Waste-grown microalgae are a potentially important biomass for wastewater treatment. The lipid accumulated in microalgae could be utilized as feedstocks for biodiesel production. The algal residues, as major by-products derived from lipid extraction, mainly consist of carbohydrate and protein, making anaerobic digestion an efficient way to recover energy. The conversion of lipid-extracted algal residues into methane plays dual role in renewable energy production and sustainable development of microalgal biodiesel industry. Therefore, an anaerobic fermentation process for investigation of the methane production potential of algal residues was conducted in this paper. The effect of inoculum to substrate ratios (ISRs) on the methane production by anaerobic digestion of Chlorella sp. residue in a single stage was evaluated. The maximum methane yield of 195.6 ml CH4/g volatile solid (VS) was obtained at an ISR of 1:1. The stability and progress of the reaction from algal residues to methane were monitored by measuring the pH, volatile fatty acids (VFAs), total ammoniacal nitrogen (TAN), and methane volume. Based on the results of one-stage experiments, two-stage technology was proposed and was found to be more suitable for high organic load. The optimum conditions for acidogenesis and methanogenesis are indicated in this paper.  相似文献

Acidogenesis of cattail using rumen cultures was carried out to produce volatile fatty acids (VFA) in this study. The influences of pH and substrate concentration on cattail degradation, VFA yield and microbial growth were investigated by using response surface methodology (RSM). Experimental results showed that a low substrate concentration and pH of 6.9 were optimal for acidogenesis of cattail. The highest cattail degradation efficiency, VFA yield and microbial yield were 75.9%, 0.41 g/g VS and 0.110 g/g VS, respectively. Further experiments confirmed that the main VFA in the acidogenesis of cattail were acetate, propionate and butyrate, while i-butyrate, valerate and i-valerate were also produced at low levels. The results suggested that acidogenesis using rumen cultures is a promising method for cattail disposal.  相似文献

In anaerobic processes, the population dynamics of methanogens in the methanogenic stage were monitored along with hydraulic retention times (HRTs) shift. Decreasing HRTs increased the loading rates of volatile fatty acids (VFAs) and ammonia. Methanomicrobiales (MMB) began to be dominant at longer than 12.5 days HRT, Methanosarcinales (MSL) were dominant at 8, 10, and 12.5 days HRT, and Methanobacteriales (MBT) were dominant at shorter than 6 days HRT. Increased loading rates of VFAs and ammonia increased MBT, decreased MMB, and had no significant effect on MSL. Maximal daily methane production was observed at 1.57 L/L when MSL copy numbers also reached 3.60 × 107 copy/mL as a peak, which were expressed as positive correlation between DMA and MSL. No sooner had methane yield (MY) increased from 1.15 to 1.32 L/g VSremoved along with HRT reduction from 25 to 22.5 days, then MY gradually decreased from 1.32 to 0.04 L/g VSremoved.  相似文献

A two step biological process for the conversion of grass biomass to the biodegradable polymer medium chain length polyhydroxyalkanoate (mcl-PHA) was achieved through the use of anaerobic and aerobic microbial processes. Anaerobic digestion (mixed culture) of ensiled grass was achieved with a recirculated leach bed bioreactor resulting in the production of a leachate, containing 15.3 g/l of volatile fatty acids (VFAs) ranging from acetic to valeric acid with butyric acid predominating (12.8 g/l). The VFA mixture was concentrated to 732.5 g/l with a 93.3 % yield of butyric acid (643.9 g/l). Three individual Pseudomonas putida strains, KT2440, CA-3 and GO16 (single pure cultures), differed in their ability to grow and accumulate PHA from VFAs. P. putida CA-3 achieved the highest biomass and PHA on average with individual fatty acids, exhibited the greatest tolerance to higher concentrations of butyric acid (up to 40 mM) compared to the other strains and exhibited a maximum growth rate (μMAX?=?0.45 h?1). Based on these observations P. putida CA-3 was chosen as the test strain with the concentrated VFA mixture derived from the AD leachate. P. putida CA-3 achieved 1.56 g of biomass/l and accumulated 39 % of the cell dry weight as PHA (nitrogen limitation) in shake flasks. The use of volatile fatty acids (VFAs) for microbial lipid accumulation was investigated in flask cultures of Cryptococcus albidus. The optimum culture temperature and pH were 25 °C and pH 6.0, respectively, and the highest lipid content (27.8%) was obtained with ammonia chloride as a nitrogen source. The lipid yield coefficient on VFAs was 0.167 g/g of C. albidus with a VFAs (acetic, propionic, butyric acids) ratio of 8:1:1, which was in good agreement with a theoretically predicted lipid yield coefficient of the VFAs as a carbon source. The major fatty acids of the lipids accumulated by C. albidus were similar to those of soybean oil and jatropha oil. A preliminary cost analysis shows that VFAs-based biodiesel production is competitive with current palm and soybean based biodiesels. Further process development for lower aeration cost and higher lipid yield will make this process more economical.  相似文献

Batch experiments were carried out on anaerobic digestion of swine manure under 10 % of total solids and 60 g/L of zeolite addition at 35 °C. Four distinctive volatile fatty acid (VFAs) evolution stages were observed during the anaerobic process, i.e., VFA accumulation, acetic acid (HAc) and butyric acid (HBu) utilization, propionic acid (HPr) and valeric acid (HVa) degradation, and VFA depletion. Large decreases in HAc/HBu and HPr/HVa occurred respectively at the first and second biogas peaks. Biogas yield increased by 20 % after zeolite addition, about 356 mL/g VSadded with accelerated soluble chemical oxygen demand degradation and VFA (especially HPr and HBu) consumption in addition to a shortened lag phase between the two biogas peaks. Compared with Ca2+ and Mg2+ (100–300 mg/L) released from zeolite, simultaneous K+ and NH4 + (580–600 mg/L) adsorptions onto zeolite particles contributed more to the enhanced biogasification, resulting in alleviated inhibition effects of ammonium on acidogenesis and methanogenesis, respectively. All the identified anaerobes could be grouped into Bacteroidetes and Firmicutes, and zeolite addition had no significant influence on the microbial biodiversity in this study.  相似文献

In this investigation, the effect of pH (4.0–11.0) on waste-activated sludge (WAS) hydrolysis and acidification in the presence of a biosurfactant rhamnolipid (RL) were studied. The results showed that the hydrolysis and acidification of WAS in the presence of RL at alkaline pH values were more efficient than that at acidic and near-neutral pH values. After 6 h of hydrolysis, the soluble protein and carbohydrate were 1,654.7 and 675.9 mg/L (pH 11.0), and 825.6 and 376.0 mg/L (pH 7.0), whereas the values were only 315.0 and 84.0 mg/L at pH 4.0 and 164.1 and 32.0 mg/L for the blank, respectively. After 2 or 3 days of fermentation, the accumulated short-chain fatty acids (SCFAs) reached the highest and then decreased with a further increase in time at all investigated pH values. The analysis of SCFA compositions showed that acetic, propionic, and iso-valeric acids were the three main products at any pH value. A higher pH contributed to a greater proportion of acetic acid and a lesser proportion of iso-valeric acid; a lower pH resulted in a greater proportion of iso-valeric and lesser proportion of acetic acid in the initial fermentation. The proportions of acetic acid for the system with biosurfactant RL addition were 16.65, 36.33, and 62.94 %, respectively, at pH 4.0, 7.0, and 11.0 after 1 day. Correspondingly, the proportions were 40.34, 12.60, and 11.01 % for iso-valeric acid.  相似文献

Hydrogen and volatile fatty acids (VFAs) were coproduced from marine macroalgae by anaerobic fermentation using a microbial community. The hydrogen and VFAs production were characterized based on inoculum heat-treatment, methanogen inhibitor addition, operating temperature, and in-situ extraction of VFAs. Maximum hydrogen of 179 mL/g-VS and VFAs concentration of 9.8 g/L were produced from 35 g/L of S. japonica within 5 days of anaerobic fermentation. Hydrogen and VFAs yields were well-correlated with carbohydrate content of substrate. Inoculum heat-treatment significantly improved hydrogen production while the VFAs productivity was affected adversely. The addition of methanogen inhibitors also enhanced the hydrogen production, but the effect on VFAs production was dependent on the type of inhibitor used. Low temperature (25°C) was found to be favorable for high hydrogen and VFAs yield, while high temperature (40°C) and programmed-temperature (35 ~ 45°C) were more favorable for hydrogen and VFAs productivity. Clostridium sp. content was found to be the most abundant at 25°C. An extractive fermentation with anion-exchange resin was tested to recover the VFAs and to control the pH during the anaerobic fermentation.  相似文献

The effluent from a continuous-flow stirred tank reactor (CSTR) treating synthetic wastewater was used as an alternative carbon source to glucose for algal biomass and biodiesel production. The influence of the effluent on microalgal cell growth and lipid yield, as well as the utilization of volatile fatty acids (VFAs) in the effluent were investigated. The results indicate that C. protothecoides can proliferate in the CSTR effluent and accumulate biolipid. The final lipid content of the culture with the effluent feeding was 27?±?1.11 % after 168 h cultivation in flasks, which was higher than that with glucose of the same COD concentration. Valeric acid, ethanol, and butyric acid were favorable carbon sources for cell growth. The soluble microbial products (SMP) can be used as a carbon source for cell growth, and the existence of SMP could protect the cell from the inhibition caused by strong VFAs and improve the utilization of VFAs.  相似文献

Bacterial fermentation of lignocellulose has been regarded as a sustainable approach to butyric acid production. However, the yield of butyric acid is hindered by the conversion efficiency of hydrolysate xylose. A mesophilic alkaline-tolerant strain designated as Clostridium butyricum B10 was isolated by xylose fermentation with acetic and butyric acids as the principal liquid products. To enhance butyric acid production, performance of the strain in batch fermentation was evaluated with various temperatures (20–47 °C), initial pH (5.0–10.0), and xylose concentration (6–20 g/L). The results showed that the optimal temperature, initial pH, and xylose concentration for butyric acid production were 37 °C, 9.0, and 8.00 g/L, respectively. Under the optimal condition, the yield and specific yield of butyric acid reached about 2.58 g/L and 0.36 g/g xylose, respectively, with 75.00% butyric acid in the total volatile fatty acids. As renewable energy, hydrogen was also collected from the xylose fermentation with a yield of about 73.86 mmol/L. The kinetics of growth and product formation indicated that the maximal cell growth rate (μ m ) and the specific butyric acid yield were 0.1466 h?1 and 3.6274 g/g cell (dry weight), respectively. The better performance in xylose fermentation showed C. butyricum B10 a potential application in efficient butyric acid production from lignocellulose.  相似文献

Batch experiments were performed to investigate the influence of cellulose particle size and pH on the anaerobic degradation of crystalline cellulose by ruminal microbes. At a particle size of 50 μm there was a higher hydrolysis and acidogenesis rate, and a reduced degradation time, than for 100-μm particles. Reduction in cellulose particle size resulted in decreased methane production, but an increase of soluble products. Cellulose degradation increased with pH from pH 6.0 to 7.5, whereas at pH⩽5.5 there was no degradation. The inhibitory effect of low pH (⩽5.5) on ruminal microbes was not completely remedied even when the pH of the medium was adjusted to a neutral range. In an anaerobic cellulosic waste degrading system inoculated with ruminal microbes the fermentation system should therefore be maintained above pH 6.0. In all cases, volatile fatty acids were the major water-soluble products of cellulose degradation; acetate and propionate accounted for more than 90% of the volatile fatty acid total.  相似文献

It is generally accepted that short (C2-C5) and medium (C6-C11) chain volatile fatty acids (VFAs) are among the primary causal molecules of axillary malodour. It is also widely acknowledged that malodour generation is attributable to the biotransformation of odourless natural secretions, into volatile odorous products, by cutaneous bacteria. However, little information is available on the biochemical origins of VFAs on axillary skin. In these studies, assay systems were developed to investigate the generation of VFAs from lipid substrates readily available to the bacteria resident on axillary skin. A major route to short and medium chain VFAs in the axilla was shown to be the partial catabolism of structurally unusual (e.g. methyl-branched) longer chain fatty acids by a previously uncharacterized sub-group of the Corynebacterium genus, corynebacteria (A). In contrast, corynebacteria (B) are incapable of growth on fatty acid. Structurally unusual fatty acids originate from the triacylglycerol component of sebum, and probably also apocrine sweat, by the action of bacterial lipases. Interestingly, VFA formation in the axilla is a dynamic process, with some cutaneous microorganisms, specifically micrococci and brevibacteria, capable of fully catabolizing these odorants. The results of these studies provide new understanding on the biochemical origins of VFA-based axillary malodour.  相似文献

Volatile fatty acids (VFAs) that can be derived from food wastes were used for microbial lipid production by Chlorella protothecoides in heterotrophic cultures. The usage of VFAs as carbon sources for lipid accumulation was investigated in batch cultures. Culture medium, culture temperature, and nitrogen sources were explored for lipid production in the heterotrophic cultivation. The concentration and the ratio of VFAs exhibited significant influence on cell growth and lipid accumulation. The highest lipid yield coefficient and lipid content of C. protothecoides grown on VFAs were 0.187 g/g and 48.7 %, respectively. The lipid content and fatty acids produced using VFAs as carbon sources were similar to those seen on growth and production using glucose. The techno-economic analysis indicates that the biodiesel derived from the lipids produced by heterotrophic C. protothecoides with VFAs as carbon sources is very promising and competitive with other biofuels and fossil fuels.  相似文献

A packed bed reactor (PBR) was fed with nitrate containing synthetic wastewater or effluent from a sequencing batch reactor used for nitrification. The C source introduced into the PBR consisted of volatile fatty acids (VFAs) produced from anaerobic acidogenesis of food wastes. When nitrate loading rates ranged from 0.50 to 1.01 kg N/m3·d, the PBR exhibited 100∼98.8% NO3 -N removal efficiencies and nitrite concentrations in the effluent ranged from 0 to 0.6 NO2 -N mg/L. When the PBR was further investigated to determine nitrate removal activity along the bed height using a nitrate loading rate less than 1.01 kg N/m3·d, 100% nitrate removal efficiency was observed. Approximately 83.2% nitrate removal efficiency was observed in the lower 50% of the packed-bed height. When reactor performance at a C/N ratio of 4 and a C/N ratio of 5 was compared, the PBR showed better removal efficiency (96.5%) of nitrate and less nitrite concentration in the effluent at the C/N ratio of 5. VFAs were found to be a good alternative to methanol as a carbon source for denitrification of a municipal wastewater containing 40 mg-N/L.  相似文献

Organic matters in sewage sludge can be converted into volatile fatty acids (VFAs) as renewable carbon sources. This work for the first time applied anthraquinone-2,6-disulfonate (AQDS) for enhancing VFA production from sewage sludge. With 0.066 or 0.33 g AQDS g−1 dried solids (DS), the yields for VFAs peak at 403 or 563 mg l−1, 1.9- or 2.7-fold to the control. The accumulated VFAs were principally composed of acetate and propionate. The AQDS enhances degradation rates of model proteins (bovine serum albumin), but had little enhancement on that of model polysaccharides (dextrans). The acidification step is proposed the rate-limiting step for VFA production from sewage sludge, in which the AQDS molecules shuttle electrons to accelerate the redox reactions associated with amino acid degradation. Methanogenic activities are inhibited in the presence of AQDS. The AQDS-assisted VFAs are renewable organic carbon sources, although their direct use for anaerobic digestion is not advised.  相似文献

The purpose of this study was to enhance the efficiency of anaerobic co-digestion with sewage sludge using pretreatment technologies and food waste. We studied the effects of various pretreatment methods (thermal, chemical, ultrasonic, and their combination) on hydrogen production and the characteristics of volatile fatty acids (VFAs) using sewage sludge alone and a mixture of sewage sludge and food waste. The pretreatment combination of alkalization and ultrasonication performed best, effecting a high solubilization rate and high hydrogen production (13.8 mL H2/g VSSconsumed). At a food waste:pretreated sewage sludge ratio of 2:1 in the mixture, the peak hydrogen production value was 5.0 L H2/L/d. As the production of hydrogen increased, propionate levels fell but butyrate concentrations rose gradually.  相似文献

Mesophilic and thermophilic anaerobic digesters (MD and TD, respectively) utilizing Gracilaria and marine sediment as the substrate and inoculum, respectively, were compared by analyzing their performances and microbial community changes. During three successive transfers, the average cumulative methane yields in the MD and TD were 222.6 ± 17.3 mL CH4/g volatile solids (VS) and 246.1 ± 11 mL CH4/g VS, respectively. The higher hydrolysis rate and acidogenesis in the TD resulted in a several fold greater accumulation of volatile fatty acids (acetate, propionate, and butyrate) followed by a larger pH drop with a prolonged recovery than in the MD. However, the operational stability between both digesters remained comparable. Pyrosequencing analyses revealed that the MD had more complex microbial diversity indices and microbial community changes than the TD. Interestingly, Methanomassiliicoccales, the seventh methanogen order was the predominant archaeal order in the MD along with bacterial orders of Clostridiales, Bacteriodales, and Synergistales. Meanwhile, Coprothermobacter and Methanobacteriales dominated the bacterial and archaeal community in the TD, respectively. Although the methane yield is comparable, both MD and TD show a different profile of pH, VFA and the microbial communities.  相似文献

The requirements for pH buffer addition for hydrogen production and acidogenesis in batch acidogenic digestion of a food waste (FW) feedstock with limited alkalinity was studied at various initial pH conditions (6.0–8.0). The results showed that, without buffer addition, hydrogen production from this feedstock was insignificant regardless of the initial pH. With buffer addition, hydrogen production improved significantly if the initial pH was greater than 6.0. Substantial hydrogen production occurred when the pH at the end of the batch digestion was higher than 5.5. The maximum hydrogen production was found to be 120 mL/g VS added when the initial pH was 6.5 and buffer addition was in the range of 15–20 mmol/g VS. The effect of pH buffering on the formation of volatile fatty acids (acetic acid, propionic acid and butyric acid) was similar to its effect on hydrogen production. The results of this study clearly indicated shifts in the metabolic pathways with the pH of fermentation. The changes in metabolic pathways impacted upon the dosage of buffer that was required to achieve maximum hydrogen generation.  相似文献

摘要:【目的】通过对污泥厌氧发酵pH调控,研究挥发性脂肪酸的累积、产酸微生物种群变化及产氢产乙酸菌群对乙酸产生的贡献。【方法】测定不同pH条件下污泥厌氧发酵过程中挥发性脂肪酸的累积;分别应用末端限制性片段长度多态性(T-RFLP)和荧光原位杂交技术(FISH)分析产酸系统中微生物种群结构的变化及产氢产乙酸菌的数量。【结果】 pH为10.0时,有机酸和乙酸的产率在发酵结束时分别达到652.6 mg COD/g-VS和322.4 mg COD/g-VS,显著高于其它pH条件。T-RFLP结果表明,pH值为12  相似文献
