Recent reviews evidence that the naturally occurring compounds containing the chromone skeleton exhibit antiradical activities, providing protection against oxidative stress. The antioxidant activities of 13 new synthesized chromonyl‐2,4‐thiazolidinediones, chromonyl‐2,4‐imidazolidinediones and chromonyl‐2‐thioxoimidzolidine‐4‐ones were evaluated using in vitro antioxidant assays, including superoxide anion radical (), hydroxyl radical (), 2,2‐diphenyl‐1‐picryl‐hydrazyl free radical (DPPH?) scavenging capacity and total antioxidant capacity ferric ion reducing activity. Superoxide anion radical was produced using potassium superoxide/18‐crown‐6‐ether dissolved in dimethylsulfoxide, and the Fenton‐like reaction (Fe(II) + H2O2) was a generator of hydroxyl radicals. Chemiluminescence, spectrophotometry, electron paramagnetic resonance (EPR) and 5,5‐dimethyl‐1‐pyrroline‐N‐oxide (DMPO) as the spin trap were the measurement techniques. The results showed that the majority of the chromone derivatives tested showed a strong scavenging effect towards free radicals, similar to the chemiluminescence reaction with superoxide anion radical with a high activity, inhibition of the DMPO‐OOH radical EPR signal (24–58%), the DMPO‐OH radical EPR signal (4–75%) and DPPH radical EPR signal (6–100%) at 1 mmol/L. Several of the examined compounds exhibited the high reduction potentials. The results obtained show that the new synthesized chromone derivatives may directly scavenger reactive oxygen species and thus may play a protective role against oxidative damage. The use of triarylmethyl (trityl) free radical, TAM OX063, for detection of superoxide in aqueous solutions by electron paramagnetic resonance (EPR) spectroscopy was investigated. TAM is paramagnetic (EPR active), highly soluble in water and exhibits a single sharp EPR peak in aqueous media. It is also highly stable in presence of many oxidoreductants such as ascorbate and glutathione that are present in the biological systems. TAM reacts with superoxide with an apparent second order rate constant of 3.1 × 103 M−1 s−1. The specific reactivity of TAM with superoxide, which leads to loss of EPR signal, was utilized to detect the generation of superoxide in various chemical (light/riboflavin/electron/donor), enzymatic (xanthine/xanthine oxidase), and cellular (stimulated neutrophils) model systems. The changes in the EPR line-width, induced by molecular oxygen, were utilized in the simultaneous determination of consumption of oxygen in the model systems. The effects of flux of superoxide and concentration of TAM on the efficiency of detection of superoxide were studied. The use of TAM for detection of superoxide offers unique advantages namely, (i) the utilization of very low concentration of the probe, (ii) its stability to bioreduction, and (iii) its use in the simultaneous determination of concentrations of superoxide and oxygen.  相似文献

Free radical activity towards superoxide anion radical (), hydroxyl radical (HO?) and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH?) of a series of novel thiazolidine‐2,4‐dione derivatives (TSs) was examined using chemiluminescence, electron paramagnetic resonance (EPR) and EPR spin trapping techniques. 5,5‐Dimethyl‐1‐pyrroline‐N‐oxide (DMPO) was applied as the spin trap. Superoxide radical was produced in the potassium superoxide/18‐crown‐6 ether dissolved in dimethyl sulfoxide. Hydroxyl radical was generated in the Fenton reaction (Fe(II) + H2O2. It was found that TSs showed a slight scavenging effect (15–38% reduction at 2.5 mmol/L concentration) of the DPPH radical and a high scavenging effect of (41–88%). The tested compounds showed inhibition of HO? ‐dependent DMPO‐OH spin adduct formation (the amplitude of EPR signal decrease ranged from 20 to 76% at 2.5 mmol/L concentration. Superoxide radicals can be measured by redox methods which utilize the oxidation/reduction reactions of specific compounds. The redox methods, however, suffer from various interferences, which limit their use in the assay of superoxide. Electron paramagnetic resonance (EPR) spectroscopy using spin traps has been widely used as an alternative and direct technique to measure superoxide radicals. In our recent study, we have demonstrated the detection of superoxide in cellular system by EPR spectroscopy with triarylmethyl (trityl) free radical, TAM Ox063. TAM is highly water-soluble and stable in the presence of many biological oxidizing and reducing agents such as hydrogen peroxide, ascorbate, and glutathione. TAM reacts with superoxide with an apparent second order rate constant of 3.1x10(3)M(-1)s(-1). In the present work, we investigated the feasibility of a spectrophotometric assay of superoxide by taking advantage of the newly formed distinct absorption peak corresponding to the product formed from the reaction between TAM and superoxide. The effects of different fluxes of superoxide and concentrations of TAM on the efficiency and sensitivity of quantification of superoxide were investigated and compared with the widely used cytochrome c method of superoxide determination. The results demonstrated that the TAM method is comparable to the cytochrome c method for the assay of superoxide and further revealed that the assay is not affected by the presence of hydrogen peroxide. In summary, the TAM spectrophotometric assay of superoxide provides a suitable alternative method to the cytochrome c assay to measure superoxide and further complements our earlier reported TAM-EPR assay of superoxide.  相似文献

Abstract

New techniques and probes are routinely emerging for detecting short-lived free radicals such as superoxide radical anion (O2?–), nitric oxide (?NO), and transient oxidants derived from peroxynitrite (ONOO/ONOOH). Recently, we reported the profiles of oxidation products (2-hydroxyethidium, ethidium, and various dimeric products) of the fluorogenic probe hydroethidine (HE) in the ?NO/O2?– system (Zielonka et al. 2012). In this study, we used HPLC analyses of HE oxidation products in combination with continuous wave electron paramagnetic resonance (CW-EPR) spin trapping with 5-tert-butoxycarbonyl-5-methyl-1-pyrroline N-oxide (BMPO) to define the identity of the oxidizing species formed in the ?NO/O2?– system. EPR spin-trapping technique is still considered as the gold standard for characterization of free radicals and their intermediates. We monitored formation of BMPO-superoxide (BMPO-?OOH) and BMPO-hydroxyl (BMPO-?OH) radical adducts. Simultaneous analyses of results from EPR spin-trapping and HPLC measurements are helpful in the interpretation of the mechanism of formation of products of HE oxidation.  相似文献

The purpose of this study was to identify the antioxidant activity of 16 compounds isolated from Piper cubeba (CNCs) through the extent of their capacities to scavenge free radicals, hydroxyl radical (HO?), superoxide anion radical () and 2,2‐diphenyl‐1‐picrylhydrazyl radical (DPPH?), in different systems. Electron paramagnetic resonance (EPR) and 5,5‐dimethyl‐1‐pyrroline‐N‐oxide, DMPO, as the spin trap, and chemiluminescence techniques were applied. Using the Fenton‐like reaction [Fe(II) + H2O2], CNCs were found to inhibit DMPO? OH radical formation ranging from 5 to 57% at 1.25 mmol L?1 concentration. The examined CNCs also showed a high DPPH antiradical activity (ranging from 15 to 99% at 5 mmol L?1 concentration). Furthermore, the results indicated that seven of the 16 tested compounds may catalyse the conversion of superoxide radicals generated in the potassium superoxide/18‐crown‐6 ether system, thus showing superoxide dismutase‐like activity. Astaxanthin when esterified with ferulic acid is better singlet oxygen quencher with k2 = (1.58 ± 0.1) 1010 L mol? 1s? 1 in ethanol at 25°C compared with astaxanthin with k2 = (1.12 ± 0.01) 109 L mol? 1s? 1. The ferulate moiety in the astaxanthin diester is a better radical scavenger than free ferulic acid as seen from the rate constant of scavenging of 1-hydroxyethyl radicals in ethanol at 25°C with a second-order rate constant of (1.68 ± 0.1) 108 L mol? 1s? 1 compared with (1.60 ± 0.03) 107 L mol? 1s? 1 for the astaxanthin:ferulic acid mixture, 1:2 equivalents. The mutual enhancement of antioxidant activity for the newly synthetized astaxanthin diferulate becoming a bifunctional antioxidant is rationalized according to a two-dimensional classification plot for electron donation and electron acceptance capability.  相似文献

Abstract

Oxidation of tyrosine moieties by radicals involved in lipid peroxidation is of current interest; while a rate constant has been reported for reaction of lipid peroxyl radicals with a tyrosine model, little is known about the reaction between tyrosine and alkoxyl radicals (also intermediates in the lipid peroxidation chain reaction). In this study, the reaction between a model alkoxyl radical, the tert-butoxyl radical and tyrosine was followed using steady-state and pulse radiolysis. Acetone, a product of the β-fragmentation of the tert-butoxyl radical, was measured; the yield was reduced by the presence of tyrosine in a concentration- and pH-dependent manner. From these data, a rate constant for the reaction between tert-butoxyl and tyrosine was estimated as 6?±?1 × 107 M?1 s?1 at pH 10. Tyrosine phenoxyl radicals were also monitored directly by kinetic spectrophotometry following generation of tert-butoxyl radicals by pulse radiolysis of solutions containing tyrosine. From the yield of tyrosyl radicals (measured before they decayed) as a function of tyrosine concentration, a rate constant for the reaction between tert-butoxyl and tyrosine was estimated as 7?±?3 × 107 M?1 s?1 at pH 10 (the reaction was not observable at pH 7). We conclude that reaction involves oxidation of tyrosine phenolate rather than undissociated phenol; since the pKa of phenolic hydroxyl dissociation in tyrosine is ~ 10.3, this infers a much lower rate constant, about 3 × 105 M?1 s?1, for the reaction between this alkoxyl radical and tyrosine at pH 7.4.  相似文献

The reduction of alloxan (A) into dialuric acid (AH2) by?COO? radical has been investigated by pulse-radiolysis. A radical intermediate ?AH is shown to be formed with a rate constant k (?COO? + A) = 3.2 × 107 M?1 s?1. ?AH absorption spectrum is given. This radical disappears by disproportionation leading to dialuric acid, with a rate constant 2 k = 1.6 × 108 M?1s?1.  相似文献

Transient spectra and kinetic data of Tiron (1,2-dihydroxybenzene-3,5-disulphonic acid) are reported, obtained after pulse-radiolytic oxidation by hydroxyl radicals (°OH), superoxide anions (O2?) or a combination of both oxygen radicals. The rate constant with °OH radicals was determined at 1.0·109 M?1·s?1. Contrary to a previous report (Greenstock, C.L. and Miller, R.W. (1975) Biochim. Biophys. Acta 396, 11–16), the rate constant with O2? of 1.0·107 M?1·s?1 is lower by one order of magnitude; also the semiquinone absorbs at 300 nm rather than at 400 nm. The ratio of the rate constants with °OH and O2? of 100 again demonstrates that any oxidation reaction by the latter radical is unspecific due to the more efficient reaction of °OH radicals, leading to the same products with catechol compounds.  相似文献

During the aerobic xanthine oxidase reaction, O2? is produced and accumulates to a steady state determined by a balance between the rate of production of this radical and its rate of dismutation. Addition of ferricytochrome c then results in a biphasic reduction, the very rapid phase of which reflects reaction of the accumulated O2?, while the slower phase corresponds to the continuing production of this radical. Superoxide dismutase suppresses the accumulation of O2? during the xanthine oxidase reaction and thus diminishes the burst of reduction seen upon addition of ferricytochrome c. This effect has been utilized, at pH 10.2, as the basis of an assay that permits measurement of picomolar levels of superoxide dismutase. The rate of reaction between superoxide anion (O¯.2) and 1,2-dihydroxybenzene-3,5-disulfonic acid (tiron) was measured with pulse radiolysis-generated O¯.2. A kinetic spectrophotometric method utilizing competition betweenp-benzoquinoneand tiron for O¯.2 was employed. In this system, the known rate of reduction ofp-benzoquinonewas compared with the rate of oxidation of tiron to the semiquinone. From the concentration dependence of the rate of tiron oxidation, the absolute second order rate constant for the reaction was determined to be 5 · 108 M?·s?1. Ascorbat reduced O¯.2 to hydrogen peroxide with a rate constant of 108 M?1 · s?1 as determined by the same method. The tiron semiquinone may be used as an indicator free radical for the formation of superoxide anion in biological systems because of the rapid rate of oxidation of the catechol by O¯.2 compared to the rate of O¯.2 formation in most enzymatic systems.Tiron oxidation was used to follow the formation of superoxide anion in swollen chloroplasts. The chloroplasts photochemically reduced molecular oxygen which was further reduced to hydrogen peroxide by tiron. Tiron oxidation specifically required O¯.2 since O2 was consumed in the reaction and tiron did not reduce the P700 cation radical or other components of Photosystem I under anaerobic conditions.  相似文献

The mechanism for the hydroxyl-radical-induced depolymerization of cellulose under alkaline conditions in air was investigated using density functional theory at the B3LYP/6-31+G(d,p) level as well as electron transfer theory. The pathway for the depolymerization of cellulose was obtained theoretically and H abstraction from the C(3) atom of the pyran ring during the cleavage of the glucosidic bond was found to be the rate-limiting step due to its high energy barrier (16.81 kcal/mol) and low reaction rate constant (4.623?×?104 mol L?1 s?1). Calculations of the electron transfer between O2 and the saccharide radical performed with the HARLEM software package revealed that following the H abstraction, the oxygen molecule approaches C(2) on the saccharide radical and obtains an electron from the radical, even though no bond forms between the oxygen molecule and the radical. The rate constant for electron transfer could be as high as 1.572?×?1011 s?1. Furthermore, an enol intermediate is obtained during the final stage of the depolymerization.  相似文献

The synthesis and antioxidant evaluation of some novel benzimidazole derivatives (1024) are described. Antioxidant properties of the compounds were investigated employing various in vitro systems viz., microsomal NADPH-dependent inhibition of lipid peroxidation (LP), interaction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and scavenging of superoxide anion radical. Compounds 12 and 13 showed very good antioxidant capacity and were 17–18 -fold more potent than BHT (IC50 2.3 × 10? 4M) with 1.3 × 10? 5M and 1.2 × 10? 5M IC50 values, respectively, by interaction of the stable DPPH free radical.  相似文献

Radiation chemical studies of thioesculetin (1), a thioketone derivative of coumarin, were performed by both pulse radiolysis technique and DFT calculations. Hydroxyl (?OH) radical reaction with 1 resulted transients absorbing at 320, 360 and 500?nm. To identify the nature of the transients, the reaction was studied with specific one-electron oxidant (N3?) radical, where 360?nm band was absent. The transient absorption at 500?nm was concentration-dependent. The overall impression for ?OH radical reaction was that the transient absorbing at 320, 360 and 500?nm was due to sulphur centred monomer radical, hydroxysulfuranyl and dimer radical of 1 respectively. The equilibrium constant between the monomer to dimer radical was 3.75?×?104 M?1. From the transients’ redox nature, it was observed that 57 and 24% of ?OH radical yielded to oxidising and reducing products respectively. Further, the product analysis by HPLC suggested that the dimer radical disproportionate to esculetin and thioesculetin. DFT energy calculation for all the possible transients revealed that dimer radical has the lowest energy. The HOMO of 1 and its monomer radical suggested that the electron density was localised on the sulphur atom. The bond length between the two sulphur atoms in dimer radical was 2.88 Å which was less than the van der Waals distance. Bond order between the two sulphur atoms was 0.55, suggesting that the bond was two centre three electron (2c–3e). From TD-DFT calculation, the electronic transition of dimer radical was at 479?nm which was in close agreement with the experimental value. The nature of the electronic transition was σ → σ* from a 2c???3e bond.  相似文献

Summary

The reaction between peroxynitrous acid (hydrogen oxoperoxonitrate) and L-tryptophan is 130 M?1s?1 at 25°C. The pH dependence of the second-order rate constant shows a maximum at pH 5.1. The enthalpy and entropy of activation at pH 7.1 are 10.6 ± 0.4 kcal.mol?1 and -16 ± 2 cal.mol?1K?1 respectively. High-performance liquid chromatography analysis revealed a number of reaction products, two of which were identified as 5- and 6- nitrotryptophan. Hydroxytryptophans were not observed, even at low peroxynitrite concentrations where most of the peroxynitrite decays to nitrate via a first-order process. These results support the hypothesis that isomerization of protonated peroxynitrite to nitrate does not involve formation of the hydroxyl radical.  相似文献

Incubation in 8M urea (pH 7.4) inactivated yeast Cu, Zn superoxide dismutase with biphasic first order kinetics (k for the decrease from 100% to 16% activity = 6.5 × 10?3 min?1; k for the decrease from 16% to 0.1% activity = 2.5 × 10?3 min?1). The inactivation was fully reversible on dilution with or dialysis against urea-free buffer. No inactivation was shown to occur in similar experiments with the bovine Cu, Zn enzyme. EPR spectra recorded immediately after addition of 8M urea showed a more axial line shape and a higher A of the copper signal typical of the native enzyme. In the case of the yeast enzyme, this change was more pronounced and further incubation led to a new type of copper signal, typical of the inactivated enzyme. All EPR changes were reversible. Comparative analysis of the amino acid sequence of the two enzymes showed substantial identity of the protein regions contributing the ligands to the metals and the disulfide bridge. Differential destabilization of active sites by urea should be due to replacements in other protein segments, such as the three C-terminal and some N-terminal residues.  相似文献

The method of Electron Paramagnetic Resonance (EPR) spectroscopy was used to study the reaction of human methaemoglabin (metHb) with hydrogen peroxide. The samples for EPR measurements were rapidly frozen in liquid nitrogen at different times after H2O2 was added at 3- and 10-fold molar excess to 100 μM metHb in 50 mM phosphate buffer, pH 7.4, 37°C. Precautions were taken to remove all catalase from the haemoglobin preparation and no molecular oxygen evolution was detected during the reaction. On addition of H2O2 the EPR signals (- 196°C) of both high spin and low spin metHb rapidly decreased and free radicals were formed. The low temperature (- 196°C) EPR spectrum of the free radicals formed in the reaction has been deconvoluted into two individual EPR signals, one being an anisotropic signal (g° = 2.035 and g° = 2.0053), and the other an isotropic singlet (g = 2.0042, AH = 20 G). The former signal was assigned to peroxyl radicals. As the kinetic Pehaviour of both peroxyl (ROO*) and nonperoxyl (P*) free radicals were similar, we concluded that ROO* radicals are not formed from P* radicals by addition of O2. The time courses for both radicals showed a steady state during the time required for H2O2 to decompose. Once all peroxide was consumed, the radical decayed with a first order rate constant of 1.42 ± 10-3 s-1 (1:3 molar ratio). The level of the steady state was higher and its duration shorter at lower initial concentration of H2O2. The formation of the rhombic Fe(III) non-haemcentres with g = 4.35 was found. Their yield was proportional to the H2O2 concentration used and the centers were ascribed to haem degradation products. The reaction was also monitored by EPR spectroscopy at room temperature. The kinetics of the free radicals measured in the reaction mixture at room temperature was similar to that observed when the fast freezing method and EPR measurement at —196°C were used.  相似文献

The reaction of hydroxyl radicals (?OH) and superoxide anions (O2?) with methional were investigated by pulse-radiolytic methods. The second-order rate constant for the attack of OH was determined at 8.2×109 M?1 sec?1. In the case of O2? a slow first-order decay rate of 5.2×103 sec?1 suggests a far less efficient reaction. The transient species were identified by comparison with published results of pulse radiolysis and EPR spectroscopy of model compounds. The mechanism for the oxidation of methional by OH was found to be more complex than a simple fragmentation reaction.  相似文献
