Seedling performance is often a limiting factor in ecological restoration. Changes in the soil microbial community generated by invasive plants contribute to seedling failure. A method to remediate invasive species‐induced changes to the soil microbial community that results in increased native species seedling performance and decreased invasive species seedling performance could have a large impact on the success of many restoration efforts. In a greenhouse experiment, we first examined the changes in the soil microbial community created by invasive compared to native grasses. Then, we investigated four microbial treatments (bacterial inoculant, fungal inoculant, fungicide, and bactericide/fungicide) to remediate microbial plant–soil feedbacks (PSFs) created by invasive species Bromus inermis and Poa pratensis and increase the performance of natives Andropogon gerardii, Elymus canadensis, Pascopyrum smithii, and Schizachyrium scoparium. We found that the PSF mitigation treatments had some context‐dependent utility for restoration. For example, all of the treatments decreased the performance of B. inermis and fungal inoculant decreased the performance of P. pratensis. However, no single treatment increased the performance of all natives. Fungicide increased the performance of A. gerardii and E. canadensis in soil previously occupied by B. inermis and the performance of S. scoparium in soil previously occupied by P. pratensis. If validated in the field, PSF mitigation treatments may have utility for restoration practitioners.

Extensive tree mortality in forests can change the community composition of soil fungi altering seedling establishment, a process critical to forest restoration. Disturbances that result in the loss of ectomycorrhizal fungi, in particular, may impede the establishment of tree species reliant on these symbionts for their survival. Inoculation of seedlings with soil from intact forests may improve the establishment of seedlings in such disturbances but the method has rarely been tested in the field. Here, we assess whether soil inoculation improves lodgepole pine (Pinus contorta var. latifolia) seedling performance in conspecific stands with high levels of tree mortality caused by a mountain pine beetle (Dendroctonus ponderosae) outbreak and whether underlying soil type modifies inoculation effects. We first inoculated seedlings in a growth chamber with small amounts of soils (5% volume) originating from either intact (<10%) or “beetle-killed” (>70% pine basal area killed) conspecific stands or added no soil inoculum and, after 4 months, transplanted them into 15 beetle-killed stands. After two growing seasons, root-associated fungal communities of seedlings receiving inoculum from intact stands differed in composition from those receiving inoculum from beetle-killed stands or no inoculum. However, inoculation had no effect on seedling survival, height, or biomass. Site properties, including soil texture and the resident fungal community composition, overwhelmed the effect of soil inoculation on seedling performance. Seedling survival and shoot mass was higher in sandy than loamy soils. Restoration to improve seedling performance in beetle-killed stands should consider stand-level treatments as soil inoculation at the level evaluated was ineffective.

Soil organisms influence plant species coexistence and invasion potential. Plant-soil feedbacks occur when plants change soil community composition such that interactions with that soil community in turn may positively or negatively affect the performance of conspecifics. Theories predict and studies show that invasions may be promoted by stronger negative soil feedbacks for native compared with exotic species. We present a counter-example of a successful invader with strong negative soil feedbacks apparently caused by host-specific, pathogenic soil fungi. Using a feedback experiment in pots, we investigated whether the relative strength of plant-soil feedbacks experienced by a non-native woody invader, Sapium sebiferum, differed from several native tree species by examining their performance in soils collected near conspecifics ('home soils') or heterospecifics ('away soils') in the introduced range. Sapium seedlings, but no native seedlings, had lower survival and biomass in its home soils compared with soils of other species (negative feedback'). To investigate biotic agents potentially responsible for the observed negative feedbacks, we conducted two additional experiments designed to eliminate different soil taxa ('rescue experiments'). We found that soil sterilization (pot experiment ) or soil fungicide applications (pot and field experiments) restored Sapium performance in home soil thereby eliminating the negative feedbacks we observed in the original experiment. Such negative feedbacks apparently mediated by soil fungi could have important effects on persistence of this invader by limiting Sapium seedling success in Sapium dominated forests (home soils) though their weak effects in heterospecific (away) soils suggest a weak role in limiting initial establishment.

We have added glucose and nutrients to manipulate soil microbial activity and nutrient availability in a boreal spruce forest to study the performance of birch and spruce seedlings in relation to the soil microbial community. The proportion of aboveground biomass in the seedlings was largest in plots amended with extra nutrients, while ectomycorrhizal (ECM) colonisation was low in these plots. ECM appeared beneficial for growth of both species, but only at low levels of colonisation (<25% ECM colonised root-tips). The soil microbial biomass, as determined by total PLFA, was largest in plots treated with glucose and there was a significant negative relationship between birch seedling size and levels of total PLFA in soil. This could be taken to suggest that poor seedling growth was due to nutrient limitation caused by microbial assimilation. However, the treatment response of the birch seedlings was generally weak, and spruce often showed no response at all to the addition of nutrients and glucose. The most consistent parameter for the variation in plant performance, as well as for the microbial soil community, was the block-effect. This suggests a strong spatial structure in the soil microbial community, and that this structure was robust with respect to our treatments even though they continued over a 3-year period.

Seedling emergence is a critical stage in the establishment of desert plants. Soil microbes participate in plant growth and development, but information is lacking with regard to the role of microbes on seedling emergence. We applied the biocides (captan and streptomycin) to assess how seed mucilage interacts with soil microbial community and physiochemical processes to affect seedling emergence of Artemisia sphaerocephala on the desert sand dune. Fungal and bacterial community composition and diversity and fungal–bacterial interactions were changed by both captan and streptomycin. Mucilage increased soil enzyme activities and fungal–bacterial interactions. Highest seedling emergence occurred under streptomycin and mucilage treatment. Members of the phyla Firmicutes and Glomeromycota were the keystone species that improved A. sphaerocephala seedling emergence, by increasing resistance of young seedlings to drought and pathogen. Seed mucilage directly improved seedling emergence and indirectly interacted with the soil microbial community through strengthening fungal–bacterial interactions and providing favourable environment for soil enzymes to affect seedling emergence. Our study provides a comprehensive understanding of the regulatory mechanisms by which soil microbial community and seed mucilage interactively promote successful establishment of populations of desert plants on the barren and stressful sand dune.

The importance of mycorrhizal network (MN)-mediated colonization under field conditions between trees and seedlings was investigated. We also determined the combined influences of inoculum source and distance from trees on the ectomycorrhizal (EM) community of seedlings. On six sites, we established trenched plots around 24 residual Pseudotsuga menziesii var. glauca trees and then planted seedlings at four distances (0.5, 1.0, 2.5, and 5.0 m) from the tree into four mesh treatments that served to restrict inoculum access (i.e., planted into mesh bags with 0.5, 35, 250 μm pores or directly into soil). Ectomycorrhizal communities were identified after two growing seasons using morphological and molecular techniques. Mesh treatments had no effect on seedling mycorrhizal colonization, richness, or diversity, suggesting that MN-mediated colonization, was not an essential mechanism by which EM communities were perpetuated to seedlings. Instead, wind-borne and soil inoculum played an important role in seedling colonization. The potential for MNs to form in these forests was not dismissed, however, because trees and seedlings shared 83 % of the abundant EM. Seedlings furthest from trees had a simpler EM community composition and reduced EM richness and diversity compared to seedlings in closer proximity.

Cheatgrass ( Bromus tectorum L.) is an exotic annual grass that has invaded approximately 40,000,000 ha of rangelands in the United States, including montane ecosystems that are important habitats for wildlife and livestock. In addition to well-understood mechanisms by which Cheatgrass gains competitive advantage, recent studies have shown that Cheatgrass may also change the associated soil microbial community to impact native perennial plants and promote the persistence of Cheatgrass. Furthermore, reducing plant-available N represents a tool for initiating conditions that accelerate successional change from annual- to perennial-dominated communities. At a montane, mixed shrub–grassland Cheatgrass-dominated site in Colorado, we applied sucrose to reduce available N, and we added soil from a native plant community in order to reestablish the microbial community. This approach tested the idea that intact native soil microbial communities may enhance the beneficial effect of reducing soil N availability in a restoration setting. By the end of the experiment, reduced N availability decreased Cheatgrass by 9.8%, non-native annual/biennial plant cover by 15.0%, and increased relative perennial plant cover by 13.4%; soil inoculation reduced Cheatgrass by 7.6% and increased perennial abundance by 11.3%. Soil inoculum additions and reduced N availability both contributed toward restoring a perennial-dominated community and demonstrates that addition of native soil inoculum may be a useful tool for restoration efforts.

The soil seed bank is the basis for community establishment and permanence and plays a primary role in natural restoration of degraded or altered ecosystems. As part of a restoration project, this study aimed to quantify the soil seed bank and to evaluate the effect of the needle litter layer on seedling emergence. Soil samples from a pine plantation were collected at random in the field and set to germinate in a greenhouse. Half of them were covered by a 6cm layer of dead pine needles simulating field conditions. In the field, 20 x 20cm plots were established, half were left intact and half were cleaned from the litter needles. All four treatments had 15 replicates and seedling emergence was recorded during six months. Soil seed bank density was 1 222/m2 from 17 morphotypes. In the field, the number of morphotypes and seedlings was only 9% and 6% respectively, of those emerged in the greenhouse, possibly due to watering and lack of predation in the latter. In both cases, herbs and graminoids were the dominant emerging seedlings, making up to 70-90% of the total. The needle layer didn't prevent seeds from reaching the soil but strongly reduced (> 50%) seedling emergence, although high variability within treatments resulted in no statistically significant differences. These results show that the needle layer hinders germination and/or emergence of seedlings from the seed bank. Its removal may be a recommended technique to accelerate natural restoration in pine plantations.

Sub-alpine environments consist of altitudinal gradients associated with dramatic changes in plant growth and community composition, but the role of soil feedbacks and microbe interactions is largely unknown. Here, we examine the influence of the overall soil microbial community, with a focus on ectomycorrhizal and dark septate endophytic root colonizing fungi, from low, mid, and high elevations on the growth of Pinus contorta and Picea glauca × engelmannii. The influence of the soil microbial community was tested on seedlings from the same three elevations in order to determine ‘home’ versus ‘away’ effects on conspecifics of differing elevations. The low elevation soil was the most fertile and harbored a soil microbial community with an overall negative effect on seedling growth. In contrast, the high elevation soil was the least fertile and had a microbial community that enhanced seedling growth. However, only the soil microbial community in the highest elevation soil resulted in a stronger influence on the native P. contorta seedlings than seedlings originating from lower elevations. Despite the overall influence of the soil microbial community, ectomycorrhizal colonization was significantly correlated with P. glauca × engelmannii growth rates, but colonization by dark septate endophytes showed no relationship with seedling growth. The results provide evidence that plant—soil microbial community relationships are dependent on soil environment. Moreover, our results provide further support for the importance of soil microbes in facilitating seedling growth toward the edge of their elevational range.

Biological soil crusts (biocrust) are microbial communities that develop at the soil surface of drylands and play an important role in erosion control and fertility. Soil surface disturbance from a broad range of natural and human processes (e.g. fire, livestock grazing, off‐road traffic) cause significant losses in biocrust cover and associated ecosystems services. Hence, biocrust restoration is emerging as an important intervention strategy to rehabilitate degraded dryland soils. In a multistep process, we designed protocols for the establishment of “microbial nurseries” to produce photosynthetic cyanobacterial inoculum for biocrust seeding at scale. We first report on the strategy for isolation, directly from the target site, of a large culture collection of cyanobacteria that included multiple representatives of the five most common biocrust taxa. After genetic pedigreeing of these isolates, we could select those that best matched field populations genetically for scale‐up cultivation. We then developed protocols for effective cyanobacterial biomass production to obtain sufficient inoculum. This was followed by conditioning treatments (hardening off) to preacclimate this inoculum to the stressful conditions expected in the field. Finally, we show that the inoculum obtained was fit to thrive in its original soil under natural outdoor conditions if sufficient water was available. We repeated this process successfully for four sites, two in the hot Chihuanuan desert and two in the cooler Great Basin Desert, and on two textural types of soils in each. The cyanobacterial biocrust nursery approach represents a versatile, viable, and safe tool for the rehabilitation of dryland soils.

Soil communities are often degraded in mined sites, and facilitating the recovery of soil mutualists such as arbuscular mycorrhizal fungi (AMF) may assist with the restoration of native plants. At a grassland mine restoration site, I compared a commercial AMF inoculum with soil collected from beneath native grasses as a source of inoculum, as well as a control treatment. Field plots were broadcast‐inoculated and seeded with native grasses, and biomass of native and non‐native species was measured in three consecutive years. In addition, greenhouse‐grown seedlings of a native bunchgrass (Stipa pulchra) were inoculated with similar treatments, transplanted into the field, and assessed after 18 months. When broadcast inoculation was used, the local soil inoculum tended to increase non‐native grass biomass, and marginally decreased non‐native forb biomass in the second year of study, but did not significantly affect native grass biomass. Broadcast commercial inoculum had no detectable effects on biomass of any plant group. Stipa pulchra transplants had greater N content and mycorrhizal colonization, and marginally higher shoot mass and K content, when pre‐inoculated with local soil (relative to controls). Pre‐inoculation with commercial AMF increased AMF colonization of the S. pulchra transplants, but did not significantly affect biomass or nutrient content. The findings indicate that at this site, the use of local soil as an inoculum had greater effects on native and non‐native plants than the commercial product used. In order to substantially increase native grass performance, inoculation of transplanted plugs may be one potential strategy.

Vegetative inoculum of Amanita ovoidea (Bull.) Link and three isolates of Suillus collinitus (Fr.) Kuntze, as well as spore inoculum of Rhizopogon roseolus (Corda) Th. M. Fr. and S. collinitus, were evaluated for the production of Pinus halepensis Mill. in nursery and for the establishment of seedlings in a degraded gypsum soil. In nursery, most of the fungi significantly improved the height of seedlings and modified the accumulation of nutrients in needles. The percentage of ectomycorrhizas (ESR) per seedling ranged from 25 to 78%, depending on the fungi. One and 2 years after planting in the field, the survival of seedlings was significantly improved by inoculation with two isolates of S. collinitus and with spores of the same fungus. Inoculation with A. ovoidea had no significant effect on seedling survival, whilst R. roseolus caused a significant mortality of seedlings. Seedling height was significantly improved by inoculation with all fungi except R. roseolus and isolate CCMA-1 of S. collinitus. One year after planting, mycorrhization of control seedlings was negligible, and percentages of ESR were under 38% for the rest of treatments. In spring of the second year, seedlings in all treatments, including the control, became highly mycorrhizal (60–77% of ESR). Low ectomycorrhizal diversity (five morphotypes described) and seasonal variation on morphotype composition were detected 2 years after plantation. From a perspective of soil restoration management under limiting environmental conditions, nursery inoculation with selected fungi can be a key advantage for tree seedlings to surmount the initial transplant stress, assuring their establishment in the field. Our results emphasise the importance of selecting compatible fungal–host species combinations for nursery inoculation and sources of inoculum adapted to the environmental conditions of the transplantation site.

Restoration practitioners have a variety of practices to choose from when designing a restoration, and different strategies may address different goals. Knowledge of how to best use multiple strategies could improve restoration outcomes. Here, we examine two commonly suggested strategies in a single tallgrass prairie restoration experiment: increased forb sowing density and prairie soil inoculation. We designed a study with two different forb seeding densities. Within these densities, we transplanted seedlings into 1‐m2 plots that had been grown in either a whole prairie soil inoculum or sterilized prairie soil. After 4 years, we found positive effects of both high forb sowing density and inoculation treatments on the ratio of seeded to nonseeded plant cover in these plots, and negative effects of both treatments on nonseeded plant diversity. No effects of either treatment were seen on seeded plant diversity. Each strategy also affected the plant community in different ways: high forb sowing density increased seeded forb richness and decreased native nonseeded plant cover, while inoculation decreased non‐native cover, and tended to increase average successional stage of the community. These effects on restoration outcome were typically independent of each other, with the result that plots with both manipulations had the most positive effects on restoration outcomes. We thus advocate the combined use of these restoration strategies, and further studies which focus on both seeding and soil community manipulation in restoration.

The montane ecosystems of northern California have been subjected to repeated manipulation and active fire suppression for over a century, resulting in changes in community structure that contribute to increased wildfire hazard. Ecosystem restoration via reduction of stand density for wildfire hazard mitigation has received substantial attention in recent years; however, many ecological questions remain unanswered. This study compares belowground effects of two alternative forest thinning treatments designed to restore the large, old tree component of late‐seral structure, one of which focuses on restoring Pinus ponderosa dominance (Pine‐preference) and the other of which promotes development of large trees regardless of species (Size‐preference). We evaluated forest floor and soil chemical and microbial parameters in six experimental thinning treatment units of 40 ha each in the Klamath National Forest of northern California 5–6 years after thinning. Inorganic N availability, soil organic C content, phenol oxidase activity, and forest floor C:N ratio were greater in the Size‐preference treatment, whereas forest floor N and soil pH were greater in the Pine‐preference treatment. Our results indicate that these two thinning strategies produce differences in the soil environment that has the potential to affect growth rates of trees that remain, as well as the growth and survivorship of newly established seedlings. Thus, which species/individuals are removed during structural restoration of these mixed‐conifer forests matters both to the belowground components of the ecosystem today and the vegetation and productivity of the ecosystem in future decades.

Arbuscular mycorrhizal (AM) fungal communities can influence the species composition of plant communities. This influence may result from effects of AM on seedling recruitment, although the existing evidence is limited to experimental systems. We addressed the impact of AM fungi on the plant community composition and seedling recruitment of two species – Oxalis acetosella and Prunella vulgaris – in a temperate forest understory. We established a field experiment over two years in which soil fertility (using fertilizer to enhance and sucrose to decrease fertility) and the activity of AM fungi (using fungicide) was manipulated in a factorial design. Species richness, diversity and community composition of understory plants were not influenced by soil fertility or AM fungal activity treatments. However, plant community composition was marginally significantly affected by the interaction of these treatments as the effect of AM fungal activity became evident under enhanced soil fertility. Suppression of AM fungal activity combined with decreased soil fertility increased the number of shoots of herbaceous plants. Unchanged activity of AM fungi enhanced the growth of O. acetosella seedlings under decreased soil fertility, but did not influence the growth of P. vulgaris seedlings. We conclude that the role of AM fungi in structuring plant communities depends on soil fertility. AM fungi can have a strong influence on seedling recruitment, especially for those plants that are characteristic of the habitat.

In contrast to the extensive understanding of seed mucilage biosynthesis, much less is known about how mucilage is biodegraded and what role it plays in the soil where seeds germinate. We studied seed mucilage biodegradation by a natural microbial community. High‐performance anion‐exchange chromatography (HPAEC) was used to determine monosaccharide composition in achene mucilage of Artemisia sphaerocephala. Mucilage degradation by the soil microbial community from natural habitats was examined by monosaccharide utilization tests using Biolog plates, chemical assays and phospholipid fatty acid (PLFA) analysis. Glucose (29.4%), mannose (20.3%) and arabinose (19.5%) were found to be the main components of achene mucilage. The mucilage was biodegraded to CO2 and soluble sugars, and an increase in soil microbial biomass was observed during biodegradation. Fluorescence microscopy showed the presence of mucilage (or its derivatives) in seedling tissues after growth with fluorescein isothiocyanate (FITC)‐labelled mucilage. The biodegradation also promoted early seedling growth in barren sand dunes, which was associated with a large soil microbial community that supplies substances promoting seedling establishment. We conclude that biodegradation of seed mucilage can play an ecologically important role in the life cycles of plants especially in harsh desert environments to which A. sphaerocephala is well‐adapted.
