosity, and swelling ratio. Among the nanofibers, 3:1 SF:PVA blend showed uniform morphology and fiber diameter. The blended scaffolds had fluid uptake and swelling ratio of 80 % and 458 ± 21 %, respectively. Primary meniscal cells isolated from surgical debris after meniscectomy were subcultured and seeded onto these hybrid nanofibrous scaffolds. Meniscal cell attachment studies confirmed that 3:1 SF:PVA nanofibrous scaffolds supported better cell attachment and growth. The DNA and collagen content increased significantly with 3:1 SF:PVA. These results clearly indicate that a blend of SF:PVA at 3:1 ratio is suitable for meniscus cell proliferation when compared to pure SF-PVA nanofibers.  相似文献

Herbivorous insects are commonly faced with host plants being distributed in scattered patches across a landscape. Immigration rates into habitat patches may strongly depend on the sensory cues used in the patch location process, and immigration rates of insects can be predicted based on the scaling of sensory cues. Here, we tested recent estimates of the scaling of olfactory information to patch size, which predicts a scaling coefficient ζ ≈ ?0.5 (A ζ , where A = patch size, ζ = scaling coefficient). We predicted that immigration rates of olfactory searching insects into patches of different sizes should scale according to the estimated slope. We investigated attraction of the weevils Cionus tuberculosus and Cionus scrophulariae to odors from figwort Scrophularia nodosa and quantified immigration rates of weevils into differently sized patches. We also investigated oviposition rates of the sawfly Tenthredo scrophulariae. The slope in the regression between density and patch size for herbivores was then compared with the predicted scaling coefficient. Using olfactometers, we found that weevils were attracted to figwort odors. Weevil densities were significantly affected by patch size, and the slope in the relationship between density and patch size was ζ = ?0.53. The slope in the relationship between larval densities of sawflies and patch size was less negative with a slope of ζ = ?0.15, indicating differences in search behavior compared with the weevils. The density–patch size relationship for the weevils closely matched the predicted slope and supported the previous estimations of the scaling of olfactory information to patch size.  相似文献

Abstract

To fully hydrate patches of Crossidium crassinerve in the Mojave Desert, a rain event of at least 2.0 mm was required. When patches were hydrated for at least 3 days, mean daily ambient maximum/minimum temperatures were only 15.6/7.5°C. During the summer, patch temperatures exceeded ambient temperatures, whereas during the cooler months patch temperatures were lower than ambient temperatures, with degree of patch hydration not a factor in patch temperature. During patch hydration, mean surface patch temperature ranged from ?3 to 14°C. With few exceptions, patch hydroperiod (duration of patch hydration) was restricted to the cooler months spanning October to April, with the mean patch hydroperiod (among five patches) ranging from 3.7 to 4.9 days. While the most commonly recorded dry period was <25 days, Crossidium patches experienced longer periods of desiccation on nine occasions, with the longest period being 191 days. During a late winter rain event, patches dried slowly over a period of several days, whereas during a summer rain event, patches dried in as few as 3 h. Over the 4-year period, 248 sporophytes were initiated, with all but four of these from two patches. Only nine of these initiated sporophytes survived to disperse spores, with mortality attributed to embryonic abortion (69%) and capsule herbivory (30%).  相似文献

Forests are naturally extensive tracts. However, in South Africa natural fires over many millennia have reduced forested areas into small remnants spread throughout a grassland matrix. Small patches, especially distant patches, are generally considered to be adverse for forest specialists, owing to decreased forest interior and increased edge. Here we test this assumption by determining the impact of forest interpatch distance and patch size on epigaeic arthropod diversity in this globally rare vegetation type. Forty patches were selected: ten large (100–435 m diameter) that are distant (500–645 m) from other patches, ten large that are close to other patches (38–97 m), ten small (30–42 m) that are distant, and ten small-close patches. Each patch had two plots: edge and interior. Arthropods were sampled using pitfall traps, Berlese-Tullgren funnels and active searches. Interiors and edges had similar species richness and composition, excluding spiders, which were richer in interiors. Patch size significantly influenced species richness of predatory beetles and arthropod assemblages, excluding spiders. Effect of the interaction between patch size and interpatch distance on species richness and composition varied among taxa. Furthermore, large patches supported similar assemblages regardless of interpatch distance. Arthropod response, particularly ants to patch size and interpatch distance, was partly shaped by the matrix type. The percentage of surrounding grassland had little effect on arthropod diversity. We can conclude that large and close patches are important for arthropod conservation. Nevertheless, it is also important to conserve a variety of patch sizes at various distances to maximize overall arthropod composition.  相似文献

Vascular calcification is a prominent feature of many diseases, including atherosclerosis, and it has emerged as a powerful predictor of cardiovascular morbidity and mortality. A number of studies have examined the association between selenium and risk of cardiovascular diseases, but little is known about the role of selenium in vascular calcification. To determine the role of selenium in regulating vascular calcification, we assessed the effect of sodium selenite on oxidative-stress-enhanced vascular smooth muscle cell (VSMC) calcification and the underlying mechanism. Oxidative stress induced by xanthine/xanthine oxidase increased apoptosis, as determined by Hoechst 33342 staining and annexin V/propidium iodide staining, and it enhanced osteoblastic differentiation and calcification of VSMCs, on the basis of alkaline phosphatase activity, the expression of Runx2 and type I collagen, and calcium deposition. These effects of oxidative stress were significantly inhibited by selenite. The following processes may explain the inhibitory effects of selenite: (1) selenite significantly suppressed oxidative stress, as evidenced by the decrease of the oxidative status of the cell and lipid peroxidation levels, as well as by the increase of the total protein thiol content and the activity of the antioxidant selenoenzyme glutathione peroxidase; (2) selenite significantly attenuated oxidative-stress-induced activation of the phosphatidylinositol 3-kinase/AKT and extracellular-signal-regulated kinase signaling pathways, resulting in decreased osteoblastic differentiation of VSMCs; (3) selenite significantly inhibited oxidative-stress-activated endoplasmic reticulum stress, thereby leading to decreased apoptosis. Our results suggest a potential role of selenium in the prevention of vascular calcification, which may provide more mechanistic insights into the relationship between selenium and cardiovascular diseases.  相似文献

Effective repair of a vascular injury depends on establishment of a stable fibrin patch at the injury site. Data presented in this study demonstrate that structural modification of fibrin occurs as a result of fibrin interaction with naturally occurring components of the vascular basement membrane and subendothelial structures. Of the basement membrane components, type IV collagen produces the greatest structural modification, generating thick fibrin fibers; a 3-fold increase in the fiber mass/length ratio occurs when type IV collagen is increased from 0 to 100 ng/ml. Laminin and dermatan sulfate decrease the fibrin fiber mass/length ratio resulting in thinner fibers. However, the overall effect of the basement membrane on fibrin is to increase the fibrin fiber diameter. Electrophoretic light scattering and the binding of type IV collagen by fibrinogen-Sepharose further establish the interaction between type IV collagen and fibrinogen. Incorporation of laminin with type IV collagen onto coated surfaces decreases the ability of type IV collagen to bind fibrinogen. These studies emphasize that the final fibrin structure is influenced by the milieu in which the clot is assembled.  相似文献

Resveratrol-phospholipid complex (Phytosome®) (RSVP) was found better aqueous soluble and permeable than free resveratrol (RSV). RSVPs were incorporated in polymeric patch prepared by solvent casting method using Eudragit RL 100, PVP K30, and PEG 400 for application on dermal sites for sustained treating of inflammation. Prepared patches were evaluated for various physicochemical properties, surface morphology by SEM, TEM, and compatibility of patch components by FT-IR and DSC studies. Optimized formulation (F9) gave 95.79?±?3.02% drug release and 51.36% (4.28?±?0.48 mg/cm2) skin permeation after 24 h. Skin extract when examined for drug accumulation showed 38.31?±?2.42% drug content. FE-SEM images of the patch taken after drug release and skin permeation studies showed that RSVPs in polymeric patch are stable and retain their structure after 24 h long exposure to physiologic environment. Sustained anti-inflammatory effect was established in carrageenan-induced paw edema model in which test formulation gave 84.10% inhibition of inflammation at 24 h as compared to 39.58% for standard diclofenac sodium gel. The CLSM study confirmed the localization of RSVPs for a longer period, thus enabling drug targeting to the dermis for sustained effect. Skin irritation test on rabbit revealed that the patches are safe for skin application. Histological observations suggested that after exposure to the permeants, the SC integrity had not altered and no evidence of presence of inflammatory cells found. RSVP (Phytosome®) containing patches abled to give sustained therapeutic effect that may be useful in treating acute and chronic inflammation.  相似文献

Honey bees (Hymenoptera: Apidae) were used as a model insect system to explore forager use of a learned color-cue memory over several subsequent days. Experiments used artificial flower patches of blue and white flowers. Two experiments were performed, each beginning with a learning experience where 2 M sucrose was present in one flower color and 1 M sucrose in the alternative flower color. The first experiment followed flower color fidelity over a series of sequential days when rewards no longer differed between flowers of different color. The second examined the effect of intervening days without the forager visiting the flower patch. Results showed that color-cue memory decline was not a passive time-decay process and that information update in honey bees does not occur readily without new experiences of difference in rewarding flowers. Further, although the color cue learned was associated with nectar reward in long term memory, it did not seem to be specifically associated with the 2 M sucrose nectar reward when intervening nights occurred between learning and revisiting the flower patch.  相似文献

The current study aimed to enhance the efficacy of peripheral nerve regeneration using a biodegradable porous neural guidance conduit as a carrier to transplant allogeneic Schwann cells (SCs). The conduit was prepared from polyurethane (PU) and gelatin nanofibrils (GNFs) using thermally induced phase separation technique and filled with melatonin (MLT) and platelet-rich plasma (PRP). The prepared conduit had the porosity of 87.17 ± 1.89%, the contact angle of 78.17 ± 5.30° and the ultimate tensile strength and Young’s modulus of 5.40 ± 0.98 MPa and 3.13 ± 0.65 GPa, respectively. The conduit lost about 14% of its weight after 60 days in distilled water. The produced conduit enhanced the proliferation of SCs demonstrated by a tetrazolium salt-based assay. For functional analysis, the conduit was seeded with 1.50 × 104 SCs (PU/GNFs/PRP/MLT/SCs) and implanted into a 10-mm sciatic nerve defect of Wistar rat. Three control groups were used: (1) PU/GNFs/SCs, (2) PU/GNFs/PRP/SCs, and (3) Autograft. The results of sciatic functional index, hot plate latency, compound muscle action potential amplitude and latency, weight-loss percentage of wet gastrocnemius muscle and histopathological examination using hematoxylin–eosin and Luxol fast blue staining, demonstrated that using the PU/GNFs/PRP/MLT conduit to transplant SCs to the sciatic nerve defect resulted in a higher regenerative outcome than the PU/GNFs and PU/GNFs/PRP conduits.  相似文献

Connectivity of habitat patches is crucial for wildlife dispersal and survival, and identifying patches with high importance for maintaining connectivity can aid effective wildlife management. Knowledge of the habitat distribution of the Tibetan antelope in the Altun Mountain National Nature Reserve, which is essential for connectivity analysis, remains limited. We analyzed potential habitat distribution and priority patches using GIS-based habitat suitability modeling with three weighting factors and evaluated the connectivity of habitat patches under four dispersal distance scenarios. Patches with high habitat suitability covered 25.39 % of the total area, and these patches were selected for connectivity analysis as resource patches. Connectivity analysis indicated that, although the overall probability of connectivity (PC) showed an upward trend with increasing dispersal distance, the importance of each patch varied considerably under different dispersal distance scenarios. Transfer analysis of patch numbers between different importance levels revealed that the number of patches becoming less important was higher than the number of patches becoming more important when dispersal distance increased. In addition, nine patches covering 38.49 % of the suitable habitat area were identified as priority patches, in particular the patches near the Kardun and the Karchuka inspection stations. We also found that the habitat distribution of the Tibetan antelope obtained from the suitability model matched the population distribution determined by a field survey. Correlation analysis between patch area and the percentage of PC index value loss (dPC) revealed that the larger patches in this region were more likely to be important for maintaining connectivity.  相似文献

Little is known about the real incidence and the clinical relevance of the enigmatic Monckeberg’s medial calcification in the patency of the femoral artery allograft. Here we present a retrospective study on 143 multiorgan donors (mean age 38 years, range 14–59 years), to describe the incidence and the morphological features of vascular calcifications in banked femoral arteries suitable for clinical use. In the present series, focal vascular calcifications were present in 36 (25 %) cases, 23 cases localized in the intima, 7 in the media, and 6 were mixed. No correlation was found between the incidence of calcifications and the classical cardiovascular clinical risk factors (n = 9); only hypertension correlated with the medial localization, but not with the incidence, of the calcification (P = 0.017). While the macroscopic exclusion criteria of vascular grafts include atheromatous and not-atheromatous lesions, we ignore the actual impact of Monckeberg’s medial calcification on vessel transplantation and allograft life. In our opinion this is a very important topic, since when the histological criteria for Monckeberg’s calcification diagnosis are used, 25 % of our young donors population was affected. Whether Monckeberg’s medial calcification is a stable arterial condition, apparently underestimated in the general population, or a dynamic process evolving with age and atherosclerosis, or a banking-related vascular alteration, still remain an open issue deserving further studies with subjects of different ages.  相似文献

Cell-attached and inside-out patch clamp recording was used to compare the functional expression of membrane ion channels in mouse and human embryonic stem cells (ESCs). Both ESCs express mechanosensitive Ca2+ permeant cation channels (MscCa) and large conductance (200 pS) Ca2+-sensitive K+ (BKCa2+) channels but with markedly different patch densities. MscCa is expressed at higher density in mESCs compared with hESCs (70 % vs. 3 % of patches), whereas the BKCa2+ channel is more highly expressed in hESCs compared with mESCs (~50 % vs. 1 % of patches). ESCs of both species express a smaller conductance (25 pS) nonselective cation channel that is activated upon inside-out patch formation but is neither mechanosensitive nor strictly Ca2+-dependent. The finding that mouse and human ESCs express different channels that sense membrane tension and intracellular [Ca2+] may contribute to their different patterns of growth and differentiation in response to mechanical and chemical cues.  相似文献

Synovial fibroblasts (SF) contribute to the pathogenesis of osteoarthritis (OA), but the effects of intra-articular cytokines on SF are not completely understood. The aim of this study was to characterize the interplay between tumor necrosis factor (TNF)α and the anti-inflammatory interleukin (IL)-10. Non-immortalized human SF and SF of the human cell line K4IM were stimulated with recombinant TNFα, IL-10, or TNFα?+?IL-10 (10 ng/ml each) for 24 h or transduced with an adenoviral vector overexpressing human IL-10 (hIL-10) and subsequently treated with 10 ng/ml TNFα for 24 h. Effects on the gene expression and protein synthesis of IL-6, IL-10, matrix metalloproteinases (MMP)-1, ?3, type I collagen, β1-integrin, and CD44 were investigated via real-time detection polymerase chain reaction, immunofluorescence labeling, flow cytometry, and Western blotting. IL-10 release by transduced SF was confirmed with enzyme-linked immunosorbent assay. Both cell populations were activated by TNFα and by TNFα?+?IL-10, increasing their gene expression and protein synthesis of IL-6, IL-10, MMP-1, and MMP?3 and altering the synthesis of type I collagen, β1-integrin, and CD44. hIL-10 overexpression greatly elevated the gene expression and protein synthesis of IL-10. However, transduction did not significantly affect the gene expression of IL-6, MMP-1, and MMP?3 in SF. The increased expression of pro-inflammatory and catabolic mediators in TNFα-activated SF indicates their role in OA pathogenesis, suggesting they are a potential therapeutic target. Although the vigorousness of the responses of non-immortalized SF and K4IM clearly differ, the K4IM cell line seems to be a suitable model for non-immortalized human SF.  相似文献

For hydrogel patches, the laboratory tests could not fully reveal the existing problems of full scale of industrial production, and there are few studies about the preparation technique for the industrial manufacturing process of hydrogel patches. So, the purpose of this work was to elucidate the effects of mainly technological operation and its parameters on the performance of hydrogel patches at the industrial-scale production. The results revealed the following: (1) the aqueous phase was obtained by polyvinylpyrrolidone (PVP) along with tartaric acid dissolved in purified water, then feeding this into a vacuum mixer as a whole in one batch, thus extended the crosslinking reaction time of hydrogel paste (matrix) and allowed the operation of coating/cutting-off to be carried out easily, and there was no permeation of backing layer; (2) the gel strength of the hydrogel patches increased with the increase of working temperature, however, once the temperature exceeded 35 ± 2 °C, the hydrogel paste would lose water severely and the resultant physical crosslinking structure which has lower gel/cohesive strength would easily bring gelatinization/residues during application; (3) the relative humidity (RH) of the standing-workshop was dynamically controlled (namely at 35 ± 2 °C, keeping the RH at 55 ± 5% for 4 days, then 65 ± 5% for 2 days), which would make patches with satisfactory characteristics such as better flexibility, higher adhesive force, smooth flat matrix surface, and without gelatinization/residues and warped edge during the using process; (4) the aging of the packaged hydrogel patches was very sensitive to storage temperature, higher temperature, higher gel strength and lower adhesiveness. The storage temperature of 10 ± 2 °C could effectively prevent matrix aging and adhesion losing, which would also facilitate the expiration date of patches extended obviously. In conclusion, this work provides an optimized and feasible preparation technique for the industrial production of the hydrogel patches and establishes the hydrogel patches as a novel carrier for transdermal drug delivery.  相似文献

Uncontrolled urban development is threatening the survival of many terrestrial species worldwide, especially those with limited dispersal capabilities. Soil invertebrates account for a high proportion of the global biodiversity but there are few studies on how soil biodiversity varies with different parameters of landscape structure, especially in fragmented tropical habitats. Millipedes are among the most abundant detritivore invertebrates in the soil-litter macrofauna. We examined the relationships between soil millipede diversity and landscape structure in 12 forest patches of approximately 30 years of age, in northeastern Puerto Rico. Spatial characteristics of the landscape were determined from aerial color photographs and were digitized into a GIS package for analysis. Millipede species diversity and composition in these forest patches showed correlations with their surroundings (e.g. amount of forest in the matrix) and with the presence of vegetation corridors that connected to other forest patches, rather than forest patch attributes such as patch area and shape. Millipede species richness correlated negatively with the degree of isolation of forest patch (within 600 m radius), while species evenness correlated positively to the amount of forest within a 50 m buffer. Millipede species composition was related with the presence of vegetation corridors and the distance to the Luquillo Experimental Forest reserve. These findings show that a low degree of patch isolation, forested buffers, and presence of vegetation corridors need to be considered for the conservation and management of forest patches surrounded by urban developments, especially to protect terrestrial invertebrate species that require forested habitats for their dispersal.  相似文献

Aim

We studied the legacy effects of shrubs during the downgrade phase of high–cover patches. Specifically, are woody species able to modify environmental attributes at patch level to such an extent as to alter the colonization once they have vacated their original position?

Methods

We monitored five environmental variables along an experimental four-stage downgrading gradient of high–cover patches during two years in cold- and warm-seasons, individual plant growth during three years, as well as the floristic composition of patches along the same gradient after 13 years.

Results

The downgrade of high–cover patches reduces the aboveground protection due to the increase in wind speed (400–500%) and evaporation rate (43–160%) associated with shrub death and senescence. In addition, high–cover patches increase the total soil nitrogen (400–600%) and reduce the infiltration rate (44–73%) on the top layer. Leaf length and flower culms of grass tussocks were lower in bare soil patches (7.5 cm and 3) compared to whatever degradation stage of high–cover patches (9–10 cm and 18–32). Floristic composition after 13 years reveals that grass species occupied the patch stages differentially, with a disjunctive pattern among species within the Poa and Pappostipa genus.

Conclusions

Legacy effects prompted by shrubs through changes in soil properties at the horizontal plane can conditioned the patch dynamics. The ability of different plant species to cope with the spatial heterogeneity at the horizontal plane should be included as a new criterion to define plant strategies from arid ecosystems according to the gap–phase dynamics and mosaic maintenance.

The aims of this study were to develop a biological large diameter vascular graft by decellularisation of native human aorta to remove the immunogenic cells whilst retaining the essential biomechanical, and biochemical properties for the ultimate benefit of patients with infected synthetic grafts. Donor aortas (n = 6) were subjected to an adaptation of a propriety decellularisation process to remove the cells and acellularity assessed by histological analysis and extraction and quantification of total DNA. The biocompatibility of the acellular aortas was determined using standard contact cytotoxicity tests. Collagen and denatured collagen content of aortas was determined and immunohistochemistry was used to determine the presence of specific extracellular matrix proteins. Donor aortas (n = 6) were divided into two, with one half subject to decellularisation and the other half retained as native tissue. The native and decellularised aorta sections were then subject to uniaxial tensile testing to failure [axial and circumferential directions] and suture retention testing. The data was compared using a paired t-test. Histological evaluation showed an absence of cells in the treated aortas and retention of histoarchitecture including elastin content. The decellularised aortas had less than 15 ng mg?1 total DNA per dry weight (mean 94% reduction) and were biocompatible as determined by in vitro contact cytotoxicity tests. There were no gross changes in the histoarchitecture [elastin and collagen matrix] of the acellular aortas compared to native controls. The decellularisation process also reduced calcium deposits within the tissue. The uniaxial tensile and suture retention testing revealed no significant differences in the material properties (p > 0.05) of decellularised aorta. The decellularisation procedure resulted in minimal changes to the biological and biomechanical properties of the donor aortas. Acellular donor aorta has excellent potential for use as a large diameter vascular graft.  相似文献
