Development of the plant body entails many pattern forming events at scales ranging from the cellular level to the whole plant. Recent evidence suggests that mechanical forces play a role in establishing some of these patterns. The development of cellular configurations in glandular trichomes and the rippling of leaf surfaces are discussed in depth to illustrate how intricate patterns can emerge from simple and well-established molecular and cellular processes. The ability of plants to sense and transduce mechanical signals suggests that complex interactions between mechanics and chemistry are possible during plant development. The inclusion of mechanics alongside traditional molecular controls offers a more comprehensive view of developmental processes.  相似文献

The endocannabinoids are endogenous agonists of the cannabinoid receptors and some members of the transient receptor potential, vanilloid type (TRPV), family of cation channels. Endocannabinoids along with their target receptors comprise a signaling system that is not well characterized. There have been many advances in our collective understanding of endocannabinoid signaling in the last decade and experimental evidence is mounting that pharmacological augmentation of endocannabinoid tone might have a significant therapeutic benefit in several disease states. However, the mechanisms responsible for the biosynthesis, cellular uptake, and intracellular processing of endocannabinoids are not well understood and have been the source of much debate. Recent studies have revealed a role for detergent insoluble membrane domains called lipid rafts in various aspects of signaling associated with the endocannabinoid anandamide. Intact detergent insoluble membrane domains appear to play a role in an anandamide-induced signaling cascade that is independent of G protein-coupled cannabinoid receptors or TRPV channels. Furthermore, detergent insoluble membrane domain-related endocytosis and recycling to lipid rafts appear to regulate the organization and localization of anandamide metabolites. We will discuss the implications that these findings have on the way we view endocannabinoid signaling, trafficking, and processing.  相似文献

Astrocytes constitute a major group of glial cells which were long regarded as passive elements, fulfilling nutritive and structural functions for neurons. Calcium rise in astrocytes propagating to neurons was the first demonstration of direct interaction between the two cell types. Since then, calcium has been widely used, not only as an indicator of astrocytic activity but also as a stimulator switch to control astrocyte physiology. As a result, astrocytes have been elevated from auxiliaries to neurons, to cells involved in processing synaptic information. Curiously, while there is evidence that astrocytes play an important role in synaptic plasticity, the data relating to calcium's pivotal role are inconsistent. In this review, we will detail the various mechanisms of calcium flux in astrocytes, then briefly present the calcium-dependent mechanisms of gliotransmitter release. Finally, we will discuss the role of calcium in plasticity and present alternative explanations that could reconcile the conflicting results published recently.  相似文献

Experimental evidence shows the importance of the immune system in controlling tumor appearance and growth. Immunotherapy is defined as the treatment of a disease by inducing, enhancing or suppressing an immune response. In the context of cancer treatment, it involves breaking tolerance to a cancer-specific self-antigen and/or enhancing the existing anti-tumor immune response, be it specific or not. Part of the complexity in developing such treatment is that cancers are selected to escape adaptive or innate immune responses. These escape mechanisms are numerous and they may cumulate in one cancer. Moreover, different cancers of a same type may present different combinations of escape mechanisms. The limited success of immunotherapeutics in the clinic as stand-alone products may in part be explained by the fact that most of them only activate one facet of the immune response. It is important to identify novel methods to broaden the efficacy of immunotherapeutics. Calcium signaling is central to numerous cellular processes, leading to immune responses, cancer growth and apoptosis induced by cancer treatments. Calcium signaling in cancer therapy and control will be integrated to current cancer immunotherapy approaches. Influenza viruses continue to pose a severe threat worldwide, causing thousands of deaths and an enormous economic loss every year. The major problem in fighting influenza is the high genetic variability of the virus, resulting in the rapid formation of variants that escape the acquired immunity against previous virus strains, or have resistance to antiviral agents. Every virus depends on its host cell and, hence, cellular functions that are essential for viral replication might be suitable targets for antiviral therapy. As a result, intracellular signaling cascades induced by the virus, in particular mitogen-activated protein kinase pathways, have recently come into focus.  相似文献

The symbiotic interaction between the soil bacteria Frankia and actinorhizal plants leads to the formation of nitrogen-fixing nodules resembling modified lateral roots. Little is known about the signals exchanged between the two partners during the establishment of these endosymbioses. However, a role for plant hormones has been suggested.Recently, we studied the role of auxin influx activity during actinorhizal symbioses. An inhibitor of auxin influx was shown to perturb nodule formation. Moreover we identified a functional auxin influx carrier that is produced specifically in Frankia-infected cells. These results together with previous data showing auxin production by Frankia lead us to propose a model of auxin action during the symbiotic infection process.Key words: lateral roots, nitrogen fixation, Frankia, AUX1, actinorhizal symbioses, phenylacetic acid, auxin influxActinorhizal symbioses result from the interaction between the soil actinomycete Frankia and plants belonging to eight angiosperm families collectively called actinorhizal plants.1 This symbiotic interaction leads to the formation of a new organ on the root system, the actinorhizal nodule, where the bacteria are hosted and fix nitrogen.2 Unlike legume nodules, actinorhizal nodules are structurally and developmentally related to lateral roots.3 Little is known about the signals exchanged between the two partners during the establishment of the symbiosis.2 Diffusible signals are emitted by Frankia at early stages of the interaction resulting in root hair deformation.2 The chemical nature of these signals remains unknown, however, detailed studies revealed that they are different from rhizobial Nod factors.4 Phytohormones are chemicals that control many developmental processes5 and have been linked to many plant-microbe interactions. In metapopulations, competitive interactions may extend beyond the confines of the local population such that members of neighbouring habitat patches affect each other adversely (quasi-local competition). We derive a model for quasi-local competition from first principles, assuming that individuals compete for shared resources and members of a population spend a certain fraction of their foraging time in the adjacent populations. Contrary to the results of Doebeli and Killingback [2003. Theor. Popul. Biol. 64, 397-416], our model does not produce spatial patterns of population densities in homogeneous environments. Quasi-local competition nevertheless contributes to pattern formation by amplifying the effect of heterogeneities in the external environment, and this amplification can be extremely strong when dispersal is absent. Postconditioning (POC), a novel strategy of cardioprotection against ischemia-reperfusion injury, is clinically attractive because of its therapeutic application at the predictable onset of reperfusion. POC activates several intracellular kinase signaling pathways, including phosphatidylinositol 3-kinase (PI3K)-Akt (RISK). The regulation of POC-induced survival kinase signaling, however, has not been fully characterized. JAK-STAT activation is integral to cardiac ischemic tolerance and may provide upstream regulation of RISK. We hypothesized that POC requires the activation of both JAK-STAT and RISK signaling. Langendorff-perfused mouse hearts were subjected to 30 min of global ischemia and 40 min of reperfusion, with or without POC immediately after ischemia. A separate group of POC hearts was treated with AG 490, a JAK2 inhibitor, Stattic, a specific STAT3 inhibitor, or LY-294002, a PI3K inhibitor, at the onset of reperfusion. Cardiomyocyte-specific STAT3 knockout (KO) hearts were also subjected to non-POC or POC protocols. Myocardial performance (+dP/dt(max), mmHg/s) was assessed throughout each perfusion protocol. Phosphorylated (p-) STAT3 and Akt expression was analyzed by Western immunoblotting. POC enhanced myocardial functional recovery and increased expression of p-STAT3 and p-Akt. JAK-STAT inhibition abrogated POC-induced functional protection. STAT3 inhibition decreased expression of both p-STAT3 and p-Akt. PI3K inhibition also attenuated POC-induced cardioprotection and reduced p-Akt expression but had no effect on STAT3 phosphorylation. Interestingly, STAT3 KO hearts undergoing POC exhibited improved ischemic tolerance compared with KO non-POC hearts. POC induces myocardial functional protection by activating the RISK pathway. JAK-STAT signaling, however, is insufficient for effective POC without PI3K-Akt activation.  相似文献

During vertebrate heart valve formation, Wnt/β-catenin signaling induces BMP signals in atrioventricular canal (AVC) myocardial cells and underlying AVC endocardial cells then undergo endothelial-mesenchymal transdifferentiation (EMT) by receiving this BMP signals. Histone deacetylases (HDACs) have been implicated in numerous developmental processes by regulating gene expression. However, their specific roles in controlling heart valve development are largely unexplored. To investigate the role of HDACs in vertebrate heart valve formation, we treated zebrafish embryos with trichostatin A (TSA), an inhibitor of class I and II HDACs, from 36 to 48 h post-fertilization (hpf) during which heart looping and valve formation occur. Following TSA treatment, abnormal linear heart tube development was observed. In these embryos, expression of AVC myocardial bmp4 and AVC endocardial notch1b genes was markedly reduced with subsequent failure of EMT in the AVC endocardial cells. However, LiCl-mediated activation of Wnt/β-catenin signaling was able to rescue defective heart tube formation, bmp4 and notch1b expression, and EMT in the AVC region. Taken together, our results demonstrated that HDAC activity plays a pivotal role in vertebrate heart tube formation by activating Wnt/β-catenin signaling which induces bmp4 expression in AVC myocardial cells.  相似文献

Starting from the Gierer–Meinhardt setting, we propose a stochastic model to characterize pattern formation on seashells under the influence of random space–time fluctuations. We prove the existence of a positive solution for the resulting system and perform numerical simulations in order to assess the behavior of the solution in comparison with the deterministic approach.  相似文献

Temporal patterns of immigration to the country were analysed using 668 alien species in the flora of the Czech Republic for which the dates of the first record were available (64.8% of the total number of 1031 so-called neophytes, i.e. aliens introduced after the year 1500). After a period of initial slow increase lasting to the 1840s, the accumulation of neophytes over time could be best fitted by a linear model that explained 97% of the variance. The intensity of floristic research, which varied between periods, did not significantly affect the overall increase in the number of aliens. The effect of species traits on the year of introduction was evaluated, with continent of origin, introduction type (deliberate or accidental), life history, Grime's life strategy, onset of flowering, mode of dispersal and propagule size as explanatory variables. Species of European origin and CSR strategists arrived earlier than those with other origins and strategies. Deliberately introduced species appeared earlier than accidental arrivals, and those cultivated for utilitary reasons on average arrived earlier than ornamentals. Species capable of early flowering were remarkably more prevalent among early newcomers. A separate analysis of accidentally introduced American species also identified life history as a significant predictor of immigration time, with annuals being introduced earlier than biennials and perennials. The data contribute to an understanding of a crucial stage of the invasion process that has received little attention in the literature. The model "early alien" to Central Europe is a European species with a CSR strategy deliberately brought for cultivation as a utilitary plant. Once it escaped from cultivation, its establishment in the wild was favoured by its ability to flower early and, therefore, complete the life cycle.  相似文献

The main function of eukaryotic microsomal monooxygenase (MMO) is thought to be the oxygenation of xenobiotics and hydrophobic endogenous substrates. However, there are important inconsistencies in the concept that the biological role of MMO is limited to the catalysis of these reactions. It is probable that MMO is also a regulated generator of reactive oxygen species (ROS) that is involved in both the initiation and execution of apoptosis. Additional support for this hypothesis came with the discovery of a role for cytochrome c (cyt c) in apoptotic signaling. This article introduces the theory that microsomal cytochrome b(5), which modulates the production of ROS in MMO, is among the principal interacting targets of cyt c. The role of this interaction in the initiation of apoptosis is discussed.  相似文献

The plant hormone auxin is central in patterning diverse plant tissues. The direction of auxin flow and the distribution of auxin within tissues are regulated by auxin efflux transporters that are polarly localized in cells.
